The morphological characteristics of the pectoral fin spine were compared in three species of sturgeon, the Persian sturgeon (Acipenser persicus), the Russian sturgeon (Acipenser gueldenstaedtii), and the Starry sturgeon (Acipenser stellatus), all sampled from the Caspian Sea. On the basis of morphological characters of the pectoral fin spine, 62.2% of the individuals were correctly classified into separate groups. The cluster analysis also divided the three species into two major subgroups. Acipenser persicus and A. gueldenstaedtii were grouped together, suggesting a similar evolutionary basis. Significant morphological heterogeneity in pectoral fin spine characteristics was observed among the three sturgeon species. Principal component analysis identified the largest differences were in the pectoral fin spine size and the angle between distal pectoral fin spine and the horizontal line (A°). The first and second principal components (PC1 and PC2) of all observations accounted for 64.19% and 14.33% of the total variation, respectively. The combination of all analyses showed the relevance of applying pectoral fin spine shape for interspecific distinction of the three species of sturgeons.  相似文献

The effect of two feeding rates (0.5 and 1.5% of total body weight) was assessed on the growth of pectoral fin spines of captive juvenile great sturgeon, Huso huso, after second year of life. The fish received Oxytetracycline (OTC) twice in the first and second years of their lives under basic diet. During the 5‐month experimental period, juveniles (mean 1,187.4 g, 0.1 standard deviation [SD]), n = 50) were reared with two feeding rates under similar conditions in 10 fiberglass tanks (1.5 m3). The fish were fed manually with a commercial diet twice a day (35% Biomar, Nersac, France) throughout the experiment. The OTC marks were distinguished in all pectoral fin spine sections under ultraviolet light. The means of the first and second annular radii were 806.6 µm (27.2 SD) and 2,246.5 µm (50.2 SD), respectively. The marginal increment analysis beyond the second OTC mark revealed a significantly smaller marginal increment for low feeding rate treatment (143.9, 11.2 SD) as compared to the high feeding rate one (269.0, 14.6 SD). The results indicate the slower growth rate in the fish fed the low feeding treatments seen in the pectoral fin spine formation, which can be used as an indicator of recent feeding history in sturgeon juveniles. The best daily feeding rate for great sturgeon of 2,460 g was determined to be 1.5% body weight/day in this study.  相似文献

Atlantic sturgeon migrate between ocean and freshwater habitats to spawn, and juveniles spend several years in fresh/brackish water before returning to the ocean. Because strontium/calcium (Sr/Ca) ratios are diagnostic for freshwater and marine environments, we examined the utility of energy-dispersive x-ray fluorescence (EDXRF) to quantify Sr/Ca ratios of Atlantic sturgeon pectoral fin spines. Atlantic sturgeon spines from wild adults and experimental juveniles were analyzed along a linear transect from the primordium to the outermost point. To verify the technique hatchery juvenile Atlantic sturgeon were held in experimental tanks at <0.5, 13?C15, or 33?C35?? and sampled after 5?months. Sr/Ca ratios of experimental hatchery fish increased with salinity, and Sr/Ca ratios in wild adults varied predictably along the measurement transect. However, the ratio decreased in the outermost region of the spine in mature fish collected during a return to freshwater for spawning. Therefore EDXRF is a useful tool to track individual movements of Atlantic sturgeons and other diadromous fish.  相似文献

We tested the hypothesis that δ13C and δ15N signatures of pectoral spines would provide measures of δ13C and δ15N similar to those obtained from fin clips for adult shovelnose sturgeon Scaphirhynchus platorynchus. Thirty‐two shovelnose sturgeon (fork length [FL] = 500–724 mm) were sampled from the lower Mississippi River, USA on 23 February 2013. Isotopic relationships between the two tissue types were analyzed using mixed model analysis of covariance. Tissue types differed significantly for both δ13C (P < 0.01; spine: mean = ?23.83, SD = 0.62; fin clip: mean = ?25.74, SD = 0.97) and δ15N (P = 0.01; spine: mean = 17.01, SD = 0.51; fin clip: mean = 17.19, SD = 0.62). Neither FL nor the FL × tissue type interaction had significant (P > 0.05) effects on δ13C. Fin clip δ13C values were highly variable and weakly correlated (r = 0.16, P = 0.40) with those from pectoral spines. We found a significant FL‐tissue type interaction for δ15N, reflecting increasing δ15N with FL for spines and decreasing δ15N with FL for fin clips. These results indicate that spines are not a substitute for fin clip tissue for measuring δ13C and δ15N for shovelnose sturgeon in the lower Mississippi River, but the two tissues have different turnover rates they may provide complementary information for assessing trophic position at different time scales.  相似文献

