Biopsies fro the vastus lateralis muscle of male weightlifters (WL; n=6; X +/- SE, age=27.0 +/- 2.1 years), and non-weight-trained men (CON; n=7; age=27.0 +/- 2.0 years) were compared for fiber types, myosin heavy chain (MHC) and titin content, and fiber type-specific capillary density. Differences (p<0.05) were observed for percent fiber types IIC (WL=0.4 +/- 0.2, CON=2.4 +/- 0.8); IIA (WL=50.5 +/- 3.2, CON=26.9 +/- 3.7); and IIB (WL=1.7 +/- 1.4, CON=21.0 +/- 5.3), as well as percent MHC IIa (WL=65.3 +/- 2.4, CON=52.1 +/- 4.2) and percent MHC IIB (WL=0.9 +/- 0.9; CON=18.2 +/- 6.1). All WL exhibited only the titin-1 isoform. Capillary density (caps.mm(-2)) for all fiber types combined was greater for the CON subjects (WL=192.7 +/- 17.3; CON=262.9 +/- 26.3), due primarily to a greater capillary density in the IIA fibers. Weightlifting performances and vertical jump power were correlated with type II fiber characteristics. These results suggest that successful weightlifting performance is not dependent on IIB fibers, and that weightlifters exhibit large percentages of type IIA muscle fibers and MHC IIa isoform content.  相似文献

Fiber composition and oxidative capacity of hamster skeletal muscle.   总被引:6,自引:0,他引:6  
The hamster is a valuable biological model for physiological investigation. Despite the obvious importance of the integration of cardiorespiratory and muscular system function, little information is available regarding hamster muscle fiber type and oxidative capacity, both of which are key determinants of muscle function. The purpose of this investigation was to measure immunohistochemically the relative composition and size of muscle fibers composed of types I, IIA, IIX, and IIB fibers in hamster skeletal muscle. The oxidative capacity of each muscle was also assessed by measuring citrate synthase activity. Twenty-eight hindlimb, respiratory, and facial muscles or muscle parts from adult (144-147 g bw) male Syrian golden hamsters (n=3) were dissected bilaterally, weighed, and frozen for immunohistochemical and biochemical analysis. Combining data from all 28 muscles analyzed, type I fibers made up 5% of the muscle mass, type IIA fibers 16%, type IIX fibers 39%, and type IIB fibers 40%. Mean fiber cross-sectional area across muscles was 1665 +/- 328 microm(2) for type I fibers, 1900 +/- 417 microm(2) for type IIA fibers, 3230 +/- 784 microm(2) for type IIX fibers, and 4171 +/- 864 microm(2) for type IIB fibers. Citrate synthase activity was most closely related to the population of type IIA fibers (r=0.68, p<0.0001) and was in the rank order of type IIA > I > IIX > IIB. These data demonstrate that hamster skeletal muscle is predominantly composed of type IIB and IIX fibers.  相似文献

This study presents data collected over the past 10 years on the muscle fiber type composition of the vastus lateralis muscle of young men and women. Biopsies were taken from the vastus lateralis muscle of 55 women (21.2+/-2.2 yr) and 95 men (21.5+/-2.4 yr) who had volunteered to participate in various research projects. Six fiber types (I, IC, IIC, IIA, IIAB, and IIB) were classified using mATPase histochemistry, and cross-sectional area was measured for the major fiber types (I, IIA, and IIB). Myosin heavy chain (MHC) content was determined electrophoretically on all of the samples from the men and on 26 samples from the women. With the exception of fiber Type IC, no significant differences were found between men and women for muscle fiber type distribution. The vastus lateralis muscle of both the men and women contained approximately 41% I, 1% IC, 1% IIC, 31% IIA, 6% IIAB, and 20% IIB. However, the cross-sectional area of all three major fiber types was larger for the men compared to the women. In addition, the Type IIA fibers were the largest for the men, whereas the Type I fibers tended to be the largest for the women. Therefore, gender differences were found with regard to the area occupied by each specific fiber type: IIA>I>IIB for the men and I>IIA>IIB for the women. These data establish normative values for the mATPase-based fiber type distribution and sizes in untrained young men and women.  相似文献

