Primary roots of soybean [Glycine max (L.), cv Harosoy 63] seedlings were inoculated with zoospores from either race 1 (incompatible, host resistant) or race 3 (compatible, host susceptible) of Phytophthora megasperma f. sp. glycinea (Pmg) and the activities of phenylalanine ammonia-lyase (PAL), chalcone synthase (CHS), isoflavone synthase, and dihydroxypterocarpan 6a-hydroxylase related to phytoalexin (glyceollin) biosynthesis, and of glucose-6-phosphate dehydrogenase (Glc-6-PDH) and glutamate dehydrogenase (Glu-DH) were determined at various times after inoculation. About 2-4 h after inoculation with race 1, the activities of PAL, CHS, and pterocarpan 6a-hydroxylase were higher than after inoculation with race 3 and increased considerably thereafter. In contrast, activities of these enzymes in the compatible interaction were equal to or only slightly higher than in the controls over the entire infection period investigated (2-8 h). Isoflavone synthase did not increase until 7 h after inoculation with race 1. There were no significant differences in activities for Glc-6-PDH and Glu-DH between inoculated roots and controls. The results show that infection of soybean roots with zoospores of Pmg race 1 causes a race:cultivar-specific early induction of enzymes involved in glyceollin synthesis, whereas such an induction does not occur with zoospores of race 3. These findings are in agreement with the race:cultivar-specific accumulation of glyceollin in soybean roots reported previously [M. G. Hahn, A. Bonhoff, and H. 小麦初生叶接种条锈菌毒性生理小种(CY29)及其弱毒突变菌系(CY29-mut3)后,呈不亲和反应的寄主叶片可溶性蛋白质合成能力在接种后24h显著高于未接种对照,但其后逐渐降低,直至接近对照;而呈亲和性反应的寄主叶片可溶性蛋白质合成在侵染早期与对照相近,但与膜结合蛋白质在96h时大大高于对照。对接种叶中核糖体的密度梯度分析证实:呈不亲和反应寄主叶片游离多聚核糖体及亲和反应的寄主内与膜结合多聚核糖体均有特异性增加。上述结果表明寄主的抗病和感病反应均与蛋白质合成能力的变化有关。  相似文献

Immersion of roots of 2-day-old soybean seedlings (Glycine max cv. Harosoy 63) into solutions of several glucan elicitors caused the accumulation to various degrees of the soybean phytoalexin glyceollin. Laminarin and polytran proved to be more effective elicitors in this system than the glucan elicitor from Phytophthora megasperma f.sp. glycinea (Pmg). Digitonin and tomatin caused, in addition to glyceollin accumulation, the deposition of callose in the rhizodermis. Pretreatment of the soybean roots with laminarin effected an increase in resistance of the seedlings against a compatible race of Pmg.  相似文献

