以IGFBP3基因作为秦川牛(Bos taurus)部分屠宰指标的侯选基因,在对60头秦川牛的IGFBP3基因进行PCR-RFLP和序列分析的基础上,对秦川牛群体中IGFBP3基因座等位基因和基因型频率的分布及其与秦川牛部分屠宰性状的关系进行了分析。结果发现,在秦川牛群体中,651 bp的PCR 产物经过限制性内切酶HaeIII消化后,表现出3种基因型,其中等位基因A、B及3种基因型AA、AB、BB的频率分别为0.84、0.16和070、0.28、0.02。经序列分析发现,第299位的C→A颠换(GGCC变成了GGAC)导致了1个HaeIII限制性酶切位点的丢失而产生了该基因座多态性。在所研究的群体中,该多态基因座处于Hardy-Weinberg平衡状态(P>005)。对13头24月龄秦川牛进行屠宰分析,发现不同基因型对秦川牛部分屠宰指标有一定影响,AA、AB及BB型个体的屠宰率、净肉率及西冷、牛柳、眼肉和嫩肩肉的产率逐渐降低,但差异不显著(P>0.05);AA型个体的眼肌面积大于BB型个体(P<0.05),AB型和BB型个体胴体脂肪含量高于AA型个体(P<0.01)。 Abstract:DNA samples from 60 Qinchuan cattle (Bos taurus) were analyzed with PCR-RFLPs and sequencing for insulin-like growth factor binding protein 3 (IGFBP3) gene.Fragments of 651 bp were amplified with two primers and the products of PCR were digested with restriction endonuclease HaeIII.The produced fragments showed three genotypes,namely AA,AB and BB after electrophoresis.Frequencies of the genotype AA,AB,BB and allele A,B were 0.7,0.28,0.02,and 0.84,0.16,respectively.Sequence analysis showed that a transversion of C→A at 299 nt resulted in loss of the cleaved site of restriction endonuclease HaeIII and produced this polymorphism.This polymorphic locus of IGFBP3 gene was at Hardy-Weinberg equilibrium (P>0.05).The genotypes of AA,AB,BB slightly affected several slaughter and carcass traits of Qinchuan cattle.Dressing percentage,net meat percentage,striplion percentage,tenderloin percentage,ribeye percentage and tender shoulder percentage were decreased with the genotypes of AA,AB and BB in Qinchuan cattle,but it was not significant (P>0.05).Average ribeye area in individuals of AA genotype was significantly higher than that in individuals of BB genotype (P<0.05),and beef fat content in individuals of genotype AB and BB was significantly higher than that in individuals of AA genotype (P<0.01).  相似文献

Binding sites for platelet-derived growth factor (PDGF) differ in their selectivity for the AA, AB and BB forms of PDGF. Human fibroblasts bind BB well and AA poorly, whereas Swiss 3T3 cells bind more similar quantities of each ligand. We found that AA PDGF was weakly mitogenic for human fibroblasts, but strongly mitogenic for 3T3 cells. Tyrosine phosphorylation of human fibroblast receptors was stimulated most by BB and least by AA, whereas the phosphorylation of 3T3 cell receptors was stimulated more uniformly by the three dimers. The receptor polypeptides that were phosphorylated were very similar. We suggest that phosphorylation of the receptor is proportional to the number of binding sites available for each ligand. Tyrosine phosphorylation of a number of other cell proteins was also proportional to receptor phosphorylation. In contrast, protein kinase C (PKC)-dependent serine and tyrosine phosphorylations were stimulated maximally by low level occupancy of PDGF binding sites, and phosphorylation of p36 required high occupancy. These data raise the possibility that differences in biological potency of AA, AB and BB forms of PDGF may be due simply to differences in the numbers of binding sites, rather than to different biochemical functions of their receptors.  相似文献

