An S-adenosyl-l-methionine: o-dihydric phenol O-methyltransferase was isolated from tobacco cell suspension culture and was partially purified by (NH4)2SO4 precipitation and successive chromatography on DEAE-Sepharose, Sephacryl S-200 and hydroxyapatite columns. It catalysed the O-methylation of 3 cinnamic acids, two coumarins and two flavonoids, but to different extents. Results obtained from polyacrylamide gel electrophoresis, m-/p-methylation ratios and mixed substrate experiments indicated the existence of two forms of the enzyme which were resolved by chromatography on DEAE-cellulose. One form (MW 74000, pI 6.1, opt. pH 7.3) catalysed the meta-methylation of caffeic acid, while the other (MW 70000, pI 6.3, opt. pH 8.3) mediated the para-methylation of quercetin, though each form exhibited some activity against other substrates. 相似文献

An enzyme preparation from suspension cultured tobacco cells oxidized IAA only in the presence of added cofactors, Mn2+ and 2,4-dichlorophenol, and showed two pH optima for the oxidation at pH 4·5 and 5·5. Effects of various phenolic compounds and metal ions on IAA oxidase activity were examined. The properties of seven peroxidase fractions separated by column chromatography on DEAE-cellulose and CM-Sephadex, were compared. The peroxidases were different in relative activity toward o-dianisidine and guaiacol. All the peroxidases catalysed IAA oxidation in the presence of added cofactors. The pH optima for guaiacol peroxidation were very similar among the seven isozymes, but the optima for IAA oxidation were different. The anionic and neutral fractions showed pH optima near pH 5·5, but the cationic isozymes showed optima near pH 4·5. With guaiacol as hydrogen donor, an anionic peroxidase (A-1) and a cationic peroxidase (C-4) were very different in H2O2 concentration requirements for their activity. Peroxidase A-1 was active at a wide range of H2O2 concentrations, while peroxidase C-4 showed a more restricted H2O2 requirement. Analysis by ion-exchange chromatography of the enzymes from cultured tobacco cells and root or leaf tissues of the tobacco plant revealed that the cultured cells contain exclusively cationic peroxidases and the leaf tissues mainly anionic and neutral peroxidases. 相似文献

A partially purified UDP-glucose: o-dihydroxycoumarin glucosyltransferase from tobacco cell culture required an intact coumarin ring system with o-dihydroxy groups for highest activity. The enzyme exhibited strict position specificity towards the 7-OH group of both esculetin and daphnetin with the formation of cichoriin and daphnin, respectively. The apparent Km values were 111 and 95 μM, while the Vmax for esculetin was 26.4 pkat/mg protein. 相似文献

Effect of norleucine on growth,protein and catechol oxidase synthesis in tobacco suspension cultures
Tobacco cells were grown in artificial media with defined amino acid composition. In such media, the addition of methionine or norleucine caused increases in the specific activity of the catechol oxidase, while in the normal medium norleucine depressed it. The differences of the effect of norleucine on synthesis of catechol oxidase and on cell growth is demonstrated, as is the reversibility of the norleucine effect by methionine. The incorporation of norleucine into a purified enzyme fraction is shown. The change in the electrophoretic patterns of the enzyme during growth in the absence and presence of norleucine was followed. [14C]-Leucine incorporation by control and norleucine treated cells was examined and it was shown that protein synthesis in the norleucine treated cells was markedly changed and total incorporation reduced. Incorporation into soluble protein was reduced, but increased in the 20 000 g precipitate fraction. Nevertheless use of autoradiography indicates that some catechol oxidase is apparently synthesised in the presence of norleucine. 相似文献

Tobacco cells grown in suspension culture contained a higher specific activity of acid phosphodiesterase than tissues of the intact plant. The activity of the cells increased with age. One component of acid phosphatase was purified from cultured tobacco cells. The purified enzyme was homogeneous on polyacrylamide gel electrophoresis with or without sodium dodecyl sulfate. The enzyme possesses high activity toward nucleoside di- and triphosphate, much less activity toward nucleoside monophosphates and sugar esters. The MWs of the phosphatase determined by Sephadex G-100 gel filtration and dodecyl sulfate gel electrophoresis were 74000 and 76000, respectively. The phosphatase showed high affinity for concanavalin A-Sepharose and single superimposed bands of protein and carbohydrate on gel electrophoresis, suggesting that it is a glycoprotein. 相似文献

