The densities of intramembranous particles (IMPs) and of sterol complexes induced by treatment of filipin were studied by freeze-fracture replication of intact intestine and/or isolated brush border membranes (BBM) of well-fed and starved rats. The density of IMPs and filipin-sterol complexes (FSCs) decrease considerably during starvation. Biochemical estimations show a decrease in the levels of cholesterol and proteins with respect to phospholipids during starvation which is in agreement with morphological findings. It is suggested that these changes may play a role in regulating membrane fluidity which in turn affects absorption of nutrients through BBM.  相似文献

本实验用小鼠血液淋巴细胞为核供体进行了核移植研究。用淋巴细胞分离液(比重1.088)分离出小鼠血液中的淋巴细胞,直接用作核移植供体细胞,采用胞质内注射法成功构建的重构胚经常规培养2h后,SrCl_2激活处理6h,然后添加mM16培养液和小鼠输卵管上皮细胞饲养层共培养。把发育至早期囊胚阶段的重构胚转移至小鼠胎儿成纤维细胞饲养层上,添加ES细胞培养液继续培养。对孵化出的内细胞团进行消化,然后接种培养。结果显示,小鼠血液淋巴细胞可以支持体细胞核移植重构胚的发育,核移植重构胚2-细胞率41.03%(128/312),桑葚胚和囊胚发育率分别为9.29%(29/312),1.92%(6/312)。重构囊胚在小鼠胎儿成纤维细胞饲养层上分离出2个内细胞团,分离率为0.64%(2/312)。实验证实利用小鼠血液淋巴细胞进行体细胞核移植是可行的,可用于深入研究。  相似文献

本文用流式细胞术(FCM)和免疫细胞化学方法,检测了实验性小鼠脾的淋巴细胞增殖周期和5-溴脱氧尿苷阳性细胞(BrdU ̄+)在脾脏内的分布.以及脾小结中IgM ̄+细胞的反应。结果显示由利血平诱发的脾虚小鼠S期细胞数最少(占5%),G_1和G_2+M期细胞增加。经过健脾汤治疗后S期细胞增至10%,与对照组比较无显著性差异.但高于自然恢复组(P<0.001)。经免疫细胞化学显示:脾虚组小鼠的脾中5-溴脱氧尿苷阳性细胞(BrdU ̄+)的数量远低于其它组。脾小结缩小,帽状区IgM ̄+B细胞基本消失,经健脾汤治疗后,复健组的脾小结与自然恢复组比较,体积明显增大,生发中心的BrdU ̄+和帽状区IgM ̄+B淋巴细胞均相应增多。提示健脾汤有促进DNA和IgM合成及细胞增殖作用。  相似文献

Androgens produced by stimulating mouse testis with gonadotropic hormones cause a rise in renal β-glucuronidase but not an increase in acid or alkaline phosphatase. All subcellular components increase in β-glucuronidase activity, with a relatively greater increment in particulate enzyme as compared with that free in the cytoplasm (non-sedimentable). A small percentage of recovered β-glucuronidase, acid phosphatase, and alkaline phosphatase is found in material which rises to the surface during centrifugation in sucrose media (fraction I). The specific activity of β-glucuronidase and acid phosphatase in this fraction is normally quite high with respect to the homogenate, while that of alkaline phosphatase is not. On the other hand, the fraction I material from androgen-stimulated mice exhibits a further increase in specific activity with respect to β-glucuronidase and not acid phosphatase. It thus appears that there is an independence in the behavior of individual enzymes in response to physiologic stimuli in spite of obvious morphologic proximity.  相似文献

 富集的人外周血T淋巴细胞经40—42℃保温(热休克)可诱导产生90kd和71kd的两种主要热休克蛋白(HSP),此外,62和34kd HSP也可在不同条件下诱导。从不同处理的T淋巴细胞中提取mRNA并在免网织细胞裂解液系统中进行体外转译,显示出相同的主要HSP。其中71和62kdHSP不仅在升温时还可在低温(4℃)下诱导,表明淋巴细胞中几种主要HSP的诱导机制不完全相同。比较L-~(35)S-Met参入实验和体外转译的结果提示淋巴细胞中HSP基因表达主要在转录水平调控。  相似文献

