Pregnancies in hamsters may be terminated with 10 μg PGF administered b.i.d. on days 4, 5 and 6 of gestation. Small (250 μg and above) daily injections of progesterone on the same days will reverse this PG effect; in contradistinction, 10 mg of progesterone per day failed to maintain normal pregnancies in hamsters spayed on day 5. Daily administration of 3 mg of progesterone and 1 μg of estrone essentially normalized the gestation; administration of PGF at 10 mg on days 5, 6 and 7 of pregnancy in steroid-maintained rats, resulted in pregnancy termination in all animals, while 1 mg was partly effective. These data demonstrate an extra-ovarian site of action of prostaglandin F on pregnancy in hamsters.  相似文献

The naturally-occurring metabolite of prostaglandin F, 15-keto prostaglandin F (15-keto PGF), elicited rapid and sustained declines in serum progesterone concentrations when administered to rhesus monkeys beginning on day 22 of normal menstrual cycles. Evidence for luteolysis of a more convincing nature was obtained in studies where a single dose of 15-keto PGF was given on day 20 of ovulatory menstrual cycles in which intramuscular injections of hCG were also given on days 18–20; serum progesterone concentrations fell precipitously in monkeys within 24 hours following intramuscular administration of 15-keto PGF. However, corpus luteum function was impaired in only 4 of 11 early pregnant monkeys when 15-keto PGF was administered on days 30 and 31 from the last menses, a time when the ovary is essential for the maintenance of pregnancy. Gestation failed in 2 additional monkeys 32 and 60 days after treatment with 15-keto PGF, but progressed in an apparently normal manner in the remaining 5 animals. Two pregnant monkeys treated with 15-keto PGF on day 42 from the last menstrual period, a time when the ovary is no longer required for gestation, continued their pregnancies uneventfully. Corpus luteum function was not impaired in 9 control monkeys which received injections of vehicle or hCG at appropriate times during the menstrual cycle or pregnancy.  相似文献

Prostaglandin F and its methyl analogues were used for induction of abortion in 598 patients with gestational age from 9 to 20 weeks. Different routes of administration were studied and varous dosages given. The incidence of gastrointestinal side effects were within acceptable range for all methods. Both intra-amniotic injections of 50 mg PGF and 2.5 mg 15-methyl PGF as well as intramuscular and vaginal administration of 15-methyl PGF or its methyl ester, respectively, were highly effective in termination of pregnancy. The intramuscular route was, however, associated with the highest frequency of gastrointestinal side effects. If both efficacy and side effects were taken into consideration, the intra-amniotic and vaginal routes were superior. The ease of administration as well as the applicability over a wider range of gestation in termination of pregnancy may, however, in many situations speak in favour of the repeated vaginal administration of 15-methyl PGF methyl ester.  相似文献

Seven rabbits were ejaculated four times once weekly, and saline or 2.5 mg PGF tromethamine salt was injected sc at 4 and 2 hours or at 8 and 4 hours before ejaculation. First ejacula taken at 2 hours after the second injection of PGF contained 150% more (P.07) sperm than those after injections of saline. The comparable difference (60%) at 4 hours after PGF was not significant. PGF did not influence sperm output in second, third or fourth ejacula. After 28 daily sc injections of 5 mg PGF in another experiment, the fertility of four treated rabbits was as high as that for four controls. Without sexual preparation in seven bulls, im injections of 40 or 80 mg PGF 30 minutes before ejaculation resulted in 33% greater (P<.05) sperm output than that after injection of 0, 7 or 20 mg PGF, but the highest sperm output after PGF was 30% less (P<.05) than that after sexual preparation in the same bulls. We conclude that injections of PGF result in increased sperm output in ejacula taken without sexual preparation within 2 hours in rabbits and in bulls.  相似文献

