Using the pulse radiolysis technique it was shown that copper(II) complexes of kinetin and 6-benzylaminopurine (6-BAP) catalyze O?2 dismutation very efficiently at physiological pH. The ‘turnover’ rate constants at pH 7 were determined to be (1.5 ± 0.3) × 109 and (2.2 ± 0.4) × 109 M?1 s?1for 6-BAP and kinetin, respectively. The system was studied at pH 3–10 in the case of 6-BAP, and the results show that this complex catalyzes also HO2 dismutation efficiently.  相似文献

Using the direct method of pulse radiolysis to determine the superoxide dismutase like activity of copper(II) cimetidine complexes, it was found that the reaction rate constant with O?2, kcat, was (8.5 ± 0.5) × 108 M?1s?1 independent of the cimetidine concentrations present in excess of 50–200 μM over the metal. The results suggest that either the 1:1 ligand to metal complex does not catalyze O?2 dismutation at a comparable rate to that of the 2:1 complex, or that the stability constant of the last species is much higher than that determined earlier by Kimura el al.,1 and only the 2:1 species is present in the solutions. With the indirect methods of cytochrome c and NBT for determining the ability of these complexes to catalyze O?2 dismutation, these compounds exhibited a much lower SOD activity. and kcat was determined to be (5.0 ± 0.3) × 106 and (7.± 0.4) × 101 M?1s?1. respectively using the two assays.  相似文献

Abstract

Azo linked salicyldehyde and a new 2-hydroxy acetophenone based ligands (HL1 and HL2) with their copper(II) complexes [Cu(L1)2] (1) and [Cu(L2)2] (2) were synthesized and characterized by spectroscopic methods such as 1H, 13C NMR, UV–Vis spectroscopy and elemental analyses. Calculation based on Density Functional Theory (DFT), have been performed to obtain optimized structures. Binding studies of these copper (II) complexes with calf thymus DNA (ct-DNA) and torula yeast RNA (t-RNA) were analyzed by absorption spectra, emission spectra and Viscosity studies and Molecular Docking techniques. The absorption spectral study indicated that the copper(II) complexes of 1 and 2 had intrinsic binding constants with DNA or RNA in the range of 7.6?±?0.2?×?103?M?1 or 6.5?±?0.3?×?103M?1 and 5.7?±?0.4?×?104 M?1 or 1.8?±?0.5?×?103 M?1 respectively. The synthesized compounds and nucleic acids were simulated by molecular docking to explore more details mode of interaction of the complexes and their orientations in the active site of the receptor.  相似文献

The electron transfer reactions of horse heart cytochrome c with a series of amino acid-pentacyanoferrate(II) complexes have been studied by the stopped-flow technique, at 25°C, μ = 0.100, pH 7 (phosphate buffer). A second-order behavior was observed in the case of the Fe(CN)5 (histidine)3? complex, with k = 2.8 x 105 M?1 sec?1. For the Fe(CN)5 (alanine)4? and Fe(CN)5(L-glutamate)5? complexes, only a minor deviation of the second-order behavior, close to the experimental error (k = 3.2 × 105 and 1.6 x 105 M?1 sec?1, respectively) was noted at high concentrations of the reactants (e.g., 6 × 10?4 M). The results are in accord with recent work on the Fe(CN)64?/cytochrome c system demonstrating weak association of the reactants. The calculated self-exchange rate constants including electrostatic interactions for the imidazole,L -histidine, 4-aminopyridine, glycinate, β-alaninate, andL-glutamate pentacyanoferrate(II) complexes were 3.3 × 105, 3.3 × 105, 2.8 × 106,4.1 × 102,5.5 × 102, and 6.0 M?1 sec?1, respectively. Marcus theory calculations for the cytochrome c reactions were interpreted in terms of two nonequivalent binding sites for the complexes, with the metalloprotein self-exchange rate constants varying from 104 M?1 sec?1 (histidine, imidazole, and 4-aminopyridine complexes) to 106 M?1 sec ?1 (glycinate, β-alaninate, and L-glutamate complexes).  相似文献

