Fucosyllactoses, including 2′-fucosyllactose (2′-FL) and 3-fucosyllactose (3-FL), are important oligosaccharides in human milk that are commonly used as nutritional additives in infant formula due to their biological functions, such as the promotion of bifidobacteria growth, inhibition of pathogen infection, and improvement of immune response. In this study, we developed a synthetic biology approach to promote the efficient biosynthesis of 2′-FL and 3-FL in engineered Escherichia coli. To boost the production of 2′-FL and 3-FL, multiple modular optimization strategies were applied in a plug-and-play manner. First, comparisons of various exogenous α1,2-fucosyltransferase and α1,3-fucosyltransferase candidates, as well as a series of E. coli host strains, demonstrated that futC and futA from Helicobacter pylori using BL21(DE3) as the host strain yielded the highest titers of 2′-FL and 3-FL. Subsequently, both the availability of the lactose acceptor substrate and the intracellular pool of the GDP-L-fucose donor substrate were optimized by inactivating competitive (or repressive) pathways and strengthening acceptor (or donor) availability to achieve overproduction. Moreover, the intracellular redox regeneration pathways were engineered to further enhance the production of 2′-FL and 3-FL. Finally, various culture conditions were optimized to achieve the best performance of 2′-FL and 3-FL biosynthesizing strains. The final concentrations of 2′-FL and 3-FL were 9.12 and 12.43 g/L, respectively. This work provides a platform that enables modular construction, optimization and characterization to facilitate the development of FL-producing cell factories.  相似文献

Aspartate 170 of the D1 polypeptide provides part of the high-affinity binding site for the first Mn(II) ion that is photooxidized during the light-driven assembly of the (Mn)(4) cluster in photosystem II [Campbell, K. A., Force, D. A., Nixon, P. J., Dole, F., Diner, B. A., and Britt, R. D. (2000) J. Am. Chem. Soc. 122, 3754-3761]. However, despite a wealth of data on D1-Asp170 mutants accumulated over the past decade, there is no consensus about whether this residue ligates the assembled (Mn)(4) cluster. To address this issue, we have conducted an EPR and ESEEM (electron spin-echo envelope modulation) study of D1-D170H PSII particles purified from the cyanobacterium Synechocystis sp. PCC 6803. The line shapes of the S(1) and S(2) state multiline EPR signals of D1-D170H PSII particles are unchanged from those of wild-type PSII particles, and the signal amplitudes correlate approximately with the lower O(2) evolving activity of the mutant PSII particles (40-60% compared to that of the wild type). These data provide further evidence that the assembled (Mn)(4) clusters in D1-D170H cells function normally, even though the assembly of the (Mn)(4) cluster is inefficient in this mutant. In the two-pulse frequency domain ESEEM spectrum of the 9.2 GHz S(2) state multiline EPR signal of D1-D170H PSII particles, the histidyl nitrogen modulation observed at 4-5 MHz is unchanged from that of wild-type PSII particles and no significant new modulation is observed. Three scenarios are presented to explain this result. (1) D1-Asp170 ligates the assembled (Mn)(4) cluster, but the hyperfine couplings to the ligating histidyl nitrogen of D1-His170 are too large or anisotropic to be detected by ESEEM analyses conducted at 9.2 GHz. (2) D1-Asp170 ligates the assembled (Mn)(4) cluster, but D1-His170 does not. (3) D1-Asp170 does not ligate the assembled (Mn)(4) cluster.  相似文献