Embryonic implantation is a complex process in which both maternal andembryonic signals are involved. In the present study, we evaluated changes in uterine prostaglandins production and nitric oxide synthase (NOS) activity during the course of early pregnancy and their interaction during implantation in rats. Uterine phospholipase A2 (PLA2) activity is increased on days 5 (day of ovoimplantation) and 6, compared to preimplantation days (3 and 4). This enhanced activity might be responsible for the observed increase in uterine PGE and PGF production observed on day 5 of pregnancy, which induces endometrial vascular permeability and decidualization. When embryo access to the uterus is impaired, the increase of PG production is suppressed. During postimplantation, PGE levels return to preimplantation values, while PGF decreased with respect to preimplantation values. Uterine NOS activity is also increased on day 4 and reaches a maximum on day 5, with a profile similar to PGE and PGF Dexamethasone administered in vivo decreased uterine NOS activity on day 4 of pregnancy but not on day 5, suggesting the presence of at least two types of NOS enzymes in the early days of pregnancy. A competitive inhibitor of NOS, L-NAME (600 and 1000 μM) induced a decrease in PGE and PGF production in uterine tissue on day 5 of pregnancy. These results suggest the existence of a physiologically relevant nitridergic system which modulates prostaglandin production in the rat uterus during embryonic implantation.  相似文献

20α-OH-SDH is a marker of luteolysis in rat corpora lutea and appearance of this enzyme is inhibited by prolactin but stimulated by LH or hCG. PGF2α induced 20 α-OH-SDH activity in corpora lutea of pregnant rats and a significant fall in peripheral plasma progesterone concentrations when administered i.m. for two consecutive days. Rats treated with PGF2 α on days 8 and 9 of pregnancy were resorbing implants by day 10. Exogenous progesterone, but not estrogen, prevented implant resorption, yet 20 α-OH-SDH appeared in the corpora marking luteolysis. HCG, LH and prolactin, but not FSH, prevented pregnancy termination and inhibited induction of 20 α-OH-SDH in rats treated with PGF2 α in early pregnancy. PGF2α also induced 20α-OH-SDH in luteal tissue of intact and hypophysectomized rats treated on days 14 and 15 of pregnancy, but neither exogenous steroids or gonadotrophins blocked the induction of the enzyme in rats treated at this time. The increase in lutein 20α-OH-SDH activity during the peripartal period was partially blocked by administration of the prostaglandin biosynthesis inhibitor, indomethacin, suggesting a role for endogenous prostaglandins in the induction of 20α-OH-SDH at term. It appears that PGF2α acts directly on the ovary to induce 20α-OH-SDH activity by preventing the luteotrophic action of prolactin. Other luteal NADPH-dependent dehydrogenase activities are not markedly stimulated following PGF2α administration.  相似文献

The effects of prostaglandin (PG)F and PGF, 1–15 lactone were compared in luteal phase, non-pregnant and in early pregnant rhesus monkeys. Animals treated with either PG after pretreatment with human chorionic gonadotropin (hCG) had peripheral plasma progesterone concentrations that were not statistically different from those in animals treated with hCG and vehicle. However, menstrual cycle lengths in monkeys treated with PGF, 1–15 lactone were significantly (P <0.02) shorter than those in vehicle treated animals. In the absence of hCG pretreatment, plasma progesterone concentrations were significantly (P <0.008) lower by the second day after the initial treatment with either PGF or PGF, 1–15 lactone than in vehicle treated monkeys. Menstrual cycle lengths in monkeys treated with either PG were significantly (P <0.04) shorter than those in animals treated with vehicle. There were no changes in plasma progesterone concentrations in early pregnant monkeys treated with PGF, and pregnancy was not interrupted. In contrast, plasma progesterone declined and pregnancy was terminated in 5 of 6 early pregnant monkeys treated with PGF, 1–15 lactone. These data indicate that PGF, 1–15 lactone decreases menstrual cycle lengths in non-pregnant rhesus monkeys. More importantly, PGF, 1–15 lactone terminates early pregnancy in the monkey at a dose which is less than an ineffective dose of PGF.  相似文献

The effect of bovine conceptus secretory proteins (CSP) on uterine prostaglandin (PG)-F production was evaluated in dairy cattle following injection of estradiol-17β. Intrauterine injections of dialyzed serum proteins (Control, n=5) or CSP (n=5) were administered from days 15 through 18 post-estrus. Following intrauterine treatments on day 18, all cows were injected with E2 (3 mg) to stimulate uterine PGF production. Plasma concentrations of progesterone (P4) and 15-keto-13,14-dihydro-PGF (PGFM) were determined by RIA. The PGFM responses following E2 challenge were decreased (p<0.01) for cows receiving CSP versus serum proteins into the uterine lumen. Individual PGFM, P4 and cycle length responses are discussed. Data suggest that proteins secreted by the bovine conceptus suppress uterine PGF production during pregnancy recognition in the cow.  相似文献

