Boldness, a measure of an individual's propensity for taking risks, is an important determinant of fitness but is not necessarily a fixed trait. Dependent upon an individual's state, and given certain contexts or challenges, individuals may be able to alter their inclination to be bold or shy in response. Furthermore, the degree to which individuals can modulate their behaviour has been linked with physiological responses to stress. Here we attempted to determine whether bold and shy rainbow trout, Oncorhynchus mykiss, can exhibit behavioural plasticity in response to changes in state (nutritional availability) and context (predation threat). Individual trout were initially assessed for boldness using a standard novel object paradigm; subsequently, each day for one week fish experienced either predictable, unpredictable, or no simulated predator threat in combination with a high (2% body weight) or low (0.15%) food ration, before being reassessed for boldness. Bold trout were generally more plastic, altering levels of neophobia and activity relevant to the challenge, whereas shy trout were more fixed and remained shy. Increased predation risk generally resulted in an increase in the expression of three candidate genes linked to boldness, appetite regulation and physiological stress responses - ependymin, corticotrophin releasing factor and GABA(A) - but did not produce a significant increase in plasma cortisol. The results suggest a divergence in the ability of bold and shy trout to alter their behavioural profiles in response to internal and exogenous factors, and have important implications for our understanding of the maintenance of different behavioural phenotypes in natural populations.  相似文献

The effects of estradiol-17β (E2) on bone resorption and formation as well as its effects on scale resorption were investigated in rainbow trout in order to elucidate the role of the hormone in calcium mobilization from calcified tissues, and to clarify the importance of scale and bone as calcium reserves during sexual maturation. Furthermore, the effects of nutritional status on calcified tissues and E2-induced calcium mobilization were studied. In fed as well as fasted rainbow trout, E2 treatment increased scale osteoclastic activity measured as tartrate-resistant acid phosphatase activity, and reduced scale calcium content, suggesting that E2 increases scale resorption in both the fed and fasted fish. Using histomorphometry, E2 treatment was found to decrease pharyngeal bone resorption in fed, but not in fasted rainbow trout. The E2 effect on rainbow trout bone is consistent with its physiological role in mammals and birds where E2 has been reported to decrease bone resorption. It appears therefore that rainbow trout protect their skeleton and instead use scales as a source of calcium during E2-induced calcium mobilization. The formation of pharyngeal bone was decreased by fasting, and the importance of the nutritional status for the activity of the bone cells in rainbow trout is therefore emphasized. Currently, we have little information concerning the involvement of β3-adrenergic receptors (AR) in cardiovascular regulation in fishes. The goal of this study was to investigate the effect of β3-AR ligands on in vivo cardiovascular function in larval and adult rainbow trout (Oncorhynchus mykiss). In adult fish, injection of BRL373443-AR agonist) resulted in an increase in heart rate (fH) (~ 31%) while stroke volume (Sv) was reduced (? 25.9%). Injection of SR59230A3-AR antagonist) and propranolol (β12-AR antagonist) resulted in increases in dorsal aorta blood pressure (PDA) with differing effect on cardiac variables (fH and Sv). To confirm specificity of the results, BRL37344 was injected following sequential injections of phentolamine (α1-AR antagonist), atropine (muscarinic antagonist), propranolol and SR59230A. While phentolamine had no effect on BRL37344, atropine completely abolished the influence of BRL37344 on fH, Sv and cardiac output (Q). In larval trout, BRL37344 (10 and 100 μM) induced a significant concentration-dependent increase in fH while SR59230A (1 and 10 μM) and propranolol (1 and 10 μM) separately caused a significant concentration-dependent decrease. These data suggest that β3-ARs have an important role in regulation of cardiovascular function, and provide evidence for a potential interaction between muscarinic and adrenergic receptors in rainbow trout.  相似文献

