As part of the development of a method to control the outbreak and persistence of red tides using mass-cultured heterotrophic protist grazers, we measured the growth and ingestion rates of cultured Oxyrrhis marina (a heterotrophic dinoflagellate) on cultured Heterosigma akashiwo (a raphidophyte) in bottles in the laboratory and in mesocosms (ca. 60 liter) in nature, and those of the cultured grazer on natural populations of the red-tide organism in mesocosms set up in nature. In the bottle incubation, specific growth rates of O. marina increased rapidly with increasing concentration of cultured prey up to ca. 950 ng C ml(-1) (equivalent to 9,500 cells ml(-1)), but were saturated at higher concentrations. Maximum specific growth rate (mumax), KGR (prey concentration sustaining 0.5 mumax) and threshold prey concentration of O. marina on H. akashiwo were 1.43 d(-1), 104 ng C ml(-1), and 8.0 ng C ml(-1), respectively. Maximum ingestion and clearance rates of O. marina were 1.27 ng C grazer(-1) d(-1) and 0.3 microl grazer(-1) h(-1), respectively. Cultured O. marina grew well effectively reducing cultured and natural populations of H. akashiwo down to a very low concentration within 3 d in the mesocosms. The growth and ingestion rates of cultured O. marina on natural populations of H. akashiwo in the mesocosms were 39% and 40%, respectively, of those calculated based on the results from the bottle incubation in the laboratory, while growth and ingestion rates of cultured O. marina on cultured H. akashiwo in the mesocosms were 55% and 36%, respectively. Calculated grazing impact by O. marina on natural populations of H. akashiwo suggests that O. marina cultured on a large scale could be used for controlling red tides by H. akashiwo near aquaculture farms that are located in small ponds, lagoons, semi-enclosed bays, and large land-aqua tanks to which fresh seawater should be frequently supplied.  相似文献

Red tide phytoplankton Chattonella marina is known to produce reactive oxygen species (ROS), such as superoxide anion (O(2)(-)), hydrogen peroxide (H(2)O(2)) and hydroxyl radical (&z.rad;OH), under normal physiological conditions. Although several lines of evidence suggest that ROS are involved in the mortality of fish exposed to C. marina, the mechanism of ROS generation in C. marina remains to be clarified. In this study, we found that the cell-free supernatant prepared from C. marina cells showed NAD(P)H-dependent O(2)(-) generation, and this response was inhibited by diphenyleneiodonium, an inhibitor of mammalian NADPH oxidase. When the cell-free supernatant of C. marina was analyzed by immunoblotting using antibody raised against the human neutrophil cytochrome b558 large subunit (gp91phox), a main band of approximately 110 kDa was detected. The cell surface localization of the epitope recognized with this antibody was also demonstrated in C. marina by indirect immunofluorescence. Furthermore, Southern blot analysis performed on genomic DNA of C. marina with a probe covering the C-terminal region of gp91phox suggested the presence of a single-copy gene coding for gp91phox homologous protein in C. marina. These results provide evidence for the involvement of an enzymatic system analogous to the neutrophil NADPH oxidase as a source of O(2)(-) production in C. marina.  相似文献

Cochlodinium polykrikoides, a harmful red tide dinoflagellate, is highly toxic to fish, but the toxic mechanism is still unknown. Recent study has suggested that C. polykrikoides generates reactive oxygen species (ROS) such as superoxide anion (O(2)(-)) and hydrogen peroxide (H(2)O(2)), and the ROS-mediated ichthyotoxicity has been proposed. In this study, we found that the levels of O(2)(-) and H(2)O(2) detected in C. polykrikoides were trace levels as compared with those of Chattonella marina which is well-known to produce ROS. Furthermore, no significant increase in O(2)(-) generation by C. polykrikoides was observed in the presence of lectins such as concanavalin A (Con A) and wheat germ agglutinin (WGA) or fish mucus prepared from skin and gill of yellowtail, whereas C. marina generated increased level of O(2)(-) responding to these stimuli. Interestingly, the cell-free aqueous extract prepared from C. polykrikoides showed toxic effect on the HeLa cells, but the extract of C. marina had no significant effect. Furthermore, gradual accumulation of polysaccharides in the medium was observed during the growth of C. polykrikoides, and the medium gradually became viscous, but no such changes were observed in the medium of C. marina. These results suggest that multiple factors may be responsible for the toxic mechanism of C. polykrikoides.  相似文献

