Tick-borne fever (TBF) is caused by the rickettsia Anaplasma phagocytophilum (formerly Ehrlichia phagocytophila) and is a common disease in sheep in areas of Norway infested by Ixodes ricinus ticks. TBF can cause both direct and indirect losses to sheep kept on tick-infested pastures. In the present work we studied a sheep flock of 26 ewes and 50 lambs on pasture from May until September. No cases of TBF had earlier been observed on this pasture. Blood samples from lambs with a reduced weekly weight gain were collected and analysed for A. phagocytophilum infection by blood smear examination. In addition, at the end of the study, sera from all lambs were analysed by an indirect fluorescent antibody assay (IFA) to determine the antibody titre to E. equi. No clinical signs of tick-borne infections were observed, except in one lamb. However, 30 (60%) of the lambs grazing on this pasture became infected with A. phagocytophilum, and the infected lambs had a reduced weight gain (mean) of 3.8 kg compared with the uninfected lambs. The present study indicates that A. phagocytophilum infection may be widespread and contribute to considerable productivity losses even on pastures with no apparent tick infestation. A long-term field study (60 consecutive weeks) was conducted to determine whether a Boophilus annulatus (Say) population could be eradicated through repeated applications of a 1% pour-on formulation of fipronil on cattle held in an infested pasture. Animals treated repeatedly over time (seven applications) were infested with significantly fewer (p < 0.05) female ticks (5 mm in size) than untreated animals in all except one of the 57 weekly tick counts conducted after the first treatment was applied. As the number of treatments increased over time, there was a progressive decrease in the number of instances in which engorging females were observed on the treated cattle. Tick numbers obtained from sentinel cattle placed in pasture with untreated and treated cattle at various intervals throughout the study indicated that a single fipronil treatment had no observable adverse effect on the field tick population. However, after two treatments had been applied, there were always dramatically fewer ticks obtained from sentinel animals placed with treated cattle than were observed on sentinel animals placed with untreated cattle. Furthermore, tick numbers obtained from sentinel cattle placed with treated cattle over time indicated that the tick population in the pasture declined by stages. While a single treatment had no effect on the tick population, the application of two to four fipronil treatments at various intervals dramatically reduced the tick population, even though substantial numbers of females were still present on the sentinel animals. Ultimately, the application of five to seven fipronil treatments resulted in an extremely low tick population in the pasture, although total eradication of the population was not achieved during the study. The mean weight gain of treated animals was significantly higher (p < 0.05) than untreated animals during the study, indicating that the fipronil treatments had a highly beneficial impact on weight gain production.  相似文献

Serum samples of 749 sheep from 75 sheep flocks in Norway, i.e. 361 lambs (6 to 7 months old) and 388 adults (>1.5 year), were analysed for antibodies to Ehrlichia equi. Ten animals from each flock were examined. Seropositive animals were found along the coast of southern Norway from Vestfold to S?r-Tr?ndelag (as far north as 63°38'N). Seropositive sheep were not found in southeast, east or northern Norway. Thirty-two flocks were seropositive, although tick-borne fever had only been diagnosed earlier in half of these. In 78% of the seropositive flocks, more than 80% of the sheep were seropositive. A total of 35.7 % and 36.3 % of lambs and adults were found seropositive, respectively. However, the overall seroprevalence among animals that had been grazing on Ixodes pastures were 0.80 for the lambs and 0.84 for the adults. Mean antibody titres (± SD) (log10) in seropositive lambs and adults were 2.59 (± 0.449) and 2.70 (± 0.481), respectively. No significant differences in either seroprevalence or mean antibody titre between sheep of different ages were obtained in this study. Based on antibodies 94% of sheep flocks on Ixodes pastures were infected with a granulocytic Ehrlichia infection. The association between seropositive flocks and Ixodes infested pasture shows a very high degree of agreement (p < 0.00001). The present study indicates that granulocytic Ehrlichia infection in sheep is underdiagnosed in Norway.  相似文献

