Summary The rabbit Graafian follicles are encircled by a capillary network between the theca interna and the avascular membrana granulosa. After injection of an ovulatory dose of human chorionic gonadotrophin (HCG) the theca interna cells showed an increase in the amount of smooth endoplasmic reticulum, lipid droplets and mitochondria with tubular cristae. In addition, considerably more junctions, similar to the abutment nexuses of granulosa cells were found; annular nexuses also appeared. At 4 hours after injection of HCG a prominent oedema was evident in the theca interna layer, particularly in the apical region.Small fenestrations in the endothelium of the blood capillaries increased in amount after HCG injection, and close to the time of ovulation, large gaps or perforations, 1–3 in diameter, were found in the thin, distended part of the endothelial cells. The surrounding basement membrane became fragmented and partly lost, so that a seemingly free passage from the capillary lumen to the interstitium was eventually established. Leakage of fluid, causing interstitial oedema, presumably proceeds until the pressure in the pericapillary interstitium has risen to the pressure in the capillaries. Some hours before and up to ovulation the pericapillary interstitium has also broad communications with the cavity of the follicles. Therefore, both pressure and fluid can be passed from the capillaries-via the interstitium-to the follicle antrum. To evaluate the relation between the steroidogenic activity and cell proliferation of individual follicles in mature hamster ovaries during the estrous cycle, the localization of enzymes involved in estrogen biosynthesis and bromodeoxyuridine (BrdU) incorporation were examined immunohistochemically. Moreover, granulosa cells from the early atretic follicle were examined by scanning and transmission electron microscopy. Immunoreactivity for aromatase was localized in the granulosa cells of healthy developing follicles and Graafian follicles, as well as in newly formed granulosa lutein cells. In the healthy follicles of an ovulation cycle, intensity of aromatase immunoreactivity was suddenly decreased on day 3. The theca interna cells of healthy developing follicles were immunopositive for 17-hydroxylase/C17–C20 lyase (17-lyase) from day 2 to the morning of day 4, but on the evening of day 4 most theca interna cells were immunonegative except for only a few cells of the large Graafian follicles. BrdU incorporation was observed in the granulosa cells of healthy developing follicles, in the endothelial cells of capillaries around the developing follicles, and of newly formed corpora lutea. Very early morphological signs of atresia was the pyknotic change of a few granulosa cells lining the antral cavity. In that follicle, the number of BrdU-incorporating granulosa cells was suddenly decreased whilst immunoreactivity of aromatase and 17-lyase were gradually decreased. These data suggest that the mechanism of the loss of aromatase activity from the granulosa cells of atretic follicles appears to differ from that in cycling follicles. Even in the early stage of atresia, the granulosa cells showed remarkable morphological change characteristic of apoptosis, as visualized by scanning and transmission electron microscopy. Cessation of granulosa cell proliferation may occur earlier than apoptotic change and the degeneration of the granulosa cells becomes rapid once atresia starts.  相似文献

Summary A systematic study of 5-3-hydroxysteroid dehydrogenase in the granulosa cells of immature and mature mice was made. The histochemical results were compared with the ultrastructural findings on the same cells in an attempt to determine whether the granulosa cells are capable of a steroidogenic role.In newborn and immature mice the granulosa cells of a great amount of follicles demonstrated a moderate or strong histochemical activity. In mature mice the granulosa cells demonstrated a weak or moderate activity normally only in preovulatory follicles and in some other atretic follicles. The granulosa cells of the normal developing follicles did not show such activity. In addition the histological control of numerous parallel sections demonstrated particularly in immature ovaries the presence of a great amount of atretic follicles.In the cytoplasm of the granulosa cells of the follicles in immature ovaries only clusters of lipid droplets with ribosomes were noted; while in the preovulatory follicles of mature animals there started to appear mitochondria with tubular cristae, smooth membranes of the endoplasmic reticulum and irregular lipid droplets. In the obviously atretic follicles several granulosa cells as well as theca interna cells showed numerous lipid droplets and ribosomes together with different degenerating organelles. The granulosa cells of the normal developing follicles showed a well developed Golgi complex, granular endoplasmic reticulum and free ribosomes.The histochemical and ultrastructural findings suggest a steroidogenic role of the granulosa cells only in the larger preovulatory follicles (probably related to an early luteinization of this layer) but this role was not demonstrated in the same cells in normal developing follicles.In addition, since an histochemical positivity was demonstrated also in the granulosa cells of some obviously atretic follicles, it is possible that many of the follicles having granulosa cells filled with lipid droplets and attached ribosomes and histochemically positive might be, in the immature ovaries, in a very precocious stage of atresia.It is to precise for these cells whether a cytoplasm with these two strictly correlated components (lipid droplets and attached ribosomes) and showing an histochemical positivity could carry-on all the biochemical steps involved in steroid biosynthesis or only has only a temporary capability to produce some precursors of steroids.The present results were partially presented to the 56ème Congrès de l'Association des Anatomistes (Nantes, 4–8/4/1971) and to the 66° Verhandlungen der Anatomischen Gesellschaft (Zagreb, 2/4/1971).  相似文献