During diestrus in three consecutive estrous cycles, each of six heifers was given (im) 30 mg, 15 mg (twice at 6-hr intervals) and 60 mg prostaglandin F (PGF) tham salt. Neither the decline in blood progesterone, the increase in blood estradiol, the duration or the peak of the LH surge, the interval to onset of estrus, nor the interval to ovulation was affected significantly by dose of PGF. Thus, relative to that after 30 mg PGF im, two injections of 15 mg at 6-hr intervals or 60 mg PGF did not hasten luteolysis. Thirty mg was an ample im dose of PGF to cause luteolysis. Regardless of im dose of PGF, blood PGF peaked at about 6.0 ng/ml within 10 minutes and returned to basal values (<1.0 ng/ml) within 90 minutes. In another trial, after a single iv injection of 5 mg PGF, blood PGF peaked (25 ng/ml) within 5 minutes and returned to basal values within 15 minutes. During a 30-minute infusion (0.5 mg/minute) of PGF, blood PGF plateaued at 29.5 ng/ml with a metabolic clearance rate of 17.0 liters per minute.  相似文献

The susceptibility of induced corpora lutea (CL) of prepuberal gilts and spontaneously formed CL of mature gilts to prostaglandin F (PGF) luteolysis was studied. Prepuberal gilts (120 to 130 days of age) were induced to ovulate with Pregnant Mare Serum Gonadotropin and Human Chorionic Gonadotropin (HCG). The day following HCG was designated as Day 0. Mature gilts which had displayed two or more estrous cycles of 18 to 22 days were used (onset of estrus = Day 0). Gilts were laparotomized on Day 6 to 9, their CL marked with sterile charcoal and totally hysterectomized. On Day 20, gilts were injected IM with either distilled water (DW), 2.5 mg PGF or 5.0 mg PGF. An additional group of prepuberal gilts was injected with 1.25 mg PGF, a dose of PGF equivalent, on a per kilogram body weight basis, to the 2.5 mg PGF dose given to the mature gilts. The percentages of luteal regression on Day 27 to 30 for mature and prepuberal gilts given DW, 2.5 mg PGF and 5.0 mg PGF were 0.0 vs 4.4, 43.5 vs 96.8 and 47.7 vs 91.6, respectively; the percentage of luteal regression for the prepuberal gilts given 1.25 mg PGF was 75.1. These results indicate that induced CL of the prepuberal gilt were more susceptible to PGF luteolysis than spontaneously formed CL of the mature gilt and that pregnancy failure in the prepuberal gilt could be due to increased susceptibility of induced CL to the natural luteolysin.  相似文献

The concentration of prostaglandin F (PGF) has been measured in the peripheral plasma of normal rhesus monkeys ( ) during the final third of gestation, and in monkeys treated with dexamethasone or PGF2α after day 145 of pregnancy. Daily administration of PGF2α (10–15 mg/day im) reliably induced abortion within 3–6 days. However, dexamethasone (8 mg/day im from day 145) had no effect on the length of gestation.The concentration of PGF in the femoral venous plasma of untreated or dexamethasone-treated monkeys was highly variable, both in serial samples taken from the same animal and in samples taken from different animals at the same time of gestation. There was no indication of an effect of dexamethasone treatment on the plasma PGF levels, nor did the concentration of PGF increase during late pregnancy before spontaneous parturition. These results show that the myometrium of the pregnant rhesus monkey is highly sensitive to exogenous PGF2α during late gestation. However, a significant increase in the peripheral plasma concentration of PGF prior to the onset of labor was not observed.  相似文献

Two subcutaneous injections of Prostaglandin F THAM salt 24 hours apart terminated pregnancies in cats after the 40th day of gestation. Injections of 0.50 or 1.00 mg. PGF THAM salt/Kg. body weight were the most effective in terminating pregnancies. Parturition or abortion occurred within 24 hours after the initial injection in 9 cats and after the 2nd injection in 4 cats.  相似文献

Pseudopregnancy, induced in rats by sterile mating, lasted 13 days (median). The subcutaneous administration of PGF produced a significant shortening of this period (< 10 days), and PG administration on day 6 appears most suitable for assay purposes. A dose-response curve for PGF administered twice on day 6 is presented; the ED50 for termination of pseudopregnancy was estimated at 577 mcg/injection with 95% confidence limits at 463 and 723 mcg. PGE2 is also effective; 40% of rats returned to estrus at a dose of 1000 mcg twice on day 6, suggesting a potency of about PGF.  相似文献

