The effect of phenols on respiratory enzymes in seed germination   总被引:1,自引:0,他引:1  
Low molecular weight phenolic compounds were identified in two soilswith different vegetative cover, Fagus sylvatica, L. andPinus laricio, Poiret, spp. calabrica, and were tested atdifferent concentrations on seed germination of Pinuslaricio, and on respiratory and oxidative pentose phosphate pathwayenzymes involved in the first steps of seed germination. The data obtained showthat there are marked differences in the phenolic acid composition of the twoinvestigated soils. All the phenolic compounds bioassayed inhibited seedgermination and those extracted from Pinus laricio soilwere particularly inhibitory. We also found that the non-germination of seedsisstrongly correlated to the inhibition of the activities of enzymes ofglycolysisand the oxidative pentose phosphate pathway.  相似文献

This study examined seasonal variations in the content of water-soluble phenolics extracted from litter and Ah horizon of forest soil under Pinus laricio Poiret trees. After having identified and quantified different phenolics, the effects of seasonal phenolic extracts, single phenolic acids and synthetic phenol mixtures on seed germination of Pinus laricio Poiret, Pinus pinaster Aiton and Pinus halepensis Mill. were evaluated, and the activity of phenolics on the main enzymes involved in the glyoxylate cycle was tested. The results indicated a seasonal variation of phenol content in soil, with largest concentrations of water-soluble phenolic acids in autumn and smallest concentrations in summer. Addition of phenol extracts to germination medium reduced seed germination showing phytotoxic effects which differed, depending on the species and the fractions tested. Phenols extracted from litter and Ah horizon in autumn and winter, vanillic acid, and synthetic mixtures reproducing phenols extracted from soil in winter inhibited seed germination of Pinus laricio, pinaster and halepensis more than other treatments. A greater inhibitory effect was observed on seed germination of Pinus pinaster and halepensis compared to Pinus laricio. The early effects of low molecular weight phenolic compounds, released by Pinus laricio and Pinus pinaster litter, on ammonium uptake and its assimilation in two Pinus species were studied. In Pinus laricio seedlings, the exposure to phenols extracted from Pinus laricio litter increased not only the ammonium uptake but also the activity of the main enzymes involved in its assimilation, whereas the phenols extracted from Pinus pinaster litter had a negative effect on these metabolic processes. In Pinus pinaster seedlings, the exposure to both phenols decreased the ammonium uptake and the activity of the main enzymes involved in its assimilation. Histological analysis carried out in Pinus laricio roots showed that phenols extracted from Pinus laricio litter induced the greatest growth of cortex, element through which occurs the ions uptake in plants, whereas phenol extracted from Pinus pinaster litter inhibited cortex development. On the other hand, in Pinus pinaster seedlings the observation showed that both phenols inhibited cortex growth indicating a strict correlation between cortex development and ammonium uptake and its assimilation.  相似文献

The aim of this research was to extract, with a sequential method, HS free from water-soluble phenols (HS-WP) and water-soluble phenols free from humic substances (WP-HS), and to evaluate and to compare the biological effects of these two extracts to humic substances (HS) and water-soluble phenols (WP) traditionally extracted. In each extract we determined the concentration of low molecular weight organic acids, soluble carbohydrates, fatty acids, phenolic acids and total proteins. We tested the biological activity of each soil extract and of the single identified compounds on different plant organs (Pinus laricio callus, Daucus carota cells, and Pinus laricio, Pinus halepensis, Lens culinaris and Cichorium intybus seeds). The results showed that eliminating from HS the WP fraction, and from WP the HS fraction we obtained extracts chemically different from HS as such, and WP as such. HS and HS-WP increased callus and cell growth and also root elongation of the different species used; in contrast, WP and WP-HS had negative effects inhibiting callus and cell growth and seed germination percentage of coniferous, herbaceous and leguminous species. The negative effects can be ascribed to the presence of phenolic acids in the extracts while the positive biological activity can be attributed to the presence of tartaric acid, and fatty acids. In conclusion, this study helps to discriminate the effects of humic substances against phenolic compounds extracted from soils explaining the different and in some way contradictory biological behaviour of these two main fractions of SOM.  相似文献

