The effects of salicylic acid (SA) and salinity on the activity of apoplastic antioxidant enzymes were studied in the leaves of two wheat (Triticum aestivam L.) cultivars: salt-tolerant (Gerek-79) and salt-sensitive (Bezostaya). The leaves of 10-d-old seedlings grown at nutrient solution with 0 (control), 250 or 500 mM NaCl were sprayed with 0.01 or 0.1 mM SA. Then, the activities of catalase (CAT), peroxidase (POX) and superoxide dismutase (SOD) were determined in the fresh leaves obtained from 15-d-old seedlings. The NaCl applications increased CAT and SOD activities in both cultivars, compared to those of untreated control plants. In addition, the NaCl increased POX activity in the salt-tolerant while decreased in the salt-sensitive cultivar. In control plants of the both cultivars, 0.1 mM SA increased CAT activity, while 0.01 mM SA slightly decreased it. SA treatments also stimulated SOD and POX activity in the salt-tolerant cultivar but significantly decreased POX activity and had no effect on SOD activity in the saltsensitive cultivar. Under salinity, the SA treatments significantly inhibited CAT activity, whereas increased POX activity. The increases in POX activity caused by SA were more pronounced in the salt-tolerant than in the salt-sensitive cultivar. SOD activity was increased by 0.01 mM SA in the salt-tolerant while increased by 0.1 mM SA treatment in the salt-sensitive cultivar.  相似文献

In order to probe into the potential of relieving senescence in the new cultivars, we have compared gas exchanges and related physiological parameters of two cultivars of winter wheat (DK961, salt-tolerant; JN17, salt-sensitive) under a series of NaCl concentrations. Through out the whole reproductive period, it was noted that net photosynthetic rate, stomatal conductance, pigment contents, ions contents, leaf area index, leaf area duration, leaf relative water content and dry matter accumulation of spikes decreased in both cultivars with saline concentrations increasing. However, the salt-tolerant cultivars showed none significant reductions in those parameters compared with control under 0.3% and 0.5% salt concentrations, with only considerable decrease happening when soil salt concentration exceeded to 0.7%. Sharply contrast to salt-tolerant cultivars, the salt-sensitive cultivars appeared linear reductions in physiological parameters under a series of salt concentrations (0.3%, 0.5% and 0.7%), with the photosynthetic duration being evidently shorter. Significantly positive correlations among K+/Na+, reproductive growth period and total growth period were noted in salt-sensitive cultivars, however, none significant relations appeared among those parameters in salt-tolerant cultivars, indicating a strong tolerant behavior happened. Our result suggested that higher salinity tolerance cultivars of winter wheat could relieve senescence at the reproductive stage.  相似文献

Changes in respiratory pathway, dry weight, contents of proline, ATP, Na+and K+were investigated under five salinity treatments in the leaves of plants of spring wheat (Triticum aestivumL.). Two cultivars differing in salt resistance, namely, 89122 (salt-tolerant) and Longchun13 (salt-sensitive), were used. The decrease in dry weight and K+content was observed with the increasing NaCl concentrations, but more in cv. 89122 plants than in Longchun13 plants. Contents of proline and Na+in both cvs increased greatly, but the former increased more in 89122 while the latter more in Longchun13 plants. In all salinity treatments tested, a salt-induced increase in the activity of the alternative pathway was found, although cytochrome pathway (CP) still remained the main electron transport pathway. ATP production changed in parallel with CP operation. Cv. 89122 plants could produce more ATP than cv. Longchun13 plants exposed to each salinity treatment and their ATP generation could even be stimulated in contrast to its rapidly decline in Longchun13 plants with increased salinity stress. The possible relationship between respiration metabolism and above mentioned physiological changes is discussed.  相似文献

Effect of paclobutrazol (PBZ) treatment on salinity tolerance of wheat (Triticum aestivum) was investigated on a salt-tolerant (Karchia-65) and salt-sensitive (Ghods) cultivars. Salinity significantly reduced the investigated growth parameters such as plant height, length and area of sixth leaf, root length, fresh and dry weight of shoot, roots and sixth leaf, water content (WC) of plant and seeds weight in the both cultivars. The negative effect of salinity in Ghods cultivar was more than Karchia cultivar. However, PBZ treatment reduced the growth in both cultivars, the differences in plant growth among various levels of NaCl decreased in PBZ-treated plants. Salt stress resulted in high accumulation of Na+ in the sixth leaf and roots in both cultivars, particularly in Ghods cultivar. Against Karchia cultivar, salt stress decreased the storage of K+, P and N in sixth leaf and roots in Ghods cultivar. In the both cultivars, PBZ treatment enhanced the K+, P and N contents in sixth leaf and roots by increasing salinity. Although PBZ treatment decreased the growth of plants, it improved the weight of seeds against stress damage. PBZ treatment reduced the accumulation of harmful Na+ ion in plant tissues while increased the K+, P and N contents. These observations suggest that PBZ treatment may increase tolerance by diminishing ionic imbalance caused by salt stress.  相似文献

