Mitochondrial DNA (mtDNA) polymorphism in the essentially non-anadromous River Usk brown trout Salmo trutta population was investigated by restriction analysis. Following mtDNA extraction and purification on caesium chloride density gradients, monomorphic restriction profiles were obtained with Hae III , Hind III , Sau 3AI and Xbal . However, the restriction endonucleases Ava II and HinfIl proved informative. The distribution of four composite genotypes found within the Usk system was heterogeneous, and a fifth genotype appeared exclusively in an outgroup sample from the adjacent River Wye drainage. The source of the observed genetic variation is discussed in relation to estimated divergence times for Usk mtDNA genotypes and the stocking history of the catchment.  相似文献

SUMMARY 1. The large microgeographical differentiation revealed by allozyme studies in brown trout ( Salmo trutta) populations is one of the most striking features of this species. Additionally, allozymes showed great genetic differences between Atlantic and Mediterranean populations on a macrogeographical scale.
2. This study was carried out in order to assess whether the great differences observed between Atlantic and Mediterranean populations persisted where the two are geographically close (the 'microgeographical scale'). Sixteen populations of brown trout, S. trutta , were screened for genetic variation at 25 allozyme loci. The sampling sites, which occupied a relatively small geographical area, were distributed across Cantabrian (Atlantic) and Mediterranean drainages in Northern Spain.
3. The neighbour-joining tree, inferred from Nei's genetic distance, showed that brown trout populations clustered into two different groups. These groups corresponded to the Cantabrian and the Mediterranean groups of populations, although no clear geographical pattern emerged within each of the groups. This geographical pattern is basically caused by significant differences in the frequency distribution of the CK-A1 * locus, with a higher frequency of * 115 in Cantabrian samples (0.586 ± 0.091) while allele * 100 was more frequent in Mediterranean samples (0.931 ± 0.038). In addition, this study revealed alleles exclusive to the Mediterranean and Cantabrian populations, agreeing with previous findings.
4. Genetic differentiation between Cantabrian and Mediterranean regions (14.19%) was similar to that estimated in Spain at a larger scale (13%), showing that most of the differences between the regions can be observed even in a small geographical area.  相似文献

The effect of the introduction of fry of anadromous sea trout, Salmo trutta L., on the genetic integrity of landlocked brown trout populations was evaluated. Samples were taken from six brown trout populations from streams above impassable waterfalls in the Conwy river system (North Wales, U.K.) in 1989 and 1990. Three of these streams had no known stocking history and three had been stocked with sea trout fry from the lower Conwy system over the last few years. Representatives of these sea trout were collected from two streams in the lower Conwy system and from a hatchery. Allele frequencies at 13 loci, six of which were polymorphic, were determined by starch gel electrophoresis.
The stocked populations were intermediate in their allele frequencies between unstocked brown trout and sea trout samples. A principal component analysis suggested significant numbers of hybrids in all of the stocked streams. This shows that some of the introduced sea trout did not migrate down the falls to the sea, but stayed in fresh water and hybridized with the local population. The significance of this finding for the conservation of the genetic resource of brown trout stocks is discussed.  相似文献

This study addressed two hypotheses: (1) that individual recognitionis used to reduce the cost of contesting resources in sea troutand (2) that the hatchery environment selects against individualrecognition. Predictions from these hypotheses were tested inlaboratory experiments where behavior and growth were studiedin juvenile sea trout. In tact groups of familiar fish werecompared with groups that contained both familiar and unfamiliarindividuals. In general, the results were in agreement withthe first hypothesis. Familiar fish in intact groups had morestable dominance ranks, higher food intake, and better utilizationof food. Familiarity also reduced the distance to the nearestneighbor. Furthermore, initiators of conflicts were more likelyto win against familiar fish than against strangers. These resultssupport game-theory-based hypotheses explaining the dear-enemyphenomenon as an effect of familiarity A picture emerges inwhich familiarity stabilizes the hierarchical structure of agroup and governs behavioral modifications that will promotefeeding and growth, in turn leading to higher fitness. The secondhypothesis, which predicted a reduced effect of individual recognitionin sea-ranched trout, was not strongly supported because familiarityaffected sea-ranched and wild trout similarly in most respects.However, familiarity was not beneficial for growth in sea-ranchedtrout, whereas it increased growth rate in wild fish. In addition,sea-ranched trout tended to maintain larger distances to theirnearest neighbors than did wild trout.  相似文献