Four complexes [Pd(L)(bipy)Cl]·4H2O (1), [Pd(L)(phen)Cl]·4H2O (2), [Pt(L)(bipy)Cl]·4H2O (3), and [Pt(L)(phen)Cl]·4H2O (4), where L = quinolinic acid, bipy = 2,2’-bipyridyl, and phen = 1,10-phenanthroline, have been synthesized and characterized using IR, 1H NMR, elemental analysis, and single-crystal X-ray diffractometry. The binding of the complexes to FS-DNA was investigated by electronic absorption titration and fluorescence spectroscopy. The results indicate that the complexes bind to FS-DNA in an intercalative mode and the intrinsic binding constants K of the title complexes with FS-DNA are about 3.5?×?104 M?1, 3.9?×?104 M?1, 6.1?×?104 M?1, and 1.4?×?105 M?1, respectively. Also, the four complexes bind to DNA with different binding affinities, in descending order: complex 4, complex 3, complex 2, complex 1. Gel electrophoresis assay demonstrated the ability of the Pt(II) complexes to cleave pBR322 plasmid DNA.  相似文献

Complex formation between Pd(II), Pt(II) and iodide has been studied at 25 °C for an aqueous 1.00 M perchloric acid medium. Measurements of the solubility of PdI2(s) in aqueous mercury(II) perchlorate and of AgI(s) and PdI2(s) in aqueous solutions of Pd2+(aq) and Ag+(aq) gave the solubility product of PdI2(s) as Kso=(7±3) × 10−32 M3, which is much smaller than previous literature values.The stability constants β1=[MI(H2O)3+]/([M(H2O)42+][I]) for the two systems were obtained as the ratio between rate constants for the forward and reverse reactions of (i).
The following values of k1 (s−1 M−1), k−1 (s−1) and β1 (M−1) were obtained at 25 °C: (1.14±0.11) × 106, (0.92±0.18), (12±4) × 105 for MPd, and (7.7±0.4), (8.0±0.7) × 10−5, (9.6±1.3) × 104 for MPt. Combination with previous literature data gives the following values of log(β1 (M−1)) to log(β4 (M−4)): 6.08, ∼22, 25.8 and 28.3 for MPd, and 4.98, ∼25, ∼28, and ∼30 for MPt. The present results show that the large overall stability constants β4 observed for the M2+I systems are most likely due to a very large stability of the second complex MI2(H2O)2, which is probably a cis-isomer. A distinct plateau in the formation curve for mean ligand number 2 is obtained both for MPd and Pt. The other iodo complexes are not especially stable compared to those of chloride and bromide.ΔH (kJ mol−1) and ΔS (JK−1 mol−1) for the forward reaction of (i), MPd, are (17.3±1.7) and (−71±5), and for the reverse reaction of (i) MPd, (45±3) and (−95±6), respectively. The kinetics are compatible with associative activation (Ia). The contribution from bond-breaking in the formation of the transition state seems to be less important for Pd than for Pt.  相似文献

To perform biological evaluations of newly-designed Pt(II) and Pd(II) complexes, the present study was conducted with targeted protein human serum albumin (HSA) and HCT116 cell line as model of human colorectal carcinoma. The binding of Pt(II) and Pd(II) complexes to HSA was analyzed using fluorescence spectroscopy and molecular docking. The thermal stability and alterations in the secondary structure of HSA in the presence of Pt(II) and Pd(II) complexes were investigated using the thermal denaturation method and circular dichroism (CD) spectroscopy. The cytotoxicity of the Pt(II) and Pd(II) complexes was studied against the HCT116 cell line using MTT assay. The binding analysis revealed that the fluorescence findings were well in agreement with docking results such that there is only one binding site for each complex on HSA. Binding constants of 8.7?×?103 M?1, 2.65?×?103 M?1, 0.3?×?103 M?1, and 4.4?×?103 M?1 were determined for Pd(II) and Pt(II) complexes (I–IV) at temperature of 25?°C, respectively. Also, binding constants of 1.9?×?103 M?1, 15.17?×?103 M?1, 1.9?×?103 M?1, and 13.1?×?103 M?1 were determined for Pd(II) and Pt(II) complexes (I–IV) at temperature of 37?°C, respectively. The results of CD and thermal denaturation showed that the molecular structure of HSA affected by interaction with Pt(II) and Pd(II) complexes is stable. Cytotoxicity studies represented the growth suppression effect of the Pt(II) and Pd(II) complexes toward the human colorectal carcinoma cell line. Therefore, the results suggest that the new designed Pt(II) and Pd(II) complexes are well promising candidates for use in cancer treatment, particularly for human colorectal cancer.

