Qualitative and quantitative characteristics of mega- and microspores from all the cytotypes of JapaneseIsoetes are described based on the voucher specimens whose chromosome numbers were known. InI. japonica, the hexaploid possessed reticulate megaspores and levigate microspores, while the octaploid and the heptaploid had echinate microspores. Mega- and microspores of the hexaploid and the octaploid were of normal appearance, while those of the heptaploid displayed polymorphism. The tetraploid and the hexaploid ofI. sinensis resembled each other, since they both possessed cristate megaspores and echinate microspores. Echinate megaspores and levigate microspores characterized the diploidI. asiatica. The spore size was largely variable within each cytotype, while the size of the megaspores varied more than that of the microspores. The microspore length was closely correlated with polyploid level. InI. sinensis, the mean microspore length of the tetraploid was 27.6 μm while that of the hexaploid was 31.9 μm, hence these two cytotypes were easily distinguishable. In the hexaploidI. japonica, variations in mega-and microspore size displayed geoclinal variation showing a positive correlation (r=0.43–0.55) with the longitude and the latitude of the populations. We name and describe two new species,Isoetes jejuensis and I.hallasanensis, from Jeju Island, South Korea. These taxa are compared with related species in East Asia, including I.coreana andI. sinensis. The rugulate megaspore ornamentation ofI. jejuensis differs from those otherIsoetes species. Furthermore,I. hallasanensis varies fromI. asiatica in its microspore ornamentation and chromosome number, although both species have an echinate megaspore ornamentation. These new species are tetraploid (2n = 44), whereasI. coreana is hexaploid (2n = 66).  相似文献

The somatic chromosome number of three Japanese species ofIsoetes, I. asiatica, I. japonica andI. sinensis, was determined in 199 individuals from 49 populations. The chromosome number ofI. asiatica was 2n=22, confirming previous reports. However,I. japonica andI. sinensis displayed a diversity in chromosome number. Six cytotypes, 2n=66, 67, 77, 87, 88 and 89, were found inI. japonica; 2n=67, 87, 88 and 89 are new counts in the genusIsoetes. The plants with 2n=66 were the most frequent (72% of total individuals examined) and were distributed throughout Honshu and Shikoku. The plants with 2n=88 occurred in western Honshu and a limited region in northeastern Honshu where the plants with 2n=77 were also found. In contrast, four cytotypes, 2n=44, 65, 66 and 68, were found inI. sinensis. The chromosome numbers ofI. sinensis were reported here for the first time. The plants with 2n=44 occurred only in Kyushu, while the plants with 2n=66 were found throughout a large area of western Japan.  相似文献

Meiotic chromosome behaviour was studied in the hexaploid Hordeum parodii (2n=6x=42) and in six haploids (2n=3x=21) obtained from a cross between H. parodii and H. bulbosum (2n=2x=14) whereby all bulbosum chromosomes were selectively eliminated. The alloploid nature of H. parodii was evident from the exclusive bivalent formation at the hexaploid level and the low and variable number of bivalents in its haploid derivatives. In haploids, both nonhomologous (intragenomic) and homoeologous (intergenomic) chromosomes paired at prophase. Foldbacks in single chromosomes, bivalents and trivalents were observed at prophase and metaphase I. At diakinesis, the associations involved a maximum of 20 chromosomes which decreased to 12 by metaphase I. This decrease was attributed to the failure of the non-homologous associations to persist until metaphase I. A hemizygous-ineffective control for the diploid-like behaviour of the hexaploid parodii is proposed to explain the homeologous chromosome pairing in its haploid derivatives.  相似文献

Abstract

The anthers are tetrasporangiate. The anther wall comprises epidermis, fibrous endothecium, middle layer and tapetal layer. The tapetum is of the Glandular type and its cells remain uninucleate. Meiosis in pollen mother cells is normal and simultaneous cytokinesis leads to the formation of tetrahedral and decussate microspore tetrads. The pollen grains are shed at 2-celled stage. The ovule is campylotropous, bitegmic and crassinucellate. Meiosis in megaspore mother cell results in the formation of linear or occasionally T-shaped megaspore tetrad. The chalazal megaspore develops into Monosporic Polygonum type of embryo sac. Endosperm development is of the Nuclear type.  相似文献

