Three populations of the leafminer, Liriomyza trifolii (Burgess), were collected from commercial ornamental production greenhouses in the United States and tested for susceptibility to three commercial insecticides. A leaf dip bioassay of leaves containing young (1-2-d-old) larvae was used. Based on larval mortality and compared with a susceptible laboratory reference colony, the three strains varied in spectrum and level of resistance to the insecticides. CA-1, collected from Gerbera daisy, was moderately resistant to cyromazine (18.1-fold) and abamectin (22.0-fold), but highly resistant to spinosad (> 188-fold). CA-2, collected from chrysanthemums, was not resistant to abamectin, had a low level of resistance to cyromazine (8.2-fold), but was extremely resistant to spinosad (1,192-fold). GA-1, collected from chrysanthemums, had very low levels of resistance to cyromazine (5.4-fold) and spinosad (1.9-fold) but was moderately resistant to abamectin (30.6-fold). When reared in the absence of insecticide selection pressure, all three strains reverted to approximately the level of the reference strain. The CA-1 strain reverted in nine generations to cyromazine; however, the lowest levels of abamectin and spinosad resistance reverted to was 3.1-fold at F8 and 3.2 at the F10, respectively. The CA-2 strain reverted in five generations to both cyromazine and spinosad. GA-1 reverted in five generations to abamectin. Based on the results, resistance to these three insecticides was unstable. Additionally, there was no cross-resistance among these three insecticides.  相似文献

Resistance of codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae), to insecticides has become a major problem in many apple and pear production areas. Our aim was to determine the level of insecticide resistance in Spanish field populations. Seven field populations collected from apple, Malus domestica Borkhausen (Rosaceae), orchards, and three laboratory susceptible strains of codling moth were studied. Damage at harvest in all the conventional orchards from which codling moth populations were collected was higher than the economic threshold. The efficacy of eight insecticides, with five modes of action, was evaluated by topical application of the diagnostic concentrations on post‐diapausing larvae. The enzymatic activity of mixed‐function oxidases (MFOs), glutathione transferases (GSTs), and esterases (ESTs) was evaluated for each population. The susceptibility to insecticides and the biochemical activity of the three laboratory strains and one organic orchard population were not significantly different. Field populations were less susceptible to the tested insecticides than the susceptible strains, especially for azinphos‐methyl, diflubenzuron, fenoxycarb, and phosalone. The efficacy of all insecticides was significantly dependent on the activity of MFOs. Only the toxicity of the three insecticides most used in Spain when the populations were collected (azinphos‐methyl, fenoxycarb, and phosalone) was also dependent on the activity of ESTs and GSTs activity. We conclude that the control failures were because of the existence of populations resistant to the main insecticides used.  相似文献

The codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae), has developed resistance to various insecticides. Relative fitness of one susceptible strain (Sv) and two strains selected for resistance to diflubenzuron (Rt) and deltamethrin (Rv), respectively, was measured in the absence of insecticide selection pressure. Mating rate, fecundity, fertility, developmental time, fifth instar weight, and adult longevity were compared. Both resistant strains were less fecund and fertile, developed more slowly, weighed less, and had shorter life-spans than the susceptible strain. These results indicate that biological constraints are associated with insecticide resistance in the codling moth. We also found that fitness estimates of the Rv strain did not differ statistically from those of the Rt strain. Enhanced mixed-function oxidase and glutathione-S-transferase activities have been shown to be involved in insecticide resistance in both Rt and Rv strains. This suggests that the fitness cost described in both resistant strains was mainly associated to metabolic resistance. The impact of such deleterious pleiotropy of insecticide resistance in C. pomonella in terms of resistance management in the field is discussed.  相似文献

