Dielectric measurements in the MHz region of bacteriorhodopsin (bR) in aqueous solution have shown the existence of 1-dispersion with five distinct parts. This dispersion and the -dispersion at low frequencies conform approximately to the Debye relaxation equations. The derived apparent relaxation times showed dependence on various physical parameters. The relaxation effect at low frequencies is attributed to the reorientation of bR chromophore within the purple membrane (PM) fragments. The mechanism which gives rise to the 1-dispersion may well be due to the Maxwell-Wagner effect, although the first two parts of the dispersion could also be attributed to counterion relaxation or to bR reorientation.  相似文献

Purine metabolism plays a major role in regulating the availability of purine nucleotides destined for nucleic acid synthesis. Allantoate amidohydrolase catalyzes the conversion of allantoate to (S)-ureidoglycolate, one of the crucial alternate steps in purine metabolism. The crystal structure of a ternary complex of allantoate amidohydrolase with its substrate allantoate and an allosteric effector, a sulfate ion, from Escherichia coli was determined to understand better the catalytic mechanism and substrate specificity. The 2.25 A resolution X-ray structure reveals an alpha/beta scaffold akin to zinc exopeptidases of the peptidase M20 family and lacks the (beta/alpha)(8)-barrel fold characteristic of the amidohydrolases. Arrangement of the substrate and the two co-catalytic zinc ions at the active site governs catalytic specificity for hydrolysis of N-carbamyl versus the peptide bond in exopeptidases. In its crystalline form, allantoate amidohydrolase adopts a relatively open conformation. However, structural analysis reveals the possibility of a significant movement of domains via rotation about two hinge regions upon allosteric effector and substrate binding resulting in a closed catalytically competent conformation by bringing the substrate allantoate closer to co-catalytic zinc ions. Two cis-prolyl peptide bonds found on either side of the dimerization domain in close proximity to the substrate and ligand-binding sites may be involved in protein folding and in preserving the integrity of the catalytic site.  相似文献

Following the rapid spread of soybean aphid, Aphis glycines in North America in the early 2000’s, biological control was identified as a cost-effective approach for management of this invasive pest. Two parasitoid species, Binodoxys communis and Aphidius colemani, were considered as potential candidates for classical and inundative biological control, respectively. The objectives of the present study were to determine the overwintering capacity of B. communis under climatic conditions prevailing in northeastern North America, and to measure parasitism and dispersal capacity of A. colemani when released in soybean fields. Field and laboratory assessments showed that the Chinese strain of B. communis, Harbin 2002, has a very poor capacity to enter into diapause (<0.8%), and thus to establish in North America. We suggest that this strain has gradually lost its ability to enter diapause during the extended periods of quarantine and laboratory confinement, during which it was continuously exposed to non-diapause rearing conditions. A. colemani did not show strong potential to control A. glycines. Following the release of approximately 8400 females in experimental plots, only 113 mummies were recovered within a radius of 60 m from the release point. Although both parasitoids were promising in controlled environments, the B. communis Harbin 2002 strain and the A. colemani commercial strain did not show strong potential to control A. glycines populations in soybean fields.  相似文献

建立了从心内膜到心外膜的一维心肌几何模型,采用心肌双域模型建立心电电位的仿真模型,通过改变缺血程度构造不同的心室复极不一致状态,利用有限差分法求解控制方程,模拟了心电兴奋在心室复极不一致状态下形成的心电电位,并从中提取QT离散度和兴奋恢复间期(activation-recovery interval,ARI)离散度。分析结果显示:缺血区与正常区电位的QT间期没有明显差异,QT离散度接近于零,不能有效地表征心室肌复极不一致;缺血区ARI明显区别于正常区,ARI离散度与缺血程度有很好的对应关系,可以用来表征心室复极不一致。  相似文献

The recent advent of dispersion-corrected density-functional theory (DFT) methods allows for quantitative modelling of molecular self-assembly processes, and we consider what is required to develop applications to the formation of large self-assembled monolayers (SAMs) on hydrophobic surfaces from organic solution. Focus is on application of the D3 dispersion correction of Grimme combined with the solvent dispersion model of Floris, Tomasi and Pascual–Ahuir to simulate observed scanning-tunnelling microscopy (STM) images of various polymorphs of tetraalkylporphyrin SAMs on highly oriented pyrolytic graphite surfaces. The most significant problem is identified as the need to treat SAM structures that are incommensurate with those of the substrate, providing a challenge to the use of traditional periodic-imaging boundary techniques. Using nearby commensurate lattices introduces non-systematic errors into calculated lattice constants and free energies of SAM formation that are larger than experimental uncertainties and polymorph differences. Developing non-periodic methods for polymorph interface simulation also remains a challenge. Despite these problems, existing methods can be used to interpret STM images and SAM atomic structures, distinguishing between multiple feasible polymorph types. They also provide critical insight into the factors controlling polymorphism. All this stems from a delicate balance that the intermolecular D3 and solvent Floris, Tomasi and Pascual–Ahuir corrections provide. Combined optimised treatments should yield fully quantitative approaches in the future.  相似文献

