The levels of prostaglandin F found in human and rabbit plasma were determined by the chemically modified bacteriophage assay.Prostaglandin F2α was coupled covalently to bacteriophage T4 using carbodiimide as cross linking agent and the conjugated phage could be inactivated by anti-prostaglandin F2α antibodies. Prostaglandins specifically inhibited the modified phage inactivation caused by antiserum and as little as 200 picograms of prostaglandin F2α could be detected by this system. Anti-prostaglandin F2α antibodies had a high specificity toward prostaglandin F2α and exhibited a very low degree of cross reaction to the other prostaglandin derivatives. The concentration of the extracted prostaglandin F2α from the plasma containing exogenous prostaglandin F2α could be determined with a high recovery.In normal human males and in male rabbits, 0.300.82 and 0.421.22 nanograms of prostaglandin F per ml of plasma were found, respectively. These levels of prostaglandin F in plasma agree with those determined by the radioimmunoassay.  相似文献

A new injectable biodegradable hydrogel system with thermosensitive sol-gel transition behavior was developed. A series of A-B-A triblock copolymers consisting of Pluronic copolymer end-capped with D- or L-lactic acid oligomers (PL-LA(n)) with various chain lengths (n = 5,12) was synthesized. It was assumed that a pair of two triblock copolymers with enantiomeric oligolactide chains, when blended in an equimolar mixture, would form more stable, self-assembled, and stereocomplexed (ST) hydrogels. A series of blend hydrogels encapsulating human growth hormone (hGH) was prepared by varying blend ratios between PL and stereocomplexed PL copolymers. They showed sustained release of hGH via an erosion-dependent mechanism. The hydrogel with a 5% blending ratio exhibited the most delayed mass erosion as well as sustained protein release patterns in vitro possibly due to the formation of a fish-net like 3-D mesh structure. The effect of incubation condition on hGH release and degradation behaviors was also assessed.  相似文献

Background and aims

7Be has been used as a powerful tracer for estimating short-term soil redistribution by virtue of its short half-life. However, the existing conversion model associated with this radionuclide means that it can only be applied to bare soils because vegetation will intercept a large proportion of 7Be fallout. A modified model which takes into consideration the impact factor of vegetation was reported in this paper and the estimation of soil redistribution was compared by using both the conventional and the modified models.

Methods

Field experiment on 7Be distribution in above-ground grasses and soils was carried out on a 100 m2 grass-covered slope. The vegetation interception factor (P) was determined and the soil redistribution rates were calculated by using the previous model and the modified model.

Results

The result shows that nearly 40 % of the atmospherically deposited 7Be will be sequestered by leaf surfaces of herbaceous plants. Soil loss rates on grassland will be remarkably overestimated by using the previous model.

Conclusions

The net soil loss estimated from the modified model is more accurate than that derived from the conventional model and the modified model will be more appropriate to estimate soil redistribution rates on soils with significant vegetation cover by using 7Be technique.  相似文献

The standard methods for computing the number of nonsynonymous substitutions (Ka) lump all amino acid changes into one single class, even though their rates of substitution vary by at least 10-fold (Tang et al., 2004). Classifying these changes by their physicochemical properties has not been suitably effective in isolating the fastest evolving classes of changes. We now propose to use the Universal index U of Tang et al. (2004) to classify the 75 elementary amino acid changes (codons differing by 1 bp) by their evolutionary exchangeability. Let Ki denote the Ka value of each class (i = 1, ..., 75 from the most to the least exchangeable). The cumulative Ki for the top 10 classes, denoted Kh (for high-exchangeability types), has two important properties: (1) Kh usually accounts for 25%-30% of total amino acid changes and (2) when the observed number of amino acid substitutions is large, Kh is predictably twice the value of Ka. This shall be referred to as the twofold approximation. The new method for estimating Kh is applied to the comparisons between human and macaque and between mouse and rat. The twofold approximation holds well in these data sets, and the signature of positive selection can be more easily discerned using the Kh statistic than using Ka. Many genes with Ka/Ks > 0.5 can now be shown to have Kh/Ks > 1 and to have evolved adaptively, at least for the high-exchangeability group of amino acid changes.  相似文献

A spectrophotometric method is described for estimating the carbohydrate content of acid-hydrolyzed glycoproteins. The method is based on the alkaline ferricyanide reaction with reducing sugars and is accurate in the 1–25 nmole range. Using a simple two-step ion-exchange column procedure both the total sugar content and the proportion of neutral and amino sugars can be determined.  相似文献