Platelet-derived growth factor (PDGF) is a dimeric protein composed of A- and B-chains (AA, AB, and BB). PDGF purified from human platelets has been shown to be composed primarily of the AB heterodimer. Immunoblots of total platelet extracts, cell-bound PDGF from the platelet extracts, and acute human wound fluid with PDGF A- and B-chain-specific antisera all demonstrate that the PDGF A-chain is the predominant peptide. Chemotactic and immunochemical assays of chromatographic fractions during PDGF isolation support these observations and demonstrate that PDGF AA can be separated from PDGF AB and BB by ion exchange chromatography. These studies indicate that the AA isoform constitutes the major PDGF dimer contained in human platelets and is the major form present at sites of injury during the acute phase of the wound repair response.  相似文献

Alpha-crystallin, a major protein of mammalian lens, consists of two subunits, alpha A-crystallin and alpha B-crystallin. They interact to form an aggregate and play a prominent role in the maintenance of lens transparency. We evaluated the interaction between these subunits via surface plasmon resonance (SPR) using four combinations of immobilized protein and analyte: 1) AA: alpha A-crystallin was ligand immobilized onto the sensor and alpha A-crystallin was passed over the ligand, 2) AB: ligand - alpha A-crystallin, analyte - alpha B-crystallin, 3) BB: ligand - alpha B-crystallin, analyte- alpha B-crystallin, 4) BA: ligand - alpha B-crystallin, analyte - alpha A-crystallin. The order of rate of dissociation was AA approximately BA>BB approximately AB. We also examined the dissociation of gamma irradiated alpha A- and alpha B-crystallins. As radiation dose increased, so did the dissociation rate of all of the crystallins. The order of rate of dissociation of irradiated crystallins was BB>AB approximately BA>AA. The results indicate that BB is the most susceptible to gamma-irradiation and that alpha B-crystallin forms a more stable aggregate than alpha A-crystallin under normal conditions. However, when alpha B is irradiated the aggregate becomes unstable.  相似文献

Four immunologically distinct subunits were characterized in glutathione (GSH) S-transferases of human liver. Five cationic enzymes (pI 8.9, 8.5, 8.3, 8.2 and 8.0) have an apparently similar subunit composition, and are dimers of 26 500-Mr (A) and 24 500-Mr (B) subunits. A neutral enzyme, pI 6.8, is a dimer of B-type subunits. One of the anionic enzymes, pI 5.5, is also a dimer of 26 500-Mr subunits. However, the 26 500-Mr subunits of this anionic enzyme form are immunologically distinct from the A subunits of the cationic enzymes, and have been designated as A'. Immunoabsorption studies with the neutral enzyme, BB, and the antibodies raised against the cationic enzymes (AB) indicate that A and B subunits are immunologically distinct. Hybridization in vitro of the A and B subunits of the cationic enzymes (AB) results in the expected binary combinations of AA, AB and BB. Studies with the hybridized enzyme forms indicate that only the A subunits express GSH peroxidase activity. A' subunits have maximum affinity for p-nitrobenzyl chloride and p-nitrophenyl acetate, and the B subunits have highest activity towards 1-chloro-2,4-dinitrobenzene. The other anionic form, pI 4.5, present in liver is a heterodimer of 22 500-Mr (C) and B subunits. The C subunits of this enzyme are probably the same as the 22 500-Mr subunits present in human lung and placental GSH transferases. The distinct immunological nature of B and C subunits was also demonstrated by immunoaffinity and subunit-hybridization studies. The results of two-dimensional polyacrylamide-gel-electrophoretic analyses indicate that in human liver GSH transferases, three charge isomers of Mr 26 500 (A type), two charge isomers of Mr 24 500 (B type) and two charge isomers of Mr 22 500 (C type) subunits are present.  相似文献

