Abstract— The concentration of beta-trace protein, a low molecular weight water-soluble protein, was significantly higher in cerebral and cerebellar white matter than in grey matter. A similar distribution was found for transferrin. The concentrations of gamma-trace protein and pre-albumin were almost constant in cerebral and cerebellar white and grey matter. A different distribution was shown for albumin, betalc/betalA globulins, and the immunoglobulins G, A and M, with the highest concentrations mostly encountered in the highly vascularized basal ganglia and grey matter, and the lowest concentrations in white matter. Thus, different parameters, hitherto unknown determine the distribution within the central nervous system of various proteins-those which originate from serum, and beta-trace protein which originates mainly from the central nervous system.
The amounts of the different proteins were higher in the choroid plexus than in brain tissue, with the exception of gamma-trace protein.
Foetal brains contained increasing concentrations of beta-trace protein and of transferrin with age.
Femoral nerve contained lower concentrations of beta-trace protein and gamma-trace protein, and higher concentrations of the other proteins, than the central nervous system.  相似文献

The free amino acid levels in the peripheral nerve of the crab, Carcinus maenas, have been measured using various extraction procedures. A comparison of the amino acid levels of the peripheral and central nervous system of the crab has revealed marked differences in the levels of certain amino acids. Aspartate and glutamate were much lower in central nervous tissue than peripheral whilst glycine was much higher. The results are discussed in relation to the extreme steepness of the concentration gradient for amino acids across the neuronal membranes of crustaceans and a comparison is made with other groups. The significance of the differences in amino acid concentration in specific areas of nervous tissue is discussed in relation to the functions of the amino acids and to possible transmitter activity.  相似文献

The effect of environmental ion concentration on the enzyme activity of astrocytes was investigated in tissue cultures of rat cerebral cortex. It was found that the oxidative enzymatic activity (succinic dehydrogenase, DPN-diaphorase, and several other enzymes) of astrocytes depended on the concentration of NaCl in the environment. This response was not specific for NaCl, but was also elicited by MgCl2 and LiCl; the response was less consistent, and often questionable for KCl. However, only NaCl could elicit enzymatic changes in astrocytes at concentrations known to be present in a living organism. Astrocytes were the only cells which responded this way; it appeared that the foot-plates were particularly involved in the response since increase of enzyme activity occurred earlier in the foot-plates than in the perikarya. It was concluded that astrocytes are metabolically involved in the maintenance of the ionic and osmotic environment of the central nervous system, particularly in regard to the active transport of sodium.  相似文献

The cephalochordate amphioxus is the closest invertebrate relative to vertebrates. In this study, using HPLC technique, free L-amino acids (L-AAs) and D-aspartic acid (D-Asp) have been detected in the nervous system of the amphioxus Branchiostoma lanceolatum. Among other amino acids glutamate, aspartate, glycine, alanine and serine are the amino acids found at the greatest concentrations. As it occurs in the nervous system of other animal phyla, glutamate (L-Glu) and aspartate (L-Asp) are present at very high concentrations in the amphioxus nervous system compared to other amino acids, whereas the concentration of taurine and gamma-aminobutyric acid (GABA) is very low. Interestingly, as it is the case in vertebrates, D-aspartic acid is present as an endogenous compound in amphioxus nervous tissues. The physiological function of excitatory amino acids, and D-aspartate in particular, are discussed in terms of evolution of the nervous system under an Evo-fun (Evolution of function) perspective.  相似文献

—The lipid composition of the central nervous system of some marine vertebrates and two mammalian species (rat and man) was analysed by one- and two-dimensional quantitative thin-layer chromatography, and the cerebroside fatty acids were analysed by gas chromatography. The concentrations of sphingomyelin and cerebrosides are higher in mammals than in fishes, while no significant differences are observed for other lipid classes. Furthermore, in mammals the ratio between hydroxy and normal fatty acids in the cerebrosides is much higher than in fishes. The cerebrosides of mammals contain more very long chain fatty acids than those of marine vertebrates.  相似文献

