Artificial insemination in deer and non-domestic bovids   总被引:1,自引:0,他引:1  
Artificial insemination technology has revolutionized the domestic cattle breeding industry and allowed for the dissemination of valuable genetics worldwide. This technology has been adapted for use in many other taxa for the conservation of threatened and endangered species, but its use for the genetic management of small populations of deer, antelope and other non-domestic bovids has met numerous challenges and limited success. In practice, adaptation of domestic bovine AI protocols to other artiodactylids for genetic management has been limited by: (1) a lack of understanding of species-specific reproductive characteristics; (2) the inability to minimize handling stress; (3) pregnancy losses; and (4) regulatory challenges in semen importation. To date, AI protocols have been developed for seven species of cervid and 14 species of non-domestic bovids; recent developments in this technology has allowed greater use of AI for dissemination of genetics in farmed deer species. However, despite decades of research in the use of assisted reproduction for the conservation of antelope and other non-domestic bovids, even this simplest technique has not been used repeatedly for genetic management.  相似文献

Assisted breeding technology (ART), including artificial insemination (AI), has the potential to advance the conservation and welfare of marsupials. Many of the challenges facing AI and ART for marsupials are shared with other wild species. However, the marsupial mode of reproduction and development also poses unique challenges and opportunities. For the vast majority of marsupials, there is a dearth of knowledge regarding basic reproductive biology to guide an AI strategy. For threatened or endangered species, only the most basic reproductive information is available in most cases, if at all. Artificial insemination has been used to produce viable young in two marsupial species, the koala and tammar wallaby. However, in these species the timing of ovulation can be predicted with considerably more confidence than in any other marsupial. In a limited number of other marsupials, such precise timing of ovulation has only been achieved using hormonal treatment leading to conception but not live young. A unique marsupial ART strategy which has been shown to have promise is cross-fostering; the transfer of pouch young of a threatened species to the pouches of foster mothers of a common related species as a means to increase productivity. For the foreseeable future, except for a few highly iconic or well studied species, there is unlikely to be sufficient reproductive information on which to base AI. However, if more generic approaches can be developed; such as ICSI (to generate embryos) and female synchronization (to provide oocyte donors or embryo recipients), then the prospects for broader application of AI/ART to marsupials are promising.  相似文献

The experience with artificial insemination (AI) and the more invasive ARTs (assisted reproductive technologies) in the propagation of non-human primates (NHPs), although limited, has included representation from the Great Apes and both Old World and New World Macaques. The application of these technologies in NHPs is impacted by high cost, substantial technical requirements and the limited captive populations of available animals. A major incentive for their use would be to propagate endangered, underrepresented individuals or valuable founder animals. Detailed protocols and a substantial experience base for the ARTs are available for rhesus and cynomolgus macaques and form the basis of this review, including sperm recovery, processing and long-term storage at low temperatures, insemination techniques and timing. Controlled ovarian stimulation and subsequent oocyte recovery required for the invasive ARTs such as intracytoplasmic sperm injection (ICSI), is also described. Three recent AI reports in Old World Macaques are reviewed, along with examples of the use of the ARTs in the propagation of valuable founder animals, in the preservation of endangered macaques, and finally in the creation of neurodegenerative disease models for biomedical research purposes.  相似文献

Artificial insemination (AI) and semen freezing have become services available to dog owners worldwide, and the demand for services to freeze semen is increasing. In other canids such as the fox, the fur industry utilizes fresh or frozen semen to artificially inseminate vixens to produce pelts. Clearly, AI facilitates the use of a male to sire several females by diluting the ejaculate, increases breeding hygiene, and allows crossing between species with slightly different breeding seasons. The African wild dog (Lycaon pictus) is currently considered by the World Conservation Union (IUCN) as one of most endangered canids. In captive populations of African wild dogs, semen has been frozen with encouraging results, using a standard cryopreservation protocol for domestic dogs, but successful AI has not been reported. In wolves, there is one report regarding the live birth of an offspring after intravaginal AI of a deslorelin-induced estrous female. In 2005, three Mexican gray wolf females were artificially bred by intrauterine insemination with freshly collected semen from unrelated males, and all females whelped. Artificial insemination may be vaginal, intrauterine or intratubal, and the semen may be fresh, fresh and chilled (diluted), or frozen-thawed, and the source of semen may be epididymal or ejaculated. In the domestic dog, the results are good to excellent for AI with all three types of processed semen when the source is ejaculated semen, whereas epididymal sperm still yields poorer results. Species differences in female physiology, as well as differences in the cryotolerance of the sperm from various canid species, warrant further research and development.  相似文献

