With the ultimate purpose of clarifying the mechanism for aluminium (Al) toxicity and for Al tolerance, we tried to isolate cDNAs whose expression is induced by Al treatment and phosphate (Pi) starvation. We performed Pi starvation and Al treatment (two-step treatment) on suspension-cultured cells of Nicotiana tabacum L. cv. Samsun and then constructed a cDNA library using poly(A)+-RNA derived from the treated cells. Four independent cDNA clones (pAL 111, 139, 141 and 142) were isolated from the library by differential screening. Northern blot hybridization analysis indicated that the expression of these clones was induced by Pi starvation. Furthermore, we found that pAL 111 and pAL 142 are also induced by Al treatment. The complete cDNA sequencing of these 4 clones was determined. The results indicated that pAL111 is identical to the parA gene of N. tabacum, which is described as an auxin-regulated gene and that pAL142 is highly homologous to the parB gene of N. tabacum whose product has glutathione S-transferase (GST, EC 2.5.1.18) activity. Furthermore, we found a cysteine-rich domain in the amino acid sequence of pAL139. No DNA and deduced amino acid sequences homologous to the pAL141 were found.  相似文献

In vitro, animal, and human experiments have shown that boron is a bioactive element in nutritional amounts that beneficially affects bone growth and central nervous system function, alleviates arthritic symptoms, facilitates hormone action and is associated with a reduced risk for some types of cancer. The diverse effects of boron suggest that it influences the formation and/or activity of substances that are involved in numerous biochemical processes. Several findings suggest that this influence is through the formation of boroesters in biomolecules containing cis-hydroxyl groups. These biomolecules include those that contain ribose (e.g., S-adenosylmethionine, diadenosine phosphates, and nicotinamide adenine dinucleotide). In addition, boron may form boroester complexes with phosphoinositides, glycoproteins, and glycolipids that affect cell membrane integrity and function. Both animal and human data indicate that an intake of less than 1.0 mg/day inhibits the health benefits of boron. Dietary surveys indicate such an intake is not rare. Thus, increasing boron intake by consuming a diet rich in fruits, vegetables, nuts and pulses should be recognized as a reasonable dietary recommendation to enhance health and well-being.  相似文献

The storage and distribution of copper, cadmium, and uranium and their effects on ionic contents in roots and shoots of Azolla filiculoides has been studied by x-ray microanalysis. The relative content of copper was eightfold higher in the root than in the shoot, suggesting low mobility of this metal in Azolla plant. Cadmium relative content in the shoot was similar to its content in the root, hence its mobility was relatively high. The absence of significant uranium quantities in the shoot and its relative high content in the root suggest the immobility of this metal from Azolla root. Cadmium formed precipitates with phosphate and calcium in xylem cells of the shoot bundle and caused a two- to threefold increase in the content of phosphate in the root. Uranium in roots and cadmium in shoots were associated with calcium. All three treatments caused losses of potassium, chloride, and magnesium from Azolla roots. Accumulation of heavy metals in Azolla and their mobility from the root to the shoot can be correlated with damage caused by the loss of essential nutrients.  相似文献

The effects of lead toxicity on leaf gas exchange, chlorophyll content, chlorophyll fluorescence, chloroplast ultrastructure, and opening of stomata were examined in tobacco (Nicotiana tabacum L.) plants. Plants were grown in a growth chamber for 7 d in Hoagland nutrient solution supplemented with 0.0 (control), 5, 10, 25, 50, 100, 300 and 500 μM Pb(NO3)2. Plants treated with 5, 10, and 25 μM Pb(NO3)2 exhibited no abnormalities. Root and shoot growth, net photosynthetic rate and stomatal conductance were significantly reduced in plants treated with 100, 300 and 500 μM Pb(NO3)2. In plants treated with 500 μM Pb(NO3)2, the majority of stomata were closed. The effect of Pb(NO3)2 on chlorophyll content and chlorophyll fluorescence parameters was negligible. However, in plants exposed to 100, 300, and 500 μM Pb(NO3)2, the mesophyll cells showed altered chloroplasts with disrupted thylakoid membranes.  相似文献

Ginkgo biloba L. pollen-derived tissue, which is made up of small, friable masses of homogeneous parenchymatous cells, was shown to require boron in the culture medium. If no boron is supplied, growth soon stops. Growth responses to additions of boron were observed up to an optimum level of 0.1 mg of boron per liter.

