It is generally believed that peasant culture is incompatible with industrial civilization. Furthermore, peasant-city relationships are often dichotomized, suggesting maximal cultural distance. In view of data gathered in a peasant village of the Lower Rhineland it is argued here that over a period of seventy years at least the villagers were able not only to survive within an industrial civilization without losing their cultural identity but also to we industrial wage-earning as a means of preserving the essentials of their peasant way of life. Their ability to adapt to technological change is related here to a long history of previous adaptations beginning in the Middle Ages and leading to the Second Agricultural Revolution. Historical comparison between the city and the village suggests cultural continuity of urban lower class and peasantry rather than cultural polarity between the two segments. This article investigates the complex interplay between religion, gender and marginalization among a group of Pakistani schoolgirls in Hong Kong. It is found that the participants experience multiple marginalization and develop various strategies against disadvantageous positions. It is also found that, while the Pakistani girls attempt to extricate themselves from the gendered practices in their heritage culture and its customs that marginalize and confine women, they simultaneously seek to establish an Islamic or Muslim identity that differentiates them from local girls. It argues that, while mainstream culture has, to a certain extent, released Pakistani girls from the oppression and pressure of religion and customs, they have not been provided sufficient guidance to realize their dreams, making the release temporary and uncertain. The implications of the findings and directions for future research are discussed.  相似文献

The renaissance of Maori culture and tradition has played a significant role in the political campaigns of New Zealand's indigenous population over the past few decades. At the same time, however, it has brought to light that many Maori youngsters are unable to construct a cultural identity in terms of the discourses of culture and tradition that dominate the political arena. This article analyses the experience of urban Maori youngsters in ceremonial settings ( marae ) by examining the question of how they mediate different representations of their cultural identity within the self. It demonstrates that many young Maori people are engaged in a psychological dialogue between, on the one hand, the classic model for a Maori identity that prescribes them to embrace traditional culture and, on the other hand, their personal identification as outcasts in daily practices of New Zealand society.  相似文献

Others, however, stress that in many places villagers assert their right to a local identity. This article discusses how the inhabitants of the Dutch island of Texel have renegotiated and articulated their identity within the context of processes of nation building and state formation in the Netherlands. Race as a mechanism of social stratification and as a form of human identity is a recent concept in human history. Historical records show that neither the idea nor ideologies associated with race existed before the seventeenth century. In the United States, race became the main form of human identity, and it has had a tragic effect on low-status "racial" minorities and on those people who perceive themselves as of "mixed race." We need to research and understand the consequences of race as the premier source of human identity. This paper briefly explores how race became a part of our culture and consciousness and argues that we must disconnect cultural features of identity from biological traits and study how "race" eroded and superseded older forms of human identity. It suggests that "race" ideology is already beginning to disintegrate as a result of twentieth-century changes.  相似文献

This paper is about Australian nationalism in Papua at the beginning of the century. It analyses the role of science and humanitarianism in sustaining the colonial project and the white man's identity. Colonialism was formulated as philanthropy operating on a global scale, where the redemption and salvation of humanity involved the ‘higher cultures’ having a ‘sacred trust’ to educate and morally uplift the natives. This duty to pacify and civilise the tribal other was also a process of reworking the boundaries of western identity so as to remove it from too close a kinship with those forms of savage subjectivity over which it claimed moral superiority. Anthropology became part of the ethical task of how to be a good coloniser; it was heavily involved in reforming state power, such that it governed through culture rather than through violence. Indeed, anthropology was used to legitimise new understandings of native culture as something that could not be overly repressed without destroying the natives. A new internal form of colonialism emerged, one which required cultural pluralism and which was worried about the dangers of over-assimilation.  相似文献

Quebec's struggle between francophones and anglophones has deeply affected other immigrant communities' identitarian practices. Within Montreal's eighties and nineties nationalist environment some Italo-Quebecois worked as a bridge between the two groups through the transcultural magazine Vice Versa. The article presents some findings from a case study that looks at how Vice Versa challenged dominant perceptions of identity and its role in society. It assumes that identity is negotiated between individuals, that language and ethnicity are key factors for social mobility and symbolic power on the marketplace, and that these function through hegemony. Through its use of untranslated French, English and Italian and its articles' contents Vice Versa's discourse on transculture undermines the assumption that identity is necessary for social interaction and negotiates new categories that dissolve ethnic boundaries. Vice Versa is a unique example of the emergence of a counter-hegemonic movement questioning identity at its roots.  相似文献

