In this study, preimplantation mouse embryos were used as a new model for the study of actin distribution in nuclei and the identification of functional forms of nuclear actin. The combination of the direct detection of actin by fluorescent-conjugated phalloidin and DNase I with indirect immunofluorecence was applied as an integrated approach to studying the localization of actin in nuclei of two-cell mouse embryos. Aggregates of monomeric actin and two oligomeric forms of actin were revealed in nuclei. Each of these forms demonstrated their own pattern of distribution. Oligomeric actin recognized by antibodies to the actin C-terminal domain was associated with condensed chromatin, as well as with metaphase chromosomes and chromatin of the second polar body. Monomeric actin and another oligomeric form recognized by antibodies to actin N-terminal domain were revealed in the area of dispersed chromatin localization.  相似文献

Actin polymerization is a prominent feature of migrating cells, where it powers the protrusion of the leading edge. Many studies have characterized the well-ordered and dynamic arrangement of filamentous actin in this submembraneous space. However, less is known about the organization of unpolymerized actin. Previously, we reported on the use of covalently coupled profilin:actin to study actin dynamics and presented evidence that profilin-bound actin is a major source of actin for filament growth. To locate profilin:actin in the cell we have now used this non-dissociable complex for antibody generation, and obtained monospecific anti-actin and anti-profilin antibodies from two separate immunizations. Fluorescence microscopy revealed drastic differences in the staining pattern generated by the anti-actin antibody preparations. With one, distinct puncta appeared at the actin-rich leading edge and sometimes aligned with microtubules in the interior of the lamella, while the other displayed typical actin filament staining. Labelling experiments in vitro demonstrated failure of the first antibody to recognize filamentous actin and none of the two bound microtubules. The two anti-profilin antibodies purified in parallel generated a punctated pattern similar to that seen with the first anti-actin antibody. All antibody preparations labelled the nuclei.  相似文献

A simple and effective technique to produce monospecific polyclonal antibodies of high affinity against actin is described. In this procedure, rabbit skeletal muscle actin in the 1:1 complex with bovine pancreatic deoxyribonuclease I is used as antigen to immunize rabbits. The antisera obtained are shown to contain antibodies against both actin and deoxyribonuclease I. By affinity chromatography the two antibody preparations were separated and characterized. The affinity-purified anti-deoxyribonuclease I and anti-actin do not show cross-reactivity. Thus, anti-deoxyribonuclease I inhibits the enzymic activity of deoxyribonuclease I and stains the enzyme after Western blotting. Affinity-purified anti-actin does not inhibit deoxyribonuclease I activity and stains only actin after Western blotting. The affinity-purified anti-actin can be used in a number of different actin-detecting techniques such as in immunohistochemistry and in immunoblotting techniques. This antibody recognizes only actins from muscular tissues with high affinity. Immunoblots of polyacrylamide gels in the presence of ampholytes (IEF) indicate that this antibody only recognizes the alpha-variants of actin. Thus, the skeletal and cardiac alpha-actins are recognized but not the smooth muscle gamma-isoform and the cytoplasmic actins. Vascular smooth muscle alpha-actin is not recognized when using immunoblotting or enzyme-linked immunosorbent techniques. On frozen sections, however, the anti-actin antibody clearly stained vascular smooth muscle cells. Epitope analysis using actin fragments generated by limited proteolysis and selective cleavage using hydroxylamine indicate that this antibody is directed against a rather limited region within the N-terminus of actin.  相似文献

We purified actin antibodies by affinity chromatography from the serum of rabbits immunized with glutaraldehyde-fixed chicken gizzard actin filaments and used this anti-actin to localize actin in myofibrils and fixed cultured cells at each stage of the cell cycle. By double immunodiffusion the anti-actin reacted with both smooth and skeletal muscle actin. Several blocking and absorption experiments demonstrated that the antibodies also bound specifically to actin in nonmuscle cells. The same structures stained using either the direct or the indirect fluorescent antibody technique; and, while the indirect method was more sensitive, the direct method was superior because there was no detectable nonspecific staining. As expected, anti-actin stained the I-band of myofibrils. It also stained stress fibers and membrane ruffles in HeLa cells. Some PtK-2 cells have straight stress fibers which stained with anti-actin, but in confluent cultures all PtK-2 cells have, instead, sinuous phase-dense fibers which stained with antibody. At prophase the whole cytoplasm stained uniformly with anti-actin. During metaphase and anaphase, anti-actin staining was concentrated diffusely in the mitotic spindle. In contrast, fluorescent heavy meromyosin stained discrete fine spindle fibers in these fixed cells. During cytokinesis, anti-actin stained the whole cytoplasm uniformly and was not concentrated in the cleavage furrow.  相似文献

