An organometallic complex of nickel has been isolated from nickel-accumulating plants of New Caledonia and has been identified by a combination of infrared spectroscopy, high-voltage paper electrophoresis, gas-liquid chromatography and mass spectrometry. Most of the nickel in Sebertia acuminata, Homalium francii, Homalium guillainii, Homalium kanaliense, Hybanthus austrocaledonicus and Hybanthus caledonicus is found as a negatively-charged citratonickelate (II) complex with Ni(H2O)62+ as the major cationic constituent.  相似文献

The nature of nickel complexes in various Ni accumulating plant species, mainly from New Caledonia, was investigated by techniques including gel chromatography, ion-exchange chromatography, high performance liquid chromatography, and a combination of gas-liquid chromatography and mass spectrometry. Psychotria douarrei contained Ni complexed mainly (63%) as a negatively-charged malate complex balanced by a cationic aquo complex. Phyllanthus serpentinus had anionic Ni bound as 42% citrate and 4O% malate. All other species studied, contained Ni as an anionic citrate complex.  相似文献

By means of two-bottle preference tests, the threshold values in four South American primate species were evaluated for citric acid and compared with those for acetic acid. Abstract. Mutual interactions between cadmium ions, citric acid and xylem cell walls were examined. Cadmium and citric acid were measured as 115Cd and [1,5-14C] citric acid, respectively. Xylem cell walls were obtained by bacterial degradation of tomato stem sections (Lycopersicon esculentum Mill, cv. Tiny Tim), and applied as ion-exchange columns. The xylem column material carried 2·4 dm3 H2O kg?1 dry weight, and was temporarily capable of buffering perfusates at pH 5·7. Sorbed cadmium and citric acid were determined from H2O and HCl rinses after perfusion periods. In all experiments, total cadmium and/or citric acid recoveries were better than 98%, indicating both the effectiveness of the rinses applied and the possibility of full regeneration of the xylem column. The results indicate that the presence of 2·45 mol m?3 citric acid causes an approximately 50% reduction of adsorbed cadmium levels, irrespective of the applied total cadmium concentrations (0·04–0·4 mol m?3 Cd(NO3)2.4H2O). This reduction is probably related to a corresponding reduction to approximately 2% of the control applied free Cd2+ concentration, the latter also independent of the total cadmium concentrations. Furthermore, without inducing positively charged citrate complexes in the applied solution, the presence of cadmium resulted in increased levels of citric acid absorbed in the xylem column. The Donnan Free Space accumulation of citric acid in the presence of Cd(NO3)2.4H2O, observed in the experiments described, could be expressed by its distribution coefficient, as approximately 15 times the control accumulation. These data indicate that the xylem column may operate as a ligand exchanger, suggesting the importance of metal ions for the longitudinal and lateral movement of organic complexing compounds in the xylem.  相似文献

The essential trace element, nickel, is readily taken up by plants. The biochemical properties of the nickel complex in intrinsically labeled potato and alfalfa were compared and contrasted to ionic nickel. Potato and alfalfa exhibit a similar in vivo nickel complex. The approximate mol wt is 2200–2400 daltons. The complex has a lower polarity than thoes ionic nickel. The majority of the complex is a cationic species with a minor anionic species. This is confirmed with electrophoresis, ion exchange chromatography and the two nickel conplexes' affinity for cationic and anionic resins. Protein does not appear to be associated with either complex.  相似文献