A thorough spectral investigation of the copper(II) complex of the antitumor compound, bleomycin, has been carried out in solution employing optical, difference optical, electron spin resonance, and circular dichroism techniques. The optical spectrum of a pH = 7 solution of the 1:1 complex between copper(II) and bleomycin is characterized by a broad weak band in the visible region (λmax = 610 nm) that cannot be resolved and intense ultraviolet bands at 317 (? = 2800), 327 (shoulder), 250 (? = 4700), and 257 nm (shoulder). The circular dichroism spectrum in the visible region shows the broad and weak visible absorption band contains at least three components (558, 675, and 880 nm) that are likely to be “d-d” in origin. The electron spin resonance spectrum is characteristic of a tetragonal d9 copper(II) system showing no rhombic distoritions at X-band frequencies (gx = gy ± 0.002). The spin Hamiltonian parameters for the pH = 7.0 solution corrected for second order effects are A = 177 × 10?4 cm?1, A ? 15 × 10?4 cm?1, g = 2.214, g = 2.039. Most interesting was the observation of extra hyperfine splitting due to endogenous nitrogen coordination in a 30% glycerol glass (AN = 12.0 × 10?4 cm?1). That pattern is best interpreted as a seven-line sequence associated with three liganded nitrogens. A dramatic change in all spectral properties occurs when the pH of the copper(II)-bleomycin complex is lowered to 2.5. All these data taken together suggest a CuN3O coordination complex in solution. Details and justifications as well as a discussion of the limitations of the interpretations are presented.  相似文献

Purified acetylcholinesterase (AChE) of the horn fly was characterized to elucidate the enzymological, inhibitory, and molecular properties of the enzyme. Maximum activity of the AChE against the substrate acetylthiocholine (ATCh) occurred when reactions were conducted at 37°C and pH 7.5. Km and Vmax values were (9.2 ± 0.35) × 10?6 M and 239.8 ± 10.8 units/mg, respectively, for ATCh and (1.5 ± 0.07) × 10?5 M and 138.5 ± 5.5 units/mg, respectively, for butyrylthiocholine (BTCh). The activity of AChE decreased when concentrations of ATCh or BTCh were higher than 1 mM. Studies of the interaction of AChE with different inhibitors revealed pl50 values of 8.88 for eserine, 6.90 for BW284C51, and 4.97 for ethopropazine. Bimolecular reaction constants (kis) for the organophosphorus (OP) anticholinesterases were (2.74 ± 0.14) × 106 M?1 min?1 for coroxon, (7.20 ± 0.28) × 105 M?1 min?1 for paraoxon, and (2.33 ± 0.12) × 105 M?1 min?1 for stirofos. Two major forms of native AChE molecules were found on non-denaturing polyacrylamide gel electrophoresis (PAGE) with Triton X-100, corresponding to bands AChE-2 and AChE-4 found on PAGE without Triton X-100. AChE-2 had an estimated molecular weight of 603,000 and was amphiphilic. AChE-4 had a molecular weight of 147,000 and was hydrophilic. Results of PAGE analyses indicated that the purified enzyme had two bands, one of about 123 kDa and the other greater than 320 kDa, prior to disulfide reduction and only one band at about 54 kDa after reduction on SDS-PAGE. © 1994 Wiley-Liss, Inc.