The cyanobacterium Acaryochloris marina is the only known phototroph harboring chlorophyll (Chl) d. It is easy to cultivate it in a planktonic growth mode, and A. marina cultures have been subject to detailed biochemical and biophysical characterization. In natural situations, A. marina is mainly found associated with surfaces, but this growth mode has not been studied yet. Here, we show that the A. marina type strain MBIC11017 inoculated into alginate beads forms dense biofilm-like cell clusters, as in natural A. marina biofilms, characterized by strong O(2) concentration gradients that change with irradiance. Biofilm growth under both visible radiation (VIS, 400 to 700 nm) and near-infrared radiation (NIR, ~700 to 730 nm) yielded maximal cell-specific growth rates of 0.38 per day and 0.64 per day, respectively. The population doubling times were 1.09 and 1.82 days for NIR and visible light, respectively. The photosynthesis versus irradiance curves showed saturation at a photon irradiance of E(k) (saturating irradiance) >250 μmol photons m(-2) s(-1) for blue light but no clear saturation at 365 μmol photons m(-2) s(-1) for NIR. The maximal gross photosynthesis rates in the aggregates were ~1,272 μmol O(2) mg Chl d(-1) h(-1) (NIR) and ~1,128 μmol O(2) mg Chl d(-1) h(-1) (VIS). The photosynthetic efficiency (α) values were higher in NIR-irradiated cells [(268 ± 0.29) × 10(-6) m(2) mg Chl d(-1) (mean ± standard deviation)] than under blue light [(231 ± 0.22) × 10(-6) m(2) mg Chl d(-1)]. A. marina is well adapted to a biofilm growth mode under both visible and NIR irradiance and under O(2) conditions ranging from anoxia to hyperoxia, explaining its presence in natural niches with similar environmental conditions.  相似文献

The digestive process of Oxyrrhis marina Dujardin feeding on Dunaliella primolecta Butcher commences with the capture of the phototroph, perhaps involving trichocysts which restrain the prey, followed by engulfing by a membrane that appears to originate from near the fiagella pit of the O. marina. The prey, still within the capture membrane, enters the O. marina and is digested over a period estimated to be of the order of 12-24h. During digestion the prey first becomes more electron dense and the contents coagulate. Later the prey becomes less distinct and disintegrates; by this stage remains from several prey may be enclosed within one vacuole. Small O. marina , captured by large prey-depleted O. marina , suffer the same fate. Absence of any correlation between changes in intracellular amino acids and digestion of prey probably resulted from each O. marina containing several prey at various stages of digestion. However, the value of the glutamine/glutamate (Gln/Glu) ratio remained at around 0-3, whilst the population changed from one dominated by N-replete D. primolecta to one dominated by prey-replete O. marina , N-deplete D. primolecta and prey-deplete O. marina both have Gln/Glu of < 0.1. The Gln/Glu ratio may therefore be of use in assessing the nutrient status of heterotrophic microflagellates.  相似文献