—Putrescine, spermidine, spermine, RNA, DNA and protein concentrations were determined in 14 parts of the rat nervous system. If the concentrations are expressed in DNA units, putrescine and spermidine concentrations change concomitantly in the different brain parts, with the exception of hypothalamus, where relatively higher putrescine than spermidine concentrations are observed. The constancy of putrescine/spermidine ratios indicates the value of putrescine concentration as an index of spermidine biosynthesis. Spermidine correlates with RNA, except in medulla, spinal cord and peripheral nerves. It is assumed that the relative excess of spermidine in these structures indicates an additional functional role. Spermine/DNA ratios are remarkably constant in the diencephalic and telencephalic regions; they are also nearly constant, but significantly lower in midbrain, medulla, spinal cord and cerebellum. This observation gives additional support for the preferential interrelation of spermidine with RNA and spermine with DNA; i.e. for different functional roles of these two narrowly related polycations.  相似文献

After the intraventricular injection of 6-hydroxydopamine (6-OHDA), there was a long lasting reduction in the brain concentrations of noradrenaline (NA) and dopamine (DA). The brain concentration of NA was affected by lower doses of 6-OHDA than were required to deplete DA. A high dose of 6-OHDA which depleted the brain of NA and DA by 81 per cent and 66 per cent respectively, had no significant effect on brain concentrations of 5-hydroxytryptamine (5-HT) or γ-aminobutyric acid (GABA). The fall in catecholamines was accompanied by a long lasting reduction in the activities of tyrosine hydroxylase and DOPA decarboxylase in the hypothalamus and striatum, areas in the brain which are rich in catecholamine containing nerve endings. There was, however, no consistent effect on catechol-O-methyl transferase or monamine oxidase activity in these brain regions. The initial accumulation of [3H]NA into slices of the hypothalamus and striatum was markedly reduced 22–30 days after 6-OHDA treatment. These results are consistent with the evidence in the peripheral sympathetic nervous system that 6-OHDA causes a selective destruction of adrenergic nerve endings and suggest that this compound may have a similar destructive effect on catecholamine neurones in the CNS.  相似文献

Vasoactive intestinal polypeptide (VIP) occurs in high concentrations throughout the gut and the nervous system. The presence of VIP has been shown in a number of species, mainly by immunohistochemistry. The aim of the present study was to develop a new, highly specific VIP radioimmunoassay to investigate the distribution of VIP in the central nervous system of various vertebrate and invertebrate species. Different areas of the brain and spinal cord were removed from rats, chickens, turtles, frogs and fishes. The cerebral ganglia and the ventral ganglionic chain were investigated in the earthworm. The tissue samples were processed for VIP radioimmunoassay. Our results show that the antiserum used in the radioimmunoassay turned to be C-terminal specific, without significant affinity to other members of the VIP peptide family. Detection limit of the assay was 0.1 fmol/ml. Highest concentrations were found in the turtle diencephalon, followed by other brain areas in the turtle and rat. All other brain areas in the examined species contained significant levels of VIP. Immunoreactivity was also shown in the cerebral and ventral ganglia of the earthworm. In summary, our results show comparative quantitative distribution in representative species of the phylogenetic line, using the same experimental conditions.  相似文献

The sympathetic nervous system and the adjuvant adrenalin-secreting system are found in their earliest form in the annelid kingdom, and consist of cells situated in the central nervous system which are the common ancestors of both, and which are both secretory and nervous in function. 2. These cells are developed in the annelid kingdom parallel with the development of a contractile vascular system, which possesses muscles comparable in physiological actions with the muscle of the vertebrate heart. 3. This vascular muscle is regulated by the processes of the common ancestral cells as well as by their secretory activity. 4. In the primitive form contractile rhythm is an intrinsic property of cardiac muscle; its nerve supply regulates the rhythm, it does not initiate it. The beat is therefore myogenic, not neurogenic. 5. The contractile vascular system of annelids is mainly branchial in function. The vertebrate heart has been derived from it by the growing around of the lateral body folds to form a new ventral surface.  相似文献