    The rate constants of the reactions between pulse radiolytically produced superoxide anions and the Cu(II) chelates of salicylate, acetylsalicylate, p-aminosalicylate and diisopropylsalicylate were determined at pH 7.5 and found to range from 0.8 to 2.4 × 109 M?1 sec?1. It was intriguing to note that they had a superoxide dismutase activity identical with that of native cuprein-copper (k245 = 1.3 × 109 M?1 sec?1 per g-atom of Cu). These measurements confirm our earlier observations using indirect assays that all copper salicylates act as perfect model superoxide dismutases and favour the proposal that the activity of anti-inflammatory agents might be assigned to their in vivo formed Cu complexes.  相似文献

    Herein, we evaluate the binding of Pb(II) and Bi(III) to cysteine-substituted versions of the TRI peptides [AcG-(LKALEEK)4G-NH2] which have previously been shown to bind Hg(II) and Cd(II) in unusual geometries as compared with small-molecule thiol ligands in aqueous solutions. Studies of Pb(II) and Bi(III) with the peptides give rise to complexes consistent with the metal ions bound to three sulfur atoms with M–S distances of 2.63 and 2.54 Å, respectively. Competition experiments between the metal ions Pb(II), Cd(II), Hg(II) and Bi(III) for the peptides show that Hg(II) has the highest affinity, owing to the initial formation of the extremely strong HgS2 bond. Cd(II) and Pb(II) have comparable binding affinities at pH > 8, while Bi(III) displays the weakest affinity, following the model, M(II) + (TRI LXC)3 3? → M(II)(TRI LXC)3 ?. While the relevant equilibria for Hg(II) binding to the TRI peptides corresponds to a strong first step forming Hg(TRI LXC)2(HTRI LXC), followed by a single deprotonation to give Hg(TRI LXC)3 ?, the binding of Cd(II) and Pb(II) is consistent with initial formation of M(II)(TRI LXC)(HTRI LXC)2 + at pH < 5 followed by a two-proton dissociation step (pK a2) yielding M(II)(TRI LXC)3 ?. Pb(II)(TRI LXC)(HTRI LXC)2 + converts to Pb(II)(TRI LXC)3 ? at slightly lower pH values than the corresponding Cd(II)–peptide complexes. In addition, Pb(II) displays a lower pK a of binding to the “d”-substituted peptide, (TRI L12C, pK a2 = 12.0) compared with the “a”-substituted peptide, (TRI L16C, pK a2 = 12.6), the reverse of the order seen for Hg(II) and Cd(II). Pb(II) also showed a stronger binding affinity for TRI L12C (K bind = 3.2 × 107 M?1) compared with that with TRI L16C (K bind = 1.2 × 107 M?1) at pH > 8.  相似文献

