Fourteen recombinant inbred lines of sunflower (Helianthus annuus L.) and their parents (PAC-2 and RHA-266) were tested for their organogenesis ability. Seeds were surface sterilized and germinated on hormone free half strength MS basal medium containing 10 g l-1 sucrose solidified with five different gelling agents: Phytagar (Gibco laboratoires) 3 g l-1, Phytagel (Sigma) 3 g l-1, Agarose (Sigma) 5 g l-1, Arcagel (Sigma) 4 g l-1 and Agar-Agar (Fisher France) 7 g l-1. Cotyledons from 2-day-old seedlings were split in half and the four explants of each seed were cultived in 55 mm diameter petri dishes containing 10 ml of MS medium supplemented with 50 μM KNO3, 1 μM myo-inositol, 5 μM casein hydrolysate, 4.4 μM of BA and 5.4 μM of NAA solidified with the same gelling agents. The experimental design was a randomized complete block with 3 replications. A replicate for each genotype consisted of ten petri dishes containing four explants. The statistical analysis showed significant differences among genotypes and gelling agents. Of the fourteen recombinant inbred lines tested `C93' presented the highest values for all regeneration traits in the five different media and it was better than the best parent. Agarose and Agar-Agar were more better than other gelling agents for shoot induction. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Crosses were made between three cytoplasmic male-sterile and five restorer sunflower inbred lines. F1 hybrids, including their parents, were studied for their embryogenetic ability. Sterilized seeds were germinated in culture tubes on agar-solidified basal medium. Seven days after germination, epidermal layers from hypocotyls were transferred into MS and B5 liquid-midia for 5 and 8 days respectively. Then they were transferred to MS-120 solid-medium with a high level of sucrose (120 g/l). The experimental design was complete randomized blocks with three replications. Each replication per genotype consisted of three Erlenmeyer flasks with 20–25 epidermal layers (explants). Analysis of variance indicated the presence of significant variation among genotypes for all traits studied. General combining ability (GCA) and specific combining ability (SCA) showed significant effects for the studied traits. The highest value of GCA for the number of embryogenic explants per 100 explants plated (EE/100EP) belongs to parental female line ’CMS-PET1 B9’. This inberd line also gave a high positive GCA effect for the number of embryos per ten embryogenic explants (E/10EE). Additionally, it had the highest values for EE/100EP and E/10EE (41.70 and 19.28 respectively) and should be a promising parent in crossing programmes for the enhancement of somatic embryogenesis in the sunflower. The highest values of specific combining ability (SCA) for EE/100EP and E/10EE belong to the F1 hybrid ’CMS-PET1B9 ’x’ RT1B11’ which has produced 53.45 embryogenic explants/100 explants and 9.67 embryos per ten embryogenic explants. Regeneration efficiency from three different regions of cotyledonary explants was examined in six sunflower inbred lines. Proximal, middle and distal regions from seedling cotyledons were cultured on regeneration medium supplemented with growth regulators. Plant regeneration by direct organogenesis was observed after four weeks. Significant differences among inbred lines were found for regeneration percentage and average number of shoots per total explants. Also a decreasing regeneration capacity was observed from proximal to distal sections for all inbred lines. Regeneration ability from cotyledonary explants in this species is strongly influenced by the genotype and by the region from which the explant was obtained. The distance to the cotyledonary node plays a preponderant role in the expression of shoot forming capacity. Shoot differentiation via seedling cotyledons depends upon the presence of the proximal region of cotyledon regardless of the genotype.  相似文献