采用PCR-SSCP方法对中国美利奴羊和哈萨克羊中IGFBP-3基因的多态性进行了检测, 并对不同基因型与中国美利奴羊部分羊毛性状间的关联性进行了分析。结果在位于内含子1区的一段178 bp的扩增产物经SSCP分析后出现了3种基因型, 基因型AA、AB和BB及等位基因A、B在中国美利奴羊中的频率分别为0.70、0.24、0.06和0.82、0.18; 在哈萨克羊中的频率分别为0.87、0.13、0.00和0.93、0.07。序列分析发现: 在该序列的122位碱基表现多态性 (g.122 G>T)。所研究的两个群体在该位点上均处于Hardy-Weinberg不平衡状态(P<0.01)。不同基因型对部分羊毛性状有一定的影响: 不同基因型个体在剪毛后体重和净毛率上没有明显差异。AA、AB及BB基因型个体的羊毛伸直长度逐渐变短, 其中AA与AB基因型之间差异极显著(P<0.01)。AA型个体的剪毛量和羊毛密度要明显低于AB型(P<0.01)和BB型个体(P<0.05); 羊毛纤维直径则明显高于AB型(P<0.01)和BB型(P<0.05)个体。  相似文献

In our study, genetic variation in coding region of cattle CCAAT enhancer binding protein alpha(namely CEBPA)gene was detected by PCR-RFLP and DNA sequencing methods in 215 individuals from Qinchuan cattle breed. Two haplotypes (A and B) and three observed genotypes (AA, AB, and BB) were detected. The result of DNA sequence showed one mutation by comparisons with NC_007316. The mutation at nt963 (T>G) were located in coding region of the CEBPA gene. Associations between the CEBPA gene genetic variation and the carcass traits were revealed in Qinchuan cattle. Least squares analysis revealed a significant statistical effect of the CEBPA gene different genotypes on slaughter weight and carcass weight in Qinchuan cattle. Individuals with BB genotype showed higher slaughter weight and carcass weight than individuals with AA and AB genotypes. Therefore, these results suggest that the CEBPA gene is a strong candidate gene that affects carcass traits in Qinchuan cattle.  相似文献

In the course of the investigation of piebald (black-white) cattle it is found that 17,62% animals produce the AA type beta-lactoglobulin, 49,52%--the AB type and 32,86%--the BB type. The higher inhibitory activity of milk with the BB type beta-lactoglobulin was found which retained in dilution 1 : 32. The total flora of teat, cyst and parenchyma milk of animals with the BB type beta-lactoglobulin as well as Enterococcus bacteria were much lower than in milk of cows with the AA type homogenous form. The animals with the first type of milk protein had mastitis more rarely as compared to those with the second type. The animals with heterogenous form of beta-lactoglobulin had intermediate values in most of their characteristics.  相似文献

Electrophoretic analysis of 794 individual samples of water buffalo hemoglobin shows the presence of three phenotypes, AA, AB and BB. Phenotype AA has only the fast hemoglobin (Hb-fast). Phenotype AB and BB have the fast as well as the slow hemoglobin (Hb-slow). The quantitative ratio Hb-fast: Hb-slow is approximately 68:32 in BB and 84:16 in AB. Several genetic mechanisms are discussed in the light of the biochemical and family data provided.  相似文献

The present meta-analysis was carried to provide the more reliable estimates of gene frequency and association of Rsa 1 generated candidate genotype of prolactin gene within exon-3 with performance traits in 1198 Indian dairy cows using data from 15 published studies. Six genetic models viz., codominant (AA vs. AB, AA vs. BB and AB vs. BB), dominant (AA+AB vs. BB), completely over dominant (AA+BB vs. AB) and recessive (AA vs. AB+BB) were used to obtain standardized mean difference (SMD) between genotypes. Meta-analysis showed that the gene frequency of A allele (156 bp) was 0.60 (95% confidence interval (CI) 0.54, 0.65). In association analysis, cows with AB genotype [SMD?=?0.65, 95% CI 0.00, 1.30] had significantly (P?<?0.05) higher lactation milk yield (LMY) as compared to BB genotype, whereas AA and AB genotypes had similar trend. Likewise, AA?+?AB also had larger effect [SMD?=?2.31, 95% CI 0.21, 4.10] on LMY as compared to BB. Cows with AB genotype had significantly lower age at first calving (AFC) with small effect [SMD (AA vs. AB)?=?1.38, 95% CI 0.06, 2.70] and medium effect [SMD (AB vs. BB)?=????3.83, 95% CI???6.41,???1.24] as compared to cows with AA and BB genotypes, respectively. This finding was confirmed under dominant and completely over dominant models. In case of fat%, AA genotype showed negative effect (SMD?=????0.51, 95% CI???0.84,???0.17) under recessive model. It was concluded that the propagation of allele A is promising to help dairy farmers to improve the genetic quality of their dairy cows.

