Acid phosphatase, esterases, and glutamate, lactate and malate dehydrogenases of 34 strains of Enterobacter cloacae and 22 strains of Enterobacter sakazakii were analysed by horizontal polyacrylamide agarose gel electrophoresis and by isoelectrofocusing in thin-layer polyacrylamide gel. The two species could be separated on the basis of distinct electrophoretic patterns of all enzymes analysed. Glutamate dehydrogenase and acid phosphatase were detected exclusively in E. cloacae, whereas esterase bands were more intensively stained in E. sakazakii. For each species, two zymotypes could be distinguished, on the basis of electrophoretic mobilities of malate dehydrogenase and banding patterns of esterase for E. cloacae, and by both isoelectric point and electrophoretic mobilities of an esterase and of lactate and malate dehydrogenases for E. sakazakii. The high degree of enzyme polymorphism within the two species permitted precise identification of strains. The variations in electrophoretic patterns might therefore provide useful epidemiological markers.  相似文献

The electrophoretic mobilities of non-specific esterases in vertical polyacrylamide slab gels were determined for 184 strains of staphylococci, representing a total of 18 proposed species and subspecies. Markedly uniform esterase patterns were seen within species demonstrating a high degree of human host specificity, while those species demonstrating a wide host range were polytypic and often showed considerable polymorphism. The unique banding patterns found in several species indicate that this technique may serve as a valuable aid to existing taxonomic schemes. Starch gel electrophoresis of representative strains usually produced sharper esterase bands than were found with polyacrylamide electrophoresis. However, the additional molecular-sieving effect produced by the polyacrylamide gels differentiated esterases to a greater extent.  相似文献

Characterization of enterobacteria by esterase specific-activity profiles   总被引:6,自引:0,他引:6  
The spectrum of specific activities and the electrophoretic mobilities of esterases produced by 550 strains of Enterobacteriaceae belonging to 36 species and subclassified into six groups (group 1, Escherichia coli, Shigella and Escherichia hermanii; group 2, genus Salmonella and genus Citrobacter; group 3, genus Klebsiella and genus Enterobacter; group 4, genus Serratia and Serratia fonticola; group 5, genus Proteus, genus Providencia and genus Morganella; and group 6, genus Yersinia) were analysed by acrylamide/agarose gel electrophoresis using standardized methods for staining and mobility comparisons. Nineteen types of esterase were defined by their respective esterase specific-activity profile (ESAP). A multiple correspondence analysis (MCA) of the ESAP data enabled 82% of the strains in the 36 species to be correctly classified. In each group, the species were clearly delineated after MCA on both ESAP and electrophoretic mobility data. In addition, the smallest number of characters providing species identification of Yersinia strains by esterase polymorphism was identified by means of a binary segmentation tree technique.  相似文献

Esterase electrophoretic polymorphism in human and animal strains of Clostridium perfringens was studied by using polyacrylamide-agarose gel electrophoresis. Five types of esterases, designated E-I to E-V and defined by their hydrolytic specificities toward five synthetic substrates, were found in protein extracts of bacteria grown without glucose (glucose-containing media allowed only the expression of esterase E-I). Mobility variants of esterase E-I, which hydrolyzes alpha- and beta-naphthyl acetates and butyrates, were used as a basis for the distribution of strains into 11 zymogroups. When all five types of esterases and their electrophoretic variants were considered, 77 electrophoretic types (ETs) could be described for the 89 strains tested. Animal strains did not constitute a distinctive subpopulation, as revealed by their distribution in the zymogroups and by clustering analysis. Statistical analysis also emphasized the importance of esterase E-IV (which hydrolyzes only naphthyl acetates) and esterase E-V (which hydrolyzes only alpha-naphthyl acetate) in clustering by the relatedness of the ETs. ETs allowed the epidemiological characterization of stool isolates recovered from elderly inpatient residents and from adolescent chronic-care psychiatric patients. These results indicate that esterase electrophoretic typing may be a marker for epidemiological and ecological analyses.  相似文献

Two American and eight European collections of silver eels were examined electrophoretically for variations in their liver esterases. Considering the results together with those obtained previously, it was confirmed that there is large variation in phenotype frequencies within both the eastern and the western Atlantic populations. Such quantitative data are, therefore, unlikely to resolve the question whether the two populations are produced from one or from two gene pools.The concept of separate gene pools, i.e., the classical two-species theory, is supported by certain differences in the esterase patterns which are not quantitative but consist in the complete absence from the one but presence in the other population of particular electrophoretic bands, or particular molecular properties reflected in consistent dissimilarities of electrophoretic mobility.A system of esterases is described which is shared by silver eel liver and whole homogenate of elvers. This system could be used in comparing phenotype frequencies in elvers and adult eels, and thus detect changes in the freshwater stages caused by differential selection.  相似文献