Potential immunostimulatory effects of orally administered β-glucan were investigated in combination with immersion vaccination against enteric redmouth disease caused by Yersinia ruckeri in rainbow trout (Oncorhynchus mykiss). A linear, unbranched and pure (purity ≥98%) β-1,3-glucan (syn. paramylon) from the alga Euglena gracilis was applied at an inclusion level of 1% β-glucan in feed administered at a rate of 1% biomass?day(-1) for 84 consecutive days. Fish were vaccinated after two weeks of experimental feeding and bath challenged with live Y.?ruckeri six weeks post-vaccination. Blood and head kidney were sampled at day 0, 13 (1 day pre-vaccination), 15, 55, 59 (day 3 post-challenge (p.c.)), 70 and 84. Vaccination induced significantly increased survival p.c., whereas the β-glucan had no effect on survival in either unvaccinated or vaccinated fish. Expression in head kidney of genes related to the acute phase response, i.e. interleukin-1β (IL-1β), serum amyloid A (SAA), precerebellin, and hepcidin, was significantly different in vaccinated fish receiving β-glucan compared to vaccinated controls at day 3 p.c., while no effect of β-glucan was observed among unvaccinated fish. Significant interaction between β-glucan and vaccination was found for the regulation of IL-1β, tumour necrosis factor-α, interferon-γ, SAA, precerebellin and hepcidin p.c. For SAA, the significant effect of β-glucan in vaccinated fish persisted at day 14 p.c. and 28 p.c. The difference in gene expression among vaccinated fish was mainly observed as down-regulations in vaccinated, β-glucan fed fish compared to up-regulations or no regulation in vaccinated controls. Slightly increased levels of plasma lysozyme activity were found in fish (both unvaccinated and vaccinated) receiving β-glucan at day 3 p.c. compared to control fed groups. This was associated with a faster clearance of Y.?ruckeri in unvaccinated fish receiving β-glucan. In contrast to the trend towards a beneficial effect of β-glucan on plasma lysozyme activity, a trend towards suppression of plasma antibodies was seen in both unvaccinated and vaccinated fish receiving β-glucan. However, the effects of β-glucan were not reflected in the survival curves, and the differences seen in plasma lysozyme activity and antibody levels may have counteracted and set off each other as well as counteracted any potential effect represented by the differences in gene expression found.  相似文献

Effects of Hilyses®, fermented Saccharomyces cerevisiae (S. cerevisiae), on growth, body composition and skin mucus immune components in rainbow trout were quantified. Ninety rainbow trout (105 ± 5 g) were randomly assigned to 2 groups in triplicates and fed dietary Hilyses® (5 g kg?1) or control diet without Hilyses® for 50 days. Results of this study demonstrated that growth performance increased significantly by the dietary yeast supplement; however body composition was not affected in treatment group. At the 45th and 50th day of feeding trial, results of mucus samples demonstrated that yeast supplementation in treatment group significantly promoted enzyme activities, namely lysozyme, protease, alkaline phosphatase and esterase compared to control group. Significant increases were also observed in hemagglutination and antibacterial activity against Yersinia ruckeri in fish fed treatment diet. The present study suggests that fermented S. cerevisiae may effectively promote the growth performance and skin non-specific immune parameters in rainbow trout.  相似文献

Primary cultures of rainbow trout brain astroglial cells, prelabelled with [1-14C] polyunsaturated fatty acids (PUFA), were treated with various agents and the effects on cell morphology and n-3 PUFA metabolism were investigated.2. The effects of dibutyryl cAMP on trout astroglial cell cultures were similar to those of dibutyryl cAMP on primary cultures of rat brain astroglia, inducing process-bearing morphology.3. Hydrocortisone had the opposite effect to dibutyryl cAMP on the morphology of the trout astroglial cells, reducing the degree of process-bearing morphology.4. Dibutyryl cAMP increased the percentage of [14C]eicosapentaenoic acid (EPA, 20:5n-3) elongated to 22:5, whereas hydrocortisone increased the percentage of [14C]linolenic acid (LNA, 18:3n-3) desaturated and elongated to 20:5.5. Peroxisome effectors also affected trout astroglial cell morphology with the peroxisomal proliferator, clofibrate, reducing the degree of process formation, whereas the peroxisomal inhibitor, 3-aminotriazole, increased the degree of process formation.6. However, the peroxisomal effectors had similar effects on n-3 PUFA metabolism, reducing the percentages of [14C]LNA converted to 20:5 and 22:5, whereas both increased the percentages of [14C]EPA converted to 22:5 and 22:6.7. We concluded that changes in the morphology of trout astroglial cells in vitro are not directly related to changes in n-3 PUFA metabolism.  相似文献

To identify virulence-associated genes of a fish pathogen Yersinia ruckeri, we screened a total of 1056 mini-Tn5-Km2 signature-tagged mutants in rainbow trout by immersion challenge. Of 1056, 25 mutants were found survival-defective as they could not be re-isolated from fish kidney 7 days after infection. Mutated gene in F2-4 mutant, one of the 25 mutants, was homologous to uvrY that encodes UvrY response regulator of BarA–UvrY two-component system (TCS). Mutant F2-4 was significantly more sensitive (P < 0.05) to H2O2-mediated killing and was less able to infect Epithelioma papulosum cyprini cells. However, UvrY mutation did not affect survival of F2-4 mutant in the presence of non-immune fish serum and its ability to grow under iron starvation. In a time-course co-infection, mutant F2-4 had lower bacterial loads on day 1 itself, and by day 5 there was nearly a 1,000-fold difference in infection levels of the parent and mutant strains. The barA homolog of Y. ruckeri was PCR-amplified and sequence analyses identified four domains that were characteristic of hybrid histidine kinases. To conclude, the BarA–UvrY TCS contributes to the pathogenesis of Y. ruckeri in its natural host rainbow trout, possibly by regulating invasion of epithelial cells and sensitivity to oxidative stress induced by immune cells.  相似文献

Background

Non-invasive micro-ultrasound was evaluated as a method to quantify intrauterine growth phenotypes in mice. Improved methods are required to accelerate research using genetically-altered mice to investigate the interactive roles of genes and environments on embryonic and placental growth. We determined (1) feasible age ranges for measuring specific variables, (2) normative growth curves, (3) accuracy of ultrasound measurements in comparison with light microscopy, and (4) weight prediction equations using regression analysis for CD-1 mice and evaluated their accuracy when applied to other mouse strains.