BACKGROUND: and Aims Submerged plants possess well-developed aerenchyma facilitating intra-plant gas-phase diffusion of O2 to below-ground tissues, which are usually buried in anoxic sediments. However, aquatic habitats differ in terms of O2 fluctuations in the water column and in O2 consumption of the sediment, and aquatic plants differ in aerenchymal volume and resistance to O2 diffusion through the plant and across leaf and root surfaces. The hypothesis that the freshwater isoetid Lobelia dortmanna and the marine seagrass Zostera marina should display pronounced contrasts in intra-plant O2 dynamics because of differences in morphology/anatomy, physiology and growth habitat was tested. METHODS: In order to determine the O2 dynamics and relate this to the anatomy and morphology of the two species, O2 microelectrodes were inserted in the aerenchyma of leaves and roots, the sediment pore-water, and the water column in the field. Manipulation of water column O2 in the laboratory was also carried out. KEY RESULTS: It was found that intra-plant transport of O2 between leaf and root tips takes place more readily in L. dortmanna than in Z. marina due to shorter distances and greater cross-sections of the aerenchyma. The major exchange of O2 across roots of L. dortmanna can be accounted for by small intra-plant resistances to diffusion, larger root than leaf surfaces, and greater radial diffusive resistance of leaves than roots. In contrast, the major O2 exchange across leaves than roots of Z. marina can be accounted for by the opposite anatomical-morphological features. The larger aerenchymal volume and the smaller metabolic rates of L. dortmanna compared to Z. marina imply that turnover of O2 is slower in the aerenchyma of L. dortmanna and O2 fluctuations are more dampened following changes in irradiance. Also, O2 accumulated in the aerenchyma can theoretically support dark respiration for a few hours in L. dortmanna but for only a few minutes in Z. marina. CONCLUSIONS: The build-up of O2 in the pore-water of L. dortmanna sediments during the day as a result of high release of photosynthetic O2 from roots and low O2 consumption of sediments means that sediment, aerenchyma and water are important O2 sources for respiration during the following night, while Z. marina relies on the water column as the sole source of O2 because its sediments are anoxic. These differences between L. dortmanna and Z. marina appear to represent a general difference between the isoetid species mainly inhabiting sediments of low reducing capacity of oligotrophic lakes and the elodeid freshwater species and marine seagrasses mainly inhabiting sediments of higher reducing capacity in more nutrient-rich habitats.  相似文献

F-actin organization during the cell cycle was investigated in two stramenopile microalgae, Ochromonas danica (Chrysophyceae; UTEX LB1298) and Heterosigma akashiwo (Raphidophyceae; NIES-6) using FITC-phalloidin. In the interphase cell of O. danica, F-actin bundles were localized forming a network structure in the cortical region, which converged from the anterior region to the posterior, whereas in the interphase cell of H. akashiwo, F-actin bundles were observed forming a network structure in the cortical region without any polarity. In both O. danica and H. akashiwo, at the initial stage of mitosis the cortical F-actin disappeared, and then during cytokinesis assembly of an actin-based ring-like structure occurred in the cell cortex in the plane of cytokinesis. The ring-like structure initiated from aster-like structures was composed of F-actin in both O. danica and H. akashiwo. Different from animal cells, later stages of cytokinesis of O. danica seemed to be promoted by microtubules, although the early stages of cytokinesis progressed with a constriction of the ring-like structure, whereas cytokinesis of H. akashiwo was apparently completed by constriction of the cell mediated by the F-actin ring, as in animal cells.  相似文献

Photoresponse in the heterotrophic marine dinoflagellate Oxyrrhis marina   总被引:1,自引:0,他引:1  
Expressed rhodopsins were detected by proteomic analysis in an investigation of potential signal receptors in the cell membrane of the marine heterotrophic dinoflagellate Oxyrrhis marina (CCMP604). We inferred these to be sensory rhodopsins, a type of G-protein-coupled receptor trans-membrane signaling molecule. Because phototactic behavior based on sensory rhodopsins has been reported in other protists, we investigated the photosensory response of O. marina. This dinoflagellate exhibited strongest positive phototaxis at low levels (2-3 μE/m(2)/s) of white light when the cells were previously light adapted and well fed. Positive phototaxis was also found for blue (450 nm), green (525 nm), and red (680 nm) wavelengths. In a further test, O. marina showed significantly greater phototaxis toward concentrated algal food illuminated by blue light to stimulate red chlorophyll-a autofluorescence in the prey, compared with using bleached algae as prey. Concentration of a cytoplasmic downstream messenger molecule, cyclic adenosine monophosphate, a component of the signaling pathway of G-protein-coupled receptor molecules, rapidly increased in O. marina cells after exposure to white light. In addition, treatment with hydroxylamine, a rhodopsin signaling inhibitor, significantly decreased their phototactic response. Our results demonstrate that a heterotrophic marine dinoflagellate can orient to light based on rhodopsins present in the outer cell membrane and may be able to use photosensory response to detect algal prey based on chlorophyll autofluorescence.  相似文献

