Summary The occurrence and effects of a gametic disequilibrium (DSE) in the first generation of a theoretical two-population synthetic variety were investigated. Theoretical development was limited to the genetics at a single locus with two alleles in an autotetraploid species with random chromosome inheritance. Algebraic expressions were developed for the differences between the mean genotypic values of the two-population synthetic variety at generation one and in random mating equilibrium (RME). For the situation where both parents of the synthetic were in RME, a numerical analysis was performed for all possible allele frequencies assuming the following types of genic action: monoplex dominance, partial monoplex dominance, duplex dominance, partial duplex dominance, and additive. The result indicated that with non-additive genic action the DSE could, in some cases, greatly depress or inflate the mean genotypic value of the first generation (Syn-1(RME)). Thus, any change of means over advancing generations with loss of DSE could be positive or negative. When additive genic action was assumed, there was no effect associated with DSE and when both parents had the same allele frequencies there was no DSE. The DSE, with only a minor exception, decreased the genetic variance and in numerous cases forced it near zero. Expressions were developed for mean genotypic values of a first generation synthetic with DSE in one parent (Syn-1(DSE/RME)) or both parents (Syn-1(DSE)). The deviation of these means from those of Syn-1(RME) was a function of digenic and quadragenic population effects. An inspection of the response equations for Syn-1(RME) indicated that in a series of crosses with one common parent the rankings of first generation means would be the same as the ranking of populations at equilibrium though the individual means would be biased. More importantly with DSE of one or both parents there are situations when a ranking of first generation mean genotypic values would not reflect relative frequency of desirable alleles in the populations. Summary Microgametophytic selection was investigated using two ecologically diverse autotetraploid clones of alfalfa. Several selection pressures (drying, aging, freezing, and high and low temperatures) were applied to microgametophytes at three stages of the life cycle, 1) during microsporogenesis, 2) post-anthesis, and 3) pollen tube growth. Pollen aging produced a progeny population with a greater mean plant size and a lower coefficient of variation than the control progeny. High temperature (29.5 °C) applied both during microsporogenesis and pollen tube growth resulted in progeny populations which were significantly taller and, in one case, had a larger leaf number than the control populations. In contrast, air dried pollen resulted in a progeny population which had significantly smaller character means and larger coefficients of variation than the control population. Also, low temperature (15 °C) during pollen tube growth yielded progeny with reduced branch number and a larger coefficient of variation than the control progeny. In cases where progeny derived from selected microgametophytes were found to differ from the control offspring, corresponding shifts in the reciprocal cross were not observed. For the temperature stress treatments, the lack of reciprocal differences may be related to the different temperature adaptations of the two ecotypes. These results suggest that microgametophytic selection can be effective in shifting the mean of the progeny generation; however, the results obtained will vary depending upon the selection pressure, stage of selection, and the parents used.  相似文献

Summary The effects of a gametic disequilibrium (DSE) in an autotetraploid population on response to selection as measured by the covariance of selection were investigated. The theoretical responses were calculated for mass selection [Mass (1)] and half-sib progeny test selection (HSPT) in a two-allele (B and b), single locus, autotetraploid population. The complexity of calculations precluded analytical expressions for the covariances so numerical analysis was used assuming the following genetic models: monoplex dominance, partial monoplex dominance, duplex dominance, partial duplex dominance, and additive gene action.The results indicated the DSE could greatly affect the covariance of selection. For a constant allele frequency the DSE might double the covariance expected with selection in a population at random mating equilibrium (RME) of gametes, but in other instances approach zero. For all genetic models and the two breeding methods investigated the covariance of selection was always increased when the frequency of BB gamete exceeded p2 (where p is frequency of allele B) and decreased when the frequency of BB gamete was less than p2. The possible incorporation of this information into a long term breeding program and some other ramifications were briefly discussed.With the DSE the covariances of selection with HSPT and Mass (1) had a proportionality of 1:2, respectively, with the additive genetic model, but this relationship rarely occurred for other genetic models. Summary Two-locus theory for recurrent selection for general combining ability in maize was developed. The theory featured: (a) recombination of the selfed progeny of selected parents; and (b) linkage disequilibrium in the initial gametic array. The theory indicated: (a) that initial linkage disequilibrium exerts a permanent influence upon selection progress; (b) that interposition of one or more generations of random mating before each cycle reduces the permanent effect in ensuing cycles; and (c) that random mating done before initiation of selection is more efficient in removing the influence of linkage disequilibrium on selection progress than random mating done between subsequent cycles.  相似文献