Chemically modified bacteriorhodopsin (BR) films embedded in gelatin matrix were fabricated. It was found that chemically modified BR/gelatin film remained the original conformation of BR and possessed homogeneity. Then, the photochemical conversion from all-trans O state to 9-cis P state in the chemically modified BR film/gelatin was investigated. For comparison purpose, the same conversion in the wild-type BR (BRwt) film was also presented here. To our know, it is the first report to show that the absorption changes of the P state in the chemically modified BR film were larger than in the BRwt film upon illumination with red light at the low actinic power density.  相似文献

A method whereby sera can be screened for the presence of lymphocytotoxic antibodies within 4 hr using lymphocytes frozen in microtest trays is described. The reactions of the sera of 48 hemodialysis patients against freshly prepared lymphocytes were compared with those against tube-frozen (384 reaction pairs) and tray-frozen (864 reaction pairs) cells. There was a better than 90% concordance, and only 3% of the reactions differed from negative to strongly positive or vice versa. Preliminary results indicate that the method is also suitable for B-cell (HLA-DR) antibody testing. Routine screening against 50- or 100-cell panels can be accomplished more rapidly and more efficiently using tray-frozen lymphocytes. The data matrix for analysis of a serum's HLA specificity is greatly reduced in comparison to the conventional screening technique, rendering the method particularly suitable for microcomputer processing.  相似文献

Summary A modified approach of the glyoxylic acid (GA) condensation reaction for the visualization of biogenic amines in tissue is described.Cryostat sections are used from brain or extracerebral tissue in dog, monkey, rat and mouse and exposed for 3 s to a room temperature solution containing sucrose-potassium phosphate-glyoxylic acid (SPG). The tissues are air dried and heated in an oven for 5 min. The complete processing time from fresh tissue to microscopic examination takes 18 min. Morphologically sharp and brightly fluorescent monoamine-containing neurons, pre-and terminal axons are seen against a dark parenchymal background without drug pre-treatment. The SPG method retains the high specific sensitivity for monoamines previously described in the original technique but is, in addition, more rapid and simple and is easily accessible as a research tool to investigators inexperienced in histofluorescence techniques.  相似文献

Determining the amount of recombination in the genealogical history of a sample of genes is important to both evolutionary biology and medical population genetics. However, recurrent mutation can produce patterns of genetic diversity similar to those generated by recombination and can bias estimates of the population recombination rate. Hudson 2001 has suggested an approximate-likelihood method based on coalescent theory to estimate the population recombination rate, 4N(e)r, under an infinite-sites model of sequence evolution. Here we extend the method to the estimation of the recombination rate in genomes, such as those of many viruses and bacteria, where the rate of recurrent mutation is high. In addition, we develop a powerful permutation-based method for detecting recombination that is both more powerful than other permutation-based methods and robust to misspecification of the model of sequence evolution. We apply the method to sequence data from viruses, bacteria, and human mitochondrial DNA. The extremely high level of recombination detected in both HIV1 and HIV2 sequences demonstrates that recombination cannot be ignored in the analysis of viral population genetic data.  相似文献

A technique is described for staining DNA in polyacrylamide gels with silver. It is rapid, requiring about 30 min for whole staining and development procedure, very simple and at least 20 times more sensitive than ethidium bromide for the staining of double-stranded DNA in polyacrylamide gels. This technique can also be applied for the staining of denatured, single-stranded DNA as well as RNA after their electrophoretic separation on polyacrylamide gels, having the same sensitivity as for double-stranded DNA fragments.  相似文献

Abstract A novel method has been developed for concentrating and enumerating bacteriophages in water. Host bacterial cells are added to water samples to adsorb phage and then the infected bacteria are collected by centrifugation at 4000 rev./min for 30 min. The pellets are resuspended in small volumes and assayed for phages using the agar-overlay method. This procedure was more successful than three established techniques in recovering Bacillus phages from seeded samples of tap water. It also gave efficient recovery of phages from samples of river, lake and sea-water.  相似文献

The fermentation-inhibition (FI) test for Mycoplasma pneumoniae was improved by using a combination of guinea pig complement and gamma globulin-depleted horse serum in place of unheated whole horse serum employed in the conventional assay system. As the test antigen for the new FI assay system, M. pneumoniae filtrated through a 3.0 microns membrane filter was used. Owing to the strong augmenting effect of guinea pig complement, the FI activity of rabbit immune serum was increased 32-fold in the new system compared with the conventional system. Furthermore, IgM antibody, which is barely detectable by the conventional system, could easily be titrated by the new system. With this sensitive method, rapid rise of FI titer was clearly demonstrable in most children with acute M. pneumoniae infections, and a prevalence of FI or growth-inhibitory antibody among healthy adults in Japan (82%) was revealed.  相似文献
