In mice X-irradiated with lethal dose (750 r) and mid-lethal dose (550 r) the protective effect of bacterial endotoxin isolated from strains ofSalmonella typhi has been found to be limited to a short period before irradiation and administration of endotoxin in repeated doses did not enhance protection. Application of endotoxin 4 days after X-rays resulted in increase of lethality of irradiation and earlier deaths of experimental animals were observed. Active intravenous and intraperitoneal immunization with endogenous strains ofEscherichia coli isolated from the intestinal flora of mice had a demonstrable protective effect when compared with passive immunization.  相似文献

Summary The genome composition of asymmetric somatic hybrids, obtained by fusion of leaf protoplasts fromLycopersicon esculentum and gamma-irradiated leaf protoplasts fromL. peruvianum, was characterised by Southern blot analysis using 29 restriction fragment length polymorphism markers. Eight low dose hybrids and seven high dose hybrids (irradiation dose 50 Gray and 300 Gray, respectively) were analysed. By densitometric scanning of the autoradiographs, the number of alleles for each locus of the component species was established. In general, elimination of alleles from the irradiatedL. peruvianum donor genome was limited and ranged from 17%–69%. ThreeL. peruvianum loci, located on chromosomes 2, 4 and 7, were present in all asymmetric hybrids, suggesting linkage to the regeneration capacity trait which was used in selecting them. The loss of donor genome was dose-dependent. Low dose hybrids contained more alleles, loci and complete chromosomes fromL. peruvianum than high dose hybrids, whereas the high dose hybrids contained more incomplete chromosomes. In most hybrids someL. esculentum alleles were lost. The possible implications of these results for the use of asymmetric hybrids in plant breeding are discussed.  相似文献

Irradiation of the adult grain mite,Acarus siro L., with substerilizing doses of fast electrons significantly reduced fecundity and egg viability. The number of spermatophores found in the bursa of treated females was also considerably reduced. However, no change in the shape of the spermatophores was observed. It is not known whether the decrease in fecundity and egg viability was the result of the drop in spermatophore production or to the death of sperm cells.Fast electrons had less effect upon, fecundity than the same dose of gamma radiation. However, irradiation by fast electrons had a similar effect on egg viability (expressed in percent), number of spermatophores produced per individual, and the number of adult F1 progeny.Fast-electron doses above 0.4 kGy caused sterility ofAcarus siro.  相似文献

Summary The growth of a transplantable mammary carcinoma in C3H and DBA mice was suppressed in animals immune to Listeria monocytogenes (LM). When tumor cells were mixed with live bacteria at the time of transplantation, experimental LM-immune animals survived a lethal dose of tumor cells. Treatment of non-immune animals with tumor cells plus LM was ineffective. Immunity to LM was therefore a prerequisite of antitumor protection.The destruction of tumor cells was most likely a nonspecific side effect of the immune reaction to LM, since administration of LM at the same site as tumor cells was necessary. Despite this nonspecific mode of tumor suppression, animals which were protected from progressive tumor growth nevertheless developed specific antitumor immunity, being able to resist a secondary challenge by a lethal dose of syngeneic tumor cells. Abstract

Background: Renalase is kidney-derived molecule initially considered as catecholamine-inactivating enzyme. However, recent studies suggest that renalase exerts potent cardio- and nephroprotective actions, not related to its enzymatic activity.

Purpose: To assess renalase level in children with chronic kidney disease (CKD).

Material and methods: Serum renalase, BMI, arterial stiffness, peripheral and central blood pressure, intima-media thickness (IMT), medications, and biochemical parameters were analyzed in 38 children with CKD (12.23?±?4.19?years) (stage G2-5). Control group consisted of 38 healthy children.