Human skeletal muscle fiber type adaptability to various workloads   总被引:1,自引:0,他引:1  
Muscle biopsy specimens were removed from the vastus lateralis muscles of three groups of human subjects: controls, weight lifters, and distance runners. The runners proved to be a unique group with respect to the variables measured (low body weight and percentage body fat, and high VO2 max). Additionally, a histochemical analysis of the biopsy specimens revealed that the runners had a significantly higher percentage of fiber types I and IIC than either the controls or the weight lifters. Using a cryostatic retrieval method, each of the fibers identified histochemically was then analyzed morphometrically using electron microscopy. The results of volume-percent mitochondria demonstrated a strong relationship between the ATPase activity and oxidative potential of the fiber types for all three groups such that the oxidative activity would be ranked I greater than IIA greater than IIB. Irrespective of fiber type, there were significant differences between the groups with regard to muscle-fiber mitochondrial (runners greater than lifters greater than controls) and lipid content (runners greater than controls greater than lifters). The lifters had a significantly greater content of mitochondria than the controls, which may suggest that inactivity rather than the lifting exercise contributes to a low volume-percent mitochondria and a high percentage of type IIB fibers.  相似文献

This study investigated the effects of ballistic resistance training and strength training on muscle fiber composition, peak force (PF), maximal strength, and peak power (PP). Fourteen males (age = 21.3 +/- 2.9, body mass = 77.8 +/- 10.1 kg) with 3 months of resistance training experience completed the study. Subjects were tested pre and post for their squat one-repetition maximum (1RM) and PP in the jump squat (JS). Peak force and rate of force development (RFD) were tested during an isometric midthigh pull. Muscle biopsies were obtained from the vastus lateralis for analysis of muscle fiber type expression. Subjects were matched for strength and then randomly selected into either training (T) or control (C) groups. Group T performed 8 weeks of JS training using a periodized program with loading between 26 and 48% of 1RM, 3 days per week. Group T showed significant improvement in PP from 4088.9 +/- 520.6 to 5737.6 +/- 651.8 W. Rate of force development improved significantly in group T from 12687.5 +/- 4644.0 to 25343.8 +/- 12614.4 N x s(-1). PV improved significantly from 1.59 +/- 0.41 to 2.11 +/- 0.75 m x s(-1). No changes occurred in PF, 1RM, or muscle fiber type expression for group T. No changes occurred in any variables in group C. The results of this study indicate that using ballistic resistance exercise is an effective method for increasing PP and RFD independently of changes in maximum strength (1RM, PF), and those increases are a result of factors other than changes in muscle fiber type expression.  相似文献

The purpose of this study was to verify the concurrent validity of a bar-mounted Myotest? instrument in measuring the force and power production in the squat and bench press exercises when compared to the gold standard of a computerized linear transducer and force platform system. Fifty-four men (bench press: 39-171 kg; squat: 75-221 kg) and 43 women (bench press: 18-80 kg; squat: 30-115 kg) (age range 18-30 years) performed a 1 repetition maximum (1RM) strength test in bench press and squat exercises. Power testing consisted of the jump squat and the bench throw at 30% of each subject's 1RM. During each measurement, both the Myotest? instrument and the Celesco linear transducer of the directly interfaced BMS system (Ballistic Measurement System [BMS] Innervations Inc, Fitness Technology force plate, Skye, South Australia, Australia) were mounted to the weight bar. A strong, positive correlation (r) between the Myotest and BMS systems and a high correlation of determination (R2) was demonstrated for bench throw force (r = 0.95, p < 0.05) (R2 = 0.92); bench throw power (r = 0.96, p < 0.05) (R2 = 0.93); squat jump force (r = 0.98, p < 0.05) (R2 = 0.97); and squat jump power (r = 0.91, p < 0.05) (R2 = 0.82). In conclusion, when fixed on the bar in the vertical axis, the Myotest is a valid field instrument for measuring force and power in commonly used exercise movements.  相似文献

