The site of inhibition of cotton-seed germination by lucernesaponins seems to be associated with tissues other than theembryo. The rate of water uptake during imbibition was similarfor intact seeds immersed in water and for those in 0.5 percent saponin solution. Seeds previously immersed in saponinsolution showed a lower rate of respiration, absorbing about60 per cent less oxygen than those pre-immersed in water. Yetthe respiration rate of excised embryos from both treatmentswas the same. The extent of the inhibition of germination wasnot affected when oxygen was either bubbled through the saponin-containingimmersion-solution or passed through the flasks in which theseeds were germinated after 24 h of pre-immersion. The rates of oxygen diffusion through the seed coats and membranesof seeds pre-immersed in water or in saponin solution were measured.They were distinctly lower for seed coats and membranes whichhad been exposed to the saponin treatment. It has been concludedthat the seed coat and/or the membrane of intact cotton seedspre-immersed in saponin solution serve as a barrier for oxygendiffusion to the embryo, resulting in inhibition of germination.  相似文献

Either d- or l-leucine (10(-3)m) and unsaturated long-chain fatty acids such as oleic, linoleic, and arachidonic (10(-4)m) significantly stimulated macroconidia germination of Microsporum gypseum. Saturated long-chain fatty acids did not affect germination, whereas saturated short-chain fatty acids such as caprylic, hexanoic, and butyric were completely inhibitory. Germination was followed by an increase in endogenous respiration and a decrease in dry weight of approximately 5% at 4 hr. Endogenous fatty acids and soluble carbohydrates were utilized significantly during germination. Tritiated leucine, uridine, and thymidine were incorporated respectively into protein, ribonucleic acid (RNA), and deoxyribonucleic acid (DNA) fractions within the first 5 min of germination. Incorporation of oleic-1-C(14) into RNA and protein was significantly increased after germ tube development. Net synthesis of RNA and protein started prior to germ tube protrusion. Increase in DNA could be detected only later. A significant increase in RNA and protein during the 4th hr of germination was correlated with vegetative development. Inhibition of respiration and incorporation of leucine-H(3) and uridine-H(3) into corresponding macromolecules by dl-fluorophenylalanine and phenethyl alcohol started before germ tube appearance. Griseofulvin significantly inhibited incorporation of uridine-H(3) and thymidine-H(3), but not of leucine-H(3). This inhibition occurred only after initial vegetative development. In contrast to the two other inhibitors, which substantially inhibited germination, griseofulvin only slightly retarded the period of germination and did not affect respiration.  相似文献

Azotobacter vinelandii cysts undergo conversion to vegetative cells in Burk's nitrogen-free medium utilizing glucose, sucrose, or acetate. In 1% glucose, this overall process was complete in 8 hr and consisted of a germination and an outgrowth phase. Respiration, ribonucleic acid, and protein synthesis began soon after the addition of the germinant, and these processes proceeded at rates characteristic of the germination. The rates of respiration and synthesis increased sharply between 4 and 5 hr, the beginning of the outgrowth, at which time deoxyribonucleic acid synthesis and nitrogen fixation began. Respiration, macromolecular synthesis and nitrogen fixation continued at high rates until the emergence of vegetative cells from the cyst coats.  相似文献