    S-(p-azidophenacyl)-glutathione, l, is a linear competitive inhibitor at pH 7.40 of beef liver glyoxalase II with Ki = 7.96 × 10?4 M. On irradiation at 340 nm it covalently inhibits glyoxalase II to a level of 42 ± 5% inhibition. This photoaffinity labelling is prevented by the presence of a glyoxalase II competitive inhibitor (the hemimercaptal of glutathione and methylglyoxal).     The reaction of Ru(XTPP)(DMF)2, where XTPP is the dianion of para substituted tetraphenylporphyrins and X is MeO, Me, H, Cl, Br, I, F, with O2 and CO were studied in DMF. The process was found to be first-order in metalloporphyrin, first-order in molecular oxygen and carbon monoxide, and second-order overall. Second-order rate constants for the CO reaction ranged from 0.170 to 0.665 M?1 s?1 at 25°C, those for the O2 reaction from 0.132 to 0.840 M?1 s?1 at 25°C. Similar activation parameters (ΔHCO± = 87 ± 1 kJ mol?1, ΔSCO± = 22 ± 4 JK?1 mol?1; ΔHO2± = 81 ± 1 kJ mol?1, and ΔSO2± = 11 ± 5 JK?1 mol?1) were found within each series. Reactivities of X substituted metalloporphyrins were found to follow different Hammett σ functions. The CO reactions correlated with σ? following normal behavior; the O2 reactions correlated with σ8° indicating O2 is π-bonded in the transition states.     A determination method for Co(II), Fe(II) and Cr(III) ions by luminol‐H2O2 system using chelating reagents is presented. A metal ion‐chelating ligand complex with a Co(II) ion and a chelating reagent like ethylenediaminetetraacetic acid (EDTA) produced highly enhanced chemiluminescence (CL) intensity as well as longer lifetime in the luminol‐H2O2 system compared to metals that exist as free ions. Whereas free Cu(II) and Pb(II) ions had a strong catalytic effect on the luminol‐H2O2 system, significantly, the complexes of Cu(II) and Pb(II) with chelating reagents lost their catalytic activity due to the chelating reagents acting as masking agents. Based on the observed phenomenon, it was possible to determine Co(II), Fe(II) and Cr(III) ions with enhanced sensitivity and selectivity using the chelating reagents of the luminol‐H2O2 system. The effects of ligand, H2O2 concentration, pH, buffer solution and concentrations of chelating reagents on CL intensity of the luminol‐H2O2 system were investigated and optimized for the determination of Co(II), Fe(II) and Cr(III) ions. Under optimized conditions, the calibration curve of metal ions was linear over the range of 2.0 × 10‐8 to 2.0 × 10‐5 M for Co(II), 1.0 × 10‐7 to 2.0 × 10‐5 M for Fe (II) and 2.0 × 10‐7 to 1.0 × 10‐4 M for Cr(III). Limits of detection (3σ/s) were 1.2 × 10‐8, 4.0 × 10‐8 and 1.2 × 10‐7 M for Co(II), Fe(II) and Cr(III), respectively. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

    Two new ruthenium(II) polypyridyl complexes [Ru(dmb)2(HECIP)](ClO4)2 (1) (HECIP = N-ethyl-4-[(1,10)-phenanthroline(5,6-f)imidazol-2-yl]carbazole, dmb = 4,4’-dimethyl-2,2’-bipyridine) and [Ru(dmp)2(HECIP)](ClO4)2 (2) (dmp = 2,9-dimethyl-1,10-phenanthroline) have been synthesized and characterized. The DNA-binding behaviors of the two complexes were investigated by absorption spectra, viscosity measurements, and photoactivated cleavage. The DNA-binding constants for complexes 1 and 2 were determined to be 8.03 (± 0.12) × 104 M?1 (s = 1.62) and 2.97 (± 0.15) × 104 M?1 (s = 1.82), respectively. The results suggest that these complexes interact with DNA through intercalative mode. The photocleavage of pBR322 DNA by Ru(II) complexes was investigated. The cytotoxicity of complexes 1 and 2 has been evaluated by the MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide)] method. Complex 1 shows higher anticancer potency than 2 against the four tumor cell lines. Apoptosis and cellular uptake were investigated. The antioxidant activities of the ligand and these complexes were also performed.  相似文献

