Summary An antifibronectin antibody has been prepared which recognises a fibronectin-like substance isolated from Helix aspersa hemolymph. By use of the indirect immunofluorescence technique, the distribution of fibronectin in embryos and in the ovotestis at selected development stages from hatching to the adult has been investigated. In embryos, the basement membranes and the epithelia were immunoreactive, whereas the mesenchyme and the gonadal rudiment were not. After hatching, the fibronectin was present in ovotestis. It was localized on the epithelia of gonadal acini and at the periphery of the periacinar vesicular tissue. This adhesive molecule is present on the nurse cells, which sustain the group of male cells, while it is absent on differentiating male cells in the lumen of acini. The membrane of oocyte did not exhibit fluorescence. By contrast the surface of follicular cells were clearly labelled. Inside the vitellogenic oocytes, granular fluorescence was also observed.The participation of fibronectin in gonadal organogenesis is discussed in relation to cellular adhesion and movement. Its role in the metabolic exchanges between the germinal cells and the surrounding tissues is suggested.  相似文献

Summary With an antiserum (aFM) against the molluscan cardio-active FMRFamide (Phe-Met-Arg-Phe-NH2) numerous immunoreactive axons were found in the outer, longitudinal, muscle layer of the anterior aorta of Helix aspersa. Immunoreactive axons were rare in the inner, circular, muscle layer. At the ultrastructural level four types of axons could be distinguished. The granules containing the immunoreactive substance (mean diameter ca. 100 nm) are present in type-2 axons. The effect of synthetic FMRF-amide was tested in vitro on preparations of ring- and tubule-shaped pieces of the anterior aorta. Physiological doses (3 × 10-7 M) provoked contractions of the circular muscle fibres, but had no effect on the longitudinal muscle cells. Apparently in vivo the FMRF-like substance diffuses from the richly innervated longitudinal muscle layer to the circular muscle layer, where it exerts its effect. This conclusion is sustained by the observation that the contents of the aFM-immunoreactive granules in type-2 axons are released by exocytosis in a non-synaptic fashion.  相似文献

Summary The epineural connective tissue sheath investing the subesophageal ganglion of Helix aspersa consists of a superficial region and a deeper region. The superficial region contains masses of globular cells intermingled with smooth muscle cells and nerve fibers all embedded in a connective tissue matrix. The histochemical and fine structural features of the globular cells show seasonal changes. During autumn to winter glycogen accumulates in their cytoplasm; this accumulation is accompanied by the appearance of dense, cytoplasmic globules which fuse together and ultimately form large pools of granular material. All the organelles and cytoplasm are displaced towards the cell periphery. Various cell-membrane invaginations containing dense material are prominent but there is no direct evidence to link these structures with the uptake of metabolites for glycogenesis. In winter there is a concentration of homogeneous, membrane-bound inclusions in the vicinity of the Golgi bodies. It is suggested that these inclusions constitute a lipid store. They decrease in number during summer. The globular cells do not bear any intimate relation to neurons and there is no reason to include these cells in the neuroglia. The muscle cells often weave around the globular cells but there is no direct contact. Nerve fibers innervate at least some of the muscle cells. The connective tissue consists of large and small diameter fibers suggesting that maturation of the fibrous components of the intercellular matrix is taking place in the superficial regions of the epineural sheath.This work has been supported by the Australian Research Grants Committee.  相似文献

Summary Ultrastructural, cytochemical and autoradiographic techniques were used to study the functional significance of Ca2+-accumulating vesicles in Helix aspersa oocytes. The organic material of these vesicles is capable of adsorbing calcium ions, thus removing them from the cytosol and forming osmotically inactive complexes that are concentrated in the core. Ca2+-accumulating vesicles likely act as subcellular detoxifying compartments that sequestrate excess cytosolic Ca2+ (and plausibly other metal cations) to maintain physiologically adequate ionic concentrations.Part of this paper was presented at the VII Congreso Nacional de Malacología (Sevilla, November 1988)  相似文献

