Cellulosic nanomaterials provide a novel and sustainable platform for the production of high performance materials enabled by nanotechnology. Bacterial cellulose (BC) is a highly crystalline material and contains pure cellulose without lignin and hemicellulose. BC offers an opportunity to provide control of the products’ properties in-situ, via specific BC production methods and culture conditions. The BC potential in advanced material applications are hindered by a limited knowledge of optimal BC production conditions, efficient process scale-up, separation methods, and purification methods. There is a growing body of work on the production of bacterial cellulose nanocrystals (BCNs) from BC fibers. However, there is limited information regarding the effect of BC fibers’ characteristics on the production of nanocrystals. This review describes developments in BC and BCNs production methods and factors affecting their yield and physical characteristics.  相似文献

Rhizotrons provide valuable information about plant root production, but measurements are usually made in units of root length per unit surface area of observation window surface. These measurement units are not easily comparable to above-ground plant growth. To address this deficiency, several techniques have been developed to convert rhizotron measurement units into root mass production per unit ground area. In this study, four different conversion methods were applied to the same dataset of rhizotron measurements. This data was used to reveal the effect of conversion method upon estimates of the temporal variation in, and annual magnitude of, gross root mass production. Application of four different conversion methods resulted in gross root production estimates ranging between 2.1 and 11.4 t ha−1 year−1. Temporal variation in gross root mass production also varied between methods. All current methods for quantifying root production are likely to cause some disturbance and bias. Based upon a comparison of the sources of error present in each conversion method, we assess which methods are likely to produce the most reliable estimates of root biomass production per unit ground area, and propose additional measurements which could further improve accuracy.  相似文献

Many different cultivation techniques and inoculum products of the plant-beneficial arbuscular mycorrhizal (AM) fungi have been developed in the last decades. Soil- and substrate-based production techniques as well as substrate-free culture techniques (hydroponics and aeroponics) and in vitro cultivation methods have all been attempted for the large-scale production of AM fungi. In this review, we describe the principal in vivo and in vitro production methods that have been developed so far. We present the parameters that are critical for optimal production, discuss the advantages and disadvantages of the methods, and highlight their most probable sectors of application.  相似文献

The production and turnover of fine roots play substantial roles in the biogeochemical cycles of terrestrial ecosystems. However, the disparity among the estimates of both production and turnover, particularly due to technical limitations, has been debated for several decades. Here, we conducted a meta-analysis to compare published estimates of fine root production and turnover rates derived from different methods at the same sites and at the same sampling time. On average, the estimates of fine root production and turnover rates were 87% and 124% higher, respectively, by indirect methods than by direct methods. The substantially higher fine root production and turnover estimated by indirect methods, on which most global carbon models are based, indicate the necessity of re-assessing the global carbon model predictions for atmospheric carbon sequestration in soils as a result of the production and turnover of fine roots.  相似文献

Hydrogen is a promising energy source that is believed to replace the conventional energy sources e.g. fossil fuels over years. Hydrogen production methods can be divided into conventional production methods which depend mainly on fossil fuels and alternative production methods including electrolysis of water, biophotolysis and fermentation hydrogen production from organic waste materials. Compared to the conventional methods, the alternative hydrogen production methods are less energy intensive and negative-value substrates i.e. waste materials can be used to produce hydrogen. Among the alternative methods, fermentation process including dark and photo-fermentation has gained more attention because these processes are simple, waste materials can be utilized, and high hydrogen yields can be achieved. The fermentation process is affected by several parameters such as type of inoculum, pH, temperature, substrate type and concentration, hydraulic retention time, etc. In order to achieve optimum hydrogen yields and maximum substrate degradation, the operating conditions of the fermentation process must be optimized. In this review, two routes for biohydrogen production as dark and photo-fermentation are discussed. Dark/photo-fermentation technology is a new approach that can be used to increase the hydrogen yield and improve the energy recovery from organic wastes.  相似文献