The purpose of this investigation was to determine if there was a difference in kinetic variables and muscle activity when comparing a squat to a box squat. A box squat removes the stretch-shortening cycle component from the squat, and thus, the possible influence of the box squat on concentric phase performance is of interest. Eight resistance trained men (Height: 179.61 ± 13.43 cm; Body Mass: 107.65 ± 29.79 kg; Age: 24.77 ± 3.22 years; 1 repetition maximum [1RM]: 200.11 ± 58.91 kg) performed 1 repetition of squats and box squats using 60, 70, and 80% of their 1RM in a randomized fashion. Subjects completed the movement while standing on a force plate and with 2 linear position transducers attached to the bar. Force and velocity were used to calculate power. Peak force and peak power were determined from the force-time and power-time curves during the concentric phase of the lift. Muscle activity (electromyography) was recorded from the vastus lateralis, vastus medialis, biceps femoris, and longissimus. Results indicate that peak force and peak power are similar between the squat and box squat. However, during the 70% of 1RM trials, the squat resulted in a significantly lower peak force in comparison to the box squat (squat = 3,269 ± 573 N, box squat = 3,364 ± 575 N). In addition, during the 80% of 1RM trials, the squat resulted in significantly lower peak power in comparison to the box squat (squat = 2,050 ± 486 W, box squat = 2,197 ± 544 W). Muscle activity was generally higher during the squat in comparison to the box squat. In conclusion, minimal differences were observed in kinetic variables and muscle activity between the squat and box squat. Removing the stretch-shortening cycle during the squat (using a box) appears to have limited negative consequences on performance.  相似文献

The purpose of this study was to determine the acute effects of a spectrum of eccentric loads on force, velocity, and power during the concentric portion of maximal-effort jump squats utilizing a repeated measures design. Thirteen resistance-trained men (age = 22.8 +/- 2.9 years, weight = 87.1 +/- 11.8 kg, 163.5 +/- 28.6 kg squat 1 repetition maximum [1RM]; mean +/- SD), who routinely incorporated back squats into their training, participated as subjects in this investigation. Jump squat performance was assessed using 4 experimental conditions. The first of these conditions consisted of an isoinertial load equal to 30% of back squat 1RM. The remaining conditions consisted of jump squats with a concentric load of 30% 1RM, subsequent to the application of experimental augmented eccentric loading (AEL) conditions of 20, 50, and 80% of back squat 1RM, respectively. All subjects performed 2 sets of 1RM of maximum-effort jump squats with all experimental conditions in a counter-balanced sequence. Forty-eight hours after completing the first testing session, subjects repeated the experimental testing protocol to establish stability reliability. Peak performance values for the reliable variables of force, velocity, and power, as well as force and power values obtained at 20-ms intervals during the initial 400 ms of the concentric jump squat range of motion, showed no statistical difference (p > 0.05) across the experimental AEL loads. These results suggest that load-spectrum AEL prior to a 30% 1RM jump squat fails to acutely enhance force, velocity, and power.  相似文献