Age-dependent accumulation of 137Cs in the muscles and bodies of the pike Esox lucius (aged two to seven years) inhabiting a section of the Yenisei River polluted with artificial radionuclides has been studied. The content of 137Cs in muscles varied from 0.5 to 7.0 Bq/kg of fresh weight. The maximum content of the radionuclide has been found in juveniles. The content of 137Cs in pike muscles and body decreased considerably with age. The high content of 137Cs in the muscles of juveniles is probably a consequence of their higher intensity of feeding as compared to older individuals, which is due to the intense growth of juveniles.  相似文献

For a wide range of proteins of high interest, the major obstacle for NMR studies is the lack of an affordable eukaryotic expression system for isotope labeling. Here, a simple and affordable protocol is presented to produce uniform labeled proteins in the most prevalent eukaryotic expression system for structural biology, namely Spodoptera frugiperda insect cells. Incorporation levels of 80 % can be achieved for 15N and 13C with yields comparable to expression in full media. For 2H,15N and 2H,13C,15N labeling, incorporation is only slightly lower with 75 and 73 %, respectively, and yields are typically twofold reduced. The media were optimized for isotope incorporation, reproducibility, simplicity and cost. High isotope incorporation levels for all labeling patterns are achieved by using labeled algal amino acid extracts and exploiting well-known biochemical pathways. The final formulation consists of just five commercially available components, at costs 12-fold lower than labeling media from vendors. The approach was applied to several cytosolic and secreted target proteins.  相似文献

Traditional techniques for stable isotope analysis (SIA) generally require sacrificing animals to collect tissue samples; this can be problematic when studying diets of endangered species such as the pallid sturgeon Scaphirhynchus albus. Our objectives were to (i) determine if pectoral fin tissue (non‐lethal) could be a substitute for muscle tissue (lethal) in SIA of juvenile pallid sturgeon, and (ii) evaluate the influence of preservation techniques on stable isotope values. In the laboratory, individual juvenile pallid sturgeon were held for up to 186 day and fed chironomids, fish, or a commercially available pellet diet. Significant, positive relationships (r² ≥ 0.8) were observed between fin and muscle tissues for both δ15N and δ13C; in all samples isotopes were enriched in fins compared to muscle tissue. Chironomid and fish based diets of juvenile pallid sturgeon were distinguishable for fast growing fish (0.3 mm day?1) using stable δ15N and δ13C isotopes. Frozen and preserved fin tissue δ15N isotopes were strongly related (r2 = 0.89) but δ13C isotopes were weakly related (r2 = 0.16). Therefore, freezing is recommended for preservation of fin clips to avoid the confounding effect of enrichment by ethanol. This study demonstrates the utility of a non‐lethal technique to assess time integrated food habits of juvenile pallid sturgeon and should be applicable to other threatened or endangered species.  相似文献