Aims:  To evaluate the effect of the extrusion-cooking process with the addition of different acids concentration on the stability of B-aflatoxins in sorghum.
Methods and Results:  Experimental units (EU) of sorghum flour contaminated with B-aflatoxins (140 ppb) were extrusion cooked with aqueous lactic or citric acid at six different concentrations. The effects of the two extrusion variables (moisture content and acid concentration) were analysed as a completely randomized factorial 3 × 6 design. Under some conditions, the aflatoxin reduction is more effective when using aqueous citric acid (up to 92%), than when using aqueous lactic acid (up to 67%).
Conclusions:  With citric acid, some extrusion treatments produced higher aflatoxin degradation rates, than those produced with lactic acid.
Significance and Impact of the Study:  Aflatoxin contamination is a great risk both for human as well as for animal health in underdeveloped countries; consequently, practical and economical detoxification procedures are needed that eliminate or at least minimize the aflatoxin risk, through lowering aflatoxin concentrations in grains. Under these considerations, extrusion process can be used for reduction in the aflatoxin content in contaminated grains.  相似文献

A study was performed to understand the physiology and biochemical mechanism of citric acid accumulation during solid state fermentation of sweet potato using Aspergillus niger Yang No.2. A low citrate-producing mutant was isolated followed by a comparative study of the fermentation process and selected physiological and biochemical parameters. In contrast with the parent strain, the mutant strain displayed lower concentrations, yields and production rates of citric acid, accompanied by higher concentrations, yields and production rates of oxalic acid. In addition, the mutant utilized starch at a lower rate although higher concentrations of free glucose accumulated in the cultures. Biochemical analyses revealed lower rates of glucose uptake and hexokinase activity of the mutant strain in comparison with the parent strain. It is proposed that, in common with submerged fermentation, over-production of citric acid in solid state fermentation is related to an increased glucose flux through glycolysis. At low glucose fluxes, oxalic acid is accumulated.  相似文献

In this paper we report the regulation of Aspergillus niger growth rate during citric acid fermentation in a stirred tank bioreactor. For this, the influence of dissolved oxygen concentration in a medium on intracellular pH values and consequently on overall microbial metabolism was emphasized. Intracellular pH of mycelium grown under different concentrations of dissolved oxygen in the medium was determined. Sensitivity of proteins toward proton concentration is well recognized, therefore pH influences on the activities of key regulatory enzymes of Aspergillus niger were determined at pH values similar to those detected in the cells grown under lower dissolved oxygen concentrations. The results have shown significantly reduced specific activities of hexokinase, 6-phosphofructokinase and glucose-6-phosphate dehydrogenase in more acidic environment, while pyruvate kinase was found to be relatively insensitive towards higher proton concentration. As expected, due to the reduced specific activities of regulatory enzymes under more acidic conditions, overall metabolism should be hindered in the medium with lower dissolved oxygen concentration which was confirmed by detecting the reduced specific growth rates. From the studies, we conclude that dissolved oxygen concentration affects the intracellular pH and thus growth rate of Aspergillus niger during the fermentation process.  相似文献

Enzymatic studies have been performed on a local strain of Aspergillus niger to find a correlation with citric acid accumulation. The activity of aconitase [aconitate hydratase, citrate(isocitrate) hydrolyase, EC 4.2.1.3] and isocitrate dehydrogenase (NADP+) [threo-ds-isocitrate:NADP+ oxidoreductase (decarboxylating) EC 1.1.1.42] decreased after 4 days whereas that of citrate synthase [citrate oxaloacetate-lyase (pro-3S-CH2COO?acetylCoA), EC 4.1.3.7] did so after 8 days, when citric acid accumulation in the medium reached a maximum (45.9 mg ml?1). In vitro studies with mycelial cell-free extracts demonstrated inhibition of citrate synthase activity by sodium azide and potassium ferricyanide on both the 4th and 8th days. Aconitase was inhibited by sodium arsenate, sodium fluoride, iodoacetic acid and potassium ferricyanide only on the 4th day. Isocitrate dehydrogenase (NADP+) activity on the 4th and 8th days was inhibited by iodoacetic acid but was stimulated by potassium ferricyanide. The possible existence of isozyme species of these enzymes is discussed.  相似文献