The flavonoid quercetin inhibits collective motility of ejaculated ram spermatozoa in the first 2 hr of incubation; during the next 3-4 hr motility is stimulated. To explain this interesting effect, we followed the influence of quercetin on sperm glycolysis, extracellular pH, ATP content, mitochondrial respiration, and lipid peroxidation. The collective motility of untreated cells is decreased to about 40% of the original motility during two hours of incubation. During this time, the rate of glycolysis is constant, respiration rate is increasing, there is no change in ATP content, the rate of lipid peroxidation is very slow, and the extracellular pH became very acidic (pH 5.5). It is concluded that motility is decreased due to this acidification. This acidification is prevented to some extent by quercetin, which indirectly inhibits glycolysis. Quercetin inhibits motility due to the inhibition of the plasma membrane calcium pump, as we showed previously (Breitbart et al., J Biol Chem 260:11548-11553, 1985). The motility of untreated cells is arrested after 3.5 hr of incubation, whereas quercetin-treated cells show high motility, which continues for additional 2-3 hr. After 3.5 hr, the control cells show no glycolytic activity, ATP content and respiration rates are decreased, and rate of lipid peroxidation is highly increased. At this time, quercetin-treated cells show no glycolytic activity, only a small decrease in ATP content and respiratory rate, and a very low rate of lipid peroxidation. Based on these data it is concluded that sperm motility after 3.5 hr of incubation is dependent mainly on mitochondrial respiration.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Adenine, guanine, uracil and the corresponding nucleosides were identified in mature wheat grain. On germination, contents of these compounds changed in a regular pattern. Amounts of the free bases rose immediately and remained at the increased levels for a short period (3–6 hr). Nucleoside content decreased at the initial phase and increased sharply 12 to 48 hr after germination.  相似文献

The growth ofTrichoderma lignorum was studied in relation to the carbon balance of the culture system and the respiratory activity at different growth phases. Conidia, after inoculation into the medium, swelled and germinated rapidly. The growth rate of the hyphae at the exponential phase was 0.46 hr−1 (2.2 hr for mass doubling) at 25 C. The yield efficiency of hyphal biomass-C at the cost of glucose-C was 67%, while those of the waste products-C excreted and of CO2-C evolved were, 6.5% and 26.5%, respectively. The yield efficiency of conidia-C to the decrease of hyphae-C was 34%. The germination, growth rate and carbon balance were not affected by different concentrations of glucose from 10 to 2×103 mg glucose-C/l. Carbon dioxide was needed as the growth factor for the initiation of the germination of conidia, but there was no increase in yield efficiency as a result of CO2 fixation. The respiratory rate of the fungus changed drastically as the growth proceeded. The rate of endogenous respiration of conidia was less than 0.2 mg CO2-C/g conidia-C/hr which increased immediately after inoculation into the medium. The highest respiratory rate of hyphae (100–110 mg CO2-C/g hyphae-C/hr) was obtained throughout the exponential phase. Thereafter, decreasing rapidly, the respiratory rate of submerged hyphae of 1-week-old showed only 1.8 mg CO2-C/g biomass-C/hr, whereas the rate of aerial hyphae forming conidia increased again, but did not exceed 10 mg CO2-C/g biomass-C/hr.  相似文献

A 10-min irradiation with red light (R) of ‘Grand Rapids’lettuce (Lactuca sativa L.) seeds increased respiratory rates(QO2 and QCO2) over those of far red (FR)-irradiated seeds within2 to 3 hr. Differences in respiration between R- and FR-irradiatedseeds became more pronounced with time. The respiration, indarkness at 25?C, of seeds air dried after germination promotingimpregnation treatments with 1.0 mM GA3+0.5 mM kinetin in acetonewas stimulated slightly by the 6th hour after planting and markedlyby the 12th and 24th hour. Results with seeds in GA3+kinetindissolved in water were similar. ABA, which inhibits germinationand seedling growth, prevented stimulation in respiration oflight-induced seeds. ABA also repressed respiration when usedwith GA3+kinetin. The germination and growth retardant, isopropylN-(3-chlorophenyl) carbamate (CIPC), did not affect the respirationof either R- or FR-treated seeds. We have assessed the activities of the cytochrome and alternative pathways in total respiration and their role in each stage of germination of Zea mays L. radicles. Throughout imbibition, the salicylhydroxamic acid (SHAM) concentration needed to inhibit the cyanide-resistant pathway, without any side effects, decreased from 15 m M in quiescent embryos to 5 m M at 72 h after imbibition. Electrons predominantly flowed through the cytochrome pathway although the alternative pathway was already present at early imbibition. The capacity of the alternative path was about 70% of the control rate of respiration. Its engagement progressively increased from 18% after 10 min of imbibition to 70% at the radicle emergence and then decreased to 50% at 96 h after imbibition, concomitant with the onset of radicle growth. The alternative pathway was, however, not essential for germination. The observed activity of the alternative path correlated with the monosaccharide (glucose + fructose) content, suggesting that the alternative pathway could be acting according to the 'energy overflow model'. On the other hand, up to 24 h after imbibition at 16°C, maize radicles tolerate a severe desiccation, becoming intolerant at 72 h. On reimbibition of tolerant radicles, respiration increased immediately and the alternative pathway was rapidly engaged. At 72 h, no respiration was measured, indicating a total loss of the respiratory systems. The possible correlation between carbohydrate content, loss of desiccation tolerance and activity of the two respiratory pathways is discussed.  相似文献