Summary Ovaries from normal adult dairy cows were obtained at all days of the estrous cycle. The largest Graafian follicle and corpus luteum were excised, prepared for light microscopy, examined morphologically, and quantitations of nuclear sizes were made using a planimetric technique.During the 3–4 days before ovulation, membrana granulosa cells ceased growing in size, and their nuclei decreased in size and frequently appeared pyknotic. Theca interna cells during this time formed two populations: large epithelioid cells with round nuclei, that enlarged significantly, and smaller fibroblast-type cells with spindle-shaped nuclei, that did not enlarge. During the 3–4 days after ovulation, the membrana granulosa cells of the ovulatory follicle and their nuclei enlarged significantly and contributed to the large luteal cell population of the corpus luteum. The spindle-shaped theca interna cells of the ovulatory follicle assumed rounded shapes and, together with some paraluteal and trabecular luteal cells (both, probably, of theca externa origin), contributed to the small luteal cell population of the corpus luteum. The epithelioid theca interna cells of the same follicle dispersed into the ovarian stroma. Eosinophils and mast cells were commonly observed among the theca cells during this time.The observations are interpreted in relation to periestrual ovarian hormone synthesis. It is suggested that the epithelioid theca interna cells during proestrus and estrus may secrete estrogens and that the large luteal cells during diestrus may secrete progesterone.This investigation was supported by a General Research Support Grant to the College of Veterinary Medicine, University of Minnesota, of the United States Public Health Service. Summary The structural changes that characterize primary, secondary and tertiary atresia in sheep Graafian follicles have been studied by means of histological, histochemical and ultrastructural techniques.In primary atresia vacuoles representing swollen endoplasmic reticulum are prominent along the antral border together with disorganized granulosa cells containing pyknotic nuclei. Phagocytic cells, which increase in number as atresia progresses, were seen within the membrana granulosa and are considered to be transformed granulosa cells. Even in follicles classified as nonatretic, a few antral vacuoles and occasional pyknotic nuclei are present.During secondary atresia there is a large increase in the number of cells with pyknotic nuclei; many of these nuclei had been extruded and had fused to form the characteristic Feulgen-positive atretic bodies found along the edge of the antral cavity. These bodies usually have a diameter of up to 15 m but occasionally reached as much as 400 m. A second area of degeneration is frequently present in the membrana granulosa, two or three cell layers from the basal lamina, and it is at this level that exfoliation of granulosa cells occurs in tertiary atresia. In contrast to the membrana granulosa, there are during secondary atresia, only slight indications of degeneration in the cumulus.In tertiary atresia the membrana granulosa is highly disorganized; the atretic bodies are often fewer in number than at earlier stages. The basal lamina remains essentially intact. It is at this stage that the first clear signs of degeneration occur in the theca interna. Despite some disintegration of the cumulus, the integrity of the oocyte is maintained and its nucleus remains vesicular.Changes in the thecal microcirculation may play a key role in atresia: adjacent to the basal lamina of non-atretic follicles, there is a well-developed capillary network which is significantly reduced as atresia progresses.The authors are greatly indebted to Dr. H.M. Currently, histological classifications of ovarian follicular atresia are almost exclusively based on the morphology of the membrana granulosa without reference to the theca interna. Atresia in the bovine small antral ovarian follicle has been redefined into antral or basal atresia where cell death commences initially within antral or basal regions of the membrana granulosa, respectively. To examine cell death in the theca interna in the two types of atretic follicles, bovine ovaries were collected and processed for immunohistochemistry and light microscopy. Follicles were classified as healthy, antral atretic, or basal atretic. Follicle diameter was recorded and sections stained with lectin from Bandeiraea simplicifolia to identify endothelial cells or with an antibody to cytochrome P450 cholesterol side-chain cleavage to identify steroidogenic cells and combined with TUNEL labeling to identify dead cells. The numerical density of steroidogenic cells within the theca interna was significantly reduced (P < 0.001) in basal atretic follicles in comparison with other follicles. Cell death was greater in both endothelial cells (P < 0.05) and steroidogenic cells (P < 0.01) of the theca interna of basal atretic follicles compared with healthy and antral atretic follicles. Thus, we conclude that the theca interna is susceptible to cell death early in atresia, particularly in basal atretic follicles.  相似文献

