- 1.1. Microtubule reassembly was studied in supernatant fluids from rat brain. Tubulin in extracts from zinc-deficient animals showed an impaired ability to repolymerize compared to extracts from controls; 10 μM zinc stimulated reassembly of tubulin in extracts from zinc-deficient animals.
- 2.2. Low zinc concentrations (250–900 μM zinc in the presence of 1 mM EGTA) stimulated reassembly oftubulin in brain extracts from control rats; similar concentrations of nickel had no effect whilst cobalt was inhibitory. In the absence of EGTA 20–40 μM zinc stimulated reassembly in brain extracts from normal rats.
- 3.3. Zinc-induced changes in reassembly were associated with changes in the free sulphydryl group content of the assembled crude microtubule protein; increased assembly was associated with a higher free sulphydryl group content, decreased assembly with a lower content.
- 4.4. 65Zn was found to bind to tubulin. This binding was partly inhibited by N-ethylmaleimide.

- 1.1. Propanol extracts of the sponge Tethya aurantia (Demospongiae) were fractionated, guided by bioassay, for a component with negative chronotropic and inotropic activity on isolated guinea pig atria.

- 3.3. This intermediate in the sequence of degradation of purines was unexpected, since it has been reported only once before to occur in marine invertebrate animals.

- 1.1. The neuroendocrine caudodorsal cells (CDCs) of Lymnaea stagnalis are a network of about 100 electrotonically coupled neurones. The CDCs release multiple peptides, including an ovulation hormone, during a period of electrical activity, the CDC-discharge.
- 2.2. In isolated brains, a similar period of electrical activity (the afterdischarge) can be induced in all CDCs by a period of intracellular repetitive suprathreshold stimulation of one CDC.
- 3.3. In order to study the regulation of this electrical behaviour in the absence of electrical interactions and in a controlled environment, experiments were performed on CDCs in dissociated cell culture.
- 4.4. Methods for isolation and cell culture are described. Cell cultures had long-term viability and outgrowth of neurities occurred under serum-free conditions.
- 5.5. CDCs in cell culture maintained their capability of producing afterdischarges upon electrical stimulation. Cells in culture appeared more excitable than cells in the intact isolated brain.
- 6.6. The characteristic responses of CDCs in intact isolated brains to acetylcholine and FMRFamide were preserved in cultured CDCs. Both agents induced a transient hyperpolarization of the membrane, inexcitability and inhibition of an ongoing discharge.
- 7.7. In experiments where isolated CDCs were closely apposed, but physically separate, it was found that an afterdischarge in one CDC could induce a discharge in the other CDC.
- 8.8. These results confirm previous results which showed that an excitatory factor is released from the brain during the afterdischarge (Ter Maat et al., 1988, Brain Res., 43, 77–82), and demonstrate that this excitatory factor is released from the CDCs themselves.

- 1.1. Babesia hylomysci has an aminopeptidase and an acid endoprotease
- 2.2. The amino-peptidase has properties very similar to the aminopeptidase in Plasmodium yoelii nigeriensis and P. chabaudi.
- 3.3. The acid endoprotease is specific towards haemoglobin and practically has no action on bovine serum albumin.
- 4.4. In mouse normal red blood cells we find an acid protease having physico-chemical properties similar to the enzyme present in B. hylomysci extracts.
- 5.5. The similarity of electrophoretic velocity between acid protease in B. hylomysci and non-infected red blood cells leads us to think that the acid protease of parasitic extracts comes from the host-cell.
- 6.6. The proteolytic system of Babesia and Plasmodium are similar.

- 1.1. An apparent effect of insulin administration on enlargement of interscapular brown adipose tissue (BAT) was found in heat-exposed rats, but not in warm-adapted or cold-acclimated rats.
- 2.2. BAT extracts from the heat-acclimated/insulin-treated (HI) rats notably increased the capillary growth in an in vitro angiogenesis model in which microvascular fragments and myofibroblastic (Mf) cells isolated from lipid tissues were grown in co-culture, although a direct effect of insulin was not high.
- 3.3. BAT extracts from the HI rats stimulated the production of endothelial cell growth factor and collagen by Mf cells.
- 4.4. It is probable that an increased angiogenic activity contributes to the capillary growth and tissue growth in BAT of HI rats.

- 1.The levels of water, Na, K, Ca and Mg in blood serum, brain and kidney and aldosterone level in blood of Naja haje haje were studied during the different phases of the annual cycle.
- 2.The water content in the tissues studied displayed only minor changes as the animals passed from one phase to the other.
- 3.A significant increase in Na was recorded in the brain during the different phases indicating a depressed sodium pump, whereas the blood Na level showed a significant decrease during hibernation.
- 4.K increased in blood serum, brain and kidney during hibernation, while a nonsignificant decrease was found in blood serum during arousal. The brain may act as a potassium reservoir.
- 5.An increase in Ca and Mg concentration was recorded in blood serum, brain and kidney during prehibernation and hibernation. The data suggested a homeostatic function in the transport and metabolism of these cations.
- 6.Aldosterone exhibited a highly significant decrease especially during hibernation. The aldosterone regulation of ionic composition is discussed.
- 7.Na/K and Ca/Mg ratios in the brain may explain the decreased excitability during winter torpor.

- 1.1. First-generation laboratory animals of three populations of the isopod Porcellio scaber, collected from a reference wood, a zinc smelter area and a lead mine site, were compared with respect to effects of Cd.
- 2.2. All populations reacted differently to Cd-contaminated food: increased Cd concentrations in the food resulted in decreased consumption and growth for the reference isopods; mine isopods were not affected by Cd and for the smelter animals Cd stimulated growth at an intermediate concentration.
- 3.3. Since Cd concentrations in the isopod did not differ between populations, adaptation is probably based on an increased detoxification capacity.
- 4.4. The assimilation of Cd did not affect the Cu or Zn content of the isopods although the adapted isopods regulated their Cu content on a lower level than reference isopods.