A unique set of affinity-purified anti-profilin and anti-actin antibodies generated against a covalently coupled version of the profilin:actin complex was used to assess the distribution of profilin and non-filamentous actin in mouse melanoma cells. In agreement with the profilin:actin complex being the principal source of actin for filament formation, we observed extensive co-distribution of both antibody preparations with vasodilator-stimulated phosphoprotein (VASP) and the p34 subunit of the Arp2/3 complex, both of which are components of actin polymer-forming protein complexes in the cell. This suggests that the localization of profilin and actin revealed with these antibodies in fact reflects the distribution of the profilin:actin complex rather than the two proteins separately. Significantly, protruding lamellipodia and filopodia showed intensive labeling. The two antibody preparations were also used to stain HeLa cells infected with Listeria monocytogenes or vaccinia virus. In both cases, the pattern of antibody staining of the pathogen-induced microfilament arrangement differed, suggesting a varying accessibility for the antibody-binding epitopes.  相似文献

Human antibodies against actin revealed on smeared lymphoblastoid cells a strong staining of numerous microvilli of different lengths extending from the cell surface. Smeared human platelets stained by anti-actin serum showed a bright cytoplasmic fluorescence and projections extending from the surface. Human fibroblasts spread on glass were multi-shaped, and anti-actin serum revealed brightly stained fibers running through the cells. After treatment with cytochalasin B, all types of cells investigated became rounded up, and surface projections could not be demonstrated. The staining pattern indicated a redistribution of the cellular contractile proteins after cytochalasin B treatment. Cytochalasin B did not impair the antigenicity of actin, since presence of the drug did not influence the antibody absorbing capacity of actin.Culture of lymphoblastoid cells and human fibroblasts in the presence of colchicin did not influence the staining pattern of actin antibodies.  相似文献

The presence, spatial distribution and forms of intranuclear and nucleus-associated cytoplasmic actin were studied in Amoeba proteus with immunocytochemical approaches. Labeling with different anti-actin antibodies and staining with TRITC-phalloidin and fluorescent deoxyribonuclease I were used. We showed that actin is abundant within the nucleus as well as in the cytoplasm of A. proteus cells. According to DNase I experiments, the predominant form of intranuclear actin is G-actin which is associated with chromatin strands. Besides, unpolymerized actin was shown to participate in organization of a prominent actin layer adjacent to the outer surface of nuclear envelope. No significant amount of F-actin was found in the nucleus. At the same time, the amoeba nucleus is enclosed in a basket-like structure formed by circumnuclear actin filaments and bundles connected with global cytoplasmic actin cytoskeleton. A supposed architectural function of actin filaments was studied by treatment with actin-depolymerizing agent latrunculin A. It disassembled the circumnuclear actin system, but did not affect the intranuclear chromatin structure. The results obtained for amoeba cells support the modern concept that actin is involved in fundamental nuclear processes that have evolved in the cells of multicellular organisms.  相似文献

Cultures of embryonic mouse spinal cord explants, alone or in combination with rat myotubes, were stained by indirect immunofluorescence using antibodies against three structural proteins to: (a) reveal the distribution of these proteins among different cell types, and (b) test the usefulness of antibody staining to reveal the gross morphology of the neurite network in complex cultures. Affinity column purified antibodies were used against chicken gizzard actin, porcine brain tubulin, and skeletal muscle alpha-actinin. Neurites were stained intensely by anti-actin as was the stress fiber pattern of underlying fibroblasts. With anti-tubulin, the staining of neurites was an order of magnitude more intense than the staining of the microtubule pattern of background fibroblasts. Neurite cell bodies and astrocyte-like glia cells were stained with anti-tubulin and their nuclei remained unstained. Anti-tubulin could thus be used to trace even the finest extensions of nerve processes in spinal cord and spinal cord-muscle cultures. Furthermore, it could be combined with the histochemical reaction for acetylcholinesterase (AChE, EC 3.1.1.7) to demonstrate AChE-positive neurons and specialized nerve-muscle contact sites. The staining of neural elements with anti-alpha-actinin was generally much weaker than with anti-actin and anti-tubulin. Neurites were stained only moderately in comparison to myotube Z lines in the same culture. However, a distinct staining of the periphery of dorsal root ganglion cells was observed. Thus, a protein immunologically related to muscle alpha-actinin is present in the nervous system. In myotubes, Z lines were stained intensely with anti-alpha-actinin while I bands were only faintly stained with anti-actin. In isolated myofibrils, both structures were stained intensely with the same antibody preparations.  相似文献