Trace element and 87Sr/86Sr isotope analyses of fish pectoral fin rays offer non-destructive methods for determining habitat use. In this study, water and fin ray samples were analyzed for Gulf Sturgeon Acipenser oxyrinchus desotoi from the Choctawhatchee River Basin (FL and AL, USA) and compared with reference samples from Atlantic Sturgeon A. o. oxyrinchus held at controlled salinities (0, 10, 33 ppt). Samples were analyzed using inductively coupled plasma mass spectrometry, with a multi-collector for 87Sr/86Sr. In water, Sr, Ba, Mn and Zn differed between freshwater and saline habitats, with increases in Sr and decreases in Ba, Mn and Zn. 87Sr/86Sr decreased upstream to downstream with lowest values in saline habitats. In the reference study, water trace element concentrations and 87Sr/86Sr corresponded to those in pectoral fin rays. 87Sr/86Sr was higher in pectoral fin ray than water, due to influence of diet, which differed with salinity. In wild fish, trace elements in pectoral fin rays indicated freshwater emigration to saline habitats primarily occurred in the second to third growth zone with some heterogeneity in the population (4% <0.3 years, 39% 0.5–1.3 years, 39% 1.5–2.3 years, 17% 2.5–3.3 years). Analyses of 87Sr/86Sr indicated initial locations of Gulf Sturgeon were in the middle river, with few fish in the upper or lower river. Most (74%) juvenile Gulf Sturgeon utilized more than one river region prior to freshwater emigration and 48% moved upstream temporarily based on increased 87Sr/86Sr. After initial freshwater emigration, fish utilized lower-river to saline habitats. Collectively, these studies demonstrate the usefulness of trace element and 87Sr/86Sr analyses in sturgeon pectoral fin rays.  相似文献

A new platycephalid, Rogadius mcgroutheri, is described on the basis of the specimens collected from eastern Australia and New Caledonia. Rogadius mcgroutheri is distinguished from other congeners by 11 second dorsal fin rays usually, 4 or 6–8 unbranched lower pectoral fin rays, larger orbital diameter, usually single preocular spine lacking the accessory spines on the anterior base, short antrorse preopercular spine, tooth band on palatine narrow, with 2 irregular tooth rows, body with indistinct or somewhat distinct brown blotches, and caudal fin with dark brown spots and bands.  相似文献

Cation/H+ exchangers (CAXs) are membrane proteins that transport Ca2+ and other cations using the H+ gradient generated by H+-ATPase or H+-pyrophosphatase. This study reports the characterization of CAX2 from Puccinellia tenuiflora with respect to molecular and functional properties. PutCAX2 was cloned from a cDNA library of P. tenuiflora seedlings. The expression of PutCAX2 in shoots and roots was induced by Ca2+ and Ba2+ treatments. A green fluorescent protein (GFP) marker revealed that PutCAX2 was located on the endoplasmic reticulum (ER) membrane. Four yeast transformants were created using GFP fusion PutCAX2 and truncated PutCAX2s, and their growth in the presence of various cations (Fe3+, Al3+, Mn2+, Cu2+, Co2+, Ni2+, Mg2+, Zn2+, Na+, Li+, Ca2+, and Ba2+) was analyzed. The N-terminally truncated PutCAX2 (GFP-ΔNPutCAX2) and the N and C-terminally truncated PutCAX2 (GFP-ΔNCPutCAX2) transformants grew well in the presence of 100 and 150 mM Ca2+ or 8 and 20 mM Ba2+, whereas the GFP-PutCAX2 and C-terminally truncated PutCAX2 (GFP-ΔCPutCAX2) transformants did not show any tolerance to Ca2+ or Ba2+. The Ba2+ content in whole yeast cells expressing GFP-ΔNPutCAX2 or GFP-ΔNCPutCAX2 was lower than that in other yeast transformants. Moreover, the efflux experiment showed that the Ba2+ efflux rate of yeast cells expressing GFP-ΔNPutCAX2 and GFP-ΔNCPutCAX2 was higher than that of other yeast cells. To our knowledge, this is the first report on the molecular and functional characterization of a novel ER-localized CAX protein from a wild halophyte plant; the results suggest that the N-terminus of PutCAX2 acts as an auto-inhibitory domain, which affects the Ca2+ and Ba2+ tolerance of yeast.  相似文献

Recently it was shown that female fish injected with enriched stable isotopes maternally transmit a chemical signature to larval otoliths. Validation of this larval marking technique requires laboratory experiments to determine appropriate injection concentrations and to assess any negative effects on larval and adult condition. This study investigated the temporal profile of 137barium assimilation and retention in tissues of adult female anemonefish Amphiprion melanopus (Pomacentridae) following intraperitoneal injection with either 2 or 4 μg 137Ba g?1 body mass. Mean barium isotope ratios (138Ba:137Ba) in the two groups of treated fish were not significantly different from each other, but were significantly different from those in the control group up to 56 days post-injection. This pattern of 137Ba retention was consistent across gonad, muscle, liver and bone tissues. Mean plasma cortisol concentration (an indicator of non-specific physiological stress) was not significantly different among groups and was considered to be representative of unstressed fish. Together, these results indicate that (1) A. melanopus suffer minimal physiological stress and cope well after treatment with 137Ba, (2) 137Ba is retained in female A. melanopus for a prolonged period (at least 56 days), such that multiple clutches of offspring are likely to be marked with an isotopic signature, and (3) a lower dosage of 2 μg 137Ba g?1 appears sufficient for transgenerational marking. It is concluded that 137Ba is suitable for use as a transgenerational marker and is a powerful tool to resolve long-standing enigmas such as larval dispersal distances and the fishery benefits of marine reserves.  相似文献