The amplified fragment length polymorphism (AFLP) assay is an efficient method for the identification of molecular markers useful in the improvement of numerous crop species. The identification of AFLP markers linked to disease resistance genes has been shown in segregating populations from crosses of inbred lines. The development of inbred lines in alfalfa is not possible, but existing breeding programs have produced populations selected for resistance to a single pest. Two such populations, UC-123 and UC-143, differing only in selection for resistance to downy mildew (Peronospora trifoliorum de Bary) isolate I-8, were used in this study. Thirty-six resistant plants from UC-143, and 36 susceptible plants from UC-123 were screened for DNA polymorphisms using fourteen AFLP primer combinations. Four AFLP fragment markers, ACACTC208, ACACTC150, ACACAT216 and ACACTC486, were found to be significantly associated with disease susceptibility or resistance. Resistant and susceptible plants were crossed in a diallel scheme and the progeny were screened for resistance to P. trifoliorum isolate I8. Two of the AFLP markers, ACACTC208 and ACACTC486 were significantly associated with resistance in the F1 and S1 progeny. The utilization of two populations, comprised of 36 resistant and 36 susceptible plants, for the identification of DNA fragments associated with disease resistance proved successful. Seventy-two plants is a very manageable number and provides a starting point for further refinement of marker-trait associations.  相似文献

Summary Estimates of general combining ability of parents for yield and girth obtained separately from seedlings and their corresponding clonal families in Phases II and IIIA of the RRIM breeding programme are compared. A highly significant positive correlation (r = 0.71***) is found between GCA estimates from seedling and clonal families for yield in Phase IIIA, but not in Phase II (r = –0.03NS) nor for girth (r= –0.27NS) in Phase IIIA. The correlations for Phase II yield and Phase IIIA girth, however, improve when the GCA estimates based on small sample size or reversed rankings are excluded.When the best selections (based on present clonal and seedling information) are compared, all five of the parents top-ranking for yield are common in Phase IIIA but only two parents are common for yield and girth in Phases II and IIIA respectively. However, only one parent for yield in Phase II and two parents for girth in Phase IIIA would, if selected on clonal performance, have been omitted from the top ranking selections made by previous workers using seedling information.These findings, therefore, justify the choice of parents based on GCA estimates for yield obtained from seedling performance. Similar justification cannot be offered for girth, for which analysis is confounded by uninterpretable site and seasonal effects.  相似文献

The effects of methyl jasmonate (MeJA) in relation to abscisic acid (ABA) on different phases of somatic embryogenesis were studied in Medicago sativa L. Different concentrations of both the growth inhibitors (0.0, 0.5, 5.0, 50.0 and 500.0 μM) were tested in five distinct phases of somatic embryogenesis, viz., induction, proliferation, differentiation, maturation and regeneration. Like ABA, MeJA also inhibited callus induction, callus growth, proliferation of embryogenic suspension as well as germination and conversion of somatic embryos. However, its inhibitory effects on various phases of somatic embryogenesis were less pronounced as compared to that due to ABA. In contrast to ABA, MeJA did not have any significant influence on the development of somatic embryos when applied in the differentiation phase. The study showed that ABA used routinely as an inducer of somatic embryo maturation in M. sativa could not be replaced by MeJA.  相似文献