We examined the frequency of proliferating cells in cystic, atretic and healthy antral follicles to determine whether a disorder of cell proliferation was responsible for the occurrence of bovine cystic follicles. Paraffin sections of healthy follicles and various stages of atretic and cystic follicles were immunostained with mouse monoclonal antibody to proliferating cell nuclear antigen (PCNA). The PCNA-positive cells were counted in 4 different regions of a follicle from the apical to the basal side. In the granulosa layer, a significantly higher frequency of PCNA-positive cells was observed in the healthy follicle in the basal region as compared with the apical region. A similar pattern of PCNA-positive cells population was observed in the granulosa layer of atretic follicles, although the frequency in the basal region was significantly lower in the atretic than the healthy follicle. The rate of cell proliferation in the granulosa layer of cystic follicles was markedly lower at the basal region than that of atretic follicles. In the theca interna, the frequency of PCNA-positive cells in atretic follicles at the early stages was higher than that in cystic follicles at the early stages. These results suggest that in the healthy follicle the proliferative activity of granulosa cells is higher in the basal than the apical region, and that the cell proliferation activity in the granulosa and theca interna may decrease in association with the induction of a follicular cyst.  相似文献

Summary The ovaries of sexually mature, pregnant mare serum gonadotropin (PMSG) stimulated, 12 week old Mongolian gerbils were investigated morphologically and enzyme histochemically for the appearance of the 3-hydroxysteroid and the 3-hydroxysteroid dehydrogenase during the estrous cycle. Up to ovulation, on day 3 of the estrous cycle, the number of vesicular follicles increases continuously. Primarily atretic follicles can be seen on day 4. On day 5 corpora lutea appear, but they degenerate already by day 6.During the entire estrous cycle, 3-hydroxysteroid dehydrogenase and 3-hydroxysteroid dehydrogenase activity can be found in the theca of tertiary follicles and in the interstitial cells, whereas the theca of secondary follicles and the granulosa of healthy follicles do not exhibit any enzyme activity. The activity decreases from day 1 till day 6. The granulosa of atretic follicles and the cells of corpora lutea show only weak activity. It may be significant that the intensity of enzyme activity in the ovary and the estrogen level in the plasma are differently correlated to the estrous cycle.This investigation was supported by the Deutsche Forschungsgemeinschaft  相似文献

Summary Immunohistochemical methods were used to show the presence and distribution of transforming growth factor-1 and 2 during folliculogenesis in quail ovarian tissues. The results indicated that both transforming growth factor- subtypes are present. Immunolabelling for transforming growth factor-1 demonstrated that prelampbrush oocytes are immunoreactive in the Balbiani complex, and developing and pre-ovulatory oocytes in the zona radiata. Immunolabelling was also associated with granulosa cells. The number of stained granulosa cells decreased during folliculogenesis. In the pre-ovulatory follicles, immunolabelling was found predominantly in the theca interna. Immunolabelling for transforming growth factor-2 was associated with the zona radiata of developing and preovulatory follicles, and with stromal interstitial cells. Moderate immunoreactivity was found in the Balbiani complex of prelampbrush oocytes. Weak immunolabelling was localized in the granulosa cells of prelampbrush follicles, and in a few cells of the theca interna of pre-ovulatory follicles. The immunolocalization of transforming growth factor-1 and-2 in the quail ovary supports their autocrine and/or paracrine role in avian ovarian processes.  相似文献