Methods

We used 30–40 MHz ultrasound to quantify embryonic and placental morphometry in isoflurane-anesthetized pregnant CD-1 mice from embryonic day 7.5 (E7.5) to E18.5 (full-term), and for C57Bl/6J, B6CBAF1, and hIGFBP1 pregnant transgenic mice at E17.5.

Results

Gestational sac dimension provided the earliest measure of conceptus size. Sac dimension derived using regression analysis increased from 0.84 mm at E7.5 to 6.44 mm at E11.5 when it was discontinued. The earliest measurement of embryo size was crown-rump length (CRL) which increased from 1.88 mm at E8.5 to 16.22 mm at E16.5 after which it exceeded the field of view. From E10.5 to E18.5 (full term), progressive increases were observed in embryonic biparietal diameter (BPD) (0.79 mm to 7.55 mm at E18.5), abdominal circumference (AC) (4.91 mm to 26.56 mm), and eye lens diameter (0.20 mm to 0.93 mm). Ossified femur length was measureable from E15.5 (1.06 mm) and increased linearly to 2.23 mm at E18.5. In contrast, placental diameter (PD) and placental thickness (PT) increased from E10.5 to E14.5 then remained constant to term in accord with placental weight. Ultrasound and light microscopy measurements agreed with no significant bias and a discrepancy of less than 25%. Regression equations predicting gestational age from individual variables, and embryonic weight (BW) from CRL, BPD, and AC were obtained. The prediction equation BW = -0.757 + 0.0453 (CRL) + 0.0334 (AC) derived from CD-1 data predicted embryonic weights at E17.5 in three other strains of mice with a mean discrepancy of less than 16%.

Conclusion

Micro-ultrasound provides a feasible tool for in vivo morphometric quantification of embryonic and placental growth parameters in mice and for estimation of embryonic gestational age and/or body weight in utero.  相似文献

We evaluated the impact of non-native rainbow trout Oncorhynchus mykiss on a population of endemic Cedarberg ghost frog Heleophryne depressa in the upper Krom River (Olifants-Doring River Catchment, Cape Fold Ecoregion). We compared H. depressa abundance (using kick-sampling and underwater video analysis) and environmental conditions between sites above and below a waterfall that marks the upper distribution limit of O. mykiss. Heleophryne depressa abundance was significantly greater above the waterfall than that below it, and, because there was no significant difference in measured environmental variables, O. mykiss presence is identified as the most likely explanation for the observed decrease in H. depressa abundance.  相似文献

Post-hatch fishes lack a functional gill and use cutaneous surfaces for exchange with the surrounding environment. The ionoregulatory hypothesis posits that ionoregulation is the first physiological process to be limited by cutaneous exchange, necessitating its shift to the gills. We hypothesized that the ontogeny of branchial ammonia excretion (Jamm) is coupled to Na+ uptake () in accordance with the current model for exchange in freshwater. Using divided chambers, branchial and cutaneous Jamm, and oxygen consumption (MO2) by larval rainbow trout were assessed. Following hatch, the skin accounted for 97% and 86% of total Jamm and , respectively. Jamm and shifted to the gills simultaneously at 15 days post-hatch (dph) and were highly correlated (R2 = 0.951) at the gills, but not the skin, over development. Contrastingly, MO2 shifted significantly later at 27 dph, in agreement with the ionoregulatory hypothesis. Moreover, the mRNA expression and/or enzymatic activity of Rhesus proteins, Na+/H+-exchanger, H+-ATPase, Na+/K+-ATPase and carbonic anhydrase, all key components of the -exchange system, increased in the gills over larval development. We propose that the ontogeny of branchial occurs as exchange and provide evidence for a novel element to the ionoregulatory hypothesis, the excretion of potentially lethal metabolic ammonia.  相似文献