Two pairs of primers (P1 and P2) were designed to detect single nucleotide polymorphisms of exon 2 and intron 2 of bone morphogenetic protein 4 (BMP4) gene in both high fecundity breed (Jining Grey goat) and low fecundity breeds (Boer, Angora and Inner Mongolia Cashmere goats) by single strand conformation polymorphism. Results showed that no polymorphism was detected for exon 2 (primer P1) of BMP4 gene in four goat breeds. For intron 2 (primer P2), three genotypes (AA, AB and BB) were detected in Jining Grey and Inner Mongolia Cashmere goats, two genotypes (AB and BB) in Angora goats, and only one genotype (AA) in Boer goats. Sequencing revealed one mutation (2203G>A) of BMP4 gene in the genotype BB in comparison to the genotype AA. The differences of litter size between AA, AB and BB genotypes were not significant (P > 0.05) in Jining Grey goats. A pair of primer (P3) was designed to detect polymorphism in the 3' flanking region of BMP4 gene that contained dinucleotide repeated sequence (CA) in the four goat breeds by microsatellite analysis. For primer P3, three genotypes (CC, CD and DD) were detected in four goat breeds. Sequencing revealed one more CA dinucleotide in genotype DD than in genotype CC. The Jining Grey does with genotype CC had 0.55 (P < 0.05) or 0.72 (P < 0.05) kids more than those with genotype CD or DD. These results preliminarily indicated that allele C of BMP4 gene is a potential DNA marker for improving litter size in goats.  相似文献

采用PCR-SSCP方法对长白猪(87头)、大白猪(79头)和马身猪(102头)的胰岛素样生长因子-I(IGF-I)基因exon3和exon4分别进行单核苷酸多态性分析。发现exon3上有多态性,且存在3种基因型(AA、AB、BB)。统计结果表明,3种基因型在各品种中的分布不一致,多重比较差异极显著(P<0.01)。固定效应模型分析结果表明,背膘厚基因型间差异显著(P<0.05),而初生重、断奶重和6月龄重基因型间差异不显著(P>0.05)。最小二乘分析结果表明,BB基因型与其它2种基因型比较有较大的初生重,同AA和AB型比较差异极显著(P<0.01),3种基因型在初生重的大小排列顺序为AA相似文献

Components of litter size in gilts with different prolactin receptor genotypes   总被引:17,自引:0,他引:17  
Behavioral estrus and components of litter size at Day 35/36 of pregnancy were studied in gilts with prolactin receptor (PRLR) genotype AA (n=9), AB (n=25), and BB (n=22). This PRLR polymorphism (two alleles, A and B) has been associated with litter size, although it is not known whether the polymorphism itself causes differences in litter size or whether it is a marker for a closely linked causative gene. Estrus length in three successive estrous cycles was not affected by genotype, but estrous cycle length tended (P<0.1) to be longer for AA gilts compared to AB and BB gilts. AA gilts had a significantly (P<0.05) higher ovulation rate (21.5+/-0.9) than BB gilts (18.7+/-0.6), resulting in a numerically higher number of embryos at Day 35/36 (17.0+/-1.3, 15.6+/-0.8, and 13.7+/-0.9 for AA, AB, and BB gilts, respectively) which may lead to a subsequent difference in litter size. Ovulation rate of AB gilts (20.0+/-0.5) was intermediate. Genotype affected the total weight of the ovaries (P<0.05). Even after subtraction of the total weight of corpora lutea, ovarian weight in AA gilts was highest (16.6+/-1.0 g), in BB lowest (13.4+/-0.6g), and in AB gilts intermediate (15.0+/-0.6g; P<0.05). Unlike AB gilts, in AA and BB gilts uterine length was adapted to litter size, which led to longer (P<0.05) uteri for AA gilts (669+/-28 cm) compared to BB gilts (566+/-18 cm). Furthermore, embryos of AA gilts had heavier placentae (52.5+/-3.4 g) and larger implantation surface areas (309+/-19 cm(2)) than embryos of BB (42.0+/-2.3g, P<0.05; 256+/-12 cm(2), P<0.1) or AB (43.2+/-2.0 g, P<0.1; 257+/-11 cm(2), P<0.05) gilts. Results of this experiment show that the PRLR gene or a very closely linked gene affects porcine ovaries, uterus, and placenta in a way that might lead to differences in litter size. Since other genes and also environmental factors, however, might change the effect within the 112 days to parturition, it is preferable to state that the PRLR gene is a candidate gene for ovulation rate rather than for litter size.  相似文献