Summary Morphologically the surface specializations of the epithelium covering the dorsal head and ventral foot regions in Helix aspersa consists either of cilia or microvilli respectively. The epithelium at the tip of the optic tentacle is a simple one. Each epithelial cell has a number of cilia-like projections from their free surfaces. These projections usually branch at their tips into two or three slender, microvilli-like structures. From the bases of the cilia-like projections arise numerous, tubular processes which form a thick, spongy layer interspersed between these projections. The microvilli-like structures are immersed in a fine, fibrous mat; unlike the fibrous mats on the dorsal head and ventral foot epithelia this material does not autofluoresce. It is suggested that it arises from the collar cells and not from typical mucocytes. The functional relationship between these surface specializations of the optic tentacle epithelium and the abundance of sensory axons in this region is discussed. These epithelial cell projections on the tentacle probably function not only as a protective covering but also to create a fluid trap for odours in the ambient air. The various contacts between epithelial cells serve to maintain the integrity of the epithelium while allowing for stretching due to protrusion of the tentacle.This work has been supported by the Australian Research Grants Committee.  相似文献

In natural populations of Thymus vulgaris in the south of France, six genetically different chemical chemotypes occur, each differentiated by a single dominant monoterpene that gives individual plants a characteristic smell and taste. Individual populations may contain all six chemotypes, but are often dominated by one to three chemotypes. We tested the hypothesis that this chemical polymorphism was associated with selective herbivory by the snail Helix aspersa, which feeds upon T. vulgaris in nature. The relative preference of juvenile and adult molluscs was investigated by presenting individual animals with a choice of six chemotypes in three experimental trials: (1) whole plants, (2) nutritive gels containing crushed thyme leaves, and (3) nutritive gels containing a distilled monoterpene as an additive. We found significant and consistent patterns of preference. Adult and immature molluscs preferred the linalol chemotype, and ate carvacrol and thymol-containing foods the least. This preference was more marked when using the gels than the real plants, particularly the monoterpene-based gels, demonstrating the importance of the monoterpene for the choice. Furthermore, molluscs fed exclusively on linalol-containing food gained weight whereas those fed on carvacrol-containing food lost weight. After being fed these exclusive diets both groups showed a significant preference for linalol. These results (1) demonstrate that food choice is based upon the presence of specific monoterpenes and is not learned, (2) suggest that phenolic monoterpenes such as carvacrol are repellent to the animals, and (3) indicate that selective herbivory may play a significant role in the maintenance of the chemical polymorphism of T. vulgaris.  相似文献

Tobacco farmers are routinely exposed to complex mixtures of inorganic and organic chemicals present in tobacco leaves. In this study, we examined the genotoxicity of tobacco leaves in the snail Helix aspersa as a measure of the risk to human health. DNA damage was evaluated using the micronucleus test and the Comet assay and the concentration of cytochrome P450 enzymes was estimated. Two groups of snails were studied: one fed on tobacco leaves and one fed on lettuce (Lactuca sativa L) leaves (control group). All of the snails received leaves (tobacco and lettuce leaves were the only food provided) and water ad libitum. Hemolymph cells were collected after 0, 24, 48 and 72 h. The Comet assay and micronucleus test showed that exposure to tobacco leaves for different periods of time caused significant DNA damage. Inhibition of cytochrome P450 enzymes occurred only in the tobacco group. Chemical analysis indicated the presence of the alkaloid nicotine, coumarins, saponins, flavonoids and various metals. These results show that tobacco leaves are genotoxic in H. aspersa and inhibit cytochrome P450 activity, probably through the action of the complex chemical mixture present in the plant.  相似文献

The distribution of neuropeptide F (NPF) immunoreactivity in the snail, Helix aspersa, has been demonstrated by immunocytochemistry using 2 regionspecific antisera. One, designated NPF3, was raised against a synthetic N-terminal fragment of Helix aspersa NPF; the other, designated PP221, was raised against the C-terminal hexapeptide amide of mammalian pancreatic polypeptide (PP) but cross-reacts fully with the analogous C-terminal region of Helix aspersa NPF. The distribution of NPF immunoreactivity has also been compared with that of FMRFamide using alternate serial sections of Helix aspersa ganglia. Results showed that NPF immunoreactivity was abundant and widespread in the central and peripheral nervous systems and the pattern of immunostaining obtained using both region-specific antisera was similar. Likewise, immunocytochemistry of neural tissues of a congeneric species, Helix pomatia, and 2 prosobranch gastropods, Buccinum undatum and Littorina littorea, produced similar staining patterns with both antisera. However, in the cephalopod mollusc, Loligo vulgaris, and the cestode, Moniezia expansa, positive immunostaining was only obtained with the C-terminal PP antiserum. Immunostaining of alternate serial sections of Helix aspersa ganglia with NPF3, and an antiserum raised to FMRFamide, showed that while a few neurones were immunoreactive with one antiserum only, in the majority, both immunoreactivities were co-localised. NPF thus appears to be an important neuropeptide of widespread distribution in Helix aspersa and the differential immunocytochemical staining obtained using the 2 region-specific antisera would suggest a high degree of primary structural conservation within the gastropod molluscs, but lack of conservation of the N-terminal region of the peptide in other invertebrate groups.  相似文献