Nucleosides are neuromodulators that have a wide range of biological roles in the brain. In order to better understand the function of nucleosides in the human central nervous system (CNS), we constructed a nucleoside map showing the concentration of various nucleosides and their metabolites using post mortem samples from 61 human brain areas and 4 spinal cord areas. We evaluated in vivo tissue levels of four nucleosides (uridine, inosine, guanosine, and adenosine) and three of their metabolites (uracil, hypoxanthine, and xanthine). The concentrations of nucleosides were unevenly distributed across different brain regions, where the highest levels were found in the cerebral cortex and basal ganglia, whereas the lowest concentrations were located in the locus coeruleus, the zona incerta, the substantia nigra, and the inferior colliculus. The regional differences in nucleoside levels in the CNS may reflect the distinct physiological functions adopted by these compounds in different brain areas.  相似文献

—A mass fragmentographic procedure is described for the simultaneous quantification of a number of deaminated metabolites derived from tyramine, octopamine, dopamine, and norepinephrine. With this method, several of the metabolites were measured in normal rat brain. The results support the central nervous system origin of tyramine, octopamine and their metabolites. The concentration of the dopamine metabolite, homovanillic acid, in the rat brain was found to be about 15% higher than that of dihydroxyphenylacetic acid. As for the metabolites of norepinephrine, vanilmandelic acid concentration was found to be about 5% that of 3-methoxy-4-hydroxyphenylglycol. The possible role of vanilmandelic acid in the CNS metabolism of norephrine is discussed.  相似文献

To explore the submicroscopic structure of the human callus by the polarization optical method, serial sections were prepared in three principal planes and the sign of double refraction as well as optic axes of various regions of the sections was determined. It was concluded that in areas under the grooves keratin is oriented parallel to the surface plane of the callus and to the direction of the grooves. In component structures of ridge areas such as in sweat duct areas a circumferential, in cross-band areas a linear, and in other parts a random structure was seen. The cross-bands of ridge areas in association with groove areas revealed a regular lattice-like submicroscopic pattern which was considered as an important mechanical system of the cornified epithelium.  相似文献

Levels of substance P immunoreactivity (SPI) were determined in several skin and mucosal areas, in parts of the sympathetic nervous system, the urinary, biliary and respiratory systems of cats, rabbits and guinea-pigs, and in various skin and mucosal areas of humans by radioimmunoassay. Salient findings are (1) The general distribution pattern of SPI in rabbits was similar to that in rodents. (2) The highest SPI tissue levels were found in the sympathetic nervous system, notably in guinea-pigs. (3) The guinea-pig also had the highest SPI levels in ureter, urinary bladder and bile duct. (4) The aorta, pulmonary artery and portal vein of the rabbit contained very low amounts of SPI, the concentration in the carotid sinus being several fold higher. (5) Skin SPI content was generally highest in the cat, especially in the hindpaw-pad, and lowest in abdominal and back skin. (6) SPI levels found in postmortem human skin and mucosal samples are comparable to those found in other mammals. The observations are discussed in view of the sensory innervation of the various tissues.  相似文献