Isogenic diploid and tetraploid alfalfa (Medicago sativa L.) was studied with molecular markers to help understand why diploid performance and breeding behavior does not always predict that of tetraploids. In a previous study of partially heterozygous alfalfa genotypes, we detected a low correlation between yields of isogenic diploid (2x) and tetraploid (4x) single-cross progenies, and genetic distances were more highly correlated with yields of tetraploids than diploids. These differences may be related to the level of RFLP heterozygosity expected among progenies derived from heterozygous parents at the two ploidy levels. The objectives of this study were to determine the relationships among genetic distance, forage yield and heterozygosity in isogenic 2 x and 4 x alfalfa populations. Four diploid genotypes were chromosome doubled to produce corresponding isogenic autotetraploids, and these genotypes were mated in 4 × 4 diallels to produce 6 single-cross families at each ploidy level for field evaluation. Allele compositions of parents were determined at 33 RFLP loci by monitoring segregation of homologous restriction fragments among individuals within progenies, and these were used to estimate RFLP heterozygosity levels for all single-cross progenies at both ploidy levels. RFLP heterozygosity rankings were identical between progenies of isogenic diploid and tetraploid parents; but significant associations (P < 0.05) between estimated heterozygosity levels and forage yield were detected only at the tetraploid level. Since tetraploid families were nearly 25% more heterozygous than the corresponding diploid families, inconsistencies in the association between molecular marker diversity and forage yields of isogenic 2 x and 4 x single crosses may be due to recessive alleles that are expressed in diploids but masked in tetraploids. The gene action involved in heterosis may be the same at both ploidy levels; however, tetraploids benefit from greater complementary gene interactions than are possible for equivalent diploids. The dynamics of biomass and N accumulation following defoliation of alfalfa and the application of N fertilization has rarely been studied under field conditions, particularly in the seeding year. Our objectives were to determine the effect of N fertilization on the dynamics of biomass and N accumulation during the first regrowth of alfalfa in the seeding year, and to determine if a model describing critical N concentration developed for established stands could be used in the seeding year. In two separate experiments conducted in 1992 and 1993, the biomass and N accumulation of alfalfa grown with three N rates (0, 40 and 80 kg N ha-1) were determined weekly. Maximum shoot growth was reached with 40 kg N ha-1 in 1992, and maximum shoot growth was not reached with the highest N fertilization rate in 1993. Nitrogen fixation, root N reserves and soil inorganic N uptake when no N was applied were, therefore, not sufficient to ensure non-limiting N conditions, particularly when growth rates were the highest between 14 to 21 d after defoliation. Nitrogen fertilization increased shoot biomass accumulation in the first 21 d of regrowth, biomass partitioning to the shoots and shoot and taproot N concentrations. The model parameters of critical N concentration developed by Lemaire et al. (1985) for established stands of alfalfa were not adequate in the seeding year. The N requirements per unit of shoot biomass produced are greater in the seeding year than on established stands, and this was attributed to a greater proportion of leaves in the seeding year.  相似文献

Summary The need to compare pressure-chamber estimates of leaf water potential with a psychrometric method has been established for several crop species. We investigated this relationship for rice (Oryza sativa L.) as well as the need to protect leaves from water loss during sampling and measuring period in the pressure chamber. Two rice cultivars grown in containers on a clay-loam soil were stressed to varying degrees by withholding water. Fully expanded leaves were sampled for estimation of leaf water potential by the dew point hygrometer and pressure-chamber techniques. The same leaf was used in both methods allowing direct comparison. Additionally, two alternative methods of leaf handling for measurement by the pressure chamber technique were compared. Protection of leaf samples against water loss during excision, transport and handling was found to be more important at higher leaf water potentials (>−1.0 MPa). The two cultivars used appeared to differ in their response to protection of the leaf sample. These results serve to further caution pressure chamber users on extrapolating comparisons between the two measurement methods and between tissue handling techniques even within a crop species.  相似文献

Summary For 25 Medicago sativa plants, measurements were made of in vitro pollen germination and growth and callus growth on mediums containing culture filtrate from two isolates of Fusarium oxysporum f. sp. medicaginis. In vivo resistance was determined by field inoculation with one isolate. There was no correlation between any in vitro measurement and in vivo resistance, and none of the in vitro measurements on control mediums were correlated. The only significant correlation for the same measurement between the two isolates was for pollen-tube length (r = 0.65). Percent of pollen germination was negatively correlated with callus growth for both isolates. Earlier work showed that callus growth in the presence of culture filtrate was linked to plant resistance, thus it appeared that percent pollen germination on culture filtrate had decreased for the more resistant plants. The haploid pollen may be used as a progeny test for identifying heterotic loci conferring resistance to Fusarium, but if the pollen of this plant species is used in direct selection by exposure to culture filtrate, the level of resistance to Fusarium may decrease.  相似文献