There are alternative methods for estimation of phytoplankton primary production (PP) that are fundamentally different in the calculation approach. The process‐oriented PP model is a mechanistic, empirically derived method based on the photosynthesis–light relationships. The population dynamics‐based PP calculation, which is a synthetic method, provides a production estimate based on population dynamics of phytoplankton. These alternative methods were here compared with regard to production estimates and linked to enhance the performance of the existing models of population dynamics applied to a wide variety of lakes worldwide in terms of morphometry, nutrient status, and light environments. Estimates of PP were shown to be sensitive to changes in phytoplankton sinking and zooplankton grazing rates in both methods. Production estimates in the process‐oriented PP model were also sensitive to light‐associated parameters such as day length. Although the production estimated from the population dynamics‐based PP calculation tended to be lower than that from the process‐oriented PP model irrespective of lake morphometry, production estimates calculated from both methods with standard parameterization were comparable when production was estimated on an annual timescale. However, it was also shown that the alternative methods could produce different production estimates when estimated on shorter timescales such as cyanobacterial blooms in summer. Cyanobacteria with low mortality due to grazing and sinking losses have been considered as trophic bottlenecks, but there is increasing evidence that their mortality is, to a considerable extent, due to parasitic pathogens. In the case of cyanobacterial blooms, an addition of parasite‐related loss term (19%–33% of standing stock) resulted in a resolution of the difference in production estimates between the methods. These analyses theoretically support the critical role of parasitism and resolve the bottleneck problem in aquatic ecosystem metabolism.  相似文献

Summary Interest of microbial production of amino acids has been increased greatly since development of biotechnological methods. These methods represent a perspective way applied in a future large-scale manufacture of inexpensive amino acids. In this context, the isolation of producing organisms that may be exploited in the desing of alternative methods for the production of amino acids could be of primary importance.In this review we will describe the liberation of amino acids (methionine, lysine, arginine, tryptophane and glutamic acid) byAzotobacter andAzospirillum during growth in culture media with different carbon sources under diazotrophic and adiazotrophic conditions. Three methods to detect biosurfactant production, drop collapse, oil spreading, and blood agar lysis, were compared for their ease of use and reliability in relation to the ability of the cultures to reduce surface tension. The three methods were used to test for biosurfactant production in 205 environmental strains with different phylogenetic affiliations. Surface tension of select strains that gave conflicting results with the above three methods was also measured. Sixteen percent of the strains that lysed blood agar tested negative for biosurfactant production with the other two methods and had little reduction in surface tension (values above 60 mN/m). Thirty eight percent of the strains that did not lyse blood agar tested positive for biosurfactant production with the other two methods and had surface tension values as low as 35 mN/m. There was a very strong, negative, linear correlation between the diameter of clear zone obtained with the oil spreading technique and surface tension (rs = -0.959) and a weaker negative correlation between drop collapse method and surface tension (rs = -0.82), suggesting that the oil spreading technique better predicted biosurfactant production than the drop collapse method. The use of the drop collapse method as a primary method to detect biosurfactant producers, followed by the determination of the biosurfactant concentration using the oil spreading technique, constitutes a quick and easy protocol to screen and quantify biosurfactant production. The large number of false negatives and positives obtained with the blood agar lysis method and its poor correlation to surface tension (rs = -0.15) demonstrated that it is not a reliable method to detect biosurfactant production.  相似文献

Water scarcity and pollution have become increasingly serious because of industrial production. Thus, a tool should be developed to evaluate the level of water use of industrial production. Several calculation methods for water use relative to water footprint have been developed by many scholars. However, most of these methods still have defects in terms of indicators, parameters, and calculation methods. In this paper, we established an evaluation tool that is suitable for the current production situation of industrial products, i.e., a water management tool of industrial products that includes two parts: direct water use caused by the production process of industrial products and indirect water use caused by the energy and material used during production. Each part contains three indicators: water withdrawal, water consumption, and water assimilation. We applied the tool in the textile and apparel industry to calculate the water use of screen and digital printing fabrics, as well as the production processes for both printing fabrics. Results show that the tool can highlight the water intensive product and production process, which can help enterprises take effective and specific measures to save water and reduce pollution.  相似文献

The objective of this study was to investigate the production of Calanus helgolandicus off Plymouth, Western English Channel, from April to August 1998 comparing a range of different methods. The abundance of all developmental stages from NI to adult was determined. Body carbon and nitrogen and length were measured on individual NVI to adults. Female egg production was also monitored. All developmental stages were generally represented but four main periods of recruitment were determined. Two periods of low egg production were observed on the 13th and on the 27th of July: (i) the first was due to the arrival of newly moulted or not yet fertilised females in the population and (ii) the second low period was possibly due to the senescence of the females. Finally, the total production of C. helgolandicus was estimated using four different methods: the weight increment, the egg production, Huntley and Lopez and Hirst and Lampitt methods. Depending on the methods used, the production accumulated over the period from May 14 to August 10 ranged between 12 and 45 mg C m−3.  相似文献