植被的降雨分配作用对理解生态系统的水文功能具有重要的意义。该文对四川岷江上游岷江冷杉(Abies faxoniana)针叶林和川滇高山栎(Quercus aquifolioides)灌丛两种生态系统的降雨分配及降雨截留的影响因素进行了研究,探讨了植被分配降雨及截留降雨的影响机制和影响因素。文中采用定位观测的方法研究降雨分配。针叶林中冠层降雨截留占33.33%,树干茎流占0.07%,穿透雨占66.60%;而灌丛的冠层截留降雨为24.95%,穿透雨为75.05%;针叶林地被物的蓄留水能力(1.746 mm)要大于灌丛地被物的持水能力(0.941 mm);针叶林土壤的容积含水率(39.66%)也要高于灌丛土壤的容积含水率(38.19%);两种生态系统中的穿透雨率与降雨量的关系均可用逻辑斯谛方程较好地模拟。文中还选取了降雨量、降雨强度、降雨持续时间、两次降雨的间隔时间和次降雨期间的气温等5种因子分析影响两种生态系统降雨截留的主要因素。根据截留降雨与上述5种因子的偏相关分析结果:针叶林冠层的降雨截留主要受降雨量、降雨持续时间和间隔时间的影响;灌丛的降雨截留主要受降雨量、气温与降雨持续时间的影响。文中从当地的降雨特征与两种生态系统微气候差异的角度分析了两种生态系统降雨分配及降雨截留影响因素差异的原因。  相似文献

目的探讨淋巴细胞浸润在肥厚性扁平苔藓发病机制中的意义。方法采用免疫组织化学SP法对10例肥厚性扁平苔藓患者皮损、20例非肥厚性扁平苔藓患者皮损及10例正常人皮肤中CD3、CD4、CD8、CD20进行检测。结果所有患者皮损均以CD3^+’T、CD4^+T、CD8^+T细胞浸润为主,散在CD20^+B细胞浸润。肥厚性皮损中浸润T淋巴细胞数少于非肥厚性皮损(P〈0.01),CD20^+B细胞占浸润淋巴细胞百分率高于非肥厚性皮损(P〈0.05)。正常人皮肤仅在真皮偶见CD3^+T细胞。结论肥厚性扁平苔藓皮损T淋巴细胞的减少,B淋巴细胞的增多表达可能与疾病的持续存在及局部过度增殖性改变密切相关。  相似文献

The neurotransmitter acetylcholine regulates cAMP concentrations in mouse neuroblastoma cells (clone NS20). In these cells, the action of acetylcholine appears to be specific: it does not alter basal concentrations of cAMP, but prevents the elevation of cAMP which is mediated by either adenosine or prostaglandin E1. The receptor for acetylcholine which is involved in this phenomenon has been identified as muscarinic. Pilocarpine and carbamylcholine, but not acetate or choline, will substitute for acetylcholine. Furthermore, the action of 10 μM-carbbamylcholine is blocked by ≥ nM concentrations of atropine, isopropamide or 3-quinuclidinylbenzilate, but not by mM concentrations of d-tubocurarine or hexamethonium. Of eight cholinergic antagonists tested, decamethonium and succinylcholine were the only two which were able to substitute for acetylcholine. These two antagonists are known to cause depolarization of post-synaptic cells. Decamethonium and succinylcholine appear to interact with the same muscarinic receptor, as their actions are also blockèd by low concentrations of 3-quinuclidinylbenzilate. In addition to these two depolarizing antagonists, the ionophores, valinomycin, A23187 and X537A, were also found to prevent elevation of cAMP concentrations. The involvement of specific membrane depolarization as being the active agent by which acetylcholine inhibits elevation of cAMP concentrations is discussed.  相似文献

Abstract— Polysomes were isolated from several different fractions of mouse brain tissue. After homogenization, the extract was centrifuged to yield a post-mitochondrial supernatant fraction and a pellet fraction. Sucrose gradient analysis of the material in the post-mitochondrial supernatant fraction indicated that 80 per cent of the ribosomes were present in polysomes and that little, if any, of the pellet fraction was present. Sucrose gradient analysis of the solution obtained after washing the pellet showed that very little polysomal material was present. The remaining pellet fraction was resuspended in a detergent mixture of deoxycholate-Tween 40. Sucrose gradient analysis of the resulting detergent-soluble solution indicated that large amounts of ribosomal material, in which 60–70 per cent of the ribosomes were associated in polysomes, were present.
In brain tissue from young animals, 20 per cent of the polysomes were found in the post-mitochondrial supernatant fraction whereas 80 per cent of the polysomes were released from the pellet fraction by detergent treatment. In contrast, in brain tissue from adult animals, 40 per cent of the polysomes were found in the post-mitochondrial supernatant fraction, whereas 60 per cent of the polysomes were released from the pellet fraction by detergent treatment.  相似文献

用辣根过氧化物酶标记的胎球蛋白(Fet-HRP)为探针小鼠角膜发育期间胎球蛋白受体(RF)在光镜水平的定位和变化。结果表明:角膜上皮于胎龄11天出现RF,主要分布在细胞表面;角膜基质自胎龄13天出现RF,15天时最多,出生后减少;角膜内皮在胚胎期未发现RF。文中讨论了RF与角膜基质组建间的关系。  相似文献