摘要: 以544头中国荷斯坦奶牛为研究对象, 以k-酪蛋白基因为产奶性状的候选基因, 扩增779 bp的片段, 结合测序结果采用PCR-RFLP方法来检测k-酪蛋白基因3个位点的多态性。结果在exon 4的第10 891 bp、10 927 bp和10 988 bp处分别发生了T/C、C/A错义突变和G/A同义突变, 据此分别选择了TaqⅠ、HindⅢ、 PstⅠ等 3种限制性内切酶检测了其多态性。发现3个位点的A、B等位基因在群体中都有分布, 且处于低度多态; A 和B 等位基因的频率分别为86.03%和13.97%; AA, AB和BB基因型频率分别为73.71%, 24.63%和1.66%; c2适合性检验表明, 该群体在这3个位点的突变达到Hardy-Weinberg平衡状态(P > 0.05); BB和AB基因型个体乳脂率显著高于AA基因型个体(P<0.05), AB基因型个体脂蛋白比显著高于AA基因型个体(P < 0.05), 但不同基因型对产奶量和乳蛋白率没有显著影响; 3个位点的酶切多态性在所研究群体中是紧密连锁的。说明在中国荷斯坦奶牛群体中, κ-酪蛋白B等位基因可作为改良奶牛乳脂率性状的分子遗传标记。  相似文献

DNA polymorphism of the ovine prolactin receptor gene (PRLR) was investigated and used to study its effect on litter size in sheep. By means of PRLR gene sequence homology between sheep and human, three primer pairs were designed for polymerase chain reaction (PCR) amplification within intron 1 and exon 10 of the PRLR gene in sheep. In these parts of the gene the single nucleotide polymorphisms were detected by PCR-single strand conformation polymorphism (SSCP) in 314 Small Tail Han ewes. These poly-morphisms were used to study the associations with litter size. The results indicated that there were three genotypes (AA, AB and BB) detected by three primer pairs. For three primer pairs the frequency of allele A was 0.96, 0.79, 0.68; and the frequency of allele B was 0.04, 0.21, 0.32, respectively. The frequency of genotype AA was 0.93, 0.62, 0.51; the frequency of genotype AB was 0.06, 0.34, 0.34; the frequency of genotype BB was 0.01, 0.04, 0.15, respectively. The Small Tail Han ewes with genotype BB or AB had 0.64-0.76 or 0.44-0.54 more lambs than those with genotype AA, respectively. These results preliminarily showed that the prolactin receptor locus is either a major gene that influences the prolificacy in Small Tail Han sheep or is in close linkage with such a gene.  相似文献

