To assess the evolutionary significance of persistent seed banks, phenotypes of naturally germinating seeds must be compared with those that remain dormant under the same environmental conditions. Dormant seeds can often be induced to germinate by application of gibberellic acid (GA). However, this method is valid only if there are no phenotypic “side effects” of GA that could confound comparisons between dormant and naturally germinating seeds. We examined this assumption in Lesquerella fendleri, a short-lived perennial mustard of the desert Southwest. We exposed 3840 seeds from 16 maternal sibships to two different GA treatments (0 or 1 g/L GA) in two different germination environments (greenhouse and growth chamber), and measured germination and postgermination traits. As expected, application of GA increased germination. GA also had strong and long-lasting effects on seedling morphology. Seeds that received GA developed into seedlings that were taller, with fewer but longer leaves, than seeds that did not receive GA. Effects of GA on both dormancy and postgermination traits varied among maternal sibships. Our results indicate that for this species and this concentration of GA, morphological effects can be substantial. Further study is required to determine whether such side effects are found for lower concentrations of GA, or under conditions that encourage faster seedling growth. Nonetheless, the present results illustrate the importance of testing potential confounding effects of GA in studies of the evolution of seed dormancy and its influence on postgermination traits.  相似文献

BACKGROUND AND AIMS: The European Chaerophyllum temulum and two North American Chaerophyllum species have a trans-Atlantic disjunct distribution. This work aimed to resolve requirements for dormancy break and germination of C. temulum seeds and to compare dormancy traits with those of the two North American congeners. METHODS: Phenology of germination and embryo growth was studied by regularly exhuming seeds sown in natural conditions. Temperature requirements for embryo growth, breaking of dormancy and germination were determined by incubating seeds under controlled laboratory conditions. Additionally the effect of GA(3) on germination was tested to determine the specific dormancy type. KEY RESULTS: In natural conditions, embryo growth starts in early winter. Seedlings emerge in late winter shortly after the embryos reached the critical ratio for embryo length to seed length (E : S) of approx. 0.95. Growth of the embryo only occurs during a prolonged incubation period at 5 degrees C. After stratification at 5 degrees C, which breaks physiological and morphological dormancy, seeds can germinate at a wide range of temperatures. GA(3) did not substitute for cold stratification in seeds placed at 23 degrees C. CONCLUSIONS: Chaerophyllum temulum has deep complex morphophysiological dormancy. This dormancy type differs considerably from that of the two North American congeners.  相似文献

Seed dormancy release and germination of Corispermum lehmannianum Bunge were tested using various treatments: temperature, cold stratification, gibberelins (GA3), dry storage and sand burial. Results showed that temperature and light did not affect the germination of fresh seeds, cold stratification and GA3 could improve seed germination, whereas dry storage and sand burial did not. The germination percentage was highest at 35/20 °C after the cold stratification and GA3 treatments. Corispermum lehmannianum seeds were classified as non-deep, Type-2, physiological dormancy (PD), whose seed dormancy could be released by cold stratification and GA3.  相似文献

Background and Aims

The duration of the plant life cycle is an important attribute that determines fitness and coexistence of weeds in arable fields. It depends on the timing of two key life-history traits: time from seed dispersal to germination and time from germination to flowering. These traits are components of the time to reproduction. Dormancy results in reduced and delayed germination, thus increasing time to reproduction. Genotypes in the arable seedbank predominantly have short time to flowering. Synergy between reduced seed dormancy and reduced flowering time would create stronger contrasts between genotypes, offering greater adaptation in-field. Therefore, we studied differences in seed dormancy between in-field flowering time genotypes of shepherd''s purse.

Methods

Genotypes with early, intermediate or late flowering time were grown in a glasshouse to provide seed stock for germination tests. Secondary dormancy was assessed by comparing germination before and after dark-incubation. Dormancy was characterized separately for seed myxospermy heteromorphs, observed in each genotype. Seed carbon and nitrogen content and seed mass were determined as indicators of seed filling and resource partitioning associated with dormancy.