Zusammenfassung Histochemische Untersuchungen am Ratten-, Mäuse- und Meerschweinchenovarium ergaben ein unterschiedliches Verhalten der Azolesterasen (= Cholinesterase und Acetylcholinesterase) in Granulosa, Theca und Thecaorgan. Die Differenzierung der genannten Fermente wurde mit Hilfe von DFP, iso-OMPA und 284 C 51 durchgeführt. Die -Hydroxybuttersäuredehydrogenase gleicht in der Verteilung weitgehend der NADH-Cytochrom-c-Reduktase, zeigt aber quantitative Speziesunterschiede. Bei der -Ketoglutaratoxydase ist im Ratten- und Mäuseovarium erst beim atresierenden Follikel eine deutliche Aktivität der Granulosa und eine Zunahme in der Theca externa feststellbar, während im Meerschweinchenovarium die Theca interna immer, die Granulosa aber nur beim Abbau reagiert.
The histochemical behaviour of the azolesterases and other oxydoreductases during atresia of follicles in the ovaries of some rodents
Summary Histochemical investigations on the ovaries of rats, mice and guinea pigs revealed a variable behaviour of the azolesterases (= cholinesterase and acetylcholinesterase) in granulosa, theca and theca organ. The differentiation of these enzymes was carried out with DFP, iso-OMPA and 284 C 51. The distribution of -hydroxybutyrate dehydrogenase closely parallels that of NADH-cytochrome-c-reductase but shows quantitative variations in the different species. In the ovaries of rats and mice -ketoglutarate oxidase shows a marked activity only in atretic follicles and an increased activity in the theca externa. In guinea pig ovaries, however, the theca interna always reacts, but the granulosa only does so in process of decomposing.

Gonadotropic hormones PMSG (15 IU/rat), FSH (3 g/rat), LH (9 g/rat) and hCG (3 g/rat) were shown to decrease the free cytosolic lysosomal enzymes during the acute phase of hormone action in rat ovaries. When isolated cells from such rats were analyzed for the cathepsin-D activity, the granulosa cells of the ovary showed a reduction in the free as well as in the total lysosomal enzyme activities in response to FSH/PMSG; the stromal and thecal compartment of the ovary showed a reduction only in the free activity in response to hCG/PMSG. Plant regeneration has been achieved from long-term cell suspension cultures established from leaf derived callus of tepary bean (Phaseolus acutifolius). The proportion of densely cytoplasmic cells in suspension culture increased when cultured in the L-6 medium with 54 M NAA and 2 M KN. Filtration of the cells at each of five consecutive subcultures resulted in the isolation of a plant regenerating cell line (TB 686), which is being maintained in L-6 medium with 4.5 M 2,4-D and 2.3 M zeatin. Differentiated green cell aggregates were obtained when cells from maintenance medium were transferred to the same medium with 10 M BA. Embryo-like structures developed from these aggregates on L-6 medium with 2.3 M zeatin, 0.69 M GA3 and 1.5 M NAA. Plantlets regenerated from these structures when they were cultured on L-6 medium with 7.0 M NAA and 1.0 M KN. Plant regeneration from the cell line remained relatively constant for 270 days. Synopsis The ovary of the domestic pigeon,Columba livia, has been assayed histochemically for the localization of 5-3-hydroxysteroid dehydrogenase (5-3-HSDH), 17-hydroxysteroid dehydrogenase (17-HSDH), 11-hydroxysteroid dehydrogenase (11-HSDH), glucose-6-phosphate dehydrogenase (G6P-DH) and NADH-diaphorase activities during different periods of the reproductive cycle. 5-3-HSDH, 17-HSDH, 11-HSDH, G6P-DH and NADH-diaphorase activity was found in the theca interna of growing, atretic and postovulatory follicles, the granulosa of ovulatory, atretic and postovulatory follicles, and interstitial gland cells during the pre-incubation and the laying periods. During the incubation and squab feeding periods only 5-3-HSDH, G6P-DH and NADH-diaphorase activities were observed in the above mentioned cells. The steroidogenic potential of atretic follicles depends upon the type of atresia a follicle undergoes.  相似文献