The aim of this work was to investigate the role of the antioxidant enzymes in salt tolerance comparing the salt-sensitive (Pérola) and a salt-tolerant (Pitiúba) cultivar of cowpea [Vigna unguiculata (L.) Walp.]. Salt stress (100 mM NaCl for 8 d) reduced the leaf growth rate more in the sensitive cultivar. The salt-induced decrease in the relative water content, Na+ accumulation and increase in leaf electrolyte leakage was similar in both cultivars. Salt stress induced a higher increase in the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and phenol peroxidase (POX) in the tolerant cultivar than in sensitive one.  相似文献

A sand culture experiment was conducted to answer the question whether or not exogenous KNO(3) can alleviate adverse effects of salt stress in winter wheat by monitoring plant growth, K(+)/Na(+) accumulation and the activity of some antioxidant enzymes. Seeds of two wheat cultivars (CVs), DK961 (salt-tolerant) and JN17 (salt-sensitive), were planted in sandboxes and controls germinated and raised with Hoagland nutrient solution (6 mM KNO(3), no NaCl). Experimental seeds were exposed to seven modified Hoagland solutions containing increased levels of KNO(3) (11, 16, 21 mM) or 100 mM NaCl in combination with the four KNO(3) concentrations (6, 11, 16 and 21 mM). Plants were harvested 30 d after imbibition, with controls approximately 22 cm in height. Both CVs showed significant reduction in plant height, root length and dry weight of shoots and roots under KNO(3) or NaCl stress. However, the combination of increased KNO(3) and NaCl alleviated symptoms of the individual salt stresses by improving growth of shoots and roots, reducing electrolyte leakage, malondialdehyde and soluble sugar contents and enhancing the activities of antioxidant enzymes. The salt-tolerant cultivar accumulated more K(+) in both shoots and roots compared with the higher Na(+) accumulation typical for the salt-sensitive cultivar. Soluble sugar content and activities of antioxidant enzymes were found to be more stable in the salt-tolerant cultivar. Our findings suggest that the optimal K(+)/Na(+) ratio of the nutrient solution should be 16:100 for both the salt-tolerant and the salt-sensitive cultivar under the experimental conditions used, and that the alleviation of NaCl stress symptoms through simultaneously applied elevated KNO(3) was more effective in the salt-tolerant than in the salt-sensitive cultivar.  相似文献

Salinity tolerance levels and physiological changes were evaluated for twelve rice cultivars, including four white rice and eight black glutinous rice cultivars, during their seedling stage in response to salinity stress at 100 mM NaCl. All the rice cultivars evaluated showed an apparent decrease in growth characteristics and chlorophyll accumulation under salinity stress. By contrast an increase in proline, hydrogen peroxide, peroxidase (POX) activity and anthocyanins were observed for all cultivars. The K+/Na+ ratios evaluated for all rice cultivars were noted to be highly correlated with the salinity scores thus indicating that the K+/Na+ ratio serves as a reliable indicator of salt stress tolerance in rice. Principal component analysis (PCA) based on physiological salt tolerance indexes could clearly distinguish rice cultivars into 4 salt tolerance clusters. Noteworthy, in comparison to the salt-sensitive ones, rice cultivars that possessed higher degrees of salt tolerance displayed more enhanced activity of catalase (CAT), a smaller increase in anthocyanin, hydrogen peroxide and proline content but a smaller drop in the K+/Na+ ratio and chlorophyll accumulation.  相似文献

为研究盐胁迫下小麦幼苗生长及Na+、K+的吸收和积累规律,以中国春、洲元9369和长武134等3种耐盐性不同小麦品种为材料,采用非损伤微测技术检测盐胁迫2 d后的根系K+离子流变化,并对植株体内的Na+、K+含量进行测定。结果表明:短期(2d)盐胁迫对小麦生长有抑制作用,且对根系的抑制大于地上部,耐盐品种下降幅度小于盐敏感品种。盐胁迫下,小麦根际的 K+大量外流,盐敏感品种中国春K+流速显著高于耐盐品种长武134,最高可达15倍。小麦幼苗地上部分和根系均表现为Na+积累增加,K+积累减少,Na+/K+比随盐浓度增加而上升。中国春限Na+能力显著低于长武134,Na+/K+则显著高于长武134。综上所述,盐胁迫下造成小麦组织器官中Na+/K+比上升的主要原因是根系K+大量外流和Na+的过量积累,耐盐性不同的小麦品种间差异显著,并认为根系对K+的保有能力可能是作物耐盐性评价的一个重要指标。  相似文献