The black immigrant population in New York City has grown exponentially since 1990, such that West Indians now compose the majority of the black population in several neighbourhoods. This article examines how this ethnic density manifests among youth in high school, and how it has influenced ethnic identity formation among second-generation West Indians. My findings are based on twenty-four interviews and eight months of participant observation in two Brooklyn high schools from 2003 to 2004. The results show that in both schools, Caribbean island identities have become a ‘cool’ commodity within peer groups. Further, although it was important to express pride in one's island identity, these young people often blurred their national origin boundaries by drawing on Jamaican popular culture as way of projecting a unified ‘West Indian identity’. The research also uncovers evidence of a de-stigmatization of Haitianness as a way to incorporate them as cultural insiders into the larger Caribbean collective.  相似文献

Governing by identity describes the emerging regime of a globalizing, mobile world. Governance depends on identification but identification increasingly depends on biometrics. This 'solution' to difficulties of verification is described and some technical weaknesses are discussed. The role of biometrics in classification systems is also considered and is shown to contain possible prejudice in relation to racialized criteria of identity. Lastly, the culture of biometric identification is shown to be limited to abstract data, artificially separated from the lived experience of the body including the orientation to others. It is proposed that creators of national ID systems in particular address these crucial deficiencies in their attempt to provide new modes of verification.  相似文献

This paper explains the development of affiliative ethnic identity: an individual identity rooted in knowledge, regular consumption and deployment of an ethnic culture that is unconnected to an individual's ethnic ancestry until that individual regards herself, and may be regarded by others, as an affiliate of a particular ethnic group. While ethnic culture remains identifiably linked to a particular ethnic ancestry, ideological, institutional and demographic changes have elasticized the link between ancestry and culture, making the formation of affiliative ethnic identity possible. Multiculturalism and its accompanying value of diversity have become institutionalized such that individuals regard ethnic difference as something to be recognized and celebrated. The prevalence of ethnic culture in schools, ethnically infused products of popular culture, demographic changes and growing interethnic contact allow individuals, regardless of ethnic ancestry, ready access to multiple ethnic cultures, providing the basis for the formation of affiliative ethnic identity.  相似文献

In biology, man is an object of research; therefore the question might be asked whether inspirations can go from biological data to the reflections on the mind-matter relation in man. The social aspect of man, as treated by sociobiology, is left out of consideration. The knowledge that man is mind, or has a mind, is no result of biological research. It is a datum from philosophy. The biologist, however, is living in a culture which knows about the mental character of man, and this is incorporated in his investigations. He knows that mental activities are connected with processes in the central nervous system and that, especially in the brain, localizations of mental activities are found. As a result of the split-brain experiments with patients and animals, some have arrived at the conclusion that there is a double consciousness. An approach from biology can lead to statements of a philosophical character, as, for example, statements about the unity, or even identity, of mind and matter. The theories of identity meet with great interest in scientific circles, and the truth value of identity statements is investigated. The system theory is taken into consideration. On a philosophical level a revaluation of the concept of matter can lead to a different sort of identity theory.  相似文献

Aerobic biological ammonia oxidation is carried out by two groups of microorganisms, ammonia-oxidizing bacteria (AOB) and the recently discovered ammonia-oxidizing archaea (AOA). Here we present a study using cultivation-based methods to investigate the differences in growth of three AOA cultures and one AOB culture enriched from freshwater environments. The strain in the enriched AOA culture belong to thaumarchaeal group I.1a, with the strain in one enrichment culture having the highest identity with "Candidatus Nitrosoarchaeum koreensis" and the strains in the other two representing a new genus of AOA. The AOB strain in the enrichment culture was also obtained from freshwater and had the highest identity to AOB from the Nitrosomonas oligotropha group (Nitrosomonas cluster 6a). We investigated the influence of ammonium, oxygen, pH, and light on the growth of AOA and AOB. The growth rates of the AOB increased with increasing ammonium concentrations, while the growth rates of the AOA decreased slightly. Increasing oxygen concentrations led to an increase in the growth rate of the AOB, while the growth rates of AOA were almost oxygen insensitive. Light exposure (white and blue wavelengths) inhibited the growth of AOA completely, and the AOA did not recover when transferred to the dark. AOB were also inhibited by blue light; however, growth recovered immediately after transfer to the dark. Our results show that the tested AOB have a competitive advantage over the tested AOA under most conditions investigated. Further experiments will elucidate the niches of AOA and AOB in more detail.  相似文献

