This paper studies aspects of the distribution of non-mutant ancestors of a sample of gametes in a two-locus infinitely-many-alleles model. The ancestral process of two gametes is considered in detail. Included are algorithms for calculating the probability that the oldest allele is from the first locus, and the expected age of the oldest allele. Extensions to an r-locus model in the cases of complete linkage and independence are also studied.  相似文献

The Bgs locus determines tissue levels of β-galactosidase in the mouse, so that enzyme levels are twice as high in mice carrying the Bgs hallele as in mice carrying the Bgs dallele (Felton et al., 1974). By immunotitration with antiserum to purified β-galactosidase, we have found that the Bgs locus influences the amount of enzyme protein present in the tissues. We have utilized recombinant inbred lines derived from a cross between C57BL/6J and DBA/2J mice to confirm the location of the Bgs locus on chromosome 9. The inhibition of mouse β-galactosidase by the active-site-directed reagent N-bromoacetyl-β-d-galactosylamine has been investigated. β-Galactosidase from the high and low Bgs strains has identical affinity for this inhibitor.  相似文献

Summary The placental phosphoglucomutase phenotypes controlled by the first locus of 235 German and 119 non-German samples were determined. Both in the pooled material and in the sample containing German subjects only there was a significant deviation from the expected Hardy-Weinberg equilibrium. A segregation analysis of 1174 families also revealed significant deviations in the phenotypes of the children, though the mating frequencies were as expected. The distribution of the children's phenotypes deviated significantly from Hardy-Weinberg equilibrium though this was not found in the parent samples. The results are discussed.
Zusammenfassung Placenta-Phosphoglucomutase-Phänotypen des ersten Locus wurden an Hand von 235 deutschen und 119 ausländischen Stichproben bestimmt. Sowohl das Gesamtmaterial als auch die deutsche Stichprobe allein zeigten eine signifikante Abweichung vom angenommenen Hardy-Weinberg-Gleichgewicht. Eine Aufspaltungsanalyse bei 1174 Familien ergab ebenfalls eine signifikante Abweichung bei den Phänotypen der Kinder, obgleich die Elternkombinationshäufigkeiten der Erwartung entsprachen. Die Verteilung der Kinderphänotypen wich signifikant vom Hardy-Weinberg-Gleichgewicht ab, während die Verteilung der Elternphänotypen mit den Erwartungswerten übereinstimmte. Die Ergebnisse werden diskutiert.

A current trend in physiology education involves the use of clinical vignettes to demonstrate the importance of knowing normal physiology to appreciate pathophysiology. Although laudable, in effect, such tactics promote the so-called "disease" model of medicine while at the same time suggesting that the only utility for the knowledge of physiology is to understand pathophysiology. This would seem to be at odds with health professions and institutions, who maintain their goal is to promote health. Yet, a search for the locus of "health" education in typical curricula is not easily found. Given the developing interest in biological systems as well as aging, it is suggested that these topics may provide a basis for locating physiology as the locus for understanding "health."  相似文献

AD7c-NTP, the only known protein entirely encoded by tandem and nested cassettes of Alu repetitive elements, is reportedly over-expressed in brains of Alzheimer's disease patients [de la Monte et al., J. Clin. Invest. 15 (1997)]. Based on these findings a commercial diagnostic assay ("7c Gold"/"AlzheimAlert" ) has been developed. We analyzed the published cDNA sequence and compared it to corresponding EST clones as well as the genomic sequences of human and chimpanzee. We come to the conclusion that the existence of the gene and in particular the predicted protein is inconsistent with EST and genomic data. Previously published data need to be reassessed.  相似文献

We have combined the circular chromosome conformation capture protocol with high-throughput, genome-wide sequence analysis to characterize the cis-acting regulatory network at a single locus. In contrast to methods which identify large interacting regions (10–1000 kb), the 4C approach provides a comprehensive, high-resolution analysis of a specific locus with the aim of defining, in detail, the cis-regulatory elements controlling a single gene or gene cluster. Using the human α-globin locus as a model, we detected all known local and long-range interactions with this gene cluster. In addition, we identified two interactions with genes located 300 kb (NME4) and 625 kb (FAM173a) from the α-globin cluster.  相似文献