Summary Several large RNA molecules that migrated to electrophoretic positions ranging from 1.7–10 kb were observed in preparation of alfalfa (Medicago sativa) mitochondria. F1 progenies inherited the RNA's from both maternal and paternal parents (Fig. 1). Treatment of intact mitochondria with RNase A failed to remove the RNA's, indicating that they were contained within an RNase impermeable compartment. Further purification of mitochondria in linear sucrose gradients failed to separate the RNA's from mitochondria. Transmission electron microscopic examination of sucrose gradient purified mitochondria revealed that mitochondria were free of contamination by virus-like particles, indicating that the RNA's were contained within the mitochondrion. Biparental inheritance of large mitochondrial RNA's in alfalfa provides evidence that mitochondria are inherited biparentally in this species.  相似文献

The complications introduced by the autotetraploid, outcrossing nature of alfalfa (Medicago sativa L.) as related to detecting associations of marker loci and traits of interest are discussed, and a new method of detecting marker-trait associations is suggested. This method utilizes plant populations that are likely to have been produced through the plant breeding process: populations selected for one trait, and the base, unselected population. Marker allele frequency shifts between the populations are indicative of genomic regions involved in trait expression, and may indicate alleles that have reached the triplex or homozygous state and do not segregate in S1 or F1 populations. However, because many, perhaps hundreds, of sequential frequency comparisons are needed to detect fragments in significantly different proportions in the two populations, the type I error rate is very high. A resampling-based analysis method is proposed to address the concern of the type I error rate, and identify marker alleles associated with this trait of interest. The utility of marker-trait associations thus defined for identifying individual plants from heterogeneous populations was investigated through model-building and conditional probability studies. Factors investigated that influenced the utility of the marker associations and (in the base population) the frequencies of the trait and marker, and the frequencies of the markers in plants exhibiting the trait and in the plants not exhibiting the trait. The frequency of occurrence of a marker in undesirable plants profoundly influenced the efficiency with which the marker could be used to select desirable plants, however, under some circumstances, markers or combinations of markers can be highly efficient for selecting rare, desirable plants from a heterogeneous base population.  相似文献

Genes conferring resistance to kanamycin are frequently used to obtain transgenic plants as spontaneous resistance to kanamycin is not known to exist in higher plants. Nevertheless, mutations conferring kanamycin resistance have been identified in Chlamydomonas reinhardtii, raising the question as to why kanamycin-resistant mutants have not been found in higher plants. While attempting plastid transformation of alfalfa, we obtained non-transgenic but kanamycin-resistant somatic embryos following 2 months of culture in the presence of 50 mg l–1 kanamycin. Sequencing of the plastid DNA region corresponding to the decoding site of the 16S rRNA in ten independent resistant events revealed an A to C transversion at position 1357 of the 16S plastid rDNA, the same site at which an A to G conversion confers kanamycin resistance to C. reinhardtii by reducing the ability of the antibiotic to bind to its target site. All plants derived from the resistant embryos through additional cycles of somatic embryogenesis in the absence of kanamycin retained the mutant phenotype, suggesting that the mutation was homoplastomic. Resistant plants produced 85% less biomass than controls; their leaves were chlorotic during early development and over time slowly turned green. The absence of kanamycin- resistant mutants in higher plants might be explained by the requirement for a regeneration system capable of resulting in homoplastomic individuals, or it may be the result of the detrimental effect of the mutation on the phenotype.Communicated by C.F. Summary The potential breeding value of 2n gametes from diploid alfalfa (2n = 2x = 16) was tested by comparing single cross alfalfa hybrids produced via 2n = 2x gametes from diploids versus n = 2x gametes from somatic-chromosome-doubled, tetraploid counterparts. Three diploid clones, designated 2x-(rprp), homozygous for the gene rp (conditions 2n gamete formation by a first division restitution mechanism) were colchicine-doubled to produce their tetraploid counterparts, designated 4x-(SCD). These six clones were crossed as males to the same cytoplasmic male sterile clone. Yield comparisons of progeny from the six clones demonstrated a significant yield increase of the hybrid progeny from 2n = 2x gametes from the diploids over the hybrid progeny from n = 2x gametes from the chromosome doubled tetraploid counterparts. The yield gain ranged from a 12% increase to a 32% increase. Theoretical comparisons indicated the 2n = 2x gametes from diploids would have 12.5 to 50% more heterozygous loci, on average, than the n = 2x gametes derived from somatic doubling. These results confirm the importance of heterozygosity on alfalfa yield, and the results demonstrate that 2n gametes formed by first division restitution offer a unique method for producing highly heterotic alfalfa hybrids.  相似文献