Key Results

Although no differences were observed in primary dormancy, secondary dormancy was weaker among the seeds of early-flowering genotypes. On average, myxospermous seeds showed stronger secondary dormancy than non-myxospermous seeds in all genotypes. Seed filling was similar between the genotypes, but nitrogen partitioning was higher in early-flowering genotypes and in non-myxospermous seeds.

Conclusions

In shepherd''s purse, early flowering and reduced seed dormancy coincide and appear to be linked. The seed heteromorphism contributes to variation in dormancy. Three functional groups of seed dormancy were identified, varying in dormancy depth and nitrate response. One of these groups (FG-III) was distinct for early-flowering genotypes. The weaker secondary dormancy of early-flowering genotypes confers a selective advantage in arable fields.  相似文献

Seed is vital to the conservation of germplasm and plant biodiversity. Seed dormancy is an adaptive trait in numerous seed‐plant species, enabling plants to survive under stressful conditions. Seed dormancy is mainly controlled by abscisic acid (ABA) and gibberellin (GA) and can be classified as primary and secondary seed dormancy. The primary seed dormancy is induced by maternal ABA. Here we found that AtPER1, a seed‐specific peroxiredoxin, is involved in enhancing primary seed dormancy. Two loss‐of‐function atper1 mutants, atper1‐1 and atper1‐2, displayed suppressed primary seed dormancy accompanied with reduced ABA and increased GA contents in seeds. Furthermore, atper1 mutant seeds were insensitive to abiotic stresses during seed germination. The expression of several ABA catabolism genes (CYP707A1, CYP707A2, and CYP707A3) and GA biosynthesis genes (GA20ox1, GA20ox3, and KAO3) in atper1 mutant seeds was increased compared to wild‐type seeds. The suppressed primary seed dormancy of atper1‐1 was completely reduced by deletion of CYP707A genes. Furthermore, loss‐of‐function of AtPER1 cannot enhance the seed germination ratio of aba2‐1 or ga1‐t, suggesting that AtPER1‐enhanced primary seed dormancy is dependent on ABA and GA. Additionally, the level of reactive oxygen species (ROS) in atper1 mutant seeds was significantly higher than that in wild‐type seeds. Taken together, our results demonstrate that AtPER1 eliminates ROS to suppress ABA catabolism and GA biosynthesis, and thus improves the primary seed dormancy and make the seeds less sensitive to adverse environmental conditions.  相似文献

Dormancy in the hard seed coats of Mediterranean species is considered a strategy that enables persistent seed banks to be formed in the soil. An important factor related to seed coat fracture and dormancy breakage in Mediterranean ecosystems is heat. Nevertheless, the effect of factors other than heat on dormancy breakage in these species has hardly been studied. To investigate the different ecological factors involved in germination, in the laboratory we applied several scarification treatments to seeds with chromatic polymorphism. We evaluated the effect of soil seed depth during experimental burns by sowing seeds at −1, −3 and −5 cm in the soil profile, and we also studied the effect of seed origin on the posterior germination of seeds from 4 and 10 year-old shrubs as well as from the soil seed bank. U. parviflorus shows clear chromatic polymorphism: its brown seeds present higher dormancy levels than its yellow seeds. The different techniques of dormancy breakage result in different degrees of germination; the highest degree of germination is generated by the mechanical treatment, followed by the acid and the heat treatments, in that order. The depth of the seeds in the soil determines the temperature thresholds and the residence times of these temperatures and whether they stimulate a massive germination at the −1 cm soil profile or only a slight germination at the −5 cm depth. Seeds recently produced by the plant show higher dormancy levels than seeds extracted from soil seed banks. Dormancy levels also depend on the shrubland age used for extracting the soil samples (3>9 years old). In effect, from the point of view of dormancy, the germination behaviour of U. parviflorus seeds seems to follow a multiresponse strategy based on different seed populations and involving both biological and abiotic processes to break dormancy.  相似文献