The differential regulation of the activities and amounts of mRNAs for two enzymes involved in isoflavonoid phytoalexin biosynthesis in soybean was studied during the early stages after inoculation of primary roots with zoospores from either race 1 (incompatible, host resistant) or race 3 (compatible, host susceptible) of Phytophthora megasperma f.sp. glycinea, the causal fungus of root rot disease. In the incompatible interaction, cloned cDNAs were used to demonstrate that the amounts of phenylalanine ammonia-lyase and chalcone synthase mRNAs increased rapidly at the time of penetration of fungal germ tubes into epidermal cell layers (1–2 h after inoculation) concomitant with the onset of phytoalxxin accumulation; highest levels were reached after about 7 h. In the compatible interaction, only a slight early enhancement of mRNA levels was found and no further increase occurred until about 9 h after inoculation. The time course for changes in the activity of chalcone synthase mRNA also showed major differences between the incompatible and compatible interaction. The observed kinetics for the stimulation of mRNA expression related to phytoalexin synthesis in soybean roots lends further support to the hypothesis that phytoalexin production is an early defense response in the incompatible plant-fungus interaction. Harosoy 63) seedlings were inoculated with zoospores from either race 1 (incompatible, host resistant) or race 3 (compatible, host susceptible) of Phytophthora megasperma f.sp. glycinea and total callose was determined at various times after inoculation. From 4 h onward, total callose was significantly higher in roots showing the resistant rather than the susceptible response. Local callose deposition in relation to location of fungal hyphae was determined in microtome sections by its specific fluorescence with sirofluor and was quantified on paper prints with an image-analysis system. Callose deposition, which occurs adjacent to hyphae, was found soon after inoculation (2, 3 and 4 h post inoculation) only in roots displaying the resistant response, and was also higher at 5 and 6 h after inoculation in these resistant roots than in susceptible roots. Abstract Over two growing seasons, 273 bacterial strains were isolated from soybean leaves without and with bacterial blight symptoms caused by Pseudomonas syringae pv. glycinea (Psg). The majority of the isolates from leaves with symptoms were identified as Psg (43%), followed by Erwinia herbicola (21%), and Enterobacter/Erwinia (19%). The isolates from leaves without symptoms included mainly a group of unidentified Gram-negative bacteria (22%), Psg (21%), and E. herbicola (18%). Psg colonized the soybean leaves prior to saprophytic bacteria, and remained dominant during both seasons on healthy, as well as infected, leaves. Eighty-two saprophytic isolates were tested in vitro for their antagonistic activities against Psg, using an agar-diffusion assay. For the in planta assay, Psg and each isolate were simultaneously inoculated into wounds of pin-pricked leaves of greenhouse-grown soybean plants. Twenty-nine isolates were antagonistic in vitro. Nineteen isolates were able to suppress the growth of Psg and prevented the formation of leaf spots in planta when mixtures of isolate and pathogen were inoculated at ratios >1. Only 9 of the 82 isolates inhibited Psg in vitro as well as in planta. Most antagonists detected belonged to the genera Pseudomonas and the species Erwinia herbicola. Maximum and minimum xylem pressure potentials of needles were measured to evaluate water status of Pinus thunbergii Parl. after inoculation with the virulent or avirulent populations of Bursaphelenchus xylophilus or B. mucronatus. In virulent B. xylophilus-inoculated pines, the water status changed abruptly and needle chlorosis occurred by day 29 after inoculation. Similar changes were not seen in B. mucronatus-inoculated and uninoculated control pines. Oleoresin flow ceased in virulent B. xylophilus-inoculated pines. Avirulent B. xylophilus-inoculated pines responded very little to nematode invasion by a slight decrease in oleoresin flow. Oleoresin flow did not vary in B. mucronatus-inoculated and uninoculated control pines. A decrease in soil water potential below field capacity seemed to accelerate the development of pine wilt disease.  相似文献

We developed a model system to study the signal transduction pathways leading to the activation of Arabidopsis thaliana genes involved in the defense against pathogen attack. Here we describe the identification and characterization of virulent and avirulent Pseudomonas syringae strains that elicit disease or resistance symptoms when infiltrated into Arabidopsis leaves. The virulent and avirulent strains were characterized by determining growth of the pathogen in Arabidopsis leaves and by measuring accumulation of mRNA corresponding to Arabidopsis phenylalanine ammonia-lyase (PAL), beta-1,3-glucanase (BG), and chalcone synthase (CHS) genes in infected leaves. The virulent strain, P. syringae pv maculicola ES4326, multiplied 10(5)-fold in Arabidopsis leaves and strongly elicited BG1, BG2, and BG3 mRNA accumulation but had only a modest effect on PAL mRNA accumulation. In contrast, the avirulent strain, P. syringae pv tomato MM1065, multiplied less than 10-fold in leaves and had only a minimal effect on BG1, BG2, and BG3 mRNA accumulation, but it induced PAL mRNA accumulation. No accumulation of CHS mRNA was found with either ES4326 or MM1065. We also describe the cloning of a putative avirulence (avr) gene from the avirulent strain MM1065 that caused the virulent strain ES4326 to grow less well in leaves and to strongly elicit PAL but not BG1 and BG3 mRNA accumulation. These results suggest that the Arabidopsis PAL and BG genes may be activated by distinct signal transduction pathways and show that differences in plant gene induction by virulent and avirulent strains can be attributed to a cloned presumptive avr gene.  相似文献