通过室内模拟试验,阐明阔叶红松林中已测得含量较高的3种酚酸物质(苯甲酸、丁香酸和香草酸)对红松种子萌发及苗木生长的影响,为探索阔叶红松林内化感作用机理及解决红松更新障碍问题提供科学依据。采用培养皿培养法及室内盆栽培养法,以红松种子和3年生红松苗为试验对象,设置不同浓度(2、20、200 mg/L)苯甲酸、丁香酸、香草酸处理液,以蒸馏水为对照(CK),进行红松种子萌发试验及红松苗木生长试验,研究3种酚酸物质对红松种子发芽、苗木生长、光合色素、抗氧化酶活性、膜脂过氧化作用及渗透调节物质的影响。结果表明,(1)不同浓度3种酚酸均抑制红松种子萌发,但酚酸浓度变化仅对红松种子发芽率影响差异显著。(2)3种酚酸对红松苗木生长及物质积累抑制作用显著。浓度变化对红松苗株高及地径影响不显著,对生物量、根干重和茎干重影响显著。(3)针叶叶绿素a、叶绿素b、类胡萝卜素含量变化对酚酸处理反应一致,20 mg/L的3种酚酸均显著抑制光合色素产生,而200 mg/L丁香酸溶液及2 mg/L香草酸溶液均显著促进叶绿素a和类胡萝卜素积累。(4)酚酸处理使红松苗针叶中POD、CAT活性降低,SOD活性增加。针叶中MDA含量显著增加,200 mg/L丁香酸溶液处理组针叶MDA含量高于CK处理组70.51%。(5)不同浓度苯甲酸溶液促进可溶性糖增加,抑制可溶性蛋白增加;不同浓度丁香酸溶液促进可溶性蛋白增加,而不同浓度香草酸溶液抑制可溶性蛋白增加,二者对可溶性糖含量影响受浓度变化影响显著。苯甲酸、丁香酸、香草酸影响红松种子萌发,通过对红松苗光合色素、抗氧化酶活性及渗透调节物质的影响导致其生长受抑制、生物量减少,产生膜脂过氧化伤害。因此,解决阔叶红松林内红松更新障碍问题时,凋落物及土壤中酚酸物质的化感作用不容忽视。  相似文献

The allelopathic effects of Juniperus rigida litter aqueous extract (LE) on wheat and Pinus tabuliformis were studied, as well as the physiological responses to the extract. High concentration LE (0.10 g Dw/ml) significantly inhibited the seed germination and seedling growth in receptor plants. The chlorophyll content and root activity in the wheat seedlings were reduced significantly across all treatments; however, those were more prominently reduced at high concentration (0.10 g Dw/ml) but received little stimulation at low concentration (0.025 g Dw/ml) in P. tabuliformis. The content of malonaldehyde (MDA) increased with increasing concentrations of LE, except at 0.025 g Dw/ml. Activities of antioxidant enzymes (POD, CAT and SOD) in receptor plants were all significantly inhibited at high concentrations but stimulated at low concentrations. These results demonstrate that the aqueous extract from J. rigida litter has allelopathic potential. Various phenolic compounds were identified in litter aqueous extract and litter ethanol extract by HPLC. The phenolic compound content in the aqueous extract was significantly lower than that in the ethanol extract. Chlorogenic acid and podophyllotoxin were the predominant phenolic compounds in both types of litter extracts. These findings suggest that the seed germination and seedling growth of P. tabuliformis and wheat would be inhibited when planted near large amounts J. rigida litter.  相似文献