Plague is a rodent disease transmissible to humans by infected flea bites, and Madagascar is one of the countries with the highest plague incidence in the world. This study reports the susceptibility of the main plague vector Xenopsylla cheopis to 12 different insecticides belonging to 4 insecticide families (carbamates, organophosphates, pyrethroids and organochlorines). Eight populations from different geographical regions of Madagascar previously resistant to deltamethrin were tested with a World Health Organization standard bioassay. Insecticide susceptibility varied amongst populations, but all of them were resistant to six insecticides belonging to pyrethroid and carbamate insecticides (alphacypermethrin, lambdacyhalothrin, etofenprox, deltamethrin, bendiocarb and propoxur). Only one insecticide (dieldrin) was an efficient pulicide for all flea populations. Cross resistances were suspected. This study proposes at least three alternative insecticides (malathion, fenitrothion and cyfluthrin) to replace deltamethrin during plague epidemic responses, but the most efficient insecticide may be different for each population studied. We highlight the importance of continuous insecticide susceptibility surveillance in the areas of high plague risk in Madagascar.  相似文献

The life-stage variations in insecticide resistance of western flower thrips, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), to selective insecticides (acrinathrin, formetanate, and methiocarb) were studied using resistant laboratory strains. In each strain, the second-instar larva was less susceptible to the insecticides tested than the adults. The lower the resistance level of the adults, the higher the difference between larva and adult susceptibility: 32-fold to methiocarb, 15.4-fold to formetanate, and 180-fold to acrinathrin in the reference strain. In laboratory-selected resistant strains, these differences were much lower: 5.8-fold to methiocarb, 4.8-fold to formetanate, and 2.0-fold to acrinathrin. In selected strains, higher resistance levels for each insecticide were found, both for larvae and adults, compared with the reference strain. These results show that after insecticide resistance selection in adults, the resistance is carried over to the larvae, but at lower levels.  相似文献

Abstract: Four populations of codling moth Cydia pomonella L. were collected as overwintering larvae from apple orchards with different pesticide pressure (S. Michele, Roncafort, Revò and Vervò) in the Trento province (northern Italy). Mortality rate caused by a predetermined discriminating concentration of tebufenozide topically applied on overwintering larvae was evaluated. Neonate F1 progeny of the same populations were assayed for susceptibility to tebufenozide by feeding them on thinning apples treated with an appropriate discriminating dose of the insecticide. The activities of the main enzyme systems involved in the detoxification of insecticides were also evaluated in each population and related to their susceptibility to tebufenozide. The topical test detected a significant loss in susceptibility to tebufenozide in two populations, S. Michele and Roncafort, while all the overwintering larvae collected in the orchards of Revò and Vervò died when treated topically with the discriminating concentration. The apple‐dipping test performed on the neonate larvae showed a highly significant reduction in the susceptibility of the two populations of S. Michele and Roncafort. A less significant reduction in mortality rate was found in the Revò population; however, no statistical difference was found between the Vervò population and the susceptible reference. None of the four field populations significantly differed from the susceptible strain for Glutathione‐S‐transferase and esterase activity. A significantly higher frequency of individuals of the S. Michele and Roncafort populations exhibited a higher mixed function oxidase activity than the susceptible strain. The small resistance ratio values found for the two populations together with the low frequency of individuals exibiting enhanced enzymatic activity, reveals that the selection process was still at the early stage. Because of its efficiency in early detection of resistance to tebufenozide, topical application on diapausing larvae can thus be considered an appropriate, simple and robust tool for implementing resistance monitoring programmes for tebufenozide.  相似文献

Abstract:  Oriental fruit moth Grapholita molesta (Busck) (Lep., Tortricidae) has recently become a key pest of apples throughout the eastern USA. Pheromone-mediated mating disruption of Oriental fruit moth was successfully used in North Carolina apple orchards in the past few years. However, low levels of late-season fruit damage occurred in some orchards treated in late May with hand-applied pheromone dispensers because of inadequate dispenser longevity. To investigate alternative pheromone application schedules for extended mating disruption control, the following pheromone treatments were compared with conventional insecticides in Henderson County (NC) in 2002: late May application of hand-applied dispensers; late June application of hand-applied dispensers; late May application of hand-applied dispensers supplemented with a late August application of sprayable pheromone dispensers; late May application of hand-applied dispensers which have a longer activity period; and conventional insecticides as a control. All treatments were sprayed with an insecticide at petal fall in late April for thinning and for control of the first generation Oriental fruit moth adults. Pheromone trap catches were significantly reduced in all mating disruption blocks compared with conventional insecticide blocks. Among pheromone treatments, the highest trap captures were recorded in the delayed hand-applied dispenser treatment in June before treatment. However, the mean percentage fruit damage did not vary with timing of application of hand-applied dispensers and the type of pheromone dispenser used. Clearly, the combination of each mating disruption treatment with insecticide application against first generation Oriental fruit moth was as effective as the conventional insecticide treatment under moderate population pressure.  相似文献