Results: In the study group, GFR was 25.74?±?8.94?mL/min/1.73 m2; 6 children were dialyzed; 26 had arterial hypertension. Renalase level was higher in the study group compared to control group (p?<?0.001). In CKD children renalase correlated (p?<?0.05) with BMI Z-score (r?=?–0.36), alfacalcidol dose (r?=?0.41), GFR (r?=?–0.69), hemoglobin (r?=?–0.48), total cholesterol (r?=?0.35), LDL-cholesterol (r?=?0.36), triglycerides (r?=?0.52), phosphate (r?=?0.35), calcium-phosphorus product (r?=?0.35), parathormone (r?=?0.58), and pulse wave velocity Z-score (r?=?0.42). In multivariate analysis GFR (β?=?–0.63, p?<?0.001), triglycerides (β?=?0.59, p?=?0.002), and alfacalcidol dose (β?=?–0.49, p?=?0.010) were determinants of renalase.

Conclusions: In children with CKD there is a strong correlation between renalase level and CKD stage. Furthermore, in these patients renalase does not correlate with blood pressure but may be a marker of arterial stiffness.  相似文献

Dependence of the broth effeot and the phenomenon of mutation frequency decline on dose of the applied UV radiation was investigated in the strainEscherichia coli B/r Hcr+ thy trp. Reversions to Trp+ were followed. The degree of the broth effect and the mutation frequency decline is minimal within the range of UV doses corresponding to a survival of cells lower than 10-1. In connection with the two effects, excision of thymine dimers, initiation of synthesis, synthesis and degradation of DNA were also investigated. It was found that stimulation or inhibition of an inaccurate postreplication repair mechanism, rather than inhibition or stimulation of excision of thymine dimers, are responsible for the broth effect and the mutation frequency decline, respectively.  相似文献

In vivo irradiation of ovaries of Pleurodeles poireti by -rays leads to structural rearrangement of lampbrush chromosomes in late vitellogenic oocytes (stages V and VI). The loops collapse into the chromomeres and the axes condense. Doses between 200 and 2,000 rads have been tested. We observed that such changes were dependent on the irradiation dose though the chronological order of the events was irrespective of the dose. The maximum effect was attained about 10 h after irradiation. The alterations are totally reversible. Over a period of 3 days, chromosomes gradually relax, regenerate loops and recover their normal appearance. In mid vitellogenic oocytes (stages III and IV) lampbrush chromosomes do not undergo radiation induced alterations. It seems that only full-grown oocytes are competent to respond to the ionizing-flow.  相似文献

The effects of long-term daily intake of mercury on its urinary and fecal excretion, whole-body retention, and blood concentration in male rats were observed. The animals were exposed to mercuric chloride labeled with 203Hg via drinking water for 8 weeks (5, 50 and 500 m Hg). 203Hg in urine, feces and blood was quantified. The blood mercury concentration did not keep a linear relationship with the increasing dose. The percentage of the total amount of mercury intake which is excreted by the fecal route in rats exposed to 500 m Hg was significantly lower than in those exposed to 5 and 50 m. The daily dose percentage of mercury excreted in urine increased with dose size. The results show that the absorption fraction of mercury through the gastrointestinal tract (30–40%) was higher than values previously reported.  相似文献

Successful infection by Beauveria bassiana as with all other entomopathogenic fungi, is accomplished only at a high conidial dose while, theoretically, a single conidium should be sufficient. Indeed, this is a major deterrent in its use as a biocontrol agent. High pathogen load for infection is required by organisms which display ‘Allee’ effect. In such organisms, a threshold exists for pathogen dose, below which no infection can be caused. B. bassiana has a semelparous life cycle and, therefore, its infection dynamics are expected to conform to the mass action principle with a linear relationship between dose and successful infection observable as mortality of the insect. Whether the need for a high conidial dose to induce insect mortality by B. bassiana is due to the operation of Allee effect was examined. A sample of 34 isolates was bioassayed on Mylabris pustulata (Coleoptera: Meloidae) at four conidial concentrations. With more than half of the isolates in the sample, the lowest dose tested (104 conidia/insect) did not cause insect mortality. Thus, a threshold pathogen load is required to cause successful infection. In these isolates, the dose–mortality relationship was sigmoid. Allee effect is thus identified in the infection dynamics of B. bassianaM. pustulata system. The isolates that induced mortality at the lowest dose tested are concluded to be highly virulent with a lower threshold dose required for successful infection. With some isolates, at high conidial dose, the infection rate decreased either due to a decrease in the proportion of insects showing mycosis, to the speed of death, or both. Such a response could result from intra scramble competition arising from overload of pathogen at very high dose.  相似文献