The effect of infection with tobacco mosaic virus on the respiration rates of detached tobacco leaves in the period immediately after inoculation differed in plants grown at different times of the year. During winter, infection increased respiration rates, and in summer decreased them. In winter-grown plants, increasing the light intensity during the period before inoculation decreased respiration rates after infection. Extending the day length for winter-grown plants did not alter the effect of infection on respiration. Respiration rates began to change in less than 1 hr. after inoculation and are unlikely to be associated with the formation of new virus.  相似文献

Sodium nitrite alone has been shown to stimulate germination of PA 3679h spores. The process was accelerated by using increased concentrations of sodium nitrite, a low pH, and a high temperature of incubation. At low concentrations of nitrite (0.01 to 0.2%), the delay of 36 to 48 hr occurred before germination commenced at 37 C. However, with 3.45% nitrite at 45 C and pH 6.0, most of the spores germinated within 1 hr. At pH 7.0, the germination rate decreased markedly, and at pH 8.0 it was nil. The greatest acceleration in germination rate occurred near 60 C. Hydroxylamine was completely inhibitory to nitrite-induced germination. Sodium nitrite, in turn, inhibited germination by l-alanine, the degree of inhibition being influenced by nitrite concentration and pH.  相似文献

The composition and metabolic activity of cysts of the marine dinoflagellate Scrippsiella trochoidea (Stein) Loeblich were examined during dormancy, quiescence, and germination. On a per cell basis, newly formed cysts contained an order of magnitude more carbohydrate but significantly less protein and chlorophyll a than did exponentially growing vegetative cells. Loss of lipid and carbohydrate from cysts during the initial dormancy period reflected a respiration rate estimated to be 10% of the respiratory activity in vegetative cells. Among older, quiescent cysts the calculated respiration rate decreased further to approximately 1.5% of the vegetative rate and appeared to proceed largely at the expense of carbohydrate reserves. These estimated rates of respiration were in good agreement with direct measurements of cyst oxygen consumption. The transfer of quiescent cysts to conditions permissive for germination resulted in a rapid increase in respiration rate, as evidenced by carbohydrate loss and O2 consumption. The increased respiratory activity was followed by an increase in protein content and, later, by an increase in chlorophyll a content and photosynthetic capacity. Just prior to germination the P/R ratio became greater than 1, and the estimated chlorophyll-specific photosynthetic activity reached 75% of the rate in vegetative cells. Complete restoration of photosynthetic and respiratory capacity apparently was not achieved until after excystment. These data confirm the common assumption that dinoflagellate cysts represent true “resting” cells, containing extensive energy reserves and displaying greatly reduced metabolic activity.  相似文献

Phaseolus mungo seeds 0 to 10 days after germination contained putrescine, spermidine, spermine, cadaverine, agmatine and tyramine. The rate of biosynthesis of total polyamines, proteins and RNA in the developing seeds follows similar profiles, reaching maxima 3 hr from germination. Putrescine, cadaverine, spermidine, spermine and agmatine were the major amines found in Pisum sativum 0–7 days after germination. RNA and proteins seem to follow the same pattern as polyamines during the first 12 hr in the developing pea seeds. RNA reaches a peak at 15 hr and polyamines and proteins peak 24 hr after germination. A rise to total polyamine concentration was also observed in seeds of Tragopogon porrifolius, Zea mays and Triticum aestivum 2–12 hr after germination.  相似文献