The sheep tick is confined almost entirely to rough, poor hill and moorland pastures. Agricultural improvement of these pastures involves (among other things) reduction in the depth of the vegetation layer and partial or complete destruction of the moisture-holding underlying 'mat' of decaying vegetable matter. Assessment of the tick populations on improved and on comparable control areas showed a reduction in tick population following upon pasture improvement. But a fairly considerable degree of improvement is required to bring about a satisfactory reduction. Ticks were not completely extirpated even by ploughing. It is probable that pasture improvement affects tick population mainly by reducing cover and thus the humidity.
Unfortunately, effective pasture improvement is not economic on hill and moorland grazings in general. Such improvement as is practicable could only be a supplement to other control measures.
Since the sheep flock supports almost all the female ticks on a pasture, control is most likely to follow from the development of an efficient acaricidal sheep dip.  相似文献

Lambs born in pens with slatted floor were brought out at 2–5 weeks of age on pastures heavily grazed by sheep the previous years. About 16 days later the oocyst output of the lambs rapidly increased to high levels. Lambs on pastures which never had been grazed by sheep earlier, had moderate oocyst counts. Between 11 and 25 days after the beginning of grazing there were significantly more lambs with diarrhoea on permanent pastures compared with pastures never grazed by sheep earlier. It was found that lambs were heavily infected during the first 2 days on permanent pastures. Thirteen housed lambs were given 10–50 g of soil from a permanent pasture as a water suspension by a stomach tube. Fifteen days later there was a steep rise in the oocyst output in most of them, and 11 of the 13 lambs developed diarrhoea and 2 died. None of 10 lambs given uninfected soil and none of 12 untreated controls showed diarrhoea and the oocyst output remained on a moderate level. It is concluded that oocysts which have survived the winter in the pasture are the main source of infection with Eimeria spp. in lambs with this kind of management. Soil-eating is the most likely source of infection during the first days on pasture.  相似文献

The efficacy of eprinomectin on Dictyocaulus filaria and Cystocaulus ocreatus in naturally infected sheep was evaluated in the present study. In total, 30 infected sheep were randomly divided into two groups: treated (n?=?15) and untreated (n?=?15). A single pour-on dose of eprinomectin (0.5?mg/kg) was administered to the treated group. No medication was used in the untreated group. Faecal larval counts were performed on pre-treatment (day 0) and post-treatment (days 7, 14, 21 and 42) days. Eprinomectin was found to be 100% effective against D. filaria on day 7 post-treatment when compared with the untreated group and it maintained this effect on days 14, 21 and 42. However, the decrease in faecal larval count of C. ocreatus was found to be 86, 86 and 91%, on days 14, 21 and 42, respectively.  相似文献

In Sicily many tick borne diseases are endemic, in particular way those that see like main carrier ticks that prefer, for their vital cycle, climatic conditions characterized by high temperatures and a warmth-humid atmosphere. The more important pathologies transmitted by ticks causing diseases in dogs are babesiosis and ehrlichiosis. Borrelia burgdorferi, Anaplasma phagocytophilum, Rickettsia conorii, Coxiella burnetii and tick transmitted encephalitis virus assume particular relevance because they are agents of zoonosis. Our centre, C.R.A.Ba.R.T, have conducted many researches and carried out many tests for diagnostic aim in order to estimate the spread of the main tick borne diseases in Sicilians' dogs. A study lead on 342 dogs has evidenced seroprevalence for Babesia canis, Ehrlichia canis and Rickettsia respective of 5.17%, 21.70% and 53.43%. A study on zoonotic agent seroprevalences in dogs gave the following percentages: C. burnetii 31.50%, R. conorii 73.60% and A. phagocytophilum 32.80%. The data carried out from IZS Sicily diagnostic service on 5,634 tests done in 2004-2005, confirm the experimental results on the presence of B. canis, E. canis, R. conorii, A. phagocytophilum, C. burnetii, Rickettsia spp., Anaplasma spp. and Ehrlichia spp. in all the Sicilian areas.  相似文献