Fifteen highly trained distance runners VO(2)max 71.1 +/- 6.0 ml.min(-1).kg(-1), mean +/- SD) were randomly assigned to a plyometric training (PLY; n = 7) or control (CON; n = 8) group. In addition to their normal training, the PLY group undertook 3 x 30 minutes PLY sessions per week for 9 weeks. Running economy (RE) was assessed during 3 x 4 minute treadmill runs (14, 16, and 18 km.h(-1)), followed by an incremental test to measure VO(2)max. Muscle power characteristics were assessed on a portable, unidirectional ground reaction force plate. Compared with CON, PLY improved RE at 18 km.h(-1) (4.1%, p = 0.02), but not at 14 or 16 km.h(-1). This was accompanied by trends for increased average power during a 5-jump plyometric test (15%, p = 0.11), a shorter time to reach maximal dynamic strength during a strength quality assessment test (14%, p = 0.09), and a lower VO(2)-speed slope (14%, p = 0.12) after 9 weeks of PLY. There were no significant differences in cardiorespiratory measures or VO(2)max as a result of PLY. In a group of highly-trained distance runners, 9 weeks of PLY improved RE, with likely mechanisms residing in the muscle, or alternatively by improving running mechanics.  相似文献

Neuromuscular and hormonal adaptations to prolonged strength training were investigated in nine elite weight lifters. The average increases occurred over the 2-yr follow-up period in the maximal neural activation (integrated electromyogram, IEMG; 4.2%, P = NS), maximal isometric leg-extension force (4.9%, P = NS), averaged concentric power index (4.1%, P = NS), total weight-lifting result (2.8%, P less than 0.05), and total mean fiber area (5.9%, P = NS) of the vastus lateralis muscle, respectively. The training period resulted in increases in the concentrations of serum testosterone from 19.8 +/- 5.3 to 25.1 +/- 5.2 nmol/l (P less than 0.05), luteinizing hormone (LH) from 8.6 +/- 0.8 to 9.1 +/- 0.8 U/l (P less than 0.05), follicle-stimulating hormone (FSH) from 4.2 +/- 2.0 to 5.3 +/- 2.3 U/l (P less than 0.01), and testosterone-to-serum sex hormone-binding globulin (SHBG) ratio (P less than 0.05). The annual mean value of the second follow-up year for the serum testosterone-to-SHBG ratio correlated significantly (r = 0.84, P less than 0.01) with the individual changes during the 2nd yr in the averaged concentric power. The present results suggest that prolonged intensive strength training in elite athletes may influence the pituitary and possibly hypothalamic levels, leading to increased serum levels of testosterone. This may create more optimal conditions to utilize more intensive training leading to increased strength development.  相似文献

This study investigated whether hyperoxic breathing (100% O(2)) or increasing oxidative substrate supply [dichloroacetate (DCA) infusion] would increase oxidative phosphorylation and reduce the reliance on substrate phosphorylation at the onset of high-intensity aerobic exercise. Eight male subjects cycled at 90% maximal O(2) uptake (VO(2 max)) for 90 s in three randomized conditions: 1) normoxic breathing and saline infusion over 1 h immediately before exercise (CON), 2) normoxic breathing and saline infusion with DCA (100 mg/kg body wt), and 3) hyperoxic breathing for 20 min at rest and during exercise and saline infusion (HYP). Muscle biopsies from the vastus lateralis were sampled at rest and after 30 and 90 s of exercise. DCA infusion increased pyruvate dehydrogenase (PDH) activation above CON and HYP (3.10 +/- 0.23, 0.56 +/- 0.08, 0.69 +/- 0.05 mmol x kg wet muscle(-1) x min(-1), respectively) and significantly increased both acetyl-CoA and acetylcarnitine (11.0 +/- 0.7, 2.0 +/- 0.5, 2.2 +/- 0.5 mmol/kg dry muscle, respectively) at rest. However, DCA and HYP did not alter phosphocreatine degradation and lactate accumulation and, therefore, the reliance on substrate phosphorylation during 30 s (CON, 51.2 +/- 5.4; DCA, 56.5 +/- 7.1; HYP, 69.5 +/- 6.3 mmol ATP/kg dry muscle) and 90 s of exercise (CON, 90.6 +/- 9.5; DCA, 107.2 +/- 13.0; HYP, 101.2 +/- 15.2 mmol ATP/kg dry muscle). These data suggest that the rate of oxidative phosphorylation at the onset of exercise at 90% VO(2 max) is not limited by oxygen availability to the active muscle or by substrate availability (metabolic inertia) at the level of PDH in aerobically trained subjects.  相似文献