Summary Cell masses of Araucaria angustifolia cultured in LP liquid medium showed different uptake and utilization patterns for different sugars. Fructose was slowly utilized
by cell growth during the lag phase. After this lag period, fructose concentration decreased quickly, corresponding to the
start of linear growth. Glucose rapidly decreased following fructose depletion. The level of extracellular proteins in the
culture medium increased for the first 5 d after cell inoculation, during the transition from the lag phase to the high-growth
phase. The pH of the medium decreased through the first half of the culture period (4.94) and increased (5.6) thereafter.
Proembryos originated from a single cell, without a cleavage process. Suspensor cells and proembryonal groups developed independently.
Only embryogenic cells divided and formed a suspensor or a proembryo group, which differentiated further along the longitudinal
axis. The pathway observed for proembryo formation in Araucaria differs from the model proposed for other conifers, which show cleavage polyembryony. 相似文献

腺病毒载体是极具发展前景的基因治疗载体之一,为获得一条新型腺病毒规模化生产工艺,研究采用5 L振荡激流式一次性生物反应器悬浮培养HEK293细胞来生产重组腺病毒载体。细胞经种子链逐步扩增后,接种至AP10生物反应器,采用CD293无血清培养基流加培养悬浮HEK293细胞,细胞密度达约2.0×106个/m L时,以30 MOI(Multiplicity of infection)感染重组Ad-IFNγ(Recombinant adenovirus-interferon gamma),48 h后收获细胞,3次冻融裂解离心收获上清病毒粗产品。采用壳蛋白免疫法快速测定粗产品滴度。结果表明,采用振荡激流式一次性生物反应器,流加HEK293细胞悬浮培养6 d,密度可达2.0×106个/m L,Ad-IFNγ粗产量达1.49×1013 IFU(Infectious unit),单细胞包装量达3 800 IFU/cell。采用阴离子交换层析法纯化重组腺病毒,回收率35.9%。建立了利用5 L激流式一次性生物反应器悬浮培养HEK293细胞生产重组腺病毒载体Ad-IFNγ的初步工艺。 相似文献

石竹细胞继代周期为 7d时 ,悬浮细胞培养系生长最快 ,生长率最高 ,而且培养物中胚性细胞较多 ,并能保持较快的分裂和生长 ,能促进已形成的大细胞团的生长和分化。转代时接种物与新鲜培养基的体积比以1∶2较好 ,悬浮系细胞生长最快 ,生长率最高 ,以 1∶2和 1∶3的高倍稀释接种有利于胚性细胞的形成及产生小的胚性细胞团 ,对悬浮系添加椰乳和水解乳蛋白的混合物 ,可较大幅度地提高悬浮细胞系的生长速率 ,单独添加上述两种物质的效果均不如二者的综合效应好。在 6种不同激素组合中 ,配方 2 (2 ,4 D 1 .5mg/L +NAA0 .5mg/L +6 BA 0 .5mg/L)最好 ,生长率最高。配方 5 (2 ,4 D 1 .5mg/L +NAA 0 .5mg/L +6 BA 1 .0mg/L)其次 ;配方 1 (2 ,4 D 1 .0mg/L +NAA 0 .5mg/L +6 BA 0 .5mg/L)次之。 相似文献

Aggregates of tobacco cells in suspension in 2,4-D (10?6 M) and kinetin (10?5 M) cultures were fractionated by size, then their O-methyltransferase (OMT) activities were assayed. Only the kinetin culture showed high OMT activity, which was higher in the larger than the smaller aggregates at all stages of cell growth. The contents of phenolic acids were also greater in the larger cell aggregates in the kinetin culture. However, when the kinetin cultured cells were transferred to a medium containing 10?6 M of 2,4-D, the relationships between the cell size of the aggregates and OMT, lignin and the phenolic acids disappeared. The importance of kinetin and cell association for OMT and the subsequent lignification of the cells is discussed. 相似文献

We analyzed the effect of Cd2+ on cell cycle progression in partially synchronized soybean (Glycine max) cell suspension culture and followed the expression of cell cycle genes (cyclin B1 and cyclin-dependent kinase A - CDK-A).
We have checked the hypothesis that Cd2+-induced impairment of cell division is connected with DNA damage. The [3H]-thymidine incorporation in cell cultures synchronized either with hydroxyurea (HU) or phosphate starvation have shown,
that Cd2+ strongly affects the S phase of soybean cell cycle, by causing the earlier entry of cells into S phase and by decreasing
the rate of DNA synthesis. RT-PCR analysis indicated that Cd2+ decreases the level of cyclin B1 mRNA and has no effect on CDK-A mRNA. The result of comet assay indicated the damaging effect
of Cd2+ on DNA of soybean cells. We suggest that Cd2+ affects plant cell cycle at two major checkpoints: the G1/S — by damaging of DNA, and G2/M - by decreasing the level of cyclin

Conditioned media of suspension cultures of Nicotiana tabacum var. Samsun contain 0.15-0.20 mmol/l glutathione. These concentrations correspond closely to the intracellular content of glutathione and represent about three to four times the amount of glutathione needed to maintain the intracellular level of glutathione.