The rates of respiration and of photosynthesis of tobacco leaves infected with potato virus X were not affected until the leaves showed symptoms; the respiration rate was then increased by more than 30% and the photosynthesis rate decreased by 20%. When local lesions appeared on the leaves of Nicotiana glutinosa infected with tobacco mosaic virus, but not before, the respiration rate was increased by an amount, up to 30%, that varied with the number of lesions. The photosynthesis rate was decreased by 20%, but there was no effect on photosynthesis or respiration until symptoms appeared. These results differ from those previously reported for tobacco leaves infected with tobacco mosaic virus, in which both respiration and photosynthesis were affected within 1 hr. of inoculation. The validity of extrapolating arguments based on the results obtained with other combinations to this commonly used combination and vice-versa is questioned.  相似文献

Sodium nitrate and nitrite induced germination in a partly after-ripened dormant line of Avena fatua L. The dose-response curves for their stimulation of germination and oxygen uptake were similar, indicating that these compounds may stimulate germination by promoting oxygen uptake. Time-sequence studies showed that nitrate and nitrite stimulated oxygen uptake as much as 70% prior to the first signs of germination. A similar ammonium chloride treatment failed to induce germination or elevate the rate of oxygen uptake, indicating that nitrate and nitrite may promote these events by acting as electron acceptors. The stimulation of germination and oxygen uptake by nitrate and nitrite were not significantly inhibited by salicylhydroxamic acid, an inhibitor of alternative respiration. Thus, stimulation of germination and oxygen uptake by nitrate and nitrite do not require the operation of the alternative pathway of respiration. The stimulation of germination and oxygen uptake by nitrate and nitrite were not inhibited by sodium azide, an inhibitor of cytochrome-mediated respiration; however both germination and oxygen uptake were prevented when salicylhydroxamic acid and sodium azide were applied together. Thus, stimulation of germination and oxygen uptake by nitrate and nitrite require the operation of only one or other of the pathways of respiration; however a specific requirement for the operation of the alternative pathway of respiration does not exist. The function of these compounds as promotors of respiration is discussed with reference to dormancy and the involvement of the Krebs cycle.  相似文献

将雄性Wistar鼠 (体重 1 80± 1 0g)随机分为正常对照组 (NC)、实验对照组 (EC)和两个实验组 (E1、E2 ) ,其中NC、EC组饮用普通水 ,E1、E2组饮用水中分别含有苦瓜皂甙 1 0 0mg·L 1和 2 0 0mg·L 1。饲养五周后 ,后三组动物禁食 2 4h后取血待测 ;取血完毕即开始进食 ,2 4h后再次取血待测。一周后杀死动物取肌肉、肝脏标本检测糖原水平。结果显示苦瓜皂甙虽不影响禁食大鼠血浆胰岛素含量 ,却可明显升高血糖和皮质醇水平 ;显著降低进食后大鼠血糖和胆固醇水平 ,并维持适度的皮质醇水平。肌糖原和肝糖原含量明显增加  相似文献

Pisum sativum seeds contain a conserved acetylcholinesterase (AChE) which is active during the early stages of germination. The enzyme activity soon disappears and reappears after 72 hr of germination. A protein devoid of catalytic ability, but exhibiting similar chromatographic and electrophoretic properties as the active AChE, could be detected after 24 hr of germination. The pattern of incorporation of labelled amino acids into AChE and the influence of cycloheximide revealed that the AChE found in the roots from 72 hr onwards was entirely new. During this period of growth, the AChE protein accounts for 4–10% of the total proteins in the root tissue.  相似文献