To understand the molecular mechanism underlying the diversity of mammalian skeletal muscle fibers, the elementary steps of the cross-bridge cycle were investigated in three fast-twitch fiber types from rabbit limb muscles. Skinned fibers were maximally Ca(2+)-activated at 20 degrees C and the effects of MgATP, phosphate (P, P(i)), and MgADP were studied on three exponential processes by sinusoidal analysis. The fiber types (IIA, IID, and IIB) were determined by analyzing the myosin heavy-chain isoforms after mechanical experiments using high-resolution SDS-PAGE. The results were consistent with the following cross-bridge scheme: where A is actin, M is myosin, D is MgADP, and S is MgATP. All states except for those in brackets are strongly bound states. All rate constants of elementary steps (k(2), 198-526 s(-1); k(-2), 51-328 s(-1); k(4), 13.6-143 s(-1); k(-4), 13.6-81 s(-1)) were progressively larger in the order of type IIA, type IID, and type IIB fibers. The rate constants of a transition from a weakly bound state to a strongly bound state (k(-2), k(4)) varied more among fiber types than their reversals (k(2), k(-4)). The equilibrium constants K(1) (MgATP affinity) and K(2) (=k(2)/k(-2), ATP isomerization) were progressively less in the order IIA, IID, and IIB. K(4) (=k(4)/k(-4), force generation) and K(5) (P(i) affinity) were larger in IIB than IIA and IID fibers. K(1) showed the largest variation indicating that the myosin head binds MgATP more tightly in the order IIA (8.7 mM(-1)), IID (4.9 mM(-1)), and IIB (0.84 mM(-1)). The purpose of this study was to examine single cell contractile mechanics of skeletal muscle before and after 12 wk of progressive resistance training (PRT) in older men (n = 7; age = 74 +/- 2 yr and weight = 75 +/- 5 kg). Knee extensor PRT was performed 3 days/wk at 80% of one-repetition maximum. Muscle biopsy samples were obtained from the vastus lateralis before and after PRT (pre- and post-PRT, respectively). For analysis, chemically skinned single muscle fibers were studied at 15 degrees C for peak tension [the maximal isometric force (P(o))], unloaded shortening velocity (V(o)), and force-velocity parameters. In this study, a total of 199 (89 pre- and 110 post-PRT) myosin heavy chain (MHC) I and 99 (55 pre- and 44 post-PRT) MHC IIa fibers were reported. Because of the minimal number of hybrid fibers identified post-PRT, direct comparisons were limited to MHC I and IIa fibers. Muscle fiber diameter increased 20% (83 +/- 1 to 100 +/- 1 microm) and 13% (86 +/- 1 to 97 +/- 2 microm) in MHC I and IIa fibers, respectively (P < 0.05). P(o) was higher (P < 0.05) in MHC I (0.58 +/- 0.02 to 0.90 +/- 0.02 mN) and IIa (0.68 +/- 0.02 to 0.85 +/- 0.03 mN) fibers. Muscle fiber V(o) was elevated 75% (MHC I) and 45% (MHC IIa) after PRT (P < 0.05). MHC I and IIa fiber power increased (P < 0.05) from 7.7 +/- 0.5 to 17.6 +/- 0.9 microN. fiber lengths. s(-1) and from 25.5 to 41.1 microN. fiber lengths. s(-1), respectively. These data indicate that PRT in elderly men increases muscle cell size, strength, contractile velocity, and power in both slow- and fast-twitch muscle fibers. However, it appears that these changes are more pronounced in the MHC I muscle fibers.  相似文献

