During cytological screening for pollen sterility in a wild population of Haplopappus gracilis (n = 2), several partially sterile plants were found that had good pachytene pairing but varying numbers of univalents. Some plants had chromosome A bivalents or A univalents, while in the same cells chromosome B had only bivalents. In other plants the reverse condition occurred; the B chromosome had B bivalents or B univalents and only A bivalents. This demonstrates a chromosome-specific effect for the desynapsis genes. Hybridization between the two homozygous mutant genotypes produced only normal bivalents; this indicates the two mutants are not alleles and each is recessive. An F2 generation showed independent assortment of the desynaptic mutations. The chromosome A bivalent is the larger of the two and normally has one or two chiasmata; the B bivalent normally has a single chiasma. Chiasmata distribution was tested in the desynaptic mutant A bivalents and showed an acceptable fit to a binomial distribution. This occurs also in heterozygous, asynaptic pairing control gene mutations. Analysis of the NOR bivalent in two hologenomic desynaptic mutations in tomato also showed a good fit to a binomial distribution of chiasmata. This indicates the same methods are applicable to diverse species.  相似文献

Summary Continued inbreeding by self pollination resulted in a proportion of sterile plants in some families of the inbred line IP 1475 of Pennisetum americanum (L.) Leeke. Cytological examinations of the sterile plants revealed mild to extreme desynapsis and also chromosome fragmentation in some plants. Segregation ratios in the selfed families did not fit into any simple Mendelian ratio; however, in one F2 family of the cross desynaptic x normal, segregation into 15 normal: 1 desynaptic was observed. Plants from a segregating family were classified as normals, desynaptics with 2–6 univalents, desynaptics with 2–10 univalents, desynaptics with 10–14 univalents and desynaptics with chromosome fragmentation. Estimation of the content of phosphate and potassium from the flag leaves did not reveal significant differences between the five groups of plants.  相似文献

Abstract The capacity to synthesize heat shock proteins (HSPs) during seed germination of sorghum (Sorghum bicolor) and pearl millet (Pennisetum americanum) has been examined. HSP synthesis is detectable in a thermotolerant genotype of sorghum during the first hour of imbibition of the seed under high temperature stress. A non-coordinate control of HSP synthesis during germination was revealed. Genotypic differences were manifest in the stage of germination at which the ability to synthesize HSPs was first apparent and this related to the thermosensitivity of that genotype.  相似文献

Pennisetum sect. Pennisetum includes two reproductively isolated species. Pennisetum purpureum Schumach. is a tetraploid (2n = 28) perennial species which occurs throughout the wet tropics of the world. Pennisetum americanum (L.) Leeke is a diploid (2n = 14) annual species, native to the semi-arid tropics of Africa and India, and contains three morphologically diverse subspecies. Subspecies americanum includes the wide array of cultivated pearl millets. Subspecies monodii from the Sahel of West Africa is identified as the wild progenitor of pearl millet. Subspecies stenostachyum is morphologically intermediate between subsp. americanum and monodii and includes the mimetic weeds often associated with the cultivation of pearl millet.  相似文献

The proper pairing and recombination of chromosomes during prophase is essential for the formation of gametes during meiosis. As part of studies to identify genes required for homologous chromosome pairing and recombination during meiosis in plants, we characterized a number of T-DNA-tagged, male-sterile mutants of Arabidopsis. Preliminary cytological studies on one line, 7219 which is male and female sterile, suggested that the mutation may disrupt meiosis and result in the formation of aberrant microsporocytes and microspores. In this report we present the results of a detailed analysis of meiosis in microsporocytes of sterile plants to elucidate the nature of the 7219 mutation. Analysis indicates that the mutation usually results in a desynaptic phenotype, with ten sister chromatids observed prior to metaphase I in most cells. Based on this, we named the mutation dsy10. The presence of several other meiotic defects suggests that dsy10 may not be a typical desynaptic mutant. Received: 15 December 2000 / Accepted: 19 April 2001  相似文献

Summary Wide hybridization is a useful tool in plant breeding, but little is known about its possible range. For the cereals, wheat, barley and rye, this was tested with 15 different species of the Poaceae and Panicoideae. Embryo formation could be obtained with Agropyron repens, Alopecurus agrestis, Dactylis glomerata, Festuca glauca, Hordeum bulbosum, Lolium perenne, Pennisetum americanum, and Zea mays. As well, haploid as diploid embryos occurred. New embryo culture techniques should enable these embryos to grow to plants.  相似文献

