Evaluation of genetic variability in choosen apple (<Emphasis Type="Italic">Malus</Emphasis> × <Emphasis Type="Italic">domestica</Emphasis> Borkh.) cultivars by ISSR-PCR analysis

The aim of the study was to determine the genetic variability in eight apple cultivars: Delikates, Cortland, James Grieve, Lired, Jonathan, Golden Delicious, Jonagold, and Idared from the collection of Fruit Growing Research Station in Rajkowo of the West Pomeranian University of Technology, Szczecin. The cultivar Delikates was obtained from the crossing of two cultivars: Cortland, and James Grieve, whereas cultivar Lired is a James Grieve’s sport. The second one cultivar—Jonagold was obtained from the crossing of Jonathan and Golden Delicious. The cultivar Idared is a hybrid obtained from the crossing of Jonathan and Wagener. Out of 40 primers, 17 were chosen for the final study. Those amplified a total of 183 loci (872 amplicons) out of which 34 (18.5%) were monomorphic, 128 (69.5%) were polymorphic and 22 (12%) cultivar-specific. Specific ISSR products were detected for each apple cultivar. A dendrogram was constructed using the UPGMA method which revealed two distinct clusters: I—Delikates, Cortland, James Grieve and Lired, II—Jonathan, Golden Delicious, Jonagold and Idared. Genetic similarity between Delikates, Cortland and James Grieve was 68.6, 70.8%, respectively and between cultivar Jonagold, Jonathan and Golden Delicous was 79.8, 85.2%, respectively.     

Creating a community resource for protein science

Helen Berman is the recipient of the Protein Society 2012 Carl Branden Award In addition to being one of the early pioneers in protein crystallography, Carl Brändén made significant contributions to science education with his elegant and beautifully illustrated book Introduction to Protein Structure (Brändén and Tooze, New York: Garland, 1991). It is truly an honor to receive this award in their names. This award and the 40th anniversary of the Protein Data Bank (PDB; Berman et al., Structure 2012;20:391–396) have given me an opportunity to reflect on the various components that have contributed to building a resource for protein science and to try to quantify the impact of having PDB data openly available.

Compartmental models for human iodine metabolism

Compartmental models for the various aspects of human iodine metabolism are reviewed, emphasizing the role of Mones Berman in the development of this field. The review first presents published submodels for the peripheral distribution of inorganic iodine, for the thyroidal iodide trapping function, and for the peripheral distribution and metabolism of the thyroid hormones. Approaches to improving understanding of the physiology of the thyroid gland itself through compartmental modeling techniques are then discussed in more detail. The three submodels described above are incorporated into overall models of thyroid iodine metabolism after being simplified to various degrees. Previously published models for thyroid-gland radioiodine metabolism, as well as current work in progress, are illustrated by attempting to fit the models to data from a single (previously unpublished) detailed prolonged ¹²⁵I feeding experiment in a normal human subject. Published thyroid gland models reviewed include: (1) the usual presentation, where the thyroid is a single homogeneous iodine compartment; (2) the model of DeGroot and colleagues, where thyroidal iodine is presented as MIT, DIT, T3, and T4, each with an active and linked storage compartment; (3) the thyroid model developed by Berman and colleagues, with less chemical subcategorization but incorporating a delay compartment, in which a fraction of the iodinated material in the thyroid is partially or completely inaccessible to secretion during the delay; and the later updating of Berman's model to include a thyroidal iodide recirculation pool. The experimental data presented fits most of these models for the first 1–2 weeks, but the fit could not be extended to longer data collection times. To overcome this shortcoming, a new thyroid gland model is introduced. It is based on the latest Berman model but describes thyroglobulin metabolism as incorporating multiple delay compartments of various time periods. The overall fit of the long term data is better with this model construct than with any of the published models. It appears that a complex thyroidal substructure, such as that of the multidelay model under development, will be required to account for overall thyroid iodine metabolism as isotopic equilibrium in man is approached.

The effect of (2-chloroethyl) trimethylammonium chloride on ethylene evolution and respiration rate of an early cultivar of apples

Ethylene production and respiration rate of an early cultivar of apples, Letní pr?svitné ?luté, is significantly increased after the application of the lowest tested concentration of CCC (0.1% in lanolin). These effects, calculated from the amount of released ethylene and carbon dioxide, were observed during the whole period of ripening. The production of ethylene is slightly inhibited by the higher concentrations of CCC while the respiration rate is not significantly affected. The rate of the ethylene production of the early cultivar of apples is decreased 4.4 to 9.7 fold by 0.1% CCC as compared with the winter cultivar Jonathan. Although these ratios rise gradually during the ripening of fruits treated with other higher concentrations of CCC, the total amount of ethylene produced by the winter cultivar Jonathan is 1.5 fold higher than that of the early one.