Four near-isonuclear polycytoplasmic versions of 81A and two of Pb 402A male-sterile lines of pearl millet (Pennisetum typhoides) were used in factorial matings with five inbred male testers in different combinations in three sets. The cytoplasmic differences were studied for several agronomic traits using mean values and general combining effects (gca) of male-sterile lines, and specific combining ability effects of hybrids. The fertility/ sterility behaviour of different male-sterile lines in crosses with common male parents was also studied. Significant differences among near-isonuclear polycytoplasmic lines were observed in mean values for a few traits such as plant height, leaf length and peduncle length, but the differences for combining ability were more pronounced. The A3 cytoplasm was a better general combiner than the A2 cytoplasm for grain yield and both A2 and A3 cytoplasms were better general combiners for leaf length and peduncle length. In addition, superiority of A3 cytoplasm for gca was observed for plant height and ear characters over the A2 cytoplasm in set II. A differential behaviour of cytoplasms, both in combination with a common pollinator and across pollinators, was observed for several traits. The results provide evidence for the distinctiveness of different cytoplasmic sources in pearl millet and for the influence of cytoplasmic factors on the phenotypic expression of nuclear genes. A diversification of male sterility sources in the breeding of pearl millet hybrids is suggested.  相似文献

Genetic variability for regeneration ability was evaluated by studying direct organogenesis from cotyledons of thirteen genotypes including three cytoplasmic male sterile, three maintenor, three restorer inbred lines, and four F1 hybrids obtained by crosses between some of these inbred lines. The experimental design was a complete randomized block with three replications. A high genetic variability for organogenesis parameters between genotypes was observed in this study. Evidence of cytoplasmic effect and nucleo-cytoplasmic interaction for some of regeneration parameters was observed. The data also showed the importance of additive genetic control for organogenesis parameters in most genotypes. Summary Triple-testcross experiments were used to analyze epistatic contributions to larva weight, pupa weight, pupa width and adult weight in Tribolium castaneum. Seven diverse inbred lines and the F1. produced by crossing the two tester lines were examined for indications of epistasis. Larva weight was the only trait for which no significant epistasis was detected. There was significant epistasis for pupa weight in three of the inbred lines; for pupa width in four of the inbred lines; for adult weight in five of the inbred lines. Only one inbred line and the F1 line failed to exhibit significant epistasis for any trait. Each inbred line had a unique pattern of epistasis, suggesting that a number of different loci were contributing to the detected epistasis.This paper (No. 76-5-158) is published with the approval of the Director of the Kentucky Agricultural Experiment Station.  相似文献

Summary A diallel cross of eight maize, Zea mays L., inbred lines was analyzed for reaction to two species of root-knot nematodes, Meloidogyne arenaria (Neal) Chitwood and M. javanica (Treub) Chitwood. Egg production following inoculation of F1 hybrid seedlings with nematode eggs was determined in a greenhouse experiment. Data were analyzed using Griffing's Method 4, Model I. General combining ability was a significant source of variation in egg production of both M. arenaria and M. javanica; specific combining ability was not a significant source of variation for either. The correlation between egg production of the two nematode species on the 28 F1 hybrids was highly significant. Hybrids with Mp313 or SC213 as one parent were the most resistant to both species. Genetic distances between seven oil sunflower inbred lines expressed in the variability of five morphological traits were analysed in terms of their effect on the specific combining ability (SCA) effects in these lines for yield component traits. The multivariate analysis of variance (MANOVA) and other related multidimensional methods were used to recognize the differences between genotypes with respect to morphological traits. Canonical analysis was applied for a graphical configuration of parental lines and diallele hybrids in the two-dimensional space. As a measure of the difference between parental lines with respect to the analysed morphological traits jointly, i.e. as a measure of the morphological distance between these forms, Mahalanobis' distance was used. Phenotypic differences between lines were estimated using Mahalanobis' distance for each pair of crossed lines calculated for all the analysed morphological traits. The effect of morphological differences between inbred lines on the mean SCA effect for individual yield components, measured by the linear regression, was significant for oil yield per plant and for the number of seeds per plant.  相似文献