This paper traces the history of artificial insemination by selected donors (AID) as a strategy for positive eugenic improvement. While medical artificial insemination has a longer history, its use as a eugenic strategy was first mooted in late nineteenth-century France. It was then developed as 'scientific motherhood' for war widows and those without partners by Marion Louisa Piddington in Australia following the Great War. By the 1930s AID was being more widely used clinically in Britain (and elsewhere) as a medical solution to male infertility for married couples. In 1935 English postal clerk, Herbert Brewer, promoted AID (eutelegenesis) as the socialization of the germ plasm in a eugenic scheme. The next year Hermann Muller, American Drosophila geneticist and eugenicist, presented his plan for human improvement by AID to Stalin. Some twenty years later, Muller, together with Robert Klark Graham, began planning a Foundation for Germinal Choice in California. This was finally opened in 1980 as the first practical experiment in eugenic AID, producing some 215 babies over the twenty years it functioned. While AID appeared to be a means of squaring a eugenic circle by separating paternity from love relationships, and so allowing eugenic improvement without inhibiting individual choice in marriage, it found very little favour with those who might use it, not least because of a couple's desire to have their 'own' children has always seemed stronger than any eugenic aspirations. Conservation is about protecting and nurturing species so that they can survive, not only now, but also into the future. Ideally this means protecting genetically diverse populations and not simply breeding a few individuals. Unfortunately, this point is often overlooked by reproductive technologists, especially if they are more accustomed to working with humans, companion animals or agricultural species, where the goals are more usually directed towards obtaining offspring from particular individuals. This approach has tended to antagonise the conservation community, who are quick to develop an unreasonable suspicion of technological solutions, partly because they are unfamiliar with the scientific principles that underpin the reproductive technology. Unfortunately, this mutual failure to recognise that all parties are actually well meaning, has led to separate cultures that barely communicate with each other and thus fail to capitalise on the potential benefits that would come from a good working relationship. Notable successes with reproductive technology have only emerged where such relationships have been forged. In this review, we highlight, mainly for the benefit of the technologist community, the need to foster good working relationships with conservation managers and to recognise that the latest hi-tech approach to animal breeding is more likely to engender suspicion than enthusiasm.  相似文献

The objective was to determine the effect of powdered coconut water extender (ACP-106c) on the proportion of female puppies born. Twenty French Bulldog bitches were subjected to natural mating (NM) and, during the subsequent two estrus periods, were bred by intravaginal artificial insemination (AI), using chilled semen (from the same males) diluted in Tris-egg yolk (AI-Tris) or ACP-106c (AI-ACP-106c). Fresh semen was cooled to 5 °C and maintained at that temperature for 6 h, rewarmed (37 °C for 30 s), and used for AI. Pregnancy and whelping rates following NM were both 100% and were both 90.0% following AI with either extender. Litter size (mean ± SD) was 5.4 ±1.1, 4.7 ± 2.0, and 5.1 ± 2.0 (P > 0.05) for NM, AI-Tris, and AI-ACP-106c, respectively. Furthermore, for these groups, the number of female vs. male puppies born were 2.6 ± 0.6 vs. 2.8 ± 1.0, 2.2 ± 1.0 vs. 2.5 ± 1.1, and 3.4 ± 1.6 vs. 1.8 ± 1.2 (P < 0.05 for AI-ACP-106c only). In conclusion, our hypothesis was supported; AI of semen in ACP-106c extender resulted in a significantly higher proportion of female puppies. Furthermore, this extender yielded acceptable litter size and rates of pregnancy and whelping.  相似文献

