Amo 1618 inhibited the incorporation of MVA-[2-14C] into sterols (in particular the 4-desmethylsterols) and promoted its accumulation in squalene-2,-3epoxide in intact seedlings, tissues from treated seedlings, and sub-cellular, membraneous fractions of treated 21-day-old tobacco seedlings. The stem tissues appeared to have a greater sterol requirement (on a per gram fresh weight basis) than leaf tissues and incorporation of radioactivity into stem sterols was more susceptible to inhibition by Amo 1618 than incorporation into leaf tissue.  相似文献

Three plant-growth retardants 2′-isopropy1-4′-(trimethylammonium chloride)-5-methylphenylpiperidine carboxylate (Amo 1618), β-chloroethyltrimethylammonium chloride, and tributyl-2, 4-dichlorobenzylphosphonium chloride were tested for their effects on sterol production in, and growth of tobacco (Nicotiana tabacum) seedlings. As the concentration of each retardant increased, there was an increased inhibition of the incorporation of dl-2-14C-mevalonic acid into sterol (particularly desmethylsterol) fractions and an increased retardation of stem growth. Growth retardation was observed with both single and repeated retardant treatments, and with Amo 1618, in particular, a close quantitative relationship between inhibition of sterol biosynthesis and stem growth was obtained. Gibberellic acid completely overcame retardant effects and application of sterols also restored normal growth. It is concluded that the concept of causality in the relationship between growth retardation and gibberellin biosynthesis is probably premature, since growth retardants have a more general inhibitory action on isoprenoid biosynthesis in plants.  相似文献

The incorporation of radioactivity into sterols by transmethylation of methionine-[14C-methyl] was studied in mung bean hypocotyl sections. Young hypocotyl sections (1 cm) synthesized 4 times more radioactive sterols than older sections (5 cm). The transmethylation reactions may be rate limiting in older tissues. Wounding has only a quantitative effect on sterol biosynthesis, as seen by incorporation experiments with MVA-[2-14C]. Naphthalene acetic acid (NAA) stimulates sterol biosynthesis in both wounded surfaces and intact tissues of mung bean hypocotyl sections.  相似文献

The incorporation of mevalonate-[2-14C] into the free sterols, steryl esters, steryl glucosides, acylated steryl glucosides and water-soluble complexes was investigated and the sterols of each fraction were separated into stanols, Δ7 sterols, Δ5 sterols, stigmasterol, clerosterol and methylene-cholesterol. The stanols and Δ7 sterols were more strongly labelled in the steryl esters than in the free sterols. The Δ5 sterols and stigmasterol were more intensively labelled in the free sterols than in the steryl esters. All sterol types were more labelled in the steryl glycosides than in the acylated steryl glucosides. Stanols were probably formed from Δ7 or Δ5 precursors.  相似文献

The radioactive precursor, [3?3H]oleanolic acid-3-O-mono-[14C]glucoside was administrated to isolated cells obtained from the leaves of Calendula officinalis. The radioactivity of the precursor was incorporated into fractions containing free oleanolic acid, individual glucosides, glucuronide F and other glucuronides. The ratio of 3H: 14C radioactivity in these fractions indicated that glucosides were formed in a process involving direct glycosylation of the precursor, whereas the glucuronides were formed from oleanolic acid released by hydrolysis of the precursor. Dynamics curves showed that glucoside II formed by direct glycosylation of the precursor was intensively transformed to other derivatives.  相似文献

By incubation of germinating soybeans with mevalonate-[2-14C] (MVA), radioactivity was incorporated into four sapogenols which were identified by TLC. Unequivocal evidence for the identity of three of the four sapogenols was provided by co-crystallization to constant specific radioactivity. The partition of incorporated radioactivity into lipid- and water-soluble fractions and the pattern of radioactivity of individual sapogenols varied with the mode of administering labeled substrates to soybean seedlings, such as incubation of germinating soybeans with MVA-[2-14C], immersion of roots into MVA-[2-14C] or foliar application of squalene-[14C]. When alfalfa seedlings were incubated with MVA-[2-14C], about two-thirds of the radioactivity incorporated into the sapogenols was associated with medicagenic acid.  相似文献

With 14CO2, d-glucose-[U-14C] and dl-mevalonate-[4R-4-3H1] used as precursors, a study was made of the labelling dynamics of the steryl glucosides (SG) and steryl acylglucosides (ASG) in Sinapis alba seedlings. The radioactivity of the sterol and sugar moieties, as well as of the fatty acid moieties in the case of ASG, was analysed separately. The course of incorporation of 14C from 14 CO2 and glucose-[U-14C] into the sugar part of SG and ASG indicated that about 23 of the whole pool of the newly synthesized sterol glycosides of both types underwent rapid deglucosylation. Likewise, fatty acids in the ASG pool were rapidly exchanged. The present results point to a high metabolic activity of the sterol glycoside derivatives in plant cells.  相似文献