水韭属(Isoëtes)是起源最为古老的水生维管植物,全属物种均被列为国家一级重点保护植物。通过对全国水韭属植物的调查和研究发现,不同产地的四倍体植株在形态上存在显著差异。基于形态学、孢粉学和细胞学证据,将分布于中国湖南省长沙地区和怀化地区的四倍体居群分别命名为隆平水韭(Isoëtes longpingii)和湘妃水韭(I. xiangfei),并详细描述了其形态特征。隆平水韭形态上与中华水韭(I. sinensis)相似,不同之处在于其大孢子具小的瘤状或冠状纹饰,叶细长而柔弱,长达60 cm; 该种也与六倍体东方水韭(I. orientalis)相似,不同之处在于其染色体44条,大孢子具瘤状或冠状纹饰。湘妃水韭的大孢子纹饰虽与二倍体云贵水韭(I. yunguiensis)相似,但在小孢子纹饰、孢子囊形状和染色体数目方面却不同。隆平水韭仅少数植株生长于湖南省宁乡市一处池塘,完全沉水生长,而湘妃水韭则分布于怀化市通道县和会同县的湿地。由于这两个新种的分布区狭窄,野生居群数量和个体数较少,栖息地环境受到人为干扰,因此根据IUCN红色名录评估标准,将隆平水韭评为极危(CR)等级,湘妃水韭评为易危(VU)等级。所编制的中国已知水韭属物种的分种检索表,为本属物种的鉴定和保护工作提供了重要参考。  相似文献

S. integrifolium (2n = 24) can easily be crossed as the pistillate parent with S. melongena (2n = 24) and S. melongena var. insanum (2n = 24). However, crosses in the other direction do not succeed. Both hybrids are vigorous. Chromosome association at diakinesis and metaphase I was studied. Chromosome associations higher than bivalents were observed in the hybrids indicating structural repatterning of chromosomes. The modal chromosome association in hybrids was twelve bivalents per PMC. This is suggestive of the retention of ancestral chromosome homeologies by the taxa concerned. Despite regular meiosis both hybrids were highly pollen-sterile (about 95%), which was attributed to segregational events of the recombined chromosomes.  相似文献

本文对小花鬼针草的体细胞染色体进行了观察和计数,并对其染色体倍性和有关的细胞学及分类学等问题略作讨论,旨在为药用植物的分类、品种鉴别和育种实践等工作提供必要的细胞学资料。笔者观察到的小花鬼针草的体细胞染色体除了2n=48国外已有报道外,其中2n=24,36,72,84和96的二倍体、三倍体、六倍体、七倍体和八倍体等多倍体系列均为首次报道,此外2n=22,26,42和47等体细胞染色体的非整倍性变异,迄今也尚未见有报道。  相似文献

New Brassica napus inbred lines with different petal colors and with canola quality and increased levels of oleic (∼70%, 10% higher than that of B. napus parent) and linoleic (28%) acids have been developed in the progenies of one B. napus cv. Oro × Orychophragmus violaceus F5 hybrid plant (2n=31). Their genetic constituents were analyzed by using the methods of genomic in situ hybridization (GISH) and amplified fragments length polymorphism (AFLP). No intact chromosomes of O. violaceus origin were detected by GISH in their somatic cells of ovaries and root tips (2n=38) and pollen mother cells (PMCs) with normal chromosome pairing (19 bivalents) and segregation (19:19), though signals of variable sizes and intensities were located mainly at terminal and centromeric parts of some mitotic chromosomes and meiotic bivalents at diakinesis or chromosomes in anaphase I groups and one large patch of chromatin was intensively labeled and separated spatially in some telophase I nuclei and metaphase II PMCs. AFLP analysis revealed that substantial genomic changes have occurred in these lines and O. violaceus–specific bands, deleted bands in ‘Oro’ and novel bands for two parents were detected. Intertribal somatic hybrids of Raphanus sativus (2n = 18, RR) and Brassica rapa spp. chinensis (2n = 20, AA) with the dye and medicinal plant Isatis indigotica (2n = 14, I I) were firstly obtained by polyethylene glycol-induced symmetric fusions of mesophyll protoplasts. One mature hybrid with R. sativus established in field had intermediate morphology but was totally sterile. It had the expected chromosome number (2n = 32, RRI I) and parental chromosomes were distinguished by genomic in situ hybridization (GISH) analysis, and these chromosomes were paired as 16 bivalents in pollen mother cells (PMCs) at diakinesis and mainly segregated equally as 16:16 at anaphase I (A I), but the meiotic disturbance in second division was obvious. Five mature hybrids with B. rapa established in field were morphologically intermediate but showed some differences in phenotypic traits and fertility, two were partially fertile. Cytological and GISH investigations revealed that these hybrids had 2n = 48 with AAIIII complement and their PMCs showed normal pairing of 24 bivalents and mainly equal segregation 24:24, but meiotic abnormalities of lagging chromosomes and micronuclei appeared frequently during second divisions. AFLP analysis showed that all of these hybrids had mainly the DNA banding pattern from the addition of two parents plus some alterations. Some hybrids should be used for the genetic improvement of crops and the dye and medicinal plant.  相似文献