    The reactions of copper(II)-ahphatic polyamine complexes with cysteine, cysteine methyl ester, penicillamine. and glutathione have been investigated, with the goal of understanding the relationship between RS?-Cu(II) adduct structure and preferred redox decay pathway. Considerable mechanistic flexibility exists within this class of mercapto ammo acid oxidations, as changes in the rate law could be induced by modest variations in reductant concentration (at fixed [Cu(II)]o), pH, and the structure of the redox partners. With excess cysteine present at 25°C, pH 5 0, I = 0 2 M (NaOAc), decay of 1:1 cys-S?-Cu(II) transient adducts was found to be first order in both cys-SH and transient. Second-order rate constants characteristic of Cu(dien)2+ (6 1 × 103M?1sec?1), Cu(Me5dien)2+ (2.7 × 103M?1 sec?1), Cu(en)22+ (2.1 × 103M?1 sec?1), and Cu(dien)22+ (4.7 × 103 M?1 sec ?1) are remarkably similar, considering substantial differences in the composition and geometry of the oxidant first coordination sphere. A mechanism involving attack of cysteine on the coordinated sulfur atom of the transient, giving a disulfide anion radical intermediate, is proposed to account for these results Moderate reactivity decreases in the cysteine-Cu(dien)2+, Cu(Me5dien)2+ reactions with increasing [H+] (pH 4–6) reflect partial protonation of the polyamine ligands. A very different rate law, second order in the RS?-Cu(II) transient and approximately zeroth order in mercaptan, applies in the pH 5.0 oxidations of cysteine methyl ester, penicillamine, and glutathione by Cu(dien)2+ and Cu(Me5dien)2+. This behavior suggests the mtermediacy of di-μ-mercapto-bridged binuclear Cu(II) species, in which a concerted two-electron change yields the disulfide and Cu(I) products. Similar hydroxo-bridged intermediates are proposed to account for the transition from first- to second-order transient dependence in cysteine oxidations by Cu(dien)2+ and Cu(Me5dien)2+ as the pH is increased from 5 to 7. Yet another rate law, second order in transient and first order in cysteine, applies in the pH 5.0 oxidation of cysteine by Cu(Me6tren)2+ (k(25°C) 7.5 × 107 M?2 sec?1, I = 0.2 M). Steric rigidity of this trigonal bipyramidal oxidant evidently protects the coordinated sulfur atom from attack in a RSSR?-forming pathway. Formation of a coordinated disulfide in the rate-determining step is purposed, coupled with attack of a noncoordinated cysteine molecule on a vacated coordination position to stabilize the (Me6(tren)Cu(I) product.  相似文献

    Removal of Pb(II) from an aqueous environment using biosorbents is a cost-effective and environmentally benign method. The biosorption process, however, is little understood for biosorbents prepared from plant materials. In this study, the biosorption process was investigated by evaluating four adsorption models. A fixed-bed column was prepared using a biosorbent prepared from the aquatic plant Hydrilla verticillata. The effect of bed height and flow rate on the biosorption process was investigated. The objective of the study was to determine the ability of H. verticillata to biosorb Pb(II) from an aqueous environment and to understand the process, through modeling, to provide a basis to develop a practical biosorbent column. Experimental breakthrough curves for biosorption of 50 mg L?1 aqueous Pb(II) using a fixed-bed column with 1.00 cm inner diameter were fitted to the Thomas, Adams-Bohart, Belter, and bed depth service time (BDST) models to investigate the behavior of each model according to the adsorption system and thus understand the adsorption mechanism. Model parameters were evaluated using linear and nonlinear regression methods. The biosorbent removed 65% (82.39 mg g?1 of biosorbent) of Pb(II) from an aqueous solution of Pb(NO3)2 at a flow rate of 5.0 ml min?1 in a 10 cm column. Na2CO3 was used to recover the adsorbed Pb(II) ions as PbCO3 from the biosorbent. The Pb(II) was completely desorbed at a bed height of 10.0 cm and a flow rate of 5.0 ml min?1. Fourier transform infrared (FT-IR) analysis of the native biosorbent and Pb(II)-loaded biosorbent indicated that the hydroxyl groups and carboxylic acid groups were involved in the metal bonding process. The FT-IR spectrum of Pb(II)-desorbed biosorbent showed an intermediate peak shift, indicating that Pb(II) ions were replaced by Na+ ions through an ion-exchange process. Of the four models tested, the Thomas and BDST models showed good agreement with experimental data. The calculated bed sorption capacity N0 and rate constant ka were 31.7 g L?1 and 13.6 × 10?4 L mg?1 min?1 for the Ct/C0 value of 0.02. The BDST model can be used to estimate the column parameters to design a large-scale column.  相似文献