Brown trout, Salmo trutta, were allowed to thermoregulate individually in an electronic shuttlebox. Pooled data for 6 fish showed a diel pattern of preferred temperature, with a diurnal minimum of 10.3°C, an early nocturnal maximum of 13.7°C, a less pronounced mid-scotophase minimum of 11.7°C, and a secondary dawn maximum of 12.8°C, in a somewhat crepuscular pattern. The Iberian Peninsula contains diverse populations of freshwater fish, with major river basins comprising differentiated biogeographic units. The Duero River flows through the North‐Western Iberian Peninsula and is one of the most important rivers within the Iberian glacial refuge. Brown trout (Salmo trutta) populate this whole basin, and studies using both allozyme and microsatellite loci have detected a geographically sorted distribution of genetic variation in this species. In this work, sequences of the mitochondrial control region obtained from 299 brown trout from the Duero River were compared with other Iberian and European datasets. Two differentiated haplotype groups were detected inside the Duero River basin. One of them was related to the Atlantic (AT) lineage that is present in Northern European populations, whereas the other comprised an unique group that was restricted to the inner region of the basin. The amount of divergence of this Duero group from the other brown trout populations studied is consistent with a new trout lineage (Duero, DU) that is endemic to this river basin and that diverged from other Atlantic populations during the Pleistocene. The distribution of the DU and AT quaternary lineages in the Duero River was consistent with the ichthyological pattern described in the basin that originated during the Miocene–Pliocene. Evidence of selective processes that favour the haplotypes of the DU lineage may explain this discrepancy.  相似文献

Major histocompatibility complex (MHC) class I-linked microsatellite data and parental assignment data for a group of wild brown trout (Salmo trutta L.) provide evidence of closer spatial aggregation among fry sharing greater numbers of MHC class I alleles under natural conditions. This result confirms predictions from laboratory experiments demonstrating a hierarchical preference for association of fry sharing MHC alleles. Full-siblings emerge from the same nest (redd), and a passive kin association pattern arising from limited dispersal from the nest (redd effect) would predict that all such pairs would have a similar distribution. However, this study demonstrates a strong, significant trend for reduced distance between pairs of full-sibling fry sharing more MHC class I alleles reflecting their closer aggregation (no alleles shared, 311.5 ± (s.e.)21.03 m; one allele shared, 222.2 ± 14.49 m; two alleles shared, 124.9 ± 23.88 m; P<0.0001). A significant trend for closer aggregation among fry sharing more MHC class I alleles was also observed in fry pairs, which were known to have different mothers and were otherwise unrelated (ML-r = 0) (no alleles: 457.6 ± 3.58 m; one allele (422.4 ± 3.86 m); two alleles (381.7 ± 10.72 m); P<0.0001). These pairs are expected to have emerged from different redds and a passive association would then be unlikely. These data suggest that sharing MHC class I alleles has a role in maintaining kin association among full-siblings after emergence. This study demonstrates a pattern consistent with MHC-mediated kin association in the wild for the first time.  相似文献

