Ten tri- and tetranucleotide microsatellite DNA markers were isolated and characterized from common carp (Cyprinus carpio L.) to estimate genetic potential. These markers were tested in the samples from two closely related carp populations (Cyprinus carpio var. xingguonensis and Cyprinus carpio var. wananensis). The number of the alleles ranged from three to nine, and observed and expected hererozygosities varied from 0.207 to 1.000 and from 0.499 to 0.900 in each population, respectively. No evidence for linkage disequilibrium was found, indicating that these markers will be useful for population studies.  相似文献

Concentrations of polychlorinated biphenyls (PCBs) in common carp, Cyprinus carpio, from the Des Moines River, Iowa, were assessed for variability related to sampling location, sampling period, fish age, and fat content. Concentrations were highest at a location near the City of Des Moines; they were substantially lower in 1981 than in 1980. Age and fat content had little influence on PCB concentrations in carp. Bighead carp Hypophthalmichthys nobilis and silver carp Hypophthalmichthys molitrix have been two economically important aquaculture species in China for centuries. In the past decades, bighead and silver carp have been introduced from the Yangtze River to many river systems in China, including the Pearl River, in annual, large-scale, stocking activities to enhance wild fisheries. Nonetheless, few studies have assessed the ecological or genetic impacts of such introductions on native conspecific fish populations. An mtDNA D-loop segment of 978 bp from 213 bighead carp samples from 9 populations and a 975 bp segment from 204 silver carp samples from 10 populations were obtained to evaluate genetic diversity and population integrity. Results from a haplotype network analysis revealed that most haplotypes of the Pearl River clustered with those of Yangtze River origin and only a small proportion were distinct, suggesting that both the native Pearl River bighead and silver carp populations are currently dominated by genetic material from the Yangtze River. The genetic diversity of Pearl River populations is high in both species because of this inter-population gene flow, but the diversity of native Pearl River populations is low. To preserve the native genetic diversity, stocking of non-native fingerlings should cease immediately and native Pearl River bighead and silver carp fish farms should be established. This research demonstrates the danger to native biodiversity across China because of the substantial, ongoing stock-enhancement activities without prior genetic assessment.  相似文献

Since its escapement into the Upper Mississippi River (UMR) and rapid expansion in the late 1900's, the common carp, Cyprinus carpio, has become a major component of the river fishery. Mean annual harvest value for this species was $270000 between 1953 and 1977. Recent evidence suggests that common carp may not be as abundant in the river as they have been in the past. We summarized historical commerical information on common carp in the UMR to put its current harvest levels in perspective. A major factor responsible for recent low annual harvests has been PCB contamination of the river in the Lake Pepin area. Market and biological factors probably have also contributed to declining harvests. To help explain why common carp populations in the UMR may be declining, we reviewed literature pertaining to the ecology of the species, particularly that from the river, its tributaries, or midwestern lakes. We included information on how common carp are controlled by environmental factors, and how they interact with other biota. Most aspects of carp ecology in the UMR and other areas are similar. Exceptions relate primarily to their response to annual water level regimes of the river. We postulate that population declines and absent year classes in the UMR may be related to poor recruitment during high- or variable-water years.  相似文献

The objective of this study was to assess the genetic diversity and population structure of goats in the Yangtze River region using microsatellite and mtDNA to better understand the current status of those goat genetic diversity and the effects of natural landscape in fashion of domestic animal genetic diversity. The genetic variability of 16 goat populations in the littoral zone of the Yangtze River was estimated using 21 autosomal microsatellites, which revealed high diversity and genetic population clustering with a dispersed geographical distribution. A phylogenetic analysis of the mitochondrial D‐loop region (482 bp) was conducted in 494 goats from the Yangtze River region. In total, 117 SNPs were reconstructed, and 173 haplotypes were identified, 94.5% of which belonged to lineages A and B. Lineages C, D, and G had lower frequencies (5.2%), and lineage F haplotypes were undetected. Several high‐frequency haplotypes were shared by different ecogeographically distributed populations, and the close phylogenetic relationships among certain low‐frequency haplotypes indicated the historical exchange of genetic material among these populations. In particular, the lineage G haplotype suggests that some west Asian goat genetic material may have been transferred to China via Muslim migration.  相似文献

用9对引物对采自长江上游合江江段的岩原鲤种群的14个个体进行了选择性扩增片段长度多态性研究,共检测出AFLP位点755个,平均每对引物扩增出84个位点,其中多态性位点354个,占46.89%。个体间的遗传距离为0.1061~0.2846,平均遗传距离为0.1793±0.0337。种群平均杂合度为0.1768。与其它一些鱼类比较表明,岩原鲤长江合江种群的遗传多样性可能不够丰富。  相似文献