Gibberellic acid-induced germination of hazel seeds was accompanied by little change in the sterol content of the cotyledons. Dormant and germinating cotyledons rapidly incorporated [2-14C]MVA into squalene which was slowly converted to sterols. Gibberellin treatment induced an increase in the incorporation of [2-14C]MVA into cotyledon esterified sterols. An increase in free sterols occurred in the germinating embryonic axes, with increased relative amounts of stigmasterol and campesterol in the free 4-desmethylsterols. Germination was accompanied by increased incorporation of [2-14C]MVA into free and esterified sterols in the embryonic axes.  相似文献

Addition of either l-[U-14C]threonine or l-[U-14C]isoleucine to 2.7-day-old shaking liquid cultures of Pseudomonas syringae pv. atropurpurea resulted in incorporation of radioactivity into coronatine, but not into N- coronafacoylvaline, another phytotoxin excreted by P.s. atropurpurea. In contrast, addition ofl-[U-14C]valine did not lead to incorporation of radioactivity into coronatine, but instead into coronafacoylvaline. Acid hydrolysis of the purified [14C] coronatine obtained after incorporation of either [14C]isoleucine or [14C]threonine demonstrated that > 94% of the radioactivity was present in the 1-amido-1-carboxy-2-ethylcyclopropyl moiety of coronatine, and < 6 % was in the coronafacoyl moiety. These findings are used to propose a biosynthetic pathway for coronatine.  相似文献

The distribution of [14C]-labelled material into subcellular fractions of 15-day-old rat brain was studied at 2 and 24 h following intraperitoneal and intracerebral injection of [2-14C]sodium acetate, [U-14C]glucose and [2-14C]mevalonic acid respectively. The total quantity of labelled isoprenoids in the brain was, except for glucose, greater when the precursor was administered intracerebrally. The intraperitoneal route was more advantageous in the case of [U-14C]glucose. The subcellular distribution of both labelled total isoprenoid material and sterol was distinct for each labelled precursor. Intracerebrally injected [U-14C]glucose at both time periods studied suggested no dominance of labelling in any fraction. After intraperitoneal injection of [U-14C]glucose the microsomes were more prominently labelled. Both methods of administration of sodium [2-14C]acetate resulted in heavy labelling of the myelin fraction after 24 h. The total labelled isoprenoids resided mainly in the microsomes 24 h after injection of [2-14C]mevalonic acid. Labelled sterol was found to be localized more in the myelin and microsomal fractions for all three precursors than was the labelled total isoprenoids. Depending on the type of experiment to be conducted, each of these precursors can give different results, which must be interpreted accordingly.  相似文献

The effect of the polypeptide antibiotic, amphomycin, on the in vitro and in vivo synthesis of polyprenyl-linked sugars and glycoproteins in plants was examined. This antibiotic blocked the transfer of mannose from GDP-[14C]mannose into mannosyl-phos-phoryl-dolichol by a particulate enzyme preparation from mung beans and also inhibited the transfer of GlcNAc from UDP-[3H]GlcNAc to GlcNAc-pyrophosphoryl-polyisoprenol. The in vitro incorporation of these sugars into trichloroacetic acid-insoluble material was also markedly inhibited by this antibiotic. Since most of the radioactivity incorporated into this insoluble material is rendered water-soluble by treatment with pronase, it seems likely that these sugars are incorporated into glycoproteins whose synthesis is sensitive to amphomycin. Amphomycin also inhibited the transfer of glucose from UDP-[14C]glucose to steryl glucosides, although this system was less sensitive to antibiotic than was synthesis of the polyprenyl-linked sugars. The antibiotic did not block the in vitro transfer of glucose from UDP-[14C]glucose to β-glucans. In carrot slice cultures, amphomycin also inhibited the incorporation of [14C]mannose into glycolipid and glycoprotein, but it did not prevent the incorporation of [14C]lysine into protein.  相似文献

Incorporation of radioactivity from [6-14C]orotate into the pyrimidine constituents of shoots of Pisum sativum, Phaseolus vulgaris and Lathyrus tingitanus was examined with special reference to the unusual pyrimidine constituents. With each species, although 80% of the orotate supplied was catabolized to β-alanine, all the pyrimidine derivatives became radioactively labelled. With Pisum, the major part of the radioactivity incorporated into pyrimidines was located in UMP and the uracil derivatives, including the uracilyl amino acids willardiine and isowillardiine. With Phaseolus, UMP and the uracil derivatives were again the major radioactive products; incorporation of radioactivity into 5-ribosyluracil (pseudouridine), which accumulates in Phaseolus tissues, was comparable to the incorporation into orotidine and twice that found in cytidine. Lathyrus incorporated a substantially larger part of the presented [6-14C] orotate into pyrimidine derivatives than did the other two species. CMP was the most highly radioactive product, followed next by lathyrine and UMP. Surprisingly, 20% of the total radioactivity incorporated into pyrimidines by Lathyrus was located in the pyrimidine amino acid lathyrine. This confirms previous evidence that lathyrine is essentially a product of the orotate pathway. The overall recovery of radioactivity in all three species was 93–95%. The data emphasize the necessity of including the less common pyrimidine constituents, as well as the common ones, in quantitative studies of pyrimidine metabolism in plants.  相似文献