Measurements were made of the relative susceptibility to a nuclear polyhedrosis virus of three populations of light-brown apple moth Epiphyas postvittana: a resistant laboratory strain (CAN), a susceptible laboratory strain (BAR), and a field population. CAN was found to be 50 times more resistant than BAR and 160 times more resistant than the field line. Experiments on hybrid crosses of resistant and susceptible strains showed that resistance is genetically determined. This serves as a warning of the possible selection of virus-resistant strains of insect pests, where viral insecticides are being used in the field.  相似文献

Insecticide resistance is frequently associated with fitness disadvantages in the absence of insecticides. However, intense past selection with insecticides may allow the evolution of fitness modifier alleles that mitigate the cost of insecticide resistance and their consequent fitness disadvantages. Populations of Sitophilus zeamais with different levels of susceptibility to insecticides show differences in the accumulation and mobilization of energy reserves. These differences may allow S. zeamais to better withstand toxic compounds without reducing the beetles' reproductive fitness. Enzymatic assays with carbohydrate- and lipid-metabolizing enzymes were, therefore, carried out to test this hypothesis. Activity levels of trehalase, glycogen phosphorylase, lipase, glycosidase and amylase were determined in two insecticide-resistant populations showing (resistant cost) or not showing (resistant no-cost) associated fitness cost, and in an insecticide-susceptible population. Respirometry bioassays were also carried out with these weevil populations. The resistant no-cost population showed significantly higher body mass and respiration rate than the other two populations, which were similar. No significant differences in glycogen phosphorylase and glycosidase were observed among the populations. Among the enzymes studied, trehalase and lipase showed higher activity in the resistant cost population. The results obtained in the assays with amylase also indicate significant differences in activity among the populations, but with higher activity in the resistant no-cost population. The inverse activity trends of lipases and amylases in both resistant populations, one showing fitness disadvantage without insecticide exposure and the other not showing it, may underlay the mitigation of insecticide resistance physiological costs observed in the resistant no-cost population. The higher amylase activity observed in the resistant no-cost population may favor energy storage, preventing potential trade-offs between insecticide resistance mechanisms and basic physiological processes in this population, unlike what seems to take place in the resistant cost population.  相似文献

San Jose scale, Diaspidiotus perniciosus (Comstock), is a serious pest in Chilean apple tree orchards, and a number of organophosphate insecticides were used to control them for decades. Recently, control failures with these insecticides were reported and linked to insecticide resistance development. In this study, 40 San Jose scale field populations were collected and their susceptibility to two commonly used organophosphate insecticides, that is chlorpyrifos and methidathion, was assessed. The obtained bioassay data suggest moderate levels of resistance to both insecticides when compared to a reference susceptible strain. The highest resistance ratio (RR) detected for chlorpyrifos and methidathion was 31‐fold and 11‐fold, respectively. The bioassay results suggest the occurrence of a significant cross‐resistance between both compounds. Biochemical measurements revealed a role for esterases in conferring resistance to organophosphates, but not modified acetylcholinesterase. The spatial spread and extend of insecticide resistance were also evaluated. Our result shows that no autocorrelation can be assumed, and then, insecticide resistance is caused by random factors.  相似文献