A mixed culture dechlorinating 1,2-dichloroethane (1,2-DCA) to ethene was enriched from groundwater that had been subjected to long-term contamination. In the metagenome of the enrichment, a 7-kb reductive dehalogenase (RD) gene cluster sequence was detected by inverse and direct PCR. The RD gene cluster had four open reading frames (ORF) showing 99% nucleotide identity with pceB, pceC, pceT, and orf1 of Dehalobacter restrictus strain DSMZ 9455(T), a bacterium able to dechlorinate chlorinated ethenes. However, dcaA, the ORF encoding the catalytic subunit, showed only 94% nucleotide and 90% amino acid identity with pceA of strain DSMZ 9455(T). Fifty-three percent of the amino acid differences were localized in two defined regions of the predicted protein. Exposure of the culture to 1,2-DCA and lactate increased the dcaA gene copy number by 2 log units, and under these conditions the dcaA and dcaB genes were actively transcribed. A very similar RD gene cluster with 98% identity in the dcaA gene sequence was identified in Desulfitobacterium dichloroeliminans strain DCA1, the only known isolate that selectively dechlorinates 1,2-DCA but not chlorinated ethenes. The dcaA gene of strain DCA1 possesses the same amino acid motifs as the new dcaA gene. Southern hybridization using total genomic DNA of strain DCA1 with dcaA gene-specific and dcaB- and pceB-targeting probes indicated the presence of two identical or highly similar dehalogenase gene clusters. In conclusion, these data suggest that the newly described RDs are specifically adapted to 1,2-DCA dechlorination.  相似文献

A prerequisite for many studies of neurons in culture is a means of determining their original identity. We needed such a technique to study the interactions in vitro between a class of spinal cord neurons, sympathetic preganglionic neurons, and their normal target, neurons from the sympathetic chain. Here, we describe how we use two highly fluorescent carbocyanine dyes, which differ in color but are otherwise similar, to identify neurons in culture. The long carbon chain carbocyanine dyes we use are lipid-soluble and so become incorporated into the plasma membrane. Neurons can be labeled either retrogradely or during dissociation. Some of the labeled membrane gradually becomes internalized and retains its fluorescence, allowing identification of cells for several weeks in culture. These dyes do not affect the survival, development, or basic physiological properties of neurons and do not spread detectably from labeled to unlabeled neurons. It seems likely that cells become retrogradely labeled mainly by lateral diffusion of dye in the plane of the membrane. If so, carbocyanine dyes may be most useful for retrograde labeling over relatively short distances. An additional feature of carbocyanine labeling is that neuronal processes are brightly fluorescent for the first few days in culture, presumably because dye rapidly diffuses into newly inserted membrane. We have used carbocyanine dyes to identify sympathetic preganglionic neurons in culture. Our results indicate that preganglionic neurons can survive in the absence of their target cells and that several aspects of their differentiation in the absence of target appear normal.  相似文献

In Hong Kong, yumcha as a pervasive form of eating in the public has become an institution which epitomises the diversity and inclusiveness that Hong Kong people think are quintessential to the civility and economic achievement of the metropolis. It stands out particularly as a practice that has come to represent both locally and overseas a Hong Kong culture which has previously been characterised as residual of ‘Chinese tradition’ and ‘Western customs’, and hence is virtually non-existent. This paper looks at the consumption of metropolitaneity and the culturalisation of the heunggongyan or Hong Kong personal identity in the form, content and social relations involved in yumcha. It points to the significance of yumcha as a key to understanding a collective identity which has become more and more apparent as Hong Kong people vehemently try to establish a city with its own culture, first against the colonial grip and then the return of sovereignty to China in 1997.  相似文献