We examined the distribution of actin in isolated nuclear matrices from mouse leukemia L5178Y cells using an anti-actin antibody and protein A-conjugated colloidal gold particles. Before immunogold staining, we partially digested the surface lamina of the nuclear matrix with trypsin (Nakayasu and Ueda, Exp. Cell Res. 143, 55-62, 1983) to allow penetration of the gold particles into the nuclear matrix. Trypsin digestion slightly modified the internal structure of the nuclear matrix, but did not affect the actin content in the nuclear matrix nor the reactivity of actin with the antibody. Many colloidal gold particles were present along fibrogranular structures in the nuclear matrix. The results reported here confirm the existence of actin in the interior of the nuclear matrices of L5178Y cells.  相似文献

以兔抗肌动蛋白抗体为一抗,FTTC偶联的羊抗兔IgG抗体为二抗进行间接免疫荧光实验,观察到蚕豆(Vicia faba L.)根端分生组织中完整的细胞核和染色体均有明亮荧光。用抗肌动蛋白抗体和蛋白A-胶体金进行标记的免疫电镜实验结果表明,金颗粒分布在蚕豆细胞核中,集缩染色质和核仁中金颗粒较多。经DNaseI消化和2 mol/L NaCl处理得到去除DNA和组蛋白的细胞核和染色体。免疫荧光实验结果指出,去除DNA和组蛋白的细胞核和染色体与抗肌动蛋白抗体呈阳性反应。上述结果说明,肌动蛋白不仅存在于完整的蚕豆细胞核和染色体中,而且存在于去除DNA和组蛋白的蚕豆细胞核和染色体中。另外,用抗原肌球蛋白抗体所做的免疫荧光标记结果表明,原肌球蛋白也存在于蚕豆细胞核和染色体中。对高等植物细胞核和染色体以及核骨架和染色体骨架是否含有肌动蛋白等问题进行了讨论。  相似文献

Meristematic cells of Vicia faba L. were labeled with rabbit anti-actin antibody and FITC-conjugated goat anti-rabbit lgG antibody and observed with fluorescence microscopy. Both the nuclei and chromosomes sent forth distinctive fluorescence, indicating that actin is present in the nuclei and chromosomes. Sections were reacted with the anti-actin antibody and protein A-colloidal gold and observed with transmission electron microscopy. Gold particles were found over the whole nuclei, and a lot of particles were concentrated in condensed chromatin areas and nucleoli, confirming the observations with the fluorescence microscopy. V. faba nuclei and chromosomes were treated with DNase Ⅰ and 2 mol/L NaC1, and DNA and histone-depleted nuclei and chromosomes were obtained. Indirect immunofluorescence tests showed that the DNA and histone-depleted nuclei and chromosomes reacted positively with the anti-actin antibody. These results demonstrated that actin exists not only in intact nuclei and chromosomes but also in DNA and histone-depleted nuclei and chromosomes of V. faba. In addition, the authors' results indicate that tropomyosin is present in the nuclei and chromosomes of V. faba. Presence of actin in nuclei and chromosomes as well as in DNA and histone-depleted nuclei and chromosomes of higher plants is discussed.  相似文献

The purpose of the present study was to investigate the influence of the actin cytoskeleton on gap junctional intercellular communication (GJIC) in cultured astrocytes. This report describes an decrease of GJIC following microinjection of anti-actin antibodies or cytochalasin D treatment. Dye transfer of microinjected neurobiotin was used to assay gap junctional permeability in cultured astrocytes. Besides this translocation of connexins to the plasma membrane we investigated subsequent anti-actin antibody injection. While control cultures exhibited intensive dye spreading of microinjected neurobiotin, GJIC was impaired by microinjection of anti-actin antibodies. Additionally, impaired GJIC was observed after cytochalasin D treatment for 15 min. After the drug had been washed out, a recovery of GJIC was achieved. Cultured astrocytes exhibited a prominent actin cytoskeleton, with strong staining of actin filaments at the plasma membrane. Confocal laser scanning microscopy revealed an impaired translocation of Cx 43 from the Golgi apparatus to the cell membrane of cell processes following anti-actin antibody injection. These results suggest that the morphological integrity of microfilaments seems to be fundamental for GJIC, probably by means of associations among actin filaments, actin binding proteins, and Cx 43 at the plasma membrane or indirectly through the transport of connexins from the cytoplasm to the cell membrane.  相似文献