Summary Two desynaptic mutants, one of spontaneous origin and the other induced through colchicine treatment, were obtained in pearl millet. The desynpatic character was a monogenically controlled recessive one. Tests for allelism have indicated that the desynaptic genes of the two sources were allelic. Both complete and medium-strong desynaptic plants followed a monogenic pattern of inheritance and were found to be allelic. Medium-strong desynpatic plants on selfing produced both complete and medium-strong desynaptics in approximately equal numbers. It is suggested that the differential expression of the desynaptic character might be due to the presence of modifying genes.  相似文献

The response to increasing NaCl concentration of seedlings of 25 accessions of Ethiopian land races of each of Pennisetum americanum (L.) Leeke (pearl millet) and Eleusine coracana (L.) Gaertn (finger millet), and 15 accessions of Eragrostis tef (Zucc.) Trotter (tef), was examined after two week's growth in NaCl solution culture. Although increasing NaCl concentration significantly reduced seedling root lengths, there was considerable variation within, and between accessions within each species.Analysis based upon a non-linear least square inversion method, using root length data, revealed significant differences in accessions of P. americanum and E. tef on the basis of the estimated salinity threshold, C t , the NaCl concentrations at which root length begins to decrease. C t did not differ significantly between E. coracana accessions. Estimates of C50 and C0, mininum concentrations causing a 50% decrease in root length, and zero root growth respectively, revealed differences between and within accessions for all three species. Overall, finger millet was more tolerant than tef, which was more tolerant than pearl millet. There is clear evidence that differences in tolerance are genetically based from broad sense heritability estimates.  相似文献

Summary Protoplasts isolated from embryogenic suspension cultures derived from immature embryos of pearl millet (Pennisetum americanum) gave rise to cell masses. Summary Chromosome association at first meiotic metaphase in tetraploid hybrids between Lolium perenne and L. multiflorum was compared with that in autotetraploid L. perenne. The hybrids were found to have significantly higher levels of bivalent frequency, and lower levels of multivalent and chiasma frequency. A significant increase in multivalent frequency with increasing chiasma formation was found in both groups, but the increase was much less in the hybrids. These differences in chromosome associations between the two groups must therefore reflect differences in chiasma distribution and it is suggested that the results indicate a significant degree of preferential bivalent pairing in the hybrids.  相似文献

The meiosis of two mutants ofBrassica oleracea var.capitata was analysed which have been isolated after gamma irradiation and hybridization.
  1. Univalents appear in different frequencies in the pollen mother cells of both these mutants attributed to a genetically conditioned reduction of the chiasmata frequency resulting in manifold irregularities in the later stages of microsporogenesis. The number of microspores per PMC varies between 1 and 8, the chromosome number of the microspores between 6 and 12. As a consequence of these meiotic disturbances a strong reduction of the fertility of male and female germ cells occurs.
  2. In principle, both mutants show the same meiotic behaviour, but the irregularities appear in a stronger degree in mutant 45 as compared with mutant 47. They are obviously caused by the same mutated gene which shows differences in its manifestation in the two mutants due to their different genotypic constitution.
  3. The mutant gene belongs to the group of desynaptic genes controlling the process of chiasmata formation. The degree of desynapsis caused by this gene is very weak as compared with ds-genes of other species.

A virus disease of pearl millet (Pennisetum americanum) in the Ivory Coast has symptoms consisting of a light-green mosaic of variable severity, followed. by dwarfing. The causal virus is mechanically transmissible and aphid-borne, but not seed-borne. It was purified, has flexuous filamentous particles about 820 nm long, and is a member of the potyvirus group. Its host range, and biological, physico-chemical and serological properties indicate that it is a strain of guinea grass mosaic virus.  相似文献

Summary Male sterility in Pennisetum americanum (L.) Leeke, inbred line IP 482, was found to be inherited as a monofactorial recessive phenotype. Homozygosity for the gene designated ms 2, produced in addition to pollen abortion, plasmodial tapetum, plasmodial pollen mother cells, delayed and asynchronous meiotic development, desynapsis and blockage of meiosis. Successful gamete production is ensured by meiotic quality control, a process in which germ cells that fail in bivalent chromosome formation are eliminated during meiotic prophase. To date, numerous meiotic mutants have been isolated in a variety of model organisms, using defects associated with a failure in bivalent formation as hallmarks of the mutant. Presumably, the meiotic quality control mechanism in those mutants is overwhelmed. In these mutants, all germ cells fail in bivalent formation, and a subset of cells seem to survive the elimination process and develop into gametes. It is possible that mutants that are partially defective in bivalent formation were missed in past genetic screens, because no evident meiotic defects associated with failure in bivalent formation would have been detectable. Meiotic quality control effectively eliminates most failed germ cells, leaving predominately successful ones. Here, we provide evidence supporting this possibility. The Caenorhabditis elegans mrg-1 loss-of-function mutant does not appear to be defective in bivalent formation in diakinesis oocytes. However, defects in homologous chromosome pairing and synapsis during the preceding meiotic prophase, prerequisites for successful bivalent formation, were observed in most, but not all, germ cells. Failed bivalent formation in the oocytes became evident once meiotic quality control was abrogated in the mrg-1 mutant. Both double-strand break repair and synapsis checkpoints are partly responsible for eliminating failed germ cells in the mrg-1 mutant. Interestingly, removal of both checkpoint activities from the mrg-1 mutant is not sufficient to completely suppress the increased germline apoptosis, suggesting the presence of a novel meiotic checkpoint mechanism.  相似文献