Summary To identify and describe ovarian follicles committed to undergo follicular degeneration (atresia), immature rats were primed with pregnant mare serum gonadotropin (PMSG). After PMSG treatment, preovulatory follicles develop but subsequently degenerate. Prior to the appearance of pyknotic nuclei (Stage I of atresia), degenerative changes were observed in focal areas of the granulosa cell layer. These changes include blebbing of the cytoplasm and alterations in the shape of the granulosa cells. The appearance of these degenerative changes coincides with a decrease in ovarian concentrations of estradiol and testosterone. Since estrogens and androgens maintain the follicle, the decline in estradiol and testosterone could be responsible for the further degenerative alterations that lead to complete deterioration of the preovulatory follicle. In Stage I atretic follicles, lysosome-derived autophagic vacuoles develop and macrophages invade both the thecal and granulosa cell layers. The combined actions of the autophagic vacuoles and macrophages could destroy both the granulosa-cell and thecal layers and thereby transform the preovulatory follicle into an ovarian cyst.  相似文献

Summary The role of ovarian adrenergic nerves in follicular growth was studied in prepubertal guinea pigs by determining the effect of sympathectomy on 1) follicle populations and 2) follicular development following exogenous gonadotropin administration. Selective unilateral ovarian sympathectomy was achieved by injecting 6-hydroxydopamine into a surgically closed periovarian bursa on day 20 postpartum. The contralateral surgically closed ovarian bursa was injected with the vehicle used for 6-hydroxydopamine. On day 25, animals were injected with pregnant mare serum or saline followed by human chorionic gonadotropin or saline 48 h later. All animals were laparotomized on day 28 and blood from utero-ovarian veins was collected bilaterally for androstenedione determination. Ovaries were processed for morphometric analysis of follicles. The sympathectomized ovary in saline-injected animals had a significant decrease in preantral follicles (characterized by 2 layers of granulosa cells without antrum formation), an increase in 310–500 m diameter atretic follicles and an increase in follicles 700 um compared to the contralateral control ovary. There were no differences in androstenedione levels from the two sides, ovarian weights or the total number of follicles per ovary. Neither ovary had corpora lutea. The sympathectomized ovary in animals injected with gonadotropins was not different from the contralateral ovary in any of the parameters measured. Both control and sympathectomized ovaries had newly formed corpora lutea in response to the exogenous gonadotropins. These results suggest that ovarian adrenergic nerves normally participate in follicular development in the prepubertal guinea pig. However, exogenous gonadotropins may override neural influences on the prepubertal ovary.  相似文献

Summary Mature virgin rats were injected daily with 50 g melengestrol acetate (17-acetoxy-6-methyl-16-methyIenepregna-4,6-diene-3,20-dione, or MGA) and 0.2 g estradiol benzoate for periods of 1, 2, 3, and 4 weeks. Ovarian structures were examined histologically, histochemically and in the electron microscope. Graafian follicles during treatment did not attain pre-ovulatory size, ovulation was inhibited, and corpora hemorrhagica were not formed. During treatment, the percentage volume of the ovary occupied by atretic Graafian follicles increased, and that occupied by developing Graafian follicles decreased. In atresia, the membrana granulosa degenerated, and the theca cells hypertrophied peripherally to contribute to the surrounding interstitial gland. The structure and the weak lipid reaction of the hypertrophied theca cells of test follicles, when compared with estrus control follicles, suggested a low level of steroid production. The strong lipid reaction and ultrastructure of the interstitial gland suggested a lipid storage rather than secretory function. Corpora lutea, containing cells with a light lipid reaction and organelles suggestive of steroid synthesis, persisted throughout treatment. On the basis of claims, which associate the theca cells with estrogen synthesis and the luteal cells with progesterone synthesis, the morphological features described are consistent with primarily progestational and low-level estrogenic activities of MGA in the rat.This investigation was supported by the Alexander von Humboldt-Stiftung. Summary The cytodifferentiation and subcellular steroidogenic sites in the theca cell of the human ovary during the follicular phase were investigated using the electron microscopic cytochemistry. Only fibroblast-like cells were seen around or near the primordial follicle. In the theca interna of the secondary and Graafian follicle however there were three different cell types: fibroblast-like cells, theca gland cells (steroid-secreting cells) and transitional cells (partially or incompletely differentiated theca cells). On the other hand the theca externa of these follicles consisted mainly of fibroblast-like cells. The hallmarks of the cytodifferentiation of the theca cells were: 1) the appearance of lipid droplets, 2) a structural change of the mitochondrial cristae from lamellar to tubular form and 3) the appearance and development of smooth endoplasmic reticulum. Reaction products of 3-hydroxysteroid ferricyanide reductase, indicating 3-hydroxysteroid dehydrogenase activity, were localized on tubular or lamellar cristae and inner membrane of the mitochondria, and on the membranes of smooth endoplasmic reticulum in the transitional cell as well as in the theca gland cell of the secondary and Graafian follicle. From these data, it is suggested that the transitional cell has a steroid-secreting activity and also plays an important role in follicular development in human reproduction.Supported by a grant from the Japanese Educational Ministry  相似文献