Chattonella marina and Heterosigma akashiwo, known as red tide phytoplankton, are naturally wall-less and have quite fragile cell structures. In this study, we found that an equilibrium dialysis technique allowed the study of lectin binding to these flagellates. The results suggested that concanavalin A (Con A) binds to these flagellate cells through the specific carbohydrate moieties on the cell surface. Interestingly, the binding of an excess of Con A on the cell surface caused morphological changes concomitant with discharge of glycocalyx, a polysaccharide-containing common structure on the external cell surface of these flagellates. Fluorescent microscopic observation using FITC-labeled Con A (F-Con A) confirmed that F-Con A molecules are localized on the discharged glycocalyx.  相似文献

In the phylogenetic tree, selenoproteins and the corresponding translation machinery are found in Archaea, Eubacteria, and animals, but not in fungi and higher plants. As very little is known about Protozoa, we searched for the presence of selenoproteins in the primitive dinoflagellate Oxyrrhis marina, belonging to the Protoctista kingdom. Four selenoproteins could be obtained from O. marina cells cultured in the presence of 75Se. Using O. marina or bovine liver cytosolic extracts, we could serylate and selenylate in vitro total O. marina tRNAs. Moreover, the existence of a tRNA(Sec) could be deduced from in vivo experiments. Lastly, an anti-serum against the specialized mammalian translation elongation factor mSelB reacted with a protein of 48-kDa molecular mass. Altogether, our data showed that O. marina contains selenoproteins and suggests that the corresponding translation machinery is related to that found in animals.  相似文献

Recently, harmful algal bloom (HAB), also termed "red tide", has been recognized as a serious problem in marine environments according to climate changes worldwide. Many novel materials or methods to prevent HAB have not yet been employed except for clay dispersion, in which can the resulting sedimentation on the seafloor can also cause alteration in marine ecology or secondary environmental pollution. In the current study, we investigated that antimicrobial peptide have a potential in controlling HAB without cytotoxicity to harmless marine organisms. Here, antimicrobial peptides are proposed as new algicidal compounds in combating HAB cells. HPA3 and HPA3NT3 peptides which exert potent antimicrobial activity via pore forming action in plasma membrane showed that HPA3NT3 reduced the motility of algal cells, disrupted their plasma membrane, and induced the efflux of intracellular components. Against raphidoflagellate such as Heterosigma akashiwo, Chattonella sp., and C. marina, it displayed a rapid lysing action in cell membranes at 1~4 μM within 2 min. Comparatively, its lysing effects occurred at 8 μM within 1 h in dinoflagellate such as Cochlodium polykrikoides, Prorocentrum micans, and P. minimum. Moreover, its lysing action induced the lysis of chloroplasts and loss of chlorophyll a. In the contrary, this peptide was not effective against Skeletonema costatum, harmless algal cell, even at 256 μM, moreover, it killed only H. akashiwo or C. marina in co-cultivation with S. costatum, indicating to its selective algicidal activity between harmful and harmless algal cells. The peptide was non-hemolytic against red blood cells of Sebastes schlegeli, the black rockfish, at 120 μM. HAB cells were quickly and selectively lysed following treatment of antimicrobial peptides without cytotoxicity to harmless marine organisms. Thus, the antibiotic peptides examined in our study appear to have much potential in effectively controlling HAB with minimal impact on marine ecology.  相似文献