Greenhouse and field experiments were carried out in order to investigate the influence of mycorrhizal inoculation on total phenolic content (TPC) and antioxidant activity, expressed as antiradical power (ARP), of artichoke (Cynara cardunculus L. var. scolymus F.) leaves and flower heads extracts. The establishment of mycorrhizal symbiosis was monitored in pot and field grown plants, and the persistence of the inoculated AMF in roots after 2 years’ growth in the field was assessed by fungal ITS sequencing. Both in the greenhouse and in the field, marked increases in TPC and ARP were detected in leaves and flower heads of artichoke plants inoculated with the AM fungal species Glomus intraradices, either alone or in mixture with Glomus mosseae. In the field, plants inoculated with Glomus mix showed flower heads ARP content increases of 52.7 and 30.0% in the first and second year, respectively, compared with uninoculated plants. After 2 years’ growth in the field ITS rDNA sequences clustering with those of G. mosseae and G. intraradices were retrieved only from inoculated plant roots. Our data show that mycorrhizal inoculation may represent an efficient and sustainable strategy to improve productivity and enhance plant biosynthesis of secondary metabolites with health promoting activities.  相似文献

Three unique classes of carbohydrates were isolated from the hyphal cell walls of Phytophthora megasperma f.sp. glycinea (Pmg) and compared with other substances for their activity as elicitors of the phytoalexin glyceollin in soybean tissues. Glucomannans extracted from cell walls with soybean β-1,3-endoglucanase were purified and proved to be the most active elicitors yet reported. They were approximately 10 times more active in soybean cotyledons than the heterogeneous β-glucan elicitor fraction extracted from Pmg walls. In addition, the glucomannan fraction gave race-specific elicitor activity in soybean hypocotyls. Pronase was found to be a suitable reagent for the mild extraction of glycopeptides from Pmg cell walls. All of the carbohydrates isolated from Pmg cell walls possessed significant elicitor activity, but other glucans, a glucomannan and mannan from other sources, were much less active. Chitin and chitosan, reported to function as elicitors in other plants, had low activity in soybean cotyledons. Arachidonic acid was inactive, despite its previously observed elicitor activity in potato tubers. The results indicated that, for Pmg, the carbohydrate elicitor most probably involved in the initiation of phytoalexinmediated defense during fungus infection of soybean plants is the glucomannan fraction liberated by endoglucanase.  相似文献

Treatment of melon hypocotyls or petioles with an elicitor from Colletotrichum lagenarium, a fungal pathogen of melons, causes an initial transitory inhibition of protein synthesis and, after 18 hours, induces the synthesis of a plant cell wall hydroxyproline-rich glycoprotein (HRGP). Microgram amounts of elicitor are sufficient for maximum elicitation of HRGP when the elicitor is injected into hypocotyls. High elicitor concentrations have a strong inhibitory effect on total protein synthesis. Ethylene is increased early in elicitor-treated plant material, and may be involved in HRGP elicitation. In the presence of aminoethoxyvinylglycine, an inhibitor of ethylene synthesis, both elicitor-induced ethylene and elicitor-induced HRGP are inhibited. On the other hand, 1-aminocyclopropane-1-carboxylic acid, the direct precursor of ethylene, triggers the synthesis of HRGP to the same extent as the elicitor of C. lagenarium, and partly restores in elicitor-treated petioles the synthesis of ethylene and of HRGP after previous inhibition by aminoethoxyvinylglycine. Elicitation of HRGP occurs in other systems, such as soybeans when inoculated with an elicitor from Phytophtora megasperma f. sp. glycinea, and when melons are incubated with an elicitor isolated from their cell walls.  相似文献

