Protein storage vacuoles (PSVs) in soybean (Glycine max (L.) Merr.) cotyledon cells are formed by subdivision of the central vacuole early in seed maturation. They persist until the fifth or sixth day after germination when the central vacuole re-forms. The major integral membrane protein of PSVs, called Tonoplast Integral Protein or TIP, is highly conserved in the seeds of higher plants (K.D. Johnson et al. 1989, Plant Physiol. 91, 1006–1013). The primary sequence of TIP indicates that it may be a pore protein, although of unknown function (K.D. Johnson et al. 1990, Plant Cell 2, 525–532). TIP is apparently seed-specific and is localized in the protein-storage-vacuole membrane of the storageparenchyma cells and the tonoplast of provascular cells. Using correlated immunoblot and electron microscopicimmunocytochemical assays, we have studied TIP accumulation during seed maturation and its disappearance during seed germination. We have determined that the accumulation of TIP in the protein-storage-vacuole membrane is not correlated with the presence or concentration of stored protein in the organelle. Accumulation of TIP occurs primarily after the division of the central vacuole into protein-storage vacuoles is complete and most of the stored protein has been deposited. Transport of TIP to the PSV membrane is apparently mediated by the Golgi apparatus. Quantitative SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis)-immunoblots indicate that, after germination is initiated, TIP abundance is unchanged for the first 4d, but that between days 5 and 7 of growth its abundance decreases drastically. TIP is removed from the PSV membrane prior to the completion of storageprotein mobilization and concurrently with re-formation of the central vacuole. The mechanism of TIP removal appears to involve autophagic sequestering of membrane inside the PSV. The delivery of proteins to the vacuole and its limiting membrane (the tonoplast) by the secretory system is thought to be a dissociative process in which vesicles bud from one compartment and fuse with another. We studied the transport kinetics of phytohemagglutinin (PHA) and tonoplast intrinsic protein (TIP) in mesophyll protoplasts obtained from transgenic tobacco plants transformed with genes encoding these two proteins. In pulse-chase experiments, arrival of PHA in the vacuole was found to be slower (completed 24 hr after synthesis) than the arrival of TIP in the tonoplast (completed 6 hr after synthesis). Brefeldin A and monensin block protein transport by interfering in specific vesicle transport steps. Brefeldin A prevents anterograde vesicle transport between the endoplasmic reticulum and the Golgi, whereas monensin inhibits correct sorting in the trans-Golgi network by disrupting the proton gradient across the membrane. Both inhibitors blocked the transport of PHA to the vacuole and altered the rate at which its complex glycan is processed by Golgi enzymes. Neither drug stopped the arrival of TIP in the tonoplast, suggesting that the flow of vesicles continues in the presence of these inhibitors. We suggest that soluble proteins like PHA and membrane proteins like TIP reach their vacuolar destinations by different paths.  相似文献

We generated fusions between three Arabidopsis (Arabidopsis thaliana) tonoplast intrinsic proteins (TIPs; alpha-, gamma-, and delta-TIP) and yellow fluorescent protein (YFP). We also produced soluble reporters consisting of the monomeric red fluorescent protein (RFP) and either the C-terminal vacuolar sorting signal of phaseolin or the sequence-specific sorting signal of proricin. In transgenic Arabidopsis leaves, mature roots, and root tips, all TIP fusions localized to the tonoplast of the central vacuole and both of the lumenal RFP reporters were found within TIP-delimited vacuoles. In embryos from developing, mature, and germinating seeds, all three TIPs localized to the tonoplast of protein storage vacuoles. To determine the temporal TIP expression patterns and to rule out mistargeting due to overexpression, we generated plants expressing YFP fused to the complete genomic sequences of the three TIP isoforms. In transgenic Arabidopsis, gamma-TIP expression was limited to vegetative tissues, but specifically excluded from root tips, whereas alpha-TIP was exclusively expressed during seed maturation. delta-TIP was expressed in vegetative tissues, but not root tips, at a later stage than gamma-TIP. Our findings indicate that, in the Arabidopsis tissues analyzed, two different vacuolar sorting signals target soluble proteins to a single vacuolar location. Moreover, TIP isoform distribution is tissue and development specific, rather than organelle specific.  相似文献