Oxyrrhis marina, a widely distributed marine protist, is used to model heterotrophic flagellate responses in microbial food webs. Although clonal variability occurs in protists, assessments of intraspecific diversity are rare; such assessments are critical, particularly where species are used as models in ecological studies. To address the extent of intraspecific variation within O. marina, we assessed diversity among 11 strains using 5.8S rDNA and ITS sequences. The 5.8S rDNA and ITS regions revealed high divergence between strains: 63.1% between the most diverse. To compare O. marina diversity relative to other alveolates, 18S rDNA sequences for five strains were analysed with sequences from representatives of the major alveolate groups. 18S rDNA also revealed high divergence in O. marina. Additionally, consistent with phylogenies based on protein coding genes, maximum likelihood analysis indicated that O. marina was monophyletic and ancestral to the dinoflagellates. To assess ecophysiological differences, growth rates of seven O. marina strains were measured at 10 salinities (10-55 per thousand). Two salinity responses occurred: one group achieved highest growth rates at high salinities; the other grew best at low salinities. There was no clear correlation between molecular, ecophysiological, or geographical differences. However, salinity tolerance was associated with habitat type: intertidal strains grew best at high salinities; open-water strains grew best at low salinities. These data indicate the need to examine many strains of a species in both phylogenetic and ecological studies, especially where key-species are used to model ecological processes.  相似文献

Chattonella marina (C. marina), a raphidophycean flagellate, is a causative organism of red tide, and highly toxic to fish. In this study, we found that the cell-free methanol extract prepared from this flagellate exhibited potent hemolytic activity against rabbit erythrocytes. Interestingly, the hemolytic activity of the extract was absolutely light-dependent, and no hemolytic activity was detected in the dark even at very high concentration. Gel filtration chromatography of the methanol extract on a column of Sephadex LH-20 revealed that the extract contained hemagglutinin as well as hemolytic agents, and the substances responsible for these activities were separately eluted. These results suggest that the hemagglutinating and hemolytic activities were derived from distinct compounds. The hemolytic fraction obtained after gel filtration (F4) caused marked inhibition of the growth of C. marina itself and other species of phytoplanktons. Furthermore, F4 showed a potent cytotoxicity toward various mammalian cultured cell lines including human tumor cells (HeLa cells) in a dose-dependent manner. The cytotoxicity was also light-dependent, and no cytotoxic effect was exhibited in any cell lines tested in the dark. After further purification procedures via preparative thin-layer chromatography and subsequent HPLC, a major hemolytic agent was obtained as highly purified form. Since the methanol extracts prepared from other raphidophycean flagellates such as Heterosigma akashiwo, Olisthodiscus luteus, and Fibrocapsa japonica showed light-dependent hemolytic activity toward rabbit erythrocytes, it was suggested that the light-dependent hemolytic agents commonly exist at least in these raphidophycean flagellates.  相似文献

Heterocapsa circularisquama (Dinophyceae), a noxious red tide dinoflagellate, is known to have a specifically lethal effect on shellfish, especially bivalves such as pearl oyster (Pinctada fucata), but no detrimental effects of this alga on fishes have not been observed so far. In this study, we found that H. circularisquama was toxic to a microzooplankton, a rotifer (Brachionus plicatilis) in a cell concentration-dependent manner, while the cultured supernatant or ultrasonic ruptured H. circularisquama had no significant toxic effect on the rotifer. Since no such toxic effects on the rotifer were observed in Chattonella marina, Heterosigma akashiwo, or Cochlodinium polykrikoides, other species of harmful red tide plankton, H. circularisquama may have a strictly specific toxic mechanism against the rotifer as well as bivalves.  相似文献