The production of biodiesel by transesterification employing acid or base catalyst has been industrially accepted for its high conversion and reaction rates. Downstream processing costs and environmental problems associated with biodiesel production and byproducts recovery have led to the search for alternative production methods. Recently, enzymatic transesterification involving lipases has attracted attention for biodiesel production as it produces high purity product and enables easy separation from the byproduct, glycerol. The use of immobilized lipases and immobilized whole cells may lower the overall cost, while presenting less downstream processing problems, to biodiesel production. The present review gives an overview on biodiesel production technology and analyzes the factors/methods of enzymatic approach reported in the literature and also suggests suitable method on the basis of evidence for industrial production of biodiesel.  相似文献

Cell and enzyme immobilization are often used for industrial production of high-value products. In recent years, immobilization techniques have been applied to the production of value-added chemicals such as 1,3-Propanediol (1,3-PDO). Biotechnological fermentation is an attractive alternative to current 1,3-PDO production methods, which are primarily thermochemical processes, as it generates high volumetric yields of 1,3-PDO, is a much less energy intensive process, and generates lower amounts of environmental organic pollutants. Although several approaches including: batch, fed-batch, continuous-feed and two-step continuous-feed were tested in suspended systems, it has been well demonstrated that cell immobilization techniques can significantly enhance 1,3-PDO production and allow robust continuous production in smaller bioreactors. We describe methods for the production of fluorescence in situ hybridization (FISH) probes and the utilization of these probes for the detection of complementary DNA sequences with accuracy and sensitivity for application in both basic research and clinical diagnosis. Due to the frequent use of FISH in many laboratories, it is important to apply the most convenient and reproducible approach. This review describes some of the most recent techniques, and includes versatile, effective and simple methods of probe production and fluorescence in situ hybridization. The effectiveness of the purification of proteolytic enzyme produced by B. subtilis production strain 3H in different nutrient media was evaluated. As revealed in this study, the use of the initial material obtained in peptone nutrient medium made it possible to obtain proteolytic enzyme with the highest specific activity by the methods of salting out, gel and ion-exchange chromatography, but the use of these methods on a production scale led to the deterioration of purity characteristics. Changes in the nutrient medium composition used for the cultivation of the production strain resulted in greater effectiveness of the purification methods. The highly purified preparation of metalloprotease with specific activity exceeding that of available commercial preparations more than twofold was obtained.  相似文献

消化率的高低是评价饲料营养价值的重要指标,但是粪便的不同收集方式会影响消化率的测定结果。研究观察了异育银鲫摄食后的排粪高峰以及排粪持续时间,探讨了粪便的不同收集方式对异育银鲫(Carassius auratus gibelio)(约100 g)表观消化率(Apparent digestibility coefficient,ADC)测定结果的影响。饲料中加入1%三氧化二铬(Cr2O3)作为指示剂,实验采用3种粪便收集方法(积粪器法、虹吸法和后肠取粪法),并且在积粪器法和虹吸法中设置了不同的收粪时间。结果表明,正常投喂的异育银鲫摄食后的排粪高峰出现在摄食结束后的5—10h;饥饿2d的异育银鲫恢复摄食后的排粪高峰出现在摄食结束后的7—9h;饲料的干物质表观消化率(ADC of dry matter,ADCd)和蛋白表观消化率(ADC of protein,ADCp)在排粪初始阶段较低,随着排粪高峰的出现而升高。粪便的不同收集方法和不同收集时间对ADCd和ADCp的测定结果均存在显著影响(P<0.05)。采用虹吸法收集粪便测得的ADCd与在排粪高峰时段内积粪器法的测定值相当;后肠取粪法收集粪便测得的ADCd低于积粪器法排粪高峰期的测定值。采用虹吸法收集粪便测得的ADCp略低于在排粪高峰时段内积粪器法的测定值;后肠取粪法收集粪便测得的ADCp低于积粪器法各时间段内的测定值。因此,在选择挑选新鲜完整粪便的前提下,在排粪高峰期采用虹吸法收集粪便用于ADC的测定是一种方便可靠的方法,反而后肠取粪法由于操作误差容易带来更大偏差。  相似文献

New developments in different branches of biotechnology are discussed. The production of peptide hormones, new interferons and other lymphokines by the microbial and cell cultures, and new enzyme inhibitors of microbial origin are the most important for health care and pharmacy. The main direction in research in the agriculture represents the development of the new, very effective methods of nitrogen fixation and the production of animal growth hormones by genemanipulated microorganisms. One of the most important field of application of biotechnology is the chemical industry, c.f. microbial production of polymers and biotransformation of compounds previously produced by chemical methods (acrylamide, adipic acid, naphtalen conversion, etc.). Several novel methods of degradation of the cellulosic materials are mentioned and exploitation of biotechnology in environmental protection is also discussed.  相似文献