Five male cyclists were studied during 2 h of cycle ergometer exercise (70% VO2 max) on two occasions to examine the effect of carbohydrate ingestion on muscle glycogen utilization. In the experimental trial (CHO) subjects ingested 250 ml of a glucose polymer solution containing 30 g of carbohydrate at 0, 30, 60, and 90 min of exercise; in the control trial (CON) they received an equal volume of a sweet placebo. No differences between trials were seen in O2 uptake or heart rate during exercise. Venous blood glucose was similar before exercise in both trials, but, on average, was higher during exercise in CHO [5.2 +/- 0.2 (SE) mmol/l] compared with CON (4.8 +/- 0.1, P less than 0.05). Plasma insulin levels were similar in both trials. Muscle glycogen levels were also similar in CHO and CON both before and after exercise; accordingly, there was no difference between trials in the amount of glycogen used during the 2 h of exercise (CHO = 62.8 +/- 10.1 mmol/kg wet wt, CON = 56.9 +/- 10.1). The results of this study indicate that carbohydrate ingestion does not influence the utilization of muscle glycogen during prolonged strenuous exercise.  相似文献

It has been established that, in the sports of Olympic weightlifting (OL) and powerlifting (PL), the relationship between lifting performance and body mass is not linear. This relationship has been frequently studied in OL, but the literature on PL is less extensive. In this study, PL performance and body mass, for both men and women, was examined by using data from the International Powerlifting Federation World Championships during 1995-2004. Nonlinear regression was used to apply 7 models (including allometric, polynomial, and power models) to the data. The results of this study indicate that the relationship between PL performance and body mass can be best modeled by the equation y = a - bx(-c), where y is the weight lifted (in kg) in the squat, bench press, or deadlift, x is the body mass of the lifter (in kg), and a, b, and c are constants. The constants a, b, and c are determined by the type of lift (squat, bench press, or deadlift) and the gender of the lifter and were obtained from the regression analysis. Inspection of the plots of raw residuals (actual performance minus predicted performance) vs. body mass revealed no body mass bias to this formula in contrast to research into other handicapping formulas. This study supports previous research that found a bias toward lifters in the intermediate weight categories in allometric fits to PL data.  相似文献

Histochemical characteristics of soleus muscle were compared in human growth hormone (hGH) transgenic mice vs their nontransgenic littermates. Plasma of transgenic mice contained hGH (7.1 +/- 0.7 and 6.7 +/- 0.4 ng/ml, mean +/- SE, at 5 and 11 months of age, respectively); hGH was not detectable in plasma of nontransgenic littermates. Body and soleus weights were greater (approximately 55 and 25%, respectively) and both type I and type IIA fibers were larger in transgenic animals. Most significantly, fiber type composition of the soleus muscle was different in hGH-transgenic animals, i.e., the percentage of type I fibers was significantly greater than in nontransgenic mice (77.2 +/- 5.1% vs 58.4 +/- 2.5%). It is generally believed that skeletal muscle fiber composition is determined predominantly by neural influences (1, 2). These data suggest hormonal factors, growth hormone, also affect the phenotype of skeletal muscle myosin.  相似文献

To determine the effects of weight loss on intramyocellular energy substrates, vastus lateralis muscle biopsies were taken from six obese subjects (body mass index 34 +/- 5 kg/m(2)) before, after 15 wk of energy restriction (ER; -700 kcal/day), and after a further average 20.7 +/- 1.6 wk of endurance training plus low-fat diet (ET-LFD). Body weight fell from 100 +/- 6 to 89 +/- 6 kg during ER and to 84 +/- 4 kg after ET-LFD. Lipids and glycogen were histochemically measured in type I, IIA, and IIB fibers. Total muscle glycogen content (MGC; per 100 fibers) decreased after ER [from 72 +/- 13 to 55 +/- 8 arbitrary units (AU)]. A similar but not significant decrease was seen in total muscle lipid content (MLC; 14 +/- 5 to 9 +/- 1 AU). After ET-LFD, MGC returned to initial values (74 +/- 8 AU), and MLC approached near-initial values (12 +/- 3 AU). Individual fiber lipid concentration did not change throughout the protocol in all fiber types, whereas glycogen concentration increased after ET-LFD. The training effects of ET-LFD were measured as increasing activities of key mitochondrial enzymes. Although total muscle energy reserves can be reduced after weight loss, their concentration within individual myofibers remains elevated. Weight loss does not appear sufficient to correct the potential detrimental effects of high intracellular lipid concentrations.  相似文献

