Detached rice(Oryza sativaL cv Mousouri)leaves were induced to senesce in darkness at 0°C(cold), 27°C(room temperature) and 40°C(heat). 2,6-dichlorophenol indophenol (DCPIP) Hill reaction activity of chloroplasts isolated from senescing leaves under all experimental temperatures with H2O, Mn2+ or diphenyl carbazide (DPC) as electron donor declined during the period of incubation. Since DPC and Mn2+ augmented 2,6-dichlorophenol indophenol photoreduction by chloroplasts from senescing leaves, damage and/or change in the conformation of a site between H2O and DCPIP in photosystem II (PS II) is suggested. Heat caused a faster decline of the Hill activity compared to cold or room temperature. However, cold treatment showed no significant effect on the photoelectron transport from H2O to DCPIP compared with room temperature.  相似文献

The contents of chlorophyll (Chl), leaf biomass, and soluble proteins were markedly decreased in phytoplasma infected apple leaves. Similar results were also observed for ribulose-1,5-bisphosphate carboxylase, 14CO2 fixation, and nitrate reductase activity. In contrast, the contents of sugars, starch, amino acids, and total saccharides were significantly increased in phytoplasma infected leaves. In isolated chloroplasts, phytoplasma infection caused marked inhibition of whole photosynthetic electron chain and photosystem 2 (PS2) activity. The artificial exogenous electron donor, diphenyl carbazide, significantly restored the loss of PS2 activity in infected leaves. Similar results were obtained when Fv/Fm was evaluated by in vivo Chl a fluorescence kinetic measurements.  相似文献

Cupric ion (Cu++) inhibits the rate of photosystem II electron transport and the intensity of the variable part of chl a fluorescence in isolated chloroplast thylakoids. The inhibition is markedly dependent on the nature of the buffer used in the assay medium. In MES and HEPES buffers, complete inhibition of photosystem II occurs at 30 M of Cu++, while in Tricine no inhibition occurred even at 200 M Cu++. In other buffers used (TES, Phosphate, Tris), the efficacy of Cu++ inhibition is intermediate. The calculated binding constants are found to correspond to the observed I50 values for the six buffers used. It is concluded that the previous reports on copper inhibition, where buffers have been used indiscriminately should be reconsidered. A fifteen minute incubation of spinach chloroplasts with the divalent Ca2+ chelator, EGTA, in concentrations 50–250 μM, inhibits electron transport through both photosystems. All photosystem II partial reactions, including indophenol, ferricyanide and the DCMU-insensitive silicomolybdate reduction are inhibited from 70–100%. The photosystem II donor reaction, diphenyl carbazide → indophenol, is also inhibited, indicating that the inhibition site comes after the Mn2+ site, and that the first Ca2+ effect noted (site II) is not on the water oxidation enzyme, as is commonly assumed, but between the Mn2+ site and plastoquinone A pool. The other photosystem II effect of EGTA (Ca2+ site I), occurs in the region between plastoquinone A and P700 in the electron transport chain of chloroplasts. About 50% inhibition of the reaction ascorbate + TMPD → methyl viologen is given by incubation with 200 μM EGTA for 15 min. Ca2+ site II activity can be restored with 20 mM CaCl2. Ca2+ site I responds to Ca2+ and plastocyanin added jointly. More than 90% activity in the ascorbate + TMPD → methylviologen reaction can be restored. Various ways in which Ca2+ ions could affect chloroplast structure and function are discussed. Since EGTA is more likely to penetrate chloroplast membranes than EDTA, which is known to remove CF1, the coupling factor, from chloroplast membranes, and since Mg2+ ions are ineffective in restoring activity, it is concluded that Ca2+ may function in the electron transport chain of chloroplasts in a hitherto unsuspected manner.  相似文献

