Background and aims

Subsoil acidity with a high aluminium (Al3+) soil content inhibits root growth and proliferation of durum wheat (tetraploid AABB, Triticum turgidum) leading to poor nutrient and water uptake. This study evaluated the impact of Al3+-tolerantTaMATE1B allele on root and shoot traits of durum wheat grown in an acidic soil with a high Al3+concentration.

Methods

Two durum wheat lines, Jandaroi–TaMATE1B with the TaMATE1B gene introgressed from Al3+-tolerant bread wheat and Jandaroi–null (a sister line lacking the Al3+-tolerant TaMATE1B allele), were grown in rhizoboxes in a glasshouse. We mapped root growth and proliferation over time and measured shoot traits and grain yield.

Results

Introgression of the Al3+-tolerant TaMATE1B allele into durum wheat enabled root growth and proliferation below 0.25 m of the soil profile, where the soil pH was low (4.1, CaCl2 extract) with high Al3+ content (16.5 mg kg−1), and increased total root length and biomass at 42 days after sowing (DAS; Z33) by 38.3 and 22%, respectively, relative to the Jandaroi–null. Differences in root growth between the two lines were apparent from tillering stage (Z33) and by 50% anthesis (Z64), respectively. Jandaroi–TaMATE1B had 69.2% greater root biomass, 76.2% greater root length, 5.89% greater leaf area and 18% greater shoot biomass than Jandaroi–null at 50% anthesis (Z64). Time to anthesis and physiological maturity was delayed 6–7 days in Jandaroi–TaMATE1B, compared to Jandaroi–null. Jandaroi–TaMATE1B tended to have relatively greater, but not significantly different, shoot biomass, grain yield and yield components than Jandaroi–null.

Conclusions

Introgression of the Al3+-tolerant TaMATE1B allele into durum wheat enabled root growth and proliferation down an acidic soil profile with a high Al3+ concentration. We assume that in the field where plants need to acquire water at depth differences in above-ground parameters would be amplified.

The thermodynamic functions of biopolymer hydration were investigated by multitemperature vapor pressure studies. Desorption measurements were performed that allowed determination of reversible isotherms in the hydration range of 0.1 to 0.3–0.5 g H2O/g dry polymer. These isotherms are accessible to thermodynamic interpretation and are relevant to the interaction of water with biopolymers in their solution conformation. The results obtained on a series of different biopolymers (lysozyme, α-chymotrypsin, apo-lactoferrin, and desoxyribonucleic acid), show the following common features of interest: (1) The differential excess enthalpies (ΔHe ) and entropies (ΔSe ) are negative, and exhibit pronounced anomalies in a well-defined low-humidity range (approx. 0.1 g H2O/g dry polymer). These initial extrema are interpretable by structural changes, induced in the native biopolymer structures by water removal below a critical degree of hydration. (2) The ΔHe and ΔSe terms exhibit statistically significant linear enthalpy–entropy compensation effects in all biopolymer–water systems investigated. The compensation temperatures \documentclass{article}\pagestyle{empty}\begin{document}$ \hat \beta = \overline {\Delta H} ^e /\overline {\Delta S} ^e $\end{document} are approximately identical for all biopolymers, ranging from 360 to 500 K. The compensation effects are attributable to phase transitions of water molecules between the bulk liquid and the inner-sphere hydration shell of native biopolymers. (3) The negative excess free energies (ΔGe ) decrease monotonically with increasing water content and are close to zero at 0.3 to 0.5 g H2O/g polymer. This result indicates that only transitions between the bulk liquid and the inner-sphere hydration shell are associated with significant net free energy effects. The outer-sphere hydration water is thermodynamically comparable to bulk water. Background and aims

Arsenate (AsV) and chromate (CrVI) inhibit each other’s uptake and translocation in As-hyperaccumulator Pteris vittata. In the present study, we extended the research to As-sensitive plant Pteris ensiformis to better understand the mechanism of their interactions.

Methods

Plants were exposed to 0, 0.75 or 7.5 mg L?1 AsV and 0, 0.52, or 5.2 mg L?1 CrVI for 7 d in hydroponics. Arsenic and Cr speciation were determined in nutrient solutions and plant biomass.

Results

P. ensiformis accumulated high levels of As and Cr in the rhizomes and roots with low levels in the fronds. However, P. ensiformis was much more effective in taking up Cr than As, as much more Cr was accumulated in the roots (306–6015 vs. 87–642 mg kg?1). AsV and CrVI increased each other’s uptake in the rhizomes and roots when co-present. The AsV and CrVI taken up by P. ensiformis were reduced to arsenite (AsIII) and chromite (CrIII), possibly serving as detoxification mechanism.

Conclusions

Uptake of As and Cr induced oxidative stress as indicated by increased lipid peroxidation and electrical conductivity. Arsenic and Cr increased each other’s uptake by P. ensiformis.