While many studies have provided significant insight into homolog pairing during meiosis, information on non-homologous pairing is much less abundant. In the present study, fluorescence in situ hybridization (FISH) was used to investigate non-homologous pairing in haploid rice during meiosis. At pachytene, non-homologous chromosomes paired and formed synaptonemal complexes. FISH analysis data indicated that chromosome pairing could be grouped into three major types: (1) single chromosome paired fold-back as the univalent structure, (2) two non-homologous chromosomes paired as the bivalent structure, and (3) three or more non-homologous chromosomes paired as the multivalent structure. In the survey of 70 cells, 65 contained univalents, 45 contained bivalents, and 49 contained multivalent. Moreover, chromosomes 9 and 10 as well as chromosomes 11 and 12 formed non-homologous bivalents at a higher frequency than the other chromosomes. However, chiasma was always detected in the bivalent only between chromosomes 11 and 12 at diakinesis or metaphase I, indicating the pairing between these two chromosomes leads non-homologous recombination during meiosis. The synaptonemal complex formation between non-homologs was further proved by immunodetection of RCE8, PAIR2, and ZEP1. Especially, ZEP1 only loaded onto the paired chromosomes other than the un-paired chromosomes at pachytene in haploid.  相似文献

Summary Triploids (2n=3X=60) were obtained from genetic male-sterile (ms1 ms1) soybean [Glycine max (L.) Merr.] plants. Meiosis, pollen fertility, and chromosome number of their progeny were studied. Studies of meiosis in fertile and sterile triploids revealed no distinguishable differences in chromosome associations. Male-sterile plants formed coenocytic microspores characteristic of the ms1 mutant. Restitution of some dyad and tetrad nuclei were observed in male-sterile plants. Chromosomes of the triploids tended to occur in trivalents during diakinesis and metaphase I (MI), but multivalents, bivalents, and univalents also were observed. Average types and frequencies of chromosome associations per cell in diakinesis and MI from 542 pollen mother cells were 0.004 IX + 0.06 VI + 0.002 V + 0.005 IV + 16.99 III + 1.79 II + 5.03 I. Some secondary associations, nonhomologous pairing, and aberrant nucleolar distributions occasionally were observed. Such behavior support the hypothesis of duplicated genomes and the polyploid origin of soybean. Pollen fertility in male-fertile triploid plants (Ms1 ms1 ms1) varied from 57% to 82%, with an average of about 71%. Chromosome numbers of progenies obtained from these fertile triploids varied from 2n=40 to 2n=71, and exhibited a near-random distribution, with the majority (about 60%) being between 56 and 65. Studies on chromosome numbers and karyotypes in Orchid taxa from Apulia (Italy) revealed triploid complements inOphrys tenthredinifera andOrchis italica. InO. tenthredinifera there is no significant difference between the diploid and the triploid karyotypes. The tetraploid cytotype ofAnacamptis pyramidalis forms 36 bivalents during metaphase I in embryo sac mother cells. Aneuploidy was noticed inOphrys bertolonii ×O. tarentina with chromosome numbers n = 19 and 2n = 38. There were diploid (2n = 2x = 36), tetraploid (2n = 4x = 72), hexaploid (2n = 6x = 108) and octoploid (2n = 8x = 144) cells in the ovary wall of the diploid hybridOphrys apulica ×O. bombyliflora. Evolutionary trends inOphrys andOrchis chromosomes are discussed.  相似文献