The land snail Helix aspersa is a partially freezing tolerant species whose supercooling ability is limited to ca. -3 to -5 degrees C. One hundred adult snails were subjected to the following two experimental conditions: (i) a starved group, provided with water; (ii) an antibiotic-treated group that was provided with a solution containing a mixture of two antibiotics. The antibiotic group exhibited a T(c) significantly lower than the starved group (-3.94 +/- 1.32 degrees C, n = 40 and -3.07 +/- 0.99, n = 30, t test, p < 0.005). This study showed that bacteria of the gut are likely to elevate animal supercooling points. It is also the first report in which a possible ice-nucleating activity of the gut microflora in a land snail has been suggested by the action of antibiotics on the T(c).  相似文献

Enzyme histochemistry and immunocytochemistry were used to determine the distribution of neurons in the snail Helix aspersa which exhibited nicotinamide adenine dinucleotide phosphate (NADPH) diaphorase activity and/or immunoreactivity to nitric oxide synthase (NOS). NADPH diaphorase-positive cells and fibres were distributed extensively throughout the central and peripheral nervous system. NADPH diaphorase-positive fibres were present in all neuropil regions of the central and peripheral ganglia, in the major interganglionic connectives and in peripheral nerve roots. NADPH diaphorase-positive cell bodies were found consistently in the eyes, the lips, the tentacular ganglia and the procerebral lobes of the cerebral ganglia; staining of cell bodies elsewhere in the nervous system was capricious. The distribution of NOS-like immunoreactivity differed markedly from that of NADPH diaphorase activity. Small clusters of cells which exhibited NOS-like immunoreactivity were present in the cerebral and pedal ganglia; fibres which exhibited NOS-like immunoreactivity were present in restricted regions of the neuropil of the central ganglia. The disjunct distributions of NADPH diaphorase activity and NOS-like immunoreactivity in the neurvous system of Helix suggest that the properties of neuronal NOS in molluscs may differ sigificantly from those described previously for vertebrate animals.  相似文献

The aim of this study was to determine the presence and prevalence of larval stages of Dicrocoelium dendriticum and Brachylaima sp. in the first intermediate host, a species of land snail, Helix aspersa, in Turkey. A total of 211 snails were collected in April-May 2014 from pastures in Mersin District. Larval stages of D. dendriticum were identified under a light microscope. Hepatopancreas from naturally infected H. aspersa snails were examined histologically. The prevalence of larval stages of D. dendriticum and Brachylaima sp. in H. aspersa snails was found to be 2.4% and 1.9%, respectively, in Mersin, Turkey. Cercariae were not matured in sporocysts at the beginning of April; however, it was observed that cercariae matured and started to leave sporocysts by early-May. Thus, it was concluded that H. aspersa acts as an intermediate host to D. dendriticumin and Brachylaima sp. in Mersin, Turkey. A digenean trematode Brachylaima sp. was seen for the first time in Turkey.  相似文献

The digestive gland of adult land snails, Helix aspersa, sampled from four different sites in São Miguel island (Azores) was submitted to chemical analyses, autometallography and haemalum/eosin staining, in order to quantify the relative abundance of heavy metals, calcium cells and connective tissue cells. Metals were visualized, through light microscopy, as black silver deposits mostly in the connective tissue cells. Metal levels, essentially of Cu and Fe, were related to the relative volumetric density of connective tissue cells but not to the relative volumetric density of calcium cells from the digestive gland epithelium. Thus, the connective tissue index presented herein is suggested as a biomarker of Cu exposure in terrestrial mollusks.  相似文献