The multiple zones of esterase activity revealed after acrylamide disc electrophoresis of hepatopancreatic extracts from the snails Cepaea nemoralis and Cepaea hortensis have been characterized according to their molecular weights by comparing their relative mobilities with those of standard proteins in acrylamide gels of varying concentrations. It was observed that zones known to be allelic from previous work had identical molecular weights, differing only in charge. Homologies between esterases in the two species were suggested in several cases when molecular weights were taken in conjunction with biochemical data. The products of what is thought to be an esterase supergene have, with one exception, been shown to have the same molecular weights, suggesting evolution by serial gene duplications. The usefulness of molecular weight knowledge when trying to detect allelic zones intraspecifically and homologous zones interspecifically is discussed.The work was supported by a Science Research Council Postgraduate award.  相似文献

Esterases of 53 strains of Yersinia enterocolitica sensu stricto, including five previously defined biotypes, and 30 strains of Yersinia pseudotuberculosis were analysed by horizontal polyacrylamide-agarose gel electrophoresis and by isoelectrofocusing in thin-layer polyacrylamide gel. Esterase bands were defined by their range of activity towards several synthetic substrates, their resistance to heat and to di-isopropyl fluorophosphate. The two species were characterized by distinct electrophoretic patterns of their esterases. The apparent molecular weights of the heat-resistant esterase of Y. enterocolitica and of the major heat-resistant esterase of Y. pseudotuberculosis, as determined by polyacrylamide gradient gel electrophoresis, were estimated to be 52 000 and 250 000, respectively. On the basis of electrophoretic mobilities and isoelectric points of esterases produced by strains of Y. enterocolitica, five principal zymotypes were observed: two for strains of biotype 1, two for strains of biotypes 2 and 3, respectively, and only one for strains of both biotypes 4 and 5. The zymotypes of strains of biotypes 2, 3, 4 and 5 appeared to be more closely related to one another than to zymotypes of strains of biotype 1. Variations in number or mobility of bands observed within each biotype of Y. enterocolitica and within some serotypes of Y. pseudotuberculosis could represent an additional marker for epidemiological analysis.  相似文献

Acrylamide gel electrophoresis of crude cellular extracts of Bacillus subtilis revealed the presence of two acetyl esterases. Esterase A, the slower migrating enzyme, was found to be present in both vegetative and sporulating cells, whereas esterase B activity was more abundant after exponential growth ceased. Both esterases were present in the supernatant fraction of lysed spheroplasts and in a disrupted spore preparation. Of four pleiotropic asporogenous mutants tested, three exhibited decreased esterase B activity. Esterases A and B were partially purified by differential precipitation and co-chromatographed on diethylaminoethyl (DEAE)-cellulose (pH 7.5) and DEAE-Sephadex (pH 8.5). By employing gel filtration chromatography, the two esterases were separated, and molecular weights of 160,000 and 51,000 were estimated for esterases A and B, respectively. Esterase A was further purified to electrophoretic homogeneity by differential heating and preparative starch block electrophoresis. Sodium dodecyl sulfate-acrylamide gel electrophoresis of purified esterase A yielded a single protein band with a molecular weight of 31,000. The pI values of esterases A and B were determined to be 6.4 and 5.4, respectively.  相似文献