A model of leaf, photosynthesis has been developed for C3–C4 intermediate species found in the generaPanicum, Moricandia, Parthenium andMollugo where no functional C4 pathway has been identified. Model assumptions are a functional C3 cycle in both mesophyll and bundle-sheath cells and that glycine formed in the mesophyll, as a consequence of the oxygenase activity of ribulose-1,5-bisphosphate carboxylase-oxygenase (Rubisco, EC 4.1.1.39), diffuses to the bundle sheath, where most of the photorespiratory CO2 is released. The model describes the observed gas-exchange characteristics of these C3–C4 intermediates, such as low CO2-compensation points () at an O2 pressure of 200 mbar, a curvilinear response of to changing O2 pressures, and typical responses of CO2-assimilation rate to intercellular CO2 pressure. The model predicts that bundle-sheath CO2 concentration is highest at low mesophyll CO2 pressures and decreases as mesophyll CO2 pressure increases. A partitioning of 5–15% of the total leaf Rubisco into the bundle-sheath cells and a bundlesheath conductance similar to that proposed for C4 species best mimics the gas-exchange results. The effect of increased salinity on photosynthesis was studied in leaves of Plantago maritima L. that developed while plants were at low and high NaCl levels. In leaves that developed while plants were grown at 50 mol·m-3, exposure to 200 and 350 mol·m-3 NaCl resulted in reductions in net CO2 assimilation and stomatal conductance. The decline in CO2 assimilation in plants at 200 and 350 mol·m-3 NaCl occurred almost exclusively at high intercellular CO2 concentrations. The initial slope of the CO2 assimilation-intercellular CO2 (A-C i) curve, determined after salinity was increased, was identical or very similar to that measured initially. In contrast to the reductions observed in CO2 assimilation, there were no significant differences in O2 evolution rates measured at 5% CO2 among leaves from plants exposed to higher salinity and plants remaining at low salinity.Leaves that developed while plants were at increased salinity levels also had significantly lower net CO2 assimilation rates than plants remaining at 50 mol·m-3 NaCl. The lower CO2 assimilation rates in plants grown at 200 and 350 mol·m-3 NaCl were a result of reduced stomatal conductance and low intercellular CO2 concentration. There were no significant differences among treatments for O2 evolution rates measured at high CO2 levels. The increased stomatal limitation of photosynthesis was confirmed by measurements of the 13C/12C composition of leaf tissue. Mass-spectrometric disequilibrium analysis was applied to investigate CO2 uptake and HCO3 transport in cells and chloroplasts of the microalgae Dunaliella tertiolecta and Chlamydomonas reinhardtii, which were grown in air enriched with 5% (v/v) CO2 (high-Ci cells) or in ambient air (low-Ci cells). High- and low-Ci cells of both species had the capacity to transport CO2 and HCO3, with maximum rates being largely unaffected by the growth conditions. In high- and low-Ci cells of D. tertiolecta, HCO3 was the dominant inorganic C species taken up, whereas HCO3 and CO2 were used at similar rates by C. reinhardtii. The apparent affinities of HCO3 transport and CO2 uptake increased 3- to 9-fold in both species upon acclimation to air. Photosynthetically active chloroplasts isolated from both species were able to transport CO2 and HCO3. For chloroplasts from C. reinhardtii, the concentrations of HCO3 and CO2 required for half-maximal activity declined from 446 to 33 μm and 6.8 to 0.6 μm, respectively, after acclimation of the parent cells to air; the corresponding values for chloroplasts from D. tertiolecta decreased from 203 to 58 μm and 5.8 to 0.5 μm, respectively. These results indicate the presence of inducible high-affinity HCO3 and CO2 transporters at the chloroplast envelope membrane.  相似文献

The antenna proteins in photosystem II (PSII) not only promote energy transfer to the photosynthetic reaction center (RC) but provide also an efficient cation sink to re-reduce chlorophyll a if the electron transfer (ET) from the Mn-cluster is inhibited. Using the newest PSII dimer crystal structure (3.0 Å resolution), in which 11 β-carotene molecules (Car) and 14 lipids are visible in the PSII monomer, we calculated the redox potentials (Em) of one-electron oxidation for all Car (Em(Car)) by solving the Poisson-Boltzmann equation. In each PSII monomer, the D1 protein harbors a previously unlocated Car (CarD1) in van der Waals contact with the chlorin ring of ChlZ(D1). Each CarD1 in the PSII dimer complex is located in the interface between the D1 and CP47 subunits, together with another four Car of the other PSII monomer and several lipid molecules. The proximity of Car bridging between CarD1 and plastoquinone/QA may imply a direct charge recombination of Car+QA. The calculated Em(CarD1) and Em(ChlZ(D1)) are, respectively, 83 and 126 mV higher than Em(CarD2) and Em(ChlZ(D2)), which could explain why CarD2+ and ChlZ(D2)+ are observed rather than the corresponding CarD1+ and ChlZ(D1)+.  相似文献