The antibacterial activity of hemin onStaphylococcus aureus is described. Hemin binding to bacteria was a rapid process, and each cell accumulated 5×105 to 1×106 molecules within 5 min. Bacterial growth was stopped completely after 30 min from addition of low concentration of hemin (3–10 g/ml). Cell viability was reduced by 99.9% in 1 h of exposure, and the effect was consistent at any stage of the growth curve whenever hemin was added. Glucose utilization was arrested immediately after hemin addition, and no CO2 was produced. The survivors of hemin treatment regrow in a time-related kinetics depending on the dose of hemin to which the cells were exposed. The recovered bacteria were again sensitive to hemin, similar to an untreated culture. We suggested that the recovery phenomenon is a result of an on-off mechanism regulating sensitivity to hemin, rather than a selection mechanism giving rise to hemin-resistant mutants.  相似文献

Our method of real-time monitoring of dopamine release from rat striatal slices revealed that endothelin (ET)-3-induced dopamine release was inhibited by N G-methyl-L-arginine (L-NMMA; 1 mM), an inhibitor of nitric oxide (NO) synthase, while N G-methyl-D-arginine (D-NMMA; 1 mM), an inactive isomer of L-NMMA, had no effect.2. The inhibition of L-NMMA (0.1 mM) became apparent when tissues were pretreated with tetrodotoxin (1 M) for 30 min and subsequently exposed to ET-3 (4 M).3. L-NMMA (0.1 and 1 mM) dose dependently protected against ET-3-triggered hypoxic/hypoglycemic impairment of striatal responses to high K+.4. Thus, NO may work as a promoter in mediation of the stimulatory and neurotoxic action of ET-3 on the striatal dopaminergic system, presumably by interacting with interneurons in the striatum.  相似文献

The antibody response to a threshold dose (10) of SE was studied in the High responder line (H) and the Low responder line (L) of mice obtained by bidirectional selective breeding for the character quantitative agglutinin response to an optimal dose of SE, and in interline hybrids: F1, F1 and both backcrosses. Whereas the interline difference in agglutinin responses at the optimal dose is due to the additive effect of about ten independently segregating loci, one of which isH-2 linked, the responsiveness to the threshold dose is determined by the effect of two loci. The direction of the dominance effect also varies with the antigen dose: high responsiveness is partially dominant at the optimal dose while at the threshold dose nonresponder character is partially dominant. The role of theH-2 linked locus was investigated. It has been demonstrated that on an identical background (equivalent to that of F1 hybrids) this locus is responsible for 12% of the interline difference at the optimal antigen dose, and for 61% at the threshold antigen dose. For the two antigen doses, the quantitative effect of theH-2 locus is in agreement with the estimate of the number of loci obtained by variance analysis. The intervention of a second gene, non-H-2 linked, in the regulation of responsiveness to 106 SE is demonstrated by appropriate assortative matings. Septic rats (as induced by cecal puncture and ligation) showed an increased rate ofin vivo leucine oxidation as measured from the formation of14CO2 from an intravenously injected [1-14C]leucine tracer dose. Acute lipopolysaccharide (LPS) administration (1 mg/kg) to rats caused a similar effect on the rate ofin vivo leucine oxidation. Additionally, both tumour necrosis factor- (TNF) and interleukin-1- (IL-1), in an acute dose of 100 g/kg, also increased the rate of the oxidation of the amino acid, although only IL-1 caused a similar increase to that observed following LPS. The observed increased leucine oxidation was related to lower leucine concentrations both in LPS- and cytokine-treated rats. Important decreases were also observed in the other branched-chain amino acids (valine and isoleucine) in the LPS- and IL-1-treated animals. Isolated incubated muscles from TNF- and IL-1-treated rats did not show any changes in the rate of leucine utilization, thus suggesting that the mechanism by which the cytokines stimulate whole-body leucine oxidation is not based on an increase in the activity of the enzymatic machinery responsible for leucine oxidation. Additionally, glucocorticoids do not seem to mediate the enhancedin vivo oxidation of the amino acid since, although they are increased by both LPS and cytokines, treatment of the animals with RU486 (a glucocorticoid antagonist) was not able to suppress the effects of the cytokine onin vivo leucine oxidation.  相似文献