The interaction between nickel and iron was confirmed in rat metabolism. In a fully-crossed, two-way, three by four, factorially designed experiment, female weanling rats were fed a basal diet supplemented with iron at 0, 25, 50, and 100 μg/g and with nickel at 0, 5, and 50 μg/g. The basal diet contained about 10 ng of nickel and 2.3 μg of iron/g. After nine weeks, dietary iron affected growth, hematocrit, hemoglobin, plasma cholesterol, and in liver affected total lipids, phospholipids, and the contents of copper, iron, manganese, and zinc. By manipulating the iron content of the diet, effects of dietary nickel were shown in rats that were not from dams fed a nickel-deprived diet. Nickel affected growth, hematocrit, hemoglobin, plasma alkaline phosphatase activity, plasma total lipids, and in liver affected total lipids, and the contents of copper, manganese, and nickel. The interaction between nickel and iron affected hematocrit, hemoglobin, plasma alkaline phosphatase activity, and plasma phospholipids, and in liver affected size, content of copper, and perhaps of manganese and nickel. In severely iron-deficient rats, the high level of dietary nickel partially alleviated the drastic depression of hematocrit and hemoglobin, and the elevation of copper in liver. Simultaneously, high dietary nickel did not increase the iron level in liver and was detrimental to growth and appearance of severely iron-deficient rats. In nickel-deprived rats fed the borderline iron-deficient diet (25 μg/g) hematocrit and hemoglobin also were depressed. However, 5 μg Ni/g of diet were just as effective as 50 μg Ni/g of diet in preventing those signs of nickel deprivation. The findings in the present study suggested that nickel and iron interact with each other at more than one locus.  相似文献

The generation of free radicals (Fenton chemistry) from various iron citrate complexes has been studied. Spin trapping methods have been used. The results can question concerning the innocence of added citric acid in foods and cold drinks. We concluded that in absence of pathological situation citric acid is probably not dangerous but it may become dangerous in situation of oxidative stress and/or iron overload.  相似文献

Aspergillus foetidus ACM 3996 (=FRR 3558) and three strains of Aspergillus niger ACM 4992 (=ATCC 9142), ACM 4993 (=ATCC 10577), ACM 4994 (=ATCC 12846) were compared for the production of citric acid from pineapple peel in solid-state fermentation. A. niger ACM 4992 produced the highest amount of citric acid, with a yield of 19.4g of citric acid per 100g of dry fermented pineapple waste under optimum conditions, representing a yield of 0.74g citric acid/g sugar consumed. Optimal conditions were 65% (w/w) initial moisture content, 3% (v/w) methanol, 30°C, an unadjusted initial pH of 3.4, a particle size of 2mm and 5ppm Fe2+. Citric acid production was best in flasks, with lower yields being obtained in tray and rotating drum bioreactors.  相似文献

The ameliorating effects of four exogenous effectors were investigated in germinating pea seeds exposed to copper excess. The results showed that the application of IAA, GA3, Ca or citric acid alleviated Cu-induced inhibition of growth and simultaneously reduced the oxidative stress injury, particularly contents of hydrogen peroxide, malondialdehyde and carbonyl groups. The improving effects can probably be mediated by the decreases in lipoperoxidation and protein oxidation as evidenced by changes in antioxidant enzyme activities. In addition, the efficiency of this recovery was compared within two types of treatments. Obtained results demonstrated that the stress abruption by the addition of effectors after three days of Cu application (treatment of type II) seems to be more effective than the simultaneous application of ‘Cu?+?effectors’ at the beginning of germination (treatment of type I). Data could provide some clues to physiological and biochemical mechanisms of the response of germinating seeds to the addition of chemicals under heavy metal stress.  相似文献