The effect of estrogen receptor (ESR) genotype (two alleles, A and B) on litter size of 275 Large White x Meishan F2 crossbred gilts (73 AA, 126 AB and 76 BB gilts) was tested. In addition, for 63 of these gilts (18 AA, 24 AB, and 21 BB) the effect of ESR genotype on average placental traits at term was tested, since individual placental information was available for 88% of the 628 liveborn piglets. Without affecting average birth weight of the piglets, ESR genotype significantly affected litter size, i.e. AB gilts had larger litters than BB gilts (P < 0.05). Total number born was 11.38+/-0.38, 11.88+/-0.28, and 10.68+/-0.35, while number born alive was 10.45+/-0.39, 11.07+/-0.29, and 9.85+/-0.36 for AA, AB and BB gilts, respectively. Since the B allele in previous research was associated with largest litters, the hypothesis that ESR is a marker rather than the major gene itself is discussed. Average placental length, surface area, and weight including and excluding amnion were not affected by ESR genotype. However, placentae of AB gilts had a significantly lower number of areolae per placenta than BB gilts and had a lower number of areolae/cm2 placenta than AA and BB gilts. Number of areolae was 8945+/-663, 7240+/-619, and 9694+/-633, for AA, AB and BB gilts, respectively. Although the reason for the low number of areolae on placentae in AB gilts is not yet known, the results suggest that the ESR linked major gene for litter size might be involved in the development and activity of endometrial glands.  相似文献

We determined the polymorphism C242T of the aromatase gene (Cyp19) and its allelic frequency, as well as the effect of the variants on productive and reproductive traits in 71 purebred Santa Inês sheep, 13 purebred Brazilian Somali sheep, nine purebred Poll Dorset sheep, and 18 crossbred 1/2 Dorper sheep. The animals were genotyped using the PCR-RFLP technique. The influence of the animal's genotype on its performance or on the performance of its lambs was analyzed by the least square method. Another factor assessed was the importance of the animal's genotype in analysis models for quantitative breeding value estimates, and whether there were differences among the averages of breeding values of animals with different genotypes for this gene. In the sample studied, no AA individuals were observed; the AB and BB frequencies were 0.64 and 0.36, respectively. All Brazilian Somali sheep were of genotype BB. All 1/2 Dorper BB animals presented a lower age at first lambing, and the Santa Inês BB ewes presented a lower lambing interval. In these same genetic groups, AB ewes presented higher litter weight at weaning. This is evidence that BB ewes have a better reproductive performance phenotype, whereas AB ewes present a better maternal ability phenotype. However, in general, animals with genotype AB presented better average breeding values than those with genotype BB.  相似文献

A monoclonal antibody (mAb), sis 1, generated against human c-sis-encoded platelet-derived growth factor (PDGF) BB, was shown by enzyme-linked immunosorbent assay and Western blot (immunoblot) analysis to recognize human PDGF BB and human platelet PDGF AB but not the human PDGF AA. This monoclonal antibody potently inhibited PDGF receptor-binding and mitogenic activities of both human PDGF BB and PDGF AB but had no effect on PDGF AA. Finally, we demonstrated that an immunoaffinity-purified anti-c-sis peptide antibody (anti-V4) which also blocked binding of PDGF BB to its cognate receptor and competed with mAb sis 1 for binding to PDGF BB. All of these results suggest that mAb sis 1 recognizes an epitope of the c-sis gene product, PDGF BB, that spatially overlaps the V4 surface domain of PDGF BB, immunochemically localizing a region of PDGF BB critical for PDGF receptor binding and activation.  相似文献

利用PCR-SSCP和DNA测序方法对帕里牦牛、工布江达牦牛、类乌齐牦牛、康布牦牛、桑日牦牛、巴青牦牛、江达牦牛、斯布牦牛、嘉黎牦牛、桑桑牦牛、丁青牦牛等西藏11个牦牛类群共483头牦牛的ADD1基因(被认为是极可能影响肉质的候选基因)第2外显子序列进行遗传多态性分析。结果表明,66 bp处碱基T缺失和256 bp处A→G突变,共有AA、AB、BB 3种基因型;其中帕里牦牛只有AA基因型,江达牦牛只有AB基因型,康布牦牛、嘉黎牦牛、巴青牦牛、桑日牦牛、斯布牦牛均缺少BB基因型,丁青牦牛、类乌齐牦牛缺少AB型,均存在严重偏态;桑桑牦牛、工布江达牦牛中存在AA、AB、BB 3种基因型。除江达牦牛外,其它10个牦牛类群中AA为优势基因型,A基因为优势等位基因,分布较高。除帕里牦牛、巴青牦牛外,其它9个牦牛类群均处于中度多态(0.25相似文献