Reduction of hyperhydricity in sunflower tissue culture   总被引:3,自引:0,他引:3  
In order to reduce the occurrence of hyperhydrated shoots, the response of three sunflower inbred lines was examined on regeneration media containing various concentrations of kinetin, silver nitrate, and casein hydrolysate, calcium nitrate and cobalt nitrate. There were differences among the inbred lines for all the parameters taken into account to outline the in vitro efficiency. Percentage of hyperhydrated primordia, average number of shoots and of primordia per total explants, and percentage of hyperhydrated shoot traits differed among all media. The genotype × culture media interaction was significant for average number of shoots and primordia per total explants, regeneration percentage and percentage of hyperhydrated primordia. Among all media tested, those containing silver nitrate significantly reduced hyperhydricity in a dose-dependent way. The addition of silver nitrate showed to be useful in improving the quality of sunflower micropropagated plants by reducing this undesirable phenomenon.  相似文献

Summary Plant root nutrient uptake efficiency may be expressed by the kinetic parameters, Vmax and Km, as well as by normal enzymatic reactions. These parameters are apparently useful indices of the level of adaptation of genotypes to the nutrient conditions in the soil. Moreover, sulfate uptake capacity has been considered a valuable index for selecting superior hybrid characterized by both high grain yield and efficiency in nutrient uptake. Therefore, the purpose of this research was to determine combining ability for sulfate uptake, in a diallel series of maize hybrids among five inbreds. Wide differences among the 20 single crosses were obtained for Vmax and Km. The general and specific combining ability mean squares were significant and important for each trait, indicating the presence of considerable amount of both additive and nonadditive gene effects in the control of sulfate uptake. In addition, maternal and nonmaternal components of F1 reciprocal variation showed sizeable effects on all the traits considered. A relatively high correlation was also detected between Vmax and Km. However, both traits displayed enough variation to suggest that simultaneous improvement of both Vmax and Km should be feasible. A further noteworthy finding in this study was the identification of one inbred line, which was the best overall parent for improving both affinity and velocity strategies of sulfate uptake.  相似文献

Summary The androgenic ability of four Medicago sativa L. genotype (Boynitza 5, Byala, 494, and 3815) was tested. Callus and organogenesis were induced in all lines studied. The percentage of anthers producing calluses and organogenesis showed wide variation (calluses—from 11% up to 77%; organogenesis—4.8% to 15.2%). It has been established that genotype, nutrient medium composition, and stage of pollen development considerably affected both callus production and organogenesis. Androgenesis in M. sativa could be achieved via callus and direct embryogenesis. About 500 morphologically different regenerants were obtained. Wide variability in chromosome number of regenerated plants was observed by cytological studies. Haploid, dihaploid, as well as mixoploid plants were obtained.  相似文献

This study was conducted to determine the reciprocal effects for anther culture response in wheat (Triticum aestivum L.) using a set of 4 × 4 full diallel crosses. Both reciprocal and nuclear genetic effects were highly significant for anther culture response and useful for selection and breeding purposes. General combining ability (GCA) effects were predominant for all investigated anther culture traits. Also, significant differences for specific combining ability (SCA) effects were detected between reciprocal crosses. Although significant reciprocal differences for responding anther, callus number and green plant regeneration were recorded in some reciprocal crosses, there were no significant reciprocal differences for albino plant regeneration. The use of one parent as male or female could lead to change at the production of green plants from the F1 hybrids and screening of inbred lines for response to anther culture, without reciprocal effects, could decrease the utilization of breeding material.  相似文献

Immature inflorescences of ten rye inbred lines (inbred degree S10 and S11) were cultured on solidified MS medium supplemented with 3.0 mg/dm3 of 2,4-D. According to their capability for callus production explants were classified into two groups : responsive (giving weak or intensive callus production) and non responsive (lack of callus formation). After transferring responsive material into hormone-free medium the regeneration of roots or shoots from the intensive growing callus was observed. Consistent differences between lines in the portion of explants with a certain response were found. They were divided into five groups reacting in the same way. Lines with different in-vitro response were crossed in an incomplete diallel. F1, F2 and F3 generations were analyzed and the following conclusions drawn: the ability for plant regeneration from immature inflorescences in rye is determined by numerous loci, has a recessive character, and both callus production and regeneration suppression may be controlled by complementary genes.  相似文献