Summary The administration of a single subcutaneous dose of clenbuterol to rats altered the level of taurine in certain tissues. Taurine levels in cardiac tissue were significantly decreased 3 h after the administration of 250g/kg of clenbuterol and remained significantly depressed at 12h post-dose only returning to control values by 24h. The level of taurine in the liver increased 3 h after clenbuterol administration but was lower than the control value at 24 h post dose. Lung taurine levels were significantly lower than the control value at 12 hr post dose and remained depressed until 24h post dose. Clenbuterol caused a significant increase in taurine levels in serum and muscle at 3 and 6 hr postdosing respectively but not at other time points. Serum creatine kinase (CK), activity was slightly but significantly raised at the 12 and 24 h time point.The effects of clenbuterol on tissue taurine content were not dose-dependent over the range studied (63–500g/kg). However taurine levels in the lung were significantly reduced at all doses and in the heart were significantly lower in the treated groups at all except the lowest dose, 12h post dosing. Liver taurine levels were significantly increased at the highest dose of 500g/kg.The reduction of taurine concentrations in the heart, caused by clenbuterol, is of concern as taurine has been shown to have protective properties in many tissues especially the heart.  相似文献

The quantities linear energy transfer or restricted linear energy transfer are utilized in calculations that link absorbed dose to the fluence distribution of a radiation field. The computations provide approximations to absorbed dose in terms of the intermediate quantity cema or reduced cema. With the definition of the restricted linear energy transfer,L , given in ICRU Report 33, the approximation remains imperfect. This study deals with the resulting need for a modified definition ofL , as proposed in a draft report of ICRU. Essential differences between the old and the new definitions are demonstrated. The changed definition permits a rigorous formulation of the dependence between fluence and absorbed dose.  相似文献

Summary The effect of cyclopropene fatty acids, sterculic and malvalic, on the lipids of yeasts grown under nitrogen limiting, lipid accumulating, conditions was studied. The ratio of stearic to oleic acid showed a dose response effect, with an increase in stearic acid content as the dose of cyclopropene fatty acid increased, and a corresponding reduction in oleic acid. Linoleic and linolenic acids were not affected to the same extent. These effects are shown for the yeasts Candida sp. 107, Trichosporon cutaneum, and Rhodosporidium toruloides.  相似文献

研究了致命鹅膏中的毒素在安全剂量下抑制小鼠皮肤肿瘤的效果,并对此毒素的药用安全性进行初步评价。采用经典的DMBA/巴豆油诱导小鼠皮肤乳头状瘤形成的二阶段致癌模型,观察和评价致命鹅膏毒素在不同剂量下抑制肿瘤的效果,并且通过测定血清中氨基己糖的含量来判断对肝损伤的情况。结果表明,致命鹅膏毒素在某一低剂量下对DMBA/巴豆油诱导小鼠皮肤乳头状瘤有较好的治愈效果,且对小鼠肝脏无损伤,证明了致命鹅膏毒素可以较好地治疗皮肤癌。  相似文献

