A solution of the direct problem of electroencephalography was carried out within the framework of multilayer sphere model consisting of homogenous omic spherical kernel covered with five omic layers modelling epicortical liquor, three-layer bone of the skull, scalp. Solutions of more simple models cited in literature are particular variants of this general one. By means of numerical simulation solutions of multilayer spheres of different complexity are compared and a notion of the simplest adequate model is worked out.  相似文献

The biochemistry of a system made up of three kinds of cell is virtually impossible to work out without the use of in silico models. Here, we deal with homeostatic balance phenomena from a metabolic point of view and we present a new computational model merging three single-cell models, already available from our research group: the first model reproduced the metabolic behaviour of a hepatocyte, the second one represented an endothelial cell, and the third one described an adipocyte. Multiple interconnections were created among these three models in order to mimic the main physiological interactions that are known for the examined cell phenotypes. The ultimate aim was to recreate the accomplishment of the homeostatic balance as it was observed for an in vitro connected three-culture system concerning glucose and lipid metabolism in the presence of the medium flow. The whole model was based on a modular approach and on a set of nonlinear differential equations implemented in Simulink, applying Michaelis-Menten kinetic laws and some energy balance considerations to the studied metabolic pathways. Our in silico model was then validated against experimental datasets coming from literature about the cited in vitro model. The agreement between simulated and experimental results was good and the behaviour of the connected culture system was reproduced through an adequate parameter evaluation. The developed model may help other researchers to investigate further about integrated metabolism and the regulation mechanisms underlying the physiological homeostasis.  相似文献

This paper presents the derivation and partial analysis of a general producer-consumer model. The model is stoichiometric in that it includes the growth constraints imposed by species-specific biomass carbon to nutrient ratios. The model unifies the approaches of other studies in recent years, and is calibrated from an extensive review of the algae-Daphnia literature. Numerical simulations and bifurcation analysis are used to examine the impact of energy enrichment under nutrient and stoichiometric constraints. Our results suggest that the variety of system responses previously cited for related models can be attributed to the size of the total system nutrient pool, which is here assumed fixed. New, more complicated bifurcation sequences, such as multiple homoclinic bifurcations, are demonstrated as well. The mechanistic basis of the model permits us to show the robustness of the system’s dynamics subject to alternate approaches to modeling producer and consumer biomass production.  相似文献

Faced with the recurrent evolution of resistance to pesticides and drugs, the scientific community has developed theoretical models aimed at identifying the main factors of this evolution and predicting the efficiency of resistance management strategies. The evolutionary forces considered by these models are generally similar for viruses, bacteria, fungi, plants or arthropods facing drugs or pesticides, so interaction between scientists working on different biological organisms would be expected. We tested this by analysing co-authorship and co-citation networks using a database of 187 articles published from 1977 to 2006 concerning models of resistance evolution to all major classes of pesticides and drugs. These analyses identified two main groups. One group, led by ecologists or agronomists, is interested in agricultural crop or stock pests and diseases. It mainly uses a population genetics approach to model the evolution of resistance to insecticidal proteins, insecticides, herbicides, antihelminthic drugs and miticides. By contrast, the other group, led by medical scientists, is interested in human parasites and mostly uses epidemiological models to study the evolution of resistance to antibiotic and antiviral drugs. Our analyses suggested that there is also a small scientific group focusing on resistance to antimalaria drugs, and which is only poorly connected with the two larger groups. The analysis of cited references indicates that each of the two large communities publishes its research in a different set of literature and has its own keystone references: citations with a large impact in one group are almost never cited by the other. We fear the lack of exchange between the two communities might slow progress concerning resistance evolution which is currently a major issue for society.  相似文献

A generalized autocatalytic model for chiral polymerization is investigated in detail. Apart from enantiomeric cross-inhibition, the model allows for the autogenic (non-catalytic) formation of left and right-handed monomers from a substrate with reaction rates epsilon L and epsilon R, respectively. The spatiotemporal evolution of the net chiral asymmetry is studied for models with several values of the maximum polymer length, N. For N = 2, we study the validity of the adiabatic approximation often cited in the literature. We show that the approximation obtains the correct equilibrium values of the net chirality, but fails to reproduce the short time behavior. We show also that the autogenic term in the full N = 2 model behaves as a control parameter in a chiral symmetry-breaking phase transition leading to full homochirality from racemic initial conditions. We study the dynamics of the N--> infinity model with symmetric (epsilon L = epsilon R) autogenic formation, showing that it only achieves homochirality for epsilon > epsilon c, where epsilon c is an N-dependent critical value. A generalization of the “constant overall organization” constraint of Eigen's quasispecies and hypercycle models, called herein “global population regulation”, is shown to lead to mathematically tractable spatial generalizations of these two models. The spatially uniform steady state of Eigen's quasispecies model is shown to be stable and globally attracting for all possible values of the mutation and replication rates. In contrast, the spatially and temporally uniform solutions to the hypercycle with fewer than five members, the only ones insensitive to stochastic perturbations, are shown to be unstable, and a lower bound to the spatial inhomogeneities is obtained. The prospect that the spatially localized hypercycle might be immune to various instabilities cited in the literature is then briefly considered. Although spatial localization makes possible a much richer dynamical repertoire than previously considered, it is also more difficult to understand how Darwinian selection of hypercycles could result in a unique genetic code.  相似文献