The esterase isozymes were surveyed in axenic stocks of syngens 1, 2, 4, 5, 6, and 8 of Paramecium aurelia by starch gel electrophoresis. In paramecia there appear to be four types of esterases which are clearer in axenic than in bacterized stocks. Each type differs in its substrate specificity and/or its response to the inhibitor eserine sulfate. Minor variations in type D esterases sometimes occur in different extracts of the same stock and may result from changes in the temperature of growth of the cells or growth cycle differences. Differences in the mobility of the A, B, or C (cathodal) types of esterases may occur in different syngens. They also occur for the A and B types among stocks within a syngen, but the frequency is low, except in the case of syngen 2. Since each of the types of esterases varies independently, at least four and possibly more genes appear to specify the esterases in the species complex. Some pairs of syngens vary in their electrophoretic positions for all types of esterases. Other pairs have identical zymograms. This observation suggests that some syngens may differ from each other by as many as four esterase genes, while others may not differ at all. The difference between P. aurelia and Tetrahymena pyriformis in the degree of intrasyngenic variation observed for enzymes is discussed in relation to other types of characters, the organization of the genetic material in the macronucleus, the presence of symbionts, and their breeding systems. 在库蚊Culex pipiens品系中,非专一性酯酶的过量产生是对有机磷杀虫药剂抗性的普遍机理。酯酶基因位于紧密连锁的A和B座位上。现已知所有酯酶B的过量产生都是基因扩增的结果。为了确定不同国家库蚊品系的酯酶B1的过量产生是否都是相同DNA单基因型扩增的结果,我们构建了酯酶B1结构基因扩增区的限制性内切酶酶切图谱,分析了限制性酶切片段长度多态性(RFLP)。研究发现不同地理位置的酯酶B1库蚊,如法属圭亚那,委内瑞拉、波多黎各岛、美国加利福尼亚和中国北京,都有着相同的单基因扩增,但在扩增水平上有较大的差异。我们认为无论在美洲或亚洲,凡是酯酶B1扩增的库蚊都为同一个起源,之后经迁移而传播到各地;同时发现酯酶B1扩增的库蚊与酯酶A2一B2扩增的库蚊相比,其迁移有一定的局限性;并且酯酶B1扩增的库蚊仅仅限于美国、加勒比和中国的一些地区,而酯酶A2-B2扩增的库蚊则广泛地分布于美国、加勒比、亚洲、非洲、太平洋各岛及欧洲等地。  相似文献

Acetylcholine esterase (AchE) and non-specific esterases were studied during the development of sea urchins Strongylocentrotus droebachiensis and S. intermedius and their hybrids by means of electrophoresis and measurements of enzyme thermostability. Two AchE fractions were found which differed by thermostability. At the late gastrula stage, the therolabile form predominated and at the mid-pluteus stage the thermostable one. Non-specific esterases in both the species of sea urchins are represented by complex isozyme systems. Their changes during development are accompanied by the changes in thermostability and electrophoretic patterns. The thermostability of esterases at the pluteus stage in the hybrids is higher than in the maternal species, apparently, due to the appearance of the thermostable enzyme which appears in the paternal species provisionally at the prism stage.  相似文献

One hundred eighty-eight stocks of Paramecium primaurelia, P. biaurelia, P. tetraurelia, and P. octaurelia were grown axenically and tested for five esterases, visualized by starch gel electrophoresis, in a search for variant stocks. The five esterases can be distinguished on the bases of their substrate specificity, sensitivity to an inhibitor, and response to different growth conditions. This paper addresses the nature of the electrophoretic change in mobility of the variant stocks in order that species relationships can be more accurately assessed. Crosses carried out in all four species show that single genes determine the differences in mobility between variant and common subtypes. Extracts of variant stocks that gave similar patterns were run against each other, tested for their sensitivity to the inhibitor, and the pattern was compared to that found in extracts of stocks with variant and common subtypes in other species. The majority of the variants in P. primaurelia, P. tetraurelia, and P. octaurelia show an electrophoretic mobility characteristic of a common subtype, or a variant, in another species. The same proportion of variant subtypes as common subtypes have mobilities similar to esterase subtypes found in other species. Of the four species examined in this paper, P. tetraurelia and P. octaurelia appear to be most closely related on the basis of shared esterase subtypes. In P. biaurelia the mobilities of most of the variants are unique, as are the common esterase subtypes in this species. P. biaurelia stands out as having the greatest number of esterase subtypes, with very few of them homologous to subtypes found in other species. This observation supports the idea of greater diversification of stocks within P. biaurelia than for the other three species.  相似文献

To determine whether enzyme electrophoretic polymorphism in Escherichia coli populations was influenced by environmental background, the mobilities of four electrophoretically variable esterases (A, B, C and I) were examined. The distinction between isolates was established by significant differences in the electrophoretic distribution and the genetic diversity coefficient of individual esterases. Principal components analysis on each population and on all strains revealed three groups of allozymes. The first, characterized by slow electrophoretic mobilities of esterase B, was frequently observed in strains obtained from human extra-intestinal infections and rarely in commensal organisms. The second, characterized by fast mobilities of esterases A and B, was frequently found in animal isolates. The third, characterized by prominence of the most common mobilities of esterases B and A, was recovered in all populations. These results were confirmed by discriminant analysis. Among the 610 strains investigated, 316 electrophoretic types (distinctive combinations of allozymes of the four varieties of esterases) were distinguished, illustrating high esterase polymorphism.  相似文献