In the present study, effects of salinity stress were evaluated in the leaves and roots of two pistachio cultivars (Badami-Rize-Zarand (BZ) and Badami-e-Sefid (BS)). In overall, salinity negatively affects growth of both cultivars with more pronounced effects on BS. The physiological reason of the reduction could be attributed to some extent to more depletion of photosynthetic pigment in BS. In both cultivars, salinity increased proline content. Moderate and high salinities increased the soluble sugar contents in BZ. In both cultivars, Na+ content increased in plant organs with increasing Na+ in the media. Salinity treatment decreased the Fe and Pi contents in BS cultivar, while they remained unchanged in BZ. These results show that BZ cultivar exhibits more tolerance to salinity stress than BS cultivar possibly by better growth performance, accumulating more osmolytes, lesser accumulation of toxic sodium ion and lower Na+/K+ in the shoots as well as maintaining nutrient contents.  相似文献

Physiological mechanisms of salinity–Cd interactions were investigated in inter- and intracellular leaf compartments of salt-tolerant wheat × Lophopyrum elongatum (Host) A. Löve (syn. Agropyron elongatum) amphiploid and its salt-sensitive wheat parent (Triticum aestivum L. cv Chinese Spring). In comparison with the intracellular fluid, only very low Na+ concentrations (up to about 4 mM) were found in the intercellular leaf compartment of wheat after a 75 mM supply of NaCl. NaCl salinity led to a higher Cd concentration in leaves of the salt-sensitive genotype. Cd in the intercellular leaf compartment was not detectable. Higher K+ concentrations in the intercellular leaf compartment of the salt-sensitive genotype suggest a higher plasma membrane permeability caused by NaCl + Cd stress. Ascorbate peroxidase (APX) activity was increased in leaves of the salt-sensitive genotype under the combined NaCl and Cd stress. The highest non-specific peroxidase activities were detected under the combined stresses. It is suggested that NaCl and Cd stress in combination enhance the production of oxygen radicals and H2O2, especially in leaves of the salt-sensitive genotype. As a consequence, disturbed membrane function may cause elevated Cd concentrations in the intracellular leaf compartment under salinity. Cd did not change protein concentration and pattern in leaves. The protein content in inter-and intracellular leaf compartments of both genotypes was increased under salinity. A different protein pattern was obtained in inter- and intracellular leaf compartments. Thus, several physiological interactions between NaCl stress and Cd were found in the two wheat genotypes.  相似文献

The effects of salt stress (50, 100, and 150 mM NaCl) on the levels of free, bound, and total polyamines were studied in the leaf tissues of salt-tolerant (Coban) and salt-sensitive (Sanbro) cultivars of sunflower (Helianthus annuus L.) plants grown for 15 or 25 days under salinity. The amounts of free, acid-soluble bound, and total spermine increased in leaf tissues of sunflower plants subjected to salt stress while the levels of other polyamines decreased or no significant changes occurred. The increase in some PA titers suggests their potential role in overcoming the adverse effect of salinity stress. The salt sensitivity of the sunflower plants was associated with the excessive accumulation of total polyamines in the leaf tissues of salt-sensitive cultivar (Sanbro) under saline condition. The present research was conducted to assess physiological responses of ‘Malas-e-Saveh’ (Malas) and ‘Shishe-Kab’ (Shishe) pomegranates to water of different salt content and electrical conductivity (1.05, 4.61, and 7.46 dS m?1). Both cultivars showed a reduced trunk length due to salinity. Relative water content and stomatal conductivity of both cultivars were significantly reduced under salt stress, but ion leakage increased. In both cultivars, total chlorophyll (Chl) and carbohydrates decreased with rise in salinity, while proline accumulation increased. With salinity increment, the Chl fluorescence parameters (maximum photochemical efficiency of PSII and effective quantum yield of PSII) declined significantly in both cultivars, with higher reduction observed in Shishe. Generally, more Na+ accumulated in shoots and more Cl? was observed in leaves. Cl? accumulation increased by salinity in leaves of Malas, but it was reduced in Shishe. The K+/Na+ ratio in leaves decreased in both cultivars by salinity increment. Malas was less affected by osmotic effects of NaCl, but it accumulated more Cl? in its leaves. Indices of oxidative stress viz., superoxide radical and H2O2 content increased in leaves of all the cultivars with the rise in salinity level, the increase was more pronounced and significant in salt-sensitive varieties and non-significant in resistant cultivars. Except for glutathione reductase (GR), basal activities of all other antioxidative enzymes viz. superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX) and glutathione reductase (GR) were significantly higher in leaves of all the resistant cultivars as compared to the sensitive ones. A differential response of salinity was observed on various enzymatic and non-enzymatic components of antioxidant system in leaves of salt-tolerant and salt-sensitive cultivars of rice (Oryza sativa L.). Activities of superoxide dismutase and glutathione reductase enhanced in all the tolerant cultivar while declined in the sensitive cultivars with increasing salinity from 0 to 100 mM. Salt-stress induced the activities of catalase and peroxidase in all the cultivars but the magnitude of increase was more pronounced in the sensitive cultivars than in the tolerant cultivars. Contrarily, APX activity increased in the salt-sensitive cultivars but showed no significant change in the salt-tolerant cultivars. The amount of ascorbic acid content, reduced glutathione (GSH), reduced/oxidized glutathione (GSSG) ratio was higher in leaves of the tolerant cultivars than that of the sensitive cultivars under saline conditions. It is inferred that leaves of salt-tolerant cultivars tend to attain greater capacity to perform reactions of antioxidative pathway under saline conditions to combat salinity-induced oxidative stress.  相似文献