D76N is the first natural variant of human β-2 microglobulin (β2m) so far identified. Contrary to the wt protein, this mutant readily forms amyloid fibres in physiological conditions, leading to a systemic and severe amyloidosis. Although the Asp76Asn mutant has been extensively characterized, the molecular bases of its instability and aggregation propensity remain elusive. In this work all Asp residues of human β2m were individually substituted to Asn; D-to-N mutants (D34N, D38N, D53N, D59N, D96N and D98N) were characterised in terms of thermodynamic stability and aggregation propensity. Moreover, crystal structures of the D38N, D53N, D59N and D98N variants were solved at high-resolution (1.24–1.70 Å). Despite showing some significant variations in their thermal stabilities, none showed the dramatic drop in melting temperature (relative to the wt protein) as observed for the pathogenic mutant. Consistently, none of the variants here described displayed any increase in aggregation propensity under the experimental conditions tested. The crystal structures confirmed that D-to-N mutations are generally well tolerated, and lead only to minor reorganization of the side chains in close proximity of the mutated residue. D38N is the only exception, where backbone readjustments and a redistribution of the surface electrostatic charges are observed. Overall, our results suggest that neither removing negative charges at sites 34, 38, 53, 59, 96 and 98, nor the difference in β2m pI, are the cause of the aggressive phenotype observed in D76N. We propose that the dramatic effects of the D76N natural mutation must be linked to effects related to the crucial location of this residue within the β2m fold.  相似文献

The putative role of phloem amino acids as negative feedback signals for root NO3- uptake was investigated in Ricinus communis L. The NO3--grown plants were subjected to N-deficiency due either to complete N-deprivation, or to localized N-deprivation on one side of a split-root system. In comparison with controls, complete N-deprivation resulted in a transient increase in 15NO3- influx, and in profound changes in downward phloem transport of amino acids. Total amino acid concentration in the phloem sap decreased by 40%, but responses markedly differed between the individual amino acids. Concentrations of Gln and Ser were rapidly lowered by 50%, while those of Val, Phe, Leu, and Ile displayed a marked increase. Localized N-deprivation on one side of the split root system also resulted in the up-regulation of 15NO3- influx in the roots still supplied with NO3-. However, the amino acid composition of the phloem sap directed to these roots was not modified by the treatment, and remained similar to that in N-sufficient control plants. Only amino acid transport to the N-deprived roots was affected as observed in response to complete N-deprivation. The results from split-root plants indicate that the response of root NO3- influx to N-deficiency is controlled by shoot-borne regulatory signals, and provide a case study where these signals are not related to a qualitative change or a significant decrease in downward phloem transport of amino acids.  相似文献

In nature, an oxo‐bridged Mn4CaO5 cluster embedded in photosystem II (PSII), a membrane‐bound multi‐subunit pigment protein complex, catalyzes the water oxidation reaction that is driven by light‐induced charge separations in the reaction center of PSII. The Mn4CaO5 cluster accumulates four oxidizing equivalents to enable the four‐electron four‐proton catalysis of two water molecules to one dioxygen molecule and cycles through five intermediate S‐states, S0 – S4 in the Kok cycle. One important question related to the catalytic mechanism of the oxygen‐evolving complex (OEC) that remains is, whether structural isomers are present in some of the intermediate S‐states and if such equilibria are essential for the mechanism of the O‐O bond formation. Here we compare results from electron paramagnetic resonance (EPR) and X‐ray absorption spectroscopy (XAS) obtained at cryogenic temperatures for the S2 state of PSII with structural data collected of the S1, S2 and S3 states by serial crystallography at neutral pH (~6.5) using an X‐ray free electron laser at room temperature. While the cryogenic data show the presence of at least two structural forms of the S2 state, the room temperature crystallography data can be well‐described by just one S2 structure. We discuss the deviating results and outline experimental strategies for clarifying this mechanistically important question.  相似文献

