The investigation of Calea urticifolia afforded eight new germacranolides. While five are heliangolides, three are of a new type and have some unusual structural features. The structures are elucidated by extensive 1H-NMR studies and some chemical transformations. In addition, a new isoeugenol and an unusual phloroglucinol derivative were isolated; the structure of the latter could only be established after some chemical transformations. Other Calea species contain typical C17 acetylenes only, also present in other species of the subtribe Galinsoginae. So far, the results on Calea indicate that the genus is chemically heterogenous.  相似文献

An unusual m-hydroxyacetophenone, named viscidone, and three new chroman-4-one derivatives were isolated from Chrysothamnus viscidiflorus ssp. lanceolatus, which also contained a known p-hydroxyacetophenone derivative. The structure of withaminimin, a new ergostane-type steroid from Physalis minima, was established by spectral analysis (1H and 13C NMR, MS) and chemical transformations, as (20S,22R)-15α-acetoxy-5α,6β,14α-trihydroxy-1-oxowitha-2,16,24-trienolide. An unusual MH2O+ quasi-molecular ion was observed in the chemical ionization mass spectrum of the natural product.  相似文献

THE unusual modified base1 of the nucleic acid of the large, generalized transducing phage of Bacillus subtilis SP-15 was predicted from the low denaturation temperature of the native nucleic acid. The base was detected as a fifth ultraviolet-absorbing component once the nucleic acid had been hydrolysed with formic acid; it partially replaces the thymine and represents 12 mol per cent of all the bases1. It is responsible for the low Tm of the nucleic acid, since once all other unusual components have been removed, the remaining material melts at the same temperature as the native nucleic acid. We report here the isolation and characterization of this new pyrimidine.  相似文献

Eleven new furostanol saponins, typaspidosides B-L (1–11), one new spirostanol saponin, typaspidoside M (12), and five known spirostanol saponins, 25S-atropuroside (13), neoaspidistrin (14), (25S)-pratioside D1 (15), 25S-aspidistrin (16) and 25S-neosibiricoside (17) were isolated from the rhizomes of Aspidistra typica Baill. The structures of the new compounds were established using 1D and 2D NMR (1H-1H COSY, HMQC, HMBC and ROESY) spectroscopy, high resolution mass spectrometry, and chemical methods. The aglycones of 1–3 (unusual furostanol saponins with opened E ring type), 9 and 10 (the methoxyl substituent at C-23 position) were found, identified from natural products for the first time. Moreover, the anti-HIV activities of the isolated steroidal glycosides were assessed, and compounds 13, 14, 16 and 17 exhibited high active against HIV-1.  相似文献

ATP-binding cassette (ABC) transporters of the cluster 9 family are ubiquitous among bacteria and essential for acquiring Zn2+ and Mn2+ from the environment or, in the case of pathogens, from the host. These rely on a substrate-binding protein (SBP) to coordinate the relevant metal with high affinity and specificity and subsequently release it to a membrane permease for translocation into the cytoplasm. Although a number of cluster 9 SBP structures have been determined, the structural attributes conferring Zn2+ or Mn2+ specificity remain ambiguous. Here we describe the gene expression profile, in vitro metal binding properties, and crystal structure of a new cluster 9 SBP from Paracoccus denitrificans we have called AztC. Although all of our results strongly indicate Zn2+ over Mn2+ specificity, the Zn2+ ion is coordinated by a conserved Asp residue only observed to date as a metal ligand in Mn2+-specific SBPs. The unusual sequence properties of this protein are shared among close homologues, including members from the human pathogens Klebsiella pneumonia and Enterobacter aerogenes, and would seem to suggest a subclass of Zn2+-specific transporters among the cluster 9 family. In any case, the unusual coordination environment of AztC expands the already considerable range of those available to Zn2+-specific SBPs and highlights the presence of a His-rich loop as the most reliable indicator of Zn2+ specificity.  相似文献

Two novel phenotypes previously associated with arl mutations of Escherichia coli, increased frequencies of genetic recombination and unusual sensitivity of DNA to the single-strand-specific nuclease S1, have been defined most completely by the properties of λ bacteriophages grown on arl bacteria (Arl? phages). We now find that plasmids maintained in arl mutants (Arl? plasmids) exhibit elevated recombination frequencies, unusual sensitivity to nuclease S1 (in a limited number of regions) and a new Arl phenotype, partially deficient methylation of the inner cytosine at C-C-(A/T)-G-G sequences.A variety of Arl? plasmids (all pBR322 derivatives) show elevated recombination (4 to 10-fold) by three different assays (frequencies of homomultimers and of heteromultimers, efficiency of intramolecular recombination). Plasmids from arl bacteria (after conversion to linear form) are nicked by nuclease S1 about 0.7 times per duplex; Arl+ plasmids are nuclease S1-resistant. Restriction endonuclease EcoRII (recognition sequence, C-C-(A/T)-G-G) cuts Arl? plasmid DNA more readily than Arl+ DNA, but Arl? plasmids are still more EcoRII-resistant than Dcm? plasmids (from E. coli dcm mutants, which lack the chromosomal cytosine methylase; recognition sequence, also C-C-(A/T)-G-G). By chromatographic analyses, Arl? plasmid DNA contains less 5-methylcytosine than Arl+ (0.07% versus 0.15%). although the 6-methyladenine content is the same (0.5mol%).  相似文献