Stable isotopes in tree rings have widely been used for palaeoclimate reconstructions since tree rings record climatic information at annual resolution. However, various wood components or different parts of an annual tree-ring may differ in their isotopic compositions. Thus, sample preparation and subsequent laboratory analysis are crucial for the isotopic signal retained in the final tree-ring isotope series used for climate reconstruction and must therefore be considered for the interpretation of isotope–climate relationships. This study focuses on wood of Corsican Pine trees (Pinus nigra ssp. laricio) as this tree species allows to reconstruct the long-term climate evolution in the western Mediterranean. In a pilot study, we concentrated on methodological issues of sample preparation techniques in order to evaluate isotope records measured on pooled whole tree-ring cellulose and whole tree-ring bulk wood samples. We analysed 80-year long carbon and oxygen chronologies of Corsican Pine trees growing near the upper tree line on Corsica. Carbon and oxygen isotope records of whole tree-ring bulk wood and whole tree-ring cellulose from a pooled sample of 5 trees were correlated with the climate parameters monthly precipitation, temperature and the self-calibrating Palmer Drought Severity Index (sc-PDSI). Results show that the offsets in carbon and oxygen isotopes of bulk wood and cellulose are not constant over time. Both isotopes correlate with climate parameters from late winter and summer. The carbon and oxygen isotope ratios of cellulose are more sensitive to climatic variables than those of bulk wood. The results of this study imply that extraction of cellulose is a pre-requisite for the reconstruction of high-resolution climate records from stable isotope series of P. nigra ssp. laricio.  相似文献

Pinus laricio Poiret is the most widespread conifer occurring in Calabria (Sila and Aspromonte Massifs), Sicily (Mount Etna) and Corsica. Particularly, Calabrian laricio pine forests are strongly related to complex geological history, lithological and climatic characteristics and long human exploitation of the Sila territory, of which they are the most emblematic element. As far as we know, the P. laricio populations from their main range areas have never been thoroughly studied at the molecular level. This article reports on the first deep analysis of the genetic variability and structure of individuals from populations located in Calabria, Sicily and Corsica using both chloroplast and nuclear microsatellite markers. Significant variation within populations and low differentiation among populations were found by means of molecular variance estimates for the both types of markers. Bayesian clustering analyses revealed an unexpected grouping of P. laricio populations with individuals from Sila and, particularly, those from the Natural Reserve of Fallistro, have been identified as genetically distinct. Temporal genetic analysis in three large P. laricio populations also showed that there were no differences in genetic diversity levels over time; however, it allowed to recognize populations that deserve to be considered as a high priority for suitable preservation and management.  相似文献

Summary Phenolic glycosides, commonly occurring allelochemicals in the plant family Salicaceae, are differentially toxic to subspecies of the eastern tiger swallowtail and responsible for striking differences in the abilities of Papilio glaucus canadensis and P.g. glaucus to utilize the Salicaceae as food plants. This research was designed to test the hypothesis that particularly high esterase activity confers resistance to phenolic glycosides in P.g. canadensis. I conducted larval survival trials in which the phenolic glycosides salicortin and tremulacin were administered with and without inhibitors of the major detoxication enzymes. Results for P.g. canadensis showed that when esterases were inhibited, toxicity of the phenolic glycosides increased greatly. None of the inhibitors significantly increased toxicity of the compounds to P.g. glaucus. I also conducted in vitro assays of the major detoxication enzymes (polysubstrate monooxygenases, esterases, and glutathione transferases) in larval midguts. Soluble esterase activity was 3-fold higher in P.g. canadensis than in P.g. glaucus. Moreover, esterase activity was inducible by prior consumption of phenolic glycosides in P.g. canadensis but not in P.g. glaucus. Glutathione transferases may also be involved in the terminal metabolism of phenolic glycosides. This is the first verified case of detoxication of an allelochemical by esterase enzymes in herbivores. The biochemical adaptation has played an important role in the evolution of food plant preferences in P. glaucus subspecies.  相似文献