There are few data reported on radionuclide contamination in Antarctica. The aim of this paper is to report 137Cs, 90Sr and 238,239+240Pu and 40K activity concentrations measured in biological samples collected from King George Island (Southern Shetlands, Antarctica), mostly during 2001–2002. The samples included: bones, eggshells and feathers of penguin Pygoscelis papua, bones and feathers of petrel Daption capense, bones and fur of seal Mirounga leonina, algae Himantothallus grandifolius, Desmarestia anceps and Cystosphaera jacquinotii, fish Notothenia corriceps, sea invertebrates Amphipoda, shells of limpet Nacella concina, lichen Usnea aurantiaco-atra, vascular plants Deschampsia antarctica and Colobanthus quitensis, fungi Omphalina pyxidata, moss Sanionia uncinata and soil. The results show a large variation in some activity concentrations. Samples from the marine environment had lower contamination levels than those from terrestrial ecosystems. The highest activity concentrations for all radionuclides were found in lichen and, to a lesser extent, in mosses, probably because lichens take up atmospheric pollutants and retain them. The only significant correlation (except for that expected between 238Pu and 239+240Pu) was noted for moss and lichen samples between plutonium and 90Sr. A tendency to a slow decrease with time seems to be occurring. Analyses of the activity ratios show varying fractionation between various radionuclides in different organisms. Algae were relatively more highly contaminated with plutonium and radiostrontium, and depleted with radiocesium. Feathers had the lowest plutonium concentrations. Radiostrontium and, to a lesser extent, Pu accumulated in bones. The present low intensity of fallout in Antarctic has a lower 238Pu/239+240Pu activity ratio than that expected for global fallout.  相似文献

Natural variability in stable isotope ratios and element concentrations in calcified structures of fish (e.g. scales and otoliths) has provided biogeochemical ‘tags’ for studying origins and movements of marine species, but has been little used in freshwater studies. We examine whether variability in scale δ15N and δ13C values of Salmo trutta L., could provide a tag of fish over small spatial scales in a small river catchment (River Dee, U.K.) and compared their performance as tags with that of scale/otolith element concentrations. Whole scale δ15N and δ13C values differed among six study sites and fish could be classified to their site of origin with a high degree of accuracy. Classifying fish to their site of capture was marginally superior using scale δ15N and δ13C values compared to that achieved using Sr, Mn, Ba and Mg in scale hydroxyapatite or otolith aragonite. Scale δ15N and δ13C values could therefore provide non-lethally collectable biogeochemical tags superior in performance to element concentrations in otoliths and scales. A comprehensive study of δ15N and δ13C values within freshwater systems would develop our understanding of factors influencing geographical variability in baseline δ15N and δ13C signatures.  相似文献

Early life stages of Artedidraco skottsbergi and A. shackletoni were collected off Adélie Land. The morphology and pigmentation pattern of nine larvae and juveniles of A. skottsbergi between 17.2 and 21.4 mm in standard length (SL), and of two juveniles of A. shackletoni measuring 25.1 mm SL were described. A. skottsbergi was characterized by a heavily pigmented body, except for the caudal peduncle, with distinctively dense pigmentation on the ventrolateral half of the body and caudal section (17.2–17.9 mm SL). Furthermore, they had no pigmentation on the pectoral fin base until they attained 21.4 mm SL. Juvenile A. shackletoni had a heavily pigmented body except for the ventral side of the abdomen and the anal fin base. The proximal part of the dorsal fin and most of the anal fin were covered with melanophores. Although knowledge of larval and juvenile Artedidraco species is limited, the distribution of melanophores on the fins, pectoral fin base and caudal peduncle at each developmental stage may be useful for species identification.  相似文献