The tonoplast mediates the transport of various ions and metabolites between the vacuole and cytosol by mechanisms that remain to be elucidated at the molecular level. The primary structure of only one tonoplast protein, the H(+)-ATPase, has been reported to date. Here we report the primary structure of tonoplast intrinsic protein (TIP), a 27-kilodalton intrinsic membrane protein that occurs widely in the tonoplasts of the protein storage vacuoles (protein bodies) of seeds [Johnson, K.D., et al. (1989). Plant Physiol. 91, 1006-1013]. Hydropathy plots and secondary structure analysis of the polypeptide predict six membrane-spanning domains connected by short loops and hydrophilic, cytoplasmically oriented N- and C-terminal regions. TIP displays significant homology with several other membrane proteins from diverse sources: major intrinsic polypeptide from bovine lens fiber plasma membrane; NOD 26, a peribacteroid membrane protein in the nitrogen-fixing root nodules of soybean; and interestingly, GIpF, the glycerol facilitator transport protein in the cytoplasmic membrane of Escherichia coli. Based on the homology between TIP and GIpF and the knowledge that the protein storage vacuolar membrane and the peribacteroid membrane are active in solute transport, we propose that TIP transports small metabolites between the storage vacuoles and cytoplasm of seed storage tissues.  相似文献

Reports from a number of laboratories describe the presence of a family of proteins (the major intrinsic protein family) in a variety of organisms. These proteins are postulated to form channels that function in metabolite transport. In plants, this family is represented by the product of NOD26, a nodulation gene in soybean that encodes a protein of the peribacteroid membrane, and tonoplast intrinsic protein (TIP), an abundant protein in the tonoplast of protein storage vacuoles of bean seeds (KD Johnson, H Höfte, MJ Chrispeels [1990] Plant Cell 2: 525-532). Other homologs that are induced by water stress in pea and in Arabidopsis thaliana and that are expressed in the roots of tobacco have been reported, but the location of the proteins they encode is not known. We now report the presence and derived amino acid sequences of two different TIP proteins in A. thaliana. α-TIP is a seed-specific protein that has 68% amino acid sequence identity with bean seed TIP; γ-TIP is expressed in the entire vegetative body of A. thaliana and has 58% amino acid identity with bean seed TIP. Both proteins are associated with the tonoplast. Comparisons of the derived amino acid sequences of the seven known plant proteins in the major intrinsic protein family show that genes with similar expression patterns (e.g. water stress-induced or seed specific) are more closely related to each other than the three A. thaliana homologs are related. We propose that the nonoverlapping gene expression patterns reported here, and the evolutionary relationships indicated by the phylogenetic tree, suggest a functional specialization of these proteins.  相似文献

Soluble proteins that reside in the lumen of the endoplasmic reticulum are known to have at their carboxyterminus the tetrapeptides KDEL or HDEL. In yeast and mammalian cells, these tetrapeptides function as endoplasmic reticulum (ER)-retention signals. To determine the effect of an artificially-introduced KDEL sequence at the exact carboxyterminus of a plant secretory protein, we modified the gene of the vacuolar protein phytohemagglutinin-L (PHA) so that the amino-acid sequence would end in LNKDEL rather than LNKIL, and expressed the modified gene in transgenic tobacco with a seed-specific promoter. Analysis of the glycans of PHA showed that most of the control PHA had one endoglycosidase H-sensitive and one endoglycosidase H-resistant glycan, indicating that it had been processed in the Golgi complex. On the other hand, a substantial portion of the PHA-KDEL (about 75% at mid-maturation and 50% in mature seeds) had two endoglycosidase H-sensitive glycans. Phytohemagglutinin with two endoglycosidase H-sensitive glycans is normally found in the ER. Using immunocytochemistry we found that a substantial portion of the PHA-KDEL was present in the ER or accumulated in the nuclear envelope while the remainder was found in the protein storage vacuoles (protein bodies). We interpret these data to indicate that carboxyterminal KDEL functions as an ER retention-retardation signal and causes protein to accumulate in the nuclear envelope as well as in the ER. The vacuolar membrane (tonoplast) of plant cells contains a polytopic integral membrane protein with six membrane-spanning domains and cytoplasmically oriented amino-terminal and carboxy-terminal domains. This protein, tonoplast intrinsic protein (TIP), is a member of the membrane intrinsic protein (MIP) family of proteins, a family of channel proteins found in a variety of organisms. In bean seeds, alpha-TIP is synthesized on the rough endoplasmic reticulum and its transport to the tonoplast is mediated by the secretory system. In this study, we report that a polypeptide segment that includes the sixth membrane domain and the cytoplasmic tail of 18 amino acids of alpha-TIP is sufficient to target the reporter protein phosphinotricine acetyltransferase to the tonoplast of stably transformed tobacco cells. To determine if the carboxy-terminal cytoplasmic tail of alpha-TIP contains important tonoplast targeting information, a deletion construct lacking the 15 carboxy-terminal amino acids was introduced for transient expression in tobacco cells; we found that the slightly truncated protein still accumulated in the tonoplast. From these results, we concluded that a transmembrane domain of a tonoplast protein probably contains sufficient information for transport to the tonoplast. Whether such transport occurs by bulk flow or involves specific cellular machinery remains to be determined.  相似文献