The influence of infection with the juvenile stages of the sea louse, Lepeophtheirus salmonis (Kr?yer) on the response of rainbow trout Oncorhynchus mykiss (Walbaum) to a net confinement protocol was investigated. The experiment consisted of two groups of seawater-adapted rainbow trout, one which was exposed to a total of 4000 nauplii/copepodid stages of L. salmonis 30, 25 and 14 days prior to confinement. Confinement elicited a greater stress response in the lice-exposed fish, than in the controls, as seen by higher plasma cortisol and glucose levels. A reduced spleen somatic index in exposed fish following 6 h confinement coincided with increased erythrocyte and lymphocyte numbers in the blood. Circulating lymphocyte numbers were significantly reduced in both groups 24 h post-confinement, when a lower alternative complement activity was recorded in control fish. Prior to confinement, lice-exposed fish had an elevated serum lysozyme activity and reduced oxygen radical production by blood leukocytes. Following confinement, lysozyme activity was gradually reduced in lice-exposed trout. During confinement, oxygen radical production decreased in control fish and increased in infested fish. Overall, transient exposure to juvenile lice altered the response to a second stressor, which has implications for management procedures of L. salmonis exposed fish.  相似文献

Biological characteristics of “Kunimasu” (Oncorhynchus kawamurae), poorly known even before the species was prematurely believed extinct, have now become apparent following the examination of 59 specimens from Lake Saiko, Yamanashi Prefecture, Japan and comparison with 58 examples of “Himemasu” (kokanee of Oncorhynchus nerka in Japan), also from Lake Saiko. Significant (p < 0.01) differences between Kunimasu and Himemasu from Lake Saiko occurred in counts of anal-fin rays, pectoral-fin rays, gill rakers (no overlap found) and pyloric caeca. Secondary sexual characters related to maturity level were also found in Kunimasu, the body being more compressed in pre-spawning and spawning males and females than in immature and maturing individuals. Furthermore, maturing male Kunimasu and Himemasu also differed in body shape. Body coloration of Kunimasu also differed according to level of maturity, the nuptial coloration in both sexes being olive-green when alive and black when fresh. Dark dots, found in ca. 40 % of Kunimasu individuals examined, were less distinct than in Himemasu. The spawning season of Kunimasu extended through winter and early spring in Lake Saiko, with spawning males and females remaining near the bottom, compared with non-spawning individuals which occupied the upper and middle profundal zones in the daytime.  相似文献

The possibility of interspecific hybridization between Kunimasu (Oncorhynchus kawamurae) and Himemasu (Oncorhynchus nerka) was investigated in a large number of specimens, in a search for basic data relevant to conservation needs. A Bayesian-based clustering method using five microsatellite DNA loci separated 144 specimens from Lake Saiko, Yamanashi Prefecture, into two genetically distinct groups, corresponding to Kunimasu and Himemasu. Application of a threshold of individual proportion of membership of 0.90, so as to separate hybrids from purebreds, resulted in all specimens except two having high probability of purebred. These results implied that Kunimasu and Himemasu have been maintained as independent genetic entities in Lake Saiko.  相似文献

The rainbow trout (Oncorhynchus mykiss) TGF-β1 sequence was one of the first fish cytokines described. Studies of its expression suggest it is constitutively expressed but displays refractory inducibility. Here we describe a second TGF-β1 (TGF-β1b) gene that is novel in several respects. TGF-β1b possesses typical TGF-β features, including a CXC motif and an integrin binding site, a tetrabasic cut site and a mature peptide of 112 amino acids (aa) containing nine conserved cysteine residues. The mature peptide is 83% identical to the first TGF-β1 sequence described in rainbow trout, that we designate TGF-β1a, and relative to TGF-β1a shows higher homology to Atlantic salmon TGF-β1b, zebrafish TGF-β1a, and sea bass and seabream TGF-β1. The gene organisation of salmonid TGF-β1b genes, as inferred from Atlantic salmon whole genome shotgun contigs, is a 6 exon/5 intron structure with exons 3 and 4 of salmonid TGF-β1a genes apparently fused together. The two trout TGF-β1 genes have a wide distribution in vivo, with highest expression found in immune tissues for both isoforms indicating that TGF-β1 has a predominant role in immunity of fish. Expression of both genes was also seen during the ontogeny of trout, with TGF-β1a relatively constant in expression level but TGF-β1b increasing over time. Immune responses in head kidney (HK) macrophages induced by pathogen associated molecular patterns (PAMPs), pro-inflammatory cytokines, mitogens and pathway activators highly elevated the expression level of TGF-β1b but not that of TGF-β1a. TGF-β1b expression was also increased by polyinosinic:polycytidylic acid (poly(I:C)) and/or lipopolysaccharide (LPS) stimulation in three different trout cell lines studied. Finally we show that TGF-β1b is potentially involved in defense against infection with viral haemorrhagic septicemia virus (VHSV), which had no effect on TGF-β1a expression. Thus, it is likely the TGF-β1b gene represents a copy which fulfils the major immune orchestrating functions of TGF-β1 as seen in other vertebrates.  相似文献