The objective of this study was to describe the clinical findings, semen characteristics, and testicular histopathology in male dromedary camels affected with impotentia generandi (IG). According to the history, 82.6% (38/46) of the cases were classified as primary-IG (P-IG; never been able to impregnate a female), whereas 17.4% (8/46) were classified as secondary-IG (S-IG; acquired infertility). Only one scrotal testis was observed in four cases, and no scrotal testis was observed in one case. Overall, testicular length, width, and depth were 6.46 ± 0.2, 3.41 ± 0.1, and 2.8 ± 0.08 cm, respectively. Within the P-IG males, 42.2% of the testes were classified as small, 47.9% as normal, and 9.9% as large. Within the S-IG males, 0.0% of the testes were classified as small, 80% as normal, and 20% as large. Ejaculate volume, total sperm number in the ejaculate, and sperm motility, viability, and abnormal morphology were 4.4 ± 0.3 mL, 25.7 ± 1.0 × 106, 18.7 ± 3.1%, 25.2 ± 3.4%, and 46.6 ± 3.7%, respectively. Azoospermia was observed in 30.4% of the cases, asthenospermia was observed in the 25% of the cases, and necrospermia was observed in 10% of the cases. The proportion of abnormal sperm was between 20% and 50%, and between 60% and 94% in 56.2% and 34.4% of the cases, respectively. Hypospermatogenesis, arrested spermatogenesis, Sertoli cell–only syndrome, and testicular degeneration were the main histopathological findings. In conclusion, IG in male dromedary camels appears to be related mainly to testicular dysfunction, which alters semen quality and reduces fertility.  相似文献

Techniques used for the artificial insemination of macaques have primarily involved instillation of the ejaculate into the vagina or cervical canal. Intrauterine insemination has been performed previously but only as a surgical procedure. The application of ultrasound-guided techniques for this purpose provides a method for efficiently inseminating macaques in a relatively noninvasive fashion. This report describes the successful transabdominal ultrasound-guided insemination of the long-tailed macaque and the potential application of this procedure to a variety of nonhuman primate species.  相似文献

The ovarian follicular wave patterns of sixty adult female camels were monitored by serial trans-rectal ultrasound examinations and when the dominant follicle reached 1.3-1.8 cm in diameter they received a single intravenous injection of 20 microg buserelin, to induce ovulation, and were inseminated with a known number of spermatozoa 24 h later. Ejaculates were collected from the male camels and diluted 1:1 in Green Buffer with 20% egg yolk (v:v) added. Sperm concentration and motility were assessed and a dose of 40, 80 or 150 x 10(6) motile spermatozoa were deposited either just through the cervix into the uterine body or at the tip of the horn ipsilateral with the ovary containing the dominant follicle. Insemination of 150, 80 and 40 x 10(6) spermatozoa into the uterine body resulted in conception rates of 53, 7 and 0%, respectively, whereas insemination at the tip of the uterine horn resulted in conception rates of 43, 40 and 7%, respectively.  相似文献

The majority of horse registries approve the use of artificial insemination, and horse breeding has widely taken benefit from the use of cooled-stored semen. New insights into cooled-semen technology open possibilities to reduce problems such as impaired semen quality after cooled-storage in individual stallions. The stallion itself has major impacts on quality and fertility of cooled-stored semen. Dietary supplementation of antioxidants and polyunsaturated fatty acids improves semen quality in a variety of species, but only few studies on this topic exist in the horse. Proper semen collection and handling is the main key to the maintenance of semen quality during cooled-storage. Semen collection should be achieved by minimal sexual stimulation with a single mount; this results in high sperm concentration, low content of seminal plasma and minimal contamination with bacteria. Milk-based semen extenders are most popular for semen processing and storage. The development of more defined extenders containing only the beneficial milk ingredients has made extender quality more constant and reliable. Semen is often centrifuged to decrease the seminal plasma content. Centrifugation results in a recovery rate of only 75% of spermatozoa in the semen pellet. Recovery rates after centrifugation may be improved with use of a "cushion technique" allowing higher centrifugation force and duration. However, this is not routinely used in cooled-semen technology. After slow-cooling, semen-storage and shipping is best performed at 5 degrees C, maintaining semen motility, membrane integrity and DNA integrity for up to 40 h after collection. Shipping containers created from Styrofoam boxes provide maintenance of semen quality at low cost.  相似文献