In order to refine the measure of karyotype asymmetry a new chromosomal parameter of dispersion index is proposed that has the potential to decipher even the minor karyotypic variations, thus permitting further evolutionary gradations to the karyotype asymmetry classes of Stebbins. The higher the dispersion index, the more specialized would be the karyotype. The dispersion index takes into account the variance lor gradual change in chromosome size within a complementvis-a-vis variance for the position of centromere in a karyotypic totality. The dispersion index is calculated as the proportionate measure of centromeric gradient to the coefficient of variation for chromosome length; wherin centromeric gradient = length of median short arm — length of median chromosome. Thus, the three most important karyotypic criteriaviz., differences in: absolute chromosome size, position of centromere and relative chromosome size, are all covered in the proposed parameter. The effectiveness of dispersion index has been tested on a plant taxa,Papaver L., where karyomorphological details, nuclear DNA content, and morphotaxonomic parameters have been amply elucidated from an evolutionary stand point. It is hoped that dispersion index would find immense utility in delimiting species interrelationships particularly in the closely related taxa, when applied in conjunction with other systematic parameters.  相似文献

AIMS: The aim of this study was to overcome the analytical problems encountered during the detection of protozoans by flow cytometry resulting from particle compaction. METHODS AND RESULTS: Malvern Mastersizer (Malvern Instruments, Malvern, UK) was used to characterize the particle distribution of four different water samples and/or particle concentrates incubated with (i) low ionic strength solution or sequestring agent, (ii) anionic or non-ionic surfactants (iii) industry detergent formulations and (iv) physical treatment. The recovery of oocysts and cysts in seeded and treated particle concentrates was estimated by cytometry and microscopy. The decrease in ionic strength of the aqueous solution was most efficient in particle dispersion for different types of water. Moreover, samples treated with deionized water or tetrasodium pyrophosphate showed the highest recovery with more than 80% of the oocysts and cysts recovered. CONCLUSIONS: Chemical treatments that act by altering the ionic strength of the medium are the most efficient for all water types tested here but the overall detergency performance cannot be predicted for all water types. SIGNIFICANCE AND IMPACT OF THE STUDY: Flow cytometric detection has been replaced largely by immunomagnetic separation but the data recorded still have relevance in this technique as well as in molecular techniques requiring DNA or RNA extraction.  相似文献

Microfluidic systems under laminar flow conditions provide in-solution information about species size and binding affinities at very modest sample costs. Flow-induced dispersion analysis directly measures the spread of the analyte profile using Taylor dispersion analysis, whereas microfluidic diffusional sizing quantifies the transfer of analyte from one phase to another. Species of sizes between 0.5 and 1000 nm can be analyzed, and different populations resolved. Both techniques also allow analysis in complex media and medium throughput analysis. These properties make them valuable complements to existing approaches to measure biomolecular interactions.  相似文献

麦秸秆纤维作为一种重要生物质资源已被广泛应用于热塑性复合材料中。纤维在复合材料中的分散性是影响复合材料力学性能的重要因素之一,而目前对其定量化的描述和分析方法仍存在一定不足。本研究基于实验获得的纤维尺寸的统计分布规律,利用随机生成算法模拟纤维在复合材料中的分布;构建描述纤维分散性的指标:分散度,单个纤维数和接触纤维数;统计分析纤维含量、纤维大小对分散性指标影响。结果表明单个纤维数随纤维含量增加而增加,但其增量随纤维含量的增加而降低,降低规律符合三次函数。纤维接触数随纤维含量增加,增加规律符合二次函数,亦符合理论估计。纤维大小影响单个纤维数和接触纤维数的增加幅度,但不影响单个纤维数的百分比。分散度随纤维含量的增加呈线性下降规律。纤维分散性的定量化描述为进一步的复合材料性能分析和建模提供了量化指标。  相似文献