Summary Vegetative apices, floral apices and flower petals of five Solanaceae (potato, tomato, tobacco, petunia and nightshade) and of corn and Nigella were examined with an electron microscope for the presence of protein bodies in the cell vacuoles. Electron-dense bodies were found in vacuoles of all plants investigated but not in every tissue examined. The bodies observed in the apices are similar to the protein bodies previously found in tomato leaves where they appear to be related to the presence of chymotrypsin inhibitor I protein (Shumway et al., 1970). The bodies appeared in very young cells in small vacuoles, disappearing as the cell matured. They are apparently related to the growth and development of the new cells. The results suggest that plants may regulate specific proteins within the apical region through selective synthesis and degradation of proteins accompanied by compartmentalization in the vacuole.Scientific Paper No. 3822, College of Agriculture, Washington State University, Pullman, Project 1791. Many plant species contain a seed-specific tonoplast intrinsic protein (TIP) in their protein storage vacuoles (PSVs). Although the function of the protein is not known, its structure implies it to act as a transporter protein, possibly during storage nutrient accumulation/breakdown or during desiccation/imbibition of seeds. As mature somatic embryos of Picea abies (L.) Karst. (Norway spruce) contain PSVs, we examined the presence of TIP in them. Both the megagametophyte and seed embryo accumulate storage nutrients, but at different times and we therefore studied the temporal accumulation of TIP during seed development. Antiserum against the seed-specific a-TIP of Phaseolus vulgaris recognized an abundant 27 kDa tonoplast protein in mature seeds of P. abies. By immunogold labeling of sectioned mature megagametophytes we localized the protein to the PSV membrane. We also isolated the membranes of the PSVs from mature seeds and purified an integral membrane protein that reacted heavily with the antiserum. A sequence of 11 amino acid residues [AEEATHPDSIR], that was obtained from a polypeptide after in-gel trypsin digestion of the purified membrane protein, showed high local identity to a-TIP of Arabidopsis thaliana and to a-TIP of P. vulgaris. The greatest accumulation of TIP in the megagametophytes occurred at the time of storage protein accumulation. A lower molecular mass band also stained from about the time of fertilization until early embryo development. The staining of this band disappeared as the higher molecular mass (27 kDa) band accumulated in the megagametophyte during seed development. Total protein was also extracted from developing zygotic embryos and from somatic embryos. In zygotic embryos low-levels of TIP were seen at all stages investigated, but stained most at the time of storage protein accumulation. The protein was also present in mature somatic embryos but not in proliferating embryogenic tissues in culture. In addition to the seed tissue material, the antiserum also reacted with proteins present in extracts from roots and hypocotyls but not cotyledons from 13-day-old seedlings.  相似文献

The expression of proteins in transgenic plants offers an elegant means to examine targeting signals used for transport to intracellular sites of accumulation. We have used electron microscopic immunogold procedures to localize several different storage proteins and lectins expressed in transgenic tobacco seeds. The objective of these studies is to characterize targeting signals which permit translocation and accumulation in protein storage vacuoles (protein bodies). Vacuolar proteins such as phaseolin and phytohemagglutinin (PHA) are correctly transported to the protein storage vacuoles of transgenic tobacco seeds. Site-directed mutagenesis was used to change Asn-linked glycosylation sites of PHA. Minus glycan PHA was accumulated in the protein storage vacuoles indicating that glycans do not confer targeting information. Zein the non-vacuolar storage protein of maize accumulates in the protein storage vacuoles indicating that deposition occurs in some proteins which may lack vacuolar targeting signals.  相似文献