Objective: The administration of the growth hormone (GH) secretagogue GH‐releasing peptide (GHRP)‐2, like ghrelin, increases food intake (FI) in lean healthy men. The aim of this study was to investigate whether this effect occurs in obese subjects and whether it is dose‐dependent. Research Methods and Procedures: Nineteen subjects (10 lean and nine obese), all healthy and weight stable, received a double‐blind randomized subcutaneous infusion of GHRP‐2 at high dose (HD; 1 μg/kg per hour), low dose (0.1 μg/kg per hour), or placebo for 270 minutes over three study visits. Blood for hormone assays was collected through an intravenous forearm catheter. Hunger and fullness were rated on visual analog scales before and after a fixed breakfast (320 kcal at 120 minutes) and a buffet lunch at 240 minutes. Before lunch, subjects received taped instructions to eat as much as they wanted. Results: GHRP‐2 infusion significantly increased ad libitum FI in a dose‐dependent manner by 10.2 ± 3.9% at low dose (p = 0.011) and by 33.5 ± 5.8% at HD (p = 0.000) compared with placebo. Obesity status did not influence the effect of GHRP‐2 on FI. All subjects had greater ratings of appetite before but similar levels of fullness after the meal with the HD GHRP‐2. Serum GH levels increased dose dependently in all subjects. Discussion: The dual stimulatory effect of GHRP‐2 on FI and human GH is dose dependent. Obese individuals retain their ability to respond to GHRP‐2 both in terms of FI and human GH.  相似文献

Illus. 1960.—The effects of x rays on the multiplication rates (MR), percent flowering (Fl%) and gross morphology of aseptically grown cultures of Lemna perpusilla, strain 6746, were studied. MR values are unaffected by doses of less than 300 r but are progressively depressed as the dose increases. The effect of a given dose on short-term MR values is the same at the 2 dosage intensities used, 500 and 2000 r/min. The inhibitory effect of intermediate doses on long-term growth is delayed; high doses have an “immediate” effect. Bleaching of fronds and cessation of growth occur in all cultures given 5,000 r or higher. Radiosensitivity is not altered by addition of yeast extract and casein hydrolysate to the medium. Depressed Fl% and seed production appear to be consequences of lowered MR. Morphological aberrations such as small size, epinasty and wrinkling are temporary. Frond-sequence may be altered by x rays but this aberration is permanent. Various aspects of the results are discussed.  相似文献

Summary The effects of a single high dose (10mg/kg) of vinblastine (vb) sulfate (Velbe, Lilly) on the ultrastructure, catecholamine (CA) content and activity of CA-synthesizing enzymes of the rat adrenal medulla were studied for up to 120h after intravenous injection of the drug.By 1 h, microtubules were virtually absent from chromaffin cells and preganglionic cholinergic axons, and typical paracrystals had appeared inside the nerve fibers. By 16h microtubules were completely reconstituted and paracrystals had disappeared. From 16h onwards, there was an increasing depletion of storage granules from adrenaline (A) — producing cells, which coincided with biochemical determinations showing a reduction of adrenal A to about 40 % of control levels by 48 h, with noradrenaline (NA) remaining in the range of controls. Both A- and NA-storing cells showed an extensive proliferation of the rough endoplasmic reticulum (ER). Vb caused a marked increase in tyrosine hydroxylase (TH; +113%) and dopamine -hydroxylase (DBH; +82%) activities after 48 h. Splanchnicotomy completely abolished the vb-mediated increase in TH and DBH activities. A smaller increase (+ 47 %) in enzyme activity was observed with phenylethanolamine N-methyltransferase (PNMT). Vb (10–5M) had no apparent effect on granule content and the amount of rough ER in chromaffin cells, which were cultured for 48 h.The results demonstrate that a single high dose of vb has relatively little short-term effects on the rat adrenal medulla, but causes drastic long-term changes in CA-content and enzyme activities that are mediated by the preganglionic nerves. These changes could be interpreted as an effort to compensate for a loss of CA-stores in peripheral adrenergic nerves (cf. Cheney et al., 1973). The differential long-term effect of vb on adrenal NA and A might be due to the lower induction of PNMT as compared to TH and DBH activities and/or to a preferential release of A versus NA, which may occur at high frequencies of stimulation of the splanchnic nerves.Supported by grants from the Deutsche ForschungsgemeinschaftDedicated to Professor G.