Histological examination and chemical analyses showed 2 general effects of boron deficiency: (1) a reduced rate of cell division, with no significant effect on cell size, and (2) some alteration in composition of the cell walls. With the exception of a reduction in fructose, the concentration of soluble and of readily hydrolyzable carbohydrates, and the concentration of protein in the tissue, were not affected by boron deficiency.

Spontaneously hypertensive rats (SHR) were separated into two groups (n = 6 per group) and, since 5 months old, received alpha-tocopherol (alpha-tocopherol acetate120 IU) or vehicle by daily gavage for 2 weeks. Blood viscosity, blood pressure (BP) and myocardial remodeling were analyzed. The SHRs treated with alpha-tocopherol showed a significant reduction of BP and a major reduction of blood viscosity in comparison with the control SHRs. The cardiac hypertrophy indices showed some differences when the two SHR groups were compared, the LV mass index was not different between the groups; however, the cardiomyocyte size was more than 20% smaller in SHRs treated with alpha-tocopherol than in control SHRs (P < .05). The intramyocardial vessels distribution was more than 45% greater in alpha-tocopherol-treated SHRs than in control rats, significantly improving the vessels-to-myocytes ratio in treated SHRs than in control SHRs (P < .05). In conclusion, present findings strongly suggest a beneficial effect of alpha-tocopherol supplementation to genetically hypertensive rats. This was observed by a reduction of both blood viscosity and BP, and a consequent cardiomyocyte hypertrophy in treated SHRs; an improvement of vessels-to-myocytes ratio in these rats was also observed.  相似文献

Toxic solutions applied at one end of a Nitella cell 6 cm. long may produce little or no visible change in the structure of the protoplasm at the place of application but if the opposite end is covered with water its protoplasm soon disintegrates. If the middle of the cell is covered with mineral oil this region remains normal in appearance for half an hour or more. The result is due to the movement of substances in the cell. The loss of substances at the end where the toxic agent is applied results in loss at the opposite end if it is covered with water since water enters and travels along inside the cell carrying substances with it. This causes injury at the spot where the water enters. The conception developed here differs fundamentally from the usual view that the effects of injury spread gradually from the region where the toxic agent is applied to the immediately adjoining regions and thence to more remote places. The change produced by loss of substances produces an interesting pattern which deserves study.  相似文献

Metals and metal compounds are constituents of our natural environment. Their distribution depends on the existence of natural sources (e.g. volcanoes or erosion) and their use in human’s activity. They are transformed naturally (e.g. by bacterial activity) with formation of organic species that influence their mobility and accumulation in abiotic as well as biotic systems. Up to date metal species are released into the environment questioning their influence on human health. Due to their widespread use in human activities such as industry, agriculture and even as medicine (e.g. As, Se, Pt), numerous health risks may be associated with exposure to these substances. Different reports on metal intoxication are documented and studies especially on neurotoxicity, genotoxicity, or carcinogenicity, are previously published in numerous articles. Stem applications of gibberellic acid (GA) to debudded tobacco plants (Nicotiona tabacum L., var. One Sucker) produce stem swellings that involve intense proliferation of, primarily, xylem tissue. Withholding boron from GA-treated debudded plants greatly reduces the GA-induced proliferation. This response offers a system for directly demonstrating the effect of boron on xylem formation and lignification that is unconfounded by the effect of boron on elongation growth.  相似文献