A study was conducted to determine whether an opioid tonus inhibitory of LH secretion is present in underfed prepubertal sheep. Ten Suffolk ewe lambs were subjected to food restriction during 60 days. During this period they were allowed to pasture only 2 hours per day while control ewe lambs were allowed for 10 hours. Body weight and plasma blood levels of glucose, urea and total proteins were measured weekly. At the end of this period, an intravenous injection of Naloxone (NAL, 1.5 mg/kg BW) was given to control and underfed animals followed 60 min later by an intravenous injection of LHRH to test the pituitary responsiveness. Underfed animals did not show an increase in plasma LH while control animals presented a rise from 0.28 +/- 0.08 to 2.02 +/- 0.6 ng/ml after the NAL stimulus (P less than 0.05). The response to LHRH was similar in both group of animals. Basal plasma levels of insulin were lower in underfed ewe lambs than in control animals (P less than 0.05). Underfed animals were placed on plain feeding with a schedule similar to control lambs for 30 days and the same experiment was repeated. During this occasion, NAL increased plasma LH concentration in both group of lambs. Levels of plasma insulin were not different in both groups. The lack of effect of NAL on LH secretion in food restricted ewe lambs suggests that the opioid modulation of LH secretion is absent by underfeeding in female prepubertal sheep.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Anaplasma phagocytophilum is a bacterium that is transmitted by Ixodes spp. ticks, in which it resides in salivary glands. Ticks inoculate the pathogen into hosts together with an array of salivary molecules that reduce host anti-tick inflammation. Sukumaran et al. recently showed that A. phagocytophilum uses a tick salivary protein, Salp16, to enhance its uptake from the host and into the salivary gland. Occupation and exploitation of tick salivary glands have implications for the maintenance and detection of A. phagocytophilum in its vector and early pathogen interactions with its hosts.  相似文献

Sheep ticks Ixodes ricinus (Acari: Ixodidae) and tick-borne diseases cause major economic losses in both upland sheep farming and moorland shoots of red grouse Lagopus lagopus scoticus. Sheep were treated with acaricide four times between March and October and double-vaccinated against louping ill virus (LIV), instead of the conventional regime of two acaricide treatments and no vaccinations, on two moors in northern England. Enhanced treatment started at Westerdale Moor in 1995 and at Danby Moor in 2000; the latter had previously represented a spatial control site. From 1992 to 2003, grouse chick condition, tick burdens, reproductive success, shooting bags and LIV seroprevalence were measured. A total of 1297 grouse chicks from 398 broods were examined for ticks. Enhanced acaricide treatment reduced tick burdens by 90%, and LIV seroprevalence decreased in relation to the number of years since treatment began. Breeding success and post-breeding densities of grouse in the current sample area remained unrelated to acaricide treatment, tick burdens or LIV seroprevalence, but 25% and 60% more grouse were shot on Westerdale and Danby, respectively, after treatment enhancement than before. By improving shooting bags, tick management schemes help to maintain the economic viability of grouse moors, which, in turn, provide upland landscape and wildlife benefits.  相似文献

The infection rate of Toxoplasma gondii was studied during 6 years in a sheep flock in central Sweden. The flock consisted of 165-249 breeding ewes of which 20-35% were lambs less than 1 year old. Most ewes were slaughtered when 5 years old. The sheep were kept indoors from end of September to early May. Lambing took place in March and April. Individual serum samples were collected twice a year, once just before turning the sheep out to pasture in the spring, and again after housing in the autumn. Sera were analysed by ELISA for antibodies to T. gondii. The seroprevalence varied between 10% and 45% during the 6 years of observation. Seroconversion was detected predominantly at the autumn sampling, indicating that in most cases infection was acquired at pasture. Subclinical effects of T. gondii infection on lamb weight, litter size, total litter weight and ewe weight were also studied. Lambs born to chronically infected ewes were lighter at birth than those of uninfected ewes, but this disparity was no longer evident at weaning.  相似文献