Aggressiveness variation and its alternation with non-race specific resistance in sunflower were studied in 19 Plasmopara halstedii isolates belonging to several races. Regarding aggressiveness criteria, percentage infection, latent period, sporulation density and dwarfing, on two sunflower inbred lines showing different levels of non-race specific resistance resistance FU and BT, there were significant differences in aggressiveness for P. halstedii isolates. The index of aggressiveness varied between 9.4 and 31.4. The inbred line BT, rather susceptible in the field, showed a higher percentage infection, a higher sporulation density, a shorter latent period and less reduced hypocotyl length than inbred line FU, which showed a greater resistance in the field. Percentage infection on FU was 1.4% less than BT, latent period on BT was 12.4% less than FU, sporulation density on FU was 22.3% less than BT and reduced hypocotyl length on BT was 15.3% less than FU. Consequently, it seems that the criteria as latent period, sporulation density and reduction of hypocotyl length could be used to measure non-race specific resistance in sunflower to P. halstedii under controlled conditions.  相似文献

The fall armyworm, Spodoptera frugiperda (J. E. Smith), and southwestern corn borer, Diatraea grandiosella Dyar, are major insect pests of maize, Zea mays L., in the southern USA. Both insects feed extensively on leaves of plants in the whorl stage of growth. A diallel cross of seven inbred lines with different levels of susceptibility to leaf feeding damage in the field was evaluated in a laboratory bioassay for fall armyworm and southwestern corn borer larval growth. Diets were prepared from lyophilized leaf tissue of field-grown plants of the inbred lines and their 21 F1 hybrids. One inbred line, Tx601, exhibited heavy leaf damage in field tests but showed moderate resistance in the laboratory bioassay. Both general and specific combining ability were highly significant sources of variation in the inheritance of fall armyworm and south-western corn borer larval growth in the laboratory bioassay. The changes in the activity of some antioxidant enzymes and endogenous H2O2 level in zygotic sunflower embryos during organogenesis and somatic embryogenesis were monitored. Pathways of regeneration were induced on media differing with sucrose concentration 87 mmol dm−3 for shoot [shoot induction medium (SIM) medium] and 350 mmol dm−3 [embryo induction medium (EIM) medium] for somatic embryo induction. Water potential of the explants cultured on SIM increased, while the embryos maintained on EIM showed middle water deficit stress. The pattern of superoxide dismutase (SOD) isoforms was similar in organogenic and embryogenic culture; however, the intensity of MnSOD bands was higher on SIM than on EIM. Differences in catalase activity were observed: high activity on SIM predominated, whereas on EIM it was reduced. The activity of guaiacol peroxidase in the explants producing shoots and somatic embryos differed at the beginning of culture, but became comparable at the time of shoot and somatic embryo formation (day 5). H2O2 content was unchanged in organogenic culture, but on EIM it increased on day 1 followed by significant decrease. The results indicate that sugar concentration per se, or via induction of different developmental pathways influences the activity of antioxidant enzymes and also H2O2 level in cultured sunflower embryos.  相似文献

Summary Combining ability studies for grain yield and its primary component traits in diallel crosses involving seven diverse wheat cultivars of bread wheat (Triticum aestivum L.) over generations F1-F5 are reported. The general and specific combining ability variances were significant in all generations for all the traits except specific combining ability variance for number of spikes per plant in the F5. The ratio of general to specific combining ability variances was significant for all the traits except grain yield in all the generations. This indicated an equal role of additive and non-additive gene effects in the inheritance of grain yield, and the predominance of the former for its component traits. The presence of significant specific combining ability variances in even the advanced generations may be the result of an additive x additive type of epistasis or evolutionary divergence among progenies in the same parental array. The relative breeding values of the parental varieties, as indicated by their general combining ability effects, did not vary much over the generations. The cheap and reliable procedure observed for making the choice of parents, selecting hybrids and predicting advanced generation (F5) bulk hybrid performance was the determination of breeding values of the parents on the relative performance of their F2 progeny bulks.  相似文献

