芹菜体细胞胚胎发生及干化体细胞胚的研究   总被引:2,自引:0,他引:2  
以芹菜无菌苗为材料,在MS附加KT 0.5mg/L,ABA0.25mg/L,脯氨酸500mg/L,CH500mg/L的培养基上,采用静止与震荡交替进行的培养方式,可获得大量健壮的子叶期胚状体(体胚)。干燥可以提高贮茂后体胚的转换率,特别是在低温,高湿中缓慢干燥,并在干燥前用ABA预处理效果更好。通过干燥和不干燥体胚扫描电镜观察和电导值及脱氢酶的比较,发现干燥体胚有助于贮藏期间细胞结构及膜系统的保持  相似文献

植物细胞离体培养中的体细胞胚发生受多种内外因素的调控,其中激素对细胞分化,发育和形态建成起着关键的作用。大量研究表明,在激素的作用过程中,Ca^2 信号系统可能是重要的介导者之一。Ca^2 和CaM在植物合子胚和体细胞胚发生中都具有重要作用,其作用机理可能是植物激素通过Ca^2 第二信号系统直接或间接地调控基因表达而实现的。  相似文献

细胞信号转导主要是指胞间通讯的激素以及外界环境因子等作用于细胞表面(或胞内受体)后,如何跨膜传递形成胞内第二信使,以及其后的信息分子级联传递、诱导基因表达和引起生理反应的过程。植物体细胞胚发生的本质是基因的判别表达,因此细胞信号转导在此起关键作用。在植物组织培养过程中,外加的激素等因子通过细胞信号转导诱导基因差别表达,使体细胞转入胚胎发生过程,最终生长发育成完整植株。  相似文献

植物通过体细胞胚胎发生途径形成再生植株已是及其普遍的现象,这一发育途径为研究植物细胞的分化、发育、全能性表达和作物品种改良、突变体筛选等提供了良好的实验体系,在理论上和应用中都具重大意义.体细胞分化为胚性细胞是受细胞内外多种因子所调控[1, 2],其中最重要的是受特定基因的调控,在胚性细胞的分化过程中伴随着特定的遗传信息表达,其分化过程的实质是基因按顺序表达调控,是相应基因产物作为胚性细胞形成的分子基础.本文结合我们的工作来探讨体细胞胚发生分子基础研究现状.  相似文献

对根芹的不同外植体在附加不同成分的MS培养基上进行离体培养,以诱导适合于制作人工种子用的高质量体细胞胚。下胚轴、子叶和叶片在含0.5mg/LKT,0.5mg/L2,4 一 D的MSO培养基上诱导与继代胚性愈伤组织,然后转移到附加有100mg/L肌醇、2g/L葡萄糖的MSO无激素培养基上悬浮培养产生体细胞胚,获得了比固体培养基及含有KT的液体培养基中产生的体细胞胚形态发育更正常的大量体细胞胚,这为人工种子制作奠定了基础。  相似文献

The effects of desiccation of Hevea somatic embryos and of sucroseand ABA concentrations in the maturation medium on their germinabilitywere investigated. Conversion into plant, water and histochemicalstatus of somatic embryos were compared systematically to thoseof the zygotic embryos used as reference. Slow desiccation ormaturation on 351 mol m–3 sucrose supplemented with 1mmol m–3 ABA strongly improved germinability and conversionof embryos into plants. The combination of the two treatmentswas the most effective, increasing the germination frequencyby 3·7 and plant conversion by 6·6 in clone PR107. Each of these two treatments increased the vigour of somaticembryos, stimulated the formation of root and shoot meristemsand the synthesis and accumulation of starch and protein reserves.At the end of maturation, the Hevea somatic embryos bore ananatomical and histochemical resemblance to mature zygotic embryos.Likewise, the two treatments brought the water status of somaticembryos closer to that of the mature zygotic embryos, but withoutachieving a perfect match. Although somatic embryogenesis has been observed in tissuesfrom a limited number of Eucalyptus species cultured in vitro,no comparisons have been made of the morphology and structureof eucalypt somatic embryos and zygotic embryos found in matureseeds. We used scanning and transmission electron microscopy,in conjunction with histological analysis, to compare maturezygotic embryos with somatic embryos of the commercially-importanttemperate eucalypt Eucalyptus nitens. Apart from differencesin the nature of the outer coating enclosing both embryo types,somatic embryos of E. nitens were observed to have strong similaritieswith zygotic embryos in seeds in terms of their overall size,morphology and internal cellular organization. Many cells inboth sexually-produced and somatic embryos contained numerouslipid-rich globular bodies. The wider significance of theseobservations is discussed with regard to their potential applicationsin eucalypt plantation biotechnology programmes. The effect of abscisic acid (ABA), non-permeating osmoticumand desiccation treatment on storage protein synthesis duringmaturation of somatic embryos of Picea glauca (Moench) Voss.was examined. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis(SDS-PAGE) and Western blot analysis demonstrated that someof the major crystalloid and matrix polypeptides were absentfrom somatic embryos maturing on medium containing ABA and lowosmoticum. However, treatment with polyethylene glycol-4000(PEG) in combination with ABA resulted in the synthesis of aspectrum of storage polypeptides resembling that of mature zygoticembryos. These storage proteins accumulated throughout an 8-weekculture period, resulting in a threefold higher protein contentthan somatic embryos maturing for the same time in the absenceof PEG. The structure and distribution of protein bodies incells of these osmotically treated somatic embryos was similarto that in cells of mature zygotic embryos. Treatment with 5·0-7·5%PEG prevented catabolism of the accumulated storage polypeptidesduring desiccation. The optimal culture conditions for somaticembryo maturation and storage protein deposition was 16 µMABA and 7·5% PEG for 8 weeks followed by desiccation.Analysis of mRNAs by in vitro translation and immunoprecipitationof translated products showed that the crystalloid protein mRNAprofiles of zygotic and those of somatic embryos maturing on16 µM ABA in the absence of PEG were similar. The differencesobserved in the pattern of accumulated polypeptides in thesesomatic embryos and those of mature zygotic embryos, therefore,indicates that storage-protein synthesis in response to osmoticumis in part regulated at the translational level. 以极东锦鸡儿未成熟合子胚子叶为外植体进行其体细胞胚胎发生和植株再生研究。在添加不同BA与NAA或2,4-D,外加500mg·L~(-1)水解酪蛋白、30g·L~(-1)蔗糖和8g·L~(-1)琼脂的MS培养基上诱导产生了体细胞胚。在5mg·L~(-1)NAA+5mg·L~(-1)BA和5mg·L~(-1)2,4-D+1mg·L~(-1)BA处理中体胚诱导率分别为14%和10%;NAA处理每外植体上诱导出的体胚数量最多为4.3个,而2,4-D为10.5个。体细胞胚经成熟培养后,在添加0.01mg·L~(-1)NAA、20g·L~(-1)蔗糖和6g·L~(-1)琼脂的MS培养基上萌发率达到58.94%。萌发的体胚在MS培养基上长成正常小植株,再生率为87%。经炼苗后的体胚苗移植到草炭土:蛭石:珍珠岩=5:4:1(V/V/V)的栽培基质中,可以正常生长,移栽成活率为40%。  相似文献