Chlorantraniliprole is the first commercial insecticide from a new class of chemistry, the anthranilic diamides. Chlorantraniliprole provides an effective alternative insecticide for control of Plutella xylostella (L.) populations resistant to other insecticides. Baseline susceptibility to chlorantraniliprole for P. xylostella was surveyed previously from 16 geographical populations sampled from China during 2008-2009, and the median lethal concentrations (LC50s) varied among populations from 1.8- to 8.9-fold higher than the LC50 of a susceptible strain (Roth). In the present work, 20 field populations of P. xylostella sampled in 2010-2011 from China were tested with laboratory bioassays to determine if resistance to chlorantraniliprole had evolved in the field. The LC50s of the 14 populations from northern China ranged from 1.7- to 5.4-fold compared with the LC50 of Roth, which indicates these populations remain reasonably susceptible to chlorantraniliprole. However, the LC50s of the six populations from southern China (Guangdong Province) were 2.6-, 12-, 18-, 81-, 140-, and 2,000-fold higher than the LC50 of Roth. The results showed that high levels of resistance to chlorantraniliprole had evolved in field populations from Guangdong Province of southern China. Intensive use and misuse of chlorantraniliprole may be responsible for the rapid evolution of high-level resistance in P. xylostella in this region. The implementation of resistance monitoring plans and resistance management strategies is urgently needed in China to preserve susceptibility to chlorantraniliprole in P. xylostella.  相似文献

Susceptibility baselines and diagnostic doses of the technical grade insecticides deltamethrin, permethrin, fenitrothion, and propoxur were established based on Aedes aegypti (L.), Bora (French Polynesia), a reference susceptible strain. Field-collected Aedes mosquitoes from each part of Thailand were subjected to bioassay for their susceptibility to the diagnostic doses of each insecticide. Almost all Ae. aegypti collected were incipient resistant or resistant to deltamethrin and permethrin, except those from some areas of Songkhla (southern) and Phan district of Chiang Rai (northern) province. Susceptibility to fenitrothion was found in mosquitoes from Bangkok (central), Chonburi (eastern), Chiang Rai, Kanchanaburi (western), and Songkhla, whereas they were resistant in almost all areas of Nakhon Sawan (north central) and Nakhon Ratchasima (northeastern) provinces. Most of Ae. aegypti were susceptible to propoxur except those from Mae Wong, Nakhon Sawan province. Various levels of insecticide resistance and susceptibility in adjacent areas revealed a focal susceptible/resistance profile in the country. It could be noted that almost all of Ae. albopictus were susceptible to the insecticides tested at the same diagnostic doses. In conclusion, resistance to pyrethroids (permethrin and deltamethrin) has developed in Ae. aegypti in most of the collected areas, suggesting that an alternative choice of insecticide or other control measures should be applied.  相似文献

Abstract.  A common assumption in models of insecticide resistance evolution is the association between resistance and fitness costs in the absence of insecticides. There is empirical evidence of such associations, but their physiological basis (and mitigation) is little investigated. Pyrethroid-resistant populations of the maize weevil Sitophilus zeamais (Coleoptera: Curculionidae) offer this opportunity. Pyrethroid resistance in this species was initially observed in five Brazilian states by 1995, but the phenomenon apparently decreased and did not spread to other regions, probably due to the occurrence of a fitness disadvantage in resistant individuals in the absence of insecticides. The present investigation aims to verify whether differences in respiration rate and fat body morphology are related to differences in rate of development in Brazilian populations of S. zeamais resistant to insecticides, and thereby provide evidence for the existence (or not) of a physiological fitness cost acting against insecticide resistance in maize weevils. This may occur due to a possible energy trade-off between insecticide resistance and other physiological processes associated with development and reproduction. To achieve this, studies of the rate of development, respiration and fat body cytomorphology are carried out in one insecticide-susceptible (from Sete Lagoas) and two resistant populations (from Jacarezinho and Juiz de Fora) of S. zeamais . The resistant population from Jacarezinho shows that higher body mass is associated with higher energy reserves (larger trophocyte area) for development and reproduction, as well as for insecticide resistance. However, the resistant population from Juiz de Fora does not appear to have large enough energy allocation for insecticide-resistance expression and development and/or reproductive performance, suggesting a trade-off between resistance and other life history traits.  相似文献