Primary roots of tomato, Lycopersicon esculentum cv. Marglobe, were cultured aseptically on agar containing a standard nutrient formulation with or without kinetin. When secondary roots developed, cultures were inoculated with the root-knot nematode, Meloidogyne incognita. Following inoculation, the cultures were divided into two groups which were incubated either in total darkness or in 16-h light-8-h dark cycles. At 24 h, 1, 2, 3, and 4 wk after incubation, roots from all cultures were processed for transmission electron microscopy. Fine structural observation of the parenchyma tissue in galls from the inoculated cultures indicated that starch containing plastids or amyloplasts, which are usually present and remain undifferentiated in these root cells, developed into chloroplasts. These chloroplasts contained a membrane system indistinguishable from those found in leaves of intact plants. Although plastid development was not affected when uninoculated cultures were incubated in the light, differentiation of the amyloplast was induced when roots were cultured on the medium containing kinetin. These results suggest that plastid differentiation in the inoculated tissue may be influenced by an accumulation of kinetin in the gall, which is induced by the nematode and serves as the nutrient sink for its feeding.  相似文献

The epiphyte Pseudomonas syringae pv. syringae 22d / 93 (Pss22d), isolated from soybean leaves, had been characterized as a promising and species‐specific biocontrol strain in vitro and in planta against the plant pathogen P. syringae pv. glycinea (Psg), which causes bacterial blight of soybean. Three toxins are known to be produced by Pss22d: syringomycin, syringopeptin and 3‐methylarginine (MeArg). In contrast to syringopeptin and syringomycin, MeArg inhibited the growth of Psg in vitro. To examine if the toxins produced by Pss22d are responsible for antagonistic effects in planta, the pathogen Psg was co‐inoculated with either Pss22d wild‐type, a syringopeptin/syringomycin‐negative double mutant (Pss22d.ΔsypA/syrE), or a MeArg‐negative mutant (Pss22d.1) into wounds of pin‐pricked leaves of greenhouse‐grown soybean plants, respectively. In all three cases, the wild‐type Pss22d and its toxin‐deficient mutants prevented development of disease symptoms normally caused by Psg. These results indicated that neither syringopeptin, nor syringomycin, nor MeArg was required for Pss22d’s antagonistic activity in planta. Consequently, factors other than the three toxins may contribute to the intra‐species antagonism in planta.  相似文献

Cucumber (Cucumis sativa) leaves infiltrated with Pseudomonas syringae pv. syringae cells produced a mobile signal for systemic acquired resistance between 3 and 6 h after inoculation. The production of a mobile signal by inoculated leaves was followed by a transient increase in phenylalanine ammonia-lyase (PAL) activity in the petioles of inoculated leaves and in stems above inoculated leaves; with peaks in activity at 9 and 12 h, respectively, after inoculation. In contrast, PAL activity in inoculated leaves continued to rise slowly for at least 18 h. No increases in PAL activity were detected in healthy leaves of inoculated plants. Two benzoic acid derivatives, salicylic acid (SA) and 4-hydroxybenzoic acid (4HBA), began to accumulate in phloem fluids at about the time PAL activity began to increase, reaching maximum concentrations 15 h after inoculation. The accumulation of SA and 4HBA in phloem fluids was unaffected by the removal of all leaves 6 h after inoculation, and seedlings excised from roots prior to inoculation still accumulated high levels of SA and 4HBA. These results suggest that SA and 4HBA are synthesized de novo in stems and petioles in response to a mobile signal from the inoculated leaf.  相似文献

The early events of Meloidogyne incognita behavior and associated host responses following root penetration were studied in resistant (cv. Moapa 69) and susceptible (cv. Lahontan) alfalfa. Ten-day-old seedlings of alfalfa cultivars were inoculated with second-stage juveniles (J2) and harvested 12, 24, 48, and 72 hours and 7, 14, and 21 days later. Both cultivars supported similar root penetration and initial J2 migration. By 72 hours after inoculation the majority of J2 were amassed inside the vascular cylinder in roots of susceptible Lahontan, while J2 had not entered the vascular cylinder of resistant Moapa 69 and remained clumped at the root apex. Nematode development progressed normally in Lahontan, but J2 were not observed in Moapa 69 after day 7. The greatest differences between RNA translation products isolated from inoculated and uninoculated roots of Lahanton occurred 72 hours after inoculation. Only minor differences in gene expression were observed between inoculated and uninoculated Moapa 69 roots at 72 hours. Comparison of translation products from inoculated versus mechanically wounded Lahontan roots revealed products that were specific to or enhanced in nematode-infected plants. Moapa 69 appears to possess a type of resistance to M. incognita that does not depend on a conventional hypersensitive response.  相似文献