A technique has been developed for artificially inseminating freshly molted female lobsters (Homarus). Using the criterion that eggs were fertilized if they showed development of normal cleavage patterns, at least 49.5% of the inseminated females extruded fertilized eggs. Sixty-two percent of the fertilized eggs developed to the eyespot stage, and 26.6% of these subsequently hatched. Eggs were not fertilized in 7.7% of the extrusions. In 42% of the samples, it was not possible to establish whether fertilization had occurred because either samples were collected prior to initiation of cleavage or eggs were too poorly preserved to assess cleavage with confidence. It is probable that many of the eggs that were evaluated as equivocal were also fertilized. This technique should be valuable in the experimental and commercial aquaculture of lobsters and might be directly applicable to other crustaceans.  相似文献

A reliable method is reported for the long-term preservation of ovaries and spermatozoa of the silkworm (Bombyx mori). Three studies are presented. In the first, ovaries were removed from larvae at either 3rd, 4th, or 5th instar, cryopreserved, and stored in liquid nitrogen. Thawed ovaries were transplanted to surgically castrated female larvae at the same or a different developmental stage. The highest percentage of recipient females producing eggs resulted into either 3rd or 4th instar larvae (respectively, 22.1 and 8.7%). Similarly, the highest levels of other measurements of successful cryopreservation and transplanted ovary, and number of eggs laid, occurred with the same combination of donor and recipient developmental stages. Other combinations of ovary/recipient developmental stages yielded lower results. In the second experiment, semen was collected from male moths, cryopreserved, and then thawed semen was diluted with trypsin solution and artificially inseminated into females obtained from the best conditions of first experiment. A small percentage of inseminated moths laid eggs (8-10.3%) compared to that of controls (100%). In addition, the fertility of eggs from experimental moths was lower than that of control females (respectively, 40.3-88% and 97.5%). In the third experiment, eggs were surgically removed from ovarian tubules of moth following transplantation of thawed ovaries and subjected to parthenogenetic activation and artificial hatching. As expected, all resulting moths were female and, following natural mating or artificial insemination with thawed semen, yielded normal offspring at high rates.  相似文献

An overview of the present status of the use of artificial insemination (AI) in South American camelids and wild equids is offered. Technical aspects of semen collection, dilution and cryopreservation have limited the development and use of AI in camelid and equid species. To-date, efficiency is low but progress has been made and viable offspring have been produced through the use of AI in domestic South American camelids using both fresh and frozen semen. The origin, composition, and function of the viscous component of camelid seminal plasma remain a mystery and an obvious area for future research. A better understanding of the normal constituents of seminal plasma will enable the rational design of semen extenders suitable for camelids. Post-thaw sperm viability is very low, and studies are needed to address questions of optimal freezing and thawing procedures as well as the insemination dose. The basis for differences in reported pregnancy rates with sexed and frozen semen in domestic equids, and the ultimate success of AI in wild equids will require continued research into the "stallion effect", extenders and cryoprotectants, optimal volume and number of spermatozoa, temperatures during handling, processing an transport, and insemination techniques. In both camelids and equids, research on domestic species under controlled conditions provides and excellent opportunity to develop effective semen handling techniques for application in wild and endangered species of the respective families.  相似文献