Abstract— Drugs possessing (chlorpromazine, haloperidol, clozapine, thioridazine and sulpiride) or lacking (benzoctamine and perlapine) antipsychotic activity were compared with respect to their ability to enhance x-methyl-p-tyrosine-induced dopamine disappearance from the mesolimbic area and corpus striutum of rat brain. In addition, their effects on the endogenous concentrations of homovanillic (HVA) and 3.4-dihydroxyphenylacetic (DOPAC) acids in these two brain areas were determined. Some of the drugs enhanced dopamine disappearance in the mesolimbic area more than in the striatum. The most active in this respect were sulpiride. perlapine and chlorpromazine. By contrast, haloperidol was slightly more active in the striatum than in the mesolimbic area. None of the drugs was more efficient in elevating HVA levels in the mesolimbic area than in the striatum. However, there were large differences in the relative extent of the HVA increases in the two regions. Benzoctamine, perlapine and chlorpromazine increased HVA concentrations in the mesolimbic area nearly as much as in the striatum. Thioridazine and haloperidol, however, elevated striatal HVA much more effectively. Haloperidol and clozapine increased the DOPAC concentration in both areas to about the same extent. The other drugs were more active in the striatum. The largest difference between both regions was shown by chlorpromazine. Perlapine and benzoctamine, both lacking antipsychotic activity, produced much larger increases of HVA than of DOPAC. This is in contrast to the results obtained with true neuroleptics and may reflect an involvement of release phenomena in the action of these two drugs on dopamine metabolism. These results suggest that a preferential increase of dopamine turnover in the mesolimbic area is not necessarily linked to a better ratio of antipsychotic activity vs. extrapyramidal side effects. Moreover, an antiacetylcholine component of dopamine receptor blocking drugs does not seem to be a prerequisite for preferential activity on dopamine turnover in the mesolimbic system.  相似文献

The concentrations of glycine and 6 other amino acids have been assayed in the CSF and plasma of the rat, and regional heterogeneity of CSF amino acid concentration has been found. Steady state flux rates into the cranial and spinal fluid compartments were determined during perfusion with amino acid free medium. The transfer of glycine from blood into both the cranial and spinal subarachnoid fluids was saturable with only several-fold elevations of plasma glycine. The results are discussed with regard to the putative neurotransmitter function of glycine in the spinal cord.  相似文献

Abstract— Studies were made on isolated ganglia of the locust in a modified O2 electrode. The basic requirements of a suitable incubation medium were determined and found to include saturation of the medium with 100% O2, and 10m m -glucose as energy source. The effects of incubation in vitro were determined for a number of biochemical characteristics including O2 uptake and the concentrations of Arg P, ATP, ADP, glycogen, glucose, lactate and pyruvate. Action potentials were recorded from a major nerve both in vivo and in vitro. On the basis of these biochemical and physiological studies, it was concluded that the tissue functions well in vitro. Some comparisons were made between the metabolic characteristics found in the locust nervous system, and those of mammalian tissue. The locust ganglia were found to have a rate of O2 uptake much higher than comparable mammalian tissue, and to contain a considerable reserve of glycogen. The low ratio of lactate/pyruvate suggests a more aerobic metabolism than is the case in mammalian brain.  相似文献

The preparation of an extract of the mealworm larvae, Tenebrio molitor L. which hydrolyses ethyl butyrate and o -nitrophenyl acetate, but not acetylcholine, is described. The inhibition of this esterase by TEPP-containing materials and parathion was determined.
An enzyme that hydrolysed o -nitrophenyl acetate and was inhibited by a TEPP-containing material was demonstrated in the five other insect species used.
The relative toxicities as contact insecticides to adult Tribolium castaneum Hbst. of ten samples of TEPP-containing materials were compared with their relative activities as esterase inhibitors. There was not an exact quantitative correlation between TEPP content estimated chemically, insecticidal activity and anti-esterase activity; but the correlation was sufficient to suggest interdependence of these factors.
Eggs of Diataraxia oleracea L. and Ephestia kühniella Zell, were shown to contain an enzyme that hydrolysed o -nitrophenyl acetate and was inhibited by the TEPP-containing materials. This enzyme was present in eggs less than 24 hr. old, i.e. before there was any visible signs of development. The TEPP was shown to be toxic to these eggs and in high concentrations kills at an early stage of development before differentiation of the nervous system. This, in conjunction with the other evidence, suggests that esterases other than the choline-esterase of the nervous system are important when considering the toxic action of these compounds.
Comparison of the anti-esterase activity and toxicity of parathion and TEPP-containing materials as insecticides showed that although the TEPP materials were the more potent enzyme inhibitors, parathion was the more potent contact insecticide to five species of insects. This appears to be due to the relative instability of TEPP. The study of the rates of action of the two poisons applied at different concentrations supports this view.  相似文献