CO2 catalyses the isomerization of the biological toxin ONOO- to NO3- via an intermediate, presumably ONOOCO2-, which has an absorption maximum near 650 nm. The reflection spectrum of solid NMe4+ ONOO- exposed to CO2 shows a similar band near 650 nm; this absorption decays over minutes. Stopped-flow experiments in which CO2 solutions were mixed with alkaline ONOO- solutions indicate the formation of at least one intermediate. The initial absorption at 302 nm is less than that of ONOO-, which indicates that reactions take place within the mixing time, and this absorption is dependent (but not linearly) on the ONOO- and CO2 concentrations. We found that reaction of peroxynitrite with carbon dioxide forms some trioxocarbonate(*1-) (CO3*-) and nitrogen dioxide (NO2*) radicals via homolysis of the O-O bond in ONOOCO2-. We determined the extent of radical formation by mixing peroxynitrite, carbon dioxide and nitrogen monoxide. The later reacts with CO3*- and NO2* radicals to form, effectively, three NO2- per homolysis; ONOOCO2- that does not undergo homolysis yields NO3- and CO2. Based on the NO3- and NO2- analyses, the extent of conversion to NO3- is 96 +/- 1% and that of homolysis is 3 +/- 1%, respectively, significantly less than that reported in the literature.  相似文献

A few species of Cymbopogon and Vetiveria are potentially important tropical grasses producing essential oils. In the present study, we report on the leaf anatomy and photosynthetic carbon assimilation in five species of Cymbopogon and Vetiveria zizanioides. Kranz-type leaf anatomy with a centrifugal distribution of chloroplasts and exclusive localization of starch in the bundle sheath cells were common among the test plants. Besides the Kranz leaf anatomy, these grasses displayed other typical C4 characteristics including a low (0–5 µl/l) CO2 compensation point, lack of light saturation of CO2 uptake at high photon flux densities, high temperature (35°C) optimum of net photosynthesis, high rates of net photosynthesis (55–67 mg CO2 dm-2 leaf area h-1), little or no response of net photosynthesis to atmospheric levels of O2 and high leaf 13C/12C ratios. The biochemical studies with 14CO2 indicated that the leaves of the above plant species synthesize predominantly malate during short term (5 s) photosynthesis. In pulse-chase experiments it was shown that the synthesis of 3-phosphoglycerate proceeds at the expense of malate, the major first formed product of photosynthesis in these plant species.  相似文献

The assimilation of 14CO2 into the C4 acids malate and aspartate by leaves of C3, C4 and C3–C4 intermediate Flaveria species was investigated near the CO2 compensation concentration * in order to determine the potential role of phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) in reducing photorespiration in the intermediates. Relative to air concentrations of CO2, the proportion of CO2 fixed by PEP carboxylase at * increased in all six C3–C4 intermediate species examined. However, F. floridana J.R. Johnston and F. ramosissima Klatt were shown to be markedly less responsive to reduced external CO2, with only about a 1.6-fold enhancement of CO2 assimilation by PEP carboxylase, as compared to a 3.0- to 3.7-fold increase for the other C3–C4 species examined, namely, F. linearis Lag., F. anomala B.L. Robinson, F. chloraefolia A. Gray and F. pubescens Rydb. The C3 species F. pringlei Gandoger and F. cronquistii A.M. Powell exhibited a 1.5- and 2.9-fold increase in labeled malate and aspartate, respectively, at *. Assimilation of CO2 by PEP carboxylase in the C4 species F. trinervia (Spreng.) C. Mohr, F. australasica Hook., and the C4-like species F. brownii A.M. Powell was relatively insensitive to subatmospheric levels of CO2. The interspecific variation among the intermediate Flaverias may signify that F. floridana and F. ramosissima possess a more C4-like compartmentation of PEP carboxylase and ribulose-1,5-bisphosphate carboxylase/oxygenase (EC 4.1.1.39) between the mesophyll and bundle-sheath cells. Chasing recently labeled malate and aspartate with 12CO2 for 5 min at * resulted in an apparent turnover of 25% and 30% of the radiocarbon in these C4 acids for F. ramosissima and F. floridana, respectively. No substantial turnover was detected for F. linearis, F. anomala, F. chloraefolia or F. pubescens. With the exception of F. floridana and F. ramosissima, it is unlikely that enhanced CO2 fixation by PEP carboxylase at the CO2 compensation concentration is a major mechanism for reducing photorespiration in the intermediate Flaveria species. Moreover, these findings support previous related 14CO2-labeling studies at air-levels of CO2 which indicated that F. floridana and F. ramosissima were more C4-like intermediate species. This is further substantiated by the demonstration that F. floridana PEP carboxylase, like the enzyme in C4 plants, undergoes a substantial activation (2.2-fold) upon illuminating dark-adapted green leaves. Purpose