Dormancy in Ambrosia artemisiifolia seeds was broken by 8 weeks of stratification. Germination of nondormant seeds was greater in light than in continuous darkness. Embryos of freshly harvested seeds were nondormant. Leaching and scarification did not stimulate germination of the dormant seeds. Exogenous gibberellin (GA3) slightly increased germination of intact dormant seeds, and the effect was greatly increased by scarification. Germination was greater in the light in both tests. Exogenous indoleacetic acid did not stimulate germination of dormant seeds. Endogenous gibberellin and auxin content increased during stratification, and there was also a significant increase in GA during post-stratification at a favorable germination temperature. Inhibitors in the dormant seeds decreased during stratification and post-stratification. The high concentration of chlorogenic acid present in dormant seeds increased slightly during stratification. An unknown phenol very similar to chlorogenic acid in fluorescence and U.V. absorption significantly increased after 2 weeks of stratification. A significant decrease in the concentration of a second unidentified phenol occurred after 2 weeks of stratification. It is proposed that dormancy in Ambrosia artemisiifolia may be controlled by an inhibitor-promoter complex. The dormant seed is characterized by high inhibitor and low promoter levels. In the nondormant seed the balance was shifted to favor the promoter. Evidence suggests that the inhibitor involved may be abscisic acid and the promoters may be gibberellin and auxin. The content of auxin may be partially controlled by the concentration of phenols.  相似文献

This research was performed to resolve temperature requirement for embryo growth, dormancy break and seed germination of Aconitum lycoctonum, an Eurasian perennial herb growing in deciduous forests. The dormancy strategy of A. lycoctonum was compared with that of other Ranunculaceae species growing in the temperate deciduous forest habitat. Seeds of A. lycoctonum germinate immediately after embryo growth is completed during winter and seedlings subsequently emerge in early spring. Experiments in controlled conditions revealed that (1) embryo growth and germination only occurred at low temperatures (<10 °C), (2) a high-temperature pre-treatment was not required for germination, and (3) application of gibberellic acid did not overcome the chilling requirement. Based on these results, seeds of A. lycoctonum can be classified as having deep complex morphophysiological dormancy. Dormancy breaking requirements of A. lycoctonum are very similar to related species studied before, suggesting stasis in seed dormancy traits has occurred in the Aconitum–Delphinium clade.  相似文献

Abstract Neotyphodium frequently occurs as an endophyte in grasses. Evidence shows enhanced fitness of endophyte infected grasses relative to non‐infected ones. Some studies of seed germination show endophyte enhancement of plant fitness in various environments, but inconsistent results indicate that further studies are needed. So far, experiments have failed to separate the confounded effects of population origin and seed management. For this reason, we evaluated the effects of endophyte infection on seed dormancy and germination in Lolium multiflorum using an experimental design controlling these factors. Depending on the year of seed production, endophyte infection modified seed response to light quality, affecting predominantly seed dormancy levels. Nevertheless, the endophyte did not affect base temperature or thermal time of germination. We concluded that endophytes were not a strong influence on germination behaviour. We speculate from our results that the presence of the endophyte changes germination by an indirect effect, in extending growth of the maternal plant during seed development and ripening. The direct effect of hyphae in the seed on seed behaviour was disregarded, because the difference between infected and non‐infected seed varied within the year of seed production. Future experiments should focus on effects of the endophyte on the canopy of parent plants during seed production and ripening, and, hence, on subsequent dormancy and germination of the seeds.  相似文献