Camurati-Engelmann disease (CED) [OMIM 131300] is an autosomal dominant sclerosing bone dysplasia recently ascribed to mutations of the transforming growth factor (TGF-beta1) gene on chromosome 19q13.1-q13.3. Five mutations consistently located in the TGF-beta1 propeptide have been hitherto identified in 21 families. Here, we report on TGF-beta1 mutations in one Australian and six European families. Three distinct mutations were identified among seven families: namely, R218H (family 1), R218C (families 2, 6, 7) and C225R (families 3, 4, 5). The three mutations identified in our pedigrees have been previously observed in families of Japanese and Israeli origin and the R218C appears to be the most prevalent mutation worldwide (17/28 reported families). No obvious correlation between the nature of the mutations and the severity of the clinical manifestations could be established, but a marked intrafamilial clinical variability was observed, supporting incomplete penetrance of CED. Interestingly, the polymorphisms in the TGF-beta1 gene showed no correlation with the severity of the disease. We conclude that CED is a clinically variable condition and that this clinical variability is not accounted for by polymorphisms at the TGF-beta1 locus.  相似文献

Background  

We have examined the evolution of the genes at the major human β-defensin locus and the orthologous loci in a range of other primates and mouse. For the first time these data allow us to examine selective episodes in the more recent evolutionary history of this locus as well as the ancient past. We have used a combination of maximum likelihood based tests and a maximum parsimony based sliding window approach to give a detailed view of the varying modes of selection operating at this locus.  相似文献

Schnyders crystalline corneal dystrophy (SCCD) is a rare autosomal dominant eye disease with a spectrum of clinical manifestations that may include bilateral corneal clouding, arcus lipoides, and anterior corneal crystalline cholesterol deposition. We have previously performed a genome-wide linkage analysis on two large Swede-Finn families and mapped the SCCD locus to a 16-cM interval between markers D1S2633 and D1S228 on chromosome 1p36. We have collected 11 additional families from Finland, Germany, Turkey, and USA to narrow the critical region for SCCD. Here, we have used haplotype analysis with densely spaced microsatellite markers in a total of 13 families to refine the candidate interval. A common disease haplotype was observed among the four Swede-Finn families indicating the presence of a founder effect. Recombination results from all 13 families refined the SCCD locus to 2.32 Mbp between markers D1S1160 and D1S1635. Within this interval, identity-by-state was present in all 13 families for two markers D1S244 and D1S3153, further refining the candidate region to 1.58 Mbp.  相似文献

DNA variability was investigated in the last intron of the Y-chromosome-specific zinc finger gene, ZFY, and its X homolog on Xp21.3, ZFX. No polymorphisms were found in the 676-bp ZFY segment in a sample of 205 world-wide-distributed Y chromosomes, other than a solitary nucleotide variant in one individual (nucleotide diversity pi = 0.0014%). In contrast, 10 segregating sites (pi = 0.082%) were identified within 1,089 bp of the ZFX sequence in a sample of 336 X chromosomes. Four of these polymorphisms, which contributed most of the diversity, were located within an Alu insert disrupting the ZFY-ZFX homology (pi Alu = 0.24%). The diversity in the homologous portion of the ZFX intron, although higher than that in ZFY, was lower than that found in genomic segments believed to evolve neutrally; interspecies divergence in both segments was also reduced. Although this suggests that the evolution of both ZFY and ZFX homologs may not be entirely neutral, both Tajima and HKA tests did not reject neutrality. The lack of statistical significance may be attributed to a lack of power in these tests (the low divergence and variability values reduce the power of the HKA and Tajima tests, respectively); furthermore, Homo sapiens has recently undergone a rapid population growth, and selection is more difficult to detect in an expanding population. Therefore, the failure to reject neutrality does not necessarily indicate the absence of selection. In this context, the phylogenetic argument was given more weight in out interpretations. The high level of sequence identity in ZFY and ZFX segments, in spite of their separation 80-130 MYA, reflects a lower mutation rate as compared with other segments believed to undergo unconstrained evolution. Thus, the possibility of weak selection contributing to the low level of nucleotide diversity in the last ZFY intron cannot be excluded and should be kept in mind in the population genetics studies based on Y chromosome variability.  相似文献

Three sets of experiments have been conducted in order to evaluate the role of natural selection at the -Gpdh locus in Drosophila melanogaster. (1) The evolution of the F-allele frequency has been followed for many generations in 13 experimental populations having different genetic backgrounds. (2) Egg-to-adult viability has been measured in synthetic populations derived from one locality (Brouilly) and the results have been compared with those of a previous experiment involving a different local population (Tostes). (3) The effects of sodium octanoate on egg-to-adult viability have been measured on the genotypes FF, FS, SF, and SS. Summary Chromosomes from 19 unrelated Southern Yugoslav families in which cystic fibrosis (CF) occurs were analysed for the presence of the ΔF508 mutation, using polymerase chain reaction amplification followed by dot blot and polyacrylamide gel analysis. Of the 38 CF chromosomes, 15 (39.5%) carry the ΔF508 deletion. Restriction fragment length polymorphism haplotypes for KM19/PstI, XV2c/TaqI and J3.11/PstI marker loci were determined and are compared for a total of 34 N and 37 CF chromosomes.  相似文献