Six-day-old tobacco seedlings rapidly incorporated and metabolized exogenously supplied [4-14C]-sitosterol, but none of the radioactivity was recovered from stigmasterol. However, exogenously supplied [2-14C]-mevalonic acid was incorporated into both sitosterol and stigmasterol. Based on these results it is suggested that the biosynthetic pathway of stigmasterol is not via sitosterol but that both sterols have a common precursor.  相似文献

U-14C-phenylalanine and 3-14C-cinnamic acid were fed to detached Polygonum leaves through the cut petioles and to the bases of detached wheat leaves. After feeding, the leaves were divided into basal, middle and terminal segments; for each treatment of each plant more than 80% of the total radioactivity incorporated remained in the basal segment. The distribution of radioactivity between ethanol-soluble and insoluble fractions in each segment was examined. The basal segments contained more insoluble radioactivity than the terminal ones; the differences were far more marked for both plants when cinnamate rather than phenylalanine was administered. In view of the gross differences in distribution of radioactivity between the basal and terminal segments of each leaf, it is concluded that basal infusion of precursors is not the most suitable technique for studies of phenolic biosynthesis.  相似文献

Rat anterior hemipituitaries incubated in vitro rapidly take up and incorporate into protein D-[6-3H]-glucosamine · HCl, D-[1-14C]mannose and L-[G-3H]fucose. The newly labeled protein was only slowly released into a Krebs-Ringer bicarbonate incubation medium. Glucosamine- or mannose-labeled protein was barely detectable in the medium after a 30–60 min incubation whereas about 4% of all fucose-labeled protein had already been released into the incubation medium by 30 min. Puromycin · 2HCl (1 mM) inhibited incorporation of glucosamine or mannose into protein to 40% or less of control values within 30 min; fucose incorporation was not significantly inhibited before 45 min. Acid hydrolysis followed by amino acid analysis of glucosamine-labeled protein yielded significant amounts of label in glucosamine, galactosamine and apparent glucosamine-degradation products but no significant amount of label in any amino acid.  相似文献

Incorporation of DL-[2-14C]mevalonic acid ([2-14C]MVA) into4-desmethylsterols in Nicotiana tabacum cv. Turkish Samson seedlingswas inhibited by SK&F 7997-A3,1 SK&F 7732-A3, AY 9944,and the plant growth retardant, Amo 1618. Reductions in 4-desmethylsterol levels resulted from treatmentwith AY 9944 and Amo 1618, but not the SK&F compounds. Amo1618 and SK&F 7997-A3 both significantly reduced the specificactivity of each of the four major 4-desmethylsterols examined.Although SK&F 7732-A3 reduced the specific activity of campesterol,and AY 9944 reduced the specific activity of stigmasterol, neitherhad an effect on the specific activity of ß-sitosterol. Stem elongation of tobacco seedlings was retarded by SK&F7997-A3, AY 9944, and SK&F 7732-A3, particularly the former,and the retarded plants thus produced were morphologically indistinguishablefrom the Amo 1618-treated plants. Application of exogenous stigmasterol,or GA3, to the chemically-retarded plants resulted in a reversalof stem growth retardation.  相似文献

Administration of 24-methylene-cholesterol-[28-3H] to Withania somnifera, yielded [3H] radioactivity in the isolated withaferin A and withanolide D, whereas administered 24-(R,S)-methyl-cholesterol-[28-3H] was not incorporated into these compounds. 24-Methylene-cholesterol is, therefore, proposed as a sterol precursor of the withanolides. A novel procedure is described for the isolation of withanolides from W. somnifera. This method in conjunction with an improved procedure for administration of labelled sterols and mevalonolactone produces a greatly increased yield of labelled withanolides.  相似文献

The effects of AMO-1618, AY-9944 and SKF-7997 on the growth of Nicotiana tabacum and on the biosynthesis of acidic and neutral isopentenoids in the seedlings were examined. All three compounds significantly retarded stem elongation but not dry wt of the plants. Incorporation of radioactivity from [2- 14C]-mevalonate into acidic and neutral isopentenoids showed a direct relationship between the radioactivity in the two fractions and stem growth. All three compounds inhibited steps before as well as after squalene formation, but the acylic and polycyclic isopentenoids which accumulated as a result of the inhibition were not necessarily the same in each of the treatment groups. This indicates that stem growth is probably influenced not by a single product of isopentenoid biosynthesis, but rather by several products, which may even act independently in their effects on developmental processes.  相似文献

Six-day-old barley seedlings were allowed to take up [4-14C]sitosterol and [22, 23-3H]sitosterol for 2.5 hr and the incorporation into the sterol fractions was determined after 0, 6, 12 and 24 hr. Sitosterol was readily incorporated into every sterol class. The 3H/14C ratio in the free forms dropped when compared with the 3H/14C ratio of the administered sitosterol. In the free sterol, radioactive stigmasterol, showing a 3H/14C ratio half that of the sitosterol 3H/14C ratio, was isolated and its radiochemical purity established by dilution with carrier material and crystallization to constant specific activity.  相似文献