Nuclear magnetic resonance spectroscopy studies of ever larger systems have benefited from many different forms of isotope labeling, in particular, site specific isotopic labeling. Site specific 13C labeling of methyl groups has become an established means of probing systems not amenable to traditional methodology. However useful, methyl reporter sites can be limited in number and/or location. Therefore, new complementary site specific isotope labeling strategies are valuable. Aromatic amino acids make excellent probes since they are often found at important interaction interfaces and play significant structural roles. Aromatic side chains have many of the same advantages as methyl containing amino acids including distinct 13C chemical shifts and multiple magnetically equivalent 1H positions. Herein we report economical bacterial production and one-step purification of phenylalanine with 13C incorporation at the Cα, Cγ and Cε positions, resulting in two isolated 1H-13C spin systems. We also present methodology to maximize incorporation of phenylalanine into recombinantly overexpressed proteins in bacteria and demonstrate compatibility with ILV-methyl labeling. Inexpensive, site specific isotope labeled phenylalanine adds another dimension to biomolecular NMR, opening new avenues of study.  相似文献

For the first time by the example of juveniles of Russian sturgeon Acipenser gueldenstaedtii Brandt, the role of growth hormone of hypophysis in the osmotic regulation of acipenserids was studied, also the process of transformation of reserve chloride was described. In Russian sturgeon juveniles, administration of growth hormone of carp Cyprinus carpio at a dose of 1 mg per 1 g body weight causes changes in the fine structure of numerous reserve (undifferentiated) gill epithelial cells and thus stimulates their transformation into mature chloride cells involved in transport of monovalent ions. Increase in the number of chloride cells, on account of reserve cells, was accompanied by higher enzymatic activity of Na+/K+-ATPase in gill homogenates of the injected fish adapting to a hyperosmotic environment of salinity 12.5‰ (403 mosm/l) than in the intact fish under the same conditions but without hormonal treatment. Morphophysiological changes observed in the injected juveniles provided the more efficient dynamics of blood serum osmolarity under saline exposure than that in the intact fish. Their serum osmolality during the maximum rise (24 hours after the transfer of fish into the water of 12.5‰ salinity) was lower (p <0.01), than that of the intact fish. The inclusion of growth hormone in the osmoregulatory process was established.  相似文献

The Ca2+-conducting pathway of myocytes isolated from the cricket lateral oviduct was investigated by means of the whole-cell patch clamp technique. In voltage-clamp configuration, two types of whole cell inward currents were identified. One was voltage-dependent, initially activated at –40 mV and reaching a maximum at 10 mV with the use of 140 mM Cs2+-aspartate in the patch pipette and normal saline in the bath solution. Replacement of the external Ca2+ with Ba2+ slowed the current decay. Increasing the external Ca2+ or Ba2+ concentration increased the amplitude of the inward current and the current–voltage (I–V) relationship was shifted as expected from a screening effect on negative surface charges. The inward current could be carried by Na+ in the absence of extracellular Ca2+. Current carried by Na+ (I Na) was almost completely blocked by the dihydropyridine Ca2+ channel antagonist, nifedipine, suggesting that the I Na is through voltage-dependent L-type Ca2+ channels. The other inward current is voltage-independent and its I–V relationship was linear between –100 mV to 0 mV with a slight inward rectification at more hyperpolarizing membrane potentials when 140 mM Cs+-aspartate and 140 mM Na+-gluconate were used in the patch pipette and in the bath solution, respectively. A similar current was observed even when the external Na+ was replaced with an equimolar amount of K+ or Cs+, or 50 mM Ca2+ or Ba2+. When the osmolarity of the bath solution was reduced by removing mannitol from the bath solution, the inward current became larger at negative potentials. The I–V relationship for the current evoked by the hypotonic solution also showed a linear relationship between –100 mV to 0 mV. Bath application of Gd3+ (10 M) decreased the inward current activated by membrane hyperpolarization. These results clearly indicate that the majority of current activated by a membrane hyperpolarization is through a stretch-activated Ca2+-permeable nonselective cation channel (NSCC). Here, for the first time, we have identified voltage-dependent L-type Ca2+ channel and stretch-activated Ca2+-permeable NSCCs from enzymatically isolated muscle cells of the cricket using the whole-cell patch clamp recording technique.Abbreviations I Ca Ca2+ current - I Na Na+ current - I–V current–voltage - NSCC nonselective cation channel Communicated by G. The Mn2+ concentrations in the sagittae otoliths of 12 fish families (and 19 species) that co-occur in a coastal area of southeastern Brazil (~21°S) were quantified using electron paramagnetic resonance (EPR). Inferences were made about the relationship between fish habitat and trace element incorporation. Inferences were made on the relationship between trace element concentration and otolith shape. The differences in Mn2+ concentrations among the species suggest that habitat (and feeding habits) might drive the incorporation of this trace element into fish otoliths, with higher values in bottom-associated fish species than in surface-associated species. In surface-associated fish species, the correlation between trace element concentrations and otolith shape was stronger than in bottom-associated species. Thus, while the Mn bioavailability in a fish’s habitat, especially from feeding resources, is a local driving influence of trace element incorporation in sagittae otoliths, species-specific requirements also have an influence. Quantitative EPR is a non-destructive technique that is very useful when the available samples cannot be damaged, like with otolith collections.  相似文献