The relative safety and effectiveness of vacuum aspiration and the intrauterine administration of 5 mg PGF for terminating pregnancies within two weeks of a missed menstrual period were evaluated in a study where subjects were randomly assigned to procedures; 100 patients were aborted with vacumm aspiration and 100 patients were aborted with PGF. All PGF-treated patients were premedicated with meperidine, diazepam and atropine. Complications were infrequent with either of the procedures. Vomiting occurred more frequently during the PGF procedure (30.0%) than during the vacuum aspiration procedure (9.0%). The intrauterine instillation of PGF successfully terminated all pregnancies. One patient continued to be pregnant after the vacuum aspiration procedure. Based on the results of this study both study procedures appeared to be safe and effective for terminating early first trimester pregnancies.  相似文献

Four antiestrogens (anordiol, tamoxifen, RU 39411, ICI 182780) and the antiprogestin, mifepristone (RU 486), were administered to the following three animal models: (1) ovariectomized rats, (2) mated rats treated post-coitally; and (3) pregnant rats treated post-implantation. The antiestrogens were administered alone or in combination with mifepristone at doses effective in preventing and/or terminating pregnancy in rats. The objective of the study was to determine whether these drugs influenced uterine concentrations of prostaglandins (PGF and PGE2).Antiestrogens administered alone to ovariectomized rats did not effect uterine PGE2 or PGF concentrations; whereas the combination of anordiol/mifepristone increased uterine PGF concentration, resulting in an increase in the PGF/PGE2 ratio.Mated rats were treated post-coitally for three consecutive days with anordiol, tamoxifen, estradiol and mifepristone alone and with the combination of anordiol/mifepristone and tamoxifen/mifepristone. An increase in uterine PGF concentrations and in the PGF/PGE2 ratio occurred only in anordiol/mifepristone treated group. A decrease in uterine PGE2 concentrations occurred in animals treated with anordiol, tamoxifen and estradiol, resulting in an increase in the PGF/PGE2 ratio.Anordiol (5.0 mg/kg/day) and mifepristone (4.0 mg/kg/day) alone and the combination of anordiol/mifepristone (2.5/1.0 mg/kg/day) administered to pregnant rats on days 7, 8 and 9 of pregnancy induced an increase in PGF levels without affecting uterine PGE2 concentration. The changes in uterine PGF concentrations induced by anordiol and the combination of anordiol/mifepristone resulted in an increase in the PGF/PGE2 ratio.The antiestrogens tested except for ICI 182780 possessed agonist activity when assayed by measuring their capacity to increase the uterine weights in ovariectomized rats. Also, ICI 182789 was the only antiestrogen that did not influence uterine PG concentrations. It can be concluded that ICI 182780 is the only “pure” antiestrogen among those tested.The present results show that antiestrogens and the combination of mifepristone plus anordiol at doses preventing implantation and terminating pregnancy increase uterine PGF and/or decrease PGE2 concentrations, resulting in an alteration of PGF/PGE2 ratio. These findings suggest that there exists a critical balance of PGF to PGE2 concentrations in the uterus required for the normal passage of fertilized ova through the oviduct, initiating implantation of the blastocysts, development of embryos, and maintenance of pregnancy.  相似文献

Pregnant hamsters were administered (SC) prostaglandin or vehicle on the morning of the 4th day of pregnancy. Serum progesterone was significantly depressed (p<.01) at 0.5, 2, and 6 hours after treatment with 100 μg PGF. Serum progesterone levels were unchanged 2 hours and 6 hours after treatment with 100 μg PGF and 2 hours after treatment with 1 mg PGF. Progesterone levels were depressed to less than 1 ng/ml 6 hours after treatment with 1 mg PGF. The specific uptake of 3H-PGF in whole hamster corpora lutea was significantly depressed 2 hours and 6 hours following 100 μg PGF treatment. A 15% depression in specific uptake occurred 0.5 hour post-treatment. Treatment with 100 μg PGF resulted in no change. Administration of 1 mg PGF resulted in depressed 3H-PGF uptake at both 2 and 6 hours post-treatment.Prostacyclin (PGI2) treatment resulted in no change in either 3H-PGF specific uptake or serum progesterone 2 hours after 100 μg treatment SC. These parameters were both reduced approximately 30% 6 hours post-treatment. Treatment with 6-keto-PGF resulted in a complete lack of measurable 3H-PGF uptake and serum progesterone levels less than 1 ng/ml at both 2 and 6 hours after treatment with 1 mg SC.  相似文献