尽管在多种哺乳动物种系中成功制备了体细胞克隆后代,但当前的克隆技术仍有许多亟待解决的问题。体细胞核移植胚胎大多存在许多发育异常,造成了妊娠早期高流产率和出生后高死亡率。有研究认为,克隆胚胎发育障碍的一个重要的原因是供体细胞的遗传重编程不完全。哺乳动物种系中,DNA甲基化是胚胎发育期转录调节的必需步骤,除了单拷贝基因序列外,在基因组很多的区域都可以观测到克隆胚胎的异常甲基化。此外,克隆胚胎的基因印迹也存在异常。  相似文献

对离体玉米胚脱水耐性的变化以及不同脱水速率对其脱水耐性的影响进行了研究。授粉后16d的玉米胚能耐轻微脱水,含水量从1.45降低到0.28gH2Og-1DW时胚的萌发率为100%,但含水量低于0.1gH2Og-1DW时胚死亡。胚的脱水耐性随着发育逐渐加强,表现为电解质渗漏速率逐渐降低,萌发率和幼苗干重逐渐增加。授粉后20d胚内超氧化物歧化酶(SOD)和抗坏血酸过氧化物酶(APX)活性较高,过氧化氢酶(CAT)活性较低;授粉后24d,这些酶的活性与授粉后20d的正好相反。脂质过氧化产物丙二醛(MDA)在种子发育过程中呈下降趋势。不同脱水速率明显地影响胚的脱水耐性:在慢速脱水到含水量0.1~0.18gH2Og-1DW时,胚的萌发率和幼苗干重比快速脱水高,电解质渗漏速率比快速脱水低;在快速脱水条件下胚中的SOD、APX活性和MDA含量也比慢速脱水高;CAT活性的变化不明显。  相似文献

Responses of Peanut Somatic Embryos to Thidiazuron   总被引:1,自引:0,他引:1  
Induction of both somatic embryogenesis and organogenesis in presence of thidiazuron is reported in peanut tissues. However the histological evidence of thidiazuron induced somatic embryogenesis was unclear. Thidiazuron triggered multiple shoot differentiation in the plumule of the embryos. Keeping in view the ability of thidiazuron to induce both organogenesis and embryogenesis in peanut tissues, experiments were conducted to define the pathway of morphogenesis in the plumule of rooted somatic embryos. On exposure to thidiazuron, projections appeared from the plumule. These projections closely resemble the somatic embryos. However histological examination revealed that these are caulogenic buds and not somatic embryos. In concurrence with the earlier reports on thidiazuron induced organogenesis it is concluded that this growth regulator induces organogenic response in peanut tissues. Extracellular compounds isolated from embryogenic carrot cell suspension cultures increase, by 1.5 to 6-fold, end-stage embryo production when added back to carrot cultures initiating embryogenesis. The causative factors related to the enhancement of embryo production are most likely to be extracellular, high molecular weight proteins found in the embryo-free medium (EFM) after somatic embryos have been formed. The addition of heat-treated EFM to fresh cultures did not result in enhancing effects on the production of end-stage embryos. However, the addition of compounds precipitated from EFM, by high concentrations of salt, accelerated by four days the formation of comparable amounts of end-stage embryos and surpassed total end-stage embryo levels by a factor of 4-6, dependent on the precipitate dose. These results suggest that heat-labile polypeptide molecules may be responsible for growth factor-like effects during somatic embryogenesis.  相似文献