摘要: 利用30对微卫星分子标记对长江中下游5个鲢群体进行了遗传多样性分析。结果表明: 在30个基因座中, 共检测到144个等位基因, 每个座位检测到的等位基因数为1~10个, 其中有25个座位具有多态性, 多态位点百分率为83.33,5个群体的平均等位基因数A为4.0/4.1, 平均有效等位基因数Ne为2.4445~2.6332, 平均观察杂合度Ho为0.3233~0.3511, 平均期望杂合度He为0.4421~0.4704, 平均多态信息含量PIC为0.4068~0.4286。对数据进行F-检验, Fst值表明群体间的遗传分化程度中等, 并对基因型进行了基于Hardy-Weinberg平衡的卡方检验, 所得P值说明5个群体均一定程度上偏离了平衡。5个群体间的遗传相似系数为0.8466~0.9146,遗传距离为0.0893~0.1665, 并根据Nei氏标准遗传距离用UPGMA方法对5个鲢群体进行亲缘关系聚类。  相似文献

Common carp (Cyprinus carpio) is an important fish for aquaculture, but genomics of this species is still in its infancy. In this study, a linkage map of common carp based on Amplified Fragment Length Polymorphism (AFLP) and microsatellite (SSR) markers has been generated using gynogenetic haploids. Of 926 markers genotyped, 151 (149 AFLPs, two SSRs) were distorted and eliminated from the linkage analyses. A total of 699 AFLP and 20 microsatellite (SSR) markers were assigned to the map, which comprised 64 linkage groups and covered 5506.9 cM Kosambi, with an average interval distance of 7.66 cM Kosambi. The normality tests on interval map distances showed a non‐normal marker distribution. Visual inspection of the map distance distribution histogram showed a cluster of interval map distances on the left side of the chart, which suggested the occurrence of AFLP marker clusters. On the other hand, the lack of an obvious cluster on the right side showed that there were a few big gaps which need more markers to bridge. The correlation analysis showed a highly significant relatedness between the length of linkage group and the number of markers, indicating that the AFLP markers in this map were randomly distributed among different linkage groups. This study is helpful for research into the common carp genome and for further studies of genetics and marker‐assisted breeding in this species.  相似文献

Cordyceps sinensis is one of the most valuable medicinal caterpillar fungi native to China. However, its productivity is extremely limited and the species is becoming endangered. The genetic diversity of eighteen C. sinensis populations across its major distributing regions in China was evaluated by inter-simple sequence repeat (ISSR) markers. A total of 141 markers were produced in 180 individuals from the 18 populations, of which 99.3% were polymorphic. The low average of Shannon (0.104) and Nei index (0.07) of the 18 populations indicates that there are little genetic variations within populations. For all 18 populations, estimates of total gene diversity (HT), gene diversity within populations (HS), coefficient of genetic differentiation (GST), and gene flow (Nm) were 0.170, 0.071, 0.583, and 0.357, respectively. This pattern suggests that the genetic diversity of C. sinensis is low and most of the ISSR variations are found among populations with little gene exchange. The 18 populations are divided into five groups based on the genetic distance and the grouping pattern matches with the geographic distribution along the latitudinal gradient. The five groups show obvious difference in the GST and Nm values. Therefore, the genetic diversification of C. sinensis populations may be determined by geographic isolation and the combined effects of life history characters and the interaction with host insect species. The information illustrated by this study is useful for selecting in situ conservation sites of C. sinensis.  相似文献

Two populations of Przewalski's naked carp Gymnocypris przewalskii, 30 individuals per population, were screened for 10 microsatellite loci. Moderate allele variation was found in these loci with two to eight alleles per locus. Abstract

We used inter-simple sequence repeat (ISSR) markers to investigate genetic variation in eight natural populations of Elephantopus scaber from South China, including Guangdong, Hainan and Hong Kong. Eleven primers produced 247 bands across all 184 individuals, of which 243 (98.4%) were polymorphic. The average genetic diversity at the species and population levels was estimated to be 0.283 and 0.103, respectively, using mean expected heterozygosity. The average gene differentiation (F ST) among populations was 0.725. AMOVA analysis showed that the partition of molecular variation between and within populations was 72.5% and 27.5%, respectively. The effective number of migrants among populations based on the F ST was relatively low (N m = 0.095). Cluster analysis based on Nei's genetic distance and the neighbour-joining method revealed the genetic relationships among the populations of E. scaber. The Mantel test indicated that there was no significant correlation between population genetic and geographic distances. The results obtained from the AMOVA analysis, the cluster analysis, and the Mantel test all suggested that fragmented local environments and human disturbance might play important roles in shaping the population structure of E. scaber.  相似文献