Seed dormancy is thought to be a key mechanism allowing annual plants to spread extinction risk in unpredictably varying environments. Theory predicts increasing germination fractions with increasing probability of reproductive success but solid empirical evidence is scarce and often confounded with environmental factors. Here we provide an empirical test of bet‐hedging via delayed germination for three annual plant species along a ‘predictability gradient’ in Israel. We excluded confounding environmental and maternal effects by raising inbred seed families and germinating them under controlled conditions. Additionally, we germinated field‐collected seeds in three consecutive seasons to compare their germination with inbred families where maternal effects were removed. Risk of reproductive failure was quantified using demographic data from the field and from second‐generation inbred lines raised in a rainfall gradient in the greenhouse. Our findings were consistent with bet‐hedging theory in that germination fraction was negatively related to species‐ and site‐specific risk of reproductive failure. Both field and hand‐raised seeds of one species exhibited higher dormancy with increasing risk of reproductive failure across sites, and hand‐raised seeds of another species showed the same pattern. The third species exhibited a rather random pattern of germination between years and sites, corresponding to the lack of site‐specific risk of reproductive failure. Species‐specific patterns of dormancy and risk could be related to alternative risk‐spreading strategies such as high adult survival, but were also affected by phylogeny. We provide strong empirical evidence for seed dormancy being a mechanism to reduce the risk of reproductive failure in highly variable environments, but a larger number of rigorous experimental tests of bet hedging germination are needed. Specifically, the genetic basis of bet‐hedging must be shown in species with different life histories, for demonstrating that dormancy is adaptive and how it is modified by other risk‐spreading traits.  相似文献

Fluctuating temperature plays a critical role in determining the timing of seed germination in many plant species. However, the physiological and biochemical mechanisms underlying such a response have been paid little attention. The present study investigated the effect of plant growth regulators and cold stratification in regulating Leymus chinensis seed germination and dormancy response to temperature. Results showed that seed germination was less than 2 % at all constant temperatures while fluctuating temperature significantly increased germination percentage. The highest germination was 71 % at 20/30 °C. Removal of the embryo enclosing material of L. chinensis seed germinated to 74 %, and replaced the requirement for fluctuating temperature to germinate, by increasing embryo growth potential. Applications of GA4+7 significantly increased seed germination at constant temperature. Also, inhibition of GA biosynthesis significantly decreased seed germination at fluctuating temperatures depending upon paclobutrazol concentration. This implied GA was necessary for non-dormant seed germination and played an important role in regulating seed germination response to temperature. Inhibition of ABA biosynthesis during imbibition completely released seed dormancy at 20/30 °C, but showed no effect on seed germination at constant temperature, suggesting ABA biosynthesis was important for seed dormancy maintenance but may not involve in seed germination response to temperature. Cold stratification with water or GA3 induced seed into secondary dormancy, but this effect was reversed by exogenous FL, suggesting ABA biosynthesis during cold stratification was involved in secondary dormancy. Also, cold stratification with FL entirely replaced the requirement of fluctuating temperature for germination with seeds having 73 % germination at constant temperature. This appears to be attributed to inhibition of ABA biosynthesis and an increase of GA biosynthesis during cold stratification, leading to an increased embryo growth potential. We suggest that fluctuating temperature promotes seed germination by increasing embryo growth potential, mainly attributed to GA biosynthesis during imbibitions. ABA is important for seed dormancy maintenance and induction but showed less effect on non-dormant seed germination response to temperature.  相似文献

Seed dormancy is a common phase of the plant life cycle, and several parts of the seed can contribute to dormancy. Whole seeds, seeds lacking the testa, embryos, and isolated aleurone layers of Arabidopsis (Arabidopsis thaliana) were used in experiments designed to identify components of the Arabidopsis seed that contribute to seed dormancy and to learn more about how dormancy and germination are regulated in this species. The aleurone layer was found to be the primary determinant of seed dormancy. Embryos from dormant seeds, however, had a lesser growth potential than those from nondormant seeds. Arabidopsis aleurone cells were examined by light and electron microscopy, and cell ultrastructure was similar to that of cereal aleurone cells. Arabidopsis aleurone cells responded to nitric oxide (NO), gibberellin (GA), and abscisic acid, with NO being upstream of GA in a signaling pathway that leads to vacuolation of protein storage vacuoles and abscisic acid inhibiting vacuolation. Molecular changes that occurred in embryos and aleurone layers prior to germination were measured, and these data show that both the aleurone layer and the embryo expressed the NO-associated gene AtNOS1, but only the embryo expressed genes for the GA biosynthetic enzyme GA3 oxidase.  相似文献