The research focused on the changes of phenolic compounds as well as their antiradical activity and reducing power isolated from Amur grape (Vitis amurensis) seeds during germination under optimal conditions and under osmotic stress. The seeds were found to contain tannins, (+) catechin, (−) epicatechin, and gallic acid (in free, ester- and glycoside-bound forms). Extracts from the seeds were also shown to contain two other phenolic acids: caffeic and p-coumaric acids, in very low levels. During a 3-day seed germination test under osmotic stress (−0.5 MPa), the content of total phenolics, tannins and phenolic acids declined as compared to the control. However, seed germination under stress conditions led to a significant increase in the amount of catechins. Because catechin is the one of the units in condensed tannins, its dynamic increase during seed germination may be involved in metabolism of tannins under osmotic stress. It is also likely that the synthesis of catechins is greater under stress conditions and these compounds may be engaged in the process of acclimatization of grapevines to stress conditions. The content of total phenolic compounds in seed extracts is positively correlated with their antioxidant properties. The extracts from seeds germinated under optimal conditions exhibited strong antiradical properties against the DPPH (2,2-diphenyl-1-picrylhydrazyl) radical as well as reducing power. As regards the extracts from grape seeds germinated under osmotic stress, this capability was much weaker. The research demonstrated that antioxidants could interfere with the oxidation process induced by various stresses by acting as oxygen scavengers, therefore the tolerance to drought stress might be correlated with an increase in the antioxidant potential.  相似文献

Germination of lupine (Lupinus luteus L.) seeds was accompanied by an increase in concentration of free radicals with g 1 and g 2 values of 2.0056 ± 0.0003 and 2.0033 ± 0.0005, respectively. The highest intensity of free radical signal was observed in embryo axes immediately after radicle protruded through the seed coat. Hydrogen peroxide accumulated in embryonic axes and cotyledons during imbibition before the onset of germination in the seed population. The activities of superoxide dismutase (SOD, EC 1.15.1.1) and catalase (CAT, EC 1.11.1.6) rose progressively in embryo axes. In cotyledons SOD activity did not change significantly, while that of CAT increased during germination. The enhancement of Cu, Zn-SODs and Mn-SOD isoforms in embryonic axes was observed. A new isoform of catalase was synthesized, suggesting that it plays a relevant role during germination. SOD and CAT activities were detected in dry seeds. Free radical generation and response of antioxidative enzymes differed between embryo axes and cotyledons during the germination timecourse.  相似文献

The effect of Paxillus involutus, Laccaria laccata, Suillus luteus, S. bovinus, Hebeloma crustuliniforme and a strain of the ectendomycorrhizal fungus Mrg X (Ascomycotina) on the content of phenolic compounds: ortho-diphenols and total soluble phenols in roots of Pinus sylvestris seedlings grown in vitro was investigated. Mycorrhizal fungi varied significantly in the ability to influence the concentration of phenolics in Scots pine roots. Plants inoculated with S. luteus accumulated the lowest amount of the phenolics, whereas the amount of phenolics in pine roots inoculated with L. laccata was the highest.  相似文献

Abstract

Nitrogen fixation was measured in a Corsican pine (Pinus laricio Poiret) forest in Calabria (Southern Italy). Acetylene reduction activity (ARA) and CO2 production levels were determined by incubation of litter and superficial (0–5 cm) soil layer samples in the field, at monthly intervals. ARA variations were not correlated to those of substrate moisture, air temperature and microbial respiration. In fact N2 fixation presented phases of different intensity which irregularly followed each other. Both litter and soil showed similar rates of N2 fixation. Based on a C2H2:N2 ratio of 3:1 0.8 Kg N ha–1 y–1 in each layer are fixed in the Pinus laricio forest, thus contributing to the N status of the soil in this nutrient–poor forest.  相似文献