Castanea squinii Dode, an endemic tree widely distributed in China, plays an important role both in chestnut breeding and forest ecosystem function. The spatial genetic structure within and among populations is an important part of the evolutionary and ecological genetic dynamics of natural populations, and can provide insights into effective conservation of genetic resources. In the present study, the spatial genetic structure of a panmictic natural population of C. sequinii in the Dabie Mountain region was investigated using microsatellite markers. Nine prescreened microsatellite loci generated 29–33 alleles each, and were used for spatial autocorrelation analysis. Based on Moran’s I coefficient, a panmictic population of C. sequinii in the Dabie Mountain region was found to be lacking a spatial genetic structure. These results suggest that a high pollen-mediated gene flow among subpopulations counteract genetic drift and/or genetic differentiation and plays an important role in maintaining a random and panmictic population structure in C. sequinii populations. Further, a spatial genetic structure was detected in each subpopulation’s scale (0.228 km), with all three subpopulations showing significant fine-scale structure. The genetic variation was found to be nonrandomly distributed within 61 m in each subpopulation (Moran’s I positive values). Although Moran’s I values varied among the different subpopulations, Moran’s I in all the three subpopulations reached the expected values with an increase in distances, suggesting a generally patchy distribution in the subpopulations. The fine-scale structure seems to reflect restricted seed dispersal and microenvironment selection in C. sequinii. Diversity in 26 microsatellite loci from section Caulorrhizae germplasm was evaluated by using 33 accessions of A. pintoi Krapov. & W.C. Gregory and ten accessions of Arachis repens Handro. Twenty loci proved to be polymorphic and a total of 196 alleles were detected with an average of 9.8 alleles per locus. The variability found in those loci was greater than the variability found using morphological characters, seed storage proteins and RAPD markers previously used in this germplasm. The high potential of these markers to detect species-specific alleles and discriminate among accessions was demonstrated. The set of microsatellite primer pairs developed by our group for A. pintoi are useful molecular tools for evaluating Section Caulorrhizae germplasm, as well as that of species belonging to other Arachis sections.  相似文献

Nineteen microsatellite markers were employed to evaluate the genetic diversity of 92 accessions of common wild rice Oryza rufipogon Griff., which represent a significant portion of the distribution range from field gene banks of China. In comparison, a total of 57 varieties from most of the rice growing areas in China were also analyzed. The microsatellite analysis revealed a considerable amount of genetic diversity resided within the preserved wild rice germplasms. In all, the nineteen microsatellites revealed 328 alleles. The number of alleles per locus varied widely among these markers, ranging from 6 at RM242 to 30 at RM206. A comparison of the genetic parameters showed that wild rice strains preserved in the field gene banks (na = 17.27; R S = 15.66; H S = 0.86; H T = 0.852; H O = 0.307) possess much higher genetic diversity than cultivated rice varieties (na = 8.27; R S = 8.14; H S = 0.75; H T = 0.758; H O = 0.051). A total of 196 alleles detected in the wild rice could not be found in cultivated rice, suggesting that about 60% of the alleles of wild rice might be lost during the process of rice domestication. This result shows that these ex situ preserved wild rice strains are of great importance for the discovery and utilization of novel genes in the future rice breeding practices. Considerably abundant genetic variability detected within the studied wild rice germplasms could be comparable to that previously found in a wide sampling of 47 natural populations (na = 16.17; H S = 0.67; H O = 0.229), demonstrating that developing field gene banks of wild rice is a necessary and efficient way for preserving genetic diversity of wild rice resources. To determine minimum microsatellites that could distinguish these wild rice accessions, the phylogenetic trees constructed by means of the combinations of different microsatellites suggested that the five highly polymorphic microsatellites could clearly identify these samples. High polymorphisms of rice microsatellite loci and their great resolving power will be particularly helpful for germplasm evaluation and evolutionary studies for better strengthening the conservation and utilization of genetic diversity of wild rice in the field gene banks.  相似文献

The Toll-like receptor 7 (TLR7) is activated by single strand RNA and RNA-like compounds (imidazoquinoline), and it induces interferon production. We identified and described carp TLR7 cDNA and its mRNA expression. The full-length cDNA of carp TLR7 gene is 3427 bp, encoding 1049 amino acids (AB553573). The similarities of carp TLR7 with zebrafish, rainbow trout, fugu, and human TLR7 were 89.6, 83.4, 80.6 and 74.6%, respectively, at the amino acid sequence level. Furthermore, the expression of TLR7 mRNA was investigated in normal tissues of carp by semi-quantitative RT-PCR analysis. Carp TLR7 expression was exhibited in healthy tissues (kidney, brain, spleen, skin, intestine, muscle, liver, gills and heart) and though the expression level in each tissue varied among healthy fish. Carp TLR7 expression was significantly increased in head kidney stimulated with TLR7 agonist, imiquimod, at 8, 24 and 48 h in vitro when compared to expression in the control group. Moreover, carp head kidney leukocytes produced elevated levels of pro-inflammatory and type 1 interferon cytokine mRNA in response to imiquimod stimulation.  相似文献