Photoinhibition was studied in osmotically broken chloroplasts isolated from spinach leaves (Spinacia oleracea L.). Both whole chain electron transport (measured as ferricyanide-dependent O2 evolution in the presence of NH4Cl) and photosystem II activity (measured as O2 evolution in the presence of either silicomolybdate plus 3-(3,4-diphenyl)-1,1 dimethylurea or parabenzoquinone) showed similar decreases in activity in response to a photoinhibitory treatment (8 minutes of high light given in the absence of an electron acceptor other than O2). Photosystem I activity was less affected. Photoinhibition of silicomolybdate reduction was largely reversible by an 8 minute dark incubation following the light treatment. Decreasing the O2 concentration during photoinhibition below 2% increased photoinhibition of whole chain electron transport. Addition of superoxide dismutase to the reaction medium did not affect photoinhibition. Photoinhibition of both photosystem I and photosystem II activity increased as the rate of electron transfer during the treatment increased, and was largely prevented when 3-(3,4-diphenyl)-1,1-dimethylurea was present during the photoinhibition period. Noncyclic photophosphorylation was decreased as a consequence of whole chain electron transfer photoinhibition. Since diphenyl carbazide added after light treatment did not relieve photoinhibition of dichlorophenol indophenol reduction, we conclude that the site of inhibition is located within or near the photosystem II reaction center.  相似文献

Direct evidence for the possible loss of photosystem II (PS II) activity in chloroplasts of Vigna sinensis L. cv. Walp after ultraviolet-B (UV-B, 280–320 nm) radiation treatment was provided by polyacrylamide gel electrophoretic analysis of PS II polypeptides. A 30 min UV-B treatment of chloroplasts caused a 50% loss of PS II activity. The artificial electron donor. Mn2+ failed to restore UV-B radiation induced loss of PS II activity, while diphenyl carbazide (DPC) and NH2OH only partially restored activity. Such a loss in PS II activity was found to be primarily due to a loss of 23 and 33 kDa extrinsic polypeptides. UV-B treatment induced the synthesis of a few polypeptides and a 29 kDa light-harvesting chlorophyll protein.  相似文献

When spinach chloroplast membranes were exposed to osmotic stress in vitro, by incubation in 1.0 M sorbitol + 10 mM MgCl2 their oxygen evolving system was suppressed. The possible reasons for such inactivation of PS II mediated oxygen evolution were examined. There were conformational changes in the chloroplast membranes, as indicated by their absorption spectra. The pattern of sensitivity to DCMU was not altered. The sensitivity of PS II to water stress remained, even after a pre-wash treatment with NaCI (which removed 18 and 24 kD proteins) but not when the thylakoids were pretreated with NH20H or CaCl2 (removed manganese and 33 kD). The manganese content of thylakoid membranes was markedly reduced under osmotic stress in presence of magnesium. We suggest that exposure of chloroplasts to 1.0 M sorbitol in presence of Mg2+ released manganese from thylakoid membranes, thereby leading to a suppression in oxygen evolution.  相似文献

The photosynthetic capacity of detached leaves of a non-yellowing mutant of Festuca pratensis Huds. declined during senescence at a similar rate to that in a normal cultivar. Respiratory oxygen uptake in the dark continued at similar rates in both genotypes during several days of senescence. In chloroplasts isolated from leaves at intervals after excision, the rate of photosystem I (PS I)-mediated methyl viologen reduction using reduced N,N,N,N-tetramethyl-p-phenylene diamine as electron donor also declined in both genotypes, possibly due to loss of integrity of the photosynthetic apparatus in the cytochrome f-plastocyanin region. There was a similar fall in PS II electron transport using water as electron donor and measured at the rate of reduction of 2,6-dichlorophenolindophenol. Partial restoration of this activity by the addition of diphenyl carbazide was evidence for lability of the oxygen-evolving complex during senescence. An accentuated difference between mutant and normal material in this case indicated that the mutant retains a greater number of functional PS II centres. The cucumber (Cucumis sativus L.) plants were sprayed with 20 mM 5-aminolevulinic acid or distilled water (control) and incubated in dark for 14 hr. The thylakoid membranes prepared from the intact chloroplasts, isolated from the above plants in dark, were illuminated with low light intensity (100 W/m2) for 30 min. Due 10 photodynamic reactions, the photochemical function of photosystem II was damaged by 50% in treated thylakoids whereas it was only slightly (8%) affected in control thylakoids. The photosystem I was, however, not affected. The exogenous electron donors, MnCl2, diphenyl carbazide and NH2OH failed to restore the photosystem II activity suggesting that the photodynamic damage had taken place very close to photosystem II reaction center. Singlet oxygen scavenger, histidine, could protect the photosystem II activity while superoxide radical scavengers, superoxide dismutase and 1, 2-dihydroxybenzene-3, 5-disulphonic acid disodium salt, and hydroxyl radical scavenger, formate, failed to protect the same.  相似文献