This study examined the changes in peak power, ground reaction force and velocity with different loads during the performance of the parallel squat movement. Twelve experienced male lifters (26.83 +/- 4.67 years of age) performed the standard parallel squat, using loads equal to 20, 30, 40, 50, 60, 70, 80, and 90% of 1 repetition maximum (1RM). Each subject performed all parallel squats with as much explosiveness as possible using his own technique. Peak power (PP), peak ground reaction force (PGRF), peak barbell velocity (PV), force at the time of PP (FPP), and velocity at the time of PP (VPP) were determined from force, velocity, and power curves calculated using barbell velocity and ground reaction force data. No significant differences were detected among loads for PP; however, the greatest PP values were associated with loads of 40 and 50% of 1RM. Higher loads produced greater PGRF and FPP values than lower loads (p < 0.05) in all cases except between loads equal to 60-50, 50-40, and 40-30% of 1RM for PGRF, and between loads equal to 70-60 and 60-50% of 1RM for FPP. Higher loads produced lower PV and VPP values than lower loads (p < 0.05) in all cases except between the 20-30, 70-80, and 80-90% of 1RM conditions. These results may be helpful in determining loads when prescribing need-specific training protocols targeting different areas of the load-velocity continuum.  相似文献

5'-AMP-activated protein kinase (AMPK) signaling initiates adaptive changes in skeletal muscle fibers that restore homeostatic energy balance. The purpose of this investigation was to examine, in rats, the fiber-type protein expression patterns of the alpha-catalytic subunit isoforms in various skeletal muscles, and changes in their respective contents within the tibialis anterior (TA) after chronic low-frequency electrical stimulation (CLFS; 10 Hz, 10 h daily), applied for 4 +/- 1.2 or 25 +/- 4.8 days. Immunocytochemical staining of soleus (SOL) and medial gastrocnemius (MG) showed that 86 +/- 4.1 to 97 +/- 1.4% of type IIA fibers stained for both the alpha1- and alpha2-isoforms progressively decreased to 63 +/- 12.2% of type IID/X and 9 +/- 2.4% of IIB fibers. 39 +/- 11.4% of IID/X and 83 +/- 7.9% of IIB fibers expressed only the alpha2 isoform in the MG, much of which was localized within nuclei. alpha1 and alpha2 contents, assessed by immunoblot, were lowest in the white gastrocnemius [WG; 80% myosin heavy chain (MHC) IIb; 20% MHCIId/x]. Compared with the WG, alpha1 content was 1.6 +/- 0.08 (P < 0.001) and 1.8 +/- 0.04 (P < 0.0001)-fold greater in the red gastrocnemius (RG: 13%, MHCIIa) and SOL (21%, MHCIIa), respectively, and increased in proportion to MHCIIa content. Similarly, alpha2 content was 1.4 +/- 0.10 (P < 0.02) and 1.5 +/- 0.07 (P < 0.001)-fold greater in RG and SOL compared with WG. CLFS induced 1.43 +/- 0.13 (P < 0.007) and 1.33 +/- 0.08 (P < 0.009)-fold increases in the alpha1 and alpha2 contents of the TA and coincided with the transition of faster type IIB and IID/X fibers toward IIA fibers. These findings indicate that fiber types differ with regard to their capacity for AMPK signaling and that this potential is increased by CLFS.  相似文献