Prostaglandin biosynthesis was studied in the rat uterus during the oestrous cycle. Uterine homogenates were incubated for 20 minutes in the presence of exogenous substrate (2.10−5M). PGF and PGE2 were measured by R.I.A.. A sharp peak PGF and a smaller peak of PGE2 were observed at prooestrus, 20 h. Another small PGE2 peak occurred at dioestrus II, 15 h. The lowest values of both PGs were found on dioestrus, 15 h. Plasma oestradiol concentration were highest at proestrus, 15 h and 20 h. A sharp progesterone peak occurred at prooestrus, 20 h. The PGF peak is next to the oestradiol peak and is superimposable or lags slightly beyond the progesterone peak.Incubation with 14C arachidonic acid and subsequent analysis of extracts by TLC and scanning showed that the major metabolite is PGI2, identified as 6 keto PGF. The conversion rate of arachidonic acid into 6 keto PGF is 5 times higher than into PGF. 6 keto PGF was further identified by GC/MS. No significant difference was observed between 6 keto PGF production during oestrus and dioestrus.  相似文献

The cerebral cortical action of prostaglandin F (PGF) has been determined by recording the effects of intracarotid injections of PGF on cerebral evoked potentials. PGF differentially reduced cortical evoked potentials. The cortical action of PGF appeared to be qualitatively identical with that of norepinephrine (NE) but weaker. A protection of the cortex from the inhibitory action of NE by a preceding dose of PGF was demonstrated. The actions of both PGF and NE appear to be on the same or related postsynaptic receptors. The actions described were at doses that did not reduce oxygen availability. PGF may act as a modulator of adrenergic transmission in the cortex. The intracellular recording in the companion paper supplies the further critical evidence that PGF has a synaptic inhibitory action.  相似文献

In a number of assay ssytems, some 17-phenyl-trinor prostaglandins were similar in activity and potency to the corresponding parent prostaglandin. In others, the 17-phenyl analogs appeared several times more potent. In the hamster antifertility assay, which is considered to measure luteolytic activity, 17-phenyl-18,19,20-trinor prostaglandin F was about 90-times PGF in potency.Rat blood pressure responses to 17-phenyl analogs were significant. The 17-phenyl-trinor PGF pressor potency was 5 times that of PGF. The 17-phenyl-trinor PGE2 blood pressure response was atypical since a pressor rebound phenomenon followed the expected depressor response. Lastly, 17-phenyl-trinor PGF was more potent than PGF in synchronizing the estrous cycle in beef cows.  相似文献

This study was conducted in vitro to examine factors that may regulate prostaglandin release by bovine trophoblast and endometrial slices. Trophoblastic tissues and endometrial slices were recovered from superovulating and normally-ovulating cattle on day 16 or 20 of pregnancy and incubated for 24 h. Release of PGF2α and 13,14-dihydro-15-keto-PGF2α (PGMF), and incorporation of [14C]-leucine into proteins were quantified and expressed per μg DNA, which gives a measure of cellular activity. Activity of trophoblastic tissue for synthesizing protein was decreased (P<.05) and for releasing PGMF was increased (P<.05) on day 20 compared to day 16 of pregnancy. Neither supercovulation nor day of pregnancy altered trophoblastic activity for releasing PGF2α. Supercovulation increased (P<.05) endometrial release of PGF2α. Endometrial release of PGF2α was less (P<.05) on day 20 than on day 16 of pregnancy. When arachidonic acid (0, 100, 200 or 400 μg) was added at the start of incubation, trophoblastic release of PGF2α changed (P<.05) quadratically with dose of arachidonic acid. When arachidonic acid was added 8 h after the start of incubation, triphoblastic release of PGF2α increased linearly (P<.01) with dose of arachidonic acid. Adding arachidonic acid to incubation medium did not affect trophoblastic or endometrial protein synthesis. Endometrial slices suppressed (P<.05) trophoblastic protein synthesis and release of PGF2α. Apparently, endometrium can modulate trophoblastic release of prostaglandins and synthesis of proteins in vitro, and trophoblastic tissue from supercovulated cattle 16 or 20 days pregnant can be used to study trophoblastic synthesis of prostaglandins and proteins.  相似文献