The distribution of actin was studied during early events of sheep fertilization by fluorescence microscopy after staining with 7-nitrobenz-2-oxal-1.3 diazole (NBD)-phallacidin and anti-actin antibody and by electron microscopy after heavy meromyosin labelling. Unfertilized and fertilized eggs exhibited a continuous band of fluorescence with both NBD-phallacidin and anti-actin antibody. Unlike in mice, no high concentration of actin overlying the spindle was detected in ovulated sheep oocytes. At the site of sperm head incorporation, the fertilization cone developed above the decondensing male chromatin and was underlined by a submembranous area rich in microfilaments. A similar actin network was observed in the cortex of the second polar body. Cytochalasin D was used to investigate the role of actin during the fertilization process. This drug did not prevent sperm fusion and incorporation but inhibited polar body abstriction and fertilization cone development and retarded sperm tail incorporation. Moreover, in the presence of the drug, the anchorage of the metaphase II spindle at the surface of the egg was destroyed. The role of microfilaments in these early events is discussed.  相似文献

在本实验中我们用优化的免疫荧光化学法结合激光共聚焦显微技术,观察了微丝在小鼠卵细胞不同期的分布情况及PKB/Akt对小鼠卵母细胞和早期胚胎的微丝聚合的影响。结果显示,在小鼠卵母细胞及早期胚胎中均有微丝的表达,且主要集中在纺锤体处的质膜处、极体及分裂沟处。注射激活型PKB/Akt mRNA能够增强微丝的聚集。相反,注射激酶失活型的PKB/AktmRNA减弱了微丝的聚合。因而我们认为PKB/Akt可以影响小鼠卵细胞和早期胚胎的微丝聚集。  相似文献

Monoclonal antibodies (IgM kappa) have been produced to actin isolated electrophoretically from L cell extracts. These monoclonal anti-actin antibodies bind to intact L cells and modulate DNA synthesis and cell proliferation, much like affinity-purified polyclonal rabbit antibody to the same Mr 42,000 actin. In addition, monoclonal antibodies specific for actin from Entamoeba histolytica also bound to and modulated the growth of L cells. A monoclonal antibody directed against a neuroblastoma surface antigen did not produce stimulation of L cells, and the binding activity of anti-actin monoclonal antibody to L cells was removed by absorption with actin covalently coupled to Sepharose. These observations demonstrate the specificity of interaction between the anti-actin monoclonal antibodies and the surface of intact L cells. We conclude that a surface actin-like molecule on the L cell, when bound by specific monoclonal antibody, initiates a stimulatory signal which results in enhanced cellular metabolism.  相似文献

Previous immunochemical and immunocytochemical studies have shown that an antibody to actin prepared from body wall muscle of the marine mollusc Aplysia californica is specific for vertebrate cytoplasmic actins. The ability of this anti-actin to distinguish between different forms of actin most likely reflects the recognition of amino acid sequences unique to cytoplasmic actins. We have confirmed the specificity of this antibody for cytoplasmic actins using nervous tissue as a source of cytoplasmic actin in further immunochemical studies. In addition to binding cytoplasmic actin in purified preparations, the antibody removed actin selectively from crude extracts of nervous tissue of some but not all of the species tested. Our results also suggest that tissue-specific differences in the distribution of cytoplasmic actins may exist. Immunofluorescence studies of Aplysia nervous tissue stained with anti-actin revealed that actin is present in the cell body and axonal processes of Aplysia neurons. Although the function of actin in nerve cells is not understood, the observed pattern of immunofluorescence staining is consistent with the idea that actin may be involved in movement within the axoplasm.  相似文献

Triplex DNA is an unusual conformation of DNA formed when two pyrimidine nucleotide strands share a common purine strand. A monoclonal antibody, demonstrated by numerous criteria to be specific for triplex DNA, was used to investigate the presence and distribution of this unique DNA configuration in nuclei and chromosomes of mouse LM cells and human lymphocytes. Indirect immunofluorescence microscopy revealed that constitutive heterochromatin in acetic-methanol fixed mouse nuclei was usually, but not always immunofluorescent, suggesting possible cell cycle related variations in the amount of triplex DNA or its accessibility in this condensed chromatin. In fixed mouse and human chromosomes, there was a positive correlation between immunofluorescent staining patterns, Hoechst 33258 banding, and G- and/or C-banding patterns. Unfixed, isolated mouse chromosomes also reacted positively with the antibody, particularly when they were gently decondensed by exposure to low ionic conditions at neutral pH. This result indicates that fixation is not mandatory for antibody staining, suggesting that some mammalian chromosomal DNA may be naturally organized in a triplex configuration. However, there is a possibility that fixation may facilitate the formation of additional triplex DNA complexes in potential sequences or expose previously inaccessible triplex DNA. The precise correspondence between the immunofluorescent patterns produced by anti-triplex DNA antibodies and G- and C-bands known to represent regions of chromatin condensation, suggests a potential role of triplex DNA in chromosome structure and regional chromatin condensation.  相似文献