通过共培养方法,以细胞密度为主要测定指标,研究了小珊瑚藻对赤潮异弯藻的克生效应及其对UV-B辐射增强的响应.结果表明:小珊瑚藻的新鲜组织和水溶性抽提液对赤潮异弯藻的生长表现出显著的抑制作用(P<0.05),说明小珊瑚藻对赤潮异弯藻具有克生效应;而小珊瑚藻干粉末和培养水过滤液对赤潮异弯藻生长则无明显影响(P>0.05).将小珊瑚藻用不同剂量的UV-B辐射预处理后,再与赤潮异弯藻共培养,除培养水过滤液外,其新鲜组织、干粉末和水溶性抽提液对赤潮异弯藻生长的作用效果均有所改变,高剂量(3.0 J·m-2)UV-B辐射使小珊瑚藻对赤潮异弯藻的抑制作用减弱(P<0.05),而低剂量(0.9 J·m-2) UV-B辐射则使抑制作用增强(P<0.05).  相似文献

研究了N、P营养浓度对赤潮异弯藻(Heterosigma akashiwo)生长的影响.结果表明,该藻的生长速率与N、P营养因子浓度的关系符合Monod公式.在NO3--N浓度达到7.5 mg·L-1时,赤潮异弯藻开始生长;浓度为3.75~75 mg·L-1时,赤潮异弯藻的比生长速率与NO3--N浓度成正比关系.N营养充足时,赤潮异弯藻的最大生长速率μm-n=0.3475·d-1,Ks-n=18.91 mg·L-1.PO4--P浓度为0~1.0 mg·L-1时,赤潮异弯藻的比生长速率与P浓度成正比关系;P营养充足时,赤潮异弯藻的最大生长速率μm-p=0.3024·d-1,Ks-p=0.4086 mg·L-1.N/P达到25后藻细胞浓度达到最大,表明N/P为25时最适合赤潮异弯藻生长.赤潮异弯藻最适合在N 37.5~225.0 mg·L-1、P 5.0~50.0 mg·L-1、N/P=25条件下生长.  相似文献

Free-living marine protists are often assumed to be broadly distributed and genetically homogeneous on large spatial scales. However, an increasing application of highly polymorphic genetic markers (e.g., microsatellites) has provided evidence for high genetic diversity and population structuring on small spatial scales in many free-living protists. Here we characterise a panel of new microsatellite markers for the common marine flagellate Oxyrrhis marina. Nine microsatellite loci were used to assess genotypic diversity at two spatial scales by genotyping 200 isolates of O. marina from 6 broad geographic regions around Great Britain and Ireland; in one region, a single 2 km shore line was sampled intensively to assess fine-scale genetic diversity. Microsatellite loci resolved between 1-6 and 7-23 distinct alleles per region in the least and most variable loci respectively, with corresponding variation in expected heterozygosities (H(e)) of 0.00-0.30 and 0.81-0.93. Across the dataset, genotypic diversity was high with 183 genotypes detected from 200 isolates. Bayesian analysis of population structure supported two model populations. One population was distributed across all sampled regions; the other was confined to the intensively sampled shore, and thus two distinct populations co-occurred at this site. Whilst model-based analysis inferred a single UK-wide population, pairwise regional F(ST) values indicated weak to moderate population sub-division (0.01-0.12), but no clear correlation between spatial and genetic distance was evident. Data presented in this study highlight extensive genetic diversity for O. marina; however, it remains a substantial challenge to uncover the mechanisms that drive genetic diversity in free-living microorganisms.  相似文献