The photosynthetic performance of the desiccation‐tolerant, intertidal macro‐algae Ulva prolifera was significantly affected by sorbitol‐induced osmotic stress. Our results showed that photosynthetic activity decreased significantly with increases in sorbitol concentration. Although the partial activity of both photosystem I (PS I) and photosystem II (PS II) was able to recover after 30 min of rehydration, the activity of PS II decreased more rapidly than PS I. At 4 M sorbitol concentration, the activity of PS II was almost 0 while that of PS I was still at about one third of normal levels. Following prolonged treatment with 1 and 2 M sorbitol, the activity of PS I and PS II decreased slowly, suggesting that the effects of moderate concentrations of sorbitol on PS I and PS II were gradual. Interestingly, an increase in non‐photochemical quenching occurred under these conditions in response to moderate osmotic stress, whereas it declined significantly under severe osmotic stress. These results suggest that photoprotection in U. prolifera could also be induced by moderate osmotic stress. In addition, the oxidation of PS I was significantly affected by osmotic stress. P700+ in the thalli treated with high concentrations of sorbitol could still be reduced, as PS II was inhibited by 3‐(3,4‐dichlorophenyl)‐1,1‐dimethylurea (DCMU), but it could not be fully oxidized. This observation may be caused by the higher quantum yield of non‐photochemical energy dissipation in PS I due to acceptor‐side limitation (Y(NA)) during rehydration in seawater containing DCMU.  相似文献

The extent of mercury (Hg) toxicity in the heterocystous cyanobacterium Nostoc muscorum grown for 72 h in three different light intensities was tested for various physiological parameters viz. growth, pigment contents, photosynthesis, respiration, reactive oxygen species (ROS), malondialdehyde formation and antioxidants. A general reduction in growth and pigments, whole cell O2-evolution, photosynthetic electron transport activities and 14CO2-fixation was observed in a metal concentration–dependent manner, and this effect was more pronounced in high light (130 μmol photon m−2 s−1)–exposed cells as compared to low (10 μmol photon m−2 s−1) and normal (70 μmol photon m−2 s−1) light intensity–exposed cells; however, carotenoids and respiration showed reverse trend. Among photosynthetic electron transport activities, whole chain activity was found to be most sensitive in comparison with photosystem II (PS II) and photosystem I (PS I). Comparing the different photosynthetic processes, 14CO2-fixation was most affected in cyanobacterial cells when exposed to Hg and different light intensities. After application of various exogenous electron donors, diphenyl carbazide was found to be more effective to restore PS II activity, suggesting that site of damage lies in between oxygen evolving complex and PS II. Level of oxidative stress (superoxide radical and lipid peroxidation) was maximum at 3.0 μM of Hg when coupled with high light intensity (except hydrogen peroxide). A dose-dependent increase in enzymatic antioxidants such as superoxide dismutase, peroxidase and catalase as well as non-enzymatic antioxidants such as proline, ascorbate, cysteine (except under high light intensity) and non-protein thiols [NP-SH] was observed, which further increased with the increase in light intensity. It was noticed that Hg intoxicates N. muscorum through ROS production, which is aggravated along with the increase in light intensity. Overall results suggest that the severity of the metal stress does increase with Hg concentrations but when coupled with light, it was the light intensity that determines the extent of Hg toxicity.  相似文献