IntroductionEpicardial exit sites of ventricular tachycardia (VT) are frequently encountered during VT ablation requiring an epicardial ablation approach for successful elimination of VT. We sought to assess the utility of repolarization markers in identifying individuals requiring an epicardial ablation approach in addition to an endocardial approach.Methods32 patients who underwent successful ablation for scar mediated VT were included in the study. Fourteen patients who required a combined endocardial and epicardial VT ablation were defined as epicardial VT group (Epi) whereas 18 patients who were successfully ablated from the endocardium alone constituted the endocardial VT group (Endo). Repolarization markers during sinus rhythm were compared between the two groups.ResultsA higher QTc max and QTc dispersion were seen in the Epi group compared to Endo group (479 ± 34 vs 449 ± 20, p = 0.008 and 63 ± 13 vs 38 ± 8, p = 0.001, respectively). Ts-p and Ts-p/Tp-e were higher in the Epi group (166 ± 23 vs 143 ± 23, p = 0.008 and 1.55 ± 0.26 vs 1.3 ± 0.21, p < 0.005). On multivariate regression, QTc dispersion was an independent predictor of the need for an epicardial approach to ablation. A QTc dispersion more than 51.5 msec identified individuals requiring a combined epicardial and endocardial approach to ablation with a sensitivity of 92.9% and a specificity of 100%.ConclusionsPatients requiring an epicardial ablation have a higher QTc dispersion. A value greater than 51.5 msec reliably differentiates between the two groups with high sensitivity and specificity.  相似文献

Dispersion of chironomid larvae between and within stands of the aquatic macrophyte Ranunculus penicillatus var. calcareus was studied by sampling individual leaves of Ranunculus. Two of the six common species exhibited no variation in density between sites, while others had significant variations. A new probability density function was applied to the data and it was revealed that random colonization was not uncommon among these epiphytic larvae. It was suggested that stochastic factors are of some importance in the system under study.  相似文献

Root-knot nematode, Meloidogyne incognita is considered as one of the major non-insect pests of crops. The management of these root feeders becomes highly challenging due to a strong host-parasitic relationship. Pochonia chlamydosporia is a nematophagus fungus that colonizes eggs of nematodes. This study aimed to test the efficacy of P. chlamydosporia (NAIMCC-SF0039) against M. incognita. An oil dispersion formulation of P. chlamydosporia was prepared using emulsifiers and vegetable oil. This formulation had a shelf-life of 90 days (3.3 × 108 CFU/mL) at room temperature (28 ± 1 °C). The inhibitory effect of oil formulation was tested against M. incognita by inoculating it on the egg mass. We found that colonization of the gelatinous matrix occurred on the third day of inoculation followed by complete egg parasitization on the seventh day. A greenhouse trial was laid out to evaluate the biocontrol potential of P. chlamydosporia in cucumber (Cucumis sativus). The results showed that the application of talc formulation of P. chlamydosporia at the rate of 1 kg per acre during planting, followed by delivery of 1 L of oil dispersion formulation through drip lines at 30-day intervals caused the highest reduction of nematode infestation. This treatment recorded 67.9 and 57.5% reduction in egg masses and soil nematode population respectively than that of control.  相似文献

Ants are effective at moving seeds toward their nests, something that may benefit the seeds. We evaluated whether seed movements that may be useful for the rehabilitation of degraded pastures in Colombia can be enhanced by local ants. An artificial aril was prepared and then evaluated in six open cattle pasture farms. Twenty paper disks (each holding seeds with an artificial aril, honey, tuna oil, and control) were set up along linear transects at each farm, and monitored five times in 48 hours. A total of 340 out of 480 seeds were moved from the experimental units by ants. Seeds with tuna oil and an artificial aril were removed twice as frequently as the control and honey smeared seeds. Ectatomma ruidum, Solenopsis geminata, and Pheidole sp. removed the majority of seeds. Advantages of the artificial aril over tuna oil are discussed. This inexpensive technique can enhance seed movement by generalist ants in degraded pastures, likely contributing to regeneration and ecological rehabilitation.  相似文献