Summary The effects of 9-tetrahydrocannabinol on the activities of lactate and succinate dehydrogenases in the theca interna and membrana granulosa of rat preovulatory follicles have been analysed microdensitometrically using the same injection regime employed in a previous study on steroidogenic enzymes. A small but statistically significant (18%) decrease in succinate dehydrogenase activity was observed in the theca interna, but none in any region of the membrana granulosa. Lactate dehydrogenase activity was unaffected by THC administration. Thus, a dosage and regimen sufficient to cause significant decreases in the activities of steroidogenic enzymes had little effect on succinate and lactate dehydrogenases in rat preovulatory follicles.  相似文献

Summary Changes in the distribution of the in vitro uptake of 125I-HCG by the ovaries of adult rats were examined histochemically throughout the estrous cycle.Only in follicles wider than 500 m, occurring mainly at diestrus and proestrus, could granulosa cells bind the labelled hormone. The labelling increased with follicular size and decreased in intensity from the peripheral granulosa cells inwards. No uptake occurred in the oocytes, in the cells of the cumulus oophorus nor in the granulosa cells of the atretic follicles.The binding capacity of the newly-formed corpora lutea of estrus was less than that of preovulatory follicles. The uptake of 125I-HCG by corpora lutea during the first cycle reached its maximum at diestrus but fell sharply by proestrus. The uptake was patchy in the corpora lutea of the second cycle and not significant in the older ones.The uptake of 125I-HCG by thecae increased with follicular size and was greater in the thecae of atretic follicles than in the thecae of growing follicles of like size. There was a greater uptake in the last formed interstitial tissue than there was in older tissue.At proestrus, the uptake of 125I-HCG was unaffected by the LH surge at 18.00h but had decreased slightly at 24.00 h.The implications of these data in relation to the regulation of receptor sites, is discussed.  相似文献

Summary Morphological alterations induced by dehydroepiandrosterone (DHA) were studied in polycystic mouse ovaries (PCO). Treated mice showed ovulatory failure and cystic changes; cysts and follicles in various stages of growth and atresia were present although corpora lutea were absent. The levels of testosterone, dihydrotestosterone, 3- and 3-androstanediol, estrone and androstenedione increased, whereas estradiol was not detectable.The ultrastructure of granulosa cells in healthy and atretic follicles was similar to that of control animals, although the membrana granulosa in cysts was reduced to a monolayer of flattened cells. The theca interna of healthy and atretic follicles and ovarian cysts showed ultrastructural signs of abnormal steroidogenic stimulation.No significant differences (0.7<P<0.8) were found between the extensive surface area of gap junctions of healthy follicles of control and DHA-treated animals. On the P-face of granulosa cells of large healthy follicles, meandering strands of tight junctional particles were observed; their average length was significantly longer than those in healthy follicles of control animals (P<0.001). This increase was probably related to the large amounts of androgens present in the treated animals.Theca interna cells possessed small gap junctions; no significant differences (P>0.9) in gap-junction surface area were observed between DHA-treated and control animals.