The cytological phenotype was studied in a desynaptic form isolated from a population of rye cultivar Vyatka. The primary defect of desynaptic plants was identified as nonhomologous (heterologous) chromosome synapsis, which was observed by electron microscopy of synaptonemal complexes (SCs) in meiotic prophase I. Synapsis defects involved switches of synapsing axial elements to nonhomologous partners, asynapsis in the switching region, and foldbacks formed by the SC lateral elements. Defective bivalent formation was observed at later stages: the univalent number varied and multivalent chromosome associations were observed in single cells in metaphase I. The desynaptic phenotype was controlled by two recessive genes, sy8a and sy8b, which acted and were inherited independently. In a hybrid combination with line Ku-2/63, the desynaptic phenotype was suppressed by the dominant allele of a third gene for inhibitor I; the segregation in hybrid families corresponded to 57:7.  相似文献

Pearl millet (Pennisetum americanum(L.)Leeke) was grown in containers at three constant rates of nitrogensupply or with the nitrogen supply increased from the lowestto the highest rate during panicle differentiation or at anthesis.We measured the rate and duration of nitrogen and dry weightgain by individual grains and nitrogen (15N) uptake by rootsand its distribution during grain filling. The total amountsof nitrogen and dry weight in all grain per plant at the lowestnitrogen supply were 8% and 14% respectively of plants growncontinuously at the highest rate of nitrogen. This was becauselow rates of nitrogen supply reduced grain number, mean grainweight and the nitrogen content of each individual grain. Theamino acid composition of the grain protein was affected onlyslightly by nitrogen treatments. Rates of grain growth were sensitive to nitrogen supply whereasthe duration of nitrogen movement to the grain was not. Nitrogenuptake by roots continued throughout grain filling; rates ofuptake per g root in plants given least nitrogen were one-halfthose of plants given the highest amount of nitrogen. A changefrom lowest to highest nitrogen supply at panicle differentiationincreased the uptake of nitrogen by roots and the rates of growthof individual grains, to the rates observed in plants whichhad been supplied continuously with the highest nitrogen. Whenthe change in supply was made at anthesis there was rapid movementof nitrogen into the plant but this was not translated intomore rapid grain growth. The formation of chiasmata in six full sib male partially asynaptic individuals of Locusta migratoria has been studied. The mean chiasma frequency per cell was 2.3 both at diplotene and metaphase I. Chiasmata tended to be distributed evenly among the bivalents. The frequency and distribution of the chiasmata in each type of bivalent (L, M, or S) depended on the level of asynapsis and on interference between the bivalents. Short bivalents were the most affected by interference, while M bivalents associated independently of L and S bivalent behaviour.  相似文献

Mitochondrial DNA (mtDNA) from cell suspension cultures of two intergeneric somatic hybrids of Pennisetum americanum (pearl millet) + Saccharum officinarum (sugarcane) was examined by restriction endonuclease digestion and hybridization with sorghum mtDNA cosmids. The mtDNA of one somatic hybrid was indistinguishable from that of pearl millet, while the second exhibited a combination of parental mtDNAs, suggesting mitochondrial fusion. Endonuclease restriction fragment patterns of Pennisetum americanum L. mitochondrial DNAs (mtDNAs) from a cytoplasmic male-sterile (CMS-A1), fertile revertants and a normal fertile cytoplasm were variable, while chloroplast DNA from those lines lacked variation. Comparisons between mtDNAs of CMS-A1 (parental) and fertile revertant lines revealed the presence of a unique 4.7 kbp PstI fragment in the sterile line that was not detected in any of the revertant lines. A 9.7 kbp PstI fragment was found in all of the revertants, but not in the CMS-A1. Neither of those fragments was found in the normal cytoplasm mtDNA. Hybridization studies revealed two sets of multiple homologies: 1) the 4.7 kbp fragment had homology with a 10.9 kbp and a 13.6 kbp fragment; and 2) the 9.7 kbp fragment was homologous with the 13.6 kbp fragment. The presence of those two repeated mitochondrial sequences on the altered fragments suggests that they may be involved in the recombinational associated events with reversion from CMS to fertility in P. americanum.Florida Agricultural Experiment Station Journal Series No.7797.  相似文献