利用测定净光合放氧速率的方法研究了光照强度、温度、pH、盐度对小球藻(Chlorella sp.XQ-200419)和海洋小球藻(Chlorella marina NJ-016)光合作用的影响。小球藻(Chlorella sp.XQ-200419)的适宜光照强度范围为100~1600μmo·lm-2·s-1,光饱和点在500μmo·lm-2·s-1附近;适宜温度范围为25~42.5℃,最适温度为37.5℃;适宜pH值范围为6.5~9.0,最适pH值为7.0;对盐度的适应范围较广,在0~0.6mol/L范围内,随着盐度的升高,净光合放氧速率有下降趋势。海洋小球藻(Chlorella marina NJ-016)的适宜光照强度范围为400~1600μmol·m-2·s-1,光饱和点在1400μmo·lm-2.s-1附近;适宜温度范围为25~42.5℃,最适温度为37.5℃;适宜pH值范围为5.0~9.0,最适pH值为8.0;对盐度有很好的适应性,在0~0.6mol/L范围内,随着盐度升高,净光合放氧速率明显上升。小球藻和海洋小球藻的净光合放氧速率随光照强度、温度、pH值和盐度变化的规律,表明了两种小球藻的基本生理生态学特性:能适应较强的光照强度、较高的温度、中性偏碱的环境和较高的盐度。研究结果有助于小球藻培养条件的优化。两种小球藻对光照强度、温度、pH值和盐度变化的反应也有所不同:与小球藻(Chlorella sp.XQ-200419)相比,海洋小球藻(Chlorella marina NJ-016)对光照强度有更好的适应性,对pH值变化有更宽的适应范围,适当提高盐度对其光合作用有明显的促进作用。这表明海洋小球藻(Chlorella marina NJ-016)在快速生长繁殖方面具有更大的潜力,这一研究结果为筛选适合于大量培养的优良藻种提供了依据。  相似文献

We studied the charge recombination characteristics of Photosystem II (PSII) redox components in whole cells of the chlorophyll (Chl) d-dominated cyanobacterium, Acaryochloris marina, by flash-induced chlorophyll fluorescence and thermoluminescence measurements. Flash-induced chlorophyll fluorescence decay was retarded in the mus and ms time ranges and accelerated in the s time range in Acaryochloris marina relative to that in the Chl a-containing cyanobacterium, Synechocystis PCC 6803. In the presence of 3-(3,4-dichlorophenyl)-1, 1-dimethylurea, which blocks the Q(B) site, the relaxation of fluorescence decay arising from S(2)Q(A)(-) recombination was somewhat faster in Acaryochloris marina than in Synechocystis PCC 6803. Thermoluminescence intensity of the so called B band, arising from the recombination of the S(2)Q(B)(-) charge separated state, was enhanced significantly (2.5 fold) on the basis of equal amounts of PSII in Acaryochloris marina as compared with Synechocystis 6803. Our data show that the energetics of charge recombination is modified in Acaryochloris marina leading to a approximately 15 meV decrease of the free energy gap between the Q(A) and Q(B) acceptors. In addition, the total free energy gap between the ground state and the excited state of the reaction center chlorophyll is at least approximately 25-30 meV smaller in Acaryochloris marina, suggesting that the primary donor species cannot consist entirely of Chl a in Acaryochloris marina, and there is a contribution from Chl d as well.  相似文献

Photosystem II (PSII) electron transfer (ET) in the chlorophyll d-containing cyanobacterium Acaryochloris marina (A. marina) was studied by time-resolved electron paramagnetic resonance (EPR) spectroscopy at room temperature, chlorophyll fluorescence, and low-temperature optical spectroscopy. To maximize the ability to measure PSII ET in the intact cells of this organism, growth conditions were optimized to provide the highest specific O(2) activity and the instrumental parameters for the EPR measurements of tyrosine Z (Y(Z)) reduction were adjusted to give the best signal-to-noise over spectral resolution. Analysis of the Y(Z)(*) reduction kinetics revealed that ET to the oxygen-evolving complex on the donor side of PSII in A. marina is indistinguishable from that in higher plants and other cyanobacteria. Likewise, the charge recombination kinetics between the first plastoquinone acceptor Q(A) and the donor side of PSII monitored by the chlorophyll fluorescence decay on the seconds time scale are not significantly different between A. marina and non-chlorophyll d organisms, while low-temperature optical absorption spectroscopy identified the primary electron acceptor in A. marina as pheophytin a. The results indicate that, if the PSII primary electron donor in A. marina is made up of chlorophyll d instead of chlorophyll a, then there must be very different interactions with the protein environment to account for the ET properties, which are similar to higher plants and other cyanobacteria. Nevertheless, the water oxidation mechanism in A. marina is kinetically unaltered.  相似文献