以不同发育时期的凤仙花花药为实验材料,采用组织化学方法,对花药发育中的结构变化及多糖和脂滴物质分布进行观察。结果表明:(1)凤仙花的花药壁由6层细胞组成,包括1层表皮细胞,2层药室内壁细胞,2层中层细胞和1层绒毡层细胞。其中绒毡层细胞的形态不明显,很难与造孢细胞区分,且在小孢子母细胞时期退化。(2)在小孢子母细胞中出现了一些淀粉粒,但减数分裂后,早期小孢子中的淀粉粒消失,又出现了一些小的脂滴;随着花粉的发育,小孢子形成大液泡,晚期小孢子中的脂滴也消失;小孢子分裂形成二胞花粉后,营养细胞中的大液泡降解、消失,二胞花粉中又开始积累淀粉;接近开花时,成熟花粉中充满细胞质,其中包含了较多的淀粉粒和脂滴。(3)在凤仙花的花药发育中,绒毡层细胞很早退化,为小孢子母细胞和四分体小孢子提供了营养物质;其后的中层细胞退化则为后期花粉发育提供了营养物质。  相似文献

为探讨小叶丁香在北京栽培条件下种子败育的问题,对其胚胎发育过程进行了解剖学观察研究。结果显示:(1)小叶丁香人工异花授粉坐果率为20.37%,天然授粉坐果率为9.13%,人工自花授粉以及去雄套袋授粉均未坐果;各种授粉方式结实率均为0。(2)小叶丁香花药壁发育为基本型,腺质绒毡层,胞质分裂为同时型,小孢子四分体排列方式以四面体型为主,2-胞成熟花粉;倒生胚珠,单珠被,薄珠心,直线型大孢子四分体,胚囊发育为蓼型,核型胚乳,胚发育为紫菀型,先后形成球形胚、心形胚、鱼雷形胚和子叶胚;9月1日以后,子叶胚发育停滞,细胞逐渐降解。研究表明,栽培条件下,小叶丁香为异花授粉植物,人工异花授粉可提高植株坐果率,但未获得种子;其大小孢子及雌雄配子体发育、传粉受精过程、胚胎发育初期均正常,但后期发育停止,是小叶丁香引种栽培条件下种子败育的关键环节。  相似文献

采用石蜡切片技术对百合科植物山麦冬大小孢子发生及雌雄配子体发育进行了观察研究。结果表明:(1)山麦冬花药具有4个花粉囊,花药壁的发育方式为基本型,花药壁完全分化时由表皮、药室内壁、中层及绒毡层组成。(2)绒毡层发育类型为分泌型,到四分体孢子彼此分离形成单细胞花粉阶段,绒毡层细胞开始解体退化,花粉成熟时绒毡层细胞完全消失;花粉母细胞减数分裂为连续型,四分体为左右对称形排列,成熟花粉为3-细胞花粉,单萌发沟。(3)子房3室,每室2枚胚珠,胚珠倒生型,双珠被,薄珠心,雌性孢原细胞不经过平周分裂而直接发育而成大孢子母细胞。(4)减数分裂后四分体大孢子呈线型或T型排列,合点端大孢子分化为功能大孢子,胚囊发育为蓼型;花粉母细胞减数分裂过程中,二分体、四分体细胞外方被胼胝质壁所包被,小孢子形成后胼胝质壁逐渐消失。该研究结果丰富了百合科植物生殖生物学研究的内容,也为探讨百合科植物的系统学研究提供了参考。  相似文献

Pachytene chromosomes of diploid (n=10), hexaploid (n=30) and heptaploid (n=35) races of Apluda mutica L. are similar in their diagnostic morphological features. Hexaploids and heptaploids possess the same ten basic types of chromosomes characteristic of the diploid in multiples of six and seven respectively. Quadrivalent associations involving four homologous chromosomes were observed for all ten types of chromosomes of the hexaploid and heptaploid. It is suggested that the polyploid races may be autoploids.This work forms part of a thesis submitted to the Andhra University, Waltair for the Degree of Doctor of Philosophy.  相似文献