We characterized the effects of subolesin and heat shock protein (HSP) expression on Ixodes scapularis Say (Acari: Ixodidae) stress responses to heat shock and feeding, questing behaviour and Anaplasma phagocytophilum (Rickettsiales: Anaplasmataceae) infection. Ticks and cultured tick cells were analysed before and after subolesin, hsp20 and hsp70 gene knock-down by RNA interference. The results of these studies confirm that HSPs are involved in the tick cell response to heat stress and that subolesin and HSPs are both involved in the tick response to blood-feeding stress and A. phagocytophilum infection. Subolesin and hsp20 are involved in the tick protective response to A. phagocytophilum infection and hsp70 expression may be manipulated by the pathogen to increase infectivity. Importantly, these results demonstrate that subolesin, hsp20 and hsp70 expression also affect tick questing behaviour. Overall, this research demonstrates a relationship between hsp and subolesin expression and tick stress responses to heat shock and blood feeding, A. phagocytophilum infection and questing behaviour, thereby extending our understanding of the tick-host-pathogen interface.  相似文献

The coevolutionary history of Ixodes spp. ticks, the obligately tick-transmitted bacterial pathogen Anaplasma phagocytophilum, and its various rodent reservoir hosts world-wide is not known. According to coevolution theory, the most recently evolved of tick-bacterial complexes could have difficulty maintaining A. phagocytophilum in nature, because transmissibility has not been efficiently maximized. This study was intended to examine the phylogeographic history of I. ricinus-subgroup ticks and A. phagocytophilum, provide an estimate for the date of the divergence of A. marginale and A. phagocytophilum, and evaluate whether there is correspondence between tick and Anaplasma spp. trees. Analysis of Ixodes spp. ticks showed a New World clade consisting of I. scapularis and I. pacificus, European I. ricinus as a sister group to this clade, and Asian I. persulcatus as basal. Of the three A. phagocytophilum genes evaluated, the most resolution was provided by the ankA gene. ankA sequences formed an Old World clade with eastern North America strains as a sister clade. California strains were highly diverse and did not form a clade. Base substitution rates were very comparable along both A. marginale and A. phagocytophilum lineages. Based on 16S rDNA analysis, maximum and minimum divergence times of A. phagocytophilum and A. marginale were calculated to be 78,296,703 and 43,415,708 years, respectively. If A. phagocytophilum did closely coevolve with specific I. ricinus-subgroup tick species, then A. phagocytophilum strains could have specialized on local tick species and optimized local infectivity in the Old World and eastern US. However, lack of absolute resolution of tick trees and conflicting prevalence data (with low prevalence in Asia and western North America) preclude us from inferring a tight coevolutionary relationship of tick species from this phylogeographic analysis.  相似文献

Human anaplasmosis is an emerging tick-borne disease in the United States, but few studies of the causative agent, Anaplasma phagocytophilum, have been conducted in southeastern states. The aim of this study was to determine if A. phagocytophilum is present in small mammals and ticks in northeast Florida. Polymerase chain reaction assays designed to amplify portions of the major surface protein 2 gene (p44), 16S rDNA, and groESL operons were used to test rodent blood and tick DNA samples for the presence of A. phagocytophilum. Positive samples were confirmed by DNA sequence analysis. Anaplasma phagocytophilum DNA was detected in less than 5% of cotton mice and 45% of cotton rats from two sites in northeast Florida. Anaplasma phagocytophilum DNA was also confirmed in 1.3% of host-seeking adult Ixodes scapularis tested and 2.7% of host-seeking adult Amblyomma americanum. This report describes the first DNA sequence data confirming strains of A. phagocytophilum in rodents and ticks in Florida. The DNA sequences of the msp2, 16S rDNA, and groESL gene fragments obtained in this study were highly similar to reference strains of human pathogenic strains of A. phagocytophilum. These findings suggest that A. phagocytophilum is present and established among some small mammal species in northeast Florida. Although the infection prevalence was low in the total number of ticks tested, the presence of A. phagocytophilum in two human biting tick species, one of which is a known competent vector, suggests that humans in this region may be at risk of granulocytic anaplasmosis caused by this pathogen.  相似文献