Initial feeding of brown trout was investigated under laboratory conditions. Fifty per cent feeding occurred when yolk constituted approximately 31% of total alevin dry weight, and feeding rate was positively correlated to developmental stage. Conservation of species should be based on knowledge of effective population sizes and understanding of how breeding tactics and selection of recruitment habitats lead to genetic structuring. In the stream‐spawning and genetically diverse brown trout, spawning and rearing areas may be restricted source habitats. Spatio–temporal genetic variability patterns were studied in brown trout occupying three lakes characterized by restricted stream habitat but high recruitment levels. This suggested non‐typical lake‐spawning, potentially representing additional spatio–temporal genetic variation in continuous habitats. Three years of sampling documented presence of young‐of‐the‐year cohorts in littoral lake areas with groundwater inflow, confirming lake‐spawning trout in all three lakes. Nine microsatellite markers assayed across 901 young‐of‐the‐year individuals indicated overall substantial genetic differentiation in space and time. Nested gene diversity analyses revealed highly significant (≤P = 0.002) differentiation on all hierarchical levels, represented by regional lakes (FLT = 0.281), stream vs. lake habitat within regional lakes (FHL = 0.045), sample site within habitats (FSH = 0.010), and cohorts within sample sites (FCS = 0.016). Genetic structuring was, however, different among lakes. It was more pronounced in a natural lake, which exhibited temporally stable structuring both between two lake‐spawning populations and between lake‐ and stream spawners. Hence, it is demonstrated that lake‐spawning brown trout form genetically distinct populations and may significantly contribute to genetic diversity. In another lake, differentiation was substantial between stream‐ and lake‐spawning populations but not within habitat. In the third lake, there was less apparent spatial or temporal genetic structuring. Calculation of effective population sizes suggested small spawning populations in general, both within streams and lakes, and indicates that the presence of lake‐spawning populations tended to reduce genetic drift in the total (meta‐) population of the lake.  相似文献

In a series of experiments, kin-biased behavior of young browntrout (Salmo trutta) was observed. The aggressiveness shownby groups of familiar siblings (siblings reared together sincefertilization) and groups of unfamiliar siblings (siblings rearedapart since fertilization) was significantly lower comparedto that of mixed groups of two unrelated sibling groups (offspringof two different pairs of parents). The rivers of the island of Corsica, whose catchment areas are on crystalline rock, have low salt contents and their invertebrate fauna is qualitatively and quantitatively poorer than on the European mainland. The growth rate of trout in Corsica was analysed on samples from of six coastal rivers: the Tavignano, the Fium Orbo and the Golo on the west coast, the Prunelli, the Taravo and the Rizzanese on the east coast. Mesological data — conductivity, temperature, calcium content and altitude and biological data — biomass and linear growth rate of the trout, and density of benthic invertebrates — were collected at each of the sampling station.Analysis of variance of the size of three year old trout revealed three groups of rivers. The first includes the Tavignano, the Rizzanese and the Taravo, where the highest linear growth rates were recorded; the second consists of the Golo and the Prunelli, and the third, of the Fium Orbo. Principal component analysis gave two main axes on the basis of temperature and benthic invertebrate density for the first, and trout biomass for the second. Linear regression showed that benthic invertebrate density accounted for 75% of size variance of three year old trout. Evidence of the limiting role of the trophic factor is provided.  相似文献

A useful genetic marker exists through the apparent fixation of the LDH-5 * 100 allele in wild populations of brown trout in rivers from Asturias, Spain, contrasted with the near fixation of the LDH-5 * 90 allele in hatchery populations used to stock these rivers. In sampling locations where natural reproduction occurred, the * 100 allele was found exclusively in all areas having no record of hatchery stocking. The * 100 allele also predominated in three stocked areas having natural reproduction; in two of these areas a few individuals of the 0 + age class were homozygous for the * 90 allele. These data indicated that all catchable and reproductive fish originated from indigenous populations and thus the policy of hatchery supplementation was a failure in these areas.  相似文献

Ten polymorphic enzyme coding loci were assayed using starch gel electrophoresis in anadromous (sea trout) and freshwater-resident trout, Salmo trutta, from two neighbouring river systems draining into Clew Bay in western Ireland. Samples were collected from the Burrishoole and from the neighbouring Newport river system. In the Burrishoole system, two samples were from above waterfalls impassable to migratory trout, two consisted of anadromous smolts or post-smolts and the remainder were freshwater-resident trout from freely accessible areas. There were large differences in gene frequency between trout from above impassable barriers and all others sampled, and these two groups also differed substantially from each other. More minor differences were evident between some other samples, particularly between trout from the upper lake and streams entering it and a stream associated with the lower lake. Two anadromous populations therefore occur in the Burrishoole system. In contrast, no differences were evident in two sampling years between anadromous trout and cohabiting freshwater-resident trout from the upper part of the Burrishoole system. Thus, in the upper part of this system, anadromous trout do not appear to comprise a separate population from non-migratory trout and interbreeding may occur.  相似文献

