The suitability of electron microscope autoradiography for sutdying the uptake and intracellular localization of lipid vesicles (liposomes) containing radiolabeled saturated phospholipids has been examined. Data are presented showing that preparation of specimens for electron microscope autoradiography by conventional methods is accompanied by significant translocation and intercellular redistribution of radiolabeled saturated lipids, causing spurious labeling patterns. Intercellular redistribution of radiolabeled lipid was demonstrated by mixing glutaraldehyde-fixed mous L1210 cells that had been incubated with sonicated lipid vesicles containing [H] dipalmitoyl phosphatidylcholine with an indicator cell population (fixed avian erythrocytes) which had not been exposed to vesicles and showing that after electron microscope processing radiolabeled grains were present in both cell types. The same redistribution artifact also probably affects the intracellular localization of radiolabeled lipids. This artifact is discussed in relation to previous work in which autoradiographic methods have been used for ultrastructural localization saturated phospholipids in cells and tissues.  相似文献

The distal aspect of the housefly ommatidium was surveyed by the scanning electron microscope. Attention was directed to the somal eminence of the superior central cell and the lens to large pigment cell junction. The underside of each lens facet exhibits six hexagonally arranged incisures. Into each of these indentations are fitted several large pigment cells. This hexagonal indentation appears to be a tenacious anchorage. Two corneal pigment cells laterally encircle the pseudocone and at their proximal extension they enclose the Semper cells and neck of the retinula. The somal eminence of the superior central cell is about 10 mum from the base of the corneal pigment cell enclosure. Micrographs were used to construct a diagram of the ommatidium above the basement membrane. Suggestions are made as to the functional correlates of the observed ommatidial structures.  相似文献

DNA-synthesizing cells in the gonads of the ascidian Styela clava were labeled with tritiated thymidine and detected with autoradiography. In the testis, spermatogonia and primary spermatocytes are labeled after 1 hr. Labeled spermatozoa occur in the lumen of the testis follicles after 10 days and in the sperm ducts after 20 days. In the ovary, only germ cells (oogonia and pre-leptotene primary oocytes) and follicle cells are labeled after 1 hr. By 60 days, oocytes with basophilic cytoplasm (15–65 μ in diameter) are labeled; test cells embedded in larger eosinophilic oocytes (150 μ in diameter) are also labeled. Germ cells give rise to both oocytes and follicle cells. Through continued cell division, follicle cells give rise to test cells.  相似文献

There is a need for quantitative data on patterns and rates of movement of organisms to understand their movement behavior and predict their rates of spread. Opportunities for studying movement of biological control agents are presented during release programs. However, despite these opportunities, patterns and range of dispersal are often not considered. For example, information about effects of wind on dispersal patterns and heterogeneities in rates of movement is critical to predicting future range expansion of biological control agents and determining proximity of multiple releases. Here, the pattern and range of movement of a fire ant parasitoid, Pseudacteon tricuspis Borgmeier, was investigated by performing a series of mass-release-resighting experiments. Flies were released at a central location surrounded by radial transects containing trays of host ants at variable distances along four axes. Resighted flies were censused at these trays at 30 min intervals, up to 2 h postrelease. The dispersal pattern of P. tricuspis in the short term was consistent with a simple diffusion model. On average, 50% of P. tricuspis dispersed ≤10 m, and 95% dispersed ≤29 m. Diffusion rates were variable, depending on release densities, but tended to decline over time after release. Drift of dispersing flies was detected in several trials, and was attributed to prevailing wind dynamics. Data from this assessment of the short term redistribution pattern of P. tricuspis could be useful in determining proximity of releases of this, and other fire ant parasitoids.  相似文献