Summary

The experimental laboratory pairing of hermaphroditic snails of different sub-species, Helix aspersa aspersa (H.a.a) and H. aspersa maxima (H.a.m.), made it possible to obtain both reciprocal hybrids for the first time and to describe their growth. The relatively small number of matings observed (20–25%) was partly responsible for the small number of clutches obtained (10–13%). The cross H.a.a.M × H.a.m.F is more fertile than the reciprocal. The observed tendency to reproductive isolation (behavioural and anatomical) suggests that H.a.a. and H.a.m. should be considered as two sub-species. Hybridization is not accompanied by a beneficial heterosis effect since significant juvenile mortality and an increased variation in growth were observed. The small size of H.a.a. is a dominant characteristic, whereas the colour of the mantle edge in the Fl individuals is intermediate between the black of H.a.m. and the white of H.a.a. The Fl generation is fertile and gives F2 snails with an adult size close to that of H.a.a. but slower growing. This approach allows investigations into the mechanisms underlying the reproductive isolation of parental sub-species.  相似文献

1. We report the isolation of a peptide from the brain of the snail Helix aspersa by radioimmunoassay using an antisomatostatin.2. The sequencing of an immunopositive fraction showed the presence of a new tridecapeptide, termed Helix cardioinhibitory peptide (HCIP), with the following primary structure : H-Val-Phe-Gln-Asn-Gln-Phe-Lys-Gly-Ile-Gln-Gly-Arg-Phe-NH2. It is structurally related to the Achatina cardioexcitatory peptide (ACEP-1) and the terminal-ammo acid sequence of HCIP is identical to that of FMRFamide family peptides.3. The synthetic HCIP was tested on heart and neuronal activities and it was found to have inhibitory actions not only on the ventricle but also on visceral neurons of the central nervous system of Helix. Immunocytochemical investigation indicates its presence in visceral and parietal ganglia, in which cells taking part in the regulation of the heartbeat have been previously identified .  相似文献

Summary The morphology, axonal arborization and ultrastructure of synaptic connections of the V21 giant neuron in the visceral ganglion of the snail Helix pomatia has been investigated following intracellular labelling with horseradish peroxidase. The V21 neuron establishes several afferent and efferent axo-axonic connections, mainly along the first half of the primary axon. Collaterals of 200–300 m length originate from the primary axon, which shows further arborization, and both afferent and efferent synaptic contacts are formed on these fine axon profiles. Afferent and efferent contacts of the cell occur within very short distances of a few micrometers. On the basis of ultrastructure and vesicle and granule content, several afferent terminals can be distinguished on V21 labelled axon profiles. The majority of these afferent terminals resembles peptidergic-(neurosecretory)-like terminals. This finding supports the possible transmitter role of neuropeptides in the central nervous system of gastropods. Our results are consistent with and provide morphological evidence for recent electrophysiological observations suggesting that, in addition to integrating input, the V21 neuron functions as an interneuron in Helix central nervous system.  相似文献

Summary At hatching, the hermaphrodite duct of Deroceras reticulatum consists of a single cell type designated the Gonadal Stem Cell (GSC). Proliferation of the GSC leads to the formation of numerous ductules each of which forms one of the acini of the gonad. The germinal and supporting cells are derived entirely from the GSC. The germ cells differentiate first, followed by the Sertoli and follicle cells. At the early sperm stage of gonadal development the hermaphrodite duct differentiates to function as a seminal vesicle. Once the GSC are committed to this change they lose their regenerative ability. The only remaining GSC are the cells of the acinar epithelium, and these retain their germinal potential until the death of the animal.Regeneration will occur from the hermaphrodite duct provided it is in the immature state, i.e., composed of GSC, and is exposed to the hormonal conditions of a young animal. Nervous connections and the presence of an artery are not necessary for this regeneration. The presence of a functional gonad does not inhibit regeneration.  相似文献

Summary

The activity of the endocrine dorsal bodies (DB) of the adult land snail Helix aspersa living in the field shows striking fluctuations during a 24 hr cycle. Quantitative electron microscopical data revealed that the number of secretory granules, the volume of the Golgi apparatus and the number of Golgi saccules containing electron-dense material were maximal at 1 am and minimal at 1 pm. The use of tannic acid indicated the exocytosis of secretory material was intense around 1 pm and only moderate at 1 am. The results suggest that, under natural conditions, the DB have a diurnal rhythm of activity, packaging secretory material into secretory granules mainly during the night and releasing it during the afternoon.  相似文献