Aims While using phylogenetic and functional approaches to test the mechanisms of community assembly, functional traits often act as the proxy of niches. However, there is little detailed knowledge regarding the correlation between functional traits of tree species and their niches in local communities. We suggest that the co-varying correlation between functional traits and niches should be the premise for using phylogenetic and functional approaches to test mechanisms of community assembly. Using functional traits, phylogenetic and environmental data, this study aims to answer the questions: (i) within local communities, do functional traits of co-occurring species co-vary with their environmental niches at the species level? and (ii) what is the key ecological process underlying community assembly in Xishuangbanna and Ailaoshan forest dynamic plots (FDPs)?Methods We measured seven functional traits of 229 and 36 common species in Xishuangbanna and Ailaoshan FDPs in tropical and subtropical China, respectively. We also quantified the environmental niches for these species based on conditional probability. We then analyzed the correlations between functional traits and environmental niches using phylogenetic independent contrasts. After examining phylogenetic signals of functional traits using Pagel's λ, we quantified the phylogenetic and functional dispersion along environmental gradients within local tree communities.Important findings For target species, functional traits do co-vary with environmental niches at the species level in both of the FDPs, supporting that functional traits can be used as a proxy for local-scale environmental niches. Functional traits show significant phylogenetic signals in both of the FDPs. We found that the phylogenetic and functional dispersion were significantly clustered along topographical gradients in the Ailaoshan FDP but overdispersion in the Xishuangbanna FDP. These patterns of phylogenetic and functional dispersion suggest that environmental filtering plays a key role in structuring local tree assemblages in Ailaoshan FDP, while competition exclusion plays a key role in Xishuangbanna FDP.  相似文献

The cultivation of transgenic crops, such as maize, requires successful gene isolation in field environments. Five spatial statistical techniques are used to evaluate the use of a regional mesoscale observation network (Iowa Environmental Mesonet) as a means to drive field-scale pollen dispersion modeling. The Nearest Neighbor Index, Fractal Dimension, Morisita Index, Thiessen Polygons, and Coefficient of Representativity are computed showing the positive and negative impacts of sequential addition of observation networks into a mesonet framework (a collection of pre-existing networks). While it is shown that the arbitrary combination of disparate observing networks increases spatial resolution, this improvement is often at the expense of increased clustering due to co-location of observation sites near urban areas. Network composition in terms of density and degree of clustering was evaluated with a grid analysis using the Barnes scheme as a means to mitigate clustering and improve prediction accuracies when mesonet data are applied to modeling. This paper shows the importance of understanding and accounting for the spatial characteristics of an observational network before applying it to a modeling effort such as field scale pollen dispersion.  相似文献

We consider the idea that foraging animals seek to achieve a certain threshold intake of food per day rather than maximising mean energy intake. Earlier experiments and theoretical work have shown that under these conditions animals should respond to variance in reward. We identify six possible sources of such variance and produce models for five of them (the sixth is already solved). These models describe (1) whether to feed in habitats possessing small or large prey, (2) whether to feed on clumped or dispersed prey, (3) which prey items to eat, (4) the effects of misidentification of prey selection, (5) whether to feed singly or join a flock and (6) when to move between patches when the travel time between them is variable. The results are often radically different from those of traditional models and we discuss their biological significance.  相似文献

The history of human occupation in Brazil dates to at least 14 kyr BP, and the country has the largest record of early human remains from the continent. Despite the importance and richness of Brazilian human skeletal collections, the biological relationships between groups and their implications for knowledge about human dispersion in the country have not been properly explored. Here, we present a comprehensive assessment of the morphological affinities of human groups from East‐Central, Coastal, Northeast, and South Brazil from distinct periods and test for the best dispersion scenarios to explain the observed diversity across time. Our results, based on multivariate assessments of shape and goodness of fit tests of dispersion and adaptation models, favor the idea that Brazil experienced at least two large dispersion waves. The first dispersive event brought the morphological pattern that characterize Late Pleistocene groups continent‐wide and that persisted among East‐Central Brazil groups until recently. Within the area covered by our samples, the second wave was probably restricted to the coast and is associated with a distinct morphological pattern. Inland and coastal populations apparently did not interact significantly during the Holocene, as there is no clear signal of admixture between groups sharing the two morphological patterns. However, these results cannot be extended to the interior part of the country (Amazonia and Central Brazil), given the lack of skeletal samples in these regions. Summary A time‐specific log‐linear regression method on quantile residual lifetime is proposed. Under the proposed regression model, any quantile of a time‐to‐event distribution among survivors beyond a certain time point is associated with selected covariates under right censoring. Consistency and asymptotic normality of the regression estimator are established. An asymptotic test statistic is proposed to evaluate the covariate effects on the quantile residual lifetimes at a specific time point. Evaluation of the test statistic does not require estimation of the variance–covariance matrix of the regression estimators, which involves the probability density function of the survival distribution with censoring. Simulation studies are performed to assess finite sample properties of the regression parameter estimator and test statistic. The new regression method is applied to a breast cancer data set with long‐term follow‐up to estimate the patients' median residual lifetimes, adjusting for important prognostic factors.  相似文献