Representative, consistent and up-to-date life cycle inventories (LCI) of electricity supply are key elements of ecoinvent as an LCI background database since these are often among the determining factors with regard to life cycle assessment (LCA) results. ecoinvent version 3 (ev3) offers new LCI data of power supply (electricity markets) in 71 geographies. This article gives an overview of these electricity markets and discusses new ecoinvent features in the context of power supply.

Methods

The annual geography- and technology-specific electricity production for the year 2008 specifies the technology shares on the high-, medium- and low-voltage level electricity markets. Data are based on IEA statistics. Different voltage levels are linked by transformation activities. Region-specific electricity losses due to power transmission and voltage transformation are considered in the market and transformation activities. The majority of the 71 power markets are defined by national boundaries. The attributional ecoinvent system model in ev3 with linking to average current suppliers results in electricity markets supplied by all geography-specific power generation technologies and electricity imports, while the consequential system model generates markets only linked to unconstrained suppliers.

Results and discussion

The availability of LCI data for 71 electricity markets in ev3 covering 50 countries reduces the “Rest-of-the-World” electricity supply not covered by country- or region-specific inventories to 17 % for the year 2008. Specific power supply activities for all countries contributing more than 1 % to global electricity production are available. The electricity markets show large variations concerning contributions from specific technologies and energy carriers. Imports can substantially change the national/regional power mix, especially in small markets. Large differences can also be observed between the electricity markets in the attributional and the consequential database calculation. Region-specific total power losses between production on the high voltage level and consumer on the low voltage level are on the order of 2.5–23 %.

Conclusions

Electricity supply mixes (electricity markets) in the ecoinvent database have been updated and substantially extended for v3. Inventories for electricity supply in all globally important economies are available with geography-specific technology and market datasets which will contribute to increasing quality and reducing uncertainties in LCA studies worldwide and to allow more accurate estimation of environmental burdens from global production chains. Future work should focus on improving the details of country-specific data, implementation of more countries into the database, splitting of large countries into smaller regions and on developing a more sophisticated approach specifying country-specific electricity mixes in consequential system models.

The efficiency of light harvesting in higher plant photosynthesis is regulated in response to external environmental conditions. Under conditions of excess light, the normally highly efficient light-harvesting system of photosystem II is switched into a state in which unwanted, potentially harmful, energy is dissipated as heat. This process, known as nonphotochemical quenching, occurs by the creation of energy quenchers following conformational change in the light-harvesting complexes, which is initiated by the build up of the thylakoid pH gradient and controlled by the xanthophyll cycle. In the present study, the evidence to support the notion that this regulatory mechanism is dependent upon the organization of the different antenna subunits in the stacked grana membranes is reviewed. We postulate that nonphotochemical quenching occurs within a structural locus comprising the PsbS subunit and components of the light-harvesting antenna, CP26, CP24, CP29 and LHCIIb (the major trimeric light-harvesting complex), formed in response to protonation and controlled by the xanthophyll cycle carotenoids.  相似文献