The codling moth, Cydia pomonella, is renowned for developing resistance to insecticides and causing significant economic damage to pome fruits worldwide. In spite of its economic importance, little is known about the patterns of movement of this pest and the effects of insecticide treatment on the population genetic structure. Here, we investigated the genetic structure of the pest in 27 orchards from France, Italy, Armenia and Chile at seven microsatellite loci and two resistance markers [biochemical activity of cytochrome P450 oxidases and proportion of knockdown resistance (kdr) alleles in the sodium channel gene]. According to the microsatellite loci, we detected isolation by distance at the supranational scale but found no evidence of geographical structure among the 24 French orchards, which were mainly structured by the intensity of the insecticide treatments. Similarly, the highest levels of metabolic resistance associated with activity of the cytochrome P450 oxidases were detected in the most treated orchards. The kdr alleles were observed in southern France and Armenia where the pyrethroid insecticides were or have been intensively sprayed. The intensity of the insecticide treatments marginally affected the allelic richness in each orchard, but not the level of inbreeding. These results suggest important and high-distance gene flow among the codling moth populations, which were mainly structured according to the history of insecticide applications. Differences in mutation-migration-drift equilibrium among treated and untreated orchards also suggest that insecticide applications are the main force regulating the local dynamics of codling moth populations.  相似文献

A diet-incorporation larval bioassay was developed to measure the response of codling moth, Cydia pomonella (L.), to the benzoylhydrazine insecticides tebufenozide and methoxyfenozide. The bioassay tested neonates and third, fourth, and fifth instars from a laboratory colony and neonates and fourth instars from a pooled population collected from five certified-organic apple orchards. Bioassays were scored after 6 and 14 d. No differences between the laboratory and field population were found for either insecticide. Significant differences were found in the response of third and fifth instars between the 6 and 14 d bioassays, primarily due to a high proportion of moribund larvae in the shorter assay. Larval age had a significant effect in bioassays and was more pronounced in 6- versus 14-d tests. Fifth instars were significantly less susceptible to both insecticides than other stages, while responses of third and fourth instars were similar. The response of neonates was significantly different from third and fourth instars to tebufenozide but not with methoxyfenozide in the 14-d test. Field bioassays excluded the use of fifth instars and were scored after 14 d. LC50s estimated for 18 field-collected populations varied five- and ninefold for tebufenozide and methoxyfenozide, respectively. The responses of all but six field-collected populations were significantly different from the laboratory strain. Five of these six populations were collected from orchards with no history of organophosphate insecticide use. The LC50 for methoxyfenozide of one field-collected population reared in the laboratory for three generations declined fourfold, but was still significantly different from the laboratory population. These data suggest that transforming current codling moth management programs in Washington from a reliance on organophosphate insecticides to benzoylhydrazines may be difficult.  相似文献

The efficacy of mating disruption by using Isomate-M 100 pheromone dispensers and two formulations of microencapsulated sprayable pheromone for management of oriental fruit moth, Grapholita molesta (Busck), was compared with conventional insecticides in large plot studies in Henderson County, North Carolina, in 2000 and 2001. In addition, experiments were conducted in small and large plots to test the response of oriental fruit moth males to different application rates of sprayable pheromone. Pheromone trap catches were significantly reduced in mating disruption blocks compared with conventional and abandoned orchards. Pheromone traps placed in the upper canopy captured significantly more moths than traps placed in the lower canopy across all treatments, and lures loaded with 100 microg of pheromone caught more moths than traps with 300 microg, but the difference between doses was statistically significant at only one location in 2001. Isomate-M 100 provided excellent trap shutdown and was significantly more effective than sprayable pheromone formulations. Fruit damage by oriental fruit moth larvae was very low (< or = 1%) in mating disruption blocks and was generally lower than in conventional and nonmanaged blocks. Based on male moth response to pheromone traps in small plots, there was little difference among doses of sprayable pheromone, ranging from 12.4 to 49.1 g (AI)/ha, but efficacy declined at 2.4 g (AI)/ha. With the exception of one orchard, there was no significant difference between 12.4 and 37.1 g (AI)/ha under low and high oriental fruit moth population pressure in large plot studies. Mating disruption proved to be an alternative to organophosphate insecticides for managing oriental fruit moth populations in North Carolina apple orchards.  相似文献