Population bottlenecks occur frequently inthreatened species and result in loss ofgenetic diversity and evolutionary potential.These may range in severity between shortintense bottlenecks, and more diffusebottlenecks over many generations. However,there is little information on the impacts ofdifferent types of bottlenecks and disagreementas to their likely impacts. To resolve thisissue, we subjected replicate Drosophilapopulations to intense bottlenecks, consistingof one pair over a single generation, versusdiffuse bottlenecks consisting of an effectivesize of 100 over 57 generations. The intenseand diffuse bottlenecks were designed to induceidentical losses of heterozygosity. However,computer simulations showed that theprobability of retaining alleles is lower inthe intense than the diffuse bottlenecktreatment. The effects of these bottlenecks ongenetic diversity at nine microsatellite lociin Drosophila were evaluated. Bottleneckssubstantially reduced allelic diversity,heterozygosity and proportion of locipolymorphic, changed allele frequencydistributions and resulted in large differencesamong replicate populations. Allelic diversity,scaled by heterozygosity, was lower in theintense than the diffuse treatments. Short-termevolutionary potential, measured as the abilityof bottlenecked populations to cope withincreasing concentrations of NaCl, did notdiffer between the intense and diffusebottlenecked populations. The effects ofbottlenecks on short-term evolutionarypotential relate to loss of heterozygosity,rather than allelic diversity.  相似文献

Wild common carp from two lakes and two rivers in Greece were genetically characterized with sequencing analysis of two mitochondrial DNA segments: cytochrome b (1119 bp) and D-loop (646 bp). A total of 9 variable singleton sites and 7 unique haplotypes were detected. A common haplotype was found in three out of the four populations examined, which seems to be the ancestral one and represents the European origin of common carp from Greece. This haplotype could be also justified by the introductions reported with individuals belonging to the Central European race, into many natural habitats in Greece. Limited genetic variation — in Evros and Aliakmonas populations — could be due to bottleneck effects and small effective population sizes, whereas the different haplotypes found in Lake Volvi could represent different common carp stocks. Values of sequence divergence among Greek haplotypes ranged from 0.0006 to 0.0023. The Neighbour-Joining (NJ) phylogenetic tree constructed based on the combined sequences, reveals that the populations of common carp from Greece belong to the European group of populations — which is highly divergent from the South East-Asia cluster — and to the subspecies Cyprinus carpio carpio.  相似文献

Broadening of the genetic base and systematic exploitation of heterosis in faba bean requires reliable information on the genetic diversity in the germ plasm. Three groups of faba bean inbred lines were examined by means of RAPDs (random amplified polymorphic DNAs) assays: 13 European small-seeded lines, 6 European large-seeded lines, and 9 Mediterranean lines. Out of 59 primers, 35 were informative and yielded 365 bands, 289 of which were polymorphic with a mean of 8.3 bands per primer. Monomorphic bands were omitted from the analyses and genetic distances (GD) were estimated via the coefficient of Jaccard. The mean GD among the European small-seeded lines was significantly greater than those among the lines of the other two groups. Repeatability of GD estimates was high. Cluster (UPGMA) and principal coordinate analyses identified European small-seeded lines and Mediterranean lines as distinct groups with European large-seeded lines located in between. The results are in harmony with published archaeobotanical findings. We conclude that RAPDs are useful for classification of germ plasm and identification of divergent heterotic groups in faba bean.  相似文献

Anthropogenic habitat alteration interferes the natural aquatic habitats and the system''s hydrodynamics in the Yangtze River floodplain lakes, resulting in a serious decline in freshwater biodiversity. Zooplankton communities possess major position in freshwater ecosystems, which play essential parts in maintaining biological balance of freshwater habitats. Knowledge of processes and mechanisms for affecting variations in abundance, biomass, and diversity of zooplankton is important for maintaining biological balance of freshwater ecosystems. Here, we analyzed that the temporal and spatial changes in the structure of zooplankton community and their temporal and spatial variations respond to changes in environmental factors in the middle reach of Yangtze River floodplain lakes. The results showed that zooplankton samples were classified into 128 species, and Rotifera was the most common taxa. Significant seasonal differences were found among the abundance and diversity of zooplankton. Similarly, we also found significant seasonal differences among the biomass of zooplankton functional groups. The spatial turnover component was the main contributor to the β diversity pattern, which indicated that study areas should establish habitat restoration areas to restore regional biodiversity. The NMDS plot showed that the structure of zooplankton community exhibited significant seasonal changes, where the community structure was correlated with pH, water temperature, water depth, salinity, total nitrogen, chlorophyll‐a, and total phosphorus based on RDA. This study highlights that it is very important to ensure the floodplain ecosystem''s original state of functionality for maintaining the regional diversity of the ecosystem as a whole.  相似文献