The effects of boron (B) deficiency on several phenolics and enzyme activities involved in the biosynthesis of these compounds were investigated in tobacco plants (Nicotiana tabacum L. cv. Gatersleben). The levels of phenylpropanoids (mainly the caffeic acid esters, chlorogenic acid and its isomers) as well as phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) and polyphenoloxidase (PPO, EC 1.14.18.1) activities were determined in plants subjected to B starvation for 1–7 d. The results presented here show that a short-term B deficiency causes both quantitative and qualitative changes in the phenolic metabolism of tobacco plants, which are especially evident after 3 d of B starvation. Although the concentration of B decreased from the onset of B starvation, root B level was less affected than leaf B by a short-term B deficiency. The concentration of phenylpropanoids as well as PAL and PPO activities increased mainly in the leaves of tobacco plants during B starvation. Moreover, leaves starved of B for 7 d showed the accumulation of new compounds, one of which was identified as caffeoylputrescine. In addition, a positive correlation between PAL activity and phenylpropanoid concentration was observed in tobacco leaves, especially after 5–7 d of B starvation, suggesting that an increase in PAL activity during B starvation could be responsible for the enhancement in the levels of phenylpropanoids.  相似文献

ABSTRACT. The toxicity and biological activity of the defensive secretions of ten species of neotropical termites were examined by bioassay on two species of predatory ant. Pseudomyrmex termitarius (F. Smith) (Pseudomyrmecinae) was more susceptible to the effects of the secretions than Ectatomma sp. (Ponerinae). The bioassay results show a progression in the nature of termite chemical weaponry and in the potency of defensive secretions. The tacky secretions used by Coptotermes and Ruptitermes are simply adhesive, entangling agents with no apparent toxic or pheromonal properties. The secretion of Syntermes calvus Emerson serves as a wound-active poison with little topical activity, while those of Armitermes spp. are repellent to ants and show activity as topical poisons. The secretions used by Nasutitermes spp. combine adhesive, entangling properties with the toxicity of a topically-active poison, and are effective also as ant-repellents.  相似文献

Shoot and root mass of tobacco plants treated with only 0.05 μM boron was decreased by 25 and 50 %, respectively, when compared to plants sufficiently supplied with B (2 and 5 μM). Leaf B content of 0.05 μM B-treated plants decreased (about 80–90 %) when compared to 2 μM B treated plants; this drop of B content were not as marked (about 25–45 %) in roots. Leaf and root nitrate contents in B-deficient plants were 45–60 % and 35–45 % lower, respectively, than those from 2 and 5 μM B treated plants. It is suggested that B deficiency might decrease nitrate uptake rather than nitrate reductase activity in tobacco plants.  相似文献

用全血细胞分析仪检测染毒小鼠新鲜血样,再用日立7020生化分析仪,配合试剂盒法对血清进行检测,研究铅对血液与肝肾功能的影响,结果表明:100mg/kg浓度的醋酸铅对小鼠的血液有显著影响,白细胞数目(WBC),淋巴细胞数目(Lymph),中性粒细胞数目(Gran)等指标显著增加(P〈0.05),此外对血小板凝血系统功能显著影响,血小板数目(PLT)等指标下降极显著(P〈0.01),对血红细胞(RBC),血红蛋白(HGB)等指标影响不显著(P〉0.05)。肝肾功能性指标:谷丙转氨酶(AIJT),谷草转氨酶(AST)尿素氮(UREA)肌酐(CREA)生化酶显著偏离正常值(P〈0.05)。表明铅对肝脏肾脏功能有显著的破坏。对检测结果做了进一步的分析讨论。  相似文献

In this study we investigated the effects of boric acid and sodium tetraborate on an acute leukemia cell line and healthy human lymphocytes. We evaluated the effects of boric acid and sodium tetraborate on the HL-60 cell line and healthy human lymphocytes by using the methods of MTT, Neutral Red, AO (flow cytometry) and transmission electron microscope. We found that there were dead cells at a concentration of 500 µM boric acid and sodium tetraborate (50 % and 40 %, respectively). An apoptotic effect was found at a concentration of 1,000 µM concentration in normal lymphocytes and HL-60 (acute leukemia cells) cells (2.5 % and 8.8 % respectively). We observed that boric acid at a concentration of 500 µM caused double nucleus and micronucleus formation in both HL-60 cells and lymphocytes. An expansion in mitochondrial dimension and deformation in cristas also appeared. Our findings suggest that boric acid is more effective than sodium tetraborate on the HL-60, and boric acid in particular showed a cytotoxic effect on HL-60 in comparison to healthy lymphocytes and it also affected the mitochondrial pathway.  相似文献