1. Chemical examination of the cell sap of Nitella showed that the concentrations of all the principal inorganic elements, K, SO4, Ca, Mg, PO4, Cl, and Na, were very much higher than in the water in which the plants were growing. 2. Conductivity measurements and other considerations lead to the conclusion that all or nearly all of the inorganic elements present in the cell sap exist in ionic state. 3. The insoluble or combined elements found in the cell wall or protoplasm included Ca, Mg, S, Si, Fe, and Al. No potassium was present in insoluble form. Calcium was predominant. 4. The hydrogen ion concentration of healthy cells was found to be approximately constant, at pH 5.2. This value was not changed even when the outside solution varied from pH 5.0 to 9.0. 5. The penetration of NO3 ion into the cell sap from dilute solutions was definitely influenced by the hydrogen ion concentration of the solution. Penetration was much more rapid from a slightly acid solution than from an alkaline one. It is possible that the NO3 forms a combination with some constituent of the cell wall or of the protoplasm. 6. The exosmosis of chlorine from Nitella cells was found to be a delicate test for injury or altered permeability. 7. Dilute solutions of ammonium salts caused the reaction of the cell sap to increase its pH value. This change was accompanied by injury and exosmosis of chlorine. 8. Apparently the penetration of ions into the cell may take place from a solution of low concentration into a solution of higher concentration. 9. Various comparisons with higher plants are drawn, with reference to buffer systems, solubility of potassium, removal of nitrate from solution, etc.  相似文献

Abstract— The natural fluorescence of hemicholinium-3 (HC-3) was used to provide a method for determining its concentration in brain tissue following intraventricular administration of the compound to rats and rabbits. When hemicholinium-3 was perfused through the cerebral ventricles of the rabbit, the highest concentration was found in the pons-medulla oblongata. The effect of a number of compounds on the uptake of hemicholinium-3 in the brain was studied in rats. Choline and tetramethylammonium significantly reduced the uptake of HC-3 from the cerebral ventricles but tetraethylammonium, physostigmine and atropine had no effect. The results are discussed in relation to the known effects of hemicholinium-3 in the central nervous system.  相似文献

为了解温度及氮磷浓度对平裂藻和栅藻生长及竞争的影响,分别在18、25和35℃条件下,以BG11培养基为基础,设置了不同的氮磷浓度组,进行了2种藻的单独培养试验和混合培养试验,结果表明:在纯培养条件下,低营养浓度组(磷浓度分别为0.1和0.5 mg/L)平裂藻在3种温度下基本停止生长,高营养盐浓度组(磷浓度分别为1.0和1.5 mg/L)除18℃1.0 mg/L组停止生长外,其余均能正常生长增殖,且细胞最大密度表现为25℃ 18℃ 35℃, 1.5和1.0 mg/L浓度组细胞生长差异不显著。栅藻在纯培养条件下,细胞密度总体表现为随着营养盐浓度升高而增加的趋势,细胞最大密度表现为18℃ 25℃ 35℃。在混合培养试验中,除35℃、1.5 mg/L组平裂藻对栅藻的竞争抑制参数大于栅藻对平裂藻的竞争抑制参数外,其余试验组均表现为栅藻受平裂藻影响较小。在混合培养体系中, 0.1 mg/L组平裂藻仍基本停止生长, 0.5 mg/L组受栅藻的刺激作用,生长优于纯培养体系。1.0和1.5 mg/L组均受栅藻的抑制作用,且浓度越高抑制作用越强。    相似文献