The horn fly, Haematobia irritans (L.) (Diptera: Muscidae), was introduced to Chile in the beginning of the 1990s. Since its introduction, farmers have controlled this pest almost exclusively with insecticides. To understand the consequences of different control strategies on the development of insecticide resistance and their persistence, a field survey was conducted at eight farms in the south of Chile to characterize insecticide resistance in field populations and resistance mechanisms. Horn fly samples were assayed to determine levels of resistance to pyrethroids and diazinon, genotyped for kdr and HialphaE7 mutations, and tested for general esterase activity. All field populations, including ones that were not treated with insecticides for the past 5 yr, showed high levels of cypermethrin resistance and high frequencies of the kdr mutation. None of the fly populations demonstrated resistance to diazinon and the HialphaE7 mutation was not detected in any of the fly samples. Esterase activities in all populations were comparable to those found in the susceptible reference strain. The findings of high frequencies of homozygous resistant and heterozygous individuals both in insecticide treated horn fly populations and in the untreated fly populations suggests complex interactions among field populations of the horn fly in Chile.  相似文献

The codling moth, Cydia pomonella (L.), and oriental fruit moth, Grapholita molesta (Busck), are two key pests of apple (Malus domestica Borkh.) in North Carolina. Growers extensively relied on organophosphate insecticides, primarily azinphosmethyl, for > 40 yr to manage these pests. Because of organophosphate resistance development and regulatory actions, growers are transitioning to management programs that use new, reduced-risk, and OP-replacement insecticides. This study evaluated the toxicity of a diversity of replacement insecticides to eggs, larvae, and adults, as well as an assessment of their residual activity, to codling moth and oriental fruit moth. Laboratory-susceptible strains of both species were used for all bioassays. Fresh field-harvested apples were used as a media for assessing the ovicidal activity of insecticides. For larval studies, insecticides were topically applied to the surface of lima bean-based diet, onto which neonates were placed. Toxicity was based on two measures of mortality; 5-d mortality and development to adult stage. Ovicidal bioassays showed that oriental fruit moth eggs were generally more tolerant than codling moth eggs to insecticides, with novaluron, acetamiprid, and azinphoshmethyl having the highest levels of toxicity to eggs of both species. In contrast, codling moth larvae generally were more tolerant than oriental fruit moth to most insecticides. Methoxyfenozide and pyriproxyfen were the only insecticides with lower LC50 values against codling moth than oriental fruit moth neonates. Moreover, a number of insecticides, particularly the IGRs methoxyfenozide and novaluron, the anthranilic diamide chlorantriliprole, and the spinosyn spinetoram, provided equal or longer residual activity against codling moth compared with azinphosmethyl in field studies. Results are discussed in relation to their use in devising field use patterns of insecticides and for insecticide resistance monitoring programs.  相似文献

利用浸叶法对南京郊区小菜蛾Plutella xylostella的抗药性进行了监测,发现其对拟除虫菊酯类药剂的抗性较高,而对氟虫腈、辛硫磷、毒死蜱、多杀菌素和虫酰肼依然处于敏感阶段。利用抗性小菜蛾测试发现,氧化胡椒基丁醚(增效醚,简称PBO)对拟除虫菊酯具有显著的增效作用。室内敏感性恢复实验表明,在不接触药剂的条件下,小菜蛾对拟除虫菊酯的抗性迅速下降,繁殖10代以后,抗性维持在低抗水平。因此, 小菜蛾的抗药性治理,应充分利用不同药剂的轮换使用,避免单一使用某一品种,以延缓抗性的发展,同时可以利用PBO增强拟除虫菊酯的防治效果,保证这类药剂在小菜蛾防治中的作用。  相似文献