Summary Changes that may have occurred over the past 50 years of hybrid breeding in maize (Zea maize L.) with respect to heterosis for yield and heterozygosity at the molecular level are of interest to both maize breeders and quantitative geneticists. The objectives of this study were twofold: The first, to compare two diallels produced from six older maize inbreds released in the 1950's and earlier and six newer inbreds released during the 1970's with respect to (a) genetic variation for restriction fragment length polymorphisms (RFLPs) and (b) the size of heterosis and epistatic effects, and the second, to evaluate the usefulness of RFLP-based genetic distance measures in predicting heterosis and performance of single-cross hybrids. Five generations (parents, F1; F2, and backcrosses) from the 15 crosses in each diallel were evaluated for grain yield and yield components in four Iowa environments. Genetic effects were estimated from generation means by ordinary diallel analyses and by the Eberhart-Gardner model. Newer lines showed significantly greater yield for inbred generations than did older lines but smaller heterosis estimates. In most cases, estimates of additive x additive epistatic effects for yield and yield components were significantly positive for both groups of lines. RFLP analyses of inbred lines included two restriction enzymes and 82 genomic DNA clones distributed over the maize genome. Eighty-one clones revealed polymorphisms with at least one enzyme. In each set, about three different RFLP variants were typically found per RFLP locus. Genetic distances between inbred lines were estimated from RFLP data as Rogers' distance (RD), which was subdivided into general (GRD) and specific (SRD) Rogers' distances within each diallel. The mean and range of RDs were similar for the older and newer lines, suggesting that the level of heterozygosity at the molecular level had not changed. GRD explained about 50% of the variation among RD values in both sets. Cluster analyses, based on modified Rogers' distances, revealed associations among lines that were generally consistent with expectations based on known pedigree and on previous research. Correlations of RD and SRD with f1 performance, specific combining ability, and heterosis for yield and yield components, were generally positive, but too small to be of predictive value. In agreement with previous studies, our results suggest that RFLPs can be used to investigate relationships among maize inbreds, but that they are of limited usefulness for predicting the heterotic performance of single crosses between unrelated lines.Joint contribution from Cereal and Soybean Research Unit, USDA, Agricultural Research Service and Journal Paper no. Under stress of iron deficiency roots of sunflower (Helianthus annuus L.) increase proton efflux which acidifies the root medium, increase the ferric reducing capacity and the exudation of phenolic compounds. Differences have been found previously among sunflower inbred lines in the capacity of their roots to lower pH and it was also found that this character is under genetic control.This work presents the results of an inheritance study made by crossing two genotypes, one (CMS HA 89) without acidification capacity and another (RHA 271) with it. Plants were grown individually in 75 mL vessels with an aerated solution low in iron. After 4 days, solutions were changed to one without iron and the pH of the medium was measured during the following days. Results from F1, F2, and backcross generations can be explained with two pairs of alleles controlling the character, being the relation between alleles of complete dominance at both gene pairs, but either gene, when dominant is epistatic to the other.  相似文献

A genetic map was constructed with specific PCRs, DALPs and AFLPs using F8-generation sunflower recombinant inbred lines. RI lines generated from a F2 population of one cross between the two cultivated inbred lines HA89 (maintainer for Pet1 CMS) and LR4 (restorer for Pet1 CMS) were used. A total of 305 markers were located using seven sPCR, 64 DALP and 301 AFLP loci. They were generated with one, seven and 14 primer pairs, respectively. The map construction consisted of a two-step strategy using 6 and 3.1 LOD scores revealed by a simulation file. Mapped markers were assembled into 18 linkage groups covering 2,168.6 cM with an average of 6.1 cM. The distribution of DALPs and AFLPs revealed that both markers tagged different regions to enable covering most of the sunflower genome. This leads to the longest map published so far for sunflower.  相似文献