Leaves of two barley (Hordeum vulgare L.) isolines, Alg-R, which has the dominant Mla1 allele conferring hypersensitive race-specific resistance to avirulent races of Blumeria graminis, and Alg-S, which has the recessive mla1 allele for susceptibility to attack, were inoculated with B. graminis f. sp. hordei. Total leaf and apoplastic antioxidants were measured 24 h after inoculation when maximum numbers of attacked cells showed hypersensitive death in Alg-R. Cytoplasmic contamination of the apoplastic extracts, judged by the marker enzyme glucose-6-phosphate dehydrogenase, was very low (less than 2%) even in inoculated plants. Dehydroascorbate, glutathione, superoxide dismutase, catalase, ascorbate peroxidase, glutathione reductase, monodehydroascorbate reductase, and dehydroascorbate reductase were present in the apoplast. Inoculation had no effect on the total foliar ascorbate pool size or the redox state. The glutathione content of Alg-S leaves and apoplast decreased, whereas that of Alg-R leaves and apoplast increased after pathogen attack, but the redox state was unchanged in both cases. Large increases in foliar catalase activity were observed in Alg-S but not in Alg-R leaves. Pathogen-induced increases in the apoplastic antioxidant enzyme activities were observed. We conclude that sustained oxidation does not occur and that differential strategies of antioxidant response in Alg-S and Alg-R may contribute to pathogen sensitivity.  相似文献

The effects of race-specific resistance as conditioned by Rps genes (rps, Rps1-k, Rps2, Rps3, Rps6) in two genetic backgrounds (Williams & Harosoy) on accumulation of soluble peroxidases were determined by a soybean peroxidase capture assay (SPCA) after inoculation with P. sojae races 2, 7, or 25. Peroxidase activity increased in all isolines during the 72 h after inoculation, but reactions varied depending on time after inoculation, genetic background, Rps gene and P. sojae race. Peroxidase activity was higher in race-specific resistant than in susceptible reactions at 72 h. after inoculation, except for plants with the Rps2 gene which confers a unique form of root resistance in addition to the whole plant race-specific resistance. Williams isolines had larger increases in peroxidase activity than Harosoy isolines when data were averaged across Rps genes, and was most evident when plants were inoculated with race 2. When soybeans were inoculated with race 7 Rps1-k resistant plants had the highest increase in peroxidase activity, but Rps2 susceptible plants had a significantly higher peroxidase activity than plants with rps, Rps3, and Rps6 that were also susceptible. Results from inoculations with race 25 were somewhat different, Rps2 resistant plants had the highest increase in peroxidase activity; however, plants with the Rps3 or Rps6 gene that were also resistant did not have a significantly higher peroxidase activity than susceptible plants with the rps or Rps1-k gene.  相似文献

Plant suspension cell cultures display many features of the innate immune responses observed in planta and have been extensively applied to the study of basal and race-specific defences. However, no single model including photosynthetic cultured cells has been used for the exhaustive characterization of both basal and race-specific defences to date. In this article, we report the activation of basal and race-specific defences in green cultured cells from Arabidopsis thaliana . Inoculation of cultured cells with isogenic virulent or avirulent strains of Pseudomonas syringae pv. tomato DC3000 (Pst) was used to evaluate race-specific defences. The proliferation of avirulent Pst was found to be lower than that of virulent Pst in the inoculated cultures. Extracellular pH changes, sustained oxidative burst (5–13 h post-inoculation), enhancement of salicylic acid, and massive cell death were specifically stimulated by the avirulent bacterium. Neither avirulent nor virulent Pst induced markers of basal resistance, such as callose deposition or early oxidative burst (1–5 h post-inoculation). However, both basal defences were activated when cells were exposed to Pseudomonas syringae pv. phaseolicola or to the Pst mutant defective in the type III secretion system (TTSS), Pst-hrpL. Thus, in these cells, basal defences may be inhibited by Pst in a TTSS-dependent manner. Recapitulation of classical defence features demonstrates the usefulness of this system for the fine characterization of plant innate immune components.  相似文献