The fine structure of rat thymic lymphocytes from early prophase to late telophase of mitosis is described, using material fixed at pH 7.3 either in 1 per cent OsO4 or in glutaraldehyde followed by 2 per cent OsO4. The structure of the centriolar complex of interphase thymocytes is analyzed and compared with that of centrioles during division. The appearance of daughter centrioles is the earliest clearly recognizable sign of prophase. Daughter centrioles probably retain a secondary relation to the primary centriole, while the latter appears to be related, both genetically and spatially, to the spindle apparatus. The nuclear envelope persists in recognizable form to help reconstitute the envelopes of the daughter nuclei. Ribosome bodies (dense aggregates of ribosomes) accumulate, beginning at late prophase, and are retained by the daughter cells. Cytokinesis proceeds by formation of a ribosome-free plate at the equator with a central plate of vesicles which may coalesce to form the new plasma membrane of the daughter cells. Cultures of human adenoidal lymphocytes exposed briefly to either phytohemagglutinin (PHA), Staphylococcus filtrate (Staph-F), concanavalin-A(Con-A), or pokeweed mitogen (PWM) incorporate increased amounts of thymidine earlier than replicate cultures exposed continuously to the mitogens. These effects can begin in the first 24 hr of culture and are seen maximally between 36 and 72 hr. Once a blastogenic response is established, PHA or PWM can diminish that response. Inhibition with PWM requires that the initial stimulation was with this mitogen, while PHA can inhibit blastogenesis to both PHA and PWM-stimulated cells. Because these mitogens can have a paradoxical effect on adenoidal lymphocytes, being capable of both initiating and inhibiting DNA synthesis, this phenomena should be kept in mind when such systems are utilized for the evaluation of antigens and drug effects.  相似文献

A tentative model describing the passage of circulatory lymphocytes through the spleen is formulated in accord with known anatomical features. In order to preserve isomorphism between the model and the splenic system, the model is formulated in compartmental form and its design allows alternative routes and modes of lymphocyte transit to be considered. The simultaneous differential equations arising from the model are solved using an analogue computer which also provides the means whereby the performance of the model may be compared with suitable dynamic data drawn from literature. This not only allows the selection of a particular configuration of the model in preference to its alternatives, but also allows the numerical determination of certain unknown parameters. In the case of the rat spleen, best agreement between model and experimental data is obtained when between 10 and 25% of the total lymphocyte flux in the model spleen passes through the marginal zone where the average dwell time of the lymphocytes is about 50 min. The white pulp receives a lymphocyte flux from the marginal zone amounting to about 10% of the total splenic flux and the white pulp lymphocytes are sequestered for a period of 4–6 hr before release to the venous circulation. The red pulp receives 90% of the total splenic flux but the majority of lymphocytes find transit through the red pulp in less than 5 min. The remaining flux of lymphocytes, amounting to 10% of the splenic input, is delayed in transit through the red pulp by 2–3 hr before release to the venous circulation.  相似文献

The DNA synthesis pattern and several kinetic parameters of in vitro PHA stimulated normal and CLL lymphocytes were determined. The DNA synthesis peak of CLL lymphocytes occurred 2–3 days later than that of normal lymphocytes. The generation time, estimated by the labeled mitoses method, was found to be 28 hr and 20 hr for CLL and normal lymphocytes respectively. This difference was mainly due to longer S and Gt periods. It was also shown that both CLL and normal lymphocytes divide several times. These data were confirmed by the chromatid labeling pattern and by the halving of the grains and the double labeling techniques. By combining continuous and pulse labeling the growth fraction of CLL lymphocytes was found to be progressively increasing, because of the recruitment of new cells in cycle, from the third day of culture. Therefore the delayed peak of DNA synthesis of CLL lymphocytes was caused by a longer cell cycle and by a longer pre-replicative phase.  相似文献

用全细胞膜片箝技术研究了小鼠心室肌细胞去极化激活的非钙依赖性外向电流的特征。短时去极化至-50mV以上电位可引发快速上升的外向钾电流,之后缓慢衰减至一平台。在改变保持电位时,该峰电流与平台电流的比值也发生改变,并测得两者翻转电位分别为-64.3±3.9和-53.3±2.9mV。外向电流的衰减过程可用双指数方式很好拟合,而在500ms预脉冲刺激后再经去极化刺激的电流时程适合单指数方式。结果提示:小鼠心室肌细胞的外向电流包括两种失活成分(Ikf,和Iks),它们对4-氨基吡啶具有相似的敏感性。Ikf的稳态失活在测试电位范围内(-80-+30mV)均不完全,这可能与小鼠心肌动作电位时程很短有关联。  相似文献