Abstract

Exposure to UV-B at ambient or enhanced levels is known to trigger a variety of responses in all living organisms, including higher plants. Here we show that in Cucumis sativus L. UV-B radiation affects enzyme activity of key oxydative pentose phosphate pathway (OPPP) enzymes glucose-6-phosphate dehydrogenase (G6P-DH) and 6-phosphogluconate dehydrogenase (6-PGlu-DH), of key phenolic compounds enzyme phenylalanine ammonia lyase (PAL) as well as erythrose-4-phosphate, tryptophan and tyrosine levels. Furthermore, we found an increased activity of antioxidant enzymes such as peroxidase (POX) and catalase (CAT) in treated plants, with respect to the controls. In order to confirm the biochemical results, we isolated total RNA from both controls and UV-B treated plants to be used for gene expression analysis. We demonstrated that UV-B increases the gene expression level of peroxidase (POX), catalase (CAT) and phenylalanine ammonia lyase (PAL). Finally, our results are useful for understanding protective strategies against UV-B radiation and for elucidating what components are involved in stress-induced signals within the plant.  相似文献

Several antioxidants namely ascorbic acid, salicylic acid, sodium benzoates, thiouria and catechol were used as seed treatment and foliar spraying to reduce the incidence of root and pod rot diseases of peanut caused by (Fusarium oxysporum, Fusarium solani and Fusarium moniliforme) as well as to determine of phenolic compounds and oxidative enzymes in the treated plants. Each antioxidant was used at different concentrations (2, 4, 6 and 8 mM) against tested pathogenic fungi in vitro. All antioxidants at 8 mM showed the greatest reduction of mycelial growth of the pathogens. In greenhouse experiments, treated seeds (Giza 5 cv.) or foliar spraying of peanut plants after 30 and 60 days from planting date with each antioxidant at 8 mM reduced severity of both diseases. The treated plants with antioxidants increased accumulation of phenolic compounds and activity levels of oxidative enzymes (catalase, peroxidase and polyphenoloxidase) in infected plants compared to healthy plants.

Functioning of antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APO), and guaiacol peroxidases (GPO)) and low-molecular organic ROS scavengers (proline and phenolic compounds) in various organs (roots, cotyledons, stem, and needle) of 6-week-old seedlings of pine (Pinus sylvestris L.) developing in the chronic presence of ZnSO4 (50, 100, and 150 μM). Pine seedlings were grown in water culture in the climate-controlled chamber at an irradiance of 37.6 W/m2 with a 16-h photoperiod, an air temperature of 23 ± 1/15 ± 1°C (day/night), and a relative humidity of 55/70% (day/night). Endogenous Zn content was a key factor determining SOD activity decomposing superoxide into H2O2 and O2. Hydrogen peroxide produced is efficiently destroyed by CAT and also by APO and GPO. At the same time, the content of proline increased (especially at 150 μM ZnSO4), but the content of phenolic compounds remained unchanged. All these processes help to maintain stable intracellular levels of O2⊙− and H2O2 at elevated zinc concentrations and to prevent generation of hydroxyl radical and development of oxidative stress.  相似文献