Esterases, and glutamate, lactate and malate dehydrogenases of 64 Aeromonas hydrophila, A. caviae and A. sobria strains, were analysed by polyacrylamide agarose gel electrophoresis and by thin layer isoelectrofocusing. On the basis of the isoelectric points of malate dehydrogenase from the three species and the mobility of lactate dehydrogenase from A. sobria, 8 species specific zymotypes were defined: three for A. hydrophila strains, three for A. caviae strains and two for A. sobria strains. These zymotypes correlated with previously established DNA hybridization groups. The other electrophoretic data were found to be less useful for distinction between A. hydrophila and A. sobria strains, but supported differentiation into zymotypes for A. caviae strains. The two-dimensional electrophoretic profile established by plotting isoelectric point against electrophoretic mobility of the major esterase illustrated the degree of enzyme polymorphism among the strains of the three species. Variation in electrophoretic patterns within A. hydrophila and A. caviae might provide useful epidemiological markers.  相似文献

Two closely related species, Drosophila aldrichi and D. mulleri, are compared on the basis of their esterase isozyme patterns after starch gel electrophoresis. Comparable esterases between the two species are identified by substrate specificity, inhibition, and enhancement of esterase activity by various agents. The extensive electrophoretic variability of most of the esterases in nature is described and data relating to the inheritance of the various enzyme forms are presented.This work was supported (in part) by a U.S. Public Health Service research grant (GM 11609 to W. Fifty-three strains of Acinetobacter, belonging to the species A baumannii, A. haemolyticus and A. johnsonii, were differentiated by electrophoretic typing of their esterases, on the basis of both the enzyme specific activity profiles and their electrophoretic mobilities. Each esterase was defined by its spectrum of hydrolytic activity toward five synthetic substrates and its sensitivity to di-isopropyl fluorophosphate. Since each enzyme was not detected in all strains of a given species, several zymotypes could be defined by the patterns of combinations of esterases. Thus, 24 zymotypes were defined in the 32 A. baumannii strains, 4 were defined in the 10 A. haemolyticus strains and 6 were defined in the 11 A. johnsonii strains. When the electrophoretic mobilities of the various esterases were included, each of the 53 strains of Acinetobacter (with the exception of three A. haemolyticus strains) showed a distinct electrotype.  相似文献

The six species were clearly separated from each other by their distinct enzyme electrophoretic polymorphism. For Y. enterocolitica, the strains of biotype 5 were differentiated from the other biotypes by the mobility of glutamate dehydrogenase. For Y. frederiksenii, six zymotypes were delineated by pI and by the mobility of the enzymes. Variation in number or mobility of esterases within each species could represent a marker for epidemiological and ecological analyses. A linear relationship was obtained between the mean genetic diversity coefficient of enzymes and the mean percentage DNA-DNA relatedness of Y. intermedia, Y. aldovae, Y. enterocolitica and Y. frederiksenii.  相似文献

We have purified a bacterial enzyme, designated esterase M, by tailoring an efficient and rapid strategy with information derived from titration curves of proteins in crude extract. The pH-dependent stability of the enzyme activity observed by titration pattern allowed an acidic pH treatment of extract and a cationic exchange chromatography at pH 4.1. These two steps were followed by an anionic exchange chromatography and a preparative electrophoresis. Thus, the enzyme was purified about 2000-fold within two days with a recovery of 13.3%. The electrophoretic variants of esterase M were investigated for their molecular relationship through the specific effect of antibodies on esterase electrophoretic pattern (immunosubtractive electrophoresis) which is applicable to large series of samples. By this process, we have demonstrated the presence of common antigenic determinants among the electromorphs of esterase M produced by the three species of motile Aeromonas.  相似文献

Esterases are involved in the susceptibility or resistance of organisms to organophosphate pesticides. We have examined the action of parathion on the marine dinoflagellates Crypthecodinium cohnii and Prorocentrum micans by looking at their esterases. One-dimensional gel electrophoresis, immunoblotting and cytochemistry plus image analysis were used to characterize the nature and distribution of the enzymes. Esterases were found in both species, but there appeared to be no particular intracellular localization. The esterase activity of the heterotrophic species Crypthecodinium cohnii was 30-fold greater than that of the autotrophic Prorocentrum micans and had an antigenic site in common with mosquito esterase. The resistance of Crypthecodinium cohnii to parathion was specific and reversible. Less parathion entered the parathion-resistant Crypthecodinium cohnii cells than the untreated control cells. Parathion-resistant cell extracts of Crypthecodinium cohnii analyzed after immunoblotting also contained an additional band of esterase activity. These results confirm the importance of esterases in toxicological studies of organophosphate insecticides, especially those of marine dinoflagellates.  相似文献