To investigate the phylogenetic relationships and geographical structure among brown trout S. trutta L. populations from the South Adriatic-Ionian and Aegean sea basins, mitochondrial DNA nucleotide sequence comparisons were used. A 310-base-pair (bp) segment of the control region (D-loop), and an additional 280-bp segment of the cytochrome b gene were sequenced from representatives of 13 brown trout populations. Phylogenetic analyses, conducted after combining the data presented with published data from other Eurasian brown trout, revealed four major phylogenetic groups, three of which were found widely distributed within the southern Balkan region. The phylogeographical patterns revealed by mtDNA represent one of the few cases where phylogenetic discontinuity in a gene tree exists without obvious geographical localization within a species' range and has most likely resulted from the differentiation of the major mtDNA clades during Messinian or early Pleistocene times. Finally, the genetic relationships among the populations suggested by mtDNA were generally not in accordance with either allozyme or morphological data.  相似文献

1. The present study was designed to characterize the genetic structure of brown trout ( Salmo trutta ) populations from the southern Balkans and to assess the spread of non-native strains and their introgression into native trout gene pools. We analysed polymorphism at nine microsatellite loci in seven supposedly non-admixed and three stocked brown trout populations.
2. The analyses confirmed the absence of immigration and extraordinarily strong genetic differentiation among the seven non-introgressed populations in parallel with low levels of intrapopulation genetic variability. In contrast, analyses of the stocked populations revealed that the genetic integrity of the local populations had been substantially changed, and the populations must be characterized as hybrid swarms. The pattern of population differentiation observed at microsatellites contrasted to that depicted previously by mtDNA variation. However, the close relationships between populations from the Danube and Axios river systems proposed solely by microsatellites could be explained by palaeogeographic events.
3. Our research showed that most of the populations examined represent unique gene pools, whose existence is critically compromised. Therefore, appropriate management and conservation strategies should be developed urgently in order to protect the subspecific biodiversity and to reverse currently negative trends.  相似文献

The diel timing of displacement of emerging brown trout ( Salmo trutta ) was studied by trapping the alevins at the downstream end of three experimental channels. The presence of phosphorescent landmarks on the substrate delayed the catches during the first hours of complete darkness, but increased them during twilight. During daylight, the presence of the landmarks was associated with greatly increased numbers of free-swimming fishes.  相似文献

Morphological studies on the effect of low pH on egg shells (chorion) of rainbow trout, Oncorhynrhus mykiss Walbaum (formerly Salmo gairdneri ), and brown trout, Salmo trutta f. fario L., showed that the plugs of the outer layer of the chorion, which block the channels of the underlying layer, developed distinctive vacuolization at low pH exposure. This may lead to loss of chorional semi-permeability. Chorions were incompletely degraded at low pH inducing hatching problems. Partially hatched alevins showed only a small local zone of dissolution of the chorion around the head of the embryo preventing the larvae from being freed.  相似文献

Cannulated brown trout, Salmo trutta , were exposed for 36 h to synthetic water with a low calcium content of pH 5 and similar synthetic water dosed with aluminium to raise the filterable A1 from 5 to 290 μg 1−1 over the 36-h period. There were no significant disturbances of plasma concentrations of glucose, cortisol or catecholamine (adrenaline and noradrenaline) in fish held in water of pH 5. The addition of aluminium to this acidic synthetic water resulted in a generalized endocrine stress response with a four-fold increase in plasma glucose concentration after 18 h and a significant increase in plasma cortisol concentration from 24 h onwards when filterable A1 exceeded 200 μg 1−1. Plasma catecholamine concentration indicated an adrenergic stress response in aluminium-exposed brown trout. A transient doubling in noradrenaline after 6 h in A1 was followed by a larger increase in both plasma adrenaline and noradrenaline concentrations in fish surviving the 36-h exposure to A1.  相似文献