The lectin, concanavalin A (Con A), was localized in the cotyledon of developing jack beans (Canavalia ensiformis (L.) DC) by electron-microscope immunocytochemistry. In mature seeds, Con A was present in protein-storage vacuoles (protein bodies) of storage-parenchyma cells. Although protein bodies could be seen in other cell types, only protein bodies in storage-parenchyma cells contained Con A. During seed development, Con A was also localized on the endoplasmic reticulum and Golgi apparatus, presumably en route toward deposition within the protein bodies. Cultured oat (Avena sativa L. — naked form) aleurone protoplasts were employed as a model system for following changes which accompany the development of vacuoles during in-vitro incubation. Over a 5-d period, the aleurone grains progressively grew and fused to form a large central vacuole and the volume of the protoplasts increased sevenfold. The growth of the vacuole was accompanied by a progressive acidification of the vacuolar sap. Vacuolation was inhibited by high concentrations of mannitol and by cycloheximide and cordycepin applied at various times during the incubation period. Neither cycloheximide nor cordycepin affected the initial phases of vacuolation but cycloheximide retarded subsequent stages, particularly if added early in the incubation. Cordycepin inhibited only the later stages of vacuolation. Strictosidine, a precursor to over 1000 indole alkaloids including the anti-tumor drugs vinblastine, vincristine, and camptothecin, is produced by the condensation of tryptamine and secologanin. Strictosidine synthase, the enzyme responsible for this condensation, is the first committed step in the indole-alkaloid pathway. We have introduced a modified cDNA encoding Strictosidine synthase from Catharanthus roseus (L.) Don. (McKnight et al. 1990, Nucl. Acids Res. 18, 4939) driven by the CaMV 35S promoter into tobacco (Nicotiana tabacum L.). Transgenic tobacco plants expressing this construct had from 3 to 22 times greater strictosidinesynthase activity than C. roseus plants. Ultrastructural immunolocalization demonstrated that strictosidine synthase is a vacuolar protein in C. roseus and is correctly targeted to the vacuole in transgenic tobacco. Immunoblot analysis of strictosidine synthase showed that two distinct forms of the enzyme were produced in transgenic tobacco plants but that only a single form was made in C. roseus. This observation indicates that the second form of the protein is not simply a result of overexpression in tobacco, but may reflect differences in protein processing between tobacco and C. roseus.Abbreviations cDNA complementary DNA - TLC thin-layer chromatography We thank Dr. C.A. Storage proteins of maize (Zea mays L.) were studied in germinated seeds, as were the proteins of protein bodies isolated from endosperms at different germination times. Major endosperm storage proteins were degraded in a sequential way, glutelin 2 being hydrolysed faster than zein 1. Immunocytochemical labelling of the different protein bodies using the antisera anti-glutelin 2 and anti-zein 1 indicates that the protein bodies were degraded by progressive hydrolysis from their surface. The digestion of glutelin 2 correlated with the disappearance of the protein-body membranes.  相似文献

Development of legume seeds is accompanied by the synthesis of storage proteins and lectins, and the deposition of these proteins in protein-storage vacuoles (protein bodies). We examined the subcellular distribution, in developing seeds of the common bean, Phaseolus vulgaris L., of the major storage protein (phaseolin) and the major lectin (phytohemagglutinin, PHA). The proteins were localized using an indirect immunocytochemical method in which ultrathin frozen sections were immunolabeled with rabbit antibodies specific for either PHA or phaseolin. Bound antibodies were then localized using goat-anti-rabbit immunoglobulin G adsorbed onto 4- to 5-nm colloidal gold particles. The sections were post-fixed with OsO4, dehydrated, and embedded in plastic on the grids. Both PHA and phaseolin exhibited a similar distribution in the storage-parenchyma cells, being found primarily in the developing protein bodies. Endoplasmic reticulum and Golgi complexes (cisternal stacks and associated vesicles) also were specifically labeled for both proteins, whereas the cytosol and other organelles, such as mitochondria, were not. Plant cell vacuoles may have storage or lytic functions, but biochemical markers specific for the tonoplasts of functionally distinct vacuoles are poorly defined. Here, we use antipeptide antibodies specific for the tonoplast intrinsic proteins alpha-TIP, gamma-TIP, and delta-TIP in confocal immunofluorescence experiments to test the hypothesis that different TIP isoforms may define different vacuole functions. Organelles labeled with these antibodies were also labeled with antipyrophosphatase antibodies, demonstrating that regardless of their size, they had the expected characteristics of vacuoles. Our results demonstrate that the storage vacuole tonoplast contains delta-TIP, protein storage vacuoles containing seed-type storage proteins are marked by alpha- and delta- or alpha- and delta- plus gamma-TIP, whereas vacuoles storing vegetative storage proteins and pigments are marked by delta-TIP alone or delta- plus gamma-TIP. In contrast, those marked by gamma-TIP alone have characteristics of lytic vacuoles, and results from other researchers indicate that alpha-TIP alone is a marker for autophagic vacuoles. In root tips, relatively undifferentiated cells that contain vacuoles labeled separately for each of the three TIPs have been identified. These results argue that plant cells have the ability to generate and maintain three separate vacuole organelles, with each being marked by a different TIP, and that the functional diversity of the vacuolar system may be generated from different combinations of the three basic types.  相似文献

