1. During the early differentiation of the cuticle the cell membranes smooth out and the cells become closely attached over most of their surface. The change seems to be due to a layer of cement which forms between them. The plasma membranes also increase in density. 2. The decreased membrane activity of the cuticle cells may prevent a phagocytosis of the melanocyte processes and thus account for the non-pigmentation of the cuticle. 3. The flattening and imbrication of the cuticle may possibly be explained by a zipper-like spread of cell contacts. 4. Keratinisation of the cuticle occurs at a late stage in its development; the keratin formed is an amorphous type, similar to the gamma-fraction of the cortex which is produced at a similar level. 5. Keratinisation is accompanied by the formation of complex intercellular layers similar to structures observed in the inner root sheath (see Part 3). 6. In the final stage of keratinisation the remaining cytoplasm condenses with the result that the cell is divided into a laminated structure with an outer keratinised layer and an inner layer, which is insoluble in keratinolytic solvents.  相似文献

1. The presumptive cortical cells of hair in the undifferentiated matrix of the bulb contain mitochondria, agranular vesicles, and many small dense R.N.P. particles, but no keratin, pigment granules, or endoplasmic reticulum. 2. In the mid-bulb region intercellular adhesion is limited to small localised areas. Intercellular gaps are common and the cell surfaces are irregularly convoluted. The melanocyte processes penetrate the cell gaps. The relation between their pigment-bearing tips and the involutions of the cell membranes suggests an active phagocytosis of the tips. 3. Fibrous keratin first appears in loose parallel strands of fine filaments (ca. 60 A diameter) in the mid-bulb. The filaments, the long mitochondria, and elongated nucleus are all parallel to the long axis of the cell and the axis of the follicle. 4. At the level of the constriction of the bulb and above, a dense amorphous substance appears between the fine filaments and apparently acts as adhesive cement. The bundles of filaments now form well defined fibrils. The packing of the filaments within the fibrils is in places hexagonal and elsewhere in the form of "whorls." 5. At higher levels further filaments and interfilamentous cement are added together and the whole cytoplasmic space becomes packed with fibrils which finally condense to massive blocks of keratin. The residual cellular material occupies the interstices. 6. The addition of the interfilamentous substance is regarded as an essential factor in keratinisation. Keratin is considered to be a complex made of fine filaments (alpha-filaments) embedded in an amorphous substance (gamma-keratin) which has the higher cystine content. 7. The wide-angle fibre-type x-ray pattern is thought to be due to scattering by the fine alpha-filaments and some low angle lateral spacings to the filament-plus-cement structure.  相似文献

This paper reviews the current knowledge about human hair photodamage and the photodegradation mechanisms proposed in the literature. It is shown that there are still a number of questions without answer regarding this issue. For example, a better understanding of the hair structural changes caused by different radiation wavelengths is still lacking. We also find controversies about the effects of sun exposure on different hair types. Explanations to these questions are frequently sustained on the amount and type of melanin of each hair, but factors such as the absence of knowledge of melanin structure and of established methodologies to use in human hair studies make it difficult to reach a general agreement on these issues.  相似文献

An in vitro preparation of hair cells from the bullfrog sacculus produces a transepithelial microphonic potential in response to well-defined mechanical stimuli. If corrected for the electrical time constant of the epithelium, the response follows a fast stimulus with a 40-microsecond delay at 22 degree C. The short latency and its modest temperature dependence limit possible models for transduction by hair cells.  相似文献

The voltage-dependent mechanical activity of outer hair cells (OHC) from the organ of Corti is considered responsible for the peripheral auditory system's enhanced ability to detect and analyze sound. Nonlinear processes within the inner ear are presumed to be characteristic of this enhancement process. Harmonic distortion in the OHC mechanical response was analyzed under whole-cell voltage clamp. It is shown that the OHC produces DC, fundamental and second harmonic length changes in response to sinusoidal transmembrane voltage stimulation. Mechanical second harmonic distortion decreases with frequency, whereas the predicted transmembrane second harmonic voltage increases with frequency. Furthermore, the phase of the second harmonic distortion does not correspond to the phase of the predicted transmembrane voltage. In contradistinction, it has been previously shown (Santos-Sacchi, J. 1992. Neuroscience. 12:1906-1916) that fundamental voltage and evoked mechanical responses share magnitude and phase characteristics. OHC length changes are modeled as resulting from voltage-dependent cell surface area changes. The model suggests that the observed harmonic responses in the mechanical response are consistent with the nonlinearity of the voltage-to-length change (V-delta L) function. While these conclusions hold for the data obtained with the present voltage clamp protocol and help to understand the mechanism of OHC motility, modeling the electromechanical system of the OHC in the in vivo state indicates that the mechanical nonlinearity of the OHC contributes minimally to mechanical distortion. That is, in vivo, at moderate sound pressure levels and below, the dominant factor which contributes to nonlinearities of the OHC mechanical response resides within the nonlinear, voltage-generating, stereociliar transduction process.  相似文献

The human type I hair keratin subfamily comprises nine individual members, which can be subdivided into three groups. Group A (hHa1, hHa3-I, hHa3-II, hHa4) and B (hHa7, hHa8) each contains structurally related hair keratins, whereas group C members hHa2, hHa5, and hHa6 represent structurally rather unrelated hair keratins. Antibodies produced against these individual hair keratins, first analyzed for specificity by one- dimensional Western blots of total hair keratins, were used to establish the two-dimensional catalog of the human type I hair keratin subfamily. The catalog comprises two different series of type I hair keratins: a strongly expressed, Coomassie-stainable series containing hair keratins hHa1, hHa3-I/II, hHa4, and hHa5, and a weakly expressed, immunodetectable series harboring hHa2, hHa6 hHa7, and hHa8. In situ hybridization and immunohistochemical expression studies on scalp follicles show that two hair keratins, hHa2 and hHa5, define the early stage of hair differentiation, i.e. hHa5 expression in hair matrix and hHa5/hHa2 coexpression in the early hair cuticle cells. Whereas cuticular differentiation proceeds without the expression of further type I hair keratins, matrix cells embark on the cortical pathway by sequentially expressing hHa1, hHa3-I/II, and hHa4, which are supplemented by hHa6 at an advanced stage of cortical differentiation, and hHa8, which is expressed heterogeneously in cortex cells. Thus, six type I hair keratins are involved in the terminal differentiation of anagen hairs. The expression of hHa7 is conspicuously different from that of the other hair keratins in that it does not occur in the large anagen follicles of terminal scalp hairs but only in central cortex cells of the rare and small follicle type that gives rise to vellus hairs.  相似文献