Bicarbonate ion, not dissolved CO2 gas, is shown to increase 4- to 5-fold the rate of dichlorophenol indophenol reduction by isolated maize (Zea mays) chloroplasts. Glutaraldehyde fixed chloroplasts continue to exhibit bicarbonate-dependent 2,6-dichlorophenol indophenol reduction. Bicarbonate is shown to act close to the oxygen-evolving site, i.e. prior to the electron donation site of diphenyl carbazide to photosystem II. Dark incubation and light pretreatment of chloroplasts in various concentrations of bicarbonate, just prior to assay, indicate that bicarbonate binds to chloroplasts in the dark and is released again as the Hill reaction proceeds in the light. It is suggested that bicarbonate ions may play a critical role in the oxygen-evolving process in photosynthesis.  相似文献

Changes in contents of pigments, chlorophyll-protein complex, and photosynthetic activities were investigated in field grown apple (Malus pumila Mill.) leaves infected by Apple Proliferation phytoplasma. The contents of chlorophyll a+b (Chl) and carotenoids (Car) markedly decreased in infected leaves. Similar results were also observed for content of total soluble proteins and ribulose-1,5-bisphosphate carboxylase activity. When various photosynthetic activities were followed in isolated thylakoids, phytoplasma infection caused a marked inhibition of whole chain and photosystem 2 (PS2) activity. Smaller inhibition of photosystem 1 (PS1) activity was observed even in severely infected leaves. The artificial exogenous electron donors, MnCl2 diphenyl carbazide, and NH2OH, did not restore the loss of PS2 activity in both mildly and severely infected leaves. Similar results were obtained by Chl fluorescence measurements. The marked loss of PS2 activity in infected leaves was due to the reduction of contents of chlorophyll and light-harvesting chlorophyll-protein 2 complexes. A cDNA clone for the extrinsic 30 kDa protein (OEC30) of photosystem II in Euglena gracilis Z was isolated and characterized. The open reading frame of the cDNA encoded a polypeptide of 338 amino acids, which consisted of a long presequence of 93 amino acids and a mature polypeptide of 245 amino acids. Two hydrophobic domains were identified in the presequence, in contrast to the presence of a single hydrophobic domain in the presequence of the corresponding proteins from higher plants. At the N- and C-terminal regions, respectively, of the presequence, a signal-peptide-like sequence and a thylakoid-transfer domain were identified. After preheating of Amaranthus chloroplasts at elevated temperatures (up to 45°C), the chlorophyll a fluorescence level under low excitation light rises as compared to control (unheated) as observed earlier in other chloroplasts (Schreiber U and Armond PA (1978) Biochim Biophys Acta 502: 138–151). This elevation of heat induced fluorescence yield is quenched by addition of 0.1 mM potassium ferricyanide, suggesting that with mild heat stress the primary electron acceptor of photosystem II is more easily reduced than the unheated samples. Furthermore, the level of fluorescence attained after illumination of dithionite-treated samples is independent of preheating (up to 45°C). Thus, these experiments indicate that the heat induced rise of fluorescence level at low light can not be due to changes in the elevation in the true constant F0 level, that must by definition, be independent of the concentration of QA. It is supposed that the increase in the fluorescence level by weak modulated light is either partly associated with dark reduction of QA due to exposure of chloroplasts to elevated temperature or due to temperature induced fluorescence rise in the so called inactive photosystem II centre where QA are not connected to plastoquinone pool. In the presence of dichlorophenyldimethylurea the fluorescence level triggered by weak modulated light increases at alkaline pH, both in control and heat stressed chloroplasts. This result suggests that the alkaline pH accelerates electron donation from secondary electron donor of photosystem II to QA both in control and heat stressed samples. The effect of iron deficiency on photosynthetic pigments, ribulose-1,5-bisphosphate carboxylase (RuBPC), and photosystem activities were investigated in field grown grapevine (Vitis vinifera L. cv. Pinot noir) leaves. The contents of chlorophyll (Chl) (a+b) and carotenoids per unit fresh mass showed a progressive decrease upon increase in iron deficiency. Similar results were also observed in content of total soluble proteins and RuBPC activity. The marked loss of large (55 kDa) and small (15 kDa) subunits of RuBPC was also observed in severely chlorotic leaves. However, when various photosynthetic electron transport activities were analysed in isolated thylakoids, a major decrease in the rate of whole chain (H2O methyl viologen) electron transport was observed in iron deficient leaves. Such reduction was mainly due to the loss of photosystem 2 (PS2) activity. The same results were obtained when Fv/Fm was evaluated by Chl fluorescence measurements in leaves. Smaller inhibition of photosystem 1 (PS1) activity was also observed in both mild and severely chlorotic leaves. The artificial electron donors, diphenyl carbazide and NH2OH, markedly restored the loss of PS2 activity in severely chlorotic leaves. The marked loss of PS2 activity was evidently due to the loss of 33, 23, 28-25, and 17 kDa polypeptides in iron deficient leaves.  相似文献