The seed is an important organ in higher plants, it is an important organ for plant survival and species dispersion. The transition between seed dormancy and germination represents a critical stage in the plant life cycle and it is an important ecological and commercial trait. A dynamic balance of synthesis and catabolism of two antagonistic hormones, abscisic acid (ABA) and giberellins (GAs), controls the equilibrium between seed dormancy and germination. Embryonic ABA plays a central role in induction and maintenance of seed dormancy and also inhibits the transition from embryonic to germination growth. Therefore, the ABA metabolism must be highly regulated at both temporal and spatial levels during phase of dessication tolerance. On the other hand, the ABA levels do not depend exclusively on the seeds because sometimes it becomes a strong sink and imports it from the roots and rhizosphere through the xylem and/or phloem. These events are discussed in depth here. Likewise, the role of some recently characterized genes belonging to seeds of woody species and related to ABA signaling are also included. Finally, although four possible ABA receptors have been reported, not much is known about how they mediate ABA signaling transduction. However, new publications seem to show that almost all these receptors lack several properties to consider them as such.Key words: ABA/GA balance, ABA in woody plants, ABA-receptors, biosynthetic ABA mutants, rhizosphere ABA, seed dormancy

Hybridisation between crops and their wild relatives may promote the evolution of weeds. Seed germination and dormancy are the earliest life‐history traits and are highly influenced by the maternal parent. However, the ecological role of the maternal effect on seed traits in the evolution of crop–wild hybrids has received little attention. In this study, we test the relative importance of maternal and hybridisation effects on seed traits of the first generation of crop–wild sunflower hybrids (Helianthus annuus). Seed germination was tested in two wild populations with contrasting dormancy, two cultivated materials and their reciprocal crosses at four different times after harvest and three different temperatures. Seed germination at each of the four times, after ripening response and secondary dormancy were recorded along with four morphological traits. Additionally, the pericarp anatomy was analysed with light and scanning electron microscopy. We observed strong maternal effects on all seed traits. Seed germination, morphology and pericarp anatomy differed largely between the crop and wild seeds and these traits in the crop–wild hybrids resembled their female parent. Slight but significant hybridisation effects were observed in germination, mainly in seeds produced on wild plants. Crop hybridisation changed seed germination, the after ripening response and secondary dormancy in the crop direction. Morphological and anatomical traits associated with domestication strongly correlated with the observed differences in seed germination and dormancy in crop–wild sunflower hybrids. The large maternal effects along with the evolutionary divergence in seed traits were responsible for the large phenotypic differences observed in crop–wild hybrids with the same genetic composition. Wild‐like seed traits of hybrids suggest that there are no barriers to crop gene introgression at the seed level whereas crop‐like seed traits could be strongly selected against, conditioning the selection of traits expressed later in the life cycle and in the next generations.  相似文献

Aim of this study was to investigate the nature of dormancy in black henbane (Hyoscyamus niger) seeds which have low germination rate under normal laboratory conditions. To do this, before placing the seeds in Petri dishes, they were soaked in 5,10 and 15 mg/L GA; 1,2 and 3% H2SO4, 15 mg/L GA + 1% H2SO4, 0.01 M KNO3 solutions, tap water, 40, 50 and 60°C hot water for 30 min. The study was performed under both continuous illumination and darkness in growth chambers to evaluate the effect of light on germination rate. The results showed that H2SO4 and GA treatments were the most important factors affecting seed germination and their germination enhancing effects were more evident in darkness. The results also suggested that black henbane seeds exhibit double dormancy involving a hard seed coat and a partially dormant embryo and have a partial dark requirement to germinate.  相似文献