Several new 4α-methyl sterols with unusual unsaturation in the Δ8(14)-or Δ14-positions, 4α,24S-dimethyl-5α-cholest-8 (14)-en-3β-ol, 4α-methyl-24ξ-ethyl-5α-cholest-8(14)-en-3β-ol, 4α-methyl-24(Z)-ethylidene-5α-cholest-8(14)- en-3β-ol, 4α,23 (or 22),24ξ-trimethyl-5α-cholesta-8(14),22-dien-3β-ol, 4α,24S(or 23ξ)-dimethyl-5α-cholest-14-en-3β-ol and 14-dehydrodinosterol, have been isolated from extracts of the cultured marine dinoflagellates Amphidinium carterae, A. corpulentum and Glenodinium sp. 4α-Methyl-24ξ-ethyl-5α-cholestan-3β-ol was isolated from the steryl ester fraction of Glenodinium sp. The structures of these new sterols are based upon extensive 360 MHz 1H NMR and MS analyses.  相似文献

The investigation of nine Vernonia species afforded in addition to known sesquiterpenes 28 new ones. The structures were elucidated by high field 1H NMR spectroscopy and the configurations were determined by NOE difference spectroscopy and, in one case, by X-ray analysis. The results indicated that configurations of several previously reported sesquiterpene lactones have had to be revised. In addition to known types two new ones, the jalcaguaianolides and the vernojalcanolides, are described. Furthermore some unusual reaction products are presented which, in part, led to some natural occurring lactones.  相似文献

Assembly of N,N′-bis(4-picolinoyl)hydrazine (H2L) with cadmium nitrate in the presence of dicyanamide anion (dca) affords a new coordination polymer {[Cd(HL)(dca)] · (H2O)0.5}n (1), in which the [Cd(HL)]n layers are extended by dca bridges to result in a three-dimensional (3-D) coordination framework. The network structure of 1 has unusual (3,5)-connectivity and represents a new type of (4·62)(4·66·83) topology. Two such identical and complementary networks are entangled to generate a twofold parallel interpenetrating supramolecular lattice.  相似文献

The Cys703 to Ile or His mutation within Saccharomyces cerevisiae oxidosqualene-lanosterol cyclase ERG7 (ERG7C703I/H) generates an unusual truncated bicyclic rearranged intermediate, (8R,9R,10R)-polypoda-5,13E,17E,21-tetraen-3β-ol, related to iridal-skeleton triterpenoid. Numerous oxidosqualene-cyclized truncated intermediates, including tricyclic, unrearranged tetracyclic with 17α/β exocyclic hydrocarbon side chain, rearranged tetracyclic, and chair–chair–chair tricyclic intermediates (compounds 39), were also isolated from the ERG7C703X site-saturated mutations or the ERG7F699T/C703I double mutation, indicating the functional role of the Cys703 residue in stabilizing the bicyclic C-8 cation and the rearranged intermediate or interacting with Phe699, and opened a new avenue of engineering ERG7 for producing biological active agents.  相似文献

Bacteremia caused by Clostridium difficile is rare. In this report, we describe a case of C. difficile bacteremia caused by an unusual strain of C. difficile. The isolate contained neither toxin A nor B genes, however, binary toxin genes were present (tcdA, tcdB, cdtA+, cdtB+) and a 7.2-kb element unrelated to the PaLoc was found inserted within the PaLoc integration site. The clinical relevance of the isolate could not be determined.  相似文献

A new seco-cadinane sesquiterpenoid (curcumane C, 1) and a pair of new nor-bisabolene enantiomers [(+)- and (−)-curcumane D, 2a and 2b] were isolated from C. longa. Compound 1 possesses an unusual 4,5-seco-cadinane skeleton with a tetrahydrophthalide moiety, while 2a and 2b contain an unusual 15-nor-bisabolene skeleton with a chromone core. All compounds exhibited significant vasorelaxant effects against KCl-induced contraction of rat aortic rings. Compound 1 also exhibited a vasorelaxant effect against phenylephrine-induced contraction of rat aortic rings. Meanwhile, compound 1 showed a stronger vasorelaxant effect in endothelium-intact rat aortic rings compared with endothelium-denuded rat aortic rings, indicating that vasodilation by 1 involved both endothelium-dependent and endothelium-independent pathways. Furthermore, compound 1 increased the NO content in human umbilical vein endothelial cells and its vasorelaxant effect could be attenuated by treatment with L-NAME, an endothelium NO synthase inhibitor. Thus, the underlying vasodilatory mechanisms of 1 may be mediated via abrogation of extracellular Ca2+ influx and regulation of NO release in vascular endothelial cells.  相似文献