There is little direct literature detailing exhaustive bacteriological studies comparing human donor faecal flora, human flora‐associated (HFA) mouse models and conventional rodent faecal flora. While there is a premise that the implanted donor faecal flora from humans is established in the rodent model the evidence is incomplete and indeed for groups such as Bifidobacterium spp. it is lacking. The reviewed bacteriology studies are generally lacking in detail with the exception of one study from which the data have mostly been overlooked when cited by other workers. While there are studies that suggest that the HFA rodent model is more relevant to man than studies with conventional rodents, the hypothesis remains to be proven. This review concludes that the established microbial flora in the HFA rodent model is different to that of donor human faecal flora, and this clearly raises the question as to whether this matters, after all a model is a model and as such models can be useful even should they fail to be a true representation of, in this case, the gastrointestinal tract. What matters is that there is a proper understanding of the limitations of the model as we attempt to unravel the significance of the components of the gastrointestinal flora in health and disease; examples of why such an analysis is important are provided with regard to obesity and nutritional studies. The data do unsurprisingly suggest that diet is an extremely influential variable when interpreting HFA and conventional rodent data. The microbiology data from direct bacteriology and indirect enzyme studies show that the established microbial flora in the HFA rodent model is different to that of donor human faecal flora. The significance of this conclusion remains to be established.  相似文献

This paper summarizes trends in Australian wetland rehabilitation on the basis of responses to a questionnaire from sixty-nine rehabilitation projects and a literature review. Project sizes ranged from 0.4 to 110,000 ha. Costs ranged from A$6 to 70,000 ha–1 yr–1, with most projects costing less than A$1,000 ha–1 yr–1 and with larger projects generally costing less per area than smaller projects. The oldest project began in 1963, but most projects began between 1990 and 1996. The most commonly cited dominant plant genera in rehabilitation projects were Eucalyptus, Melaleuca, Muehlenbeckia, Juncus, Avicennia, and Typha. Filling and draining and altered hydrology were the most commonly cited impacts that led to a need for rehabilitation, while excavation and planting were the most commonly cited rehabilitation methods. About 65% of projects undertake rehabilitation methods that address the causes of impacts. Habitat improvement was the most commonly stated objective in wetland rehabilitation. Of the projects that supported monitoring programs, fewer than 25% appeared to monitor variables that were closely linked to project goals. Thirty-six projects reported involvement in research other than monitoring, with vegetation research most frequently cited. Most projects encouraged some level of community involvement. Wetland rehabilitation in Australia could benefit from a centralized register of projects and a library of rehabilitation literature, including government and in-house reports.  相似文献

A time-dependent simulation model, based on the Coburn-Forster-Kane equation, was written in Advanced Continuous Simulation Language to predict carboxyhemoglobin (HbCO) formation and dissociation in F-344 rats during and after exposure to 500 parts/million CO for 1 h. Blood-gas analysis and CO-oximetry were performed on samples collected during exposure and off-gassing of CO. Volume displacement plethysmography was used to measure minute ventilation (VE) during exposure. CO diffusing capacity in the lung (DLCO) was also measured. Other model parameters measured in the animals included blood pH, total blood volume, and Hb concentration. Comparisons between model predictions using values for VE, DLCO, and the Haldane coefficient cited in the literature and predictions using measured VE, DLCO, and calculated Haldane coefficient for individual animals were made. General model predictions using values for model parameters derived from the literature agreed with published HbCO values by a factor of 0.987 but failed to simulate experimental data. On average, the general model overpredicted measured HbCO level by nearly 9%. A specific model using the means of measured variables predicted HbCO concentration within a factor of 0.993. When experimentally observed parameter fluctuations were included, the specific model predictions reflected experimental effects on HbCO formation.  相似文献

A spectrofluorometric technique for bacterial DNA base composition has been developed. This fast and simple technique requires two fluorescent dyes and a few inexpensive reagents. The data from this assay indicate that the guanine-cytosine content obtained was within acceptable statistical limits in comparison to commonly cited literature values. The spectrofluorometric technique is reliable and reproducible.  相似文献

Studies of the organization of double-stranded DNA within bacteriophage heads during the past four decades have produced a wealth of data. However, despite the presentation of numerous models, the true organization of DNA within phage heads remains unresolved. The observations of toroidal DNA structures in electron micrographs of phage lysates have long been cited as support for the organization of DNA in a spool-like fashion. This particular model, like all other models, has not been found to be consistent will all available data. Recently we proposed that DNA within toroidal condensates produced in vitro is organized in a manner significantly different from that suggested by the spool model. This new toroid model has allowed the development of an alternative model for DNA organization within bacteriophage heads that is consistent with a wide range of biophysical data. Here we propose that bacteriophage DNA is packaged in a toroid that is folded into a highly compact structure.  相似文献

