A commonly used measure of ordinal association in two-way contingency tables is Goodman and Kruskal's gamma. In a randomized clinical trial setting, the row variable may consist of increasing doses of a drug and placebo (i.e., treatments) and the column variable may be an ordinal response variable (e.g., physician's global evaluation of treatment effectiveness). Typically, patients are randomly assigned to treatments (rows) such that the row totals are fixed and the rows form a product-multinomial sample of the ordinal response variable. For the above situation, we provide the asymptotic variance of gamma using the delta method. The population version of gamma and its standard error can be estimated by their sample counterparts, enabling us to construct approximate tests and confidence intervals using the normal distribution. 相似文献

We consider the problem of testing for independence against the consistent superiority of one treatment over another when the response variable is binary and is compared across two treatments in each of several strata. Specifically, we consider the randomized clinical trial setting. A number of issues arise in this context. First, should tables be combined if there are small or zero margins? Second, should one assume a common odds ratio across strata? Third, if the odds ratios differ across strata, then how does the standard test (based on a common odds ratio) perform? Fourth, are there other analyzes that are more appropriate for handling a situation in which the odds ratios may differ across strata? In addressing these issues we find that the frequently used Cochran–Mantel–Haenszel test may have a poor power profile, despite being optimal when the odds ratios are common. We develop novel tests that are analogous to the Smirnov, modified Smirnov, convex hull, and adaptive tests that have been proposed for ordered categorical data. (© 2006 WILEY‐VCH Verlag GmbH & Co. The stratified Cox proportional hazards model is introduced to incorporate covariates and involve nonproportional treatment effect of two groups into the analysis and then the confidence interval estimators for the difference in median survival times of two treatments in stratified Cox model are proposed. The one is based on baseline survival functions of two groups, and the other on average survival functions of two groups. I illustrate the proposed methods with an example from a study conducted by the Radiation Therapy Oncology Group in cancer of the mouth and throat. Simulations are carried out to investigate the small‐sample properties of proposed methods in terms of coverage rates. 相似文献

We consider a semiparametric inference procedure for data from epidemiologic studies conducted with a two-component sampling scheme where both a simple random sample and multiple outcome- or outcome-/auxiliary-dependent samples are observed. This sampling scheme allows the investigators to oversample certain subpopulations believed to have more information about the regression model while still gaining insights about the underlying population through the simple random sample. We focus on settings where there is no additional information about the parent cohort and the sampling probability is nonidentifiable. We motivate our problem with an ongoing study to assess the association between the mutation level of epidermal growth factor receptor (EGFR) and the antitumor response to EGFR-targeted therapy among nonsmall cell lung cancer patients. The proposed method applies to both binary and multicategorical outcome data and allows an arbitrary link function in the framework of generalized linear models. Simulation studies show that the proposed estimator has nice small sample properties. The proposed method is illustrated with a data example. 相似文献