A review of the species and subspecies of Ditrema from East Asia recognized the following taxa: D. jordani Franz, 1910, D. temminckii pacificum subsp. nov., D. t. temminckii Bleeker, 1853, and D. viride Oshima, 1940. Ditrema jordani is characterized by a coppery-red body when fresh, the posterior end of the dorsal-fin base anterior to that of the anal-fin base, a dark rounded marking on the anterior suborbital area margined with white lines or a broad dark inverse trapezoid marking on the anterior part of the suborbital region, the lower half of the spinous portion of the dorsal fin often with a longitudinal black stripe, and a faint longitudinal dark line along the anal-fin base. Ditrema temminckii is characterized by a silvery body, usually bluish dorsally when fresh, a black oblique band on the anterior suborbital area, the spinous portion of the dorsal fin with a black distal margin, and no dark line along the anal-fin base. Ditrema t. pacificum, mainly distributed along the Pacific Ocean coast, differs from D. t. temminckii, mainly distributed along the coast of Sea of Japan, in lacking a dark spot on the anterior portion of the preopercle (vs. dark spot present in the latter), but having a black line along the posterior margin of the pelvic-fin spine (vs. black spot anteriorly on base of pelvic fin), fewer dorsal-fin spines (usually 9 or 10 vs. 10 or 11), more dorsal-fin soft rays (mode 21 vs. mode 20), and longer pectoral, pelvic, and caudal fins. The two nominal species (D. smitti and D. leave) are regarded as junior synonyms of D. t. temminckii. Ditrema viride is characterized by a silvery, dorsally yellowish-green body when fresh, 9–11 dorsal-fin spines (mode 10), a dark triangular marking on the anterior suborbital area, the spinous portion of the dorsal fin with a black distal margin, a longitudinal black line along the anal-fin base and the pelvic fin slightly darkish, and lacking a black spot anteriorly on the base. Neotypes are designated for D. jordani and D. viride. A key to the species and subspecies of Ditrema is provided.  相似文献

The potential use of pectoral fin spines for ageing purposes was examined in Aristotle’s catfish (Silurus aristotelis) and validated for growth proportionality and growth mark periodicity. Monthly samples (n = 436, total length range: 11.10–36.70 cm) were collected in Lake Pamvotis, Greece from November 2002 to December 2003. Marginal increment analysis showed that the spine width (SW) radius grew proportionally to total body length (TL) (TL = 9.581 + 6.83SW, r2 = 0.66, n = 431, P < 0.01), and was at a minimum in January and February, indicating a single growth mark formation per year (i.e. annulus). Further analyses showed that the morphometric characteristics of the spines did not differ significantly (r2 > 0.80 in all cases) when removed from one side of the fish or the other. The procedure was based on individuals no older than 5 years of age; extrapolation to higher age classes may be critical.  相似文献