A selected Glycine max (L.) salt-tolerant calluscell line (R100) was significantly more tolerant to salt than a salt-sensitiveline (S100) during exposure to salt stress. Growth (Fresh and Dry weights) ofthe R100 cell line declined significantly at NaCl concentrations greater than 75mM, while growth of the S100 cell line was already impaired at 25mM NaCl. Levels of Na+ and Cl inthe callus were elevated as the salt concentration increased, whileK+, Ca2+ and Mg2+ levels weremarkedly reduced. The lower s reduction and Na+accumulation found in the S100 callus corresponded with the higher callusdehydration during salinity. Calli grown on Miller's basal medium weresupplied with 100 mM NaCl for 12 days and then supplied with mediumwithout NaCl to relieve salinity stress. The Na+ andCl content decreased in both R100 and S100 cell lines duringthe first 24 h and reached normal levels four days after transferto the normal medium. This lower concentration was maintained until the end ofthe experiment. Concurrently, the K+ content andK+/Na+ ratio increased sharply and reached theirhighest levels within 24 h in both salt-sensitive and salt-tolerantcell lines. These data suggest that the inhibitory effects of salinization ongrowth and accumulation of potentially toxic ions (Na+,Cl) can be readily reversed when salinity is relieved.  相似文献

The molybdenum cofactor (MoCo) is a component of aldehyde oxidase (AO EC 1.2.3.1), xanthine dehydrogenase (XDH EC 1.2.1.37) and nitrate reductase (NR, EC 1.6.6.1). The activity of AO, which catalyses the last step of the synthesis of abscisic acid (ABA), was studied in leaves and roots of barley (Hordeum vulgare L.) plants grown on nitrate or ammonia with or without salinity. The activity of AO in roots was enhanced in plants grown with ammonium while nitrate-grown plants exhibited only traces. Root AO in barley was enhanced by salinity in the presence of nitrate or ammonia in the nutrient medium while leaf AO was not significantly affected by the nitrogen source or salinity of the medium.Salinity and ammonium decreased NR activity in roots while increasing the overall MoCo content of the tissue. The highest level of AO in barley roots was observed in plants grown with ammonium and NaCl, treatments that had only a marginal effect on leaf AO. Plants of two wheat (Triticum aestivum L.) cultivars differing in salt tolerance were grown in sand with nutrient solutions. 35-d-old plants were subjected to 5 levels of salinity created by adding NaCl, CaCl2 and Na2SO4. Growth reduction caused by salinity was accompanied by increased Na+ and Cl- concentrations, Na+/K+ ratio, and decreased concentration of K+. The salt tolerant cv. Kharchia 65 showed better ionic regulation. Salinity up to 15.7 dS m-1 induced increased uptake of Na+ and Cl- but higher levels of salinity were not accompanied by further increase in uptake of these ions. Observed increases in Na+ and Cl- concentrations at higher salinities seemed to be the consequence of reduction in growth. Uptake of K+ was decreased; more in salt sensitive cultivar. 以冬小麦(Triticum aestivum L.)品种周麦18(Zhoumai18)和豫麦49(Yumai49)为材料,采用盆栽培养,研究100、250、350 mmol/L和450 mmol/L NaCl 浓度胁迫下,小麦幼苗干物质分配、根系特征、叶绿素含量、游离脯氨酸含量和根系活力变化规律.结果表明,随着盐分浓度的增加,两个品种小麦的叶面积、地上干重以及根的长度显著减小;根系干重、根直径、根表面积、根体积、根系活力以及叶绿素含量呈先上升后下降趋势,在250 mmol/L NaCl处理下达最大.叶绿素a/b随NaCl浓度升高而上升.随盐分浓度变化周麦18叶片游离脯氨酸含量高而变化幅度大,450mmol/L处理组的含量高于对照组1.5倍.供试品种冬小麦耐盐阈值为250~350 mmol/L.  相似文献