Phosphoribulokinase (PRK; EC 2.1.7.19) is active in illuminated chloroplasts and inactive in darkened chloroplasts. This regulatory mechanism is mediated by thioredoxin-dependent reduction of a kinase disulfide in vivo. Extracts of spinach (Spinacia oleracea L.) leaves in the presence of 10 mM dithiothreitol contain a single 80-kDa form of PRK as judged by gel filtration. Gel filtration of thiol-free extracts of light-harvested tissue shows the presence of two inactive forms of PRK, the 80-kDa form and an aggregate (> 550 kDa) form, but treatment of both forms with dithiothreitol restores kinase activity. Gel filtration following extraction of dark-harvested tissue in the absence of dithiotreitol demonstrates the presence of only the heavier form. Inclusion of 400 mM (NH4)2SO4 in the homogenization buffer during extraction of light-harvested tissue suppresses the formation of the high-M r form of PRK, but does not eliminate the aggregate form observed in extracts of dark-harvested leaves. However, prolonged treatment of extracts from dark-harvested tissue with 400 mM (NH4)2SO4 results in conversion of the high-M r form of phosphoribulokinase to the low-M r form. Two integral membrane proteins, MP28 and MP23, were found in protein bodies isolated from pumpkin (Cucurbita sp.) seeds. Molecular characterization revealed that both MP28 and MP23 belong to the seed TIP (tonoplast intrinsic protein) subfamily. The predicted 29 kDa precursor to MP23 includes six putative membrane-spanning domains, and the loop between the first and second transmembrane domains is larger than that of MP28. The N-terminal sequence of the mature MP23 starts from residue 66 in the first loop, indicating that an N-terminal 7 kDa fragment that contains one transmembrane domain is post-translationally removed. During maturation of pumpkin seeds, mRNAs for MP28 and MP23 became detectable in cotyledons at the early stage, and their levels increased slightly until a rapid decrease occurred at the late stage. This is consistent with the accumulation of the 29 kDa precursor and MP28 in the cotyledons at the early stage. By contrast, MP23 appeared at the late stage simultaneously with the disappearance of the 29 kDa precursor. Thus, it seems possible that the conversion of the 29 kDa precursor to the mature MP23 might occur in the vacuoles after the middle stage of seed maturation. Both proteins were localized immunocytochemically on the membranes of the vacuoles at the middle stage and the protein bodies at the late stage. These results suggest that both MP28 and the precursor to MP23 accumulate on vacuolar membranes before the deposition of storage proteins, and then the precursor is converted to the mature MP23 at the late stage. These two TIPs might have a specific function during the maturation of pumpkin seeds.  相似文献

DiP , a gene from Antirrhinum majus , which encodes a protein with striking homology to other integral membrane proteins, was cloned. The gene was specifically expressed in mature seeds and during seedling germination, particularly in cotyledons of seedlings grown in the dark. The deduced product, called DiP, for dark intrinsic protein, shows strong homology with the MIP family of channel transporters which include; the bovine major intrinsic protein (MIP), the Escherichia coli glycerol facilitator (GlpF), the peribacteroid nodulin-26 (Nod26), and the tonoplast protein from kidney bean (TIP). DiP is most similar to other plant members of this family, and in particular to the tobacco protein TobRB7 which is expressed specifically in roots. However, the expression pattern of diP suggests that its product is functionally more similar to the tonoplast intrinsic protein from kidney bean since it is most highly expressed in the cotyledons of germinating seedlings, before the cells undergo expansion growth and become photosynthetic.  相似文献