It was to be shown whether during the biogenesis of microbodies some of their components were already present in the cell prior to the organelle's assembly. To this end, the occurrence and properties of catalase in soluble and particular fractions of ripening cucumber seeds were examined. Homogenates of seeds from ripening fruits were fractionated by isopycnic density gradient centrifugation, and thus catalase was found in three different fractions: as a soluble enzyme in the gradient supernatant, as a membrane fraction at density d=1.18 kg l-1, and in association with microbodies. In the early steps of seed formation, catalase was detected at density d=1.18 kg l-1 and in the gradient supernatant. At a later stage of seed maturation, however, catalase was primarily associated with microbodies which exhibited an equilibrium density of d=1.23 kg l-1. M r as well as subunit M r of catalase were determined, and their close immunological relationship to leaf peroxisomal catalase and glyoxysomal catalase was demonstrated. Biosynthesis of catalase at different stages of seed maturation was investigated by in vivo labeling with l-[35S]methionine, l-[14C]leucine and -[3H]aminolaevulinic acid. Electrophoretic analysis of de novo synthesized catalase subunits revealed the occurrence of a heavy form (M r 57,500) in the soluble fraction; this form was preferentially labeled. A light form, M r 53,500, was detected in microbodies and also in the soluble fraction. The findings lend support to the hypothesis that the rate of catalase synthesis is highest in an early stage of seed formation, when globulins have already been formed, but before de novo synthesis of malate synthase has commenced. Compounds that promoted the growth of pollen tubes were isolated from the style of Rhododendron mucronatum and were identified as azalein, (+)-catechin, and (?)epicatechin. Among these compounds, 50–100 ppm of (+)-catechin or (?)-epicatechin increased the growth of pollen tubes in Camelliajaponica, R. mucronatum, Styrax japonica, and Pinus densijlora by 16–20%. These compounds had no significant effects on the growth of pollen tubes in Lilium auratum and Narcissus pseudo-narcissus. Among the phenolic compounds with similar chemical structures as the promoters, catechol-and pyrogallol-type compounds had the activity and the latter types were more effective. As α-tocopherol also showed growth promoting activity, it was thought that the antioxidative activity of the phenolic compounds was responsible for the promotion of the pollen tube growth.  相似文献

Aims: To investigate the effect of seven wine phenolic compounds and six oenological phenolic extracts on the growth of pathogenic bacteria associated with respiratory diseases (Pseudomonas aeruginosa, Staphylococcus aureus, Moraxella catarrhalis, Enterococcus faecalis, Streptococcus sp Group F, Streptococcus agalactiae and Streptococcus pneumoniae). Methods and Results: Antimicrobial activity was determined using a microdilution method and quantified as IC50. Mor. catarrhalis was the most susceptible specie to phenolic compounds and extracts. Gallic acid and ethyl gallate were the compounds that showed the greatest antimicrobial activity. Regarding phenolic extracts, GSE (grape seed extract) and GSE‐O (oligomeric‐rich fraction from GSE) were the ones that displayed the strongest antimicrobial effects. Conclusions: Results highlight the antimicrobial properties of wine phenolic compounds and oenological extracts against potential respiratory pathogens. The antimicrobial activity of wine phenolic compounds was influenced by the type of phenolic compounds. Gram‐negative bacteria were more susceptible than Gram‐positive bacteria to the action of phenolic compounds and extracts; however, the effect was species‐dependent. Significance and Impact of Study: The ability to inhibit the growth of respiratory pathogenic bacteria as shown by several wine phenolic compounds and oenological extracts warrants further investigations to explore the use of grape and wine preparations in oral hygiene.  相似文献

The sexual reproductive process in Pinus lambertiana has not been completely described, and previous attempts to generate hybrids with Pinus monticola and other North American pines have not been successful. The nature of incompatibility barriers between P. lambertiana and P. monticola is unknown. This needs to be understood if strategies are to be developed to overcome the said barriers. In this paper, development on interspecific crosses is compared with that on intraspecific crosses on the same parent trees. Pollen grains of both species germinated on the nucellus of both species within a week after pollination. Seed cone receptivity in P. lambertiana came approximately 2 weeks after receptivity in P. monticola, and this delay was perpetuated throughout ovule development in the first year of the reproductive process. Development of the second-year seed cones proceeded more gradually in P. lambertiana. However, seed cones reached maturity only for P. monticola x P. lambertiana. In both crosses, the barriers to hybridization occurred during the second year of the reproductive process. With the P. lambertiana as the seed parent, it was manifested through the failure of the megaspores at the free-nuclear stage to resume development. When P. monticola was used as the seed parent, the male and female gametes failed to fuse. Our results clearly show that the barriers to hybridization in these species occur before or at fertilization. However, the exact mechanisms behind these are still unknown. Based on the results of this study, we present several strategies to bypass the developmental barriers and possibly produce hybrid progenies.  相似文献