A new example of a racemate showing unusual enantiomeric resolution phenomenon, in which simple recrystallization of the racemate leads to remarkable enantiomeric enrichment of either enantiomer up to 100% ee in the mother liquor, has been found. This compound is (±)-[2-[4-(3-ethoxy-2-hydroxypropoxy)phenylcarbamoyl]-ethyl]dimethylsulfonium p-nitrobenzenesulfonate [EtOCH2CH(OH)CH2OC6H4NHCOCH2CH2SMe2+O2NC6H4SO3] [(±)-SN]. By repeating recrystallization of (±)-SN and the resulting deposited crystals successively and collecting the resulting enantiomerically enriched mother liquors with the same chirality sense, highly efficient enantiomeric resolution of the racemate into its separate enantiomers has been accomplished. The relationship between the occurrence of this enantiomeric resolution phenomenon and the crystal properties has been investigated with respect to SN and its aryl- and alkylsulfonate derivatives. The mode of enantiomeric resolution of (±)-SN was similar to that of para-substituted benzenesulfonate derivatives (±)-ST (4-MeC6H4SO3) and (±)-SC (4-ClC6H4SO3) previously reported, whereas the unsubstituted derivative (±)-SB (C6H5SO3) and alkysulfonate derivatives (±)-SO (n-C8H17SO3) and (±)-SM (CH3SO3) did not show such an enantiomeric resolution phenomenon. The crystalline form of the former racemates that underwent the enantiomeric resolution was racemic compounds, while the latter were mixed crystals (solid solutions) composed of the respective optical antipodes. An investigation with the scanning electron microscope into the microstructure of the surface layer covering the oral teeth of Latimeria chalumnae has shown this tissue to be enamel of the type found in amphibians, reptiles and mammals. It is not comparable with enameloid, cuticular enamel or terminal membrane enamel as described in scanning electron microscope accounts of the teeth of actinopterygians. Equidistant lamellations parallel to the enamel-dentine junction are a distinctive feature, interpreted in this study as phasic appositional growth, a characteristic of the ectodermal type of enamel. These conclusions together with those from histological and microradiographic studies confirm that enamel is present in coelacanths.
The microstructure of enamel in the teeth of Latimeria an extant actinistian compares well with that of fossil crossopterygian teeth described from polarized light studies and indicates that enamel, homologous with the ectodermal type of enamel is found in both actinistians and rhipidistians.  相似文献

Here, we hypothesized that the microbial gut flora of animals/pests living in polluted environments, produce substances to thwart bacterial infections. The overall aim of this study was to source microbes inhabiting unusual environmental niches for potential antimicrobial activity. Two cockroach species, Gromphadorhina portentosa (Madagascar) and Blaptica dubia (Dubia) were selected. The gut bacteria from these species were isolated and grown in RPMI 1640 and conditioned media were prepared. Conditioned media were tested against a panel of Gram‐positive (Methicillin‐resistant Staphylococcus aureus, Streptococcus pyogenes, Bacillus cereus) and Gram‐negative (Escherichia coli K1, Salmonella enterica, Serratia marcescens, Pseudomonas aeruginosa, Klebsiella pneumoniae) bacteria, as well as the protist pathogen, Acanthamoeba castellanii. The results revealed that the gut bacteria of cockroaches produce active molecule(s) with potent antibacterial properties, as well as exhibit antiamoebic effects. However, heat‐inactivation at 95°C for 10 min had no effect on conditioned media‐mediated antibacterial and antiamoebic properties. These results suggest that bacteria from novel sources i.e. from the cockroach's gut produce molecules with bactericidal as well as amoebicidal properties that can ultimately lead to the development of therapeutic drugs.

Significance and Impact of the Study

The bacteria isolated from unusual dwellings such as the cockroaches' gut are a useful source of antibacterial and antiamoebal molecules. These are remarkable findings that will open several avenues in our search for novel antimicrobials from unique sources. Furthermore studies will lead to the identification of molecules to develop future antibacterials from insects.  相似文献

The distribution and quantitation of 125I-alpha-bungarotoxin (alpha-BTX) binding sites and thus acetylcholine receptor (AChR) were determined in mouse sternomastoid muscle by electron microscope autoradiography. We found that a valid criterion for receptor saturation at the neuromuscular junction was the complete elimination of neurally evoked tetanic muscle contractions, since, when such a criterion was used for the endpoint of toxin incubation, alpha-BTX was bound to approximately 90% of total available endplate sites. When, without implying localization, the presynaptic axonal membrane was used as a convenient reference structure, the concentration of alpha-BTX relative to this membrane was determined to be 46,000 +/- 27% sites/mum2.  相似文献

Summary The huge numbers of molluscs, worms and bacteria found in the sheltered tidal flats of the Dutch Wadden Sea, decompose large amounts of organic matter. In this process nutrients incorporated in the particulate organic matter are transferred to the dissolved state. The importance of this process for nutrient budgets was first demonstrated by POSTMA (1954) and later by various authors with emphasis on tidal watersheds and sheltered bays (see Neth. J. Sea Res., 8 (2/3). 1974).The nutrient budgets of the Balgzand tidal flats, as a typical example of a sheltered bay, have been studied during one tidal period by TIJSSEN and VAN BENNEKOM (1976) and recently for a longer period in a cooperative project by the Studiedienst Hoorn of Rijkswaterstaat (DE BOER, 1978) and the Netherlands Institute for Sea Research (MANUELS, 1978). A qualitative measure for the high intensity of mineralization are the high concentrations of nutrients in the water on the Balgzand, compared to those in the Marsdiep (Table I).Simple box models can be used to derive the intensity of nutrient production from the concentration differences in which also the water depth and the exchange velocity of the water with adjacent areas contribute. As a first approximation all mineralization is thought to be situated in the surface sediment (VAN BENNEKOM et al., 1974) and the characteristics parameter becomes the nutrient production per m2 per day. For reactive silicate this parameter shows a pronounced seasonal cycle. In January, February and Marchin situ production is hardly detectable while in July and August 500 mg SiO2 m–2d–1 is found. For comparison, weathering of soil by rain water gives 4–30 mg SiO2.m–2d–1 with only slight seasonal variations. The difference is at least partially explained by assuming a different source: diatom frustules in the case of the Balgzand and silicate minerals in the case of normal weathering.The scarce data available indicate that the seasonal cycles of in situ production of phosphate, nitrate and ammonia on the Balgzand are less pronounced than for reactive silicate. The picture is further complicated because ammonia is always produced, but sometimes nitrate is consumed, apparently by denitrification.The nutrient ratios change during the year (Table II). Especially in summer the contribution of silicate is relatively large. This is in marked contrast with the nutrient ratios in the coastal sea water where during the summer the amount of silicate is relatively low. The nutrient ratios are probably an important factor in the seasonal and regional species composition of the phytoplankton.  相似文献