Ethanol induced germination in several partly after-ripened dormant lines of Avena fatua L. The dose-response curves for the stimulation of germination and for oxygen uptake were similar, indicating that ethanol may stimulate germination by promoting oxygen uptake. A time-sequence study showed that ethanol stimulated oxygen uptake by as much as 70% prior to the first visible signs of germination. A similar methanol treatment failed to induce germination or significantly elevate oxygen uptake, indicating that the promotive effects of ethanol are not common to all alcohols. The stimulation of both germination and oxygen uptake by ethanol was not inhibited significantly by salicylhydroxamic acid, an inhibitor of alternative respiration. Thus, stimulation of germination and oxygen uptake by ethanol does not require the operation of the alternative pathway of respiration. Similarly, the stimulation of germination and oxygen uptake by ethanol were not inhibited by sodium azide, an inhibitor of cytochrome-mediated respiration. However, both germination and oxygen uptake were prevented when salicylhydroxamic acid and sodium azide were administered together. Thus, stimulation of these events by ethanol requires only the operation of one or other of these pathways of respiration; a specific requirement for the operation of the alternative pathway of respiration does not exist. The function of ethanol as a promoter of respiration is discussed with reference to dormancy and involvement of the Krebs cycle.  相似文献

We have examined germination, protein synthesis and ribonucleic acid (RNA) synthesis by microcysts of the fruiting myxobacterium Myxococcus xanthus. The morphological aspects of microcyst formation were completed at about 2 hr after induction had begun. In such microcysts, germination, RNA synthesis, and protein synthesis were inhibited by actinomycin D (Act D). At 6 hr after induction, germination and protein synthesis had become relatively resistant to Act D, whereas RNA synthesis was inhibited by about 95%. Experiments with (3)H-Act D indicated that the deoxyribonucleic acids of both young and old microcysts bind Act D equally. Resistance of germination to Act D was acquired 4 to 5 hr after induction of microcyst formation, and was due to an Act D-sensitive synthesis at that time. Vegetative cells and microcysts were pulsed with uridine-5-(3)H and chased for 60 min; the RNA was extracted and analyzed by means of sucrose density gradient centrifugation and gel electrophoresis. Both microcysts and vegetative cells were found to contain grossly the same types of RNA in the same proportions. RNA pulse-labeled in microcysts was more stable than that in vegetative cells. No particular portions of the microcyst pulse-labeled RNA were selectively stabilized. These data indicate that a stable messenger RNA required for synthesis of germination proteins was synthesized during microcyst formation. This may be the same as the RNA synthesized 4 to 5 hr after initiation of microcyst formation. We suggest that the existence of such stable messenger RNA in microcysts is consistent with the limited biosynthetic activities of such cells.  相似文献

The effect of O2 partial pressure on the germination and the respiration of 12 cultivated species was studied. The reciprocal of the time necessary to observe rootlet emergence in 50% of the seeds was used to approach the germination rate. The maximum germination and respiration rates were reached in most seeds at O2 pressures close to that of air. Decreasing the O2 pressure produced a gradual decrease of the germination rate. The seeds could be classed in two groups according to their response to low O2 pressures. Group I includes lettuce, sunflower, radish, turnip, cabbage, flax, and soybean: at O2 pressures close to 2 kilopascals, the germination in this group was stopped and the adenylate energy charge was lower than 0.6. Group II includes rice, wheat, maize, sorghum, and pea. The germination rate of these seeds was also gradually decreased by lowering the O2 partial pressure but germination still occured, very slowly, at 0.1 kilopascal; the adenylate energy charge remained higher than 0.6. These differences in the germination rates and adenylate energy charge values could not be explained by differences in the sensitivity of respiration to O2.  相似文献

The RNA content and RNase activity were determined during maturation and germination of rice seeds. The RNA content reached a maximum after the 16th day from anthesis but RNase activity steadily increased up to the last stage of maturation. During germination RNA content was greatest after 24 hr and associated with a very low level of RNase activity. Maximum RNase activity was observed at 72 hr from germination and it afterwards gradually declined. During germination, exogenous application of polyamines decreased the level of RNase activity. RNase was partially purified (448-fold) from 72 hr germinated embryonic axis. Effects of polyamines and other divalent cations were observed on the purified enzyme.  相似文献