We examined the effect of inseminating mixed parity sows (n = 231) once with fewer sperm at different times relative to ovulation. Lactation length was 19 days and sows received an IM injection of 600 IU equine chorionic gonadotrophin (eCG) 12 h before weaning. At 80 h after eCG injection, sows received an IM injection of 5 mg porcine luteinizing hormone (pLH). Predicted time of ovulation (PTO) was 38 h after pLH injection. Sows were assigned by parity to receive a single transcervical artificial insemination (AI) at either 6 or 24 h before PTO with semen doses containing either 2.5 or 1.25 × 109 sperm. A positive control group of sows (n = 49) was subject to conventional AI 24 and 6 h before PTO. Detection of estrus was performed in the presence of a boar and only sows exhibiting estrous behavior at the assigned time of AI were included in the study. Farrowing rate for sows receiving 2.5 × 109 sperm at 6 h before PTO was greater than that for sows receiving 1.25 × 109 sperm at 24 h before PTO (85% versus 61%, P < 0.05). All other groups were intermediate. There was no effect of time of AI or sperm numbers on subsequent litter size. These data indicate that single insemination of fewer sperm may compromise sow fertility, even when performed transcervically, if not appropriately timed relative to ovulation.  相似文献

This study was undertaken to investigate serum and follicular fluid (FF) concentrations of some biochemical metabolites during the low (May to October) and the peak breeding season (November to April) in female camels with small and large follicles. For this purpose, ovaries from 92 female camels aged 3–7 years (young) or 8–15 years (adult) with clinically normal reproductive tract and slaughtered over a 24-month period were collected. Jugular blood samples and FF aspirated from small (5–9 mm) and large (10–20 mm) follicles were analyzed for various metabolite concentrations, using the commercial kits.The effect of season, age and follicular size on serum glucose levels was not significant. However, FF glucose concentration in small follicles (136.79 ± 4.05 mg/dl) was higher (P < 0.05) than that of large follicles (77.09 ± 4.31 mg/dl). Serum cholesterol contents were neither affected by the season, nor by age of the animal or the size of the ovarian follicles. The FF cholesterol concentration during the low breeding season (21.08 ± 1.11 mg/dl) was higher (P < 0.05) than 6.25 ± 1.14 mg/dl recorded during the peak breeding season. The serum and FF total protein and albumin concentrations were neither affected by the season, nor by the age of the animal or the size of the ovarian follicles. The FF globulin concentration during the peak breeding season (2.46 ± 0.06 g/dl) was higher (P < 0.05) than 1.56 ± 0.06 g/dl recorded during the low breeding season. Serum and FF activities of AST and ALT did not differ between the two seasons, age groups or follicle classes. Serum triglycerides (56.12 ± 1.28 mg/dl) and HDL (45.12 ± 0.12 mg/dl) levels during the peak breeding season were higher (P < 0.01) than 31.91 ± 1.25 and 42.60 ± 0.11 mg/dl, respectively, observed during the low breeding season. Serum and FF triglycerides were neither affected by age nor by follicle size. Serum HDL concentration was higher (P < 0.05) in adult than young camels. The concentration of HDL in FF was higher (P < 0.05) during the peak (41.92 ± 0.06 mg/dl) than the low (40.80 ± 0.06 mg/dl) breeding season. It was concluded that serum contents of triglycerides and HDL were influenced by the breeding season. Similarly, FF contents of cholesterol, globulin and HDL were influenced by season, while FF glucose contents were influenced by the size of the follicle. However, no correlation could be established between serum and follicular fluid contents of various biochemical metabolites included in the study.  相似文献