The chromosome morphology and meiotic pairing behavior in the pollen mother cells (PMCs) of Metasequoia glyptostroboides were investigated. The results showed that: (1) The chromosome number of the PMCs was 2n=22. (2) The PMCs developed in the successive manner, and the nucleoids in the dynamic development were similar to those of the other gymnosperms. (3) At prophase, most of the chromosomes were unable to be identified distinctively because the chromosomes were long and tangled together. The chromosome segments were paired non-synchronously. At pachytene, the interstitial or terminal regions of some bivalents did not form synapsis and the paired chromosomes showed difference in sizes, indicating that there were structure differences between the homologous chromosomes. (4) At diakinesis, the ring bivalents showed complicated configurations due to the differences in location and number of chiasmata. In addition, there were cross-linked bivalents. (5) At metaphase I, the chromosome configuration of each cell was 8.2II 0 + 1.1II + 1.3II + + 0.8I. Most of the chromosomes were ring bivalents, but some were cross-linked bivalents, rod bivalents, or univalents. (6) 15\% PMCs at anaphase I and 22\% PMCs at anaphase II presented chromosome bridges, chromosome fragments, micronuclei, and lagging chromosomes. Twenty seven percent microspores finally moved into one to three micronuclei. Twenty five percent pollens were abortive. The results indicated that the observed individual of M. glyptostroboideswas probably a parpcentric inversion heterozygote, and there were structural and behavioral differences between the homologous chromosomes. The chromosomal aberration of M. glyptostroboidesmay play an important role in the evolution of this relict species, which is known as a living fossil. Further evidence is needed to test whether the differences between homologous chromosomes were due to hybridization.  相似文献

采用常规压片法和去壁低渗法,以生长于广西凭祥和广东深圳的广东万年青(Aglaonema modestumSchott ex Engl.)为材料,对二者的体细胞染色体、花粉母细胞减数分裂染色体配对行为和花粉发育过程进行了观察。结果表明:(1)野生植株为二倍体2n=40,栽培植株为三倍体2n=60;(2)野生种的小孢子母细胞减数分裂前期I终变期均为二价联会,栽培种偶见三价联会;(3)中期I,野生种为20个二价体排列在赤道板上,未见单价体,栽培种的20个二价体排列于赤道板上,20个单价体随机分布于两极,证实其为三倍体;(4)后期I,野生种二价体分离,出现单染色单体桥和断片,几率为10%,栽培种几率为3%,还存在落后染色体,部分不能进入两极的落后染色体和染色体断片在末期I形成微核;(5)四分体时期,野生种未观察到异常孢子,栽培种出现大量败育的四分体或多分体;(6)小孢子进入正常的发育分化,通过两次有丝分裂形成三细胞型花粉。野生种成熟花粉败育率为2.18%,栽培种为88.29%;(7)野生种正常结实,栽培种果实中未发现种子,为高度不育。  相似文献

Meiosis and mitosis of eight species of Polygonatum from Sichuan, China, were investigated. Ten bivalents (n=10) at meiosis of pollen mother cells were observed ih P. odoratum (Mill.) Druce. Meiotic observation on pollen mother cells in P. punctatum Royle ex Kunth shows 16 bivalents (n=16) at diakinesis, P. kingianum Coll. et Hemsl. 13 bivalents (n=13) at diakinesis. P. zanlanscianense Pamp. 15 bivalents (n=15) in MI, P. cirrhifolium (Wall.) Royle 28 bivalents at diakinesis. Somatic chromosomes were observed in root tip cells. The karyotype formulae are as follows: P. odoratum (Mill.) Druce K(2n)=20=4st+6sm+10m; P. cyrtonema Hua K(2n)=20=6sm+14m; P. punctatum Royle ex Kunth K(2n)=32=2t+8st+ 2sm+20m; P. kingianum Coll. et Hemsl. K(2n)=26=8st(2SAT)+14sm+4m; P. alternicirrhosum Hand.-Mzt. K(2n)=32=6st+8sm+18m(2SAT); P. zanlanscianense Pamp. K(2n)=30= 2t+6st+6sm+16m(2SAT); P. sibiricum Delar. ex Redouté K(2n)=24=2t+14st(2SAT)+6sm +2m; P. cirrhifolium (Wall.) Royle K(2n)=56=18st+10sm+28m. The karyotypes of P. punctatum, P. kingianum, P. alternicirrhosum and P. cirrhifolium are reported for the first time. Both the chromosome number and structure in our materials are certainly different from thoses in previous reports. Of numerical variation polyploidy and aneuploidy have been recorded, and aneuploidy is frequent. According to degree of asymmetry of the karyotype, the 8 species are divided into three types: 2B, 3B, 2C. The increasing asymmetry is correlated with the increasing of the chromosome number. There are no obvious differences in chromosome number and karyotype between Alternifolia Baker and Verticillate Baker. They are not two natural groups. This opinion is different from that of Therman and Suomalainen.  相似文献