Photoinhibition under irradiance of 2 000 μmol m−2 s−1 (HI) was studied in detached control (C) and water deficit (WD) leaves of grapevine (Vitis vinifera L.) plants. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. The potential efficiency of photosystem (PS) 2, Fv/Fm, marginally declined under HI in WD-leaves without significant increase of F0. In contrast, Fv/Fm ratio declined markedly with significant increase of F0 in C-leaves. In isolated thylakoids, the rate of whole chain and PS2 activity under HI were more decreased in C-than WD-leaves. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in both C-and WD-leaves. Thus HI operates at the acceptor side of PS2 in both leaf types. Quantification of the PS2 reaction centre protein D1 following HI exposure of leaves showed pronounced differences between C-and WD-leaves. The marked loss of PS2 activity under HI of C-leaves was due to the marked loss of D1 protein of the PS2 reaction centre.  相似文献

The influence of vanadium compounds (vanadate, vanadyl citrate) on photosynthesis in Chlorella fusca and in algal and spinach chloroplasts has been investigated. It was found that: 1. At moderately high concentrations (at least 0.1 mM) both vanadate and vanadyl citrate enhance photosynthetic O2 production in intact C. fusca cells. At lower V concentration (about 2 μM) only vanadate stimulates photosynthesis. The increase is dependent on culture conditions and on light intensity. 2. Up to 1 mM V, neither vanadium compound influences PS II activity, either in intact cells or in algal or spinach chloroplasts. 3. The PS I reaction in algal and spinach chloroplasts is maximally enhanced (3-fold) in presence of vanadium (20 μM). The increase is independent of light intensity. 4. Cr(VI), Mo(VI), and W(VI) (1 mM) stimulate photosynthesis in intact C. fusca cells, but do not influence the photosystems of isolated chloroplasts. Vanadium is suggested to act as a redox catalyst in the electron transport from PS II to PS I.  相似文献

Chlorpromazine, phenothiazine and trifluoperazine, known as calmodulin antagonists, inhibit electron transport in Photosystem II of spinach chloroplasts in concentrations from 20–500 μM. The inhibition site is located on the diphenyl carbazide to indophenol pathway in Tris-treated chloroplasts, indicating that water oxidation is not affected by these drugs. Ca2+ ions, bound to chloroplast membranes before the addition of calmodulin antagonists, can protect against inhibition up to 25% of the electron transport rate. In presence of A23187, the Ca2+-specific ionophore, Ca2+ ions provide less protection against inhibition by the 3 calmodulin antagonists used. A possible role of a calmodulin-like protein in spinach chloroplasts is postulated.  相似文献

In grapevine (Vitis vinifera L.) leaf chlorophyll (Chl) a and Chl b and carotenoid contents were higher in plants grown at low photon flux densities (PFD) than in those grown at medium and high PFD. The highest Chl a variable to maximum fluorescence ratio Fv/Fm was observed in plants grown at medium PFD while the minimum fluorescence F0 was highest in those at high PFD. In isolated thylakoids, both high and low PFD caused marked inhibition of whole chain and photosystem 2 (PS2) activities. The artificial exogenous electron donor diphenyl carbazide significantly restored the loss of PS2 activity in low PFD leaves.  相似文献