This research aimed to evaluate two concentrations of egg yolk inclusion rates (20 and 2.5%) in the semen extender of goat semen cryopreserved during two seasons of the year. The study was conducted during a light-induced breeding season (Experiment 1), and during the natural breeding season (Experiment 2), in the southern hemisphere. Four ejaculates from each buck (n = 2) were collected in each experiment. After collection, semen was divided, with each sample being diluted in the semen extender – according to the treatments (T1 – 20% egg yolk or T2 – 2.5% egg yolk, using a glucose–EDTA extender). For T1 treatment in Experiment 2, the semen was also washed before the semen cryopreservation process. The semen samples were frozen, and after thawing evaluated for seminal characteristics i.e. sperm motility, vigor, morphology and membrane integrity. The fertilising capacity of the frozen-thawed semen was evaluated following a single artificial insemination 12 h after the onset of estrus in 50 (Experiment 1) and 60 does (Experiment 2). In Experiments 1 and 2, the mean values for sperm motility and membrane integrity of the frozen-thawed semen did not differ between the T1 and T2 treatments. However, the mean sperm vigor and morphological normal sperm were greater (P < 0.05) in T2 than T1 treatment. The fertility rates recorded did not differ between T1 and T2 treatments in Experiment 1, however, it was greater (P < 0.05) in the T2 than in the T1 treatment, in Experiment 2. According to obtained results, it can be recommended to use a glucose–EDTA extender with a low egg yolk concentration (2.5%) inclusion, for superior fertility results in goats.  相似文献

The aim of this study was to optimize the laparoscopic intrauterine insemination (LAI) methodology by testing different time intervals between the natural heat detection and ewe insemination. Three experiments were performed in the breeding conditions of Southern Kazakhstan. Ewes (n = 243) were exposed for one hour to direct contact with the teaser rams (once a day, morning or evening). Ewes expressing behavioral symptoms of heat were detected and inseminated with the use of LAI during 210–1290 min (3.5–21.5 hrs) after heat detection. Reproductive traits of lambing rate (LR) and litter size (LS) were recorded according to the births registered at 137 to 152 days post insemination. Our statistical model showed significance only for the effects of ewe age category and the time interval from heat detection to LAI on the LR attribute. The highest LR (38.8%) was detected in ewes at 2.5–3.5 years of age. Corrected least-square means of LR indicated 18.5 hrs. as an optimal time for LAI of ewes in natural heat. In the present study, the percentage value of lambing rate obtained at 18.5 h interval was 70.7%. Therefore, our study suggested an effective methodology to spread valuable genetic information in the sheep population in the regions of extensive farming where heat cycle synchronization is not usually performed. Importantly, our study is among the first ones that follow the European strategy to eliminate the occurrence of hormones in livestock production and the environment.  相似文献

The objective was to improve pregnancy per artificial insemination (P/AI; 35-42 d after AI) in virgin Jersey heifers bred by AI of sex-sorted semen after being detected in estrus. Giving 100 μg of GnRH at first detection of estrus, with AI 12 h later, did not affect P/AI in Experiment I [GnRH = 47.2% (100/212) vs. No GnRH = 51.7% (104/201); P = 0.38] or Experiment II [GnRH = 53.1% (137/258) vs. No GnRH = 48.6% (122/251); P = 0.43]. In these two experiments, estrus detection was done with tail-head chalk or a HeatWatch® system, respectively. In Experiment III, a single insemination dose (2.1 × 106 sperm) 12 h after estrus detection (n = 193), a double dose at 12 h (n = 193), or a double dose involving insemination 12 and 24 h after estrus detection (n = 190) did not affect P/AI (87/193 = 45.1%, 85/193 = 44.0%, and 94/190 = 49.5%, respectively; P = 0.51). However, P/AI was influenced by the number of AI service (First, 115/208 = 55.3%a; Second, 94/204 = 46.1%a; and Third, 57/165 = 34.8%b; P = 0.004). In Experiment IV, the P/AI of heifers inseminated from 12 to 16 h after the onset of estrus (40/106 = 37.7%) was less (P = 0.03) than those inseminated from 16.1 to 20 h (85/164 = 51.8%), and 20.1 to 24 h (130/234 = 55.6%). However, the P/AI for heifers inseminated from 24.1 to 30 h (61/134 = 45.5%) did not differ from that of any other interval. In conclusion, in Jersey heifers inseminated with sex-sorted semen, P/AI was not significantly affected by giving GnRH at detection of estrus or a double insemination dose, but it was higher with AI 16.1 to 24 h vs. 12 to 16 h after the onset of estrus.  相似文献