Control of female parthenogenetic apomixis and androsporogenesis of Douglas-fir embryonal initials was studied using an experimental culture system in which changes in growth condition can mediate changes in cell identity and outcomes. This culture system constitutes an artificial sporangium in which myriad culture conditions can be simulated and should be applicable for research on a variety of gymnosperms. In this study, embryonal initials from developing seeds from two Douglas-fir trees were rescued and became reprogrammed for female parthenogenetic apomixis (fPA) and parthenogenetic androsporogenesis (mPA). Female PA was initiated by endomitosis forming a binucleate cell with a diploid egg-equivalent and an apoptotic ventral canal nucleus in an archegonial tube. Egg-equivalent nuclei formed cells (parthenotes) that were discharged into an aqueous culture medium. Parthenotes developed axial tiers atypical of early embryogenesis in seeds. Earlier in the year, androsporangial tubes were parthenogenetically differentiated and released monads, dyads, triads, and tetrads into the culture medium. Spores showed chromosomal aberrations. PA demonstrated a temporal separation in gender expression (dichogamy). Embryonal initials brought forward and by-passed the long juvenile phases normally needed for cells to develop into trees and express reproductive maturity. Expressions of fPA and mPA indicated that the specialized culture flasks served as an artificial sporangium (AS). Awareness is raised for the value of an AS for research in gymnosperm life cycles and as a teaching and research laboratory.  相似文献

Phytases catalyze the release of phosphate from phytic acid. Phytase-producing microorganisms were selected by culturing the soil extracts on agar plates containing phytic acid. Two hundred colonies that exhibited potential phytase activity were selected for further study. The colony showing the highest phytase activity was identified as Aspergillus niger and designated strain 113. The phytase gene from A. niger 113 (phyI1) was isolated, cloned, and characterized. The nucleotide and deduced amino acid sequence identity between phyI1 and phyA from NRRL3135 were 90% and 98%, respectively. The identity between phyI1 and phyA from SK-57 was 89% and 96%. A synthetic phytase gene, phyI1s, was synthesized by successive PCR and transformed into the yeast expression vector carrying a signal peptide that was designed and synthesized using P. pastoris biased codon. For the phytase expression and secretion, the construct was integrated into the genome of P. pastoris by homologous recombination. Over-expressing strains were selected and fermented. It was discovered that ~4.2 g phytase could be purified from one liter of culture fluid. The activity of the resulting phytase was 9.5 U/mg. Due to the heavy glycosylation, the expressed phytase varied in size (120, 95, 85, and 64 kDa), but could be deglycosylated to a homogeneous 64 kDa species. An enzymatic kinetics analysis showed that the phytase had two pH optima (pH 2.0 and pH 5.0) and an optimum temperature of 60 degrees C.  相似文献

An exopolygalacturonase (exo-PGase; EC 3.2.1.82) was found in the culture broth of a Bacillus isolate. The gene encoding the exo-PGase, pehK, was cloned by polymerase chain reaction using mixed primers designed from N-terminal and internal amino acid (aa) sequences of the enzyme (PehK). The determined nucleotide (nt) sequence of pehK revealed a 2940 bp open reading frame (980 aa) that encoded a putative signal sequence (27 aa) and a mature protein (953 aa; 103810 Da). The recombinant enzyme was purified to homogeneity from a culture broth of Bacillus subtilis harboring a pehK-containing plasmid. It had a molecular mass of 105 kDa and a pI value of 5.0. The maximum activity was observed at pH 8 and 55 degrees C in Tris-HCl buffer. The degradation products from polygalacturonic or oligogalacturonic acids were digalacturonic acid, like the exo-PGases, PehX of Erwinia chrysanthemi and PehB of Ralstonia solanacearum. The deduced aa sequence of PehK exhibited moderate homology to those of PehX and PehB with approx. 30% identity for both. High homology was observed in a suitably aligned internal region of the three enzymes (65% identity), and some of the conserved aa residues appeared to form the catalytic core of the enzymes.  相似文献