Fall dormancy results in decumbent, slow shoot growth of alfalfa (Medicago sativa L.) in autumn and reduced shoot regrowth rates after herbage removal in summer. Although fall dormancy is used to predict alfalfa adaptation, we possess a poor understanding of the biological mechanisms underlying fall dormancy. Our objective was to examine growth and carbohydrate metabolism of suspension cell cultures derived from contrasting alfalfa cultivars that genetically differed in fall dormancy. Suspension cells were grown in B5h media containing 2% sucrose. Cells derived from fall non-dormant plants accumulated sugars more rapidly after transfer to fresh media and to higher concentrations than did cells derived from fall dormant alfalfa cultivars. Dark respiration rates of cells derived from non-dormant plants were similar to those derived from fall dormant plants when growth was limited at low cell sugar concentrations. However, both cell growth and dark respiration rates increased in cells derived from non-dormant cultivars in response to greater cell sugar concentrations. High growth rates of cells derived from rapid growing, fall non-dormant alfalfa cultivars were associated with rapid sugar uptake and higher cell respiration rates when compared to cells derived from dormant alfalfa cultivars.  相似文献

The trait for somatic embryogenesis is being introduced sexually into alfalfa (Medicago sativa) breeding populations to facilitate genetic transformation of this crop. Cocultivation experiments were conducted with an agronomically-improved embryogenic clone from one such population as well as with two other embryogenic clones, one of which was the source of the embryogenic trait in the breeding populations. Transgenic plants were produced from the agronomically-improved clone whereas none were produced from the other two clones. Among the 16 transgenic plants analyzed there was a range in both copy number and number of integration sites for the NPT-II gene; those plants regenerated after a prolonged selection phase in vitro generally had the highest numbers in both respects. There was no evidence of sectoral chimerism of the transgene in a subsample of transgenic plants analyzed by PCR.  相似文献

The heteroplasmy of chloroplast DNA (cpDNA) observed in Medicago sativa L., which involves the presence (type B) or absence (type A) of an Xba I restriction site, was examined using closed fragments covering the variable XbaI site from type-A and type-B cpDNA. The 6.2-kb PstI fragment of DNA from type-A cpDNA (–XbaI) and from type-B cpDNA (+XbaI) was cloned into pUC19 plasmids. EcoRI fragments bearing the variable XbaI site from the type-A and type-B 6.2-kb PstI fragments were subcloned into pUC19. DNA sequences of both types of the 696-bp EcoRI fragments were determined and computer-assisted analysis of the sequence data carried out. Type-A cpDNA was found to differ from type-B cpDNA by 1 base, a G to T conversion, which results in a non-recognition site for XbaI in the type-A cpDNA. The sequence difference was in a non-coding region. Cloning and sequencing of the fragments verified the individual identity of the type-A and type-B cpDNA.  相似文献

Summary Tetraploid alfalfa (Medicago sativa L.) clones were derived from the same diploid genetic background by four different methods. A phenotypically superior clone was selected from each method and compared for herbage yield and fertility. The four methods and their best clones were: a) In vitro somatic chromosome doubling of one diploid hybrid (HG2-4x); b) selection within a two allele tetraploid synthetic population derived from HG2-4x (HAG); c) somaclonal variant selection from cell suspension culture of the diploid hybrid (NS1); and d) sexual polyploidization of a sibling hybrid (HXG). Clones HG2-4x, HAG, and NS1 were likely diallelic or monoallelic at all loci. Clone HXG was probably tetrallelic or triallelic at most loci. Experiments measured fertility, clonal herbage yield, and herbage yield of test cross progeny for each selected clone. Fertility rankings were HXG = HAG > NS1 > HG2-4x. Clonal herbage yield rankings were HXG = HAG > NS1 > HG2-4x. Test cross progeny herbage yield rankings varied depending on the tester, but, in general, HXG HAG NS1 HG2-4x. Overall the best clones from the sexual methods exceeded the best somaclonal variant which, in turn, was better than the chromosome doubled clone.  相似文献