    Pholcodine is an opiate derivative drug which is widely used in pediatric medicine. In this study, a chemiluminescence (CL) method is described that determines pholcodine in human plasma and syrup samples. This method is based on the fact that pholcodine can greatly enhance the weak CL emission of reaction between tris(1,10 phenanthroline)ruthenium(II), Ru(phen)32+, and acidic Ce(IV). The CL mechanism is described in detail using UV–vis light, fluorescence and CL spectra. Effects of chemical variables were investigated and under optimum conditions, CL intensity was proportional to the pholcodine concentration over the range 4.0 × 10?8 to 8.0 × 10?6 mol  L?1. The limit of detection (LOD) (S/N = 3) was 2.5 × 10?8 mol  L?1. Percent of relative standard deviations (%RSD) for 3.0 × 10?7 and 3.0 × 10?6 mol  L?1 of pholcodine was 2.9 and 4.0%, respectively. Effects of common ingredients were investigated and the method was applied successfully to the determination of pholcodine in syrup samples and human plasma.  相似文献

    Kinetic studies of the reduction of ferrioxamine B (Fe(Hdesf)+) by Cr(H2O)62+, V(H2O)62+, and dithionite have been performed. For Cr(H2O)62+ and V(H2O)62+, the rate is ?d[Fe(Hdesf)+]/dt = k[Fe(Hdesf)+][M2+]. For Cr(H2O)62+, k = 1.19 × 104 M?1 sec?1 at 25°C and μ = 0.4 M, and k is independent of pH from 2.6 to 3.5. For V(H2O)62+, k = 6.30 × 102 M?1 sec?1 at 25°C, μ = 1.0 M, and pH = 2.2. The rate is nearly independent of pH from 2.2 to 4.0. For Cr(H2O)62+ and V(H2O)62+, the activation parameters are ΔH = 8.2 kcal mol?1, ΔS ?12 eu and ΔH = 1.7 kcal mol?1, ΔS = ?40 eu (at pH 2.2) respectively. Reduction by Cr(H2O)62+ is inner-sphere, while reduction by V(H2O)62+ is outer-sphere. Reduction by dithionite follows the rate law ?d[Fe(Hdesf)+]/dt =kK12[Fe(Hdesf)+][S2O42?]12 where K is the equilibrium constant for dissociation of S2O42? into SO2? radicals. The value of k at 25°C and μ = 0.5 is 2.7 × 103 M?1 sec?1 at pH 5.8, 3.5 × 103 M?1 sec?1 at pH 6.8, and 4.6 × 103 M?1 sec?1 at pH 7.8, and ΔH = 6.8 kcal mol?1 and ΔS = ?19 eu at pH 7.8.  相似文献

    The interactions between cytochrome P450 2B4 (d-2B4), NADPH:cytochrome P450 reductase and cytochrome b5 have been investigated in the monomeric reconstituted P450 2B4-containing monooxygenase system in the presence of a substrate (7-pentoxyresorufin) and an electron donor, NADPH. Each partner was immobilized via its amino groups on the carboxymethyldextran biochip surface of the optical biosensor IAsys+. Such mode immobilization was not accompanied by any loss of activities of the immobilized proteins. The formation of binary d-Fp/d-2B4 complexes was registered. The association/dissociation rate constants (kon/koff) were (0.013 ± 0.005) × 106 M?1 s?1/0.05 ± 0.02 s?1, and dissociation constant (KD) was (0.26 ± 0.13) × 10?6 M. Comparison of kon, koff and KD values for d-Fp/d-2B4 complexes formed under hydroxylation (O-dealkylation) with corresponding constants obtained for the oxidized proteins of (0.10 ± 0.03) × 106 M?1 s?1/(0.14 ± 0.06) s?1, and (0.71 ± 0.37) × 10?6 M, respectively shows that the decrease in kon and an insignificant decrease in KD are associated with the increase of complex lifetime during transition from the oxidized to hydroxylation conditions. Complex formation between d-Fp and d-b5 was not registered in both hydroxylation conditions and in the case of oxidized forms of these proteins. In both cases formation of the ternary d-Fp/d-2B4/d-b5 complexes occurred.  相似文献