This paper provides an analysis of the literature on trunk muscle recruitment in low-back pain patients. Two models proposed in the literature, the pain-spasm-pain model and the pain adaptation model, yield conflicting predictions on how low- back pain would affect trunk muscle recruitment in various activities. The two models are outlined and evidence for the two from neurophsysiological studies is reviewed. Subsequently, specific predictions with respect to changes in activation of the lumbar extensor musculature are derived from both models. These predictions are compared to the results from 30 clinical studies and three induced pain studies retrieved in a comprehensive literature search. Neither of the two models is unequivocally supported by the literature. These data and further data on timing of muscle activity and load sharing between muscles suggest an alternative model to explain the alterations of trunk muscle recruitment due to low-back pain. It is proposed that motor control changes in patients are functional in that they enhance spinal stability.  相似文献

We report here a mathematical model using computer simulation to solve the phase fractionation coefficient (f) of instantaneous insulin release on glucose infusion. By extensive model testing with the cited parameters obtained from the literature, the values of the factor f were shown to lie in range of 0.93+/-0.02 (mean+/-2S.D., n=15), indicating that the high pulsatile bolus of glucose by i.v. infusion may trigger acute insulin release (AIR) corresponding to a fraction of more than 90% of the stored insulin release in the first phase from the secretory granules of pancreatic beta cells. In addition, the value of the factor f was shown to be independent of both the glucose infusion method and the non-insulin-dependent uptake of glucose.  相似文献

The most commonly accepted secondary structure models for 5S RNA differ for molecules of eubacterial origin, where the four-helix model of Fox and Woese is generally cited, and those of eukaryotic origin, where a fifth helix is assumed to exist. We have carefully aligned all available sequences from eukaryotes, eubacteria, chloroplasts, archaebacteria and plant mitochondria. We could thus derive a unified secondary structure model applicable to all 5S RNA sequences known to-date. It contains the five helices already present in the eukaryotic model, extended by additional segments that were not previously assumed to be universally present. One of the helices can be written in two equilibrium forms, which could reflect the existence of a flexible, dynamic structure. For the derivation of the model and the estimation of the free energies we followed a set of rules optimized to predict the tRNA cloverleaf. The stability of the unified model is higher than that of nearly all previously proposed sequence-specific and general models.  相似文献

Pushout tests can be used to estimate the shear strength of the bone implant interface. Numerous such experimental studies have been published in the literature. Despite this researchers are still some way off with respect to the development of accurate numerical models to simulate implant stability. In the present work a specific experimental pushout study from the literature was simulated using two different bones implant interface models. The implant was a porous coated Ti-6Al-4V retrieved 4 weeks postoperatively from a dog model. The purpose was to find out which of the interface models could replicate the experimental results using physically meaningful input parameters. The results showed that a model based on partial bone ingrowth (ingrowth stability) is superior to an interface model based on friction and prestressing due to press fit (initial stability). Even though the present study is limited to a single experimental setup, the authors suggest that the presented methodology can be used to investigate implant stability from other experimental pushout models. This would eventually enhance the much needed understanding of the mechanical response of the bone implant interface and help to quantify how implant stability evolves with time.  相似文献

Fatigue failure of the cement mantle has been proposed as one of the failure processes contributing to aseptic loosening of cemented joint replacements. It has also been suggested that fatigue failure is dramatically accelerated by residual stress generated during the cement polymerisation process. Previous computational models of the polymerisation process have investigated only the latter part of polymerisation by assuming both instantaneous hardening of the material (a stress locking point) and that all residual stress results from thermal shrinkage after this stress locking point. In this study, finite element models which use the local degree of polymerisation to calculate material properties and shrinkage have been used to predict residual stresses in two models of total hip replacement cement mantles. Results indicate that the final value of cement mantle stress may not be the highest stresses that the cement is subjected to during the polymerisation process. Two models are presented, a 2-dimensional model, which was adapted from a similar model in the literature (Lennon and Prendergast, 2002) and a 3-dimensional concentric-cylinders model. In both cases a chemical kinetics model was used to predict the progress of the polymerisation reaction and a second linear model used to predict cement mechanical properties and density, and so stress generation and volume change, over time. There was good agreement of the results of the 2D model with its counterpart in the literature. For the 3D model, the final residual stress magnitudes and patterns showed good agreement with similar physical and computational models in the literature.  相似文献

Experiments were conducted on the duration of life in sea water of three species of cystophorous cercariae from the Barents sea. The expenditure of reserve nutritive substances (glycogene) in the process of the vital activity of cercariae was followed. The life duration of cystophorous cercariae is considerably longer than that cited in literature for other groups of cercariae that is associated with their morphological and biological peculiarities.  相似文献
