Firstly, this paper analyzes the dynamic between horse, rider, and landscape in foxhunting culture. Secondly, it considers nonhuman animals as active elements within the research process. The methodology principally involved extensive, in-depth participation as a rider within mounted foxhound packs, as I originally participated as a rider/ethnographer in order to understand the nexus between foxhunting culture and the landscape. However, the equine focus in my research emerged unexpectedly as my fieldwork drew me into a collaboration with an unanticipated character in this network; the “made hunter,” a horse seasoned for hunting. These animals acted as my equine gatekeepers literally incorporating me into this hunting world. I relied on and respected their experience and capabilities as novice and experienced foxhunters have done for centuries. This paper presents an analysis of the quadripartite synthesis of foxhunting culture, rider, horse, and landscape and involves a corporeal and sensory exploration of the human/nonhuman inter-Hunt dynamic that defines foxhunting. Hunting cultures rely upon the capabilities of animals and have developed an ability to translate animal narratives and semiotics. Thus, they provide us with a platform for developing the multi-species perspective in social science. This paper calls for an ethnographic approach which draws upon these epistemic systems and incorporates the superhuman abilities of animals into our understanding of our relationship with space. Animals, I argue, offer more than potential subjects to study. By incorporating their knowledge, experiences, and perspectives, and by translating their engagements with the world, we have the potential to augment our perception of our shared environment.  相似文献

Venezuelan equine encephalitis (VEE) virus was purified and concentrated by chromatography of tissue culture supernatant fluids on diethylaminoethyl-cellulose columns. Stepwise gradient elution studies indicated a broad elution pattern for the virus, with recovery occurring from 0.05 to 0.7 m NaCl. Optical density, infectivity, hemagglutination (HA), and complement fixation (CF) assays indicated that complete recovery of input virus in highly purified form was possible. Single-step elution with 0.7 m tris(hydroxymethyl)aminomethane-succinate-salt buffer resulted in a virus volume decrease of 85% with a concomitant increase in infectivity and antigenicity. Recoveries consistently equaled or exceeded 100% of the input preparations. Additional purification of column-recovered virus was obtained by sedimentation of pooled virus eluates on 50% sucrose cushions. Exposure of borate saline and 0.5% histidine suspensions of purified VEE virus preparations to 6 x 10(6) r of gamma radiation resulted in a loss of infectivity for tissue culture and a loss of lethality for weanling and suckling mice. Inactivation was an exponential function of the dosage. In contrast to infectivity, antigencity (HA and CF) of both saline and histidine preparations was retained after irradiation with doses as high as 6 x 10(6) r. Purified and irradiated VEE virus preparations have been successfully used for routine serological tests and are being evaluated as vaccines.  相似文献

Three viral proteins were separated from the TC-83 strain of Venezuelan equine encephalomyelitis virus by discontinuous polyacrylamide gel electrophoresis after disruption with sodium dodecyl sulfate and beta-2-mercaptoethanol. These proteins were inoculated into rabbits and the resultant antisera were tested for immunological activity by gel precipitation, plaque reduction neutralization, hemagglutination inhibition (HI), complement fixation, fluorescence microscopy, and mouse protection studies. All proteins were capable of stimulating precipitating antibody in rabbits, but the largest protein (VP 1), which is contained in the envelope, stimulated the production of detectable neutralizing and HI antibody against the intact virion. The other two proteins yielded little or no neutralizing or HI antibody.  相似文献

Calcium-binding equine lysozyme (EL) combines the structural and folding properties of c-type lysozymes and alpha-lactalbumins, connecting these two most studied subfamilies. The structural insight into its native and partially folded states is particularly illuminating in revealing the general principles of protein folding, amyloid formation and its inhibition. Among lysozymes EL forms one of the most stable molten globules and shows the most uncooperative refolding kinetics. Its partially-folded states serve as precursors for calcium-dependent self-assembly into ring-shaped and linear amyloids. The innate amyloid cytotoxicity of the ubiquitous lysozyme highlights the universality of this phenomenon and necessitates stringent measures for its prevention.  相似文献

普氏野马(Equus przewalskii)和野驴(Equus hemionus)是许家窑遗址动物群中的优势属种。本文基于对这两种动物牙齿材料的测量与分析,确定了遗址中马科动物的死亡年龄,并对上、下文化层的死亡年龄分布进行了研究,以期探知古人类获取肉食资源的方式与特点。通过与马科动物在自然生存状态下以及死于不同原因(如疾病或营养衰竭、食肉动物猎杀、现代人类狩猎等)的年龄结构对比,结果表明:古人类在许家窑文化早期(下文化层)可能通过捡拾自然死亡的动物尸体、与食肉类动物抢夺猎物、主动狩猎等多种方式获取马科动物,而在许家窑文化晚期(上文化层)可能以主动狩猎作为获取马科动物的主要方式。此外,古人类在遗址的早期就可能已经具有捕获整个马科动物居群中任意年龄个体的能力,并能做出最优化判断,有选择地去捕猎脂肪和肉量较高的壮年动物群体。  相似文献