Multi drug resistant Klebsiella pneumoniae showed stepwise adaptation when grown in increasing concentration of Cefotaxime eventually reaching a maximum of 2 mg/ml. The resultant Cefotaxime resistant mutant strain was stable and did not revert to susceptibility on frequent subculturing. The response of the cells to different concentration of Cefotaxime was examined by scanning electron microscope which showed that the size of the bacterium increased with increasing concentration of Cefotaxime.  相似文献

【目的】本研究旨在明确访花昆虫红腹毛蚊Bibio rufiventris触角感器的类型和分布。【方法】通过扫描电镜(scanning electron microscope, SEM)观察红腹毛蚊雌、雄成虫触角感器的种类、数量和形态,比较雌雄个体间的差异。【结果】红腹毛蚊雌、雄成虫触角均包含3部分,分别为柄节、梗节和鞭节,其中鞭节由8个亚节组成。雌成虫触角平均总长度为862.556±78.662μm,雄成虫触角平均总长度为880.361±83.253μm,雌、雄成虫触角各亚节的长度几乎相似,只有鞭节第8亚节长度有显著差异。红腹毛蚊的触角感器共有4大类,即刺形感器、锥形感器、毛形感器和B?hm氏鬃毛。其中,雌性红腹毛蚊触角感器共有6种亚型,即刺形感器、毛形感器2型、锥形感器(1, 2和4型)和B?hm氏鬃毛;雄性红腹毛蚊触角感器共有5种亚型,即刺形感器、毛形感器1型、锥形感器(2和3型)和B?hm氏鬃毛。【结论】红腹毛蚊雌、雄成虫触角感器在种类、数量以及形态特征方面存在一定差异。本研究为进一步探究红腹毛蚊触角感器的生理功能及其行为活动的分子机制提供了形态学基础。  相似文献

Sponges (Porifera) currently represent one of the richest sources of natural products and account for almost half of the pharmacologically active compounds of marine origin. However, to date very little is known about the pharmacological potential of the sponges from polar regions. In this work we report on screening of ethanolic extracts from 24 Antarctic marine sponges for different biological activities. The extracts were tested for cytotoxic effects against normal and transformed cell lines, red blood cells, and algae, for modulation of the activities of selected physiologically important enzymes (acetylcholinesterase, butyrylcholinesterase, and α-amylase), and for inhibition of growth of pathogenic and ecologically relevant bacteria and fungi. An extract from Tedania (Tedaniopsis) oxeata was selectively cytotoxic against the cancer cell lines and showed growth inhibition of all of the tested ecologically relevant and potentially pathogenic fungal isolates. The sponge extracts from Isodictya erinacea and Kirkpatrickia variolosa inhibited the activities of the cholinesterase enzymes, while the sponge extracts from Isodictya lankesteri and Inflatella belli reduced the activity of α-amylase. Several sponge extracts inhibited the growth of multiresistant pathogenic bacterial isolates of different origins, including extended-spectrum beta-lactamase and carbapenem-resistant strains, while sponge extracts from K. variolosa and Myxilla (Myxilla) mollis were active against a human methicillin-resistant Staphylococcus aureus strain. We conclude that Antarctic marine sponges represent a valuable source of biologically active compounds with pharmacological potential.  相似文献

The relationship of chromosome pairing to chiasma localisation in the grasshopper Stethophyma grossum was investigated by a combined light and electron microscope study. Observations on the extent of synaptonemal complex formation in spermatocytes suggest that pairing is complete in all chromosome regions and that localised chiasmata do not therefore follow from localised pairing of homologues. This study also revealed an unprecedented variant of synaptonemal complex structure. Each bivalent was found to contain an asymmetrical region within which one lateral element was enormously enlarged while the other lateral element retained normal dimensions. This asymmetrical region is probably confined to one end of each bivalent and seems to extend constantly about 4 from the nuclear membrane attachment site. The possible significance of this variant of synaptonemal complex structure is briefly discussed.  相似文献