Using a cuvette for simultaneous measurement of net photosynthesis in above ground plant organs and root respiration we investigated the effect of reduced leaf glucokinase activity on plant carbon balance. The gin2–1 mutant of Arabidopsis thaliana is characterized by a 50% reduction of glucokinase activity in the shoot, while activity in roots is about fivefold higher and similar to wild type plants. High levels of sucrose accumulating in leaves during the light period correlated with elevated root respiration in gin2–1. Despite substantial respiratory losses in roots, growth retardation was moderate, probably because photosynthetic carbon fixation was simultaneously elevated in gin2–1. Our data indicate that futile cycling of sucrose in shoots exerts a reduction on net CO2 gain, but this is over-compensated by the prevention of exaggerated root respiration resulting from high sucrose concentration in leaf tissue.  相似文献

The blue light dependent utilization of nitrate by green algae under common air and high irradiances, besides its assimilatory nature, is associated with the release of NO2 and NH4 + to the culture medium. If the CO2 content of the sparging air was increased up to 2%, previously excreted NO2 and NH4 + were rapidly assimilated. When under air and high irradiances the cell density in the culture reached values corresponding to 25 g Ch 1.ml-1, no further growth was observed and the highest values of NO3 consumption and NO2 and NH4 + release were attained. Besides low CO2 tensions, increasing NO3 concentrations in the medium stimulated the release of NO3 and NH4 +. Under CO2-free air the consumption of NO3 and the release of NO2 and NH4 + on a total N bases were almost stoichiometric and their rates saturated at much lower irradiances than under air. Under CO2-free air high rates of NO2 release were only observed under the blue radiations that were effectively absorbed by photosynthetically active pigments, i.e. 460 nm, but not under 404 and 630 nm radiations. However, the simultaneous illumination of the cells with 404 and 630 nm monochromatic light showed a remarkable synergistic effect on NO2 release.The results are discussed in terms of the close relationship between C and N metabolism, the photosynthetic reducing power required to convert NO inf3 sup± -N into R – NH2-N and the blue light activation of nitrate reductase.  相似文献

The availability of a complete genome database for the cyanobacterium Synechocystissp. PCC6803 (glucose-tolerant strain) has raised expectations that this organism would become a reference strain for work aimed at understanding the CO2-concentrating mechanism (CCM) in cyanobacteria. However, the amount of physiological data available has been relatively limited. In this report we provide data on the relative contributions of net HCO3 uptake and CO2 uptake under steady state photosynthetic conditions. Cells were compared after growth at high CO2 (2% v/v in air) or limiting CO2 conditions (20 ppm CO2). Synechocystishas a very high dependence on net HCO3 uptake at low to medium concentrations of inorganic carbon (Ci). At high Ci concentrations net CO2 uptake became more important but did not contribute more than 40% to the rate of photosynthetic O2 evolution. The data also confirm that high Ci cells of Synechocystissp. PCC6803 possess a strong capacity for net HCO3 uptake under steady state photosynthetic conditions. Time course experiments show that induction of maximal Ci uptake capacity on a shift from high CO2 to low CO2 conditions was near completion by four hours. By contrast, relaxation of the induced state on return of cells to high CO2, takes in excess of 230 h. Experiments were conducted to determine if Synechocystissp. PCC6803 is able to exhibit a `fast induction' response under severe Ci limitation and whether glucose was capable of causing a rapid inactivation in Ci uptake capacity. In Prophenoloxidase (ProPO) cascade, two targets namely serine protease and α-2-macroglobulin are key regulators involved in the defense system of crustaceans. In biological systems, routine role of cell systems requires the understanding in protein–protein interactions through experimental and theoretical concepts, which might yield useful insights into the cellular responses. Response of cells to regulating the immune system is governed by the interactions-involved biomolecular simulations. Unfortunately, studies on the inhibitors (SP and α-2M) that negatively regulate the proPO system or melanization in penaeid shrimp are not yet available. In order to understand how these interactions change the proPO mechanism in Indian white shrimp Fenneropenaeus indicus was determined. In F. indicus, innate immune system is in a sensitive balance of intricate interactions; elucidating these interactions by the integration of in silico and in vitro has great potential. We have determined the expression of both the SP and α-2M enzymes in regulatory mechanism, which are analyzed through qRT-PCR, protein–protein docking, and simulation studies. From this work, we propose a novel approach for studying an organism at the systems level by integrating genome-wide computational analysis and the gene expression data.  相似文献