Cytogenetic analyses of horses have benefited the horse industry by identifying chromosomal aberrations causing congenital abnormalities, embryonic loss and infertility. Technical advances in cytogenetics enabled the identification of chromosome specific aberrations. More recently, advances in genomic tools have been used to more precisely define chromosome abnormalities. In this report we review the history of equine clinical cytogenetics, identify historical landmarks for equine clinical cytogenetics, discuss how the current use of genomic tools has benefited this area, and how future genomics tools may enhance clinical cytogenetic studies in the horse.  相似文献

Equine arteritis virus (EAV) is a small, enveloped, positive-stranded RNA virus, in the family Arteriviridae , W.H.ich can infect both horses and donkeys. While the majority of EAV infections are asymptomatic, acutely infected animals may develop a wide range of clinical signs, including pyrexia, limb and ventral edema, depression, rhinitis, and conjunctivitis. The virus may cause abortion and has caused mortality in neonates. After natural EAV infection, most horses develop a solid, long-term immunity to the disease. Marzz and geldings eliminate the virus within 60 days, but 30 to 60% of acutely infected stallions will become persistently infected. These persistently infected animals maintain EAV within the reproductive tract, shed virus continuously in the semen, and can transmit the virus venereally. Mares infected venereally may not have clinical signs, but they shed large amounts of virus in nasopharyngeal secretions and in urine, which may result in lateral spread of the infection by an aerosol route. The consequences of venereally acquired infection are minimal, with no known effects on conception rate, but mares infected at a late stages of gestation may abort. Identification of carrier stallions is crucial to control the dissemination of EAV. The stallions can be identified by serological screening using a virus neutralization (VN) test. If positive at a titer of >/= 1:4, the stallion should be tested for persistent infection by virus isolation from the sperm-rich fraction of the ejaculate, or by test mating Shedding stallions should not be used for breeding, or should be bred only to mares seropositive from a natural infection or from vaccination, the mares should be subsequently isolated from seronegative horses for three weeks after natural or artificial insemination. A live attenuated (ARVAC) and a formalin-inactivated (ARTERVAC) vaccine are available. Both vaccines induce virus-neutralizing antibodies, the presence of which correlates with protection from disease, abortion, and the development of a persistent infection. Serological investigations indicate that EAV has a worldwide distribution and that its prevalence is increasing. As a consequence, an increasing number of equine viral arteritis (EVA) outbreaks is being reported. This trend is likely to continue unless action is taken to slow or halt the transmission of this agent through semen.  相似文献

Scientists and equestrians continually seek to achieve a clearer understanding of equine learning behaviour and its implications for training. Behavioural and learning processes in the horse are likely to influence not only equine athletic success but also the usefulness of the horse as a domesticated species. However given the status and commercial importance of the animal, equine learning behaviour has received only limited investigation. Indeed most experimental studies on equine cognitive function to date have addressed behaviour, learning and conceptualization processes at a moderately basic cognitive level compared to studies in other species. It is however, likely that the horses with the greatest ability to learn and form/understand concepts are those, which are better equipped to succeed in terms of the human-horse relationship and the contemporary training environment. Within equitation generally, interpretation of the behavioural processes and training of the desired responses in the horse are normally attempted using negative reinforcement strategies. On the other hand, experimental designs to actually induce and/or measure equine learning rely almost exclusively on primary positive reinforcement regimes. Employing two such different approaches may complicate interpretation and lead to difficulties in identifying problematic or undesirable behaviours in the horse. The visual system provides the horse with direct access to immediate environmental stimuli that affect behaviour but vision in the horse is of yet not fully investigated or understood. Further investigations of the equine visual system will benefit our understanding of equine perception, cognitive function and the subsequent link with learning and training. More detailed comparative investigations of feral or free-ranging and domestic horses may provide useful evidence of attention, stress and motivational issues affecting behavioural and learning processes in the horse. The challenge for scientists is, as always, to design and commission experiments that will investigate and provide insight into these processes in a manner that withstands scientific scrutiny.  相似文献

Some retroviruses contain monoubiquitinated Gag and do not bud efficiently from cells treated with proteasome inhibitors, suggesting an interaction between the ubiquitin-proteasome system and retrovirus assembly. We examined equine infectious anemia virus (EIAV) particles and found that approximately 2% of the p9(Gag) proteins are monoubiquitinated, demonstrating that this Gag protein interacts with an ubiquitinating activity. Different types of proteasome inhibitors were used to determine if proteasome inactivation affects EIAV release from chronically infected cells. Pulse-chase immunoprecipitation and time course immunoblot analyses showed that proteasome inactivation slightly decreased virus release (at most a twofold effect), while it did not affect Gag processing. These results contrast with those obtained with other viruses which are sensitive to these inhibitors. This suggests that, although its Gag is monoubiquitinated, the requirements for EIAV release are somewhat different from those for retroviruses that are sensitive to proteasome inhibitors.  相似文献