Phytoextraction, the use of plants to extract heavy metals from contaminated soils, could be an interesting alternative to conventional remediation technologies. However, calcareous soils with relatively high total metal contents are difficult to phytoremediate due to low soluble metal concentrations. Soil amendments such as ethylene diaminetetraacetate (EDTA) have been suggested to increase heavy metal bioavailability and uptake in aboveground plant parts. Strong persistence of EDTA and risks of leaching of potentially toxic metals and essential nutrients have led to research on easily biodegradable soil amendments such as citric acid. In our research, EDTA is regarded as a scientific benchmark with which degradable alternatives are compared for enhanced phytoextraction purposes. The effects of increasing doses of EDTA (0.1,1,10 mmol kg(-1) dry soil) and citric acid (0.01, 0.05, 0.25, 0.442, 0.5 mol kg(-1) dry soil) on bioavailable fractions of Cu, Zn, Cd, and Pb were assessed in one part of our study and results are presented in this article. The evolution of labile soil fractions of heavy metals over time was evaluated using water paste saturation extraction (approximately soluble fraction), extraction with 1 M NH4OAc at pH 7 (approximately exchangeable fraction), and extraction with 0.5 M NH4OAc + 05 M HOAc + 0.02 M EDTA at pH 4.65 (approximately potentially bioavailable fraction). Both citric acid and EDTA produced a rapid initial increase in labile heavy metal fractions. Metal mobilization remained constant in time for soils treated with EDTA, but a strong exponential decrease of labile metal fractions was noted for soils treated with citric acid. The half life of heavy metal mobilization by citric acid varied between 1.5 and 5.7 d. The chemoautotrophic theory concerning the origin of life postulates that a central role is played in the prebiotic chemical machinery by a reductive citric acid cycle operating without enzymes. The crucial point in this scenario is the formation of pyrite from hydrogen sulfide and ferrous sulfide, a reaction suggested to be linked to endergonic reactions, making them exergonic. This mechanism is believed to provide the driving force for the cycle to operate as a carbon dioxide fixation network. The present paper criticizes the thermodynamic calculations and their presentation in the original version of the archaic reductive citric acid cycle [W?chtersh?user, 1990. Evolution of the first metabolic cycles. Proc. Natl Acad. Sci. USA 87, 200-204.]. The most significant differences between the W?chtersh?user hypothesis and the present proposal: W?chtersh?user did not consider individual reactions in his calculations. A particularly questionable feature is the involvement of seven molecules of pyrite which does not emerge as a direct consequence of the chemical reactions presented in the archaic reductive citric acid cycle. The involvement of a considerable number of sulfur-containing organic intermediates as building blocks is also disputed. In the new scheme of the cycle proposed here, less free energy is liberated than hypothesized by W?chtersh?user, but it has the advantages that the free energy changes for the individual reactions can be calculated, the number of pyrite molecules involved in the cycle is reduced, and fewer sulfur-containing intermediates are required for the cycle to operate. In combination with a plausible route for the anaplerotic reactions [Kalapos, 1997a. Possible evolutionary role of methylglyoxalase pathway: anaplerotic route for reductive citric acid cycle of surface metabolists. J. Theor. Biol. 188, 201-206.], this new presentation of the cycle assigns a special meaning to hydrogen sulfide formation in the early stage of biochemical evolution.  相似文献

Three experiments were conducted with chicks to examine the effects of dietary iron and ascorbic acid on the accumulation of lead in various organs. Lead was fed as PbCl2, 500 or 1000 ppm Pb, iron as FeSO4·7H2O, 1000 ppm Fe, and ascorbic acid at 0.5%. Iron was effective in reducing the accumulation of lead in the femur and kidneys at both levels of lead. Ascorbic acid reduced the lead level in the kidneys when the concentration of lead in the diet was 500 ppm, but not at 1000 ppm. The effects of ascorbic acid on bone accumulation was variable. In two experiments the lead concentration was increased and in one it was decreased. These findings may reflect two influences of ascorbic acid found by others, namely an increase in absorption and an increase in urinary excretion. The rapid accumulation of lead in chick bones suggests that it may be an excellent experimental animal for lead studies.  相似文献