Abstract This paper examines the efficacy of 10% lambdacyhalothrin-impregnated plastic tags and a deltamethrin pour-on preparation in protecting red grouse chicks from parasitism by ticks and subsequent infection with the louping-ill virus. In 1995, ten red grouse hens ( Lagopus lagopus scoticus ) in a free-living population in north-east Scotland were fitted with lambdacyhalothrin-impregnated plastic tags, glued to radio transmitters. Chicks of more than 10 days of age from a further ten untreated radio-collared hens were caught and fitted with individual tags to the plagium. Both treatments significantly reduced tick burdens in the short term. The number of larvae and nymphs on chicks up to 45 days was less under both treatments than on control chicks and tagged chicks had fewer nymphs than chicks from treated hens. Nevertheless, treatments did not reduce viral infection rates nor increase survival to 10 weeks, possibly explained by incomplete treatment of tagged broods and/or direct or indirect mortality due to tags. In 1996 chicks in ten broods from hens with radio transmitters were individually treated at 14 days of age at a rate of lmg/kg of chick with a deltamethrin pour-on preparation. This preparation significantly reduced the number of larvae and nymphs on grouse chicks 7–10 days after application below the number on untreated controls. At 20 days from application, however, only larval numbers were lower on treated chicks. Nevertheless louping-ill virus infection prevalences were significantly reduced at 35 days of age and survival of chicks to 10 weeks increased.  相似文献

Ixodes ticks harbour several human pathogens belonging to the order Rickettsiales, including Anaplasma phagocytophilum, the agent of human anaplasmosis. When ticks feed on A. phagocytophilum-infected mice, the pathogen enters the ticks' gut. The bacteria then migrate from the gut to infect the salivary glands of the ticks and are transmitted to the next host via the saliva. The molecular mechanisms that enable the migration of A. phagocytophilum from the gut to the salivary glands are poorly understood. Here we show that a secreted tick protein, P11, is important in this process. We show that P11 enables A. phagocytophilum to infect tick haemocytes, which are required for the migration of A. phagocytophilum from the gut to the salivary glands. Silencing of p11 impaired the A. phagocytophilum infection of tick haemocytes in vivo and consequently decreased pathogen infection of the salivary glands. In vitro experiments showed that P11 could bind to A. phagocytophilum and thus facilitate its infection of tick cells. This report provides new insights into A. phagocytophilum infection of ticks and reveals new avenues to interrupt the life cycle of Anaplasma and related Rickettsial pathogens.  相似文献

A three-year survey (1997–99) was carried out on organically reared sheep flocks throughout Sweden. The aim was to determine the prevalence and intensity of nematode infections and to establish relationships between sheep management practices and parasite infections. Faecal samples from ewes and lambs were collected from 152 organic flocks around lambing-time and during the grazing-period for analysis. Results were compared with the different management practices that farmers use to prevent parasitism in their flocks. A high proportion of the flocks was infected with nematodes. The most prevalent species were Haemonchus contortus, Teladorsagia circumeincta, Trichostrongylus axei, T. colubriformis and Chabertia ovina and infections progressively increased during summer in lambs grazing on permanent pastures. Severity of parasitic infection in lambs was highly dependent on egg output from the ewes. H. contortus was found in 37% of the flocks, even at latitudes approximating the Polar Circle. Nematodirus battus was recorded for the first time in Sweden during the course of this study. Lambs turned out onto permanent pasture showed higher nematode faecal egg counts (epg) than lambs that had grazed on pastures, which had not carried sheep the previous year. This beneficial effect of lambs grazing non-infected pastures persisted if the ewes were treated with an anthelmintic before turn-out and if the lambs were kept on pastures of low infectivity after weaning. In lambs, the prevalence and the magnitude of their egg counts were higher during autumn in flocks where lambs were slaughtered after 8 months of age, compared with flocks where all lambs were slaughtered before this age. These results will be used in providing advice to farmers of ways to modify their flock management in order to minimise the use of anthelmintics, but at the same time efficiently produce prime lambs.  相似文献