Biosynthesis of the heterocycle, 1H-2,3,5,6-tetrahydro-5-oxo-9,10-dihydroxyimidazo[3,4,5-de]pyrimido[1,2-a]quinolinium-1- carboxylate, produced by iron-deficient Azotobacter vinelandii, Strain 0 was investigated using 14C-labeled aminto acids and formate. The incorporation of 14C is consistent with this unusual heterocycle beinig derived from l-3(3,4-dihydroxyphenyl)alanine, l-asparagine, formate and glycine.  相似文献

Two species, Hymenorus bifurcatus, and H. excavatus are described as new from Guatemala and the new species H. balli from both the state of Chiapas in southern Mexico and Guatemala. These three species are unique among the species of Hymenorus Mulsant, 1851 in the unusual and highly modified fifth ventrites of the male and the modified shape of the female ninth tergites. The unusual sexual characters of the males and females are illustrated with photographs. The usage of the generic names Hymenorus Mulsant versus Hymenophorus Mulsant is discussed.  相似文献

The desmethyl sterol composition of the oomycete Dictyuchus monosporus is unusual in that it is a mixture of 56.9 % Δ5-sterols and 42.6 % Δ7-sterols. The Δ5-sterols are cholesterol, 24 methylenecholesterol and fucosterol; the Δ7-sterols are cholest-7-enol, ergosta-7,24(28)-dienol and stigmasta-7,E-24(28)-dienol. Stigmasta-7,E-24(28)-dienol, is identified for the first time from natural sources. In addition, traces of lanosterol are present.  相似文献

The su+7 nonsense suppressor of Escherichia coli is a mutant tRNATrp that can be aminoacylated with either tryptophan or glutamine. We have compared the ternary complexes of glutaminyl and tryptophanyl-su+7 tRNA with elongation factor Tu and GTP. Glutaminyl-su+7 tRNA binds more strongly than tryptophanyl-su+7 tRNA to EF Tu · GTP. The greatest distinction between the two species of the tRNA is seen in their dissociation rates from the complex, which differ by as much as fivefold. The distinction is affected by pH values around neutrality. These results show that EF Tu can distinguish between two aminoacyl-tRNAs which differ only in the aminoacyl group. The implications for the unusual amino acid specificity of su+7 tRNA are discussed.  相似文献

The specific physicochemical and spectral properties (dipole moments, NMR and fluorescence spectra) of 2- and 4-substituted 9-isothiocyanatoacridines were studied and compared with the series of m, p-substituted phenyl and benzoyl isothiocyanates. The high reactivity of heterocumulene bonds in the NCS group and that of 9 and 10 position in the acridine skeleton were utilized for the synthesis of new types of compounds, e.g. spiro dihydroacridine 9(10H),4′-thiazolines. Iminothiocarbonates or dithiocarbamates are the intermediates of these unusual addition-cyclization reactions. A new reagent, 1-azonium-4-azabicyclo[2.2.2]octane hydrogen-sulfide [DABCOH](+)SH(?), was developed for the preparation of hitherto inaccesible 9-acridinyl dithiocarbamates as well as 3-(9′-acridinyl)-5-substituted tetrahydro-1,3,5-thiadiazine-2-thiones. The reaction of iminothiocarbonates with bromoacetyl bromide represents a general method for the synthesis of 3-substituted 1,3-thiazolidine-2,4-diones from various type of isothiocyanates. The reaction of 9-hydrazinoacridine with 1-isothiocyanato-1-ethoxy-propane affords, instead of the expected 1,2,4-thiazoline derivative, the N-acridinium-9-yl-N’-propylidenehydrazine thiocyanate. The structures of the synthesized compounds were confirmed with IR, NMR, MAS spectra and X-ray analysis.  相似文献

Two-component oxygenases catalyze a wide variety of important oxidation reactions. Recently we characterized a novel arylamine N-oxygenase (PrnD), a new member of the two-component oxygenase family (J. Lee et al., J. Biol. Chem. 280:36719-36728, 2005). Although arylamine N-oxygenases are widespread in nature, aminopyrrolnitrin N-oxygenase (PrnD) represents the only biochemically and mechanistically characterized arylamine N-oxygenase to date. Here we report the use of bioinformatic and biochemical tools to identify and characterize the reductase component (PrnF) involved in the PrnD-catalyzed unusual arylamine oxidation. The prnF gene was identified via sequence analysis of the whole genome of Pseudomonas fluorescens Pf-5 and subsequently cloned and overexpressed in Escherichia coli. The purified PrnF protein catalyzes reduction of flavin adenine dinucleotide (FAD) by NADH with a kcat of 65 s−1 (Km = 3.2 μM for FAD and 43.1 μM for NADH) and supplies reduced FAD to the PrnD oxygenase component. Unlike other known reductases in two-component oxygenase systems, PrnF strictly requires NADH as an electron donor to reduce FAD and requires unusual protein-protein interaction with the PrnD component for the efficient transfer of reduced FAD. This PrnF enzyme represents the first cloned and characterized flavin reductase component in a novel two-component arylamine oxygenase system.  相似文献