- 1.1. In brush border membrane vesicles isolated from eel kidneys, adapted either to sea water or freshwater environments, a Na+/H+ antiporter is present.
- 2.2. Using a calibration plot it is possible to evaluate the amount of protons that this antiporter can accumulate inside the vesicular space.
- 3.3. The activity of the antiporter seems to be affected by the salinity of the water; it is higher in animals adapted to seawater.
- 4.4. This adaptation seems to occur by a Jmax regulation of the antiporter.

- 1.1. A potentiometric method for the assay of cholinesterase has been proposed and compared with a colorimetric assay.
- 2.2. Main kinetic parameters of cholinesterase from Hypostomus punctatus brain were determined indicating that true acetylcholinesterase is by far the predominant enzyme in the brain of this fish.
- 3.3. We have compared our data with published results described from other fish species.
- 4.4. The enzyme inhibition achieved after 3 hr incubation of brain homogenates with ethyl-parathion have indicated that this enzyme shows a characteristic organophosphorous sensitive behavior.

- 1.1. The effect of cold (8 ± 2°C) acclimation on the lactate dehydrogenase activities and isoenzyme patterns from sartorius muscle, liver, heart and brain of adult Discoglossus pictus pictus (Otth.) was studied.
- 2.2. Two groups of animals were studied: one set of animals was trapped in October and another set in December. In both cases some of the animals were sacrificed upon collection and some others subjected to 5 months of acclimation at 8 ± 2°C before being sacrificed for analysis.
- 3.3. A general trend towards a decrease in LDH specific activity was observed during cold acclimation. The magnitude of change, but not the direction, depends on both the tissue examined and the season at which the experiment was initiated.
- 4.4. A complex LDH isoenzyme reorganization was also found in liver, heart and brain. In liver from Experiment 1 and in heart from both experiments, a relative maintenance in M-type LDH activity during cold acclimation was observed. However, in brain there was a relative maintenance of LDH3 activity in both experiments.
- 5.5. The low behavioral activity (and its metabolic consequences) and the existence of an intrinsic annual rhythm in D. pictus metabolism are suggested as responsible for the observed enzymatic changes.

- 1.1. The yolk proteins of hermaphrodite Dolichorhabditis sp. (Nematode, Rhabditida) are composed of at least three polypeptides: VT1, VT2 and VT3 with molecular masses of 175.2, 107 and 82 kDa respectively.
- 2.2. All three yolk polypeptides make up at least one native protein complex which can be resolved by PAGE.
- 3.3. The yolk proteins are glycosylated and can be isolated by chromatography in Con A-Sepharose.
- 4.4. Partial chymotryptic hydrolysis shows that VT2 in different from its C. elegans homologue, YP115.
- 5.5. The main polypeptides synthesized by whole animals are the yolk components which are actively secreted in the incubation medium.

- 1.1. Creatine kinase (CPK) isozymes of extracts from the electric organ, dorsal muscle and brain of Electrophorus electricus (L.) were analysed with Cellogel electrophoresis. A single component corresponding to the MB-form was obtained for both electric organ and the dorsal muscle. The BB-form was present in the brain extract.
- 2.2. Upon acetone fractionation of the aqueous extract of electric organ, the final fraction was submitted to gel filtration and presented a single peak of CPK activity.
- 3.3. Characterization of this fraction by thin-layer gel filtration indicated an apparent molecular weight of 80,000 which corresponds to the enzyme dimeric structure.
- 4.4. The implications of this finding with the muscular origin of the electric organ are discussed.

- 1.1. Three monoclonal antibodies have been produced which neutralize in vitro the haemolytic activity present in tentacle extracts of the box jellyfish (Chironex fleckeri).
- 2.2. Two of these monoclonal antibodies bound specifically to a component of relative molecular mass 50,000 in tentacle extract on Western blots.
- 3.3. This binding only occurred when the extracts were electrophoresed under non-reducing conditions.
- 4.4. The third monoclonal antibody did not display binding to Western blots of tentacle extract under any of our experimental conditions.

- 5.5. Three different centrifugation regimes were tested: self-generated density gradients in a fixed angle rotor; self-generated density-gradients in a swinging-bucket rotor; pre-formed density-gradients in a swinging-bucket rotor.
- 6.6. Genomic DNA isolated by the CsTFA-procedure was found to have higher purity as compared to genomic DNA isolated in a conventional CsCl gradient.
- 7.7. Isolated DNA was shown to be of a quality suitable for applications in molecular biology.

- 1.1. Tubulin has been isolated from brain of carp (Cyprinus carpio), acclimated to summer temperatures (16–20°C), and its in vitro reassembly behavior has been characterized.
- 2.2. Among the striking properties of this tubulin preparation is the temperature profile showing a high level of polymerization at the environmental temperature of carp.
- 3.3. The critical tubulin concentration for assembly was 0.8 mg/ml, which was higher than mammalian tubulin purified by the cycle procedure.
- 4.4. The microtubular protein showed three high mol.wt component and a minor component of about 43,000 daltons was also found to copurify with tubulin.

- 1.1. The tissue specificity and ontogeny of alcohol dehydrogenase (ADH) are reported for the leopard danio, Brachydanio nigrofasciatus.
- 2.2. Of the seven adult tissues assayed (eye, brain, kidney, liver, ovary, skeletal muscle and stomach), only liver extracts showed ADH activity.
- 3.3. The activation of the Adh locus is correlated with the first functioning of liver. It is suggested that the state of liver cell differentiation may be the stimulus necessary for Adh locus expression.