Over a 2-year period, numbers of infective larvae in samples of pasture herbage, and numbers of worms in previously worm-free “tracer” lambs allowed 4 weeks grazing, were compared as estimators of the abundance of infective larvae on pastures.Transformation of sample estimates of infective larval numbers per 100 g herbage dry matter (DM) and of worm numbers in tracer sheep, according to the expression y = log10 (x+25), was effective in stabilizing variances. Estimates of error variance for each technique did not differ significantly among the genera Haemonchus, Ostertagia or Trichostrongylus and the pooled estimate for the tracer sheep method was 4 times greater than that for pasture sampling. From these results, more tracer sheep than pasture samples would be required to achieve the same level of precision with the two techniques. Using conventional statistical methods, the effects of numbers of pasture samples or tracer sheep on the size of the difference between two means which can be detected as significant and on the width of the confidence interval about a single mean, are illustrated. These can be used as a guide in the choice of sample sizes. Error variances for Nematodirus spp. were significantly less than for the other genera by pasture sampling, and greater by the tracer sheep technique. Possible reasons for this are discussed, but it is concluded that pasture sampling is likely to be much the more precise method for estimating Nematodirus spp. infective larval availability.Changes with time in infective larval abundance, for Haemonchus, Trichostrongylus and Nematodirus spp. which were present in moderate to low numbers, followed similar trends by both techniques. However, for Ostertagia spp. larvae, which were much more abundant, peak levels were defined more sharply and occurred earlier by pasture sampling than by the tracer method. It is suggested that worm counts from tracer sheep, especially those grazing for 4 weeks rather than shorter periods, may systematically underestimate the infective larval population on pasture at high levels of abundance owing to density-dependent worm loss.  相似文献

Twelve Romney lambs and 10 Angora goats were infected with 7000 infective third-stage larvae (89% Trichostrongylus, 11% Ostertagia) collected from goats suspected of harbouring ivermectin-resistant nematodes. On 28 days p.i., the lambs and goats were divided into treatment and control groups of six and five animals, respectively. The animals in the treatment groups were treated with ivermectin (0.2 mg/kg) and necropsied 35 days p.i. Faecal egg counts were estimated on days 28 and 35 p.i. and larval development assays (LDAs) were conducted on 22, 24, 26, 28, 30, 32, 34 and 35 days p.i. The ivermectin treatment reduced Trichostronglus colubriformis burdens by 39% and 13% and Ostertagia circumcincta by 33% and 0% in lambs and goats, respectively. When compared with a susceptible strain, the LDAs indicated a resistance factor before treatment in lambs for T. colubriformis of 2.6 and 1.5 with ivermectin and avermectin B2, respectively, which rose to 3.4 and 2.0 after treatment. The LD50 values of the two control groups were relatively constant throughout the experiment. Prior to ivermectin treatment the LD50 values of the treated groups were similar (P > 0.05) to the control groups but following ivermectin treatment their LD50 values increased steadily until the animals were killed on 35 days p.i. The LD50 values for ivermectin and avermectin B2 of sheep were always slightly higher and significantly different (P < 0.01) than those of goats indicating a host effect on this parameter. The greater reduction in worm counts in goats suggests a difference in the efficacy of ivermectin between lambs and goats. This is the first confirmed report of ivermectin resistance in a field strain of T. colubriformis.  相似文献

Hard ticks are a perennial problem in livestock systems throughout Mexico. No data are currently available on tick prevalence, infestation intensity and population dynamics for sheep in the humid tropics of Mexico. Blackbelly sheep (n = 30) in Tabasco state, Mexico, were examined every 3 weeks for 1 year, and all Ixodidae ticks were counted and removed for analysis. Tick species were identified, infestation prevalence and intensity calculated per animal, and infestation per body zone determined. Overall infestation on the studied animals was 51.9%. Four tick species were identified (Amblyomma mixtum, A. imitator, Rhipicephalus microplus and R. annulatus), the most abundant being A. mixtum (94.9%). Prevalence was highest during the northwinds season (61.9%), followed by the dry (48.5%) and rainy (47.3%) seasons; however, the intensity (2.01–2.07 ticks/sheep) did not differ between seasons. Infestation was most frequent in certain zones including the axilla, crotch, udder, abdomen, thorax and rib area. Sheep in the humid tropics of Tabasco, Mexico, are parasitized by four hard tick species and prevalence is highest in the northwinds season. The present data constitute an important baseline for developing sustainable tick control programs for sheep in the humid tropics of Mexico.  相似文献
